We present a method for increasing the accuracy and acquisition rate of the initial location data of plants within fixed areas. Using a personal digital assistant (PDA) to create a link between various electronic measurement devices (pantograph, micrometers etc.) and a database of the study individual's location information, has significantly increased measurement reliability and speed in a demographic field study. The method also provides a means to quickly and accurately re-identify the same individuals during subsequent visits to the study plot. Application of the same method can be used in all ecological field studies with sessile organisms in permanent plots, increasing speed and accuracy of coordinate measurement.  相似文献

Chloroplast DNA (ctDNA) from the marine chromophytic alga, Olisthodiscus luteus, has been isolated using a whole cell lysis method followed by CsCl-Hoechst 33258 dye gradient centrifugation. This DNA, which has a buoyant density of 1.691 grams per cubic centimeter was identified as plastidic in origin by enrichment experiments. Inclusion of the nuclease inhibitor aurintricarboxylic acid in all lysis buffers was mandatory for isolation of high molecular weight DNA. Long linear molecules (40 to 48 micrometers) with considerable internal organization comprised the majority of the ctDNA isolated, whereas supertwisted ctDNA and open circular molecules averaging 46 micrometers were occasionally present. Also observed in this study were folded ctDNA molecules with electron dense centers (“rosettes”) and plastid DNA molecules which have a tightly wound “key-ring” center. The ctDNA of Olisthodiscus has a contour length that is median to the size range reported for chlorophytic plants.  相似文献

Digital fringe projection (DFP) techniques provide dense 3D measurements of dynamically changing surfaces. Like the human eyes and brain, DFP uses triangulation between matching points in two views of the same scene at different angles to compute depth. However, unlike a stereo-based method, DFP uses a digital video projector to replace one of the cameras1. The projector rapidly projects a known sinusoidal pattern onto the subject, and the surface of the subject distorts these patterns in the camera’s field of view. Three distorted patterns (fringe images) from the camera can be used to compute the depth using triangulation.Unlike other 3D measurement methods, DFP techniques lead to systems that tend to be faster, lower in equipment cost, more flexible, and easier to develop. DFP systems can also achieve the same measurement resolution as the camera. For this reason, DFP and other digital structured light techniques have recently been the focus of intense research (as summarized in1-5). Taking advantage of DFP, the graphics processing unit, and optimized algorithms, we have developed a system capable of 30 Hz 3D video data acquisition, reconstruction, and display for over 300,000 measurement points per frame6,7. Binary defocusing DFP methods can achieve even greater speeds8.Diverse applications can benefit from DFP techniques. Our collaborators have used our systems for facial function analysis9, facial animation10, cardiac mechanics studies11, and fluid surface measurements, but many other potential applications exist. This video will teach the fundamentals of DFP techniques and illustrate the design and operation of a binary defocusing DFP system.  相似文献

Although the consequences of Ras activation have been studied extensively in the context of oncogenesis, its regulation in physiological modes of signal transduction is not well understood. A fluorescent indicator, Raichu-Ras, was fused to the C-terminal hypervariable regions of H-Ras and K-Ras to create indicators for Ras activation within caveolae/rafts (Raichu-tH) and non-raft domains (Raichu-tK) of the plasma membrane, respectively. Raichu-tH was also found abundantly in endomembranes. To monitor Ras activation with high spatial resolution, it is imperative to observe sectioned images of the signals. We have developed a wide-field fluorescence microscope equipped with a digital micromirror device (DMD) to acquire optically sectioned images using fringe projection. This system provides reliable signals from fluorescence resonance energy transfer (FRET) between cyan and yellow mutants of green fluorescent protein. We have used this system to demonstrate that, upon stimulation with growth factors, the two indicators are activated in spatially and temporally unique patterns.  相似文献

Sarcocysts were found by light microscopic examination of muscle in 199 (51%) of 390 white-tailed deer (Odocoileus virginianus) from the southeastern United States. Sarcocystis infections were detected more frequently in histologic sections of tongue (45%) than of heart (9%). Sarcocysts were significantly more prevalent in adult deer (54%) than fawns (26%) (P less than .01). Statistically significant differences in prevalence were not found in deer from different physiographic provinces or between sexes. Artificial digestion was more sensitive in detecting Sarcocystis infections than examination of histologic sections when both techniques were used to examine tongues of 35 deer. Three different size sporocysts, possibly representing at least two species of Sarcocystis, were recovered during feeding trials. Seven dogs (Canis familiaris) shed sporocysts 9 to 12 days after eating infected venison. Sporocysts measured 13.4-16.8 x 9.0-12.3 micrometers with an average measurement of 15.2 x 10.9 micrometers (N = 195). One of three cats (Felis catus) and one of two red foxes (Vulpes vulpes) first shed sporocysts of Sarcocystis 10 days after eating infected venison. Sporocysts from the cat measured 11.2-13.4 x 6.72-8.96 micrometers (avg 12.0 x 8.7 micrometers, N = 18), and those from the fox measured 11.2-15.7 x 9.0-11.2 micrometers (avg 13.6 x 10.2 micrometers, N = 7).  相似文献

An automated device for reading the fringe patterns produced by interference optics in an analytical ultracentrifuge has been designed and tested. The device significantly reduces the time and work required to read these photographs while improving the precision over manual reading. Fringe edges are detected by a differential system and fringe centers estimated from the edge positions. Both semiautomatic and automatic modes of operation are described. In the latter, the device has been interfaced to a PDP-7 minicomputer.  相似文献

Summary The present study describes for the first time the development of early acellular extrinsic fiber cementum (AEFC) until its establishment on human teeth. Precisely selected premolars with roots developed to 50%–100% of their final length were prefixed in Karnovsky's fixative and most of them were decalcified in EDTA. Their roots were subdivided into about 10 blocks each, cut from the mesial and distal root surfaces. Following osmication, these blocks were embedded in Epon and sectioned for light-and transmission electron microscopy. Some blocks were cut non-demineralized. From semithin stained sections, the density of the collagenous fiber fringe protruding from the root surface was measured by using the Videoplan-system. After initiation of this fiber fringe and its attachment to the dentinal root surface followed by mineralization, the fringe gradually increased in length and subsequently became mineralized. Fringe elongation and the advancement of the mineralization front appeared to progress proportionally. Thus, in all stages of AEFC development, a short fiber fringe covered the mineralized AEFC. Its density remained constant, irrespective of AEFC thickness. The latter gradually increased and reached an early maximum of 15–20 m in the cervical region. At this stage, the AEFC fringe appeared to fuse with the future dentogingival or other collagen fibers of the tooth supporting apparatus. Telepathology, the practice of pathology at a long distance, has advanced continuously since 1986. Today, fourth-generation telepathology systems, so-called virtual slide telepathology systems, are being used for education applications. Both conventional and innovative surgical pathology diagnostic services are being designed and implemented as well. We have a successful experience in Egypt in applying the static & dynamic techniques in a pilot project between the Italian Hospital in Cairo (NPO) and the Civico Hospital in Palermo This project began in 2003 and continued till now. In 2004, centers in Venice, London and Pittsburgh participated actively in our project. During the past seven years we consulted on many problematic pathological cases with these different specialized pathological centers in Italy, UK & USA. In addition to the highly specialized scientific value of consulting on the cases and exchanging knowledge, we saved a lot of time and money and succeeded in providing our patients with a better medical service. In view of this success we have already established a new Digital Telepathology unit (DTU) in the pathology department, Cairo University, using the latest technique of telepathology which is Whole Slide Imaging (WSI) since one year. This unit is considered the first Digital pathology unit in all the universities of the whole Middle East. During the passed year we created a digital pathology library for the under graduate students using the WSI technique and changed the teaching method of the histopathology slides to be completely digital. We are building another digital pathology library (for post graduate candidates) which will be available to all pathology candidates in Egyptian universities & universities in the surrounding Arabic countries. We are also creating a digital pathology network between pathology centers in the Middle East for exchanging knowledge & telepathology.  相似文献

In dentistry, mechanical articulators with which mandibular movements can be reproduced in dentals casts play a major role. Commonly used semiadjustable articulators, however, have major limitations: On the one hand, the movement of the mandible is not reproduced exactly, on the other, they do not provide time-related information on jaw movement. Both problems can be solved by replacing the mechanical articulator by a digital simulation ("virtual articulator") based on digitized plaster casts and electronically recorded masticatory movements. We present a system for the 3D measurement of plaster casts in a skull-related, anatomical coordinate system using the fringe projection technique, and electronically recorded condylar movements. Using numerical algorithms, the contacts between upper and low jaw, and the angle of rotation of the temporomandibular joint can be computed for each movement in dynamic occlusion. Taking the data recorded from a patient as an example, the influence of the accuracy of the digitization of plaster casts on the computation of the rotation of the temporomandibular joint is discussed in relation to the anatomy of the masticatory apparatus.  相似文献

The conventional method of studying mass transport in membranes by spot photobleaching and then following the recovery of fluorescence has disadvantages. Among them, the need for a high density of fluorescent molecules, the measurement of the beam profile, and a knowledge of the photobleaching processes are of a crucial importance. The application of a planar fringe pattern of light both for the bleaching and the monitoring of the fluorescent molecules solves these three major difficulties. Brownian diffusion coefficients and flow velocities can be measured independently and are averaged over the whole fringe pattern volume. These transport coefficients are explored over the wide range of experimentally accessible distances (from an interfringe spacing 0.5-50 micron). The quantification of the mobile and immobile components is further simplified by scanning the fringe pattern and detecting only a modulated fluorescence recovery signal. The fringe pattern photobleaching method is particularly adapted to the measurements of diffusion coefficients and flow velocity of membrane components, as well as of cytoplasmic proteins. The theoretical results and the test experiments with fluorescent bovine serum albumin are described.  相似文献

The effect of short fiber reinforcements on the mechanical properties of cement has been examined using a splitting tensile – digital image correlation (DIC) measurement method. Three short fiber reinforcement materials have been used in this study: fiberglass, nylon, and polypropylene. The method outlined provides a simple experimental setup that can be used to evaluate the ultimate tensile strength of brittle materials as well as measure the full field strain across the surface of the splitting tensile test cylindrical specimen. Since the DIC measurement technique is a contact free measurement this method can be used to assess sample failure.  相似文献

The ellipsoidal to slightly ovoid oocysts of Eimeria caprina sp. n. from the domestic goat, Capra hircus, are 27--40 x 19.5--26 micrometers (mean 31.8 x 23.1 micrometers); their sporocysts are 13--17 x 7--10 micrometers (mean 15.3 x 8.5 micrometers). The oocyst wall is 1.7 micrometers thick, smooth, dark brown to brownish-yellow, and 2-layered. Micropyle, polar granule, and sporocyst residuum are present; micropylar cap and oocyst residuum are absent. These features distinguish the new species from other species in the genus which have a micropylar cap, or are smaller, or have a small micropyle with a small internal plug.  相似文献

Various ways of applying differential interferometry to ultracentrifugal analyses are examined and several analytical techniques are established. In transport and moving boundary methods, the sedimentation coefficient is more precisely determined in the differential interference system than in the schlieren optical system because fringe measurement accuracy is much higher in the former system. Compared to interference and absorption optics, the differential interferometer provides a more exact s value in the transport method since an accurate calculation procedure can be adopted. Moreover, the following advantages of differential interferometry are noted. Determination of the initial solute concentration, which must be done in the usual interference method, is unnecessary in this sedimentation equilibrium method. Regardless of the partial loss of solute from the observed system due to rapid precipitation or adsorption to the cell wall during centrifugation, the molecular weight of the rest of the solute can be determined exactly. The diffusion coefficient can be determined accurately by fringe displacement analysis at the hinge point during the transient state. Together with the molecular weight and diffusion coefficient, the partial specific volume and sedimentation coefficient of a solute can be obtained from the result of a single low-speed centrifugation when the sample solutions in H2O and D2O are compared.  相似文献

The plasmolytic response of Bacillus licheniformis 749/C cells to the increasing osmolarity of the surrounding medium was quantitated with stereological techniques. Plasmolysis was defined as the area (in square micrometers) of the inside surface of the bacterial wall not in association with bacterial membrane per unit volume (in cubic micrometers) of bacteria. This plasmolyzed surface area was zero when the cells were suspended in a concentration of sucrose solution lower than 0.5 M, but increased linearly when the sucrose molarity rose above 0.5 M, reaching a plateau value of 3.61 micrometers2/micrometers3 in 2 M sucrose. In contrast, when the bacterial cells were treated with lysozyme plasmolysis increased abruptly from 0.06 micrometers2/micrometers3 in 0.75 M sucrose to 4.09 micrometers2/micrometers3 in 1 M sucrose. When the time of exposure was prolonged, the degree of plasmolysis increased gradually for the duration of the experiment (30 min) after exposure to 1 M sucrose without lysozyme, whereas with lysozyme plasmolysis reached a maximum (4.09 micrograms2/micrometers3) in 2 to 5 min. The examination of ultrastructure showed that the protoplast bodies of lysozyme-treated cells in 1 M sucrose and untreated cells in 2 M sucrose are maximally retracted from the intact wall of the bacteria; hardly any retraction of protoplasts could be seen for untreated cells in 1 M sucrose. The data suggest that the B. licheniformis cells are isoosmotic to 800 to 1,100 mosM solutions, but are able to withstand much greater osmotic pressure with no signs of plasmolysis because the cell wall and the plasma membrane are held in close association, perhaps by a covalent bond. It is likely that lysozyme weakens this bond by degradation of the peptidoglycan layer. Cellular autolysis also weakens this wall-membrane association.  相似文献

Lunatic fringe is a vertebrate homologue of Drosophila fringe, which plays an important role in modulating Notch signaling. This study examines the distribution of chick lunatic fringe at sites of neural crest formation and explores its possible function by ectopic expression. Shortly after neural tube closure, lunatic fringe is expressed in most of the neural tube, with the exception of the dorsal midline containing presumptive neural crest. Thus, there is a fringe/non-fringe border at the site of neural crest production. Expression of excess lunatic fringe in the cranial neural tube and neural crest by retrovirally mediated gene transfer resulted in a significant increase ( approximately 60%) in the percentage of cranial neural crest cells 1 day after infection. This effect was mediated by an increase in cell division as assayed by BrdU incorporation. Infected embryos had an up-regulation of Delta-1 in the dorsal neural tube and redistribution of Notch-1 to the lumen of the neural tube, confirming that excess fringe modulates Notch signaling. These findings point to a novel role for lunatic fringe in regulating cell division and/or production of neural crest cells by the neural tube.  相似文献

Mitochondria, important energy centers in the cell, also control sperm cell morphogenesis. Drosophila spermatids have a remarkably large mitochondrial formation called the nebenkern. Immediately following meiosis during sperm development, the mitochondria in the spermatid fuse together into two large aggregates which then wrap around one another to produce the spherical nebenkern: a giant mitochondrion about 6 micrometers in diameter. The fused mitochondria play an important role in sperm tail elongation by providing a structural platform to support the elongation of sperm cells. We have identified a novel testis-specific protein, Spermitin (Sprn), a protein with a Pleckstrin homology-like (PH) domain related to Ran-binding protein 1 at its C-terminus. Fluorescence microscopy showed that Sprn localizes at mitochondria in transfected Kc167 cells, and in the nebenkern throughout spermatid morphogenesis. The role of Sprn is unclear, as sprn mutant males are fertile, and have sperm tail length comparable to the wild-type.  相似文献

生态-地理过程(Eco-geographic process,EGP)是现代生态学、地理学与人居环境科学交叉与融合的重要研究方向。阐述了人居环境系统演变的内涵,并指出人居环境系统演变一般是从乡村人居环境,到边缘区人居环境,再到核心区人居环境,由此形成一个不可逆的演替序列。在此基础上,以广州市为例,综合运用生态学与地理学等分析方法,从边缘区与核心区、乡村人居环境系统及外部环境相互作用的角度,将人居环境系统演变的生态-地理过程划分为4类,即入侵(渗透、跳跃、蚕食、延伸)、竞争(分化和共生)、反应和调控,并构建了大城市边缘区人居环境系统演变的生态-地理过程概念模型。生态-地理过程研究对理解大城市边缘区演变机理与生态调控具有重要的理论价值与实践意义。  相似文献

The metameric organization of the vertebrate trunk is a characteristic feature of all members of this phylum. The origin of this metamerism can be traced to the division of paraxial mesoderm into individual units, termed somites, during embryonic development. Despite the identification of somites as the first overt sign of segmentation in vertebrates well over 100 years ago, the mechanism(s) underlying somite formation remain poorly understood. Recently, however, several genes have been identified which play prominent roles in orchestrating segmentation, including the novel secreted factor lunatic fringe. To gain further insight into the mechanism by which lunatic fringe controls somite development, we have conducted a thorough analysis of lunatic fringe expression in the unsegmented paraxial mesoderm of chick embryos. Here we report that lunatic fringe is expressed predominantly in somite -II, where somite I corresponds to the most recently formed somite and somite -I corresponds to the group of cells which will form the next somite. In addition, we show that lunatic fringe is expressed in a highly dynamic manner in the chick segmental plate prior to somite formation and that lunatic fringe expression cycles autonomously with a periodicity of somite formation. Moreover, the murine ortholog of lunatic fringe undergoes a similar cycling expression pattern in the presomitic mesoderm of somite stage mouse embryos. The demonstration of a dynamic periodic expression pattern suggests that lunatic fringe may function to integrate notch signaling to a cellular oscillator controlling somite segmentation.  相似文献
