At fluence rates below 45 W· m-2 cells of the flagellate stage of Haematococcus lacustris react only positively phototactically with a rather high degree of orientation (indicated by r values up to 0.66 with the Rayleigh test). The directedness of orientation decreases with decreasing irradiance. The degree of directedness of the phototactic response depends on the intensity of preirradiation: Low light intensity applied after strong light application results in a dark reaction (low r values), low light given after darkness stimulates a rather high degree of directedness of positive phototaxis. Weak blue light (=483 nm; 0.4 W · m-2) stimulates positive phototactic response, whereas comparable red light (=658 nm; 0.5 W · m-2) does not.Cells which were grown in a medium containing 10-4 M Norflurazon (effective in inhibition of carotenoid biosynthesis) although maintaining motility completely lose the ability to react positively phototactically. The possible role of carotenoids in the phototactic orientation is discussed.  相似文献

The responses to light among the fish Gambusia affinis, permanent residents of hypogean brackish water basins of the Spunnulate dolinas system of Torre Castiglione, in southeastern Italy, were studied at light intensities of 180 and 430 lux. These fish showed, on the average, a negative phototactic behaviour (scotophilia) with a low coefficient of variability. On the other hand, epigean, spring-dwelling individuals of G. affinis from the same region showed a positive phototactic behaviour with a greater variability in the responses when compared with cave-dwelling conspecifics. Possible causes for the detachment of hypogean from epigean forms are discussed.  相似文献

UV-B irradiation has a detrimental effect on the survival of populations of the filamentous cyanobacterium, Phormidium uncinatum, at levels slightly higher than those currently measured at the surface of the earth. The organisms are not damaged or killed by UV-B radiation at 300 nm of 200 Wm-2 for up to 20 h; but slightly increased levels of UV-B irradiation (2 h of 200 Wm-2 at 300 nm) drastically impair motility, phototactic orientation and photophobic responses. These photosynthetic organisms require a narrow light intensity range for growth so that any decrease in their ability to actively search for and move into areas of favorable light conditions is bound to affect the survival of a population. The fluorescence yield of both phycobilins and chlorophyll is not altered even after 20 h of UV-B irradiation (200 Wm-2 at 270 nm) indicating that UV-B at that dose does not affect the photosynthetic apparatus. The organisms are killed either by too bright intensities which bleach the photosynthetic pigments or by the lack of energy when they are unable to avoid moving into dark areas.  相似文献

Sporisorium reilianum, the fungus that causes sorghum head smut, was grown with sucrose, lactose, trehalose or raffinose in liquid suspension or on a solid medium. Liquid culture media were analyzed for hydrolysis products of these carbohydrates to determine extracellular enzyme activity of the fungus. Increased amounts of glucose and fructose in the culture medium ofS. reilianum grown with sucrose or raffinose indicated that invertase (-fructofuranosidase, 3.2.1.26) activity was present. No evidence of extracellular galactosidase or trehalase activity was found. Enhanced sporidial colony formation on carbohydrates that can be hydrolyzed to hexoses, and specific forms of mycelial growth on lactose, trehalose or on a carbohydrate-deficient medium might suggest that mycelial growth is a way of foraging for food sources. However, the rapid and profuse mycelial growth on the host cell wall glycoprotein appears to be in response to abundant food supply (probably of a different type). Therefore availability of different kinds of carbon sources in the environment of the growing fungus might determine dimorphism and associated pathogenesis byS. reilianum.Technical Article No: 30699 from the Texas Agricultural Experiment Station.  相似文献

Zusammenfassung Bei, in kleinen Käfigen mit Zugang zu einer Lauftrommel, einzeln gehaltenen Tieren der beiden ArtenFunambulus palmarum (Palmenhörnchen) undEutamias sibiricus (asiatisches Streifenhörnchen) wurde der Einfluß verschiedener Beleuchtungsstärke unter Dauerlichtbedingungen auf die circadiane Aktivitätsperiodik untersucht.4 Palmenhörnchen wurden in verschiedenen Beleuchtungsstärken von nicht meßbarer Intensität bis etwa 1000 Lux gemessen. Von etwa 1 Lux bis 1000 Lux war die Periodendauer () positiv mit der Beleuchtungsstärke korreliert. Über den gesamten Bereich der untersuchten Lichtintensitäten war die Aktivitätsmenge positiv mit dem Logarithmus der Beleuchtungsstärke (logI LL ) korreliert. Es bestand jedoch ebenso wie zwischen und logI LL keine lineare Beziehung zwischen beiden Größen (Fig. 1). Das Verhältnis Aktivitätszeit zu Ruhezeit (:) war bei allen untersuchten Tieren weder mit noch mit der Aktivitätsmenge korreliert.Von insgesamt 15 unter Dauerlichtbedingungen zwischen 0,01 und 100 Lux gemessenen asiatischen Streifenhörnchen zeigten nur 7 Tiere circadiane Perioden der Aktivität. Infolge der großen Streuung der Aktivitätsanfänge und -enden sowie des Aktivitätsmusters konnten a und : bei den meisten Tieren nicht bestimmt werden. Die -Werte streuten um einen mittleren Wert von 23,6 Std; sie zeigten jedoch keine regelhafte Abhängigkeit von der Beleuchtungsstärke (Fig. 5).Bei einzelnen Tieren beider Arten wurde bei Beleuchtungsstärken von etwa 3–4 Lux eine Aufspaltung (temporäre Dissoziation) einzelner Aktivitätskomponenten mit verschiedenen Frequenzen beobachtet (Figs. 2 und 4).Aufgrund der unterschiedlichen Abhängigkeit der Aktivitätsparameter (Aktivitätszeit und Aktivitätsmenge) von bei verschiedenen Tiergruppen wird vermutet, daß das Licht sowohl verschiedene Parameter der circadianen Aktivitätsperiodik als auch einzelne Komponenten des der Aktivität zugrunde liegenden circadianen Systems unabhängig voneinander beeinflußt. Tag- und nachtaktive Säuger unterscheiden sich im wesentlichen nur in den kritischen Beleuchtungsstärken, d.h. den Beleuchtungsstärken, bei welchen die kürzesten Perioden gemessen werden (Fig. 6).

Summary Palm squirrels (Funambulus palmarum) and Sibirian chipmunks (Eutamias sibiricus) were kept individually in small cages attached to a running wheel. Under continuous illumination the influence of different light intensities on the circadian rhythm of activity was measured.Four palm squirrels were exposed to light intensities between unmeasurable intensity (total darkness) and about 1000 lux. Between 1 and 1000 lux the period length () of the circadian rhythm of activity was positively correlated with light intensity. The amount of activity per period showed a positive relationship with light intensity over the range of intensities tested (Fig. 1). In all 4 animals the ratio activity-time to rest-time (: ) was neither correlated with the amount of activity per period nor with .From a total of 15 Sibirian chipmunks tested under continuous illumination between 0.01 and 100 lux only 7 animals showed circadian rhythms of activity. As a consequence of the great variability of the beginning and the end of activity as well as in the temporal activity pattern, a (activity time) and : could not be determined in most animals of this species. The values of varied around a mean of 23.6 hr, but did not change as a function of light intensity (Fig. 5).In some individuals of both species a splitting or temporary dissociation of single components of activity with different frequencies was observed at light intensities of about 3 to 4 lux (Figs. 2 and 4).On the basis of the different relationships between the activity functions (amount of activity and activity-time) and in various groups of animals (cf. day-active mammals and birds), it is assumed that light acts separately on the parameters of the circadian activity rhythm as well as on single components of the circadian system underlying activity. Day- and night-active mammals differ essentially only in respect to their critical light intensities, i.e. the light intensities at which the shortest circadian periods are measured (Fig. 6).

This preliminary mechanistic model of normal swimming and phototactic behaviour in individual Daphnia was constructed using data and assumptions based on experiments and observations. Swimming under constant light intensity is characterized by short periods of upward movements alternating with equal periods of downward movements. Two oscillators are proposed that generate these phases in swimming. Unexpected shifts in depth, as observed in D. magna and D. longispina, are also present in the swimming of the computer daphnid and thus seem to be inherent to the underlying mechanism. As in real daphnids, during relative decreases in light intensity of low velocity, positive phototactic upward swimming is stepwise. With increasing velocity in the change in light, these steps disappear. When the model is triggered by a natural increase in light at dawn, a small downward movement results. Migration distance can be increased to commonly found depths of migrating Daphnia by the introduction of a fish exudate factor into the model, which enhances the phototactic response. Since attenuation of light in the water affects the phototactic swimming response, it also influences migration distance. The results of model calculations agree quite well with an empirical relationship between Secchi disc depth and amplitude of diel vertical migration in a number of lakes.  相似文献

Summary Feeding of Styropor lamellae treated with 0.25 M maltose, sucrose, raffinose or melezitose byS. littoralis larvae induced amylase activity to 5–6 fold and invertase and protease activity to 2–3 that of the control. Melibiose induced amylase activity to about 4-fold and invertase and protease activity to 188% and 173%, respectively, that of the control. -lactose stimulated the various digestive enzymes to various extents; cellobiose had practically no effect. Glucose and fructose induced the various digestive enzymes to about the same level but to a much higher extent than galactose.The feeding rate of the various sugar compounds except maltose, correlates well with their stimulating activity on the digestive enzymes. Maltose, which induced the larval digestive enzymes to about the same level as that of sucrose, elicited a considerably lower feeding response. The excretion index of sugars stimulating digestive enzymes and eliciting a feeding response, such as maltose, sucrose, raffinose and melezitose, is considerably lower than that of the other sugars.In a comparative test, sucrose, maltose and raffinose induced the various digestive enzymes to about the same level as their equivalent monosaccharide components. Summary The spermatozoon of the holothurian Leptosynapta clarki has a small circular head measuring about 3.0 at the greatest diameter, a midpiece containing a single mitochondrion and a tail flagellum measuring between 35 and 45 in length. The acrosomal region contains a granule measuring 0.7 in diameter which consists of electron dense material arranged in concentric lamellae. Five concentric very electron dense lamellae alternate with areas of much less electron dense material in the central region of the granule. This granule rests in an anterior nuclear depression.The nucleus is circular in shape and contains one or two unbound vacuoles which frequently contain a fine granular material. Posteriorly the nucleus is bounded by a large mitochondrion and an occasional Golgi complex. The proximal centriole which contains a lateral arm of dense material lies in a deep fossa projecting into the nucleus. The distal centriole lies posteriorly in the mitochondrial mass and gives rise to nine satellite projections and their Y-shaped connective extensions.The tail contains the 9 + 2 tubule arrangement and tapers at its distal end.This investigation was supported by a National Research Council grant to F. Abstract

In Marchantia palmata Nees, normal growth of thalli occurs on Nitsch's basal medium with 1% sucrose. With 4% sucrose, dark green callus is initiated by light intensities of 150–4,500 lux after 13–15 days of growth. In the dark only rhizoids develop from callus and at low light intensity (150 lux) rhizoids as well as knob-like green outgrowths differentiate. Increased light intensities induce differentiation of thalli and rhizoids from callus. The callus inducing effect of increased sucrose level can be replaced by higher concentrations of mannitol. Histological studies reveal that callus initially arises from the lower epidermis.  相似文献

Although light trap can be used to control pest populations, they can also kill the natural enemies of pests. Scleroderma guani (Hymenoptera: Bethylidae) is a parasitoid of a bark-weevil Pissodes punctatus (Coleoptera: Curculionidae). To understand the phototactic behavior of S. guani, we investigated its diurnal and nocturnal behavior, then examined its phototactic response to nine monochromatic lights and to five intensities of the two most attractive lights. Our results showed that S. guani is most active during the day, while remain still in a dark room or at night. S. guani showed a positive response both to a broad spectrum of monochromatic light and total light (natural light), which implies a broad sensitivity to the light spectrum. S. guani was most sensitive to blue (450 nm) and green (549 nm) lights, suggesting its visual system composed of blue and green receptors. S. guani was least sensitive to ultraviolet (340 nm) light, which may be caused by long-term mass rearing and propagating under artificial conditions. Furthermore, low intensities elicited a positive phototactic response, while high intensities showed a decreased trend under both blue and green lights. Thus, S. guani is a phototactic insect which shows preferences for light in both color and intensity. This study suggests that light trap can only be utilized to control the adult P. punctatus during and after its peak emergence, due to the overlap in the spectral sensitivity of both pest and parasitoid adults.  相似文献

Forty-six strains of tempe-forming Rhizopus species were screened for their ability to grow on raffinose as the sole carbon source. Six of the strains showed good growth and sporulation. These isolates were one Rhizopus oligosporus, one Rhizopus microsporus var. chinensis, three Rhizopus oryzae and one Rhizopus stolonifer. These six moulds and R. oligosporus strain NRRL 2710 were investigated for their metabolism of the raffinose family of -galactoside carbohydrates. Degradation experiments were performed in submerged culture in a medium containing soybean -protein, sodium phytate and either stachyose, raffinose or melibiose. R. oryzae and R. stolonifer completely consumed the tested carbohydrates as carbon source. R. microsporus var. chinensis failed to hydrolyse the -galactosidic bonds of raffinose, stachyose or melibiose, whereas it was able to use sucrose and the fructose moiety of raffinose or stachyose for growth. R. oligosporus NRRL 2710 was unable to hydrolyse any of the tested carbohydrates. The results of the oligosaccharide degradation experiments could be verified during tempe production from soybeans with the selected fungal species.  相似文献

A culture system for the rotifer Brachionus plicatilis was designed to maintain higher food conversion rates and stable population densities. Two 2001 plastic tanks were employed in the culture experiments, tank A for feedback culture and tank B for a control culture. The experiments were carried out for 70 days at 24 °C, light intensity, 1500 lux, and a photoperiod of L:D 15:9. B. plicatilis were fed once a day on baker's yeast and Chlorella.Food conversion rates in tanks A and B were 24.7% and 10.1%, respectively. Population density of B. plicatilis in tank A was consistently stable at 100–150 ind. ml–1 throughout the culture period. Density in tank B, however, showed large fluctuations after 40 or 50 days and by the end of the experiment, declined to zero.  相似文献

The development of chlorosomes and their pigmentation were studied by growing Chloroflexus aurantiacus strain Ok-7o-fl first under conditions under which BChl c-synthesis is low (50°C, 2000 lux and 30°C, 1500 lux) and subsequently under conditions promoting high BChl c-synthesis (50°C, 400 lux). Electron microscopic observations on and chemical analyses of isolated cell components showed that in BChl c-depleted cells chlorosome-like structures (chlorosome bags) are attached to fragments of cytoplasmic membranes. These chlorosome bags exhibit a periodic fine structure caused by the construction of the baseplates of the chlorosomes. The baseplates are closely attached to the cytoplasmic membrane, they are rich in phospholipids and apparently contain a 790 nm-BChl a-complex. Chlorosome bags of BChl c-depleted cells always contain a limited amount of light-harvesting pigment complexes (BChlc, - and -carotene). A new cDNA (Osfruct2) encoding an acid –d-fructofuranosidase from rice has been cloned, sequenced and expressed in Pichia pastoris. The full-length cDNA is 2453 base pairs long and encodes a pre-pro-protein of 682 amino acids. The cDNA fragment coding for mature enzyme was sub-cloned into vector pPICZA for extracellular expression in the methylotrophic yeast. The recombinant product was purified by Ni2+-nitrilotriacetic acid agarose column and biochemically characterized. The enzyme could hydrolyze sucrose and raffinose. Molecular mass is 66 kDa. The activity optimum was at pH 4.8 and 40 °C.  相似文献

The effect of different light intensities on growth, and the amino acid, fat and sugar concentrations ofCaulerpa prolifera (Foerskal) Lamouroux were studied. The results showed that 2,500 lux was the optimum light intensity for growth coupled with a minimum in total free amino acids and maximum in peptides, total soluble sugars and total lipids. Any deviations above or below the optimum light intensity caused a drop in all components studied.  相似文献

Leaf dises of Xanthium pensylvanicum floated on 0.01 M sucrose solutions inside Petri dishes incubated in a growth chamber at 22°G with illiumination of 3000 lux for 16 to 24 hours of light lost 90% of their chlorophyll. Similar dises floated on water under the same conditions lost 60% of their chlorophyll by the end of one week. chlorophyll degradation calculated as percent of the optical density of the original chlorophyll content accelerated with time. A minimum light intensity of 2000 lux is required for such degradation. Higher intensities accelerated chlorophyll degradation. Glucose, ribose, and xylose have effects similar to sucrose on chlorophyll destruction in the presence of light. chlorophyll degradation was found to be temperature sensitive. There is no difference in chlorophyll content in the presence or absence of sucrose at 10 °C.  相似文献

Während der Zugzeiten im Frühjahr und Herbst 1973 und Herbst 1974 wurde unter Laborbedingungen an gekäfigten Garten- und Mönchsgrasmücken (Sylvia borin undS. atricapilla) der Einfluß der nächtlichen Beleuchtungsstärke auf die Zugunruhe untersucht.2. Bei beiden Arten war die Zugunruhe-Menge über den gesamten Intensitätsbereich (0,0000–100,0 Lux) positiv mit der Beleuchtungsstärke korreliert. Eine Optimal-Beleuchtungsstärke für die Zugunruhe konnte für keine der beiden Arten festgestellt werden.3. Die Zugunruhe-Dauer stieg bei beiden Arten bis zu einer Beleuchtungsstärke von 0,005 Lux an und blieb bei weiterem Anstieg der Beleuchtungsstärke konstant.4. Das Schwirren erreichte bei beiden Arten einen Maximalwert für 0,005 Lux. Eine statistisch sicherbare Abnahme des Schwirrens trat erst wieder bei 5,0 Lux auf.5. Die Zugunruhe war bei beiden Arten, unabhängig von den Lichtverhältnissen, symmetrisch um die Nachtmitte verteilt.6. Die Mönchsgrasmücken zeigten bei völliger Dunkelheit keinerlei Aktivität mehr, wogegen die Gartengrasmücken unter dieser Bedingung im Mittel noch 1,5 Stunden pro Nacht schwirrten.7. Gartengrasmücken waren im Mittel pro Nacht länger zugunruhig als die Mönchsgrasmücken.
The influence of light intensity on the migratory restlessness of garden warblers and blackcaps (Sylvia borin andS. atricapilla)
Summary 1. The effects of night light intensity on migratory restlessness during the migratory periods of spring and autumn 1973, and autumn 1974 were investigated under laboratory conditions using caged garden warblers and blackcaps.2. The amount of migratory restlessness was positively correlated with the light intensity in both species within the entire intensity range investigated (0,0000 to 100,0 lux). No optimal value for the effects of light intensity on the amount of migratory restlessness could be determined in either species.3. In both species the duration of the migratory restlessness increased with light intensity to 0,005 lux; further increase in light intensity produced no further increase in duration.4. In both species whirring reached a maximum at 0,005 lux. A statistically significant decrease in whirring first occured at 5,0 lux.5. The migratory restlessness was symmetrically distributed about the middle of the night regardless of the light intensities.6. The blackcaps ceased all activity in complete darkness, while the garden warblers showed an average of 1,5 hours of whirring per night under the same conditions.7. The garden warblers showed on average longer migratory restlessness per night than the blackcaps.

Cloning and characterization of an exoinulinase from Bacillus polymyxa   总被引:2,自引:0,他引:2  
A gene encoding an exoinulinase (inu) from Bacillus polymyxa MGL21 was cloned and sequenced. It is composed of 1455 nucleotides, encoding a protein (485 amino acids) with a molecular mass of 55522 Da. Inu was expressed in Escherichia coli and the His-tagged exoinulinase was purified. The purified enzyme hydrolyzed sucrose, levan and raffinose, in addition to inulin, with a sucrose/inulin ratio of 2. Inulinase activity was optimal at 35°C and pH 7, was completely inactivated by 1 mM Ag+ or Hg2+. The K m and V max values for inulin hydrolysis were 0.7 mM and 2500 M min–1 mg–1 protein. The enzyme acted on inulin via an exo-attack to produce fructose mainly.  相似文献

Porphyra vietnamensis Tanaka & Pham-Hoang Ho is a tropical species of high potential for farming. Studies of the life cycle have been conducted for many years but have not been successful until recently. Mature thalli were collected from Songkhla, in the southern part of Thailand, and were used to obtain Conchocelis in the laboratory in Bangkok. Conchocelis in shells as well as free-floating filaments could be observed after one week of incubation at 25 °C, 25 salinity and 350–500 lux light intensity, and covered the culture shell surface within 2 months. Conchosporangia were formed after being incubated for 10 days at 30 °C, 20 salinity under light intensities of 350–500 lux with a photoperiod of 12 hours a day. Induction of conchospore release was achieved by lowering the temperature to 25 °C and the salinity to 10–15 and increasing the light intensity to 800–1000 lux. Liberated conchospores germinated into young thalli which became mature after 70 days.  相似文献

Changes in soluble carbohydrate composition and concentration in leavesand petioles of watermelon (Citrullus lanatus (Thunb)Matsum and Nakai cv. Sugar Baby) seedlings during early stages ofphytochrome-regulated petiole elongation were investigated. Watermelon seedlingswere grown in a controlled environment with 350 molm–2 s–1 photosynthetically activeradiation (PAR) during a 12-h photoperiod. Low intensity end-of-day(EOD) light treatments (for 15 min) of red (R), far-red (FR) and FRfollowed by R (FR/R) were initiated when the seedlings were 14 days old.Seedling growth, and soluble carbohydrate concentration and composition inleaves and petioles were determined after 3 and 6 days of EOD light treatments.The EOD FR increased the petiole length and dry mass partitioned to petioles asearly as 3 days into the treatment. This increased petiole dry mass inFR-treated plants was accompanied with an increase in reducing sugar (glucoseand fructose) concentration in the petioles. Although both leaves and petiolesshowed this effect, the relative increase was greater in petioles than leaves.While the most abundant sugars in petioles were fructose and glucose, thepredominant sugars in leaves were sucrose, raffinose, and stachyose. Thephotoreversion of FR induced changes in growth and sugar concentrations by Rindicates the involvement of phytochrome in these processes.  相似文献
