首次以体积排阻液相色谱法进行质粒提取液浓度的测定研究,通过纯质粒溶液在260 nm处的紫外吸峰面积与质粒浓度之间数据的线性最小二乘拟合,得到了反映质粒浓度和紫外吸收峰峰面积之间关系的标准曲线:Y=2×10~(-5) X,通过验证实验,确定了质粒提取液中不同组分,如核糖核酸,蛋白质等在色谱图中的出峰位置.结合标准曲线测定了自制的质粒提取液的质粒浓度为0.32 mg/mL.该方法方便,快捷,准确.  相似文献

Recently, it was shown that the cell surface antigen defined by the monoclonal antibody 12E7 is expressed by both the human X and Y chromosomes; the gene loci on the X and Y chromosomes are referred to as MIC2X and MIC2Y, respectively. It was also shown that MIC2X is located in the region Xp22.3----Xpter and MIC2Y is in the region Ypter-Yq1.1. Here, we show that MIC2X escapes X-inactivation on structurally normal and abnormal inactive human X chromosomes.  相似文献

在胚泡表面表达的Lewis寡糖抗原 (LewisX ,LewisY)在胚胎发育以及着床过程中起重要作用 .应用免疫印迹和免疫荧光等方法对着床前小鼠胚泡表面的Lewis寡糖抗原进行分析 .结果发现 :小鼠胚泡LewisX寡糖蛋白有 2 7kD、2 9kD、6 8kD和 80kD 4种 ,LewisY寡糖蛋白有 70kD和 90kD 2种 ;2种寡糖抗原均在 8细胞时期开始表达 ,其中 ,LewisY寡糖抗原在胚泡表面的表达持续升高 ,直至胚泡着床 ;而LewisX寡糖抗原的表达则在桑椹期后逐渐降低 ,但仍在胚胎期的囊胚腔侧的顶端可见有部分表达 ;应用RT PCR的分析结果显示 ,LewisX合成的关键糖基转移酶FUT9基因在 4细胞及桑椹期高表达 ,到胚泡期虽然强度明显减弱 ,但仍有表达 ;而LewisY合成关键酶FUT1基因在 4细胞未见表达 ,在桑椹和胚泡阶段均有表达并逐渐升高 ,表达趋势与相应寡糖的表达趋势基本一致 .结果说明 ,在小鼠胚泡表面表达的Lewis寡糖抗原的表达受到相应糖基转移酶基因转录的调控  相似文献

Aims

Signal averaged electrocardiography is a noninvasive method to evaluate the presence of the potentials that are generated by tissues, activated later than their usual timing in the cardiac cycle. The purpose of this study was to demonstrate the correlation of data obtained via signal averaged electrocardiography and left ventricular dyssynchrony.

Methods

We included the patients with advanced systolic left ventricular dysfunction (ejection fraction ≤ 35%) and intraventricular conduction delay. All patients underwent surface 12-lead electrocardiography, signal averaged ECG, and tissue Doppler echocardiography.

Results

The study included 72 patients with mean age of 56.45±13.59 years. Mean QRS duration was 0.14 ± 0.02 sec; 63.9% of patients had left bundle branch block. Linear regression demonstrated significant correlations between filtered QRS duration and interventricular mechanical delay (P<0.000, Y= 0.41X-24.76), root mean square 40 and peak velocity difference (P: 0.001, Y=-0.39X+109.72), root mean square 40 and Ts-SD-12 (P:0.026, Y=-o.26X+40.08), low amplitude signals duration and peak velocity difference (P<0.000, Y=0.44X+67.3) and finally low amplitude signals duration and Ts-SD-12 (P:0.31, Y=0.26X+28.23) as well. Area under the curve in ROC of filtered QRS duration was significant for the detection of interventricular mechanical delay. Areas under the curves in ROC of low amplitude signal duration and root mean square 40 were significant for the detection of peak velocity difference.

Conclusions

Signal averaged electrocardiography can have a role in predicting the amount of ventricular dyssynchrony. The duration of low amplitude signals and root mean square 40 have significant linear relations to some indices of intraventricular dyssynchrony.  相似文献

Hand surface area (HSA) has been utilized for burned skin area estimation in burn therapy, heat exchange in thermal physiology, exposure assessment in occupational toxicology, and the development of manual equipment/ protective gloves in ergonomics. The purpose of this study was to determine the hand surface area to the total body surface area (BSA) and derive a formula for estimating HSA. Thirty-four Korean males (20-60 years old; 158.5-187.5 cm in height; 48.5-103.1 kg in body weight) and thirty-one Korean females (20-63 years old; 140.6-173.1 cm; 36.8-106.1 kg) participated as subjects. The HSA and BSA of 65 subjects were directly measured using alginate. The measurements showed 1) the surface area of the hand had a mean of 448 (371-540) cm(2) for males, and 392 (297-482) cm(2) for females. 2) The hand as a percentage of the total body surface area for males and females was 2.5% and 2.4% respectively, showing no significant difference. 3) The hand as a percentage of BSA by body shape was 2.5% for the lean group and 2.3% for overweight people (p=0.001). 4) When estimating the surface area of a hand, formulae based on hand length or hand circumference were more valid than formulae based on height and body weight. We obtained the following formula for estimating HSA: Estimated HSA(cm(2))=1.219 Hand length(cm) x Hand circumference(cm).  相似文献

We wanted to study the expression of P2-receptors at the mRNA-level in the heart and if it is affected by congestive heart failure (CHF). To quantify the P2 receptor mRNA-expression we used a competitive RT-PCR protocol which is based on an internal RNA standard. The P2 receptor mRNA-expression was quantified in hearts from CHF rats and compared to sham-operated rats. Furthermore, the presence of receptor mRNA was studied in the myocardium from patients with heart failure. In the sham operated rats the G-protein coupled P2Y-receptors were expressed at a higher level than the ligand gated ion-channel receptor (P2X1). Among the P2Y-receptors the P2Y6-receptor was most abundantly expressed (P2Y6 > P2Y1 > P2Y2 = P2Y4 > P2X1). A prominent change was seen for the P2X1- and P2Y2-receptor mRNA levels which were increased 2.7-fold and 4.7-fold respectively in the myocardium from the left ventricle of CHF-rats. In contrast, the P2Y1-, P2Y4- and P2Y6-receptor mRNA levels were not significantly altered in CHF rats. In human myocard the P2X1-, P2Y1-, P2Y2-, P2Y6- and P2Y11-receptors were detected by RT-PCR in both right and left atria and ventricles, while the P2Y4-receptor band was weak or absent. In conclusion, most of the studied P2-receptors were expressed in both rat and human hearts. Furthermore, the P2X1- and P2Y2-receptor mRNA were upregulated in CHF, suggesting a pathophysiological role for these receptors in the development of heart failure.  相似文献

Suaeda physophora was distributed on the saline soil with a salt contents 3.0%~ 6.0% in the surface layer (0420 cm underground). The plant height was 45~60 cm and the average length of the one-year twig was 12 cm. The succulent leave was strip shaped. The well developed lateral roots was distributed in the deeper soil (EOb 60 cra) with decreased salt contents and increased water content. The multiregression model for individual plant's aboveground biomass was: Y = 312. 00 + 3.44X1 + 4. 85X2 - 2.16X3 (Xl, length; X2, width: X3, height). The contents of Na, S and K in leave of S. physophora were very high. That of Na was 91255 μg · g-1. The element contents varied in the different parts of plant. K, Na, Mg, S and P contents were the highest respectively in the leaf and ear of flower, but they were the lowest in the old twig. Total salt content of water extracts from tender twig and leaf was 23.17%, being more than in ear of flower and old twig. The element and salt contents varied in different seasons of plant growth. The S. physophora community was a formation on the succulent halophytic dwarf sub-shrub desert. Nine associations were recorded at this area. The coverage of community was 6 % ~25 %.  相似文献

黑肩绿盲蝽Cyrforhinus livisipennis(Reuter)取食白背飞虱卵时,其未成熟期的发育速率与温度呈逻辑斯蒂曲线,世代发育起点和有效积温分别为9‘83℃,359.05日度。在21-29℃时,世代存活率y1,种群内禀增长力Y2:和成虫产卵量y3:粒/雌)较高,高低温区均呈下降趋势,各参数与温度X的关系方程分别为 Y1=cxp(-3.0031+0.5223X-0.0105X2), Y2=-0.5971+0.0535X-0.0010X2,Y3=exp(-12.0791十1.3869X-0.0278X2 理论上26℃时周限增长率又最大,达1.1044倍/天。以褐飞虱卵、白背飞虱卵、褐飞虱低龄若虫和白背飞虱低龄若虫四种食物作为猎物下,黑肩绿盲蝽成虫的产卵量分别为247.63±94.73、237.25±118.62、196.83±69.08、128.14±81.29粒/雌,其卵孵化率分别为59.83、58.26、44.35、46.96%。结果表明,褐飞虱卵是黑肩绿盲蝽发育、存活和繁殖的适宜食物。  相似文献

Cell surface molecules undergo specific changes during apoptosis, including the expression of phosphatidylserine (PS) and some proteins and alterations in sugar chains. Among the various sugar chains on the cell surface, Lewis X (Le(X)) and Lewis Y (Le(Y)) antigens are key determinants for a variety of biological processes. We studied the changes in Le(X) and Le(Y) expression in Jurkat cells, a human T cell line, during apoptosis. Flow cytometry showed that Le(X) and Le(Y) antigen expression was enhanced on the cell surface during apoptosis induced by anti-Fas antibody. To clarify the mechanism of enhanced Le(X) and Le(Y) expression, we assessed the expression levels of fucosyltransferase (FUT1, 2, 3-5-6, 4, and 9) mRNAs that are predominantly expressed in Jurkat cells and which are considered to form Le(X) and Le(Y). The expression of FUT4 mRNA was up-regulated after exposing cells to anti-Fas antibody. Moreover, the increase in Le(X) and Le(Y) antigen levels was significantly suppressed by caspase 3 or 8 inhibitors. These results indicated that the induction of FUT (mainly FUT4), the gene expression of which is mediated by signals downstream of caspase 3, increases Le(X) and Le(Y) expression in apoptotic cells.  相似文献

The gill surface area of 0-group, post-metamorphic Pleuronectes flesus L. was examined using digital image analysis software and expressed in relation to body mass according to the equation log Y=loga+c logW ( a =239·02; c =0·723). The components that constitute gill area, total filament length, interlamellar space and unilateral lamellar area were measured. The measurement of the length of every filament on all eight arches showed that commonly used methods of calculation can lead to an under-estimation of up to 24% of total filament length. Direct measurements of unilateral lamellar area with digital image analysis showed that previously reported gill area data for the same species was over-estimated by as much as 58%. In addition, in this species the neglect of gill pouch asymmetry after metamorphosis, can bring about a 14% over-estimation of total gill area.  相似文献

We investigated the expression and function of P2 receptors and ecto-nucleotidases on human monocyte derived dendritic cells (DC). In addition we analyzed the effect of extracellular ATP on the maturation of DC. By RT-PCR, DC were found to express mRNA for several P2X (P2X1, P2X4, P2X5, P2X7) and P2Y (P2Y1, P2Y2, P2Y4, P2Y5, P2Y6, P2Y10, P2Y11) receptors. As shown by FURA-2 measurement, triggering of P2 receptors resulted in an increase in free intracellular Ca2+. In combination with Tumor necrosis factor-alpha, ATP increased the expression of the DC surface markers CD80, CD83 and CD86 indicating a maturation promoting effect. DC expressed the ecto-apyrase CD39 and the ecto-5'-nucleotidase CD73 as demonstrated by RT-PCR. Extracellular ATP was rapidly hydrolyzed by these ecto-enzymes as shown by separation of 3H-labeled ATP metabolites using a thin layer technique. These data suggest that ATP acts as a costimulatory factor on DC maturation.  相似文献

 根据近几年塔里木河中下游地区的大量监测结果,分析了不同地下水位对植被的影响。结果显示:1)在塔里木河中下游,地下水位与植被盖度和植物种类的回归模型分别可以表示为:Y=159.32e-0.314 8X,R2=0.819 3, p<0.01;Y=9.113e-0.162 3X,R2=0.606 7, p<0.01。2)地下水位对植被的影响在很大程度上是通过改变土壤含水率来实现的,当地下水位在1~4 m时,其回归模型为:Y=64.898e-0.515X,R2=0.727,p<0.01;当地下水位在4~12 m时,Y=21.147e-0.178X,R2=0.658,p<0.01。当地下水位在3.5~4.0 m时,土壤含水率出现明显变化,因此认为3.5 m是塔里木河中下游地区草本植被生态恢复的最低水位。3)通过胡杨叶脯氨酸(Pro)和脱落酸(ABA)在不同水位条件下的含量变化,可以认为引起胡杨水分胁迫的地下水位出现在5.0 m。其后随着水位的降低,胡杨脱落酸的积累更加明显,地下水位与胡杨叶脱落酸含量可以表示为:Y=0.703 5e0.408X,R2=0.830 4, p<0.01。4)通过塔里木河下游输水后的植被调查,当地下水位出现明显升高后,植被的地表生态响应非常明显,乔灌草植被在水位升高至不同水位后均出现相应的变化,说明以上的分析是符合实际的。  相似文献

Studies on the morphometric characteristics and the age and growth of Mugil cephalus (L.) of Bardawil Lake, Egypt, showed that the regression of body measurements on total length was linear and revealed that the standard length, pectoral fin depth and dorsal fin depth were different than the caudal peduncle and head length. Age and growth studies also showed that a straight line relation was determined between scale radius and body length as expressed by the following equation: Y  =−1.2127 + 1.0850 X . Accordingly, M. cephalus attained mean lengths of 14.2, 20.6, 23.9, 26.4, 28.5 and 30.0 cm at 1 to 6 years of age, respectively. Growth of the species is represented by the von Bertalanffy growth equation as: L t=45[1−exp−( t − t o)].  相似文献

A new simple but general estimation method for survival time in a hot environment is presented in this study. Even in heat-tolerant rats showing a triphasic heat response, an accurate estimation of survival time (ST) is possible. Rat groups, which included some heat-tolerant individuals, were exposed to 42.5 degrees C, 40% rh. Colonic temperature (Tco) was measured continuously by copper-constantan thermocouple. The ST (Y) of male and female rats were expressed as a linear function of time (X) until the Tco of 42.5 degrees C was reached: Y = 0.976X + 30.6 and Y = 0.968X + 31.6, respectively. A Tco of 42.5 degrees C at rest was just below the maximum survivable body temperature and above the steady-state equilibrium Tco levels during the second phase of the triphasic heat-response curve. Heat-tolerant individuals showed lower equilibrium temperatures than heat-intolerant rats. All 140 rats survived the Tco of 42.5 degrees C and lived for more than 8 wk, thus enabling them to be used for future experiments on thermoregulation. The heat survivors were able to reproduce, and their genetically controlled offspring could be used for thermoregulatory experiments.  相似文献

The purpose of this study was to determine the body surface area (BSA) based on the alginate method, to derive formulae for estimating BSA, and to compare the error of the present formula to previous formulas obtained from other countries. We directly measured the entire body surface area of 34 males (20-60 years old, 158.5-187.5 cm in height, 48.5-103.1 kg in body weight) and 31 females (20-63 years old, 140.6-173.1 cm, 36.8-106.1 kg) using alginate. The measurements showed that the BSA had a mean of 18,339 cm(2) (15,416-22,753 cm(2)) for males, and 16,452 cm(2) (12,825-22,025 cm(2)) for females. Based on these measurements, a regression model to estimate BSA was derived: Estimated BSA (cm(2))=73.31 Height (cm)(0.725) x Weight (kg)(0.425) (r(2)=0.999). The mean error of the formula was -0.1%, and did not show any significant difference by gender or body shape. When applied to the datasets (n=506) composed of various races (Caucasians, Africans, and Asians), the mean error of the formula was 0.4% and was smaller than that of DuBois & DuBois's, Gehan & George's, and Mosteller's formulas when applied to the same datasets. The errors of the three previous formulas were also within 2%. Overall, formulas based on the DuBois exponent (Weight(0.425) Height (0.725)) did not show any tendency of overestimation or underestimation by body shape, but other BSA-formulae showed differences by body shape. The present BSA formula has shown good accuracy in Korean adults of all weight categories compared to traditional formulas.  相似文献

The purpose of this study was to validate the accuracy and reliability of the Weightlifting Video Overlay System (WVOS) used by coaches and sport biomechanists at the United States Olympic Training Center. Static trials with the bar set at specific positions and dynamic trials of a power snatch were performed. Static and dynamic values obtained by the WVOS were compared with values obtained by tape measure and standard video kinematic analysis. Coordinate positions (horizontal [X] and vertical [Y]) were compared on both ends (left and right) of the bar. Absolute technical error of measurement between WVOS and kinematic values were calculated (0.97 cm [left X], 0.98 cm [right X], 0.88 cm [left Y], and 0.53 cm [right Y]) for the static data. Pearson correlations for all dynamic trials exceeded r = 0.88. The greatest discrepancies between the 2 measuring systems were found to occur when there was twisting of the bar during the performance. This error was probably due to the location on the bar where the coordinates were measured. The WVOS appears to provide accurate position information when compared with standard kinematics; however, care must be taken in evaluating position measurements if there is a significant amount of twisting in the movement. The WVOS appears to be reliable and valid within reasonable error limits for the determination of weightlifting movement technique.  相似文献

A thermally regulated Plexiglas chamber was designed for investigation of transcutaneous diffusion of N2 and helium (He) in the human hand. Influence of cutaneous blood flow in this process was studied simultaneously with gas diffusion measurements. Changes in cutaneous blood flow (Q, in ml X min-1 X 100 ml tissue-1) were effected by altering ambient temperature (T) from 20 to 40 degrees C (Q = 0.08 X 100.07T). We found that the rate of inert gas diffusion through human skin, expressed as conductance (G, in ml STPD X h-1 X m-2 X atm-1), increases exponentially as a function of blood flow, and was indistinguishable between He and N2 (G = 21.19 X 100.0124Q). The permeability, diffusion coefficient per unit diffusion distance (D/h, in cm/h), also rose exponentially as a function of blood flow. But permeability for He (D/h = 0.1748 X 100.0203Q) was greater than that for N2 (D/h = 0.1678 X 100.0114Q). As cutaneous blood flow rises, because of increased temperature, the apparent diffusion distance falls linearly for both N2 and He. The change is more prominent for He than for N2 diffusion. Estimated replacement time for the body stores of N2 by transcutaneous diffusion alone was shortened from 26.8 h at 31 degrees C to 15.1 h at 37 degrees C. It is suggested from this study that beneficial results may be derived during decompression procedure 1) by maintaining an appropriate transcutaneous pressure gradient of inert gases, and 2) by elevating ambient temperature.  相似文献

It has been argued on both evolutionary and functional grounds that genes must be shared by the mammalian sex chromosomes. The only direct evidence for such genes is our previous finding that loci on the human X (MIC2X) and Y (MIC2Y) chromosomes encode a species-specific cell surface antigen recognised by the monoclonal antibody 12E7. These loci map to the regions of the sex chromosomes which pair at meiosis, and MIC2X has been shown to escape X-inactivation. We have used immunoprecipitation and Western blot analysis combined with one- and two-dimensional polyacrylamide gel electrophoresis to compare the products of MIC2X and MIC2Y. The human specific molecule recognised by the 12E7 antibody is a membrane-associated polypeptide of mol. wt. 32.5 kd and pI = 5.0. No difference in size or charge has been detected between X and Y encoded forms of this molecule confirming that MIC2Y is a functional homologue of MIC2X. An intracellular polypeptide of mol. wt. 29 kd and pI = 7.0 present in the cytoplasm of both human and mouse cells is also recognised by the 12E7 antibody.  相似文献

【目的】明确黑翅土白蚁Odontotermes formosanus巢群的觅食群体大小与工蚁体长之间的关系,为进一步研究其生态学特点以及更好地开展防治提供依据。【方法】在杭州植物园内选择5处有黑翅土白蚁活动区域作试验点,诱捕并以中性红(neutral red)标记捕获的白蚁工蚁,通过“捕获-标记-释放-重捕”试验测定黑翅土白蚁的觅食群体大小。【结果】5个试验点中有4个成功实施了“捕获-标记-释放-重捕”试验,4个试验点黑翅土白蚁巢群的觅食群体个体数量分别为443 133±45 469, 495 360±67 429, 674 345±101 703和1 224 662±93 112头。黑翅土白蚁觅食群体大小(Y)与觅食工蚁的体长(X)呈正相关,拟合的指数函数方程式为:Y=1.8389e0.7185X(R2=0.7834)。【结论】本研究明确了黑翅土白蚁的觅食群体大小以及工蚁体长之间的函数关系,为开展区域防治提供了依据。  相似文献
