Summary The columnar epithelial cells of the gas gland in the swimbladder of the toadfish, Opsanus tau L., contain lamellar bodies that resemble the lamellar bodies found in epithelial cells of vertebrate lungs. Cytochemical assays indicate that swimbladder lamellar bodies are soluble in chloroform-methanol solution, react with tricomplex flocculation solution (indicating a phospholipid component), exhibit a positive reaction for cholesterol when exposed to digitonin, and contain acid phosphatase.The anterior chamber of the toadfish swimbladder is lined by an extracellular layer. Digitonin-cholesterol crystals are found in this layer when the swimbladder is treated with digitonin. A ruthenium red positive layer is also present in the anterior chamber of the toadfish swimbladder.The structure and cytochemistry of swimbladder lamellar bodies are compared with those of vertebrate lung lamellar bodies. We analyzed by different cytochemical and immunocytochemical approaches the biochemical compositon of coiled bodies in three different cultured cell lines. Coiled bodies are stained by the AgNOR staining method and by the EDTA regressive staining method preferential for ribonucleoprotein (RNP). Using the in situ polyadenylate nucleotidyl transferase-immunogold technique or anti-RNA antibodies, we decisively demonstrated the presence of appreciable amounts of RNA in coiled bodies. Neither the in situ terminal deoxynucleotidyl transferase-immunogold technique nor anti-DNA antibodies revealed any DNA in coiled bodies. Coiled bodies thus appear as distinct regions of cell nuclei involved in some steps of RNA metabolism but not directly in RNA synthesis. Their relationships with the dense fibrillar component of the nucleolus and with interchromatin granule clusters are discussed. 相似文献

The present study was carried out to characterize two distinct types of nuclear bodies, the nucleolus-associated bodies (NABs) and the satellites (SATs) using some specific staining, enzyme and immunogold labeling techniques in Cicer arietinum L. These bodies are of interest as the functional components of plant nucleus. DNA-specific staining and labeling with anti-DNA, a monoclonal antibody, were employed to verify the presence of DNA in NABs as well as in SATs. The enzyme-gold labeling technique was used to compare the relative amounts of RNase-gold and protease-gold labeling over the NABs. In NABs, RNase-gold labeling was relatively low compared to the protease-gold labeling. Ag-NOR staining revealed a similar content of NOR-silver proteins in both NABs and granular zone of the nucleolus. The NABs do not contain any DNA as they show negative response to DNA-specific stains and also when incubated with anti-DNA, only few gold particles are found over these structures. The SATs, on the other hand, react positively with DNA-specific stains, and high labeling is recorded with anti-DNA along with the dense chromatin masses. 相似文献

Previous immunohistochemical studies have identified several regulatory peptides in the carotid body chief cells in both humans and animals. These peptides, together with amines, may be important in the modulation of the chemoreflex by the carotid body. We report the localization and distribution of calcitonin and cholecystokinin-like (CCK) immunoreactivity in chief cells of human infant carotid body by light- and electron-microscopic immunohistochemical techniques. Consecutive sections immunostained with calcitonin and/or CCK antibodies revealed positively stained chief cells, both alone and in clusters, scattered throughout the carotid body lobule. Generally more chief cells were positive for calcitonin than for CCK. This was confirmed by quantiative analysis showing that the ratio of calcitonin to CCK immunoreactive cells was consistently >2:1 in all cases studied. There was no apparent correlation between the immunoreactivity for the two peptides and the age, sex, or postmortem interval. Calcitonin-like and CCK-like immunoreactivities were localized electron-microscopically over the dense core granules of the chief cells. Calcitonin and CCK-like peptides in carotid body chief cells may act as neutransmitters or neuromodulators involved in chemoreception. 相似文献

Wild plaice and flounders with lymphocystis were sampled and their lesion distribution noted. Regression of the lymphocystis cells was seen by histology and a host response including macrophages and immune cells occurred. Antibodies were detectable by a double diffusion precipitin test. Summary The formation of apoptotic cells and their phagocytosis by viable neighbouring cells in the gastric epithelium of 2-to 6-day-old mice was analysed. In order to observe the topographic relationship between apoptotic and normal epithelial cells using scanning electron microscope, the critical-point dried tissues was cracked before coating with gold. Cytochemical methods for the identification of surface carbohydrates and different tracers for apical and lateral cell membranes were applied for the analysis using the transmission electron microscope. Apoptotic cells were found on apical and lateral surfaces; this indicates the presence of tight connections with viable cells at some points. Ruthenium red strongly stained all accessible surfaces of normal cells and of apoptotic bodies. The quantity of neutral mucosubstances, as revealed by staining with tannic acid-uranyl acetate, seemed to decrease in the glycocalyx of apoptotic cells. The scanning and transmission electronmicroscopic results suggest that the phagocytotic vacuoles arise at the lateral side of the cells. The phagocytotic activity is not dependent upon a definite differentiation step of the mucoid cell. 相似文献

Multivesicular bodies were observed frequently in electron microscope photographs of Leydig cells from normal adult rat testes. Their formation, evolution and fate were analyzed morphologically in preparations treated to show cytidine monophosphatase (CMPase) activity and in animals sacrificed at various time intervals ranging from 5 min to 2 hrs after a single intratesticular injection of cationic ferritin (CF). Analysis of morphological and cytochemical data led to the following interpretation for the origin and fate of the multivesicular bodies in Leydig cells. The formation of multivesicular bodies in Leydig cells can be divided into three steps. Step 1, some endocytic vacuoles in Golgi region fuse with small vesicles to form pre-multivesicular bodies. Step 2, the pre-multivesicular bodies fuse together to form pale multivesicular bodies which are characterized by their large size, pale matrix and paucity of internal vesicles. Step 3, the pale multivesicular bodies remove their surplus enveloping membrane to become dense multivesicular bodies which are characterized by their smaller size, dense matrix and filling with internal vesicles. The pre-multivesicular bodies and pale multivesicular bodies do not contain hydrolytic enzymes, the dense multivesicular bodies acquire their hydrolytic enzymes by fusion with lysosomes and show CMPase activity. The dense multivesicular bodies often show a very close association with autophagosomes, and they might be involved in the autophagic activity of Leydig cells. 相似文献

Roach, rudd, bream and their natural hybrids of 2 cm standard length or larger can be definitively identified by their enzyme electrophoretic patterns. Zymograms of lactate dehydrogenase and esterases as produced by vertical starch gel electrophoretic analysis of whole fry or adult eye extracts are the most useful in this respect. The lactate dehydrogenase isozymes, containing B sub-units, migrate more anodally in rudd and bream than in roach. Due to the tetrameric structure of lactate dehydrogenase, in hybrid rudd x roach and roach x bream, eleven isozymes can be observed as compared with six in the parental patterns. Esterases show unique patterns for all species and hybrids. With the exception of one fraction in rudd x bream, the esterase patterns of hybrids show summations of the parental phenotypes. 相似文献

The tomato (Solanum lycopersicum L.) is one of the world's most important vegetable crop species. Among the many tomato accessions available, only a few are tolerant to abiotic stresses, which are responsible for the majority of the crop losses worldwide. Wild tomato species are then secondary gene pool in the breeding of more resistant tomato cultivars. In the current study, the composition of leaf cuticular waxes from fourteen tomato accessions, including S. lycopersicum, Solanum pennellii, Solanum pimpinellifolium, and their interspecific hybrids was studied in order to select the most adequate chemotaxonomic markers. Total cuticular wax load of S. pennellii plants was much higher than in the other plant species. Hydrocarbons were usually the most abundant wax components, followed by minor quantities of triterpenes and other compounds. Interspecific hybrids showed intermediate wax characteristics. The amount and composition of surface waxes were not correlated with the abiotic stress tolerance in S. lycopersicum. The composition of the hydrocarbon fraction was the least variable both within a single accession and between all the plants studied. Based on the results, cuticular hydrocarbons are proposed as potential chemotaxonomic markers in the classification of tomato and related species. 相似文献

Summary Cytological and histochemical characterization of the structures from which an obscure substance is secreted via open stomata to the abaxial surface of Japanese morning glory (Pharbitis nil Choisy cv. Violet) cotyledons has been carried out. Observation of intact cotyledons using the light microscope revealed randomly distributed semi-transparent structures. These structures, which were shown to be the same as those previously described as giant oil cells are referred to here as cotyledonary bodies. These bodies can be easily isolated and purified after enzymatic digestion of the cotyledons. Using different staining procedures we have confirmed that each cotyledonary body originates from an individual mesophyJl cell during embryo development. Purified bodies consist of (i) a thick shell-like envelope; (ii) a transparent, hydrophilic zone; (iii) a hydrophobic core. Hydrophobic contents of the bodies were readily extracted with methanol and shown to contain fatty acids and phenolic compounds using the gas chromatography/mass spectrometry (GC/MS) technique. Methanolic extracts of cotyledonary bodies showed high fluorescence with two excitation and emission maxima. Using a fluorescence microscope we have shown that the bodies isolated from seedlings grown in continuous light, conditions non-inductive for flowering, and those grown under conditions inductive for flowering (a single 16 h, long dark period) have different fluorescence emission spectra. Different levels of free Ca2+ inside cotyledonary bodies isolated from light-grown and single dark-period treatedP. nil seedlings were found using the fluorescent calcium indicator dye Fluo-3 under a confocal scanning laser microscope. We have investigated Ca2+ activity during pollen germinationand the possibility that it may be responding to a phosphoinositidesignal transduction pathway, by employing inhibitors of Ca2+channels (verapamil and TMB-8), EGTA as a Ca2+ scavenger andthe inositol 1-phosphatase inhibitor lithium chloride. We havefound that at least two Ca2+ pools are utilized during pollengermination. Influx of extracellular Ca2+ appears to be necessaryfor the germination of apple and tobacco pollen, but it doesnot appear to be required for the germination of potato pollen.Conversely, activation of intracellularly stored Ca2+ was necessaryfor optimal germination of all three pollen species. LiCI hadstrong effects on pollen germination. At 5 mM LiCI, pollen germinationwas inhibited by 78% for apple, 84% for tobacco, and 74% forpotato. Li+ inhibition was overcome by the addition of Ca2+,which restores germination of all three species to 85100%of that observed in controls, myo-lnositol also partially overcomesLi+ inhibition of pollen germination, thus providing some evidencefor a link between Li+ inhibition and Ca2+ rescue, myo-lnositolrescue of Li+ inhibition was most effective for potato pollen.Chlorotetracycline (CTC) spectroscopy revealed a higher levelof membrane-Ca2+ in Li + -treated pollen grains than in controls,and the short pollen tubes which did emerge did not accumulatemembrane-associated Ca2+. The results suggest that Li+ inhibitionmay interfere with the release (activation) or partitioningof membrane-Ca2+ during pollen germination and that this Ca2+activity may be responding, at least in part, with a phosphoinositidesignal transduction pathway. The distribution and growth of roach and dace in the R. Exe catchment was studied. Distribution conformed to the classic theories of zonation, although human interference extended or reduced the range of species. Distinct differences were found in the growth rates of roach and dace between different regions of the catchment. Both species achieved their fastest growth in zones where the river topography portrayed their 'preferred' habitat characteristics, i.e. the fast-flowing middle and slow-flowing lower reaches for dace and roach, respectively. Female roach were found to grow considerably faster than male fish, whilst no sexual differentiation in growth rates was found in dace. Data on the parasite diversity in Salmonidae fish parr from different parts of the Utsjoki River obtained during 1993-1995 and 2006-2007 are presented. Three fish species, Salmo salar L., S. truttae L., and Salvelinus alpinus L., were examined on the presence of helminthes. Twenty species of salmon parasites were found, the majority of which are the parasites with complicated life cycles. Infusorians C apriniana piscium, myxosporidia Chloromyxum januaricus and Myxobolus neurobius, metacercaria of the genera Diplostomum and Apatemon, and the nematode Raphidascaris acus larvae were the most numerous in salmon parasite fauna. Brown trout had the most number of specific parasite species, whereas char was infested by protozoan parasites only. 相似文献

Summary This paper examines the reproductive cycles of three ecologically important marine bivalves-Modiolus modiolus (L.), Cerastoderma edule (L.), and Mytilus edulis L. in Strangford Lough, Northern Ireland over a period of almost five years.Whilst Modiolus does not appear to become sexually mature until it is several years old, Mytilus and Cerastoderma can reproduce in their first and second years of life respectively. In Cerastoderma and Modiolus sexual maturity is preceded by a period of rapid somatic growth.The subtidal Modiolus population remained in a more or less fully ripe condition virtually throughout the period of this investigation suggesting that this particular population lacked any marked cyclical reproductive activity. We interpret this as evidence of slow but almost continuous release of gametes throughout much of the year, a suggestion which is supported by recruitment data. A small intertidal population of Modiolus in Belfast Lough monitored over a period of two years exhibited a much more seasonal cycle. Here spawning occurred mainly during the autumn and winter. These data suggest that localised environmental factors are exceedingly important in controlling the annual reproductive cycle of this species.Cerastoderma from the mid-tidal sand flats ripened rapidly during the spring and spawned over a relatively restricted period in the summer. In Strangford Lough Mytilus occurs predominantly in the low-shore and while it spawns mainly in the spring and summer the annual cycle is considerably more protracted and variable than in Cerastoderma.Variations in the duration of the spawning periods in these bivalves can perhaps be explained in terms of both environmental stability and the immediate physical conditions experienced by these particular populations. The reproductive strategies exhibited by Cerastoderma, Modiolus, and Mytilus in Strangford Lough are considered in relation to population stability and to the different patterns of mortality which characterise these species in their respective local habitats. 相似文献

Plant species differ in nutrient uptake efficiency. With a pot experiment, we evaluated potassium (K) uptake efficiency of maize (Zea mays L.), wheat (Triticum aestivum L.), and sugar beet (Beta vulgaris L.) grown on a low-K soil. Sugar beet and wheat maintained higher shoot K concentrations, indicating higher K uptake efficiency. Wheat acquired more K because of a greater root length to shoot dry weight ratio. Sugar beet accumulated more shoot K as a result of a 3- to 4-fold higher K influx as compared to wheat and maize, respectively. Nutrient uptake model NST 3.0 closely predicted K influx when 250 mg K kg?1 were added to the soil, but under-predicted K influx under low K supply. Sensitivity analysis showed that increasing soil solution K concentration (CLi) by a factor of 1.6–3.5 or buffer power (b) 10- to 50-fold resulted in 100% prediction of K influx. When both maximum influx (Imax) and b were increased by a factor of 2.5 in maize and wheat and 25 in sugar beet, the model could predict measured K influx 100%. In general, the parameter changes affected mostly calculated K influx of root hairs, demonstrating their possible important role in plant K efficiency. 相似文献
