Bacteria, yeasts, and molds which grew in a medium containing a synthetic lignin — a dehydrogenation polymer (DHP) of coniferyl alcohol — as a sole carbon source, were isolated from soil. One fungus, Fusarium solani M-13-1, was found to degrade the DHP most vigorously among the isolated organisms. It was shake-cultured in a medium containing dehydrodiconiferyl alcohol (DHCA) (I), an important lignin model compound, and the following six metabolic products were isolated and identified: 1) Phenylcoumaran--aldehydic (II) and -carboxylic compounds, 2) phenylcoumaran--aldehydic compound (IV), formed by release of a 2-carbon fragment from the phenylcoumaran--carboxylic compound, 3) 5-acetylvanillyl alcohol (V), formed by cleavage of the coumaran ring and reduction of the -aldehyde group, 4) 5-carboxyvanillyl alcohol (VI), formed by subsequent oxidation of the acetyl group, and 5) the -ether of DHCA (VII), considered to be a by-product. -1,3-Glucanase (Lu) was isolated from unfertilized eggs of the sea urchin Strongylocentrotus intermedius. A comparative study of some properties of -1,3-glucanase Lu and -1,3-glucanases with different action types—endo--1,3-glucanase from crystalline style of the marine mollusk Spisula sachalinensis (LIV) and exo--1,3-glucanase from the terrestrial snail Eulota maakii (LII)—was performed. It was found that -1,3-glucanase Lu hydrolyzes laminaran with a high yield of glucose in the reaction products. The enzyme hydrolyzes substrates with retention of the glycosidic bond configuration, is able to cleave modified substrates, and exhibits transglycosylating activity. All properties of -1,3-glucanase from S. intermedius were more similar to those of the endo--1,3-glucanase from the marine mollusk (LIV) than exo--1,3-glucanase LII from the terrestrial snail. The differences in the effect of LIV and Lu on laminaran are probably related to the functions of -1,3-glucanase Lu from sea urchin eggs (which, in contrast to LIV, is not a digestive enzyme).  相似文献

Pulse sequences are presented for the measurement of3JCC and3JNC scalar couplings for allC containing residues in15N,13C uniformly labeled proteins. The methodsdescribed are based on quantitative J correlation spectroscopy pioneered byBax and co-workers [Bax et al. (1994) Methods Enzymol., 239, 79–105].The combination of 3JCC and3JNC scalar coupling constants allows theassignment of discrete rotameric states about the 1 torsion angle in cases where such states exist or, alternatively,facilitates the establishment of noncanonical 1conformations or the presence of rotameric averaging. An efficient and reliable micropropagation system for Persian clover (Trifolium resupinatum L.) was developed using different explants and media. Node, hypocotyl and cotyledonary node explants were cultured on Murashige and Skoog (MS) medium supplemented with combinations of either 6-benzyladenine (BA) and indole-3-butyric acid (IBA) or BA, Kinetin (KIN) and IBA. Direct multiple shoots developed within 6weeks in all explants in most media tested. The best shoot multiplication capacity was obtained from cotyledonary node explants on MS medium containing 7.1M BA and 1M IBA or 14.1M BA and 1M IBA. Elongated shoots were rooted on either MS medium alone or combination with different concentrations of indole-3-butyric acid (IBA), indole-3-acetic acid (IAA) and -naphthaleneacetic acid (NAA). High rooting was achieved in half strength MS medium containing 8M IBA.  相似文献

Summary The systematic study of heterozygotes for the g locus controlling G serotype in Paramecium aurelia syngen 1, shows that a phenomenon of allelic exclusion exists. This phenomenon of exclusion happens either systematically, almost systematically or randomly, depending on the studied combination of alleles (Table 2). For a given combination of alleles, it is always the same allele which is excluded. Back-cross experiments indicate that the observed allelic modulation is not dependent upon a simple or classical type of regulatory system. It seems to be characteristic of a given allelic interaction.This phenomenon of allelic exclusion resembles the well-known phenomenon of mutual exclusion occurring between different loci which govern the surface antigens corresponding to the different serotypes of Paramecium aurelia. Both cases of exclusion (inter-allelic or intergenic) might involve an original mechanism of inter-regulation between proteins belonging to the same molecular family and fulfilling similar functions but under different physiological conditions.  相似文献

An in frame gene fusion containing the coding region for mature -lactamase and the 3-end of hylA encoding the haemolysin secretion signal, was constructed under the control of a lac promoter. The resulting 53 kDa hybrid protein was specifically secreted to the external medium in the presence of the haemolysin translocator proteins, HlyB and HlyD. The specific activity of the -lactamase portion of the secreted protein (measured by the hydrolysis of penicillin G), approximately 1 U/g protein, was close to that of authentic, purified TEM--lactamase. This is an important example of a hybrid protein that is enzymatically active, and secreted via the haemolysin pathway. Previous studies have indicated that haemolysin is secreted directly into the medium, bypassing the periplasm, to which -lactamase is normally targeted. This study indicated, therefore, that normal folding of an active -lactamase, can occur, at least when fused to the HlyA C-terminus, without the necessity of entering the periplasm. Despite the secretion of approximately 5 g/ml levels of the active -lactamase fusion into the medium, there was maximally only a 50% detectable increase in the LD50 for resistance to ampicillin at the individual cell level. This result suggests that, normally, resistance to ampicillin requires a high concentration of the enzyme close to killing targets, i.e. in the periplasm, in order to achieve significant levels of protection.These authors made an equal contribution to this work  相似文献

Immobilization of Chlamydomonas reinhardtii in alginate increases its photorespiration rate. In the immobilized cells, the photorespiratory enzyme, phosphoglycolate phosphatase, was 75% higher than in freely suspended cells. Thus, the immobilized cells produced glycolate at twice the rate than in freely suspended cells when treated with aminooxyacetate (a transaminase inhibitor). Cell adhesion molecules, some of which are lectins, play a key role in the control of normal and pathological processes of various living organisms. We found herein that NacetylDglucosaminespecific lectin, isolated from the ascidian Didemnum ternatanum (DTL), alters the growth properties of HeLa tumor cells depending on the anchorage. DTL was shown to increase the proliferation of HeLa cells grown in soft agar greatly (in anchorageindependent fashion). In contrast, DTL inhibits the proliferative activity of HeLa cells grown on solid substrate and acts as inductor of differentiation, slowing cell growth, increasing the cell attachment and spreading. Scanning electron microscopic data have demonstrated that DTL treatment resulted in pronounced changes of the shape and surface of HeLa cells. Changes of cellular morphology correlated with essential redistribution of actin microfilaments.  相似文献

Zusammenfassung Zweijährige Selektion auf Frostresistenz bei einer Kreuzung der argentinischen Tomatensorte Platense mit der nordamerikanischen Pearl Harbour unter Freilandbedingungen erwies sich bei Überprüfung in der Kühlkammer im Vergleich zu frostempfindlichen Kultursorten als ergebnislos. Es wird daher der Schluß gezogen, daß auch bei Tomaten eine erfolgreiche Züchtung auf Frostresistenz nur durch Einkreuzung mit frostresistenten Wildarten möglich ist, die auch vorhanden sind. Transforming growth factor- (TGF-) sequestered in dentine matrix has an important role in dental tissue repair after injury and its exposure at sites of injury may stimulate tertiary dentinogenesis. This study aimed to investigate the expression of TGF- isoforms in mature human dentine matrix and the ability of chemical treatments to expose TGF- on the cut surface of dentine using gold immunolabelling and subsequent scanning electron microscopy examination. TGF-1 was the only isoform that could be detected in human dentine and the nature of the chemical treatment of the tissue influenced its detection. EDTA treatment provided good exposure of TGF-1 on the dentine surface, whilst citric acid and sodium hypochlorite treatments revealed lesser amounts of this isoform. Only minimal staining for TGF-1 was observed in samples treated with phosphate-buffered saline. TGF-2 and -3 could not be detected in the specimens with any of the treatments. This study suggests that TGF-1 is the only TGF- isoform expressed by human odontoblasts to be sequestered in dentine implying that differences in isoform–extracellular matrix interactions may exist. Information on chemical treatment of tissue specimens for immunostaining may provide a useful basis for selection of tissue preparation techniques for clinical restorative treatment procedures to facilitate TGF- mediated reparative processes at sites of dental injury.  相似文献

Cultured human and rat endothelial cells were used to study cellular toxicity and Ca2+ signalling upon exposure to reactive oxygen species. Superoxide and hydrogen peroxide (O2·–/H2O2) were produced by the hypoxanthine/xanthine oxidase system (HX/XO) and caused intracellular Ca2+ concentration ([Ca2+]i) to rise steadily when activities above 2 mU/ml were used. These Ca2+ increases were also measured when the glucose/glucose oxidase (G/GO) system above 5 mU/ml was used to produce hydrogen peroxide (H2O2). Gross morphological changes appeared to parallel elevated [Ca2+]i levels preceding cell death. However, when HX/XO or G/GO were used at non toxic doses rapid and transient changes in [Ca2+]i were measured. These treatments did not alter subsequent receptor mediated Ca2+ signalling induced by ATP (10 M) or histamine (100 M). Superoxide dismutase (50 U/ml), which dismutates O2·minus; into H2O2 al ient [Ca2+]i responses. H2O2 added directly was able to induce similar Ca2+ transients when concentrations of at least 500 M were used. Buffering trace amounts of iron (o-phenanthroline; 200 M) in order to inhibit úOH radical formation was not effective to alter Ca2+ changes. Experiments performed in Ca2+-free buffer showed a similar rise in [Ca2+]i and readdition of Ca2+ to the extracellular medium indicated the activation of store operated Ca2+ entry. Blocking Ca2+-ATPases of the endoplasmatic reticulum with thapsigargin (1 M) inhibited ROS induced transient increases and cells preincubated with pertussis toxin (200 nM) showed unchanged Ca2+ transients after exposure to both enzyme systems. Phospholipase C inhibitor U73122 (2 M) effectively reduced hydrogen peroxide induced emptying of intracellular stores. Taken together, we demonstrate that enzymatically produced non-toxic H2O2 rather than O· ndash; or · OH causes calcium signalling from thapsigargin sensitive stores, and activates store operated Ca2+ entry at least partially by activating phospholipase C. These changes clearly differ from pathological oxidative stress associated with a progressive increase in [Ca2+]i.  相似文献

The specific uptake rate of -glucose by yeast (S. cerevisiae) and recombinant yeast is higher than that of -glucose. On the other hand, the specific uptake rate of -glucose byE.coli and recombinantE.coli is less than that of -glucose. These results imply that the unequal specific uptake rates of optical isomers of glucose should be included in modeling and optimizing high-cell density fermentations, fed batch fermentations, and immobilized cell systems, where cell density is maintained high and glucose concentration is low.  相似文献

Photosynthesis-irradiance (P-E) curves are widely used to describe photosynthetic efficiency and potential. Contemporary models assume maximal photosynthetic quantum yield () at low irradiances. But P-E observations made with both oxygen evolution and carbon uptake techniques show that this is not always the case. Using new and published data in conjunction with modeling exercises, we demonstrate that regardless of the mechanism there can be reductions in at low irradiances that are not readily observable using conventional P-E analyses. We also show that analytical errors, such as inaccurate estimation of dark oxygen consumption or carbon uptake, can markedly affect the structure of -E curves with negligible effect on P-E curve structure. Whether from respiration `corrections' or other mechanisms, these deviations in at low light levels from the maximum quantum yield of photosynthesis (max) can lead to significant errors (> 50%) in the estimation of the linear portion of the P-E curve and ultimately max. Non-linear models of P-E, such as the rectangular hyperbola, quadratic, exponential and hyperbolic tangent that are commonly used to estimate the initial slope () of the P-E curve assume that is maximal at low light levels and therefore can err in the estimation of max when is reduced at low light levels. Using a diverse data set of 622 P-E curves with a total of 7623 points, we show that although model skills are high (r 2 = 0.96 ± 0.05, 0.97 ± 0.04, 0.97 ± 0.04 and 0.97 ± 0.04, respectively), a large fraction of the model-predicted max differ by greater than 10% from true max values (91%, 50%, 82% and 46%, respectively). Data from these observations and modeling exercises lead us to suggest that max be determined by directly estimating the true maximum of a -E curve rather than using the more conventional methodology employing the initial slope of the P-E curve.This revised version was published online in October 2005 with corrections to the Cover Date.  相似文献

Polyclonal antibodies to mouse - and /-caseins were raised in rabbits. These antibodies display tissue- and species specificity as shown by immunoblotting. Immunohistochemical analyses demonstrate that both - and /-caseins were synthesized and secreted from virtually all lactating mammary epithelial cells, in a pattern very similar to that of the mouse -lactalbumin. Residual amounts of caseins were located also in the apical surface of epithelial cells surrounding the ducal lumen of virgin mammary gland sections. In contrast to the significant level of -casein in the milk, the amount of this protein compared to - or -caseins was extremely low in medium conditioned for 24 h by mammary explants of mid-pregnant mice immediately after explantation or after 4 days.  相似文献

Summary Four types of chromosomes with a deletion between the human embryonic and globin genes were identified among 2.8% of 321 Black Americans from Georgia. Two deletions of approximately 11 kb which differed by about 300 bp occurred on chromosomes with or without a polymorphic Xba I site 5 to the globin gene [(X+) or (X-)]. The deletions are identifiable in Xba I digests of genomic DNA using an or a globin gene probe which yield fragments of 23 kb from (X+)–* chromosomes or 27 kb from (X)–* chromosomes. Digestion with other enzymes and probing with both and probes gave fragments typical of the two globin gene deletions previously identified in Polynesians. Among Black Americans, these globin gene deletions have been found in combination with globin gene deletions in trans but not in cis. Homozygotes have not been found. Hematologic data on carriers of the globin gene deletions in association with Hb AS, SS, and SC suggest that these deletions have no effect on the function of the adult globin genes.  相似文献

In this essay I argue that Ernst Mayr's idea that the emergence of evolutionary biology in Western thought was delayed by the pernicious influence of the false ideologies of Platonism, Christianity, and physicalism is ahistorical and anti-evolutionary, that similar ideas, especially his antipathy to physicalism, prejudice his account of the transformation of natural history and medical science into biology, that his organicist resolution of the perennial conflict between mechanism and vitalism is an unstable compound of semi-holism and semi-mechanism, that his conception of biology as the true bridge between the sciences and the humanities, ethics, and social theory is open to question (especially as to the adequacy of the theory of natural selection to account for every aspect of human nature), and that his depiction of science as the sovereign key to understanding everything known to exist or happen in this universe cannot be justified at the bar of reason.  相似文献

Idioms of distress refers to the popular expression of emotional tension that arises in the relationship between sickness and kinship. By reference to case studies and discussions among the Polynesian people of Tonga, the author shows where such tension arises and how it influences the sickness process. Sickness is necessarily a collective phenomenon which can best be understood not simply as a clinical event, but as an experience that is part of the experience of family. Various ways of expressing distress as a reflexive encounter between personal and cultural meaning systems are reviewed, as are several new concepts such as doing sickness as kinship, and turning in the process of decision making in the kinship management of sickness. The explanatory models of sickness in Tonga are shown to encompass culturally sanctioned expressions of distress as part of the adaptive coping mechanisms in that society. Distress frequently emerges in somatic form, as a number of studies have shown. However, the author emphasizes the kinship meaning of sickness, kinship management and sickness therapy, the adaptive process of idiomatic expressions of distress, which are expanded here and offered as potential avenues for elaboration in other cultural milieu. Two aspects of the notion idioms of distress are noted, and the phenomenon is understood as a process which acts as a prime mover in social change.  相似文献

In the legume Phaseolus vulgaris L., glutamine synthetase (GS) (EC.6.3.1.2.) occurs as three cytosolic polypeptides, , and , and a plastidic polypeptide, . This paper describes the subunit composition of active octameric GS isoenzymes from root nodules and plumules using ionexchange high-performance liquid chromatography followed by two-dimensional denaturing gel electrophoresis and Western immunodetection. Root nodules contained four separable GS activities, three of which were composed mainly of cytosolic , / and GS polypeptides, whereas the fourth activity, consisted of plastidic GS polypeptides. The increase in GS activity during nodulation was due largely to the appearance of -containing isoenzymes, and to a lesser extent on the isoenzyme, whereas the -isoenzyme activity remained approximately constant throughout. Plumule GS from imbibed seeds was found to be composed of separate and isoenzymes, but 2 d after germination, plumule GS consisted of a mixture of , / and isoenzymes. The results from both nodules and plumules indicate that different cytosolic GS polypeptides in P. vulgaris are able to assemble into both homo-octameric and heterooctameric isoenzymes. Summary An opr24 mutation decreasing the degradation of RNA polymerase subunits was found among the Ts+ revertants of Ts strain carring an rpoC1 mutation which stimulates overproduction of these polypeptides. The opr24 mutation is allele-non-specific, for it decreases the degradation of the mutant and subunits, the amberfragment of the subunit and the non-mutant subunit. Moreover, opr24 reduces the proteolysis of abnormal proteins containing canavanine.The opr24 mutation maps between 17 and 21 min on the E. coli genetic map.The results demonstrate that a slow proteolysis rate in the bacterial cell can supress conditional lethal mutations.In addition, the slower degradation and the ensuing accumulation of subunits does not effect the rate of the subunit synthesis.  相似文献

GlcNAc1-2Man and GlcNAc1-6Man were synthesized using the reverse hydrolysis activity of -N-acetylglucosaminidase from both jack beans and Bacillus circulans. In turn, Gal1-4GlcNAc1-2Man and Gal1-4GlcNAc1-6Man were synthesized regioselectively using the transglycosylation activity of -galactosidase from Diplococcus pneumoniae and B. circulans, respectively. These di- and trisaccharides are important components of complex type sugar chains and will be used as intermediates in our synthetic studies.
