Ascidian hemolymph contains various types of blood cells (hemocytes), which are believed to be involved in defense mechanisms. We have studied nitric-oxide (NO) synthase activity in hemocytes of the ascidian Styela plicata after exposure to lipopolysaccharide (LPS). To investigate which cell types are involved in NO production, we first identified, by electron microscopy, the types of hemocytes previously described, mainly by light microscopy, by others. Five types of blood cells could be recognized in the hemolymph: granulocytes, hemoblasts, lymphocyte-like cells, morula cells, and pigment cells. The lymphocyte-like cells produced the most NO. In agreement with studies of other invertebrates, nitrite generation did not change after LPS stimulation in assays in vitro, under either different concentrations of LPS or different time periods. Therefore, we performed an in vivo assay by injecting a known quantity of Escherichia coli into the tunic of the ascidians in order to investigate possible differences in NO levels. No increase of NO occurred accompanying the inflammatory reaction suggesting that another molecule in the pathway was involved. We found that nuclear factor κB (NFκB) was activated. Since NFκB is involved in the production of many substances related to immune responses, additional molecules might also be generated in response to E. coli infection. These observations may improve our understanding of the reaction of animals to eutrophic conditions. 相似文献

The occurrence of neuropeptide Y-like substances has been verified in the nervous system and alimentary tract of the ascidian Styela plicata. Neuropeptide Y-like immunoreactivity is present in a few small neurons and in a network of beaded nerve fibres of the cerebral ganglion. Neuropeptide Y-like immunoreactive material can be also localized in the endostyle and in a few cell bodies of the branchial walls. Moreover, immunofluorescent endocrine-like cells of the "open" type occur in the gastric folds. Finally, some possible functions of the ascidian neuropeptide Y are discussed. 相似文献

The solitary ascidian Styela clava Herdman, 1882 is considered to be native to Japan, Korea, northern China and the Russian Federation in the NW Pacific, but it has spread globally over the last 80 years and is now established as an introduced species on the east and west coasts of North America, Europe, Australia and New Zealand. In eastern Canada it reaches sufficient density to be a serious pest to aquaculture concerns. We sequenced a fragment of the cytochrome oxidase subunit I mitochondrial gene (COI) from a total of 554 individuals to examine the genetic relationships of 20 S. clava populations sampled throughout the introduced and native ranges, in order to investigate invasive population characteristics. The data presented here show a moderate level of genetic diversity throughout the northern hemisphere. The southern hemisphere (particularly New Zealand) displays a greater amount of haplotype and nucleotide diversity in comparison. This species, like many other invasive species, shows a range of genetic diversities among introduced populations independent of the age of incursion. The successful establishment of this species appears to be associated with multiple incursions in many locations, while other locations appear to have experienced rapid expansion from a potentially small population with reduced genetic diversity. These contrasting patterns create difficulties when attempting to manage and mitigate a species that continues to spread among ports and marinas around the world. 相似文献

The solitary ascidian Styela plicata is an introduced species in harbors of temperate and tropical oceans around the world. The invasive potential of this species has been studied through reproductive biology and population genetics but no study has yet examined the microbial diversity associated with this ascidian and its potential role in host ecology and invasiveness. Here, we used 16S rRNA gene tag pyrosequencing and transmission electron microscopy to characterize the abundance, diversity and host-specificity of bacteria associated with 3 Mediterranean individuals of S. plicata. Microscopy revealed low bacterial abundance in the inner tunic and their absence from gonad tissues, while pyrosequencing revealed a high diversity of S. plicata-associated bacteria (284 OTUs from 16 microbial phyla) in the inner tunic. The core symbiont community was small and consisted of 16 OTUs present in all S. plicata hosts. This core community included a recently described ascidian symbiont (Hasllibacter halocynthiae) and several known sponge and coral symbionts, including a strictly anaerobic Chloroflexi lineage. Most recovered bacterial OTUs (79.6 %) were present in single S. plicata individuals and statistical analyses of genetic diversity and community structure confirmed high variability of bacterial communities among host individuals. These results suggest that diverse and variable bacterial communities inhabit the tunic of S. plicata, including environmental and host-associated bacterial lineages that appear to be re-established each host generation. We hypothesize that bacterial communities in S. plicata are dynamic and have the potential to aid host acclimation to new habitats by establishing relationships with beneficial, locally sourced bacteria. 相似文献

Decorin is an extracellular matrix dermatan sulfate/chondroitin sulfate proteoglycan found in a variety of vertebrate species. In the extracellular matrix of mammals, decorin interacts with fibrillar collagen and regulates its morphology. We report here the occurrence and distribution of collagen type I and the peptide, CEASGIGPEVPDDRD, which is present in the human decorin proteoglycan, in the extracellular matrix of different tissues of the primitive invertebrate chordate Styela plicata. The content of collagen was estimated by hydroxyproline, and its distribution in the tissues by histochemistry. Collagen was detected biochemically in intestine, heart, pharynx and mantle, occurring in higher amounts in the heart, followed by pharynx, mantle and intestine. Histochemical analysis with Sirius red indicates that collagen is present in the extracellular matrix of intestine and pharynx. Further ultrastructural immuno-gold assays using polyclonal antibodies raised against the decorin-specific peptide CEASGIGPEVPDDRD and collagen type I showed a co-localization of these molecules. These data suggest the occurrence of a protein containing a decorin-like peptide sequence, which may be interacting with fibrillar collagen in this primitive chordate. 相似文献

Mannose-binding lectin (MBL) is a C-type lectin involved in the first line of host defense against pathogens and it requires MBL-associated serine protease (MASP) for activation of the complement lectin pathway. To elucidate the origin and evolution of MBL, MBL-like lectin was isolated from the plasma of a urochordate, the solitary ascidian Halocynthia roretzi, using affinity chromatography on a yeast mannan-Sepharose. SDS-PAGE of the eluted proteins revealed a major band of approximately 36 kDa (p36). p36 cDNA was cloned from an ascidian hepatopancreas cDNA library. Sequence analysis revealed that the carboxy-terminal half of the ascidian lectin contains a carbohydrate recognition domain (CRD) that is homologous to C-type lectin, but it lacks a collagen-like domain that is present in mammalian MBLs. Purified p36 binds specifically to glucose but not to mannose or N-acetylglucosamine, and it was designated glucose-binding lectin (GBL). The two ascidian MASPs associated with GBL activate ascidian C3, which had been reported to act as an opsonin. The removal of GBL-MASPs complex from ascidian plasma using Ab against GBL inhibits C3-dependent phagocytosis. These observations strongly suggest that GBL acts as a recognition molecule and that the primitive complement system, consisting of the lectin-proteases complex and C3, played a major role in innate immunity before the evolution of an adaptive immune system in vertebrates. 相似文献

Cell distribution and tunic morphology in the ascidian Styela canopus were examined by electron microscopy. The observations showed that the outer covering is composed of a thin sinuous cuticle with several protrusions and a deep layer of ground substance. The fibrous component and its arrangement in the tunic were demonstrated: elementary fibrils exhibit a 'microtubular' structure and an elliptical cross-sectional shape. Four types of cells were described: clear vesicular tunic granulocytes, tunic microgranulocytes, unilocular tunic granulocytes, and globular tunic granulocytes. Morphofunctional aspects of the tunic tissue and certain phylogenetic relationships are discussed. 相似文献

The hemolymph of ascidians (Chordata-Tunicata) contains different types of hemocytes embedded in a liquid plasma. In the present study, heparin and a sulfated heteropolysaccharide were purified from the hemolymph of the ascidian Styela plicata. The heteropolysaccharide occurs free in the plasma, is composed of glucose ( approximately 60%) and galactose ( approximately 40%), and is highly sulfated. Heparin, on the other hand, occurs in the hemocytes, and high performance liquid chromatography of the products formed by degradation with specific lyases revealed that it is composed mainly by the disaccharides DeltaUA(2SO(4))-1-->4-beta-d-GlcN(SO(4)) (39.7%) and DeltaUA(2SO(4))-1-->4-beta-d-GlcN(SO(4))(6SO(4)) (38.2%). Small amounts of the 3-O-sulfated disaccharides DeltaUA(2SO(4))-1-->4-beta-d-GlcN(SO(4))(3SO(4)) (9.8%) and DeltaUA(2SO(4))-1-->4-beta-d-GlcN(SO(4))(3SO(4))(6SO(4)) (3.8%) were also detected. These 3-O-sulfated disaccharides were demonstrated to be essential for the binding of the hemocyte heparin to antithrombin III. Electron microscopy techniques were used to characterize the ultrastructure of the hemocytes and to localize heparin and histamine in these cells. At least five cell types were recognized and classified as univacuolated and multivacuolated cells, amebocytes, hemoblasts, and granulocytes. Immunocytochemistry showed that heparin and histamine co-localize in intracellular granules of only one type of hemocyte, the granulocyte. These results show for the first time that in ascidians, a sulfated galactoglucan circulates free in the plasma, and heparin occurs as an intracellular product of a circulating basophil-like cell. 相似文献

Species distribution, abundance, and long-term survival are determined by biotic and abiotic regimes. However, little is known about the importance of these factors in species range expansion. Styela plicata is a solitary ascidian introduced all over the world by ship fouling, including salt marsh habitats, where introduced populations must tolerate high seasonal variations in temperature and salinity. To determine the seasonal stress levels in a salt marsh population of S. plicata, we quantified heat shock protein (hsp70) gene expression using quantitative real-time PCR throughout a 2-year cycle. Results showed that hsp70 expression varied over time, with higher stress levels recorded in summer and winter. Periodic conditions of high temperatures, particularly when coupled with low salinities, increased hsp70 gene expression. Mortality events observed every year around June were concurrent with sharp increases in temperature (>6°C), indicating that drastic changes in abiotic factors may overwhelm the observed stress response mechanisms. Determining the ability of introduced species to cope with stress, and the thresholds above which these mechanisms fail, is fundamental to predict the potential expansion range of introduced species and design efficient containment plans. 相似文献

Summary This study identifies three discrete processes responsible for the rejection of tunic tissue transplanted between individuals of the solitary ascidian Styela plicata. The first stage of rejection is characterized by the destruction of blood vascular components within incompatible allografts. In the second phase, dense boundaries of extracellular material are deposited between grafts and the surrounding host tunic, effectively amputating the transplanted tissues. Finally, detached transplants undergo a gradual necrosis which results in the total degeneration of extracellular graft matrices. Of these three phases, the initial cellular depletion of allografts is responsible for the immunological specificity that is characteristic of histocompatibility in S. plicata. The subsequent amputation and necrosis of extracellular graft matrices are taken to be non-specific consequences of the initial cellular reactivity. 相似文献

Summary A small group of granulated endocrine cells have been described in the endostylar region of the pharynx in Styela. These cells are argyrophilic and exhibit calcitonin-like immunofluorescence. Tests with antisera to other peptides all proved negative. Cells from animals exposed to elevated calcium levels showed a degree of degranulation. Summary Ultrastructural studies have shown the presence of two types of granulated endocrine cell in the gut of Styela clava. Type I, which occurs in the stomach and intestine contains small irregular granules, each with a distinct halo. Type II, found only in the oesophagus contains larger rounded granules, often with little or no halo. The characteristics of these two cell types are compared with those of endocrine cells found in the digestive tracts of other protochordates and discussed with special reference to the evolution of gastrointestinal endocrine cells in vertebrates.The authors are grateful to Mr. R. This study characterised a 90 kDa lectin from an invertebrate chordate, the tunicate Styela plicata. One- and two-dimensional electrophoresis showed that the apparent molecular weight of this protein is maintained under both reducing and non-reducing conditions, suggesting that its native form is a monomer. The 90 kDa lectin was localised within a single type of hemocyte (morula cells), but was secreted from those cells when tunicates were challenged with the inflammatory elicitor, zymosan. Functional studies showed that the 90 kDa protein binds to galactose-based sugars in a divalent cation-dependent manner. Amino acid composition analysis and N-terminal amino acid sequencing indicated that the 90 kDa lectin is related to a previously characterised, collagenous lectin from S. plicata, splic43. However, peptide mass fingerprinting identified numerous differences between the two proteins. This suggests that the 90 kDa molecule represents a novel protein that is involved in host defence. 相似文献

This study investigated the effects of two environmental factors, temperature and light, on larval settlement and metamorphosis in the solitary ascidian Styela canopus. The results revealed that larval settlement rates decreased with increasing temperature in the range 12–30°C. We also demonstrated for the first time that pre-settlement metamorphosis of ascidian larvae can occur as a function of temperature. We suggest this could be an adaptation to avoid the greater energetic cost of active larval swimming, presumably resulting from the increasing temperature. They are able to metamorphose into passive drifting post-larvae and to continue planktonic life. This finding has implications for larval dispersal, especially under conditions of ocean warming. In addition, the effect of light intensity on larval settlement and metamorphosis was significantly different between photoperiods of 24 L : 0 D and 12 L : 12 D. These results provide some insight into the complex cues affecting settlement and metamorphosis of ascidian larvae and ascidian distribution in nature. 相似文献

Previous reports of tunicate endostyles have suggested that they contain little or no acidic glycoproteins in the glandular zones. The endostyles of Ciona intestinalis and Styela plicata were examined after anhydrous fixation with cyanuric chloride. Polyanions were stained with alcian blue (AB) at pH 2.5 or azure A, while sulfomucins were stained with high-iron diamine (HID) or AB at pH 1.0. Endostyles were also tested for sensitivity to acid hydrolysis (AH) and saponification. In Ciona zones 2 and 4 sometimes demonstrated positive HID and AB 1.0 responses. Almost invariably zone 6 was AB+ at pH 2.5; zones 2 and 4 were frequently responsive to AB, but less intense. Each of these 3 zones, when AB+, was sensitive to AH. Responses by zones 3 and 5 to AB (pH 2.5), azure A and saponification suggest that these zones contain mostly nuclear material. In secretory zones 2, 4 and 6 histological responses are consistent with the histochemistry of sialomucins. Zones 1 and 8 had sulfated material in the apical edges in both animal groups. Among the fixatives used for Ciona, only anhydrous fixation demonstrated most of the positive responses to polyanion-sensitive stains. 相似文献

We performed a biochemical and histochemical study of sulfated glycosaminoglycans in the tissues of the ascidian Styela plicata. A highly sulfated dermatan sulfate and a heparin-like polymer, identified by incubation with specific lyases, occur at different concentrations in intestine, heart, pharynx, and cloak. Dermatan sulfate prevails in the pharynx, whereas the heparin-like polymer abounds in the intestine. Staining of tissues sections with the cationic dye 1,9-dimethylmethylene blue before and after incubation with specific lyases revealed that the dermatan sulfate occurs in the extracellular matrix, while the heparin-like polymer is located within cytoplasmic granules of cells in the lumen of intestine and pharynx. The dermatan sulfate has a similar disaccharide composition in all tissues studied, whereas the heparin-like polymer differs in sulfate content. A direct relationship between sulfate content of the heparin-like polymer and antithrombin activity was observed. Analysis of the repeating disaccharide units of the heparin-like polymer indicates the presence of relatively high amounts of the disulfated disaccharide namely DeltaUA-1-->4-GlcN(SO(4))-(6SO(4)), which may suggest the occurrence in ascidians of regulatory biosynthetic mechanisms different from those observed for heparin in mammals. 相似文献

The ascidian Styela clava is widely distributed in northern China and is thought to be important in the functioning of estuarine systems. This ascidian may experience highly variable physiological conditions, and its physiological responses to these are of interest considering its ecological role. This study presents data on the physiological parameters in relation to body size and temperature (12-28 degrees C) of S. clava. Respiration, excretion, feces, ingestion and absorption energy were positive related to size and its mass exponents (b-values) varied from 0.2930 to 0.7488. Respiration energy increased with increasing, but critical values were found at 20 degrees C for energy of ingestion and absorption, while 24 degrees C for energy in feces and excretion. At the range of 16-24 degrees C, the scope for growth, gross and net growth efficiencies of ascidians increased with increasing temperature. The energy budget equations of different sized ascidians were obtained at different temperatures. Excretion energy shared a minimal fraction in ingestion energy (1.30-2.47%), the ratios of feces energy and respiration energy to ingestion energy were 46.53-64.27% and 10.26-80.75%, respectively. The physiological data obtained in the present study indicated that S. clava could adjust its energy budget according to the environment and its physiological conditions to meet their nutritional and energetic demands. In the range of experimental temperature (12-28 degrees C), 16-20 degrees C was suitable for the rearing of S. clava to achieve optimum development, while deficient metabolic adjustment induced a negative scope for growth of S. clava at 28 degrees C. 相似文献
