A 39-year bumblebee data base was used to study the codistribution of six cuckoo bumblebees in the subgenus Psythirus of Bombus (hereafter called Psythirus) and their free-living bumblebee hosts in the British Isles. 2. A model of nest parasitism predicted host threshold densities and stable deterministic dynamics, with fluctuations only emerging as a result of environmental or demographic stochasticity. 3. Standardized transects indicated that variation in total number of records could be largely attributed to variation in observer effort; analyses were therefore carried out using relative abundance. 4. Spatially, parasite-free zones were evident in areas of low host abundance, but the host threshold for parasite presence differed among species and locations. 5. Temporally, the relative numbers of the parasite and host species remained relatively constant, except that the nest parasite P. campestris declined significantly since 1990. 6. There were consistent negative effects of the parasitic species on the numbers of hosts in the following year, and this pattern was seen over large geographic areas. 7. The spatio-temporal patterns confirmed a high degree of host specificity, except that P. campestris may be parasitizing not only B. pascuorum but also other species in the subgenus Thoracobombus. 相似文献

Much of the ecology of rare bumblebee species remains poorly understood and in need of further study. It has recently been suggested that differences in the range and rate of decline among bumblebee species may relate to differences in their degree of habitat specialization. We examine biotope use by 17 bumblebee species in the Hebrides, southern UK and South Island, New Zealand. We identify a cluster of widespread and abundant species that occur in almost all biotopes and exploit man-made environments such as gardens and arable margins, this group corresponding to the “mainland ubiquitous” species of previous studies. A second grouping of species includes those associated to varying degrees with heathland. It is notable that some species occupy markedly different biotopes in different parts of their range; for example B. soroeensis is found largely on upland heaths in the Hebrides, but on calcareous grassland in the south. Some species, such as B. subterraneus and B. distinguendus, now survive only in specific rare biotopes and could be mistaken for habitat specialists, but it is clear from their historic distributions that they formerly occupied a broader range of biotopes. Surviving populations of several of the species that have declined most (B. distinguendus, B. sylvarum, B. muscorum sladeni, B. humilis) exhibit a markedly coastal distribution, when once they were widespread inland. We suggest that this is probably simply because some coastal biotopes are less amenable to agricultural improvement, and so more have escaped the detrimental effects of intensive farming. Our results concur with previous suggestions that bumblebees are generally not habitat specialists, so that the conservation of most bumblebee species could be achieved by restoration of flower-rich unimproved meadows. 相似文献

This report describes the isolation procedure and properties of tightly coupled flight muscle mitochondria of the bumblebee Bombus terrestris (L.). The highest respiratory control index was observed upon oxidation of pyruvate, whereas the highest respiration rates were registered upon oxidation of a combination of the following substrates: pyruvate + malate, pyruvate + proline, or pyruvate + glutamate. The respiration rates upon oxidation of malate, glutamate, glutamate + malate, or succinate were very low. At variance with flight muscle mitochondria of a number of other insects reported earlier, B. terrestris mitochondria did not show high rates of respiration supported by oxidation of proline. The maximal respiration rates were observed upon oxidation of α-glycerophosphate. Bumblebee mitochondria are capable of maintaining high membrane potential in the absence of added respiratory substrates, which was completely dissipated by the addition of rotenone, suggesting high amount of intramitochondrial NAD-linked oxidative substrates. Pyruvate and α-glycerophosphate appear to be the optimal oxidative substrates for maintaining the high rates of oxidative metabolism of the bumblebee mitochondria. 相似文献

Characterisation of microsporidian species and differentiation among genetic variants of the same species has typically relied on ribosomal RNA (rRNA) gene sequences. We characterised the entire rRNA gene of a microsporidium from 11 isolates representing eight different European bumblebee (Bombus) species. We demonstrate that the microsporidium Nosema bombi infected all hosts that originated from a wide geographic area. A total of 16 variable sites (all single nucleotid polymorphisms (SNPs)) was detected in the small subunit (SSU) rRNA gene and 42 (39 SNPs and 3 indels) in the large subunit (LSU) rRNA sequence. Direct sequencing of PCR-amplified DNA products of the internal transcribed spacer (ITS) region revealed identical sequences in all isolates. In contrast, ITS fragment length determined by PAGE and sequencing of cloned amplicons gave better resolution of sequences and revealed multiple SNPs across isolates and two fragment sizes in each isolate (six short and seven long amplicon variants). Genetic variants were not unique to individual host species. Moreover, two or more sequence variants were obtained from individual bumblebee hosts, suggesting the existence of multiple, variable copies of rRNA in the same microsporidium, and contrary to that expected for a class of multi-gene family under concerted evolution theory. Our data on within-genome rRNA variability call into question the usefulness of rRNA sequences to characterise intraspecific genetic variants in the Microsporidia and other groups of unicellular organisms. 相似文献

Bumblebees (Bombus spp.) are important pollinators of many economically important crops and microsporidia are among the most important infections of these hosts. Using molecular markers, we screened a large sample (n=1,009 bees) of workers of 27 different Bombus spp. from China (Sichuan, Qinghai, Inner Mongolia, and Gansu provinces). The results showed that 62 individuals representing 12 Bombus spp. were infected by microsporidia with an overall prevalence of 6.1%. Based on the haplotypes (ssrRNA sequences), we confirmed the presence of Nosema bombi, Nosema ceranae and (likely) Nosema thomsoni. In addition, four new putatively novel taxa were identified by phylogenetic reconstruction: Nosema A, Nosema B-complex, Nosema C-complex and Nosema D-complex. In many cases, hosts were infected by more than one Nosema taxon. Possible caveats of sequence analyses are discussed. 相似文献

It was found that the succinate oxidation rate in mitochondria of flight muscles of Bombus terrestris L. increased by a factor of 2.15 after flying for 1 h. An electrophoretically homogenous preparation of succinate dehydrogenase with a specific activity of 7.14 U/mg protein and 81.55-fold purity was isolated from B. terrestris flight muscles. It is shown that this enzyme is represented in the muscle tissue by only one isoform with R f = 0.24. The molecular weight of the native molecule and its subunits A and B was determined. The kinetic characteristics of succinate dehydrogenase (K m = 0.33 mM) and the optimal concentration of hydrogen ions (pH 7.8) were established, and the effect of salts on the enzyme activity was studied. The role of succinate as a respiratory substrate in stress and the structural and functional characteristics of the succinate dehydrogenase system in the flight muscles of insects are discussed. 相似文献

The notion of a dynamic compensatory system is discussed, characterized by the alternation of species occupying the leading position in bumblebee assemblages, while the total density of these pollinators in island ecosystems remains at similar levels. The functioning of the compensatory system is regulated by both abiotic factors (the weather and climate) and biotic factors (competition for trophic resources). Red clover genotypes capable of regenerating plantlets in vitro from non-meristematic tissue-derived callus are rare. Selection
for genotypes capable of somatic embryogenesis identified a clone comprised of a group of plantlets regenerated from a hypocotyl-derived
callus culture on L2-based media and another group of plantlets originating from crown divisions of the epicotyl-derived plant.
The callus-derived plants of this clone were highly regenerative when reintroduced to callus culture, but the epicotyl-derived
plants produced nonregenerative callus cultures. F1, F2 and BC1 populations were evaluated to determine the mode of inheritance of the regeneration trait. Reciprocal crosses did not differ,
indicating a lack of maternal effects. Results were compatible with genetic control of regeneration by two complementary genes.
We propose the genotype Rn1-Rn2- for regenerative plants. Three petiole segment explants were sufficient to evaluate regenerative ability in seedlings. Regenerative
ability was often associated with abnormal leaf morphology in a few to several leaves.

Insect chitin possessing shell-like structure was prepared from the bumblebee corpses by a consequent treatment with 1M HCl and 1M NaOH. The bumblebee chitin was compared with crustacean (shrimp) chitin by using elemental analysis, Fourier-transform infrared (FT-IR) and solid-state (13)C cross-polarization magic angle spinning nuclear magnetic resonance (CP/MAS)-NMR spectroscopy and confocal microscopy. Bumble bees are an important group of wild pollinators in North America and considerable concern has been expressed over declines in their populations. However, before causes for declines can be assessed, it is essential that the geographical and chronological patterns of decline be discovered. Hitherto a lack of assessment of historical data has hindered our efforts to determine which species are most at risk. Here, the status of 21 North American bumble bee species (Hymenoptera: Apidae) occurring in the eastern nearctic biogeographic region is assessed using a specimen-level database from compiled museum and survey records dating back to the late nineteenth century from various institutional collections. Using a combination of measures, bumble bee declines were assessed over their entire native ranges. We report here that half of the selected fauna is in varying levels of decline (especially Bombus ashtoni, B. fervidus, and B. variabilis), with the remaining species exhibiting stable or increasing trends (e.g., B. bimaculatus, B. impatiens, and B. rufocinctus). Suggestions for prioritizing conservation efforts for this important group of pollinators are given. 相似文献

Relationships between gibberellins and floral initiation were investigated in a conditional non-flowering mutant of red clover, Trifolium pratense. Untreated mutant plants will not flower under long-days, but will do so when certain GAs are applied. Gibberellins, A3, A1, A7, and A5 all resulted in both stem elongation and flowering whilst GA4 produced the elongation only. Applications of GA20, GA8 and GA13 under long-days had no detectable effect. Thus, by combining the use of the mutant with the application of different GAs, the correlation between the processes of stem elongation and floral initiation, which is normally strongly expressed in this species, was broken. Endogenous gibberellins shown to be present in normal plants were also found in the mutant genotype. Gibberellins alone were not sufficient to initiate floral development in the mutant, there being an essential element of interaction with long-days. These results are discussed in relation to the nature of the lesion in the mutant and the signal provided by the applied gibberellin. 相似文献

Host–parasite systems are characterised by coevolutionary arms races between host and parasite. Parasites are often the driving force, as they replicate much faster than their hosts and have shorter generation times and larger population sizes, resulting in higher mutation rates per time interval. This scenario does not fit all host–parasite systems. Socially parasitic cuckoo bumblebees (Bombus (Psithyrus) vestalis) parasitise colonies of Bombus terrestris share most life history characteristics with their hosts. As they parasitise only a subset of all available colonies, their population size should be lower than that of their hosts. This might have strong negative effects on the genetic diversity of B. vestalis and their adaptability. Here, we study for the first time the population structure of a Bombus/Bombus (Psithyrus) system. Highly polymorphic DNA markers were used to reconstruct sibships from individuals collected in the wild. The analysis of the host and parasite populations revealed a rate of parasitism of about 42% (range 33–50%). The population size of B. vestalis was lower compared to their hosts, which was also reflected in low within-group genetic distance. An analysis of the reconstructed queen genotypes revealed more supersisters amongst the B. vestalis queens when compared to the B. terrestris host. The data suggest that B. vestalis females and males do not disperse over long distances. This shows a potential for local adaptation to their hosts. 相似文献

Cultivars of red clover (Trifolium pratense L.), an important forage crop in temperate regions, are often characterised by an unsatisfactory level of seed yield, leading
to high production costs. This complex trait is influenced by many components and negatively correlated with other important
traits, such as forage yield or persistence. Therefore, seed yield has proven to be difficult to improve. Thus, the objectives
of this study were to assess association among seed yield components and to provide the basis for identifying molecular markers
linked to QTLs for seed yield components to assist breeding for improved red clover cultivars. A total of 42 SSR and 216 AFLP
loci were used to construct a molecular linkage map with a total map length of 444.2 cM and an average distance between loci
of 1.7 cM. A total of 38 QTLs were identified for eight seed yield components. The traits seed number per plant, seed yield
per head, seed number per head, head number per plant and percent seed set were highly correlated with seed yield per plant,
and QTLs for several of these traits were often detected in the same genome region. Head number per plant may present a particularly
useful character for the improvement of seed yield since it can easily be determined before seed maturity. In addition, two
genome regions containing four or five QTLs for different seed yield components, respectively, were identified representing
candidate regions for further characterisation of QTLs. This study revealed several key components which may facilitate further
improvement of seed yield. The QTLs identified represent an important first step towards marker-assisted breeding in red clover. 相似文献

Inferring the evolutionary and biogeographic history of taxa occurring in a particular region is one way to determine the processes by which the biodiversity of that region originated. Tree boas of the genus Corallus are an ancient clade and occur throughout Central and South America and the Lesser Antilles, making it an excellent group for investigating Neotropical biogeography. Using sequenced portions of two mitochondrial and three nuclear loci for individuals of all recognized species of Corallus, we infer phylogenetic relationships, present the first molecular analysis of the phylogenetic placement of the enigmatic C. cropanii, develop a time-calibrated phylogeny, and explore the biogeographic history of the genus. We found that Corallus diversified within mainland South America, via over-water dispersals to the Lesser Antilles and Central America, and via the traditionally recognized Panamanian land bridge. Divergence time estimates reject the South American Caribbean-Track as a general biogeographic model for Corallus and implicate a role for events during the Oligocene and Miocene in diversification such as marine incursions and the uplift of the Andes. Our findings also suggest that recognition of the island endemic species, C. grenadensis and C. cookii, is questionable as they are nested within the widely distributed species, C. hortulanus. Our results highlight the importance of using widespread taxa when forming and testing biogeographic hypotheses in complex regions and further illustrate the difficulty of forming broadly applicable hypotheses regarding patterns of diversification in the Neotropical region. 相似文献

The volatile components from the labial glands of males of six Japanese bumblebee species were analyzed and compared. Clear species‐specificity was found. Ethyl dodecanoate was identified as the major component from the glands of Bombus (Bombus) hypocrita hypocrita and Bombus (Bombus) hypocrita sapporoensis while dihydrofarnesal and dihydrofarnesol were the major components from Bombus (Bombus) ignitus. Citronellol and trans,trans‐farnesol were found from Bombus (Pyrobombus) ardens ardens and Bombus (Diversobombus) diversus diversus, respectively. trans,trans‐Farnesol was also found from Bombus (Diversobombus) diversus tersatus in Hokkaido. Such differences strongly suggest that these chemicals play an important role in reproductive isolation between sympatric species of Japanese bumblebees. 相似文献

In 1976, red clover necrotic mosaic virus (RCNMV) was identified in red clover variety trials at the Scottish Colleges of Agriculture and at the trial centres of the National Institute of Agricultural Botany (NIAB) in Northumberland, Dyfed, Devon and Cambridge. In 1977, RCNMV was also found in two commercial crops of red clover in South Wales. The only previous finding of this virus in Britain was in 1971.
In red clover leaves RCNMV causes veinal chlorosis, often followed by severe necrosis and deformation; the plants become stunted. All cultivars tested were infected either in field or glasshouse experiments and three of the four most susceptible cultivars were tetraploids. Yield losses in cv. Hungaropoly averaged 57% over three cuts. RCNMV was transmitted manually but not through seed or by aphids {Acyrthosiphon pisum and Myzus persicae) or weevils (Apion spp. and Sitona lineatus). Seedlings became infected when grown in pots containing RCNMV-infected plants or soil from infected sites, and the roots of infected test seedlings contained an Olpidium sp. which may be the vector.
