Cryptomonad strain CBD phycoerythrin 566 carries four open-chain tetrapyrrole (bilin) prosthetic groups: three singly thioether-linked bilins at alpha-19, beta-82, and beta-158 and a bilin linked through two thioether bonds at beta-50,61 (amino acid sequence numbering from Wilbanks, S. M., Wedemayer, G.J., and Glazer, A.N. (1989) J. Biol. Chem. 264, 17860-17867). The structures of all four peptide-linked prosthetic groups were determined by 1H NMR spectroscopy. The bilin at beta-82 was identified as phycoerythrobilin (PEB), a common prosthetic group in cyanobacterial and red algal phycobiliproteins. The structures of the remaining bilins were novel. The bilin at alpha-19, designated Cys-bilin 618, differed from PEB in having additional double bonds between C-2 and C-3 of ring A and between C-12' and C-12", i.e. an acryloyl substituent at C-12 of ring C. The doubly linked bilin at beta-50,61 designated DiCys-bilin 584, differed from doubly linked PEB (Schoenleber, R.W., Lundell, D.J., Glazer, A.N., and Rapoport, H. (1984) J. Biol. Chem. 259, 5481-5484) in possessing an acryloyl substituent at C-12 of ring C in place of a propionyl substituent. Similarly, the bilin at beta-158, designated Cys-bilin 584, differed from singly-linked PEB in possessing an acryloyl substituent at C-12 of ring C in place of a propionyl substituent. The three novel cryptomonad bilins join heme d1 and chlorophylls c1, c2, and c3 as the only known porphyrin-derived natural products with acryloyl substituents.  相似文献

Structures of the open-chain tetrapyrrole (bilin) prosthetic groups of the cryptophycean biliproteins phycocyanin 645 (Cr-PC 645; from strain UW374), phycoerythrin 566 (Cr-PE 566; from strain Bermani) and phycoerythrin 545 (Cr-PE 545; from Proteomonas sulcata Hill & Wetherbee) were examined by absorption, 1H NMR spectroscopy, and mass spectrometry. These biliproteins carry the following covalently attached bilins: Cr-PC 645 (alpha subunit) has one mesobiliverdin, (beta subunit), two phycocyanobilins and a doubly linked 15,16-dihydrobiliverdin; Cr-PC 566 (alpha), bilin 584, (beta), phycoerythrobilin and two bilin 584 chromophores (Wedemayer, G.J., Wemmer, D.E., and Glazer, A.N. (1991) J. Biol. Chem. 266, 4731-4741); Cr-PE 545 (alpha) has one 15,16-dihydrobiliverdin and (beta), only phycoerythrobilins. This is the first report of naturally occurring biliproteins carrying either 15,16-dihydrobiliverdin or mesobiliverdin chromophores. Native cryptomonad phycobiliproteins have been classified on the basis of the position of their long wavelength absorption maxima. However, comparison of the bilins of Cr-PE 566 from strain Bermani with those of Cr-PE 566 of strain CBD shows that the two proteins carry different bilins on the alpha subunit. Consequently, the identity of the bilin prosthetic groups on cryptophycean phycobiliproteins cannot be unambiguously inferred from simple inspection of the visible absorption spectra.  相似文献

Gymnodinium aeruginosum has the usual fine structure of a dinoflagellate but does not seem to contain a well elaborated peduncle or a microtubular basket. Naked cells are surrounded by a single large amphiesmal vesicle. It houses an endosymbiont with typical blue-green cryptophycean chloroplasts (generally only one), cryptophycean starch grains in the periplastidal cytoplasm without a nucleomorph, and two membranes separating the periplastidal cytoplasm from the cryptophycean cytoplasm which contains mitochondria, ER, vesicles and ribosomes, but no eukaryotic nucleus. The endosymbiont is surrounded by a single membrane. Possible ways of the acquisition of the endosymbiont and the problem of the existence of ribosomes within a compartment without nucleus are discussed.Devoted to Prof. Dr.L. The present study is focused on purification, characterization and comparison of phycoerythrins from three different marine cyanobacterial cultures--hormidium sp. A27 DM, Lyngbya sp. A09 DM and Halomicronema sp. A32 DM. 'Phycoerythrin' was successfully purified and characterized. On SDS-PAGE, the PE purified from all three young cultures showed four bands--corresponding to α and β subunits of each of PE-I and PE-II. However, phycoerythrin purified after prolonged growth of Phormidium sp. A27 DM and Halomicronema sp. A32DM showed only one band corresponding to 14 kDa whereas Lyngbya sp. A09 DM continued to produce uncleaved phycoerythrin. The absorption spectra of purified PEs from all the three young and old cultures showed variations however the fluorescence studies of the purified PEs in all cases gave the emission spectra at around 580 nm. The described work is of great importance to understand the role of phycoerythrin in adapting cyanobacteria to stress conditions.  相似文献

Porphyra drewiana Coll et Oliveira, sp. nov., is described from plants collected on the south‐east coast of Brazil. The species proposed is monostromatic, monoecious, monoplastidial, without marginal microscopic teeth and does not produce monospores. Both phases, leafy and filamentous, have three chromosomes. Morphologically the most similar species is Porphyra spiralis Oliveira et Coll var. amplifolia Oliveira et Coll, from which it differs by: (i) thallus gross morphology; (ii) scattered pluristromatic areas of vegetative cells; (iii) division of the plastids prior to the nucleus at the first division of the carpospores mother cell; (iv) the number of carpospores and spermatia produced per mother cell; and (v) morphology and behavior of the filamentous phase in cultures. The following five species of microscopic tuft-forming/encrusting brown algae (Phaeophyta) are newly recorded for the Island of São Miguel (Azores): Nemoderma tingitana Schousboe ex Bornet, Pseudolithoderma roscoffense Loiseaux (Lithodermataceae), Hecatonema terminalis (Kützing) Kylin (Punctariaceae), Compsonema saxicolum(Kuckuck) Kuckuck, and Microspongium gelatinosum Reinke (Scytosiphonaceae). Microcystis andNodularia species produce cyclic hepta- and pentapeptides, microcystins and nodularin, respectively, both containing the same unusual C20 amino acid, abbreviated Adda. Biosynthesis of nodularin fromNodularia and especially of Adda employs a pathway similar to that employed byMicrocystis for producting microcystins. Nearly 30 new microcystins have been isolated in our laboratory from cyanobacteria species and their structures assigned, largely employing tandem FAB mass spectrometry (FABMS/CID/MS). Acyclic peptides, some of them presumed precursors of nodularin and microcystins, have now been isolated and characterized. The numerous analogs identified or synthesized allow the identification of important parameters in a structure-activity relationship study.  相似文献

Protein unfolding of eight different phycocyanins was investigated utilizing circular dichroism and visible spectra. The phycocyanin samples were extracted from algae that are normally found in vastly different environments, and are classified as mesophilic, thermophilic, halophilic and psychrophilic. The ability of these proteins to resist the denaturant urea is in the order of thermophile greater than mesophile, halophile greater than psychrophile. Based on a two-state approximation the apparent free energies of protein unfolding at zero urea denaturant concentration, deltaGH2Oapp, were found to range from 2.4 to 8.8 kcal/mole for the eight phycocyanins at pH 6 and 25 degrees C. The proteins from the thermophile are generally more stable than those from the mesophile. An extra stability of the halophile is believed due to the specific interaction of the proteins and the ions in solution. A correction for deltaGH2Oapp due to minor amino acid differences reveals that the stability and the structural properties of these proteins are primarily affected by this minor difference in amino acid compositions.  相似文献

The taxonomic revision of the carbonate microbiota of the limestone lenses intercalated into the Carboniferous siliciclastic series of Balia-Maden (Turkey) shows that most lenses are early or middle Brigantian in age (latest Visean), only a single lens being of younger age (late Serpukhovian). Calcareous microbiota are abundant in the Balia-Maden lenses carbonates. Microfacies analysis shows the dominance of shallow water environments. The Brigantian assemblage is accurately illustrated. The new foraminiferal taxon Cribrospira baliamadeni nov. sp. is morphologically similar to type-material of Cribrospira panderi von Möller, but has a porous wall with wider pores, almost keriothecal. C. baliamadeni nov. sp. corresponds to the misinterpreted Bradyina and Janischewskina of the previous literature on Balia-Maden lenses. These limestones show a great diversity of algosponges (carbonate microproblematica). Among them, (1) small, atypical Fasciella previously confused with Eosigmoilina; (2) an abundant form described for the first time, Frustulata reticulata nov. sp.; and (3) typical Falsocalcifolium punctatum (Maslov), important for the biostratigraphic implications, are also mentioned. The single Serpukhovian lens consists of a grainstone and contains the age-sensitive alga Archaeolithophyllum johnsoni Racz, and the foraminifers Monotaxinoides gracilis and Janischewskina sp.  相似文献

Comparison of 10 nm filaments from three bovine tissues   总被引:4,自引:0,他引:4  
Enriched fractions of 10 nm filaments were isolated from three bovine tissues and were compared using morphological biochemical, and immunological techniques. We studied keratin filaments from hoof epidermis, 10 nm filaments from corneal epithelium, and 10 nm filaments from brain white matter. The parameters of comparison and results were as follows.
1. 1. Corneal epithelial filaments and keratin filaments repolymerized after a buffered 8 M urea extract of the tissue was dialyzed against a low ionic strength (0.005 M) buffer. However, a greater yield of repolymerized corneal epithelial filaments was obtained if the urea-soluble fraction was dialyzed against the same buffer containing 0.17 M NaCl. Brain filaments harvested by cell fractionation did not repolymerize when similarly treated.
2. 2. Electrophoretic patterns of proteins of filament-enriched fractions from the three sources were different in sodium dodecyl sulphate (SDS) polyacrylamide gels, except for one co-migrating band.
3. 3. Peptide mapping by limited proteolysis of the eluted co-migrating proteins showed few similarities.
4. 4. Amino acid analysis of the co-migrating proteins revealed numerous differences.
5. 5. Antibodies to the co-migrating corneal epithelial filament and brain filament proteins reacted only with their own antigen and whole filament type, and antibody to total keratin filament protein cross-reacted only with keratin filaments.

Mutation to virulence has been measured in intact bacteriophage lambda 15 infected into host cells pre-treated with UVC (254 nm), UVB (313 nm), UVA (334 nm, 365 nm) or visible (405 nm) radiations. We have confirmed that UVC radiation leads to a large enhancement (maximum enhancement factor of 140 in wild-type) of the background spontaneous mutation frequency (non-targeted mutagenesis) and have further shown that this is at least partially dependent on excision repair (maximum enhancement factor of 14 in uvrA strain). In contrast, UVB (313 nm) radiation enhances the mutation frequency by less than a factor of 2. Longer wavelength UVA radiation (334 nm, 365 nm) actually reduces the mutation frequency to 25% of the background levels presumably by reducing the levels of viral replication occurring in the host cells. A visible wavelength (405 nm) has no effect on mutation frequency over the fluence range employed.  相似文献

Red algae genus Laurencia (Rhodomelaceae, Ceramiales) are known to produce a wide range of chemically interesting secondary halogenated metabolites. This investigation delves upon extraction, isolation, structural elucidation and antibacterial activity of inherently available secondary metabolites of Laurencia majuscula Harvey collected from two locations in waters of Sabah, Malaysia. Two major halogenated compounds, identified as elatol (1) and iso-obtusol (2) were isolated. Structures of these compounds were determined from their spectroscopic data such as IR, 1H-NMR, 13C-NMR and optical rotation. Antibacterial bioassay against human pathogenic bacteria was conducted using disc diffusion (Kirby-Bauer) method. Elatol (1) inhibited six species of bacteria, with significant antibacterial activities against Staphylococcus epidermis, Klebsiella pneumonia and Salmonella sp. while iso-obtusol (2) exhibited antibacterial activity against four bacterial species with significant activity against K. pneumonia and Salmonella sp. Elatol (1) showed equal and better antibacterial activity compared with tested commercial antibiotics while iso-obtusol (2) only equaled the potency of commercial antibiotics against K. pneumonia and Salmonella sp. Further tests conducted using dilution method showed both compounds as having bacteriostatic mode of action against the tested bacteria.  相似文献

A rich association of calcareous algae is described from the Lower Cretaceous (Barremian–? Lower Aptian) deposits of the Taft Formation, located south of Aliabad (in the vicinity of Yazd, central Iran). The association mainly consists of dasycladaleans and bryopsidales (udoteaceans and gymnocodiaceans); additionally, rare “solenoporaceans” and cyanophyceans are present. Three new species are described: two are dasycladaleans (Pseudoactinoporella? iranica and Holosporella farsica) and one is an udoteacean (Boueina minima). Many of the described and illustrated algae are mentioned for the first time from Iran.  相似文献

In the Eastern Southern Alps of northern Italy (Carnic Prealps, Friuli region), the shallow-water carbonate platform deposits of the Dolomia Principale Fm. (Norian–Rhaetian, Upper Triassic) show best-preserved platform to basin facies transition. The palaeontological study of an algal-rich level recovered from the platform margin facies (Mt. Pramaggiore) has displayed a very interesting association of Dasycladales. Two new genera (Bystrickyella and Elliottporella) and four new species (Bystrickyella ottii, Elliottporella morelloae, Palaeodasycladus lorigae and Holosporella conradii) have been described. These new data suggest that the Norian represents a period of turnover in the evolutionary history of the green algae community. This stage, placed between two extinctions, end-Ladinian and end-Norian, is here interpreted as a re-organization period of the evolutionary schemes of Dasycladales. The new lineages originated in the Norian developed further and characterized the Early Jurassic scenery. Two new species of didemnid ascidians, Diplosoma ooru sp. nov. and Diplosoma simileguwa sp. nov., are described from coral reefs on Okinawajima (Ryukyu Archipelago, Japan). These two species form green colonies, having a symbiotic association with a prokaryotic alga Prochloron sp. The former species was found at the reef edges in the subtidal zone and the latter was found in a shallow reef lagoon. In these species, the colonies are thinner and the zooids are smaller than those of any other Prochloron-bearing Diplosoma species so far described. Moreover, each of the present new species has a unique combination of stigmatic numbers: 5 stigmata in the first and third rows, 6 in the second row, and 4 in the fourth in D. ooru; 4 stigmata in the first and third rows, 5 in the second row, and 3 in the fourth in D. simileguwa. In both of the new species, the retractor muscle emerges from the underside of the thorax. Larval morphology of D. ooru is also described.  相似文献

In northern Shiraz (SW Iran), Lower Cretaceous carbonate was studied in detail. In this study, nine species of dasycladacea algae were classified. There are different species of dasycladacea algae which belong to seven different genera: Actinoporella, Cylindroporella, Dissocladela, Heteroporella, Neomeris, Salpingoporella, Trinocladus; one species of udoteaceae belongs to Bouenia; one species of acetabulariaceae belongs to Clypeina and the microproblematicum Coptocampylodon was also seen. Among the green algae, dasycladaceae and acetabulariceae are the most frequent and udoteaceans are rare in Zagros Mountains. The genus of Trinocladus is a new record for Lower Cretaceous (Upper Albian) in SW Iran.  相似文献

The selenium-containing phycocyanin from the selenium-rich algae (Spirulina platensis) has been crystallized in two crystal forms by the hanging-drop vapor diffusion techniques. A chromatographic procedure of gel filtration and anion exchange was used for purification. Form I crystal with space group P21 and cell parameters a =108.0 Å , b = 117.0 Å , c = 184.0 Å , β = 90.2° and 12(αβ ) units in the asymmetric unit was obtained by using (NH4)2SO4 as precipitant. These crystals diffract up to 2.8 Å . Form II crystal obtained by using PEG4000 as precipitant belongs to space group P63 with unit cell constants a = 155.0 Å , c = 40.3 Å , γ =120.0° and one(αβ ) unit in the asymmetric unit. The crystals diffract beyond 2.9 Å . The possible stacking forms of phycocyanin molecules in the first crystal form were discussed.  相似文献

Summary The vacuolar equilibrium potential of the lipophilic cation TPMP+ (triphenyl methyl phosphonium) in the giant algaeChara australis andGriffithsia monilis was directly measured. The TPMP+ equilibrium potential was approximately 100mV less negative than the measured vacuolar electrical potential. Thus TPMP+ does not act as a probe of the vacuolar electrical potential and appears to be extruded against an electrochemical gradient. Measurement of the plasmalemma equilibrium potential of TPMP+ showed that extrusion of TPMP+ apparently occurred at both the tonoplast and plasmalemma inChara and at the plasmalemma inGriffithsia. It is concluded that TPMP+ cannot be used as a membrane potential probe inChara orGriffithsia.  相似文献
