A seroepidemiological study on human parvovirus (B19) in Japan was undertaken with serum samples randomly collected from healthy Japanese populations (416 in 1973, 675 in 1984 and 508 in 1987/88). All samples were tested for anti-B19 IgG antibody by the indirect antigen-capture ELISA. The antibody prevalence for ages 0-9 years old in 1984 was significantly higher (16%) than that in 1973 (2%), whereas those for ages 20-29 years and 30-39 years were significantly lower in 1984 (20% and 56%) than in 1973 (67% and 80%) (p < 0.005). After the erythema infectiosum (EI) outbreak in 1986/87, the antibody prevalences for ages 5-9, 10-14 and 15-19 years were 40-85% in Fukuoka, 0-10% in Gunma, and 21-41% in Chiba reflecting each EI incidence in these three prefectures, whereas those for ages 20-29 years remained low (< 20%). These data indicate that B19 virus was transmitted mainly among children and no significant incidence of B19 virus infection in adults has occurred since 1973, resulting in a notable shifting of B19 susceptibility toward older ages including child-bearing females.  相似文献

The dramatic world-wide trend towards increasing body weight seems to be less obvious in the Japanese population. The aim of this study is to extract potentially useful information regarding childhood and adolescence obesity in Japan from series of mean height and mean body mass index between 1948 and 2003. Mean values for height and weight of Japanese boys and girls aged 5+ to 17+ years were obtained from the "Reports on School Health Survey", Ministry of Education, Culture, Sports, Science and Technology, comprising approximately 4.5% of all children and adolescents in Japan between 1948 and 2003. The data were fitted by the Preece and Baines model (Preece & Baines 1978) in order to obtain estimates of the age of peak height velocity (APHV) and final height. Isochrones for height and BMI were calculated based upon measurements that were obtained at the same chronological ages at different historic epochs. The APHV as estimated by Preece & Baines (1978) has decreased from 14.07 to 12.03 years in Japanese boys, and from 11.80 to 9.92 years in Japanese girls, indicating that the tempo of child and adolescent maturation (maturational tempo) has accelerated. Body height increased by 10.1 cm in near adult 17+ year old Japanese males and by 5.7 cm in 17+ year old Japanese females since 1948. Due to the acceleration and the earlier attainment of adult stature, isochrones for height tend to diverge for prepubertal ages and to converge for postpubertal ages. The same is true for weight. Body weight has increased by 11.8 kg in near adult males, and by 4.4 kg in near adult females. Also BMI has increased since 1948. But in contrast to height and weight, the rise in BMI only reflects the acceleration of the maturational tempo. Tempo-conditioned isochrones for BMI are almost horizontal, and even tended to temporarily decrease during the 60ies and the 70's. The BMI of Japanese children and adolescents dramatically contrasts the recent and historic BMI changes in the Western populations. The present study provides no evidence of any major rise in the prevalence of obesity in Japan as expressed by mean BMI for age and time of birth, and suggests that Japanese children and adolescents may be more resistant against those environmental factors that have caused obesity in the affluent Western societies.  相似文献

OBJECTIVE--To determine the changes between 1979 and 1990 in demography and dependency levels in elderly people in residential care. DESIGN--Censuses of those aged 65 years and over in any type of residential care at midnight on 11 December 1979 and 27 November 1990. SETTING--Leicestershire District Health Authority (population 865,133, 1991 census), coterminous with county and social services boundaries. MAIN OUTCOME MEASURES--Age, sex, length of stay, and dependency levels (measured by activities of daily living). RESULTS--In 1990 (1979), 6079 (4678) elderly people were enumerated in 241 (133) establishments, a 30% increase in the numbers of elderly people in residential care and an 82% increase in the number of establishments between 1979 and 1990. Dependency levels rose between 1979 and 1990 in all but the geriatric sector, the greatest increases being found in private residential homes where the largest percentage increase in the number of residents had occurred. CONCLUSIONS--Dependency levels in residential care have risen substantially, particularly in the private sector, even beyond levels expected from the greater numbers of elderly people. With the impending move to community care, dependency levels are likely to rise further, and more appropriate staff training and medical input to homes will become necessary.  相似文献

The cause of multiple sclerosis (MS) is unknown. Despite indications from epidemiological and identical-twin studies that MS is infectious, no virus or other infectious agent has been tightly linked to disease. The isolation of Chlamydia pneumoniae from the cerebrospinal fluid (CSF) of MS patients and the detection of both Chlamydia-specific DNA and antibody in MS CSF have been reported. Other analyses of brain and CSF have shown no significant difference in C. pneumoniae-specific DNA or antibody between MS and control subjects. Recent work has revealed intrathecal production of C. pneumoniae-specific IgG in only 24% of MS patients compared with 5% of control patients. More importantly, the major CSF oligoclonal bands from MS patients did not react to C. pneumoniae.  相似文献

Because broad genetic diversity has recently been detected in Torque teno sus viruses (TTSuV1 and TTSuVk2), the viral genome detection method needs to be improved to understand the prevalence of these viruses. Here, we established single PCR-based detection methods for the TTSuV1 and TTSuVk2a genomes with newly designed primer pairs and applied them to investigate the prevalence of TTSuV1 and TTSuVk2a in Japanese pig populations. The results revealed that 98.2% and 81.7% of the pig farms tested positive for the TTSuV1 and TTSuVk2a genomes, respectively, indicating that both TTSuV1 and TTSuVk2a are widespread in Japan.  相似文献

To determine the type of cell(s) that contain latent varicella-zoster virus (VZV) DNA, we prepared pure populations of neurons and satellite cells from trigeminal ganglia of 18 humans who had previously had a VZV infection. VZV DNA was present in 34 of 2,226 neurons (1.5%) and in none of 20,700 satellite cells. There was an average of 4.7 (range of 2 to 9) copies of VZV DNA per latently infected neuron. Latent VZV DNA was primarily present in large neurons, whereas the size distribution of herpes simplex virus DNA was markedly different.  相似文献

Skin biopsy samples from varicella-zoster virus (VZV)-infected patients examined by immunohistochemistry demonstrated VZV replication in nonepithelial cell types. ORF29p, a nonstructural nuclear protein, was found in nerves of two of six patients with chickenpox. In tissue culture, ORF29p was secreted by VZV-infected fibroblasts. Extracellular ORF29p can be taken up through endocytosis by human neurons, implying a novel role for this protein in pathogenesis.  相似文献

Varicella-zoster virus (VZV) gene 51 encodes a protein which is homologous to UL9, the origin of DNA replication-binding protein of herpes simplex virus type 1. No genetic information is available on VZV gene 51, but its product has been shown to bind to virtually the same recognition sequence as does UL9 (D. Chen and P. D. Olivo, J. Virol. 68:3841-3849, 1994; N. D. Stow, H. M. Weir, and E. C. Stow, Virology 177:570-577, 1990). We report here that gene 51 can complement a UL9 null mutant (hr94) (A. K. Malik, R. Martinez, L. Muncy, E. P. Carmichael, and S. K. Weller, Virology 190:702-715, 1992), but at a level which is only 20% of that of UL9. Quantitation of viral DNA synthesis suggests that this phenotype is due to a defect in viral DNA synthesis. Regardless, the ability of VZV gene 51 to complement UL9 suggests that alphaherpesviruses have a highly conserved mechanism of initiation of viral DNA synthesis.  相似文献

Introduction

Calcium pyrophosphate deposition (CPPD) may cause severe arthropathy, major joint destruction and treatment options are limited. The aim of this study was to test the therapeutic efficacy of methotrexate (MTX) in chronic or recurrent CPPD arthropathy.

Methods

Patients with CPPD arthropathy were randomized to receive either weekly subcutaneous injections of 15 mg/week of MTX or placebo (PBO) for three months, in a double-blind, crossover randomized controlled trial. Inclusion criteria comprised definite CPPD disease, recurrent arthritis or persistent polyarthritis, and an insufficient response to NSAIDs, glucocorticoids or colchicine. The primary outcome was an improvement in the disease activity scores based on 44 joints (DAS44). The analysis was performed on an intent-to-treat basis.

Results

We randomized 26 patients, and compared 25 treatment periods on MTX with 21 treatment periods on PBO. Baseline characteristics were balanced between the groups. The evolution of the DAS44 was not statistically significantly different between groups (median DAS44 decreased by −0.08 on MTX versus −0.13 on PBO, after three months, P = 0.44). Furthermore, pain levels remained stable in both groups (median change in VAS Pain −1 unit on MTX and 0 on PBO, P = 0.43), and none of the secondary outcomes was significantly different between the two groups. Minor adverse events (AE) did not differ in frequency between the groups, but the only serious AE occurred on MTX (bicytopenia).

Conclusions

The results of this trial with MTX in this older population with chronic or recurrent CPPD arthropathy suggest no strong effect of MTX on disease activity.

A monitoring study of serum selenium concentration was carried out on healthy blood donors from different regions of Switzerland. With an overall mean serum concentration of 98 microg/L (n=1847), the selenium status of the healthy adults can be assessed as adequate. The lowest measured concentration of 62 microg/L denotes that the risk of a marginal selenium deficiency in the Swiss population is small. Compared to a similar study carried out in 1993, the status improved for both genders, although the increase is higher among women. It is noteworthy that the supplementation practice has increased during this period with ca. 2% of the population taking Se-micronutrients. No correlation between the age and the serum concentration is detected, even though the age-range was enlarged in this study to 18-68 years for the women and to 19-72 years for the men. The place of residence only marginally influences the selenium concentration, which implies a certain uniformity of the dietary habits between the various regions of the country.  相似文献

Varicella-zoster virus (VZV) encodes at least six glycoproteins. Glycoprotein I (gI), the product of open reading frame 67, is a 58- to 62-kDa glycoprotein found in VZV-infected cells. We constructed two VZV gI deletion mutants. Immunoprecipitation of VZV gE from infected cells indicated that cells infected with VZV deleted for gI expressed a gE that was larger (100 kDa) than that expressed in cells infected with the parental virus (98 kDa). Cell-associated or cell-free VZV deleted for gI grew to lower titers in melanoma cells than did parental VZV. While VZV deleted for gI replicated in other human cells, the mutant virus replicated to very low titers in primary guinea pig and monkey cells and did not replicate in Vero cells. When compared with the parental virus, rescued viruses, in which the gI deletion was restored with a wild-type allele, showed a similarly sized gE and comparable growth patterns in melanoma and Vero cells. VZV deleted for gI entered Vero cells; however, viral DNA synthesis was impaired in these cells. The VZV gI mutant was slightly impaired for adsorption to human cells. Thus, VZV gI is required for replication of the virus in Vero cells, for efficient replication of the virus in nonhuman cells, and for normal processing of gE.  相似文献

To observe trends in the surgical therapy of pulmonary tuberculosis, the records of patients treated during the last ten years at the Tuberculosis Division of the San Diego County General Hospital (Vauclain Home) were reviewed. In this decade, a chemotherapeutic revolution permitted more patients to be treated with fewer beds, lower mortality and shorter hospitalization. Pneumoperitoneum has replaced other forms of temporary collapse. Pneumothorax, phrenic nerve interruption and pneumonolysis have been abandoned in favor of extraperiosteal plombage, particularly in older, poor risk patients. The use of permanent collapse measures as definitive treatment has decreased, thoracoplasty and extrapleural pneumothorax having been virtually abandoned. The use of resection in patients with permanent collapse failure, residual cavities, bronchostenosis and destroyed lobes or lungs has become common, and good results have been obtained.  相似文献

Global warming leads to shifts in vegetation types in given temperate environments. The fastest species movement is due to the globalized supply and use of exotic plants in gardening and urban landscaping. These standard practices circumvent dispersal limitations and biological and environmental stresses; they have three major global impacts: (i) the enhancement of biological invasions, (ii) the elevation of volatile organic compound emissions and the resulting increase in photochemical smog formation, and (iii) the enhancement of CO(2) fixation and water use by gardened plants. These global effects, none of which are currently considered in global-change scenarios, are increasingly amplified with further warming and urbanization. We urge for quantitative assessment of the global effects of gardening and urban landscaping.  相似文献

The sugarcane borerDiatrea saccharalis (F.) is the most serious and destructive insect attacking sugarcane in Louisiana. Losses in sugar yields ascribed to damage by this pest were estimated to have averaged 13% anally from 1937 to 1957. Biological and cultural control measures have been of little benefit against economically damaging infestations ofD. saccharalis and insecticides have historically assumed a major role in control programs. The maximum number of insecticide applications recommended to growers for full-season control ofD. saccharalis on sugarcane has been reduced from 12 to 3 within the past decade. Management practices most responsible for reductions in applications are: 1) discontinuance of insecticidal control of the first generation, 2) improvement of field survey methods for detecting potentially damaging infestations, 3) utilization of an economic injury threshold to ascertain need for insecticide treatment, 4) replacement of ryania and cryolite in control programs with synthetic organic insecticides that provide more effective and longer periods of control, and and 5) more emphasis on host plant resistance to reduce insecticide use.  相似文献
