We present 10 microsatellite markers for the buttonwood mangrove, Conocarpus erectus, a wide-range mangrove associate species. Polymorphism was assessed among individuals from six different populations along the Pacific Coast of Mexico and Costa Rica, as well as in two individuals from the Yucatan Peninsula in the Atlantic. The number of alleles detected in the Pacific ranged from two to five. All loci amplified in the Yucatan samples and seven loci revealed a unique Atlantic allele. These markers will be useful for studies in the conservation of the species and to study the basic biology of C. erectus.  相似文献

Stem discs from trees of known age were used to determine the periodic nature of the growth rings formed in Laguncularia racemosa and to describe the anatomical features of these rings. The growth rings were scarcely distinct on microscopic examination, but they were well distinguishable macroscopically, with alternating light brown and dark brown layers. Cross-dating analysis revealed the occurrence of annual growth rings in L. racemosa. The existence of annual growth rings in L. racemosa suggests that it may have great potential for dendrochronology and should encourage age-related studies on the dynamics of mangrove forests. These studies can be important for the evaluation of climate change impact on mangrove ecosystems, as well as for the analysis of effects related to climate variability on plant communities.  相似文献

Sonneratia caseolaris is a typical non-viviparous mangrove species and a key component of mangrove community in the Indo-West Pacific region. Here we isolated nine microsatellite simple sequence repeat (SSR) loci from the genome of S. caseolaris. Our isolated loci provided SSR markers with polymorphism of 2–6 alleles per locus. The expected and observed heterozygosities ranged from 0.242 to 0.745 and from 0.083 to 0.417, respectively. Cross-species amplification in S. alba and S. ovata showed that a subset of these markers holds promise for these congeneric species. These polymorphic SSR markers would be useful tools for population genetics studies on S. caseolaris as well as other congeneric species.  相似文献

This study assessed if mature leaves of Laguncularia racemosa were able to demonstrate salt secretion, and if the magnitude of secretion was a function of soil salinity. Thus, salinity influence on the osmolality of leaf tissue, xylem sap and leaf secretion was assessed in field and glasshouse experiments. As salinity increased, solutes were accumulated in sufficient quantity to decrease osmotic potential over the whole range of water potential. In the field, xylem osmolality (mol m–3) increased with salinity from 32.4±2.9 at 17 to 38.2±0.6 at 28. Similarly, in the glasshouse, xylem sap osmolality (mol m–3) increased from 33.4±1.8 (15) to 40.6±1.5 (30). Changes in Na+ concentration explained about 51–58% of increase in xylem osmolality. Rates of secretion (mmol m–2 day–1) in the field increased from 0.80±0.12 (17) to 1.16±0.14 (28), and in the glasshouse the secretion increased from 0.73±0.07 (15) to 1.25±0.07 (30). The Na+ accounted for 40–53% of total secretion. This study presented evidence of the capability of mature leaves of L. racemosa to secrete salt for the first time, and that the rates of secretion were enhanced as soil salinity increased.  相似文献

Twenty-five polymorphic microsatellite markers were developed for the endangered red-cockaded woodpecker (Picoides borealis). The number of alleles ranged from two to five and observed heterozygosities ranged from 0.036 to 0.750. 为了探究外来红树植物拉关木对乡土红树植物的化感作用,该研究观察了不同浓度(0.1、0.3、0.5g·mL~(-1))的拉关木根、叶水浸提液对乡土红树植物桐花树和正红树的胚轴(种子)萌发、幼苗生长及叶片抗氧化酶活性的影响。结果表明:(1)拉关木水浸提液对桐花树种子的成苗率、萌发指数和根长均存在抑制作用,其中对根长的抑制作用随水浸提液浓度的提高而增强。(2)根水浸提液对桐花树幼苗的根长、苗高、生物量等生长指标的影响总体上均表现为低浓度促进,高浓度抑制。(3)拉关木水浸提液对正红树胚轴的萌发率、萌发指数、生长指标均表现为促进作用,且根水浸提液0.1、0.3 g·mL~(-1)处理组的芽长以及根、叶水浸提液0.1、0.3 g·mL~(-1)处理组的生物量显著大于对照组;拉关木水浸提液对正红树幼苗的生物量也表现为促进作用。(4)抗性生理方面,随着拉关木水浸提液浓度的升高,桐花树和正红树幼苗SOD活性降低,正红树幼苗POD活性在根水浸提液0.3 g·mL~(-1)和叶水浸提液0.1 g·mL~(-1)处理组显著高于对照组。以上结果表明,不同乡土植物对拉关木化感作用的敏感性不同,拉关木水浸提液抑制了桐花树的生长,而对正红树的生长则表现出一定程度的促进作用。  相似文献

The marine phytoplankton, Karenia mikimotoi, causes severe red tides which are associated with mass mortality of marine fish, and have expanded their distributions in the coastal waters of western Japan. To assess the dispersal mechanism, a population genetic study using highly polymorphic genetic markers is one of the crucial approaches. Here we developed 12 polymorphic microsatellite markers from K. mikimotoi. These loci provide a class of highly variable genetic markers, as the number of alleles ranged from 5 to 23, and the estimate of gene diversity was from 0.551 to 0.933 across the 12 microsatellites. We consider these loci potentially useful for detailing the genetic structure and gene flow among K. mikimotoi populations.  相似文献

Cycas hainanensis is an endangered cycad species endemic to Hainan Island, China. To enrich our scientific conservation for this species, we developed eight microsatellite markers using repetitive DNA enriched libraries. The number of alleles per microsatellite locus varied from 8 to 17. The expected (H E ) and observed (H O ) heterozygosities varied from 0.4754 to 0.8846 and from 0.3636 to 0.9600, respectively. These markers will be employed to determine whether the ex situ C. hainanensis individuals in Sourh China Botanical Garden capture a representative portion of genetic diversity of the wild populations.  相似文献

Sixty-eight primer sets for microsatellite loci were developed from microsatellite motif enriched genomic libraries of pooled DNA from the polyploid green and white sturgeon (Acipenser medirostris and A. transmontanus). Four individuals from each species were screened for polymorphism at these loci. Forty-eight loci amplified in both species, and some exhibited species-specific amplification for white or green sturgeon (8 and 12 loci, respectively). The number of alleles per locus ranged from one to 12. At least 68% of the green and 65% of the white sturgeon loci we developed are polysomic.  相似文献

Olive ridleys, one of the widely distributed marine turtle species has undergone declines in recent years due to multiple anthropogenic factors warranting conservation efforts for which assessment of genetic variability in existing populations become critical. Here we describe development of ten new microsatellite markers from a short sequence repeat-enriched partial genomic DNA library, which are found to be highly informative for genetic studies. Eight of these markers when tested on 83 olive ridley turtles revealed high allelic diversity (4–27 alleles per marker), and high observed and expected heterozygosity estimates that ranged from 0.29 to 0.82 and 0.62 to 0.94, respectively. Two microsatellites were monomorphic in the tested olive ridley samples, but were found to be informative/polymorphic when tested on related marine turtle species. More importantly, nine of the new markers showed robust cross-species amplifications in three related species Dermochelys coriacea, Chelonia mydas and Eretmochelys imbricata. Thus, this study describes ten new microsatellite markers and also demonstrates their potential as efficient genetic markers in studies related to parentage analysis, population structure, phylogeography and species relationships of olive ridleys and other marine turtle species.  相似文献

Panax ginseng C.A. Meyer, commonly known as Korean or Asian ginseng, is a perennial herb native to Korea and China. Its roots are highly prized for several medicinal properties. The present study describes development and characterization of twenty-two polymorphic microsatellite markers for this species. A total of 99 alleles were detected with an average of 4.5 alleles per locus across 20 accessions. Values for observed (H O ) and expected (H E ) heterozygosities ranged from 0.05 to 1.00 and from 0.18 to 0.73, respectively. Eleven loci deviated from Hardy–Weinberg equilibrium (P < 0.001). Significant (P < 0.05) heterozygote deficiency was observed at 13 loci. Exact test for linkage disequilibrium showed significant values (P < 0.05) between 12 pairs of loci. These microsatellite markers provide powerful tools for understanding population and conservation genetics of this species and also for genetic differentiation and authentication of different Panax species being used in commercial ginseng products.  相似文献

Codonopsis pilosula Nannf., as an important traditional Chinese medicinal plant species, has been used to treat fatigue, thirst and loss of appetite. In this study, we developed 10 new microsatellite loci primers from the genome of this species using the combined biotin capture method. The polymorphism of each locus was further assessed through 27 individuals from four geographically distant populations. The number of alleles per locus ranged from 6 to 11. The observed and expected heterozygosity ranged from 0.15 to 0.24 and 0.27 to 0.40, respectively. We further performed cross-priming tests of these loci in the other three congeneric species. These microsatellite markers will be useful for investigating genetic diversity within and between populations of these species.  相似文献

This paper reports the development of microsatellite primers for Nelumbo nucifera Gaerten. By screening genomic libraries enriched with 10 kinds of probes, Seventeen polymorphic loci were isolated and primers were designed. Polymorphism of these 17 loci was assessed in 24 individuals. All the 17 loci are polymorphic and the number of alleles ranged from two to seven. Observed heterozygosity and expected heterozygosity ranged from 0.0000 to 0.9176 and from 0.2837 to 0.7917 respectively. These microsatellite loci should be useful for studying the genetic diversity of N. nucifera.  相似文献

Quercus miyagii is an endemic tree species in the Ryukyu Islands, Japan. We isolated and characterized 15 microsatellite loci in this species. The number of alleles ranged from 2 to 16 and expected heterozygosities from 0.07 to 0.92. This set of markers is potentially useful to investigate the genetic structure, gene flow, and the biogeographic history of Q. miyagii in the Ryukyu Islands, Japan.  相似文献

Fourteen microsatellite markers were isolated from the red mangrove Rhizophora mangle (Rhizophoraceae), a widely distributed mangrove plant in the New World and West Africa. The range of expected heterozygosity of these markers was 0.000–0.672 in the two populations of R. mangle. Cross-species testing was examined for five other species of Rhizophora, and Kandelia candel and Bruguiera gymnorrhiza. All 14 markers could be amplified in R. samoensis, thirteen in R. racemosa, and six markers in all other species of Rhizophora. Our findings greatly increase the utility of these markers.  相似文献

A genomic library enriched with (gT)n repeats from tartary buckwheat (Fagopyrum tataricum) was constructed using 5′-anchored PCR for the development of microsattellite markers. Sequencing analysis of 5 clones from the library showed that they all contained microsatellites (totally 10 loci), and each was unique. An additional locus-specific primer was designed according to flanking sequence. Two of the microsatellite loci of 10 tartary buckwheat varieties were amplified using an anchored primer and a locus-specific primer, which revealed a clear polymorphic pattern. The data confirmed that the degenerate primer was reliably anchoring at the 5′-end of the microsatellite, and the primers developed based on this technology could be used for diversity analysis of tartary buckwheat.  相似文献

Reproduction, development and morphological variation of the marine green algaCaulerpa racemosa var.peltata from the southern part of Japan were studied in culture in the laboratory. Anisogamous biflagellate male and female gametes were produced monoeciously and copulated with each other. Settled zygotes became spherical and increased in volume. After five weeks, they formed two germ tubes which extended in opposite directions. Both germ tubes became elongated and branched, resulting in the formation of creeping, filamentous, protonema-like plants. These plants formed primary shoots which differentiated into creeping rhizomes and upright axes. Each upright axis successively formed ramuli and developed into an assimilator. The morphology of assimilators, i.e., shape and arrangement of ramuli, varied with culture coditions. The effects of temperature and light intensity on the formation of assimilators were investigated with 25 combinations of 5 temperatures (20.0–30.0C) and 5 light intensities (0.5–8.0 klux). The morphological plasticity of this alga is identical to that ofCaulerpa racemosa var.laetevirens, which was previously described by the present authors. Thus, apparently, the plasticity of this taxon is correlated with environmental factors. It appears, moreover, thatC. racemosa var.peltata andC. racemosa var.laetevirens are ecophenes (ecads) of a single species.  相似文献

Papaya has a relatively small genome, displays high levels of phenotypic diversity, and is amenable to transformation, making it attractive as a fruit tree model system. The high level of phenotypic diversity seen among papaya cultivars in the field does not correlate with the low levels of genotypic polymorphism thus far elucidated. The highly mutable nature of microsatellites or simple sequence repeats (SSRs) make them potentially powerful markers for distinguishing deoxyribonucleic acid (DNA) polymorphisms between closely related genotypes. Genomic research for papaya has resulted in a significant quantity of sequence data. We mined 28.1 Mb of bacterial artificial chromosomes end sequences, 5.8 Mb of complementary DNA, and 1.6 Mb of random genomic sequences for SSRs. We generated 938 SSR markers and tested for polymorphism among seven varieties that had been used to produce five mapping populations. The level of polymorphism was highest for Kaek Dum × 2H94 with 210 markers, followed by UH928 × SunUp with 194, AU9 × SunUp with 189, UH918 × SunUp with 177, and Kapoho × SunUp displaying the lowest level with 97. Variation in levels of polymorphism, motif predominance, and motif length between the genomic and genic fractions indicated differential selection pressures acting on the microsatellites in these two fractions. The microsatellites developed in this study will greatly assist in the genetic and physical mapping of the papaya genome as well as enhance breeders’ ability to improve the crop. The study of AFLP analysis in Kandelia obovata, one of the major mangrove species in Japan, revealed the existence of a unique fragment showing stuttered peaks. We cloned this fragment and found a novel microsatellite locus. We report the method used for isolation and the polymorphic nature of this locus among the populations on Iriomote Island.  相似文献
