The present study was aimed to evaluate the effect of different seed priming methods to enhance the sodium chloride (NaCl) and polyethylene glycol-8000 (PEG-8000) stress tolerance in Indian mustard (Brassica juncea L.). Seeds subjected to different priming treatments such as water (hydro-priming), calcium chloride (CaCl2) (chemo-priming), and abscisic acid (ABA) (hormonal-priming) showed increased rate of germination as compared to non-primed seeds. The primed and non-primed seeds were grown for 15 days and then the seedlings were independently subjected to iso-osmotic salt (150 mM NaCl) or PEG-8000 (20%) stress. The different biochemical responses were studied 10 days after treatment. Under NaCl and PEG stress, the dry weight and total chlorophyll content were higher in primed sets as compared to non-primed treatment which was also evident by the phenotype of the seedlings. In general, the higher activities of superoxide dismutase and glutathione reductase resulted in lower oxidative damage, in terms of malondialdehyde content, under NaCl and PEG stress in hydro-primed set as compared to non-primed, ABA-, and CaCl2-primed treatments. Besides, the level of total phenolics and accumulation of osmolytes such as free proline, glycine betaine, and total soluble sugars was also lower in hydro-primed set as compared to other primed and non-primed treatments. The study thus suggests the use of hydro-priming as a simple and cost-effective strategy to alleviate the NaCl and PEG induced stress in B. juncea.  相似文献

In order to discriminate between the ionic and osmotic components of salt stress, sugarcane (Saccharum officinarum L. cv. Co 86032) plants were treated with salt-NaCl or polyethylene glycol-PEG 8000 solutions (?0.7 MPa) for 15 days. Both the salt and PEG treatments significantly reduced leaf width, number of green leaves, and chlorophyll stability index. Osmotic adjustment (OA) indicated that both the stresses led to significant accumulation of osmolytes and sugars. Salt stressed plants appeared to use salt as an osmoticum while the PEG stressed plants showed an accumulation of sugars. Oxidative damage to membranes was not severe in plants subjected to salt or PEG stress. The salt stressed plants showed an increase in the activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX), while PEG stress led to an increase in SOD but not APX activity as compared to the control. Thus, results indicate that the iso-osmotic salt or PEG stress led to differential responses in plants especially with respect to growth, OA, and antioxidant enzyme activities.  相似文献

Though seed priming has been emerged as an effective and pragmatic approach, efforts are being made to discover and optimize the new priming agents which are cheaper and easily accessible to the farmers. Here, we established two independent experiments, to ascertain the role of aspirin priming in salinity tolerance of wheat. In the first experiment, various concentrations of aspirin (125, 250, 375, and 500 ppm) were examined for emergence and seedling growth of wheat. A non-primed control, hydropriming, and hydrogen peroxide priming treatments were also maintained for comparison. Among the different treatments, seeds primed with 125 and 250 ppm aspirin depicted better emergence, vigorous seedling growth, and higher starch metabolism. Therefore, these treatments were further used in the second experiment under salinity stress (10 dS m?1). Salinity stress caused delayed and erratic emergence hampered the shoot and root growth, chlorophyll contents, and enhanced the lipid peroxidation and phenolics content in wheat seedlings. However, wheat seed priming particularly with aspirin effectively alleviated the negative effects of salinity on most of the observed parameters. Aspirin priming also significantly enhanced the activities of antioxidant enzymes (catalase, peroxidase, and superoxide dismutase), and reduced oxidative stress in wheat seedlings. Vigorous growth and greater salinity tolerance of wheat seedlings derived from aspirin primed seeds were related with better starch metabolism, strong antioxidative defense system, and lower lipid peroxidation.  相似文献

In order to discriminate between the ionic and osmotic components of salt stress, sugarcane (Saccharum officinarum L. cv. Co 86032) calli were cultured on media containing NaCl or polyethylene glycol (PEG) 8000 that exerted the same osmotic pressure (−0.7 MPa). PEG stress exposure for 15 days led to significant growth reduction and loss in water content than salt stressed and control tissues. Osmotic adjustment (OA) was observed in callus tissues grown on salt, but was not evident in callus grown on PEG. Oxidative damage to membranes, estimated in terms of accumulation of thiobarbituric acid reactive substances-TBARS and electrolytic leakage was significantly higher in both the stressed calli than the control however, the extent of damage was more in the PEG stressed calli. The stressed callus tissues showed inhibition of ascorbate peroxidase activity, while catalase activity was increased. These results indicate sensitivity of cells to PEG-mediated stress than salt stress and differences in their OA to these two stress conditions. The sensitivity to the osmotic stress indicate that expression of the stress tolerance response requires the coordinated action of different tissues in a plant and hence was not expressed at the cellular level.  相似文献

Salt stress impedes the productivity of wheat (Triticum aestivum L.) in many parts of the world. This study evaluated the potential role of benzyl aminopurine (BAP) and sorghum water extract (SWE) in improving the wheat performance under saline conditions. Seeds were primed with BAP (5 mg L?1), SWE (5% v/v), BAP + SWE, and distilled water (hydropriming). Soil filled pots maintained at the soil salinity levels of 4 and 10 dS m?1 were used for the sowing of primed and non-primed seeds. Salt stress suppressed the wheat growth; seed priming treatments significantly improved the wheat growth under optimal and suboptimal conditions. Total phenolics, total soluble sugars and proteins, α-amylase activity, chlorophyll contents, and tissue potassium ion (K+) contents were increased by seed priming under salt stress; while, tissue sodium ion (Na+) contents were decreased. Seed priming with SWE + BAP was the most effective in this regard. Under salt stress, the tissue Na+ contents were reduced by 5.78, 28.3, 32.2, 36.7% by hydropriming, seed priming with SWE, seed priming with BAP, and seed priming with SWE + BAP, respectively over the non-primed control. Effectiveness of seed priming techniques followed the order SWE + BAP > BAP > SWE > Hydropriming. In conclusion, seed priming with SWE + BAP may be opted to improve the tolerance against salt stress in wheat.  相似文献

Damage to plant productivity due to soil salinity is a major agricultural problem, necessitating the development of effective salinity management measures. Here, we sought the potential effects of yeast and carrot extracts, and their associated mechanisms in the alleviation of seawater-induced salt stress in maize. Pretreatment of maize seeds with yeast or carrot extract provided maize plants with enormous abilities in reducing growth inhibition and biomass loss when exposed to seawater. The better growth performance of yeast extract- and carrot extract-primed plants under saline conditions coincided with improved protection of the photosynthetic pigments, chlorophylls and carotenoids. The primed plants also restricted Na+ accumulation in both roots and shoots while maintaining a higher K+ content and lower Na+/K+ ratio when compared with that of non-primed plants. Yeast extract and carrot extract also potentiated salt tolerance mechanisms by accelerating the production of osmolytes, as evidenced by accumulating levels of total free amino acids and soluble sugars, especially in the roots of primed plants during salinity. The enhanced levels of ascorbic acid and phenolic compounds, and the heightened activities of reactive oxygen species-detoxifying enzymes superoxide dismutase, catalase, and ascorbate peroxidase with concurrent reduction of lipid peroxidation in the leaves of primed plants clearly indicated a positive impact of yeast extract- and carrot extract-priming on the antioxidant system of maize under salt stress. Our results together suggest decisive roles of yeast extract and carrot extract in the management of salt-induced adverse effects in economically important maize, and perhaps other crops.

Cytokinins are often considered abscisic acid (ABA) antagonists and auxins antagonists/synergists in various processes in plants. Seed enhancement (seed priming) with cytokinins is reported to increase plant salt tolerance. It was hypothesized that cytokinins could increase salt tolerance in wheat plants by interacting with other plant hormones, especially auxins and ABA. The present studies were therefore conducted to assess the effects of pre-sowing seed treatment with varying concentrations (100, 150 and 200 mg l−1) of cytokinins (kinetin and benzylaminopurine (BAP)) on germination, growth, and concentrations of free endogenous auxins and ABA in two hexaploid spring wheat (Triticum aestivum L.) cultivars. The primed and non-primed seeds of MH-97 (salt-intolerant) and Inqlab-91 (salt-tolerant) were sown in both Petri dishes in a growth room and in the field after treatment with 15 dS m−1 NaCl salinity. Both experiments were repeated during 2002 and 2003. Among priming agents, kinetin was effective in increasing germination rate in the salt-intolerant and early seedling growth in the salt-tolerant cultivar when compared with hydropriming under salt stress. Thus, during germination and early seedling growth, the cytokinin-priming induced effects were cultivar specific. In contrast, kinetin-priming showed a consistent promoting effect in the field and improved growth and grain yield in both cultivars under salt stress. The BAP-priming did not alleviate the inhibitory effects of salinity stress on the germination and early seedling growth in both cultivars. The increase in growth and grain yield in both cultivars was positively correlated with leaf indoleacetic acid concentration and negatively with ABA concentration under both saline and non-saline conditions. The decrease in ABA concentration in the plants raised from kinetin-primed seeds might reflect diminishing influence of salt stress. However, the possibility of involvement of other hormonal interactions is discussed.  相似文献

Three year data on the effect of water- and mannitol (4%) priming of chickpea seeds (12 h at 25°C) showed higher number and biomass of nodules in the plants from primed seeds than from non-primed seeds. The biomass of nodules increased to 75 DAS but decreased by 90 DAS. Activities of sucrose metabolism enzymes (sucrose synthase (SS) and alkaline invertase) and of nitrogen metabolism (glutamine synthetase (GS), glutamate synthase (GOGAT) and glutamate dehydrogenase (GDH)) in nodules of primed and non-primed crops during development are reported. SS and alkaline invertase activities increased to 70 DAS and then decreased. In primed plants, the higher SS activity in nodules at 60 and 70 DAS might be responsible for providing more energy and carbon skeleton for nitrogen fixation and for ammonium assimilation in primed plants. At 85 DAS, though the SS activity decreased in comparison with the earlier growth stages, it was still higher in nodules of the primed crops than the non-primed crop. Activity of alkaline invertase was maximum at 70 DAS in the nodules of primed and non-primed crops. Priming increased nodule GS activity at 70 and 85 DAS. GOGAT activity was unaffected by priming but GDH activity was greater in nodules from primed crops at 50 DAS. Elevated SS and GS nodule activities in primed chickpeas might be responsible in increasing nodule biomass and metabolic activity thereby increasing seed fill.  相似文献

To our knowledge, little attention has been paid to evaluating ZnO nanoparticles (ZNPs) roles in plants grown under salinity stress. In this study, seeds of lupine (Lupinus termis) plants were grown in plastic pots and exposed to 0 (control) and 150 (S) mM NaCl with or without priming with different concentrations of ZnO [20 mg L?1 (ZNPs1), 40 mg L?1 (ZNPs2), and 60 mg L?1 (ZNPs3)] for 20 days. Salinized plants showed a reduction in plant growth parameters (root length, shoot length, fresh weight, and dry weight) and in the contents of photosynthetic pigments (chlorophyll a and b, and carotenoids) and Zn, as well as in the activity of catalase (CAT) against control plants. On the other side, salinity stress boosted the contents of organic solutes (soluble sugar, soluble protein, total free amino acids, and proline), total phenols, malondialdehyde (MDA), ascorbic acid and Na, as well as the activities of superoxide dismutase (SOD), peroxidase (POD), and ascorbate peroxidase (APX) in stressed plants over control plants. However, seed-priming with ZNPs mostly stimulated growth of stressed plants, which was accompanied by reinforcement in the levels of photosynthetic pigments, organic solutes, total phenols, ascorbic acid and Zn, as well as in the activities of SOD, CAT, POD, and APX enzymes over stressed plants alone. On the contrary, priming with ZNPs caused a decrement in the contents of MDA and Na in stressed plants relative to salinized plants alone. It is worthy to mention that, this improvement in salt tolerance of plants primed with ZNPs was more obvious in plants primed with ZNPs3 and grown both in unstressed and stressed regimes. Thus, our findings suggest that seed-priming with ZNPs, especially 60 mg L?1 ZnO is an effective strategy that can be used to enhance salt tolerance of lupine plants.

The influence of pre-sowing seed treatment with polyamines (2.5 mM putrescine, 5.0 mM spermidine and 2.5 mM spermine) on growth, photosynthetic capacity, and ion accumulation in two spring wheat (Triticum aestivum L.) cultivars MH-97 (intolerant) and Inqlab-91 (tolerant) was examined. The primed seeds of each treatment and non-primed seeds were sown in a field containing 15 dS m−1 NaCl. Although all three polyamines were effective in improving shoot growth and grain yield in both cultivars under saline conditions, the effect of spermine was very pronounced particularly in improving grain yield. Different priming agents did not affect the net CO2 assimilation rate and transpiration rate of either cultivar. However, pre-treatment with spermidine increased stomatal conductance (gs) in the tolerant cultivar, whereas with spermine stomatal conductance decreased in the intolerant cultivar under salt stress. Priming agents had different effects on the accumulation of different ions in wheat plant tissues. When spermidine and distilled water were used as priming agents, they were effective in reducing shoot [Na+] in the tolerant and intolerant cultivars, respectively under saline conditions. Although all priming agents caused an increase in shoot [K+], distilled water was more effective in improving shoot [K+] in both cultivars under salt stress. Pre-treatment with spermidine was very effective in reducing shoot [Cl] under saline conditions particularly in the tolerant cultivar. However, the pattern of accumulation of different ions in roots due to different seed priming treatments was not consistent in either cultivar except that root Na+ decreased due to priming with spermine and spermidine in the intolerant and tolerant cultivars under saline conditions. In conclusion, although all three priming agents, spermine, spermidine and putrescine, were effective in alleviating the adverse effect of salt stress on wheat plants, their effects on altering the concentration of different ions and growth were different in the two cultivars differing in salt tolerance.  相似文献

Plants of both cultivars raised from Trp-and IAA-treated seeds accumulated free salicylic acidin their leaves when under salt stress. Overall, the Trp priming-induced improvement in germination and the subsequentgrowth of wheat plants could be related to ion homeostasis when under salt stress. The possible involvement of salicylicacid in the Trp priming-induced better growth under conditions of salt stress is discussed.  相似文献

Ubiquitin (Ub) is regarded as a stress protein involved in many stress responses. In this paper, sense and antisense transgenic tobacco plants, as well as the wild type and vector control, were used to study the role of Ub in salt tolerance of plants. In sense Ta-Ub2 transgenic tobacco plants, there was higher expression of Ub protein conjugates than in the wild type and vector control, but the reverse trend was observed in antisense Nt-Ub1 transgenic plants. The germination rate of tobacco seed, growth status and photosynthesis of the tobacco plants suggested that over-expressing Ub promoted the growth of transgenic tobacco plants and enhanced their salt tolerance, but the opposite effect was seen in plants with repressed Ub expression. Changes in antioxidant capacity may be one of the mechanisms underlying Ub-regulated salt tolerance. Furthermore, improved tolerance to a combination of stresses was also observed in the sense transgenic tobacco plants. These findings imply that Ub is involved in the tolerance of plants to abiotic stress.  相似文献

To investigate the relationship between nuclear factor Y (NF-Y) and stress tolerance in garlic, we cloned a NF-Y family gene AsNF-YC8 from garlic, which was largely upregulated at dehydrate stage. Expression pattern analyses in garlic revealed that AsNF-YC8 is induced through abscisic acid (ABA) and abiotic stresses, such as NaCl and PEG. Compared with wild-type plants, the overexpressing-AsNF-YC8 transgenic tobacco plants showed higher seed germination rates, longer root length and better plant growth under salt and drought stresses. Under drought stress, the transgenic plants maintained higher relative water content (RWC), net photosynthesis, lower levels of malondialdehyde (MDA), and less ion leakage (IL) than wild-type control plants. These results indicate the high tolerance of the transgenic plants to drought stress compared to the WT. The transgenic tobacco lines accumulated less reactive oxygen species (ROS) and exhibited higher antioxidative enzyme activities compared with wild-type (WT) plants under drought stress, which suggested that the overexpression of AsNF-YC8 improves the antioxidant defense system by regulating the activities of these antioxidant enzymes, which in turn protect transgenic lines against drought stress. These results suggest that AsNF-YC8 plays an important role in tolerance to drought and salt stresses.  相似文献

In this study, the effect of seed priming using ascorbic acid (ASA) on three commercial wheat cultivars i.e., Punjab-2011, Faisalabad-2008, and Ujala-2016 under salinity stress in both homogenous and heterogeneous environments has been investigated. It revealed that different levels of salinity have significantly reduced the growth attributes like percent germination, germination index, radical & plumule length, seed vigor index (In-vitro), seedling length, fresh & dry weight, and total chlorophyll content (In-vivo) with subsequent treatments. Salinity stress was induced by using NaCl in three different concentrations (100, 150, and 200 mM). Seeds of the three cultivars primed with 50, 100, and 150 mg/L ascorbic-acids have not only improved percent germination but also considerably reduced germination time and increased germination index (GI) indicating the potential for tolerating saline conditions. Seedling growth (seedling length, Fresh weight, and dry weight) of seeds primed with 50, 100, and 150 mg/L (ASA) was significantly higher than other non-primed seeds under the prevailing saline conditions. Hormonal priming with different concentrations of ascorbic acid was effective, nevertheless, the best results were obtained with 100 and 150 mg/L (ASA) concentrations. We concluded that the delay in germination and seedling growth was mainly due to excessive Na+ accumulation in the seeds of wheat cultivars. On the other hand, seed priming with various concentrations of ascorbic acid has proved to be effective in inducing salt tolerance in terms of germination parameters, seedling characteristics, and chlorophyll retention in the three local commercial wheat cultivars.  相似文献

氧化铈纳米颗粒(CeO2NPS),因具有较强的自由基清除能力和抗氧化酶特性,已被证明可提高植物的耐盐性,但其对辣椒种子引发作用和机制尚不明确。为揭示CeO2NPS种子引发处理辣椒对盐胁迫下的萌发及幼苗生长的影响,以辣椒品种(Capsicum annuum)茂蔬360为试验材料,设置了7个CeO2NPS浓度(0、0.05、0.1、0.2、0.3、0.4、0.5 mmol·L-1),以未引发处理组为对照,研究不同浓度CeO2NPS种子引发处理后对盐胁迫下辣椒种子萌发、幼苗生物量和生理生化指标的影响。结果表明:(1)0.5 mmol·L-1 CeO2NPS种子引发处理后的种子,其可溶性蛋白质、脯氨酸含量和过氧化氢酶(CAT)活性、抗坏血酸(AsA)含量和AsA/DHA比值显著提高,超氧阴离子(O2-)含量显著降低; 盐胁迫下,该处理种子的发芽率、发芽势、发芽指数、活力指数最大。(2)0.4 mmol·L-1 CeO2NPS种子引发处理的幼苗在盐胁迫下的鲜重、干重和根长最大,幼苗的可溶性蛋白质、AsA含量和AsA/DHA比值均显著提高。综上认为,CeO2NPS引发处理不仅可通过降低种子水势、促进贮藏物质代谢和提高抗氧化能力提高种子在盐胁迫下的发芽率,还可在苗期通过增强蛋白合成和抗坏血酸-谷胱甘肽循环(AsA-GSH)促进盐胁迫下幼苗的生长。  相似文献

Ascorbate (AsA) is a major antioxidant and free-radical scavenger in plants. Monodehydroascorbate reductase (MDAR; EC 1.6.5.4) is crucial for AsA regeneration and essential for maintaining a reduced pool of AsA. To examine whether an overexpressed level of MDAR could minimize the deleterious effects of environmental stresses, we developed transgenic tobacco plants overexpressing Arabidopsis thaliana MDAR gene (AtMDAR1) in the cytosol. Incorporation of the transgene in the genome of tobacco plants was confirmed by PCR and Southern-blot analysis and its expression was confirmed by Northern- and Western-blot analyses. These transgenic plants exhibited up to 2.1-fold higher MDAR activity and 2.2-fold higher level of reduced AsA compared to non-transformed control plants. The transgenic plants showed enhanced stress tolerance in term of significantly higher net photosynthesis rates under ozone, salt and polyethylene glycol (PEG) stresses and greater PSII effective quantum yield under ozone and salt stresses. Furthermore, these transgenic plants exhibited significantly lower hydrogen peroxide level when tested under salt stress. These results demonstrate that an overexpressed level of MDAR properly confers enhanced tolerance against ozone, salt and PEG stress.  相似文献
