Pigmy elephants inhabited the islands from the Mediterranean region during the Pleistocene period but became extinct in the course of the Holocene. Despite striking distinctive anatomical characteristics related to insularity, some similarities with the lineage of extant Asian elephants have suggested that pigmy elephants could be most probably seen as members of the genus Elephas. Poulakakis et al (2006) have recently challenged this view by recovering a short mtDNA sequence from an 800 000 year old fossil of the Cretan pigmy elephant (Elephas creticus). According to the authors of this study, a deep taxonomic revision of Cretan dwarf elephants would be needed, as the sequence exhibits clear affinities with woolly mammoth haplotypes. Poulakakis and colleagues (Poulakakis et al. 2006: Biol. Lett. 2, 451-454), report the recovery of 'authentic' mammoth DNA from an 800,000-year-old fragment of bone excavated on the island of Crete. In light of results from other ancient DNA studies that indicate how DNA survival is unlikely in samples, which are recovered from warm environments and are relatively old (e.g. more than 100,000 years), these findings come as a great surprise. Here, we show that problems exist with the methodological approaches used in the study. First, the nested PCR technique as reported is nonsensical--one of the second round 'nested' primers falls outside the amplicon of the first round PCR. More worryingly, the binding region of one of the first round primers (Elcytb320R) falls within the short 43 base pair reported mammoth sequence, specifically covering two of the three reportedly diagnostic Elephas polymorphisms. Finally, we demonstrate using a simple BLAST search in GenBank that the claimed 'uniquely derived character state' for mammoths is in fact also found within modern elephants.  相似文献

Analysis of sequence data from the internal transcribed spacers (ITS) and 5.8S region of nuclear ribosomal DNA show that Canarian and Madeiran brooms (Genisteae) of the genera Teline, Adenocarpus, and Genista are related to Mediterranean species and not to species from adjacent parts of Morocco. Each separate colonization of the islands has resulted in contrasting patterns of adaptation and radiation. The genus Teline is polyphyletic, with both groups (the "T. monspessulana group" and the "T. linifolia group") separately nested within Genista. Genista benehoavensis (La Palma) and G. tenera (Madeira) form, with G. tinctoria of Europe, a single clade characterized by vestigially arillate seeds. The Canarian species of Adenocarpus have almost identical sequence to the Mediterranean A. complicatus and are likely to be the result of island speciation after a very recent colonization event. This Canarian/Mediterranean A. complicatus group is sister to the afrotropical montane A. mannii which is probably derived from an earlier colonization from the Mediterranean, possibly via the Red Sea hills. The independent colonization and subsequent radiation of the two Teline groups in the Canary Islands make an interesting comparison: the phylogenies both show geographical structuring, each with a central and western island division of taxa. Within the "T. monspessulana group" there is some evidence that both continental and Madeiran taxa could be derived from the Canary Islands, although it is likely that near contemporaneous speciation occurred via rapid colonization of the mainland and islands. The finding of two groups within Teline also has implications for patterns of hybridization in those parts of the world where Teline species are invasive; in California members of the T. monspessulana group hybridize readily, but no hybrids have been recorded with T. linifolia which has been introduced in the same areas.  相似文献

European tits of the genus Parus constitute a complex group of coexisting boreal birds. Here we present a survey of the distribution of three coniferous-living Parus species and one of their main predators, the pygmy owl ( Glaucidium passerinum ), on nine isolated islands in Scandinavia. On all islands the coal tit ( Parus ater ) is the sole tit species when the pygmy owl is absent. The two larger species, the willow tit ( P. montanus ) and the crested tit ( P. cristatus ), only coexist with the coal tit when pygmy owls are present. We suggest that the coexistence of willow tits, crested tits and coal tits is the result of a combination of competition for food and predator-safe foraging sites. The smaller coal tit is superior in exploitation competition for food, while the two larger species have an advantage in interference competition for predator-safe foraging sites. The association between the distribution of the pygmy owl and the two larger tit species on isolated islands in Scandinavia is consistent with the idea that the pygmy owl is a keystone predator.  相似文献

The abilities of some cave-nesting swiftlets to echolocate has traditionally been used to separate the genus Aerodramus , which includes echolocating species, from the genus Collocalia , thought to lack echolocation. Here we report the discovery of echolocation in a member of the latter genus, the pygmy swiftlet Collocalia troglodytes . We also present a well-supported molecular phylogeny for the swiftlets and their relatives based on DNA sequence data from two mitochondrial genes, which we use to reconstruct the evolution of echolocation. Our data provide strong evidence that the swiftlets are a monophyletic group. This monophyly plus the presence of echolocation in C. troglodytes indicate that either (1) echolocation evolved much earlier in the swiftlets than previously thought and has since been lost in most Collocalia taxa, or (2) this ability evolved independently in Aerodramus and Collocalia . Based on our results, echolocation can no longer be considered a useful character for distinguishing these two genera.  相似文献

The Mediterranean Basin as one the world's most biologically diverse regions provides an interesting area for the study of plant evolution and spatial structure in plant populations. The dioecious moss Pleurochaete squarrosa is a widespread and common bryophyte in the Mediterranean Basin. Thirty populations were sampled for a study on molecular diversity and genetic structure, covering most major islands and mainland populations from Europe and Africa. A significant decline in nuclear and chloroplast sequence and allozyme variation within populations from west to east was observed. While DNA sequence data showed patterns of isolation by distance, allozyme markers did not. Instead, their considerable interpopulation genetic differentiation appeared to be unrelated to geographic distance. Similar high values for coefficients of gene diversity (G(ST)) in all data sets provided evidence of geographic isolation and limited gene flow among populations (i) within islands, (ii) within mainland areas, and (iii) between islands and mainland. Notably, populations in continental Spain are strongly genetically isolated from all other investigated areas. Surprisingly, there was no difference in gene diversity and G(ST) between islands and mainland areas. Thus, we conclude that large Mediterranean islands may function as 'mainland' for bryophytes. This hypothesis and its implication for conservation biology of cryptogamic plants warrant further investigation. While sexually reproducing populations were found all over the Mediterranean Basin, high levels of multilocus linkage disequilibrium provide evidence of mainly vegetative propagation even in populations where sexual reproduction was observed.  相似文献

The hexaploids Carthamus creticus and C. turkestanicus are noxious weeds with wide but non-overlapping Mediterranean distributions, and C. creticus, together with another polyploid, C. lanatus, have also invaded similar climatic regions in North and South America, South Africa and Australia. Here we infer their origins using sequences of the plastid intergenic spacer trnH-psbA and the intron trnK and three introns of nuclear low-copy genes of the RNA Polymerase family (RPD2 and the duplicated RPC2), as well as RAPD markers (random amplified polymorphic DNA). Phylogenetic analyses of the nuclear introns and additivity analysis of the RAPD markers support the hypotheses that the two hexaploids are allopolyploids sharing a tetraploid progenitor lineage represented by the broadly Mediterranean C. lanatus, combined with different diploid progenitor lineages consistent with the different geographic distributions of the hexaploids. Whereas C. leucocaulos from the south-eastern Greek Islands represents the diploid progenitor lineage of the western C. creticus, the Irano-Turanian C. glaucus represents the diploid progenitor lineage of the eastern C. turkestanicus. The plastid data suggest that the diploid lineages served as the maternal progenitors of the hexaploids.  相似文献

The tribe Inuleae (Asteraceae) has 10 species endemic to the Macaronesian islands, including the three endemic genera Allagopappus, Schizogyne, and Vierea. Phylogenetic analyses of DNA sequence data from the internal transcribed spacers (ITS) of the nuclear ribosomal DNA of 47 taxa were performed using all Macaronesian endemics and representative species from 21 of the 36 genera of the Inuleae. The resulting ITS phylogeny reveals that Allagopappus is sister to a large clade that contains all genera with a predominantly Mediterranean distribution. This finding suggests that Allagopappus may represent an ancient lineage that found refuge in the Canary Islands following the major climatic and/or geologic changes in the Mediterranean basin after the Tertiary. The Macaronesian endemic genus Schizogyne is sister to Limbarda from the Mediterranean. The third Macaronesian endemic genus, Vierea, is sister to Perralderia, which is restricted to Morocco and Algeria. Pulicaria canariensis is sister to P. mauritanica, a species endemic to Morocco and Algeria. In contrast, P. diffusa from the Cape Verde Islands is sister to a broadly distributed species, P. crispa, that occurs from North Africa to the Arabian peninsula. Based on the ITS data, the genera Blumea, Inula, and Pulicaria are not monophyletic. The ITS trees suggested that Blumea mollis belongs to the tribe Plucheeae, a finding that is congruent with recent morphological evidence. A possible southern African origin for the core of the Laurasian taxa of the Inuleae is also suggested.  相似文献

Intrastrain internal transcribed spacer (ITS) heterogeneity is first reported from Ganoderma, a fungal genus within Basidiomycetes. ITS amplification products from 4 strains, representing 4 Ganoderma species, were cloned and sequenced. Two to five different ITS types were found within a single strain. The clone sequences were analyzed along with other sequences from Ganoderma retrieved from GenBank. The results show that sequence variation within strains varies considerably with species and the heterogeneity may occur in the 3 parts (ITS1, ITS2, and 5.8S) of the ITS region.  相似文献

We examined the molecular phylogeny and chromosomal features of European Helictotrichon species to explore the relationships within the genus and to investigate the origin of several polyploids. Using both approaches, molecular and cytogenetic, revealed the strong impact of allopolyploidization on genome organization from chromosome structure to sequence level. Our research focused on Mediterranean and endemic species of the Alps. Altogether, the molecular phylogenetic analyses include a sample of 17 Helictotrichon species and subspecies, used DNA sequences from the nuclear ribosomal (nr) internal transcribed spacer region (ITS) and the single copy gene topoisomerase 6 (Topo6), and were analysed by maximum parsimony and Bayesian methods. Karyotype structures were investigated by fluorescence in situ hybridization (FISH) and fluorochrome banding. Cytogenetic characters were mapped on the combined phylogenetic tree. The absence or comparatively rare occurrence of different ITS sequence types in some (allo-) polyploid species of Helictotrichon suggests frequent intergenomic homogenization of ribosomal DNA (rDNA) loci due to the phenomenon of concerted evolution. This result implies that the ITS region is not an ideal marker to study polyploid evolution of these grasses. The phylogenetic analysis of the Topo6 region revealed three major clades that concur with three different copy types (termed SAR, SET, PAR), representing the major genome groups in Helictotrichon. A comparison of the molecular phylogenetic trees with the chromosome and karyotype structure supports allopolyploidy of several Helictotrichon species and identifies potential genome donors. A correlation between molecular phylogenetic/cytogenetic results and geographic distribution is expressed by a west-east disjunction, in the narrower or wider sense, of the analysed species. While SAR represents a geographically narrowly distributed southwest Mediterranean genome group, PAR and SET are very widespread (Mediterranean to Asia) and encompass several instances of west-east disjunctions.  相似文献

Aim The Mediterranean region is often regarded as a crossroads where species of various origins meet. However, the biogeographical relationships between this region and contiguous Saharan, Macaronesian and Irano‐Turanian regions have not been investigated in detail. The aim of this study was to characterize the phylogeography of the circum‐Mediterranean species Myrtus communis and to investigate the origin of isolated central Saharan populations of Myrtus nivellei. Location The distribution ranges of M. communis from Macaronesia to the Irano‐Turanian region (173 sampled populations) and of M. nivellei in the mountains (Hoggar, Tassili n’Ajjer, Immidir, Tibesti) of the central Sahara (23 populations). Methods Nuclear ribosomal DNA (nrDNA) sequences of Myrtaceae were used to root the phylogeny of Myrtus, and to date its crown node, according to a detailed review of the palaeobotanical records used for multiple fossil calibration. Chloroplast DNA (cpDNA) sequences were analysed through the determination of genetic diversity indices and by statistical phylogeography. Results Both cpDNA and nrDNA markers indicated east–west genetic differentiation within M. communis. During the late Miocene, a key vicariance event affected the previous circum‐Mediterranean distribution of Myrtus, leading to the isolation of eastern populations. During the late Miocene or early Pliocene, two clades diverged: one is now scattered in the Mediterranean Basin and adjacent regions, whereas the other evolved in the western Mediterranean region. The differentiation of lineages during the Plio‐Pleistocene occurred mainly in the western part of the Mediterranean Basin, which has been at the origin of migrations towards Macaronesian islands and Saharan mountains. This is one of the first plant phylogeographical studies to report migrations from the Mediterranean to the Sahara. Main conclusions The genus Myrtus has persisted in the Mediterranean region since at least the Neogene and its biogeography reflects the cumulation of the species’ responses to successive palaeoenvironmental changes. The current distribution of the genus Myrtus in the Mediterranean Basin and in isolated areas, such as the Macaronesian islands and Saharan mountains, can be explained by the striking ability of this plant not only to persist locally in various refugia, but also to migrate.  相似文献

Lathyrus (Leguminosae; Papilionoideae) is the largest genus in tribe Fabeae and exhibits an intriguing extratropical distribution. We studied the systematics and biogeography of Lathyrus using sequence data, from accessions representing 53 species, for the internal transcribed spacer plus 5.8S-coding region of nuclear ribosomal DNA as well as the trnL-F and trnS-G regions of chloroplast DNA. Our results generally supported recent morphology-based classifications, resolving clades corresponding to sections Lathyrus and Lathyrostylis, but question the monophyly of the large, widespread section Orobus sensu Asmussen and Liston. Sections Orobus, Aphaca, and Pratensis form a predominantly northern Eurasian-New World clade. Within this clade, the North American and eastern Eurasian species, including both Holarctic species (L. palustris and L. japonicus), form a transberingian clade of relatively recent origin and diversification. The South American Notolathyrus group is distant from this transberingian lineage and should be reinstated as a distinct section within the northern Eurasian-New World clade. The Notolathyrus lineage reached the New World most probably through long-distance dispersal from Eurasia. The remaining sections in the genus are centered on the Mediterranean region.  相似文献

A partial sequence of cytochrome b (228 bp) gene of mitochondrial DNA was successfully determined from rib bones of the dwarf elephant Palaeoloxodon antiquus falconeri BUSK, which were excavated from Charkadio cave of the island of Tilos, Dodekanisa, Greece. This is the first report of DNA sequence of a dwarf elephant. The sequences were used to examine the phylogenetic relationships among Elephantidae. Phylogenetic trees reconstructed by the neighbor-joining and maximum parsimony methods provided identical topologies. The results support the "Palaeoloxodon–Elephas" clade, which is consistent with previous morphological reports according to which Palaeoloxodon is more closely related to Elephas than to Loxodonta or Mammuthus.  相似文献

The objective of the present study was to compare the plant morphology, water relations and photochemical efficiency of photosystem 2 in two wild. Mediterranean species Lotus creticus creticus and Lotus creticus cytisoides. L. creticus creticus showed higher density of trichomes and stomatal density on the adaxial leaf surface than L. creticus cytisoides, whereas L. creticus cytisoides showed higher stomatal density in abaxial surface than L. creticus creticus. These morphological traits promoted clear differences in leaf surface water retention and leaf reflectance. Leaf water potential and photochemical efficiency were lower in L. creticus cytisoides than in L. creticus creticus.  相似文献

Aim To investigate the degree of phylogeographical divergence within pygmy whitefish (Prosopium coulterii) and to test hypotheses concerning the origin of disjunct populations within North America. Location North America from western Alaska to Lake Superior. Methods Mitochondrial (ATPase subunit VI) and nuclear (ITS‐1, ITS‐2) DNA sequence variation was assessed across the species’ North American range to test for the existence of distinct phylogeographical groupings of pygmy whitefish associated with known glacial refugia. Coalescent simulations of the mitochondrial DNA (mtDNA) data were used to test hypotheses of population structure. Results This species is composed of two monophyletic mitochondrial clades across its North American range. The two mtDNA clades differed by an average 3.3% nucleotide sequence divergence. These clades were also distinguished by ITS‐2, but the relationships among lineages were not resolved by the ITS‐1 analysis. Coalescent analyses rejected the null hypothesis of the current disjunct distributions being a result of fragmentation of a single widespread ancestral lineage across a variety of effective population sizes and divergence times. Main conclusions The current range disjunctions of pygmy whitefish in North America probably resulted from isolation, genetic divergence, and selective dispersal from at least two major Pleistocene glacial refugia: Beringia and Cascadia. More recent isolation and dispersal from an upper Mississippi refugium is suggested by relationships within one of the clades and by distributional evidence from co‐distributed species. The Beringian and Cascadian refugia have played major roles in the zoogeography of Nearctic temperate aquatics, but the roles of smaller refugia appear more variable among other species.  相似文献

A new onchocercid species, Loxodontofilaria caprini n. sp. (Filarioidea: Nematoda), found in subcutaneous tissues of 37 (33%) of 112 serows (Noemorhedus crispus) examined in Japan, is described. The female worm had the characteristics of Loxodontofilaria, e.g., the large body size, well-developed esophagus with a shallow buccal cavity, and the long tail with three caudal lappets. The male worm of the new species, which was first described in the genus, had unequal length of spicules, 10 pairs of pre- and post-caudal papillae, and three terminal caudal lappets. Deirids were present in both sexes. Among four species of the genus loxodontofiloria: one from the hippopotamus and three from the Elepantidae, L. caprini n. sp. appears close to L. asiatica Bain, Baker & Chabaud, 1982, a subcutaneous parasite of Elephas indicus in Myanmar (Burma). However, L. caprini n. sp. is distinct from L. asiatica in that the Japanese female worm has an esophagus half as long and the microfilariae also half as long with a coiled posterior. The microfilariae were found in the skin of serows. The new parasite appears to clearly illustrate a major event in the evolution of onchocercids: the host-switching. This might have occurred on the Eurasian continent, where elephantids and the lineage of rupicaprines diversified during the Pliocene-Pleistocene, or in Japan, into which some of these hosts migrated.  相似文献

Biological invasions have become one of the main drivers of habitat degradation and a leading cause of biodiversity loss in island ecosystems worldwide. The spread of invasive species poses a particular environmental threat on the islands of the Mediterranean Basin, which are hot spots of biodiversity and contain rare habitats and endemic species, especially on small islands, which are highly vulnerable to biodiversity loss. Following a recent survey, in this paper we aim to provide an overview of the present-day non-native vascular flora of small Mediterranean islands based on a sample of 37 islands located in the middle of the Mediterranean Sea, off the coast of Italy. By comparing the current data with those gathered during a previous survey conducted in the same study area, we also aim to highlight the main changes that have occurred in non-native plant species diversity, establishment and distribution in recent years and to present a first general overview of the most prominent plant taxa in the island’s introduced flora, focusing on those most responsible for these changes and those that pose the greatest environmental threats. We recorded 203 non-native plant species, 147 of which have established on at least one of the islands investigated. Overall, we detected a sharp increase in the number of species, in their levels of establishment and in the extent of their distribution within the study area in recent years. This may be explained by the intensification of research on plant invasions, as well as to new introduction, escape, establishment and invasion events on the islands in recent decades. The most remarkable plants detected include acacias and succulents, two groups that appear to be emerging very rapidly and to be posing new threats to the conservation of the islands’ natural environment, especially the genus Carpobrotus, whose spread into natural habitats containing rare and endemic taxa is seriously threatening biodiversity on both a local and global scale. On the whole, our results show that the plant invasion phenomenon in the study area has in recent years intensified considerably. As this process seems likely to continue, we should expect more establishment events in the future and the further spread of species that are already present. This is of particular conservation concern on the islands investigated in this survey, which are rich in endemisms, but have been facing deep socio-economic and environmental transformations in these last decades as a consequence of the abandonment of traditional management practices and the development of tourism. Our study thus confirms that plant invasions on Mediterranean islands are a serious environmental problem that threatens biodiversity conservation not only in the Mediterranean biogeographic region, but also on the global scale, and highlights the need to further increase efforts aimed at preventing, controlling or mitigating the effects of plant invasions in island ecosystems.  相似文献

Narrow endemics constitute the cornerstone of Mediterranean plant diversity. Naufraga balearica (Apiaceae) is a critically endangered, extremely narrow endemic plant from the western Mediterranean island of Majorca. Because the species belongs to a monotypic genus, N. balearica was hypothesized to be a palaeoendemism. Here we conducted phylogenetic dating, population genetic and climatic niche analyses in order to understand the evolutionary history and conservation perspectives of this flagship species. Phylogenetic dating analysis of nuclear and plastid DNA sequences revealed a late Miocene to early Pliocene divergence between Naufraga and its sister genus Apium, supporting the palaeoendemic status of the former. Amplified fragment length polymorphism (AFLP) markers and plastid DNA sequences of the five Naufraga populations revealed moderate genetic diversity. This diversity is in line with that of other palaeoendemisms from western Mediterranean islands, as revealed by a comparison with 22 other narrow endemic species from this region. Despite the fact that all Naufraga populations are located at a maximum distance of 10 km in a straight line, a strikingly strong population differentiation was found for AFLP markers, which is explained by long-term isolation likely related to short-range pollination and dispersal strategies of the species. While the species is not genetically impoverished, species distribution modelling and microclimatic monitoring revealed that narrow ecological requirements underlie the current extreme rarity of Naufraga and may jeopardize its long-term survival. Our results indicate that a multidisciplinary approach provides powerful tools to develop conservation strategies for evolutionarily singular lineages.  相似文献
