Evidence for Cometabolism in Sewage   总被引:2,自引:2,他引:0       下载免费PDF全文
A procedure was developed to demonstrate cometabolism in models of natural ecosystems. The procedure involves showing the formation of metabolic products in high yield and the lack of incorporation of substrate carbon into cellular constituents. Samples of four 14C-labeled herbicides (trifluralin, profluralin, fluchloralin, and nitrofen) were incubated with sewage aerobically and under discontinuous anaerobiosis for 88 days, and fresh sewage was added at intervals. Products were formed from each of the herbicides in nonsterile, but not in sterile, sewage. The yield of recovered products reached 87% for profluralin and more than 90% for fluchloralin and trifluralin, and the number of products ranged from 6 for nitrofen to 12 for fluchloralin. Concentrating the sewage microflora 40-fold greatly enhanced the rate of conversion. None of the radioactivity from the herbicide entered the nucleoside pool of the sewage microflora. The lack of incorporation of substrate carbon into cells and the almost stoichiometric conversion of the substrate to organic products indicate that members of the microbial community were cometabolizing the test compounds.  相似文献

A fractional factorial design (FFD) and a response surface methodology (RSM) were used to optimize the inoculum composition of six strains for treatment of synthetic domestic wastewater. The model predicted the highest overall specific substrate utilization rate (q) of 6.88 g TOC/(d-gVSS). The value is in accordance with the actual maximum q, and is 1.5 and 1.97 times greater than those without optimization for 4 and 6 strains respectively. Additionally, the shortest time to reach stationary phase (3.5 h) and highest maximum total organic carbon (TOC) removal efficiency (92%) were also achieved under the optimum condition. The results indicated that the FFD and RSM are powerful screening and optimizing tools for the microbial community. The experimental approaches enhance the overall specific rate of substrate utilization as well as other biodegradation parameters.  相似文献

A model for cometabolism is verified experimentally for a defined methanotrophic mixed culture. The model includes the effects of cell growth, endogenous cell decay, product toxicity, and competitive inhibition with the assumption that cometabolic transformation rates are enhanced by reducing power obtained from oxidation of growth substrates. A theoretical transformation yield is used to quantify the enhancement resulting from growth substrate oxidation. A systematic method for evaluating model parameters independently is described. The applicability of the model is evaluated by comparing experimental data for methanotrophic cometabolism of TCE with model predictions from independently measured model parameters. Propagation of errors is used to quantify errors in parameter estimates and in the final prediction. The model successfully predicts TCE transformation and methane utilization for a wide range of concentrations of TCE (0.5 to 9 mg/L) and methane (0.05 to 6 mg/L). (c) 1997 John Wiley & Sons, Inc. AIMS: To investigate the biodegrading ability and cometabolism of synthetic pyrethroid (SP) utilizing bacteria in cultures with various minerals and carbon sources. METHODS AND RESULTS: Previously isolated SP-degrading Pseudomonas sp. and Serratia sp. were used in cultures containing either flumethrin SP or cypermethrin SP formulations. The culture media consisted of either (i) water only, (ii) water and sucrose, (iii) mineral broth or (iv) mineral broth and sucrose. The growth of both organisms was greatest in the mineral broth and sucrose medium, but the growth-limiting factor for Pseudomonas sp. strain Circle was the mineral content whereas for Serratia sp. strain White it was the carbon substrate. CONCLUSION: The greatest extent of degradation of both SP-based compounds occurred with Pseudomonas sp. strain Circle but was dependant on the medium. SIGNIFICANCE AND IMPACT OF THE STUDY: This investigation could lead to the development of a relatively inexpensive medium supplement to enhance the microbial biodegradation of undesirable compounds, either in situ or ex situ. Municipal wastewaters contain a multitude of organic trace pollutants. Often, their biodegradability by activated sludge microorganisms is decisive for their elimination during wastewater treatment. Since the amounts of micropollutants seem too low to serve as growth substrate, cometabolism is supposed to be the dominating biodegradation process. Nevertheless, as many biodegradation studies were performed without the intention to discriminate between metabolic and cometabolic processes, the specific contribution of the latter to substance transformations is often not clarified. This minireview summarizes current knowledge about the cometabolic degradation of organic trace pollutants by activated sludge and sludge-inherent microorganisms. Due to their relevance for communal wastewater contamination, the focus is laid on pharmaceuticals, personal care products, antibiotics, estrogens, and nonylphenols. Wherever possible, reference is made to the molecular process level, i.e., cometabolic pathways, involved enzymes, and formed transformation products. Particular cometabolic capabilities of different activated sludge consortia and various microbial species are highlighted. Process conditions favoring cometabolic activities are emphasized. Finally, knowledge gaps are identified, and research perspectives are outlined.  相似文献

Propachlor, alachlor, and cycloate were extensively metabolized, but the products were organic. Little conversion of propachlor and alachlor was evident in sterilized sewage or lake water. The cometabolism of propachlor was essentially linear with time in lake water and was well fit by zero-order kinetics in short periods and by first-order kinetics in longer periods in sewage. The rate of cometabolism in sewage was directly proportional to propachlor concentration at levels from 63 pg/ml to more than 100 ng/ml. Glucose but not aniline increased the yield of products formed during propachlor cometabolism in sewage. No microorganism able to use propachlor as a sole source of carbon and energy was isolated, but bacteria isolated from sewage and lake water metabolized this chemical. During the metabolism of this herbicide by two of the bacteria, none of the carbon was assimilated. Our data indicate that cometabolism of these pesticides takes place at concentrations of synthetic compounds that commonly occur in natural waters.  相似文献

The process of nitrification has the potential for the in situ bioremediation of halogenated compounds provided a number of challenges can be overcome. In nitrification, the microbial process where ammonia is oxidized to nitrate, ammonia-oxidizing bacteria (AOB) are key players and are capable of carrying out the biodegradation of recalcitrant halogenated compounds. Through industrial uses, halogenated compounds often find their way into wastewater, contaminating the environment and bodies of water that supply drinking water. In the reclamation of wastewater, halogenated compounds can be degraded by AOB but can also be detrimental to the process of nitrification. This minireview considers the ability of AOB to carry out cometabolism of halogenated compounds and the consequent inhibition of nitrification. Possible cometabolism monitoring methods that were derived from current information about AOB genomes are also discussed. AOB expression microarrays have detected mRNA of genes that are expressed at higher levels during stress and are deemed “sentinel” genes. Promoters of selected “sentinel” genes have been cloned and used to drive the expression of gene-reporter constructs. The latter are being tested as early warning biosensors of cometabolism-induced damage in Nitrosomonas europaea with promising results. These and other biosensors may help to preserve the tenuous balance that exists when nitrification occurs in waste streams containing alternative AOB substrates such as halogenated hydrocarbons.  相似文献

A mechanistic understanding of microbial assimilation of soil organic carbon is important to improve Earth system models’ ability to simulate carbon‐climate feedbacks. A simple modelling framework was developed to investigate how substrate quality and environmental controls over microbial activity regulate microbial assimilation of soil organic carbon and on the size of the microbial biomass. Substrate quality has a positive effect on microbial assimilation of soil organic carbon: higher substrate quality leads to higher ratio of microbial carbon to soil organic carbon. Microbial biomass carbon peaks and then declines as cumulative activity increases. The simulated ratios of soil microbial biomass to soil organic carbon are reasonably consistent with a recently compiled global data set at the biome level. The modelling framework developed in this study offers a simple approach to incorporate microbial contributions to the carbon cycling into Earth system models to simulate carbon‐climate feedbacks and explain global patterns of microbial biomass.  相似文献

Biodegradation of nitroglycerin (NG), an inhibitory, nongrowth substrate present in a multicomponent munition wastewater, was investigated in a pilot-scale batch reactor operated with both aerobic and anoxic cycles. A mixed culture was initially acclimated by gradual introduction of NG into influent and subsequently exposed to actual NG-laden production wastewater. System performance revealed that NG was amenable to aerobic biodegradation without adverse impact on removal efficiencies of other pollutants. Temporal NG concentration profiles indicated that an influent concentration of approximately 200 mg/L of NG was reduced to below detection limits in less than 5 h of aeration with no appreciable (<4%) biosorption. Failure of NG-acclimated cultures to utilize NG as a sole carbon source in bench-scale reactors suggested that NG behaved as a non-growth substrate and its degradation possibly occurred by cometabolism. Ethyl acetate present in the waste stream was an adequate growth substrate in terms of both biological and physicochemical properties. High concentrations of NO(3)-N, produced as a result of aerobic degradation of NG and other nitrogenous compounds of the waste, were treated in an anoxic phase. Approximately 95 mg/L of NO(3)-N was denitrified to below detection limits in 5 h of anoxia without the addition of external carbon sources. The aims of the study were to determine group specificity in microbial utilization of root-exudate compounds and whole rhizodeposition; quantify the proportions of carbon acquired by microbial groups from soil organic matter and rhizodeposition, respectively; and assess the importance of root-derived C as a driver of soil microbial community structure. Additions of 13C-labelled root-exudate compounds to organic soil and steady-state labelling of Lolium perenne, coupled to compound-specific isotope ratio mass spectrometry, were used to quantify group-specific microbial utilization of rhizodeposition. Microbial utilization of glucose and fumaric acid was widespread through the microbial community, but glycine was utilized by a narrower range of populations, as indicated by the enrichment of phospholipid fatty acid (PLFA) analysis fractions. In L. perenne rhizospheres, high rates of rhizodeposit utilization by microbial groups showed good correspondence with increased abundance of these groups in the rhizosphere. Although rhizodeposition was not the quantitatively dominant C source for microbes in L. perenne rhizospheres, relative utilization of this C source was an important driver of microbial group abundance in organic soil.  相似文献

The biodegradability of chlorinated methanes, chlorinated ethanes, chlorinated ethenes, chlorofluorocarbons (CFCs), chlorinated acetic acids, chlorinated propanoids and chlorinated butadienes was evaluated based on literature data. Evidence for the biodegradation of compounds in all of the compound categories evaluated has been reported. A broad range of chlorinated aliphatic structures are susceptible to biodegradation under a variety of physiological and redox conditions. Microbial biodegradation of a wide variety of chlorinated aliphatic compounds was shown to occur under five physiological conditions. However, any given physiological condition could only act upon a subset of the chlorinated compounds. Firstly, chlorinated compounds are used as an electron donor and carbon source under aerobic conditions. Secondly, chlorinated compounds are cometabolized under aerobic conditions while the microorganisms are growing (or otherwise already have grown) on another primary substrate. Thirdly, chlorinated compounds are also degraded under anaerobic conditions in which they are utilized as an electron donor and carbon source. Fourthly, chlorinated compounds can serve as an electron acceptor to support respiration of anaerobic microorganisms utilizing simple electron donating substrates. Lastly chlorinated compounds are subject to anaerobic cometabolism becoming biotransformed while the microorganisms grow on other primary substrate or electron acceptor. The literature survey demonstrates that, in many cases, chlorinated compounds are completely mineralised to benign end products. Additionally, biodegradation can occur rapidly. Growth rates exceeding 1 d-1 were observed for many compounds. Most compound categories include chlorinated structures that are used to support microbial growth. Growth can be due to the use of the chlorinated compound as an electron donor or alternatively to the use of the chlorinated compound as an electron acceptor (halorespiration). Biodegradation linked to growth is important, since under such conditions, rates of degradation will increase as the microbial population (biocatalyst) increases. Combinations of redox conditions are favorable for the biodegradation of highly chlorinated structures that are recalcitrant to degradation under aerobic conditions. However, under anaerobic conditions, highly chlorinated structures are partially dehalogenated to lower chlorinated counterparts. The lower chlorinated compounds are subsequently more readily mineralized under aerobic conditions.  相似文献

This paper demonstrates the functionality, laboratory testing and field application of a microbial sensor that is capable of monitoring the organic pollution extent of wastewaters both off-line in a laboratory and on-line in a wastewater treatment plant. The biosensor was first developed in the laboratory using synthetic wastewater and then applied to monitor the effluent of the unit. The basic working principle of the biosensor is based on the on-line measurement of CO2 concentration in the off gas produced during carbon compound degradation by microbial respiration activities. CO2 concentration under operation conditions (constant oxygen flow rate, residence time and pH) is closely related to the extent of organic pollution (biochemical oxygen demand, chemical oxygen demand). CO2 monitoring is carried out by an infrared spectrometer, whereas current organic pollution is determined off-line according to the conventional 5-day lasting BOD analysis. Off gas analysis of CO2 concentration strongly correlates with off-line biochemical oxygen demand measurements allowing continuous on-line monitoring of the organic load within a wastewater treatment plant. Carbon use efficiency (CUE) is being intensively applied to quantify carbon (C) cycling processes from microbial cell to global scales. Energy use efficiency (EUE) is at least as important as the CUE because (i) microorganisms use organic C mainly as an energy source and not as elemental C per se, and (ii) microbial growth and maintenance are limited by energy, but not by C as a structural element. We conceptualize and review the importance of EUE by soil microorganisms and focus on (i) the energy content in organic compounds depending on the nominal oxidation state of carbon (NOSC), (ii) approaches to assess EUE, (iii) similarities and differences between CUE and EUE, and (iv) discuss mechanisms responsible for lower EUE compared to CUE. The energy content per C atom (enthalpy of combustion, the total energy stored in a compound) in organic compounds is very closely (R2 = 0.98) positively related to NOSC and increases by 108 kJ mol−1 C per one NOSC unit. For the first time we assessed the NOSC of microbial biomass in soil (−0.52) and calculated the corresponding energy content of −510 kJ mol−1 C. We linked CUE and EUE considering the NOSC of microbial biomass and element compositions of substrates utilized by microorganisms. The mean microbial EUE (0.32–0.35) is 18% lower than CUE (0.41) using glucose as a substrate. This definitely indicates that microbial growth is limited by energy relative to C. Based on the comparison of a broad range of processes of C and energy utilization for cell growth and maintenance, as well as database of experimental CUE from various compounds, we clearly explained five mechanisms and main factors why EUE is lower than CUE. The two main mechanisms behind lower EUE versus CUE are: (i) microbial recycling: C can be microbially recycled, whereas energy is always utilized only once, and (ii) chemical reduction of organic and inorganic compounds: Energy is used for reduction, which is ongoing without C utilization.  相似文献

Microbial fuel cells represent a new method for producing electricity from the oxidation of organic matter. A mediatorless microbial fuel cell was developed using Escherichia coli as the active bacterial component with synthetic wastewater of potato extract as the energy source. The two-chamber fuel cell, with a relation of volume between anode and cathode chamber of 8:1, was operated in batch mode. The response was similar to that obtained when glucose was used as the carbon source. The performance characteristics of the fuel cell were evaluated with two different anode and cathode shapes, platinised titanium strip or mesh; the highest maximum power density (502mWm(-2)) was achieved in the microbial fuel cell with mesh electrodes. In addition to electricity generation, the MFC exhibited efficient treatment of wastewater so that significant reduction of initial oxygen demand of wastewater by 61% was observed. These results demonstrate that potato starch can be used for power generation in a mediatorless microbial fuel cell with high removal efficiency of chemical oxygen demand.  相似文献

Over the past 30 years, research on the microbial degradation of polycyclic aromatic hydrocarbons (PAHs) has resulted in the isolation of numerous genera of bacteria, fungi and algae capable of degrading low molecular weight PAHs (compounds containing three or less fused benzene rings). High molecular weight PAHs (compounds containing four or more fused benzene rings) are generally recalcitrant to microbial attack, although some fungi and algae are capable of transforming these compounds. Until recently, only a few genera of bacteria have been isolated with the ability to utilise four-ring PAHs as sole carbon and energy sources while cometabolism of five-ring compounds has been reported. The focuss of this review is on the high molecular weight PAH benzo[a]pyrene (BaP). There is concern about the presence of BaP in the environment because of its carcinogenicity, teratogenicity and toxicity. BaP has been observed to accumulate in marine organisms and plants which could indirectly cause human exposure through food consumption. This review provides an outline of the occurrence of BaP in the environment and the ability of bacteria, fungi and algae to degrade the compound, including pathways for BaP degradation by these organisms. In addition, approaches for improving microbial degradation of BaP are discussed.  相似文献

在水-二氧化碳-碳酸盐岩-生物的相互作用下,岩溶碳循环活跃,在全球形成8.24×108 t C/a的岩溶碳汇,约占全球遗漏汇的29.4%,其中部分岩溶碳汇以土壤有机碳的形式固存,因此碱性土壤固碳是未来碳中和的主要途径。微生物作为土壤碳循环的重要驱动者,影响着土壤有机碳主要赋存形式即植物残体碳与微生物残体碳的动态变化。本文通过综述岩溶土壤有机碳库储量、岩溶土壤有机碳库的来源与构成、影响岩溶土壤有机碳库动态的微生物因素以及岩溶土壤有机碳库更新的微生物机制,探讨了微生物对岩溶土壤植物残体碳与微生物残体碳的影响,并提出亟待解决的关键科学问题。这为深入研究岩溶区土壤有机碳库分配、更新及其维持的微生物机制,深化对岩溶土壤碳循环及其微生物机理认识,进而为应对千分之四全球土壤增碳计划提供了参考。  相似文献

Aerobic granules in a sequencing batch reactor (SBR) are subjected to alternative feast and famine conditions, and are able to take up carbon substrate in wastewater rapidly and to store it as intracellular storage products when the substrate is in excess. This phenomenon could not be described by the widely used activated sludge model No.3 (ASM3). In this work, taking adsorption process, microbial maintenance, and substrate diffusion into account, the simultaneous growth and storage processes occurring in an aerobic-granule-based SBR are investigated with experimental and modeling approaches. A new model is established and successfully validated with the experimental results of an SBR fed with soybean-processing wastewater. Simulation results show that our approach is appropriate for elucidating the fates of major model components. Comparison between ASM3 and the model established in this work demonstrates that the latter is better to describe the substrate removal mechanisms and simultaneous growth and storage processes in aerobic granules.  相似文献
