Prolonged storage of apple fruits ( Pyrus malus L. cv. Golden Delicious) at different temperatures (0, 12 and 35°C) decreased the water content in seed coats and endosperms, higher temperatures being much more effective than the lower (0°C) one. No effect of the temperature on the embryo hydration was found. However, a pronounced decrease in water potential in the embryos was observed during the first 9 weeks. The decrease was much faster and the water potential reached lower levels in embryos isolated from seeds pretreated with higher temperatures (12 or 35°C) than from cold-pretreated (0°C) material. Higher temperatures of fruit storage also resulted in a decreased permeability of the embryo membranes to electrolytes and sugars. At the same time, membrane permeability to water was not modified. It is proposed that the previously observed occurrence of the discontinuous type of freezing in apple seeds (Nguyen and Kacperska, Physiologia Plantarum (X): 000-000, 1989) is associated with the temperature-induced dehydration of seed coat and endosperm, whereas a higher super cooling ability of the high-temperature-pretreated embryos is due to a decrease in the free energy of water in the system, and to the effective protection of embryo cells against heterogenous ice nucleation. The changes in water potential showed a high negative correlation with the embryo phospholipid content determined in the other work (Nguyen et al. Plant Physiol. Bio chem. 25: 697–703, 1987). Therefore, it is proposed that changes in matrix potential play an important role in the regulation of the water potential in the embryo cells.  相似文献

Apples ( Pyrus mains L., ev. Golden Delicious) were placed at 0, 12 and 35°C. At different intervals, seeds were taken from fruits, and the ability of isolated embryos to germinate was determined. In parallel, the survival of similar embryos frozen for 30 min at -7°C was estimated. Storage of apples at 0°C led to complete removal of embryo dormancy within ca 10 weeks. At 12°C, dormancy was only partly eliminated, and at 35°C breaking of dormancy did not occur. The onset of secondary embryo dormancy was thus evident as the result of 35 and 12°C treatments. On the other hand, the storage of apples at 35°C was the most effective treatment for induction of frost resistance in the embryos. At 12°C the same level of frost resistance was reached much slower. Even a prolonged storage of apples at 0°C did not allow frost survival of all embryos. The above relations were confirmed in experiments with transfer of fruits from one temperature of storage to another. The temperature of fruit storage affected the dormancy removal in embryos and induction of resistance to rapid freezing in opposite directions.  相似文献

Dormant embryos of Pyrus malus L. cv. Golden delicious, isolated from the fruits at harvest time or after a few months storage at 10 to 15°C, were kept under anaerobic conditions in order to eliminate primary dormancy. Germination tests were then carried out at different temperatures, using three modes of culture depending on the nature of the contact between the embryo and the medium. In CM the distal part of the two cotyledons was immersed in the medium. In RM only the embryonic axis was immersed. In C/2M the embryo was placed flat on the medium, the radicle and the external surface of one cotyledon being in contact with it.
Results showed that primary dormancy was released progressively depending on the duration of the anaerobic treatment. After a treatment of 11 or 13 days the last symptoms of primary dormancy were only apparent when germination tests were carried out at high temperatures (26–30°C) or in CM mode of culture.
When the embryos were kept at 4°C during 3 months inside the fruits, subsequent germination was inhibited at high temperature and in CM mode of culture. When the embryos were kept under anaerobic conditions (7 days) after the chilling treatmem inside the fruits, germination was no longer inhibited. It is concluded that the inhibition of germination at high temperature and in CM mode of culture is due to the persistence of traces of primary dormancy. Therefore, these conditions do not seem to induce secondary dormancy in apple embryos.
After elimination of primary dormancy by anaerobiosis. only application of (±) abscisic acid (3.8 and 19 μM) inhibited germination. These results support the idea that ABA is an important factor in the induction of dormancy. However, the question remains whether this secondary embryo dormancy has the same characteristics as the original primary dormancy.  相似文献

Polyphenols are the predominant ingredients in apple seeds. However, few data are available on the phenolic profile or antioxidant activity in apple seeds in previous researches. In this study, low-molecular-weight phenolic compounds and antioxidant activity in seeds, peels, and flesh of seven apple cultivars grown in northwest China were measured and analyzed using HPLC and FRAP, DPPH, ABTS assays, respectively. HPLC analysis revealed phloridzin as the dominant phenolic compound in the seeds with its contents being 240.45–864.42 mg/100 gDW. Total phenolic content (TPC) measured by the Folin–Ciocalteu assay in apple seed extracts of seven cultivars ranged from 5.74 (Golden Delicious) to 17.44 (Honeycrisp) mgGAE/gDW. Apple seeds showed higher antioxidant activity than peels or flesh; antioxidant activity in seeds varied from 57.59 to 397.70 μM Trolox equivalents (TE)/g FW for FRAP, from 37.56 to 64.31 μM TE/g FW for DPPH, and from 220.52 to 708.02 μM TE/g FW for ABTS. TPC in apple seeds was significantly correlated with all three assays. Principal component analysis (PCA) indicated that Honeycrisp was characterized with high contents of total polyphenols and phloridzin. Our findings suggest that phenolic extracts from apple seeds have good commercial potential as a promising antioxidant for use in food or cosmetics.  相似文献

ABSTRACT. Survival time at subzero temperatures is related to both long-term thermal history and the rate at which the insects are cooled. Insects cooled at one degree per hour survive for up to 3 times as long at a given test temperature than do insects cooled at 1C/min. Survival times are significantly shorter than times taken to freeze. Polyamines and ethylene in the removal of embryonal dormancy in apple seeds   总被引:2,自引:0,他引:2  
Putrescine (Put), spermidine (Spd) and spermine (Spm) were found in seeds of apple ( Malus domestica Borkh. cv. Antonovka), in amounts that increased in the order given. The levels slowly decreased during 30 days of stratification. Exogenous polyamines (PAs) affected germination of isolated embryos in a way dependent on the type of polyamine, its concentration, and the state of the embryo dormancy. The effect of Put and Spd on germination was stimulatory, while that of Spm was inhibitory. Stimulation of germination was also observed when embryos were treated with arginine, ornithine and methionine. Canavanine inhibited germination, and this effect was reversed by arginine or Put. Ethephon, aminooxyacetic acid (AOA) and aminoethoxyvinylglycine (AVG) present during seed stratification had no effect on the levels of endogenous PAs. Put and Spd did not change ethylene production, neither during seed stratification nor during embryo germination, whereas Spm reduced ethylene evolution. The data suggest that Spm plays a role in the maintenance of dormancy by preventing ethylene production, while Put and Spd participate in dormancy removal, independently of ethylene.  相似文献

Geraniol was injected into the core of Jonathan apples susceptible to storage breakdown. It was rapidly metabolized to geranyl β-d-glucoside which was transported to the flesh and metabolized into other compounds. When geraniol was injected into the flesh and prevented from diffusing into the core area, metabolism of geraniol was completely inhibited. The results suggest that some factor associated with the apple seeds is involved in the formation of the glucoside and hence with the induction of breakdown.  相似文献

This paper deals with investigations on fungal infection, moisture content, germinability and deterioration of three seeds, viz., maize (starchy), groundnut (oily) and soybean (proteinaceous) in storage at the locality of Santiniketan, West Bengal, India, under natural condition for 1 year. The airspora of storage environment was trapped using culture plate method. Different species of Aspergillus (A. candidus, A. flavus, A. niger, A. terreus, and A. ruber) were dominant followed by Rhizopus, Penicillium, Curvularia, Fusarium, Alternaria, etc. Seed moisture was maximum in the rainy season followed by a gradual decrease during longer storage. A gradual decrease in field fungi with simultaneous increase in storage fungi accompanied by a reduction in germinability occurred in all seeds as storage proceeded. A gradual loss of carbohydrate (both soluble and insoluble) content in all the seeds were recorded. A loss of protein content was recorded followed by a small increase. Oil content decreased in prolonged storage with simultaneous increase in fatty acid. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

The purpose of this study was to investigate supposedly positive biological effects of coloured hailnets on microclimate, including photosynthetically active radiation (PAR), UV-B, air, soil, fruit and leaf temperature as well as humidity, which in turn may affect leaf anatomy, tree growth and fruit quality; apple was chosen as a model crop at Klein-Altendorf near Bonn, Germany; adjacent uncovered trees served as control. Red and green hailnets transmitted 3–6% more red or green light, without alteration of the red:far red (R–666 nm:FR–730 nm) ratio (0.99–1.01:1) and hence without affecting the phytochrome system. The microclimate was changed with a reduction by 12–23% in PAR and, to a larger extent, by 20–28% in UV, viz. shading. Light measurements at a 45° angle, to mimic the fruit or leaf position, showed that PAR was 90–210 µmol m−2 s−1 larger outside on a sunny summer day than under the white or red-white and 150–340 µmol m−2 s−1 larger than under red-black and green-black hailnets. Air temperature and relative humidity under coloured hailnets decreased by ca. 1.3°C and by ca 2% rh (cloudy) to 5% rh (sunny day), respectively, compared with outside; leaf temperature was decreased by up to 3°C and fruit temperature by up to 6°C. Soil temperatures at 5 cm depth were 0.5–1°C colder under red-black and green-black hailnets, but up to 0.9°C warmer under white and red-white hailnets compared with the uncovered control outside. Alternate bearing had a larger impact on vegetative growth in the affected year than the coloured hailnets; annual trunk diameter increments in cv. ‘Fuji’, i.e. the variety susceptible to alternate bearing, showed a larger variation than cv. ‘Pinova’ without alternate bearing. Reproductive growth, viz. return bloom and leaf anatomy were impaired by the coloured hailnets. Apple trees under dark hailnets developed thinner leaves with a thinner epidermis and fewer layers of palisade cells. These leaves were 3.5°C (dark hailnets) and 2.5°C (white hailnets) cooler than outside on a sunny day compared with ca. 1.5°C (dark hailnets) and 0.85°C (white hailnets) on a cloudy day. Transpirational cooling of cv. ‘Fuji’ leaves was 0.3–0.6°C outside and 1.4–1.6°C under the green-black hailnet on sunny days compared to <0.1°C on cloudy days. As a practical application, apple fruit colouration was dependent on light (PAR and UV-B) transmission of the respective hailnet colour.  相似文献

Phospholipid (PL) and fatty acid composition of chloroplasts of pine needles ( Pinus sylvestris L.) and apple bark tissue ( Malus sylvestris Mill. cv. Golden Delicious) was determined in a series of experiments in which growth temperature and daylength were changed. Trees were exposed to 0 and 20°C and to daylength conditions of 9 and 14 h. All 16 possible combinations were investigated by transfer of the trees from the original condition to each of the other conditions. There was no direct relation between cold hardiness and PL composition in apple bark and pine chloroplasts, when temperature and/or daylength were changed. PL composition seemed to be strongly determined by the sequence of the imposed sets of daylength and temperature. The effect of these environmental factors on PL composition strongly differed from that for cold hardiness. The correlation between the levels of PL (and phosphatidylcholine) and cold hardiness, as reported in the literature, was also evident in this experiment, when treatments, presenting the normal seasonal order, were compared. It seems that the yearly cycle of temperature and daylength is important in determining the PL composition of apple bark and pine chloroplasts.  相似文献

Water relations, desiccation tolerance and longevity of Taxus brevifolia (Nutt.) seeds were studied to determine the optimal stage of development and storage conditions for seeds of this species. Seeds equilibrated to a range of relative humidities (RHs) had unusually low water contents which can be accounted for by the high lipid content of gametophyte tissues (71% of the dry mass). Water relations of embryonic tissue were more typical of those reported for other seed species. The water content below which freezing transitions were not observable in the embryo was ca 0.24 g H2O (g dry weight)−1 (g g−1) for all maturity classes studied. Embryos did not achieve significant levels of desiccation tolerance (survival to water contents less than 0.5 g g−1) until the latter stages of development when dry matter was maximal. Mature embryos could be dried to 0.025 g g−1 (seed water content of 0.010 g g−1) with no loss of viability. Thus, at the latter stages of development, embryo water content could be optimized to avoid both desiccation and freezing damage. Survival of mature seeds declined over a 2-year period when seeds were stored at temperatures between 5 and 35°C and RHs between 14 and 75%, corresponding to seed water contents between 0.015 and 0.07 g g−1. The deterioration rate was slowest for seeds stored at the lowest RH and temperature. Our data indicate that seeds of Taxus brevifolia show orthodox rather than recalcitrant storage characteristics, but that the optimum water content for storage was extremely low. The results suggest that even if stored at optimal water contents and low temperatures, T. brevifolia seeds will be relatively short lived. The high quantity of lipids or reducing sugars may be contributing factors in the poor storage characteristics.  相似文献

High temperature (45°C) inhibits seed germinition and seedling sunflower ( Helianthus annuus L. cv. Mirasol). Treatment of imbibed seeds at 45°C for more than 48 h induces a secondary dormancy, which is associated with progressive decrease of germination ability at optimal temperature (25°C) as well as with abnormal seedling growth. Ethylene (55μl l−1) and 2-chloroethylphosphonic acid (ethephon) (2.5 m M ) improve germination of thermodormant seeds at 25°C. but the abnormal growth of the seedlings remains. O2-enriched atmosphere and dry storage improve germination and normal seedling growth. The induction of thermodormancy in sunflower seeds seems associated with loss of their ability to convert 1-aminocyclopropane-1-carboxylic acid (ACC) to ethylene. Possible effects of high temperature on membranes and ethylene forming enzyme (EFE) are discussed.  相似文献

The photosynthetic contribution of a fruit to its carbon requirement throughout ontogeny and under different growing conditions was quantified in cucumber ( Cucumis sativus L. cv. Corona). In addition, the effects of shading on fruit dry matter accumulation and the diurnal course of the elongation rate were studied. Fruit darkening had no photomorphogenic effect on fruit growth, while the cumulative photosynthetic contribution of a fruit to its own carbon requirement ranged from 1 to 5%. During the day there was always a net CO2 efflux. The photosynthetic rate per fruit, calculated as the difference between rates of CO2 exchange in light and dark, increased during fruit ontogeny, while the photosynthetic rate per unit fruit surface area declined. The latter was not dependent on fruit size. The photosynthetic activity per unit surface area of fruits was estimated to be about 20–30% as efficient as that of leaves. The rate of calculated photosynthesis was reduced by 60–65% when the photosynthetically active radiation incident on the fruit decreased from 200 to 50 μmol m−2 s−1. Temperature (20–30°C) had no pronounced effect on the rate of calculated fruit photosynthesis when fruits of the same developmental stage (temperature sum) were compared. However, the relative photosynthetic contribution of a fruit to its carbon requirement increased when temperature decreased. Moreover, this contribution increased when irradiance increased or fruit growth was reduced by competing fruits. During fruit ontogeny the daily photosynthetic contribution was highest (up to 15%) in young and old fruits, with a small growth rate.  相似文献

Availability of assimilates in apple trees ( Malus domestica cv. Jonagored) was affected by removing young fruits to obtain 3 ranges of fruit/leaf ratios with average values of 130, 268 and 381 fruits per kg leaf dry matter. Fruit analyses were carried out at fruit harvest and 4 times during a 3-week ripening period. The analyses included detection of volatile aroma components from the juice by headspace gas chromatography. At a low fruit/leaf ratio, higher concentrations of total dry matter, soluble solid and titrateable acids were found. The flesh was also firmer, and ethylene development proceeded at a lower rate and reached a lower maximum value. Aroma compounds consisted of ca 20% esters, 73% alcohols and 6% C-6 aldehydes. The production of butylacetate and hexylacetate, which were the dominating esters, peaked during the ripening period and was most pronounced at the lowest fruit/leaf ratios. At the last sampling date this was also the case for butanol, which was the dominating alcohol. Other esters and alcohols behaved similarly, while C-6 aldehydes showed no significant differences in the fruit/leaf ratio. We suggest that the greater availability of assimilates when internal competition is relieved at a low fruit/leaf ratio causes increased accumulation of fatty acid aroma precursors and aroma compounds as well as of sugars, acids and other compounds in the fruits.  相似文献

Miscanthus x ogiformis Honda 'Giganteus' shoot cultures were stored in vitro on proliferation or rooting medium for up to 27 weeks at temperatures of 8, 12, 16, or 20 °C and photosynthetic photon flux densities of 5, 10, or 20 μmol m−2 s−1. Plants survived storage much better on rooting medium than on proliferation medium. Plants stored on rooting medium for 1 week survived well when survival was assessed immediately after storage or after 14 days of acclimatization, but had the lowest survival 28 days after transplantation. With increasing storage period on rooting medium increasing survival was found 28 days after transplantation. This was probably a result of the development of rhizomes and/or roots during storage. Best survival was observed at 20 μmol m−2 s−1 and a temperature of 8-16 °C. Increasing the temperature to 25 °C during the last week of storage improved survival considerably. Root formation was slow at 8 °C, but after 27 weeks of storage the rooting percentage was the same at all storage temperatures. An increasing number of shoots per plant 28 days after transplantation was found with increasing PPFD during storage.Miscanthus shoot cultures can be stored in vitro for at least 27 weeks with limited losses when stored on rooting medium at 20 μmol m−2 s−1, a temperature of 16 °C, and given a 1-week end-of-storage treatment of 25 °C. 温度是苹果花期最为敏感的生态因子,选择两种在黄土高原区具有代表性的不同树龄和树形结构(盛果期小冠开心形和初挂果期自由纺锤形)的富士苹果园,利用小气候梯度自动测定系统在2011—2014年苹果花期进行定位观测,分析花期不同天气类型下(晴天、阴天或多云、雨天)的果园温度梯度及树体温度的变化特征,并基于气象站温度(TM)建立了果园冠下温度(TL)的推算模型.结果表明: 花期果园温度的垂直分布及与园外的差异主要取决于树形结构,而不同天气类型下的差异不显著.平均温度、日最低温度从树冠下到顶部递增,日最高温度、日较差递减.小冠开心形冠层下部晴天日较差最大,多云或阴天冠层中部和顶部日较差较自由纺锤形小.园内外温差的日变化自由纺锤形呈现高 低 高的单波谷形态、而小冠开心形呈单峰形态,园外最低温度高于冠层下部而与冠层中部的温度接近,小冠开心形冠层下的最低温度较园外最低温度更低,特别是多云或阴天更明显,而冠层中部和顶部与园外的温度差异则较自由纺锤形小.线性模型能够较好地推算树冠下部的温度,误差绝对值在1 ℃以内,特别是自由纺锤形果园和雨天条件下效果更好.  相似文献

The lipid composition of leaves has been investigated in different genotypes of cucumber ( Cucumis sativus L.), which differ in temperature requirement for cultivation. In addition the effects of hardening by low but non-chilling temperature, soil heating and grafting (on the chilling-resistant C. ficifolia L.) on lipid composition have been studied. Content and composition of phospholipids and sterols were determined as well as phospholipid/sterol ratio, and fatty acid composition of total lipids and the different phospholipids.
The effects of genetic differentiation and of the various culture treatments on lipid composition of the leaves were very different. Genetic differentiation was evident as higher levels of Iinolenic acid in several phospholipids in the more cold-tolerant cultivars. Hardening the plants by low temperature resulted in a higher phospholipid level (especially phosphatidyl choline), more unsaturated phospholipid, and lowering of the sterol/phiospholipid ratio, all properties which may contribute to a higher membrane fluidity and lower growth temperature limit. Soil healing reduced the phospholipid level of the leaves slightly, and a higher content of 3- trans -hexadece-noic acid in phosphatidyl glycerol was observed. Grafting cucumber on the cold-resistant rootstock of C. ficifolia also raised the level of trans -hexadecenoic acid in phosphatidyl glycerol. The role of this fatty acid in the functioning of the chloroplast is discussed.  相似文献

Twenty day-old winter rape ( Brassica napus L. var. oleifera L. cv. Jantar) seedlings, grown in nutrient solution, were exposed to different shoot/root temperature (i. e. 20/20, 20/3, 3/20 and 3/3°C) for 2 or 4 weeks. Chilling treatments modified markedly the pattern of plant growth as indicated by changes in dry matter accumulation in individual plant parts (leaves, hypocotyls, roots) and decreased leaf specific area. Growth of roots was less sensitive to low temperature than that of shoots. This was reflected by a decrease in shoot/root biomass ratio. Chilling treatments increased freezing resistance, decreased water content and water potential and modified reducing sugar, soluble protein and phospholipid contents in the leaves. A biphasic character of tissue responses to chilling temperature was observed, the most remarkable changes being registered during the first 7 or 14 days of the treatment. Effects of root or shoot exposure to chilling temperature on ice nucleation temperature, LT50, water potential, accumulation of sugars and phospholipids in leaves were additive. All the observations point to the important role of the root system in plant acclimation to cold. Its impact on water status of leaves is emphasized and some mechanisms of root involvement in acclimation processes are proposed.  相似文献
