Summary Bicarbonate presence in the bathing media doubles Na+ and fluid transepithelial transport and in parallel significantly increases Na+ and Cl intracellular concentrations and contents, decreases K+ cell concentration without changing its amount, and causes a large cell swelling. Na+ and Cl lumen-to-cell influxes are significantly enhanced, Na+ more so than Cl. The stimulation does not raise any immediate change in luminal membrane potential and cannot be due to a HCO 3 -ATPase in the brush border. The stimulation goes together with a large increase in a Na+-dependent H+ secretion into the lumen. All of these data suggests that HCO 3 both activates Na+–Cl cotransport and H+–Na+ countertransport at the luminal barrier.Thiocyanate inhibits Na+ and fluid transepithelial transport without affecting H+ secretion and HCO 3 -dependent Na+ influx. It reduces Na+ and Cl concentrations and contents, increases the same parameters for K+, causes a cell shrinking, and abolishes the lumen-to-cell Cl influx. It enters the cell and is accumulated in the cytoplasm with a process which is Na+-dependent and HCO 3 -activated. Thus, SCN is likely to compete for the Cl site on the cotransport carrier and to be slowly transferred by the cotransport system itself.  相似文献

Summary The Na+ requirement for active, electrogenic Cl absorption byAmphiuma small intestine was studied by tracer techniques and double-barreled Cl-sensitive microelectrodes. Addition of Cl to a Cl-free medium bathingin vitro intestinal segments produced a saturable (K m =5.4mm) increase in shortcircuit current (I sc) which was inhibitable by 1mm SITS. The selectivity sequence for the anion-evoked current was Cl=Br>SCN>NO 3 >F=I. Current evoked by Cl reached a maximum with increasing medium Na concentration (K m =12.4mm). Addition of Na+, as Na gluconate (10mm), to mucosal and serosal Na+-free media stimulated the Cl current and simultaneously increased the absorptive Cl flux (J ms Cl ) and net flux (J net Cl ) without changing the secretory Cl flux (J sm Cl ). Addition of Na+ only to the serosal fluid stimulatedJ ms Cl much more than Na+ addition only to the mucosal fluid in paired tissues. Serosal DIDS (1mm) blocked the stimulation. Serosal 10mm Tris gluconate or choline gluconate failed to stimulateJ ms Cl . Intracellular Cl activity (a Cl i ) in villus epithelial cells was above electrochemical equilibrium indicating active Cl uptake. Ouabain (1mm) eliminated Cl accumulation and reduced the mucosal membrane potential m over 2 to 3 hr. In contrast, SITS had no effect on Cl accumulation and hyperpolarized the mucosal membrane. Replacement of serosal Na+ with choline eliminated Cl accumulation while replacement of mucosal Na+ had no effect. In conclusion by two independent methods active electrogenic Cl absorption depends on serosal rather than mucosal Na+. It is concluded that Cl enters the cell via a primary (rheogenic) transport mechanism. At the serosal membrane the Na+ gradient most likely energizes H+ export and regulates mucosal Cl accumulation perhaps by influencing cell pH or HCO 3 concentration.  相似文献

Summary Gluconate substitution for serosal Cl reduces the transepithelial short-circuit current (I sc) and depolarizes shortcircuited frog skins. These effects could result either from inhibition of basolateral K+ conductance, or from two actions to inhibit both apical Na+ permeability (P Na ap ) and basolateral pump activity. We have addressed this question by studying whole-and split-thickness frog skins. Intracellular Na+ concentration (C Na c ) andP Na ap have been monitored by measuring the currentvoltage relationship for apical Na+ entry. This analysis was conducted by applying trains of voltage pulses, with pulse durations of 16 to 32 msec. Estimates ofP Na ap ) and CNa/c were not detectably dependent on pulse duration over the range 16 to 80 msec. Serosal Cl replacement uniformly depolarized short-circuited tissues. The depolarization was associated with inhibition ofI sc across each split skin, but only occasionally across the whole-thickness preparations. This difference may reflect the better ionic exchange between the bulk medium and the extracellular fluid in contact with the basolateral membranes, following removal of the underlying dermis in the split-skin preparations.P Na ap was either unchanged or increased, and CNa/c either unchanged or reduced after the anionic replacement. These data are incompatible with the concept that serosal Cl replacement inhibitsP Na ap and Na, K-pump activity. Gluconate substutition likely reduces cell volume, triggering inhibition of the basolateral K+ channels, consistent with the data and conclusions of S.A. Lewis, A.G. Butt, M.J. Bowler, J.P. Leader and A.D.C Macknight (J. Membrane Biol. 83:119–137, 1985) for toad bladder. The resulting depolarization reduces the electrical force favoring apical Na+ entry. The volume-conductance coupling serves to conserve volume by reducing K+ solute loss. Its molecular basis remains to be identified.  相似文献

This study investigated the action of enprostil, a synthetic analog of PGE2, on gastric HCO3 secretion in humans and on duodenal HCO3 secretion in the anesthetized rat. A previously validated 2-component model was used to calculate gastric HCO3 and H+ secretion in 10 human subjects. Compared to placebo, a single 70 μg oral dose of enprostil increased basal gastric HCO3 secretion from 1810 +- 340 to 3190 ± 890 μmol/hr (P < 0.05). In addition, enprostil reduced basal gastric H+ secretion from 5240 ± 1140 to 1680 ± 530 μmol/hr (P < 0.02). Enprostil also increased HCO3 and reduced H+ secretion during intravenous pentagastrin infusion. In the rat, duodenal HCO3 secretion was measured by direct titration in situ using perfused segments of duodenum just distal to the Brunner gland area dn devoid of pancreatic and biliary secretions. Addition of enprostil(10 μg/ml) to the duodenal bathing solution increased duodenal HOC3 secretion from 6.3 ± 1.3 to 15.1 ± 2.0 μmol/cm·hr (P < 0.01, n = 6). The stimulatory action of enprostil on duodenal HCO3 secretion at 10 μg/ml was comparable in magnitude and duration to that of 10 μg/ml natural PGE2. In summary, the PGE2 analog enprostil stimulated gastroduodenal HCO3 secretion, effects which may be beneficial in protection of the gastroduodenal mucosa against luminal acid.  相似文献

Summary The characteristics of Cl movement across luminal and basolateral membranes ofAmphiuma intestinal absorptive cells were studied using Cl-sensitive microelectrodes and tracer36Cl techniques. Intracellular Cl activity (a Cl i ) was unchanged when serosal Cl was replaced; when luminal Cl was replaced cell Cl was rapidly lost. Accordingly, the steady statea Cl i could be varied by changing the luminal [Cl]. As luminal [Cl] was raised from 1 to 86mM,a Cl i rose in a linear manner, the mucosal membrane hyperpolarized, and the transepithelial voltage became serosa negative. In contrast, the rate of Cl transport from the cell into the serosal medium, measured as the SITS-inhibitable portion of the Cl absorptive flux, attained a maximum whena Cl i reached an apparent value of 17mm, indicating the presence of a saturable, serosal transport step. The stilbeneinsensitive absorptive flux was linear with luminal [Cl], suggestive of a paracellular route of movement. Intracellulara Cl was near electrochemical equilibrium at all but the lowest values of luminal [Cl] after interference produced by other anions was taken into account.a Cl i was unaffected by Na replacement, removal of medium K, or elevation of medium HCO 3 . Mucosae labeled with36Cl lost isotope into both luminal and serosal media at the same rate and from compartments of equal capacity. Lowering luminal [Cl] or addition of theophylline enhanced luminal Cl efflux. It is concluded that a conductive Cl leak pathway is present in the luminal membrane. Serosal transfer is by a saturable, stilbene-inhibitable pathway. Luminal Cl entry appears to be passive, but an electrogenic uptake cannot be discounted.  相似文献

Our previous studies with a line of Madin-Darby canine kidney (MDCK) cells (FL-MDCK) transfected with FLAG-labeled alpha, beta, and gamma subunits of epithelial Na(+) channel (ENaC) showed that, although most of the short-circuit current (I (sc)) was amiloride sensitive (AS-I (sc)), there was also an amiloride-insensitive component (NS-I (sc)) due to Cl(-) secretion (Morris and Schafer, J Gen Physiol 120:71-85, 2002). In the present studies, we observed a progressive increase in NS-I (sc) and a corresponding decrease in AS-I (sc) during experiments. There was a significant negative correlation between AS-I (sc) and NS-I (sc) both in the presence and absence of treatment with cyclic adenosine monophosphate (cAMP). NS-I (sc) could be attributed to both cystic fibrosis transmembrane conductance regulator (CFTR) and a 4, 4'-diisothiocyano-2, 2'-disulfonic acid stilbene (DIDS)-sensitive Ca(2+)-activated Cl(-) channel (CaCC). Continuous perfusion of both sides of the Ussing chamber with fresh rather than recirculated bathing solutions, or addition of hexokinase (6 U/ml), prevented the time-dependent changes and increased AS-I (sc) by 40-60%, with a proportional decrease in NS-I (sc). Addition of 100 muM adenosine triphosphate (ATP) in the presence of luminal amiloride produced a transient four-fold increase in NS-I (sc) that was followed by a sustained increase of 50-60% above the basal level. ATP release from the monolayers, measured by bioluminescence, was found to occur across the apical but not the basolateral membrane, and the apical release was tripled by cAMP treatment. These data show that constitutive apical ATP release, which occurs under both basal and cAMP-stimulated conditions, underlies the time-dependent rise in Cl(-) secretion and the proportional fall in ENaC-mediated Na(+) absorption in FL-MDCK cells. Thus, endogenous ATP release can introduce a significant confounding variable in experiments with this and similar epithelial cells, and it may underlie at least some of the observed interaction between Cl(-) secretion and Na(+) absorption.  相似文献

The calcium ionophore A23187 stimulates luminal alkalinization and inhibits Cl absorption in short-circuited urinary bladders of postprandial or alkalotic turtles. The ionophore appears to mimic the action of the phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine (IBMX) by its similar effects on HCO3 secretion and Cl absorption and by increasing cytosolic cAMP levels of isolated bladder epithelial cells. However, only A23187 (or ionomycin), but not IMBX or cAMP, elevated cytosolic Ca2+ of aequorin- or quin2-loaded cells. Since A23187, but not IBMX or cAMP inhibits luminal acidification, we postulate that cytosolic Ca2+ (1) regulates the acidification process by a cAMP-independent mechanism and (2) controls HCO3 secretion as well as Cl absorption, at least in part, via cAMP-mediated pathways.  相似文献

This study examines the effects of acute in vitro acid-base disorders on Na+/H+ and H+-ATPase transporters in rabbit kidney proximal tubules (PT). PT suspensions were incubated in solutions with varying acid base conditions for 45 min and utilized for brush border membrane (BBM) vesicles preparation. BBM vesicles were studied for Na+/H+ exchange activity (assayed by 22Na+ influx) or abundance (using NHE-3 specific antibody) and H+-ATPase transporter abundance (using antibody against the 31 kDa subunit). The Na+/ H+ exchanger activity increased by 55% in metabolic acidosis (pH 6.5, HCO 3 3 mm) and decreased by 41% in metabolic alkalosis (pH 8.0, HCO 3 90 mm). The abundance of NHE-3 remained constant in acidic, control, and alkalotic groups. H+-ATPase abundance, however, decreased in metabolic acidosis and increased in metabolic alkalosis by 57% and 42%, respectively. In PT suspensions incubated in isohydric conditions (pH 7.4), Na+/H+ exchanger activity increased by 29% in high HCO 3 group (HCO 3 96 mm) and decreased by 16% in the low HCO 3 groups (HCO 3 7mm. The NHE-3 abundance remained constant in high, normal, and low [HCO 3 ] tubules. The abundance of H+-ATPase, however, increased by 82% in high [HCO 3 ] and decreased by 77% in the low [HCO 3 ] tubules. In PT suspensions incubated in varying pCO2 and constant [HCO 3 ], Na+/H+ exchanger activity increased by 35% in high pCO2 (20% pCO2, respiratory acidosis) and decreased by 32% in low pCO2 (1.5% pCO2, respiratory alkalosis) tubules. The NHE-3 abundance remained unchanged in high, normal, and low pCO2 tubules. However, the H+-ATPase abundance increased by 74% in high pCO2 and decreased by 69% in low pCO2 tubules.The results of these studies suggest that the luminal Na+/H+ exchanger is predominantly regulated by pH whereas H+-ATPase is mainly regulated by [HCO 3 ] and/ or pCO2. They further suggest that the adaptive changes in H+-ATPase transporter are likely mediated via endocytic/exocytic pathway whereas the adaptive changes in Na+/H+ exchanger are via the nonendocytic/exocytic pathway.The excellent technical assistance of Yollanda J. Transepithelial fluid transport was measured gravimetrically in rabbit gallbladder (and net Na+ transport was calculated from it), at 27 degrees C, in HCO(3-)-free bathing media containing 10(-4) M acetazolamide. Whereas luminal 10(-4) M bumetanide or 10(-4) M 4-acetamido-4'-iso-thiocyanostilbene-2,2'-disulfonate (SITS) did not affect fluid absorption, 25 mM SCN- abolished it; hydrochlorothiazide (HCTZ) in the luminal medium reduced fluid absorption from 28.3 +/- 1.6 (n = 21) to 8.6 +/- 1.6 microliters cm-2 hr-1 (n = 10), i.e., to about 30%. This maximum effect was already obtained at 10(-3) M concentration; the apparent IC50 was about 2 x 10(-4) M. The residual fluid absorption, again insensitive to SITS, was completely inhibited by SCN- or bumetanide. Cl- influx at the luminal border of the epithelium, measured under the same conditions and corrected for the extracellular space and paracellular influx, proved insensitive to 10(-4) M bumetanide, but was slowly inhibited by 10(-3) M HCTZ, with maximum inhibition (about 54%) reached after a 10-min treatment; it subsequently rose again, in spite of the presence of HCTZ. However, if the epithelium, treated with HCTZ, was exposed to 10(-4) M bumetanide during the measuring time (45 sec), inhibition was completed and the subsequent rise of Cl- influx eliminated. Intracellular Cl- accumulation with respect to the predicted activity value at equilibrium decreased significantly upon exposure to 10(-3) M HCTZ, reached a minimum within 15-30 min of treatment, then rose again significantly at 60 min. Simultaneous exposure to HCTZ and bumetanide decreased the accumulation to a significantly larger extent as compared to HCTZ alone, already in 15 min, and impeded the subsequent rise. Intracellular K+ activity rose significantly within 30 min treatment with HCTZ; the increase proved bumetanide dependent. The results obtained show that Na(+)-Cl- symport, previously detected under control conditions, is the HCTZ-sensitive type; its inhibition elicits bumetanide-sensitive Na(+)-K(+)-2Cl- cotransport. Thus, the three forms of neutral Na(+)-Cl(-)-coupled transport so far evidenced in epithelia, Na+/H+, Cl-/HCO3- double exchange (in the presence of exogenous bicarbonate), HCTZ-sensitive Na(+)-Cl- symport and bumetanide-sensitive Na(+)-K(+)-2Cl- cotransport, are all present in the apical membrane of rabbit gallbladder.  相似文献

The proximal tubule Na+-HCO 3 cotransporter is located in the basolateral plasma membrane and moves Na+, HCO 3, and net negative charge together out of the cell. The presence of charge transport implies that at least two HCO 3 anions are transported for each Na+ cation. The actual ratio is of physiological interest because it determines direction of net transport at a given membrane potential. To determine this ratio, a thermodynamic approach was employed that depends on measuring charge flux through the cotransporter under defined ion and electrical gradients across the basolateral plasma membrane. Cells from an immortalized rat proximal tubule line were grown as confluent monolayer on porous substrate and their luminal plasma membrane was permeabilized with amphotericin B. The electrical properties of these monolayers were measured in a Ussing chamber, and ion flux through the cotransporter was achieved by applying Na+ or HCO 3 concentration gradients across the basolateral plasma membrane. Charge flux through the cotransporter was identified as difference current due to the reversible inhibitor dinitro-stilbene disulfonate. The cotransporter activity was Cl independent; its conductance ranged between 0.12 and 0.23 mS/cm2 and was voltage independent between −60 and +40 mV. Reversal potentials obtained from current-voltage relations in the presence of Na+ gradients were fitted to the thermodynamic equivalent of the Nernst equation for coupled ion transport. Summary Experiments were performed usingin vitro perfused medullary thick ascending limbs of Henle (MTAL) and in suspensions of MTAL tubules isolated from mouse kidney to evaluate the effects of arginine vasopressin (AVP) on the K+ dependence of the apical, furosemide-sensitive Na+:Cl cotransporter and on transport-related oxygen consumption (QO2). In isolated perfused MTAL segments, the rate of cell swelling induced by removing K+ from, and adding onemm ouabain to, the basolateral solution [ouabain(zero-K+)] provided an index to apical cotransporter activity and was used to evaluated the ionic requirements of the apical cotransporter in the presence and absence of AVP. In the absence of AVP cotransporter activity required Na+ and Cl, but not K+, while in the presence of AVP the apical cotransporter required all three ions.86Rb+ uptake into MTAL tubules in suspension was significant only after exposure of tubules to AVP. Moreover,22Na+ uptake was unaffected by extracellular K+ in the absence of AVP while after AVP exposure22Na+ uptake was strictly K+-dependent. The AVP-induced coupling of K+ to the Na+:Cl cotransporter resulted in a doubling in the rate of NaCl absorption without a parallel increase in the rate of cellular22Na+ uptake or transport-related oxygen consumption. These results indicate that arginine vasopressin alters the mode of a loop diuretic-sensitive transporter from Na+:Cl cotransport to Na+:K+:2Cl cotransport in the mouse MTAL with the latter providing a distinct metabolic advantage for sodium transport. A model for AVP action on NaCl absorption by the MTAL is presented and the physiological significance of the coupling of K+ to the apical Na+:Cl cotransporter in the MTAL and of the enhanced metabolic efficiency are discussed.  相似文献

Summary Cl influx at the luminal border of the epithelium of rabbit gallbladder was measured by 45-sec exposures to36Cl and3H-sucrose (as extracellular marker). Its paracellular component was evaluated by the use of 25mm SCN which immediately and completely inhibits Cl entry into the cell. Cellular influx was equal to 16.7eq cm–2 hr–1 and decreased to 8.5eq cm–2 hr–1 upon removal of HCO 3 from the bathing media and by bubbling 100% O2 for 45 min. When HCO 3 was present, cellular influx was again about halved by the action of 10–4 m acetazolamide, 10–5 to 10–4 m furosemide, 10–5 to 10–4 m 4-acetamido-4-isothiocyanostilbene-2,2-disulfonate (SITS), 10–3 m amiloride. The effects of furosemide and SITS were tested at different concentrations of the inhibitor and with different exposure times: they were maximal at the concentrations reported above and nonadditive. In turn, the effects of amiloride and SITS were not additive. Acetazolamide reached its maximal action after an exposure of about 2 min. When exogenous HCO 3 was absent, the residual cellular influx was insensitive to acetazolamide, furosemide and SITS. When exogenous HCO 3 was present in the salines, Na+ removal from the mucosal side caused a slow decline of cellular Cl influx; conversely, it immediately abolished cellular Cl influx in the absence of HCO 3 . In conclusion, about 50% of cellular influx is sensitive to HCO 3 , inhibitable by SCN, acetazolamide, furosemide, SITS and amiloride and furthermore slowly dependent on Na+. The residual cellular influx is insensitive to bicarbonate, inhibitable by SCN, resistant to acetazolamide, furosemide, SITS and amiloride, and immediately dependent on Na+. Thus, about 50% of apical membrane NaCl influx appears to result from a Na+/H+ and Cl/HCO 3 exchange, whereas the residual influx seems to be due to Na+–Cl contranport on a single carrier. Whether both components are simultaneously present or the latter represents a cellular homeostatic counterreaction to the inhibition of the former is not clear.  相似文献

Summary Active HCO 3 t- secretion in the anterior rectal salt gland of the mosquito larva,Aedes dorsalis, is mediated by a 11 Cl/HCO 3 exchanger. The cellular mechanisms of HCO 3 and Cl transport are examined using ion- and voltage-sensitive microelectrodes in conjunction with a microperfused preparation which allowed rapid saline changes. Addition of DIDS or acetazolamide to, or removal of CO2 and HCO 3 from, the serosal bath caused large (20 to 50 mV) hyperpolarizations of apical membrane potential (V a) and had little effect on basolateral potential (V bl). Changes in luminal Cl concentration alteredV a in a repid, linear manner with a slope of 42.2 mV/decaloga Cl l –. Intracellular Cl activity was 23.5mm and was approximately 10mm lower than that predicted for a passive distribution across the apical membrane. Changes in serosal Cl concentration had no effect onV bl, indicating an electrically silent basolateral Cl exit step. Intracellular pH in anterior rectal cells was 7.67 and the calculated was 14.4mm. These results show that under control conditions HCO3 enters the anterior rectal cell by an active mechanism against an electrochemical gradient of 77.1 mV and exits the cell at the apical membrane down a favorable electrochemical gradient of 27.6 mV. A tentative cellular model is proposed in which Cl enters the apical membrane of the anterior rectal cells by passive, electrodiffusive movement through a Cl-selective channel, and HCO 3 exits the cell by an active or passive electrogenic transport mechanism. The electrically silent nature of basolateral Cl exit and HCO3 entry, and the effects of serosal addition of the Cl/HCO3 exchange inhibitor, DIDS, on and transepithelial potential (V ic) suggest strongly that the basolateral membrane is the site of a direct coupling between Cl and HCO 3 movements.  相似文献

Na+, Li+ and Cl− transport by brush border membranes from rabbit jejunum   总被引:1,自引:0,他引:1  
Na+, Li+, K+, Rb+, Br-, Cl- and SO4(2-) transport were studied in brush border membrane vesicles isolated from rabbit jejunum. Li+ uptakes were measured by flameless atomic absorption spectroscopy, and all others were measured using isotopic flux and liquid scintillation counting. All uptakes were performed with a rapid filtration procedure. A method is presented for separating various components of ion uptake: 1) passive diffusion, 2) mediated transport and 3) binding. It was concluded that a Na+/H+ exchange mechanism exists in the jejunal brush border. The exchanger was inhibited with 300 microM amiloride or harmaline. The kinetic parameters for sodium transport by this mechanism depend on the pH of the intravesicular solution. The application of a pH gradient (pHin = 5.5, pHout = 7.5) causes an increase in Jmax (50 to 125 pmol/mg protein . sec) with no change in Kt (congruent to 4.5 nM). Competition experiments show that other monovalent cations, e.g. Li+ and NH4+, share the Na+/H+ exchanger. This was confirmed with direct measurements of Li+ uptakes. Saturable uptake mechanisms were also observed for K+, Rb+ and SO4(2-), but not for Br-. The Jmax for K+ and Rb+ are similar to the Jmax for Na+, suggesting that they may share a transporter. The SO4(2-) system appears to be a Na+/SO4(2-) cotransport system. There does not appear to be either a Cl-/OH- transport mechanism of the type observed in ileum or a specific Na+/Cl- symporter.  相似文献

Isolated segments of jejunum from Amphiuma absorb HCO3? electrogenically by a process sensitive to acetazolamide. Using a tissue chamber of special design which permits isolation of villus or intervillus epithelium the transepithelial electrical potential (ψms) of each region was measured. The serosa-negative ψms generated by the villus epithelium was more negative than that of the intervillus and exhibited greater sensitivity to acetazolamide. Both regions were responsive to other agents which alter epithelial ion transport. The results indicate that intestinal HCO3? absorption is localized predominantly within cells lining the villus epithelium.  相似文献

Summary Recent studies in hepatocytes indicate that Na+-coupled HCO 3 transport contributes importantly, to regulation of intracellular pH and membrane HCO 3 transport. However, the direction of net coupled Na+ and HCO 3 movement and the effect of HCO 3 on Na+ turnover and Na+/K+ pump activity are not known. In these studies, the effect of HCO 3 on Na+ influx and turnover were measured in primary rat hepatocyte cultures with22Na+, and [Na+] i was measured in single hepatocytes using the Na+-sensitive fluorochrome SBFI. Na+/K+ pump activity was measured in intact perfused rat liver and hepatocyte monolayers as Na+-dependent or ouabain-suppressible86Rb uptake, and was measured in single hepatocytes as the effect of transient pump inhibition by removal of extracellular K+ on membrane potential difference (PD) and [Na+] i . In hepatocyte monolayers, HCO 3 increased22Na+ entry and turnover rates by 50–65%, without measurably altering22Na+ pool size or cell volume, and HCO 3 also increased Na+/K+ pump activity by 70%. In single cells, exposure to HCO 3 produced an abrupt and sustained rise in [Na+] i , from 8 to 12mm. Na+/K+ pump activity assessed in single cells by PD excursions during transient K+ removal increased 2.5-fold in the presence of HCO 3 , and the rise in [Na+] i produced by inhibition of the Na+/K+ pump was similarly increased 2.5-fold in the presence of HCO 3 . In intact perfused rat liver, HCO 3 increased both Na+/K+ pump activity and O2 consumption. These findings indicate that, in hepatocytes, net coupled Na+ and HCO 3 movement is inward and represents a major determinant of Na+ influx and Na+/K+ pump activity. About half of hepatic Na+/K+ pump activity appears dedicated to recycling Na+ entering in conjunction with HCO 3 to maintain [Na+] i within the physiologic range.  相似文献

Summary We investigated intracellular pH (pH i ) regulation in cultured human ciliary muscle cells by means of the pH-sensitive absorbance of 5(and 6)-carboxy-4,5-dimethylfluorescein (CDMF). The steady-state pH i was 7.09±0.04 (n = 12) in CO2/ HCO 3 -buffered and 6.86±0.03 (n = 12) in HEPES-buffered solution. Removal of extracellular sodium for 6 min acidified the cells by 1.11±0.06 pH units (n = 12) in the presence of CO2/ HCO 3 and by 0.91±0.05 pH units (n = 8) in its absence. Readdition of external sodium resulted in a rapid pH i recovery, which was almost completely amiloride-sensitive in the absence of CO2/ HCO 3 but only slightly influenced by amiloride in its presence. Application of DIDS under steady-state conditions significantly acidified the ciliary muscle cells by 0.25±0.02 (n = 4) in 6 min, while amiloride had no effect. The pH i recovery after an intracellular acid load was completely dependent on extracellular sodium. In HEPES-buffered solution the pH i recovery was almost completely mediated by Na+/H+ exchange, since it was blocked by amiloride (1 mmol/liter). In contrast, a marked amilorideinsensitive pH i recovery was observed in CO2/HCO 3 -buffered solution which was mediated by chloride-independent and chloride-dependent Na+ HCO 3 cotransport. This recovery, inhibited by DIDS (0.2 mmol/liter). was also observed if the cells were preincubated in chloride-free solution for 4 hr. Analysis of the sodium dependence of the pH i recovery after NH4Cl prepulse revealed V max = 0.57 pH units/min, K m= 39.7 mmol/liter extracellular sodium for the amiloride-sensitive component and V max = 0.19 pH units/min, K m= 14.3 mmol/liter extracellular sodium for the arniloride-insensitive component. We conclude that Na+/H+ exchange and chloride-independent and chloride-dependent Na+HCO 3 cotransport are involved in the pH i regulation of cultured human ciliary muscle cells.The expert technical assistance of Astrid Krolik is gratefully acknowledged. The effects of changes in secretory concentrations of K+, Na+ and HCO3 on transmucosal potential difference (PD) and resistance in Cl-free (SO42−) solutions were compared for secreting fundus and resting fundus of Rana pipiens. In the resting fundus experiments, histamine was not present in the nutrient solution and cimetidine was primarily used to obtain acid inhibition. Increase of K+ from 4 to 80 mM, decrease of Na+ from 156 to 15.6 mM and decrease of HCO3 from 25 to 5 mM gave, 10 min after the change, in the secreting fundus Δ PD values of 39.7, −11.9 and 3.2 mV, respectively. In the resting fundus, 1.5 to 2 h after the addition of cimetidine, the same changes in secretory ion concentration gave Δ PD values of 12.2, −5.6 and 1.5 mV, respectively. Replacement of cimetidine with SCN and without histamine yielded a Δ PD somewhat lower than that in cimetidine, namely 9 mV for a K+ change from 4 to 80 mM. Subsequent addition of histamine with SCN present gave a Δ PD of about 21 mV. The change in PD was attributed to histamine increasing the secretory membrane area, leading to an increase in K+ conductance. Another possibility is that histamine increases the K+ conductance per se.  相似文献

Our previously published whole-cell patch-clamp studies on the cells of the intralobular (granular) ducts of the mandibular glands of male mice revealed the presence of an amiloride-sensitive Na+ conductance in the plasma membrane. In this study we demonstrate the presence also of a Cl conductance and we show that the sizes of both conductances vary with the Cl concentration of the fluid bathing the cytosolic surface of the plasma membrane. As the cytosolic Cl concentration rises from 5 to 150 mmol/liter, the size of the inward Na+ current declines, the decline being half-maximal when the Cl concentration is approximately 50 mmol/liter. In contrast, as cytosolic Cl concentration increases, the inward Cl current remains at a constant low level until the Cl concentration exceeds 80 mmol/liter, when it begins to increase. Studies in which Cl in the pipette solution was replaced by other anions indicate that the Na+ current is suppressed by intracellular Br-, Cl and NO 3 - but not by intracellular I-, glutamate or gluconate. Our studies also show that the Cl conductance allows passage of Cl and Br- equally well, I-less well, and NO 3 - , glutamate and gluconate poorly, if at all. The findings with NO 3 - are of particular interest because they show that suppression of the Na+ current by a high intracellular concentration of a particular anion does not depend on actual passage of that anion through the Cl conductance. In mouse granular duct cells there is, thus, a reciprocal regulation of Na+ and Cl conductances by the cytosolic Cl concentration. Since the cytosolic Cl concentration is closely correlated with cell volume in many epithelia, this reciprocal regulation of Na+ and Cl conductances may provide a mechanism by which ductal Na+ and Cl transport rates are adjusted so as to maintain a stable cell volume.This project was supported by the National Health and Medical Research Council of Australia.
