Four proline-derived chiral receptors 5-8 were readily synthesized starting from L-proline. The enantiomeric recognition ability of chiral receptors was examined with a series of carboxylic acids by (1) H NMR spectroscopy. The molar ratio and the association constants of the chiral compounds with each of the enantiomers of guest molecules were determined by using Job plots and a nonlinear least-squares fitting method, respectively. The Job plots indicate that the hosts form 1:1 instantaneous complexes with all guests. The receptors exhibited different chiral recognition abilities toward the enantiomers of racemic guests. Among the chiral receptors used in this study, prolinamide 6 was found to be the best chiral shift reagent and is effective for the determination of the enantiomeric excess of chiral carboxylic acids.  相似文献

The resolution of seven enantiomeric pairs of chiral derivatives of xanthones (CDXs) on (S,S)‐Whelk‐O1 and l ‐phenylglycine chiral stationary phases (CSPs) was systematically investigated using multimodal elution conditions (normal‐phase, polar‐organic, and reversed‐phase). The (S,S)‐Whelk‐O1 CSP, under polar‐organic conditions, demonstrated a very good power of resolution for the CDXs possessing an aromatic moiety linked to the stereogenic center with separation factor and resolution factor ranging from 1.91 to 7.55 and from 6.71 to 24.16, respectively. The chiral recognition mechanisms were also investigated for (S,S)‐Whelk‐O1 CSP by molecular docking technique. Data regarding the CSP–CDX molecular conformations and interactions were retrieved. These results were in accordance with the experimental chromatographic parameters regarding enantioselectivity and enantiomer elution order. The results of the present study fulfilled the initial objectives of enantioselective studies of CDXs and elucidation of intermolecular CSP–CDX interactions. Four diastereomeric chiral stationary phases (CSPs) based on quinine, quinidine, epiquinine, and epiquinidine tert‐butyl carbamate selectors were synthesized and evaluated under ion exchange HPLC conditions with a set of racemic N‐acylated and N‐oxycarbonylated α‐amino acids as selectands. The enantioseparation potential of quinine‐ and quinidine‐derived CSPs proved to be far superior to that of their C9‐epimeric congeners. The absolute configuration of C9 stereogenic center of the cinchonan backbone of these selectors was identified as the structural feature controlling the elution order. Guided by an X‐ray structure of a most favorable selector–selectand complex and the observed chromatographic enantioseparation data, a chiral recognition model was advanced. The enantioresolution and determination of the enantiomeric purity of 32 new xanthone derivatives, synthesized in enantiomerically pure form, were investigated on (S ,S )‐Whelk‐O1 chiral stationary phase (CSP). Enantioselectivity and resolution (α and RS) with values ranging from 1.41–6.25 and from 1.29–17.20, respectively, were achieved. The elution was in polar organic mode with acetonitrile/methanol (50:50 v/v ) as mobile phase and, generally, the (R )‐enantiomer was the first to elute. The enantiomeric excess (ee ) for all synthesized xanthone derivatives was higher than 99%. All the enantiomeric pairs were enantioseparated, even those without an aromatic moiety linked to the stereogenic center. Computational studies for molecular docking were carried out to perform a qualitative analysis of the enantioresolution and to explore the chiral recognition mechanisms. The in silico results were consistent with the chromatographic parameters and elution orders. The interactions between the CSP and the xanthone derivatives involved in the chromatographic enantioseparation were elucidated.  相似文献

A new 2,5‐diphenyl‐1,3,4‐oxadiazole‐based derivative (L) was synthesized and applied as a highly selective and sensitive fluorescent sensor for relay recognition of Cu2+ and S2? in water (Tris–HCl 10 mM, pH = 7.0) solution. L exhibits an excellent selectivity to Cu2+ over other examined metal ions with a prominent fluorescence ‘turn‐off’ at 392 nm. L interacts with Cu2+ through a 1:2 binding stoichiometry with a detection limit of 4.8 × 10–7 M. The on‐site formed L–2Cu2+ complex exhibits excellent selectivity to S2? with a fluorescence ‘off–on’ response via a Cu2+ displacement approach. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

A series of novel fluoroionophore bearing derivatives of benzo‐15‐crown‐5 were synthesized by the amination of benzo‐15‐crown‐5 followed by condensation with different quinones in the presence of titanium tetrachloride (TiCl4) and 1,4‐diazabicyclo‐[2.2.2]octane. The compounds were characterized by infrared, 1H and 13C nuclear magnetic resonance, mass spectroscopy and elemental analysis. Absorption and fluorescence spectral characteristics of these compounds were studied. It was observed that the anthraquinone derivative was acting as an Hg2+ ion sensor. This article reports a new chiral separation method-biphasic recognition chiral extraction for the separation of mandelic acid enantiomers. Distribution behavior of mandelic acid enantiomers was studied in the extraction system with O,O'-di-benzoyl-(2S,3S)-4-toluoyl-tartaric acid (D-(+)-DTTA) in organic phase and beta-CD derivatives in aqueous phase, and the influence of the types and concentrations of extractants and pH on extraction efficiency was investigated. Hydroxypropyl-beta-cyclodextrin (HP-beta-CD), hydroxyethyl-beta-cyclodextrin (HE-beta-CD), and methyl-beta-cyclodextrin (Me-beta-CD) have stronger recognition abilities for S-mandelic acid than those for R-mandelic acid, among which HP-beta-CD has the strongest ability. D-(+)-DTTA preferentially recognizes R-mandelic acid. pH and the concentrations of extractants have great effects on chiral separation ability. A high enantioseparation efficiency with a maximum enantioselectivity of 1.527 is obtained at pH of 2.7 and the ratio of 2:1 of [D-(+)-DTTA] to [HP-beta-CD]. The obtained results indicate that the biphasic recognition chiral extraction is of stronger chiral separation ability than the monophasic recognition chiral extraction. It may be very helpful to optimize the extraction systems and realize the large-scale production of pure enantiomers.  相似文献

A novel semi‐rigid latent chromophore E1, containing an amide subunit activated by an adjacent semi‐rigid intramolecular hydrogen‐bonding (IHB) unit, was designed for the detection of fluoride ion by the ‘naked‐eye’ in CH3CN. Comparative studies on structural analogs (E2, E3, and E4) provided significant insight into the structural and functional role of the amide N–H and IHB segment in the selective recognition of fluoride ions. The deprotonation of the amide N–H followed by the enhancement of intramolecular charge transfer (ICT) induced the colorimetric detection of E1 for fluoride ion. Chiral monoaza-15-crown-5 ethers (1, 2) were prepared from (R)-(-)-2-amino-1-butanol in high yield. The chiral monoaza-15-crown-5 ethers were purified directly as NaClO(4) complexes. Molecular recognition by these chiral monoaza-crown ethers of (R)- and (S)-PhEtHClO(4) and (R)- and (S)-NapEtHClO(4) as characterized by UV-vis spectroscopy. The order of enantiomeric selectivity is (R)- > (S)- PhEtHClO(4) and (S)- > (R)-NapEtHClO(4) for 1. In the case of 2 it was (R)- > (S)-PhEtHClO(4) and (R)- > (S)- NapEtHClO(4). The cavity of macrocycle and steric hindrance of the benzene units appears to play an important role in recognition.  相似文献

Two novel Rhodamine–pyrazolone‐based colorimetric off–on fluorescent chemosensors for Fe3+ ions were designed and synthesized using pyrazolone as the recognition moiety and Rhodamine 6G as the signalling moiety. The photophysical properties and Fe3+‐binding properties of sensors L1 and L2 in acetonitrile–aqueous solution were also investigated. Both sensors successfully exhibit a remarkably ‘turn‐on’ response, toward Fe3+, which was attributed to 1: 2 complex formation between Fe3+ and L1/L2. The fluorescent and colorimetric response to Fe3+ can be detected by the naked eye, which provides a facile method for the visual detection of Fe3+. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

The drug chirality is attracting increasing attention because of different biological activities, metabolic pathways, and toxicities of chiral enantiomers. The chiral separation has been a great challenge. Optimized high‐performance liquid chromatography (HPLC) methods based on vancomycin chiral stationary phase (CSP) were developed for the enantioseparation of propranolol, atenolol, metoprolol, venlafaxine, fluoxetine, and amlodipine. The retention and enantioseparation properties of these analytes were investigated in the variety of mobile phase additives, flow rate, and column temperature. As a result, the optimal chromatographic condition was achieved using methanol as a main mobile phase with triethylamine (TEA) and glacial acetic acid (HOAc) added as modifiers in a volume ratio of 0.01% at a flow rate of 0.3 mL/minute and at a column temperature of 5°C. The thermodynamic parameters (eg, ΔH, ΔΔH, and ΔΔS) from linear van 't Hoff plots revealed that the retention of investigated pharmaceuticals on vancomycin CSP was an exothermic process. The nonlinear behavior of lnk′ against 1/T for propranolol, atenolol, and metoprolol suggested the presence of multiple binding mechanisms for these analytes on CSP with variation of temperature. The simulated interaction processes between vancomycin and pharmaceutical enantiomers using molecular docking technique and binding energy calculations indicated that the calculated magnitudes of steady combination energy (ΔG) coincided with experimental elution order for most of these enantiomers.  相似文献

As a new acidic selector (resolving agent), we synthesized an enantiopure O‐alkyl phenylphosphonothioic acid with a seven‐membered ring ((R)‐ 5 ), which was designed on the basis of the results for the enantioseparation of 1‐arylethylamine derivatives with acyclic O‐ethyl phenylphosphonothioic acid ( I ). The phosphonothioic acid (R)‐ 5 showed unique chirality‐recognition ability in the enantioseparation of 1‐naphthylethylamine derivatives, aliphatic secondary amines, and amino alcohols; the ability was complementary to that of I . The X‐ray crystallographic analyses of the less‐ and more‐soluble diastereomeric salts showed that hydrogen‐bonding networks in the salt crystals are 21‐column‐type with a single exception which is cluster‐type. In the cases of the 21‐column‐type crystals, stability of the crystals is firstly governed by hydrogen bonds to form a 21‐column and secondly determined by intra‐columnar T‐shaped CH/π interaction(s), intra‐columnar hydrogen bond(s), inter‐columnar van der Waals interaction and/or inter‐columnar T‐shaped CH/π interaction(s). In contrast, the cluster‐type salt crystal is stabilized by the assistance of inter‐cluster T‐shaped CH/π and van der Waals interactions. To realize still more numbers of intra‐ and inter‐columnar and ‐cluster T‐shaped CH/π interactions, the seven‐membered ring of (R)‐ 5 plays a considerable role. A small amount of 4‐(trimethoxysilyl)phenyl groups was randomly introduced onto the 3,5‐dimethylphenylcarbamates of cellulose and amylose by a one‐pot method. The obtained derivatives were then effectively immobilized onto silica gel as chiral packing materials (CPMs) for high‐performance liquid chromatography through intermolecular polycondensation of the trimethoxysilyl groups. The effects of the amount of 4‐(trimethoxysilyl)phenyl groups on immobilization and enantioseparation were investigated. Also, the solvent durability of the immobilized‐type CPMs was examined with the eluents containing chloroform and tetrahydrofuran. When these eluents were used, the chiral recognition abilities of the CPMs for most of the tested racemates were improved to some extent depending on the compounds. Glycolate oxidase (GO; (S)‐2‐hydroxyacid oxidase, EC 1.1.3.15) is a flavin mononucleotide (FMN)‐dependent enzyme, which catalyzes the oxidation of 2‐hydroxy carboxylic acids to the corresponding 2‐keto acids. Catalase has been used as cocatalyst to decompose hydrogen peroxide produced in the reaction, thus limiting peroxide‐based side reactions and GO deactivation. GO from spinach and catalase T from Saccharomyces cerevisiae previously coexpressed in Pichia pastoris strain NRRL Y‐21001, was permeabilized and used for the oxidation of 3‐phenyllactic acid, 3‐indolelactic acid, 3‐chlorolactic acid, 2‐hydroxybutanoic acid, and 2‐hydroxydecanoic acid to demonstrate high degree of selectivity to the (S)‐enantiomers, leaving (R)‐isomers intact. The rates of oxidation ranged from 1.3 to 120.0%, relative to the oxidation of lactic acid to pyruvic acid. The best substrates were 3‐chlorolactic acid (110%) and 2‐hydroxybutanoic acid (120%). Oxidation was carried out with (R)‐, (S)‐, and (RS)‐3‐phenyllactic acid, (RS)‐lactic acid, and (RS)‐2‐hydroxybutanoic acid in 500 mL scale to characterize the products and stoichiometry of the reaction. All (RS)‐ and (S)‐2‐hydroxy acids produced 2‐keto acids at close to the theoretical yield in 1–9 h. (R)‐3‐Phenyllactic acid was not oxidized over a period of 9 h. Addition of exogenous FMN and catalase were not required for this oxidation using double recombinant Pichia pastoris whole cells. As GO is absolutely specific to (S)‐enantiomers, it can be used for resolution of racemic 2‐hydroxy acids to (R)‐2‐hydroxy acids as well as for production of 2‐keto acids. Two multimode Hg(II) sensors, L‐MethBQA and L‐CysBQA, were obtained by fusing methionine or S‐methyl cysteine, into a bis‐quinolyl amine‐based chiral podand scaffold. Quinolyl groups serve as the fluorophore and possess nitrogen lone pairs capable of chelating metal ions. On exposure to Hg2+ or Zn2+, these sensors show signal enhancement in fluorescence. However, Cu2+ quenches their fluorescence in 30:70 acetontrile/water. L‐CysBQA complexes with Hg2+, producing an exciton‐coupled circular dichroism spectrum with the opposite sign to the one that is produced by Cu2+ or Zn2+ complexation. L‐CysBQA binds Hg2+ more strongly than Zn2+ and is shown to differentiate Hg2+ from other metal ions, such as Zn2+, Cu2+, Ni2+, and Pb2+, exceptionally well. The synergistic use of relatively soft sulfur, quinoline‐based chiral ligands and chiroptically enhanced fluorescence detection results in high sensitivity and selectivity for Hg2+. Cholesterol‐dependent cytolysins (CDCs) are bacterial pore‐forming toxins secreted mainly by pathogenic Gram‐positive bacteria. CDCs generally recognize and bind to membrane cholesterol to create pores and lyse target cells. However, in contrast to typical CDCs such as streptolysin O, several atypical CDCs have been reported. The first of these was intermedilysin, which is secreted by Streptococcus intermedius and has human cell‐specificity, human CD59 (huCD59) being its receptor. In the study reported here, the diversity of receptor recognition among CDCs was investigated and multi‐receptor recognition characteristics were identified within this toxin family. Streptococcus mitis‐derived human platelet aggregation factor (Sm‐hPAF) secreted by S. mitis strain Nm‐65 isolated from a patient with Kawasaki disease was previously shown to hemolyze erythrocytes in a species‐dependent manner, its maximum activity being in human cells. In the present study, it was found that Sm‐hPAF recognizes both membrane cholesterol and huCD59 as receptors for triggering pore‐formation. Moreover, vaginolysin (VLY) of Gardnerella vaginalis showed similar characteristics to Sm‐hPAF regarding receptor recognition. On the basis of the results presented here, the mode of receptor recognition of CDCs can be categorized into the following three groups: (i) Group I, comprising typical CDCs with high affinity to cholesterol and no or very little affinity to huCD59; (ii) Group II, including atypical CDCs such as ILY, with no or very little affinity to cholesterol and high affinity to huCD59; and (iii) Group III, which contains atypical CDCs such as Sm‐hPAF and VLY with affinity to both cholesterol and huCD59.  相似文献

Four groups of organophosphonate derivatives enantiomers were separated on N‐(3,5‐dinitrobenzoyl)‐S‐leucine chiral stationary phase. The three‐dimensional structures of the complexes between the single enantiotopic chiral compounds and chiral stationary phase have been studied using molecular model and molecular dynamics simulation. Detailed results regarding the conformation, auto‐docking, and thermodynamic estimation are presented. The elution order of the enantiomer could be determined from the energy. Aggregation of the disordered protein α‐synuclein into amyloid fibrils is a central feature of synucleinopathies, neurodegenerative disorders that include Parkinson's disease. Small, pre‐fibrillar oligomers of misfolded α‐synuclein are thought to be the key toxic entities, and α‐synuclein misfolding can propagate in a prion‐like way. We explored whether a compound with anti‐prion activity that can bind to unfolded parts of the protein PrP, the cyclic tetrapyrrole Fe‐TMPyP, was also active against α‐synuclein aggregation. Observing the initial stages of aggregation via fluorescence cross‐correlation spectroscopy, we found that Fe‐TMPyP inhibited small oligomer formation in a dose‐dependent manner. Fe‐TMPyP also inhibited the formation of mature amyloid fibrils in vitro, as detected by thioflavin T fluorescence. Isothermal titration calorimetry indicated Fe‐TMPyP bound to monomeric α‐synuclein with a stoichiometry of 2, and two‐dimensional heteronuclear single quantum coherence NMR spectra revealed significant interactions between Fe‐TMPyP and the C‐terminus of the protein. These results suggest commonalities among aggregation mechanisms for α‐synuclein and the prion protein may exist that can be exploited as therapeutic targets.  相似文献
