Developmental Regulation of Low-temperature Tolerance in Winter Wheat   总被引:11,自引:4,他引:7  
Vernalization and photoperiod genes have wide-ranging effectson the timing of gene expression in plants. The objectives ofthis study were to (1) determine if expression of low-temperature(LT) tolerance genes is developmentally regulated and (2) establishthe interrelationships among the developmental stages and LTtolerance gene expression. LT response curves were determinedfor three photoperiod-sensitive LT tolerant winter wheat (Triticumaestivum L. em Thell) genotypes acclimated at 4 °C under8 h short-day (SD) and 20 h long-day (LD) photoperiods from0 to 112 d. Also, three de-acclimation and re-acclimation cycleswere used that bridged the vegetative/reproductive transitionpoint for each LD and SD photoperiod treatment. A vernalizationperiod of 49 d at 4 °C was sufficient for all genotypesto reach vernalization saturation as measured by minimum finalleaf number (FLN) and confirmed by examination of shoot apicesdissected from crowns that had been de-acclimated at 20 °CLD. Before the vegetative/reproductive transition, both theLD- and SD-treated plants were able to re-acclimate to similarLT50(temperature at which 50% of the plants are killed by LTstress) levels following de-acclimation at 20 °C. De-acclimationof LD plants after vernalization saturation resulted in rapidprogression to the reproductive phase and limited ability tore-acclimate. The comparative development of the SD (non-flowering-inductivephotoperiod) de-acclimated plants was greatly delayed relativeto LD plants, and this delay in development was reflected inthe ability of SD plants to re-acclimate to a lower temperature.These observations confirm the hypothesis that the point oftransition to the reproductive stage is pivotal in the expressionof LT tolerance genes, and the level and duration of LT acclimationare related to the stage of phenological development as regulatedby vernalization and photoperiod requirements. Investigation of low-temperature (LT) tolerance in cereals has commonly led to the region of the vyn-A1 vernalization gene or its homologue in related genomes. Two cultivars, one a non-hardy spring wheat and one a very cold-hardy winter wheat, whose growth habits are determined by the Vrn-A1 (spring habit) and vrn-A1 (winter habit) alleles, were chosen to produce reciprocal near-isogenic lines (NILs). These lines were then used to determine the relationship between rate of phenological development and the degree and duration of LT tolerance gene expression. Each allele was isolated in the genetic backgrounds of the non-hardy spring wheat 'Manitou' and the very cold-hardy winter wheat 'Norstar'. The effects of each allele on phenological development and low-temperature tolerance (LT50) were determined at regular intervals over a 4 degrees C acclimation period of 0-98 d. The vegetative/reproductive transition, as determined by final leaf number (FLN), was found to be a major developmental factor influencing LT tolerance. Possession of a vernalization requirement increased both the length of the vegetative growth phase and LT tolerance. Similarly, increased FLN in spring Norstar and winter Manitou NILs delayed their vegetative/reproductive transition and increased their LT tolerance relative to Manitou. Although the winter Manitou NILs had a lower FLN than the spring Norstar NILs, they were able to extend their vegetative stage to a similar length by increasing the phyllochron (interval between the appearance of successive leaves). Cereal plants have four ways of increasing the length of the vegetative phase, all of which extend the time that low-temperature tolerance genes are more highly expressed: (1) vernalization; (2) photoperiod responses; (3) increased leaf number; and (4) increased length of the phyllochron.  相似文献

The initiation of flowering is a crucial trait that allows temperate plants to flower in the favourable conditions of spring. The timing of flowering initiation is governed by two main mechanisms: vernalization that defines a plant's requirement for a prolonged exposure to cold temperatures; and photoperiod sensitivity defining the need for long days to initiate floral transition. Genetic variability in both vernalization and photoperiod sensitivity largely explains the adaptability of cultivated crop plants such as bread wheat (Triticum aestivum L.) to a wide range of climatic conditions. The major genes controlling wheat vernalization (VRN1, VRN2, and VRN3) and photoperiod sensitivity (PPD1) have been identified, and knowledge of their interactions at the molecular level is growing. However, the quantitative effects of temperature and photoperiod on these genes remain poorly understood. Here it is shown that the distinction between the temperature effects on organ appearance rate and on vernalization sensu stricto is crucial for understanding the quantitative effects of the environmental signal on wheat flowering. By submitting near isogenic lines of wheat differing in their allelic composition at the VRN1 locus to various temperature and photoperiod treatments, it is shown that, at the whole-plant level, the vernalization process has a positive response to temperature with complex interactions with photoperiod. In addition, the phenotypic variation associated with the presence of different spring homoeoalleles of VRN1 is not induced by a residual vernalization requirement. The results demonstrate that a precise definition of vernalization is necessary to understand and model temperature and photoperiod effects on wheat flowering. It is suggested that this definition should be used as the basis for gene expression studies and assessment of functioning of the wheat flowering gene network, including an explicit account of the quantitative effect of environmental variables.  相似文献

东北、华北和西北50a来的平均气温增幅高于全国平均水平,气候变暖明显,尤其冬季增温最显著。区域增暖的极端最低气温远比极端最高气温的贡献大。东北、华北大部、西北东部降水量明显减少,平均每10a减少20—40mm,尤其春夏季减少最明显。这种趋势一直延续到20世纪90年代以后,干旱化趋势非常突出。在综述我国北方现代气候变化基本特征是暖干化的基础上,重点阐述了喜凉作物冬小麦、春小麦、马铃薯和喜温作物水稻、玉米、谷子、糜子等7种主要粮食作物的生长发育、品种熟性、种植区域与面积、产量与品质等对气候暖干化的响应特征。揭示了气候暖干化使春播作物播期提早,苗期生长发育速度加快,营养生长期提前,生殖生长期和全生育期延长;秋作物发育期推迟,生殖生长期和全生长期延长;越冬作物播期推迟,越冬死亡率降低,种植风险减少,春初提前返青,生殖生长期提早,全生育期缩短。使作物适宜种植区域向高纬度高海拔扩展;品种熟性向偏中晚熟高产品种发展;喜温作物和越冬作物以及冷凉气候区的作物种植面积迅速扩大;在旱作区种植不较耐旱的玉米、春小麦等作物种植面积受到制约。对雨养农业区的作物气候产量影响严重,尤其对不够耐旱的小麦和玉米的气候产量受影响最大;对较耐旱的谷子、糜子、马铃薯等影响较轻。从作物属性而言,对喜温作物水稻、玉米和越冬作物冬小麦有利于气候产量提高;对喜凉作物春小麦和马铃薯的气候产量将产生不利影响。同时,提出了从5个方面应对气候暖干化的技术措施,调整作物种植结构,确保粮食生产安全;根据不同气候年型调整各种作物种植比例;针对不同气候区域发展优势作物和配置作物种植格局;采取不同栽培技术和管理模式应对气候变化;采取综合配套技术提髙抵御灾害能力。为粮食作物安全生产和种植结构调整与布局提供科学依据。  相似文献

BACKGROUND AND AIMS: Frost tolerance of wheat depends primarily upon a strong vernalization requirement, delaying the transition to the reproductive phase. The aim of the present study was to learn how saturation of the vernalization requirement and apical development stage are related to frost tolerance in wheat. METHODS: 'Mironovskaya 808', a winter variety with a long vernalization requirement, and 'Leguan', a spring variety without a vernalization requirement, were acclimated at 2 degrees C at different stages of development. Plant development (morphological stage of the shoot apex), vernalization requirement (days to heading) and frost tolerance (survival of the plants exposed to freezing conditions) were evaluated. KEY RESULTS: 'Mironovskaya 808' increased its frost tolerance more rapidly; it reached a higher level of tolerance and after a longer duration of acclimation at 2 degrees C than was found in 'Leguan'. The frost tolerance of 'Mironovskaya 808' decreased and its ability to re-acclimate a high tolerance was lost after saturation of its vernalization requirement, but before its shoot apex had reached the double-ridge stage. The frost tolerance of 'Leguan' decreased after the plants had reached the floret initiation stage. CONCLUSIONS: The results support the hypothesis that genes for vernalization requirement act as a master switch regulating the duration of low temperature induced frost tolerance. In winter wheat, due to a longer vegetative phase, frost tolerance is maintained for a longer time and at a higher level than in spring wheat. After the saturation of vernalization requirement, winter wheat (as in spring wheat) established only a low level of frost tolerance.  相似文献

Phenotypic manifestations of Vrn(vernalization) and Ppd (photoperiod) genes responsible for transition of bread wheat Triticum aestivumL. to generative growth (flowering) are mutually related. Since the mechanism of phytochrome-induced photoperiodism involves the enzymes of cyclic adenosine monophosphate metabolism, and phosphodiesterase in particular, we tested involvement of phosphodiesterase in the process of winter wheat vernalization and formation of flowering competence in alternate wheat requiring a long photoperiod but no vernalization for its transition to flowering. We studied temperature dependence of phosphodiesterase activity in vernalized and unvernalized winter wheat on the one hand and in etiolated and red light illuminated seedlings of alternate wheat on the other hand. Short-term experiments demonstrated that red light illumination is similar to long photoperiod by the effect on the long-day plants. Both influences induced a pronounced inversion of the temperature profile of phosphodiesterase activity in the 28–45°C range. We propose that phosphodiesterase is involved in vernalization and can serve as a receptor of low temperature in winter wheat. Changed temperature profile is a radical control mechanism of phosphodiesterase activity in response to the influences (red light and vernalizing temperatures) responsible for competence of various bread wheat forms for generative growth.  相似文献

The vulnerability and adaptation of major agricultural crops to various soils in north‐eastern Austria under a changing climate were investigated. The CERES crop model for winter wheat and the CROPGRO model for soybean were validated for the agrometeorological conditions in the selected region. The simulated winter wheat and soybean yields in most cases agreed with the measured data. Several incremental and transient global circulation model (GCM) climate change scenarios were created and used in the study. In these scenarios, annual temperatures in the selected region are expected to rise between 0.9 and 4.8 °C from the 2020s to the 2080s. The results show that warming will decrease the crop‐growing duration of the selected crops. For winter wheat, a gradual increase in air temperature resulted in a yield decrease. Incremental warming, especially in combination with an increase in precipitation, leads to higher soybean yield. A drier climate will reduce soybean yield, especially on soils with low water storage capacity. All transient GCM climate change scenarios for the 21st century, including the adjustment for only air temperature, precipitation and solar radiation, projected reductions of winter wheat yield. However, when the direct effect of increased levels of CO2 concentration was assumed, all GCM climate change scenarios projected an increase in winter wheat yield in the region. The increase in simulated soybean yield for the 21st century was primarily because of the positive impact of warming and especially of the beneficial influence of the direct CO2 effect. Changes in climate variability were found to affect winter wheat and soybean yield in various ways. Results from the adaptation assessments suggest that changes in sowing date, winter wheat and soybean cultivar selection could significantly affect crop production in the 21st century.  相似文献

Winter wheat “Nong Da 139” and spring wheat “Zhong 8022” were used for this experiment. The effect of low temperature treatment at different periods on protein content and composition of the seedling shoots as well as subsequent development was studied. The main results are as follows: 1. More than 40 days of vernalizing treatment were required for a rapid and uniform earing by winter wheat “Nong Da 139”. After 14–21 days vernalization, the winter wheat is possible to ear in summer, but in an irregular manner. The results indicate that during the whole vernalization process, the effect of low temperature on earing development varies with the duration of treatment. At the earlier stage, it is likely that the Iow temperature induces a change in physiological process, and in the later stage, it only accelerates the development. It is there- fore, suggested that there are two distinct processes existing during vernalization. The transformation from the former to the latter state appears to occur at the middle period of vernalization process. 2. The protein metabolic inhibitors, such as ethionine and p-fluorophenylalanine, interfere with the vernalizing process of winter wheat also at its middle period. 3. With low temperature treatment for different periods, the soluble protein content and composition are found to be changed in winter wheat shoots. At the middle-stage of vernalization (after 14 days low temperature treatment), not only is the content of protein increased twice as compared with the control, but new proteins (on electrophorestic gel) are also produced. On the contrary, there was no difference in protein bands for spring wheat. Spring wheat not to be treated by low temperature, has already possessed the proteins that appeared in vernalized winter wheat shoots. These results demonstrated that the days of 14–21 are the critical time for the vernalization of the winter wheat. The new proteins synthesized at this stage might be the factor of the determination whether or not the plants will transform from vegetative stage into reproductive phase.  相似文献

Background and Aims

Crop models for herbaceous ornamental species typically include functions for temperature and photoperiod responses, but very few incorporate vernalization, which is a requirement of many traditional crops. This study investigated the development of floriculture crop models, which describe temperature responses, plus photoperiod or vernalization requirements, using Australian native ephemerals Brunonia australis and Calandrinia sp.

Methods

A novel approach involved the use of a field crop modelling tool, DEVEL2. This optimization program estimates the parameters of selected functions within the development rate models using an iterative process that minimizes sum of squares residual between estimated and observed days for the phenological event. Parameter profiling and jack-knifing are included in DEVEL2 to remove bias from parameter estimates and introduce rigour into the parameter selection process.

Key Results

Development rate of B. australis from planting to first visible floral bud (VFB) was predicted using a multiplicative approach with a curvilinear function to describe temperature responses and a broken linear function to explain photoperiod responses. A similar model was used to describe the development rate of Calandrinia sp., except the photoperiod function was replaced with an exponential vernalization function, which explained a facultative cold requirement and included a coefficient for determining the vernalization ceiling temperature. Temperature was the main environmental factor influencing development rate for VFB to anthesis of both species and was predicted using a linear model.

Conclusions

The phenology models for B. australis and Calandrinia sp. described development rate from planting to VFB and from VFB to anthesis in response to temperature and photoperiod or vernalization and may assist modelling efforts of other herbaceous ornamental plants. In addition to crop management, the vernalization function could be used to identify plant communities most at risk from predicted increases in temperature due to global warming.  相似文献

Low temperature (LT) tolerance in cereals needs developmental regulation of metabolites, a process which is associated with vernalization requirement. This study was initiated to investigate the relationships among stage of phenological development, final leaf number (FLN), the activities superoxide dismutase, catalase, guaiacol peroxidase, ascorbate peroxidase and polyphenol oxidase, the contents of proline, photosynthetic pigments, and hydrogen peroxide (H2O2) during vernalization and LT acclimation in spring and winter wheat. Six genotypes with different vernalization requirements were grown under greenhouse and field conditions. The spring-habit parent, “Pishtaz” and line 4021, rapidly entered the reproductive phase and had a limited capacities to LT acclimate. They also had the lowest antioxidative activities and accumulation of proline among genotypes. Lines 4002 and 4014, with a short vernalization requirement and higher FLN, remained in the early stages of phenological development longer and developed a higher level of LT tolerance and metabolites compared to spring habit genotypes. In contrast, the winter habit “Norstar” and line 4023 spent a longer time in the vegetative stage and accumulated higher levels of metabolites. Maximum LT tolerance and metabolite accumulations occurred near the vegetative/reproductive transition in all genotypes. The longer periods of vernalization and increased FLN that happened along with increased defense mechanisms and decreased damage indices (H2O2 content and LT50) ensured LT tolerance in wheat. These results demonstrate that both genetic and environmental factors via developmental regulation of metabolites play important roles in creating LT tolerance in long mild winters of Iran. Significant correlations coefficients for many of the metabolites considered in this study and Lethal temperature 50 (LT50) also suggest that they could be useful as indirect measures of plant LT tolerance potential in wheat breeding programs.  相似文献

在自然界中许多高等植物需要通过冬季的低温阶段实现从营养生长到生殖生长的时期转化,这一生物学过程称作春化作用。小麦(Triticum aestivum L.)和油菜(Brassica napus L.)等作物以种子为产品器官,生产上往往通过茬口安排和栽培措施使植株尽早通过春化作用,以促进花芽形成和花器官发育,而大白菜(B rapa ssp.pekinenesis)和甘蓝(B.oleracea)等作物以叶球等营养器官作为产品器官,生产上则设法避免低温引起的春化作用,以保证产品器官的充分生长。FLOWERING LOCUS C(FLC)作为一种重要的开花抑制蛋白负调控春化作用,参与植株从营养生长向生殖生长的转化过程。文章综述了春化中FLC表达受抑制主要通过低温诱导表达FLC基因区域的非编码RNA以及VRN1、VRN2、VIN3等蛋白参与介导组蛋白甲基化,从而在表观遗传上控制春化作用的进程和产品器官的正常发育。  相似文献

Vernalization is a decisive physiological process for heading, flowering and graining of biennial plants. Variable duration of low-temperature treatment has effects on lateral morphogenesis, such as spike initiation, floral development and graining rate in winter wheat ( Triticum aestivum L.). The investigation data showed that the duration of vernalization treatment was a decisive factor for the initiation of spike relevant to the time of initiation; the longer the duration at low temperature, the earlier the spike initiation in winter wheat. In the process of the spike differentiation, relatively lower temperature and longer differential time benefited for spike differentiation. Under laboratory condition, a low-temperature treatment for 45 d was optial for flower differentiation and graining in winter wheat. It is novelly recognized that vernalization treatment is essential for development of both spikes and spikelets, besides for promoting initiation of differentiation in winter wheat.  相似文献

The recent whole-plant research reviewed suggests the commonly applied paradigms about vernalization and photoperiodism should be replaced. A simple equation based on new paradigms predictively models with excellent fit the published days to flowering of at least six plant species. The paradigm that the response to photoperiod of the days to flowering (DTF) of crop plants is revealed adequately by comparing a range of photoperiods at just one temperature should be replaced with the following concepts. There is a base (lowest) temperature below which photoperiod gene activity does not occur, and, when the temperature is high enough to allow activity, there is always a photoperiod × temperature × genotype interaction effect on the days to flowering. Similarly, the paradigm that vernalization gene activity occurs at low temperature and promotes development should be replaced as follows. Vernalization gene activity occurs only if the temperature is above a base (lowest) temperature that allows activity of the vernalization gene(s), and this activity delays development to flowering. Development to flowering is accelerated by low-temperature vernalization, because the low temperature prevents vernalization gene activity, thereby preventing delay of the DTF. The phenomena called long-day (LD) vernalization and short-day (SD) vernalization are reinterpreted as follows. The apparent replacement by short or long daylength of a requirement for low-temperature vernalization is actually a replacement by the low temperature of a requirement for long or short day. Just as true low-temperature vernalization results from prevention of vernalization gene activity, these SD and LD promotions of the DTF occur because the photoperiod gene activity is prevented by the low temperature. Rather than requiring an environment that induces flowering, an inherent capability for rapid development to flowering is expressed, if there is no delay of the DTF by the activity of either or both of the vernalization and photoperiod gene(s). All the above-mentioned effects of temperature are due to the Q10 effect on the specified photoperiod or vernalization gene activity. The effect of thermal time (due to the accumulated growing degree days) is the integrated Q10 effect on all additional genes that partially control the rate of development to the reproductive stage.  相似文献

Background and Aims

Accurately representing development is essential for applying crop simulations to investigate the effects of climate, genotypes or crop management. Development in wheat (Triticum aestivum, T. durum) is primarily driven by temperature, but affected by vernalization and photoperiod, and is often simulated by reducing thermal-time accumulation using vernalization or photoperiod factors or limiting accumulation when a lower optimum temperature (Toptl) is exceeded. In this study Toptl and methods for representing effects of vernalization and photoperiod on anthesis were examined using a range of planting dates and genotypes.

Methods

An examination was made of Toptl values of 15, 20, 25 and 50 °C, and either the most limiting or the multiplicative value of the vernalization and photoperiod development rate factors for simulating anthesis. Field data were from replicated trials at Ludhiana, Punjab, India with July through to December planting dates and seven cultivars varying in vernalization response.

Key Results

Simulations of anthesis were similar for Toptl values of 20, 25 and 50 °C, but a Toptl of 15 °C resulted in a consistent bias towards predicting anthesis late for early planting dates. Results for Toptl above 15 °C may have occurred because mean temperatures rarely exceeded 20 °C before anthesis for many planting dates. For cultivars having a strong vernalization response, anthesis was more accurately simulated when vernalization and photoperiod factors were multiplied rather than using the most limiting of the two factors.

Conclusions

Setting Toptl to a high value (30 °C) and multiplying the vernalization and photoperiod factors resulted in accurately simulating anthesis for a wide range of planting dates and genotypes. However, for environments where average temperatures exceed 20 °C for much of the pre-anthesis period, a lower Toptl (23 °C) might be appropriate. Heading time in bread wheat ( Triticum aestivum L.) is determined by three characters – vernalization requirement, photoperiodic sensitivity and narrow-sense earliness (earliness per se) – which are involved in the phase transition from vegetative to reproductive growth. The wheat APETALA1 ( AP1 )-like MADS-box gene, wheat AP1 ( WAP1 , identical with VRN1 ), has been identified as an integrator of vernalization and photoperiod flowering promotion pathways. A MADS-box gene, SUPPRESSOR OF OVEREXPRESSION OF CO 1 ( SOC1 ) is an integrator of flowering pathways in Arabidopsis . In this study, we isolated a wheat ortholog of SOC1 , wheat SOC1 ( WSOC1 ), and investigated its relationship to WAP1 in the flowering pathway. WSOC1 is expressed in young spikes but preferentially expressed in leaves. Expression starts before the phase transition and is maintained during the reproductive growth phase. Overexpression of WSOC1 in transgenic Arabidopsis plants caused early flowering under short-day conditions, suggesting that WSOC1 functions as a flowering activator in Arabidopsis . WSOC1 expression is affected neither by vernalization nor photoperiod, whereas it is induced by gibberellin at the seedling stage. Furthermore, WSOC1 is expressed in transgenic wheat plants in which WAP1 expression is cosuppressed. These findings indicate that WSOC1 acts in a pathway different from the WAP1 -related vernalization and photoperiod pathways.  相似文献

Winterhardiness has three primary components: photoperiod (day length) sensitivity, vernalization response, and low temperature tolerance. Photoperiod and vernalization regulate the vegetative to reproductive phase transition, and photoperiod regulates expression of key vernalization genes. Using two barley mapping populations, we mapped six individual photoperiod response QTL and determined their positional relationship to the phytochrome and cryptochrome photoreceptor gene families and the vernalization regulatory genes HvBM5A, ZCCT-H, and HvVRT-2. Of the six photoreceptors mapped in the current study (HvPhyA and HvPhyB to 4HS, HvPhyC to 5HL, HvCry1a and HvCry2 to 6HS, and HvCry1b to 2HL), only HvPhyC coincided with a photoperiod response QTL. We recently mapped the candidate genes for the 5HL VRN-H1 (HvBM5A) and 4HL VRN-H2 (ZCCT-H) loci, and in this study, we mapped HvVRT-2, the barley TaVRT-2 ortholog (a wheat flowering repressor regulated by vernalization and photoperiod) to 7HS. Each of these three vernalization genes is located in chromosome regions determining small photoperiod response QTL effects. HvBM5A and HvPhyC are closely linked on 5HL and therefore are currently both positional candidates for the same photoperiod effect. The coincidence of photoperiod-responsive vernalization genes with photoperiod QTL suggests vernalization genes should also be considered candidates for photoperiod effects.  相似文献

Monitoring nitrogen use efficiency (NUE) in plants is becoming essential to maintain yield while reducing fertilizer usage. Optimized NUE application in major crops is essential for long-term sustainability of agriculture production. Here, we report the precise identification of 11 major chromosomal regions controlling NUE in wheat that co-localise with key developmental genes such as Ppd (photoperiod sensitivity), Vrn (vernalization requirement), Rht (reduced height) and can be considered as robust markers from a molecular breeding perspective. Physical mapping, sequencing, annotation and candidate gene validation of an NUE metaQTL on wheat chromosome 3B allowed us to propose that a glutamate synthase (GoGAT) gene that is conserved structurally and functionally at orthologous positions in rice, sorghum and maize genomes may contribute to NUE in wheat and other cereals. We propose an evolutionary model for the NUE locus in cereals from a common ancestral region, involving species specific shuffling events such as gene deletion, inversion, transposition and the invasion of repetitive elements.  相似文献
