An improved and scalable protocol for the synthesis of 3′-O-succinates of nucleosides has been developed using succinic anhydride. As a result, formation of unwanted dimer has been completely eliminated and use of toxic and smelly reagents have been avoided during synthesis of nucleoside succinates. All succinates were isolated in pure state without silica gel column chromatography.  相似文献

A more convenient and efficient method is described for the preparation of 3′-O-succinates of 2′-deoxyribo- and ribonucleosides, in a variety of solvents. Tocopheryl succinates (TOSs) are, in contrast to tocopherols, highly cytotoxic against many cancer cells. In this study the enzyme activity of secretory phospholipase A(2) towards various succinate-phospholipid conjugates has been investigated. The synthesis of six novel phospholipids is described, including two TOS phospholipids conjugates. The studies revealed that the TOS conjugates are poor substrates for the enzyme whereas the phospholipids with alkyl and phenyl succinate moieties were hydrolyzed by the enzyme to a high extent.  相似文献

The reactions of moraprenol and dolichol with succinic and maleic anhydrides in the presence of pyridine or triethylamine were studied, and the conditions were found for the efficient synthesis of moraprenyl and dolichyl hydrogen succinates and maleates. An alternative solid support for oligonucleotide synthesis was developed by coupling a polymer colloid to a modified polyethylene filter disc. The functions on the polymer colloid not used for attachment to the surface were derivatized with a Jeffamine diamine and loaded with appropriate deoxynucleoside succinates. The performance of this support system was evaluated and compared to existing resins.  相似文献

Novel aspartate and succinate CGRP full antagonists were identified through core modification of a potent lead CGRP antagonist, BMS-694153. While aspartates were much less active and had a flat SAR, some of the succinates were very potent CGRP full antagonists and matched the potency of BMS-694153. The most potency resides in the S enantiomer as demonstrated through an asymmetric synthesis.  相似文献

Effect of succinylation on the rheological profile of starch pastes   总被引:2,自引:0,他引:2  
Rheological properties of native corn and amaranth starches, and the changes therein on succinylation have been evaluated. The degree of substitution (DS) was varied from 0.05 to 0.20 at concentrations up to 5%. A strong shear thinning behavior was observed in all the starch pastes, as described by the power law parameters, i.e. the consistency index, K, and the flow behavior or power law index, n. The effect of concentration and DS on the apparent viscosity is described. Amaranth starch succinates showed a greater shear thinning behavior vis-à-vis corn starch succinates. The bulky hydrophilic succinate group seem to influence the rheological properties.  相似文献

A series of transdermal permeation enhancers based on dicarboxylic acid esters was studied. Single-chain amphiphiles were markedly more effective than the double-chain ones. Monododecyl maleate, that is a cis derivative, was a more potent enhancer than its trans isomer, while the activity of succinates strongly depended on the donor vehicle. No difference between diastereoisomeric tartaric and meso-tartaric acid derivatives was found.  相似文献

Fumaric, palmitic, oleic and abscisic acids and methyl and ethyl hydrogen succinates were isolated from seedlings of dwarf pea (Pisum sativum L., var. Progress No. 9), grown under red light, as growth inhibitors which interfered with the responses of these plants to GA3. Of the six compounds, fumaric acid did not show any inhibitory effect on stem elongation in the absence of GA3.  相似文献

Accessible triterpenoids of ursane and lupane series, the flavonoid dihydroquercetin and their synthetic derivatives with polar substituents were tested in vitro for inhibition of collagenase 1 (MMP-1) in UVB irradiation assay. Ursolic acid and uvaol disuccinate were the most active inhibitors in the ursane series. In the lupane series, the best inhibition was manifested by carboxymethyl ester of betulonic acid and betulin succinates. Down- regulation of MMP-1 by dihydroquercetin and its synthetic derivatives surpassed the activity of a standard (retinoic acid).  相似文献

Mannose-capped lipoarabinomannan (ManLAM) is a complex lipoglycan abundantly present in the Mycobacterium tuberculosis cell envelope. Many biological properties have been ascribed to ManLAM, from directly interacting with the host and participating in the intracellular survival of M. tuberculosis, to triggering innate and adaptive immune responses, including the activation of CD1b-restricted T cells. Due to its structural complexity, ManLAM is considered a heterogeneous population of molecules which may explain its different biological properties. The presence of various modifications such as fatty acids, succinates, lactates, phosphoinositides and methylthioxylose in ManLAM have proven to correlate directly with its biological activity and may potentially be involved in the interactions between CD1b and the T cell population. To further delineate the specific ManLAM epitopes involved in CD1b-restricted T cell recognition, and their potential roles in mediating immune responses in M. tuberculosis infection, we established a method to resolve ManLAM into eight different isoforms based on their different isoelectric values. Our results show that a ManLAM isoform with an isoelectric value of 5.8 was the most potent in stimulating the production of interferon-γ in different CD1b-restricted T-cell lines. Compositional analyses of these isoforms of ManLAM revealed a direct relationship between the overall charge of the ManLAM molecule and its capacity to be presented to T cells via the CD1 compartment.  相似文献

The rate of oligomer synthesis on mineral surface is higher than that in solution. The mechanism of synthesis has been described. The requirement for phage protein synthesis for the inhibition of host deoxyribonucleic acid synthesis has been investigated by using a phage mutant unable to catalyze the production of any phage deoxyribonucleic acid. It has been concluded that the major pathway whereby phage inhibit host syntheses requires protein synthesis. The inhibition of host syntheses by phage ghosts is not affected by inhibitors of protein synthesis.  相似文献

The synthesis of globin proteins in blood reticulocytes of homozygous beta-thalassemic patients from Tadzhikistan has been previously studied. beta-thalassemia with sharp repression of beta-globin protein synthesis (alpha/beta greater than 10) has been shown to be most representative for the region. In this work, the synthesis of globin proteins has been studied in bone marrow cells of homozygous beta-thalassemic patients. Comparison of data on globin synthesis in bone marrow cells and in blood reticulocytes of the patients has revealed that in some cases the disbalance of chain synthesis in both cell types is equal. In other cases the disbalance in bone marrow cells is less than in blood cells, indicating the instability of beta-globin mRNA that is partially degrading in the process of cell maturation. Homozygous beta-thalassemic cases with low content of Hb F in blood cells (5-10%), with substantial disbalance of alpha and beta-globin synthesis and marked production of gamma-globins in bone marrow cells and in blood reticulocytes are of special interest. It has been assumed that parallel to beta-thalassemia some instability of gamma-globin proteins takes place in these patients.  相似文献

Tetrathiomolybdate has been used as an efficient sulfur-transfer reagent in the synthesis of a number of thiolevomannosan derivatives having an axial-rich 1C4 conformation. An unprecedented synthesis of a novel thioorthoester and its synthetic utility in glycosylation has been demonstrated. Based on the torsional mechanism of ATP synthesis by ATP synthase, a kinetic scheme has been developed in this work. The scheme considers adenine nucleotide transport, binding of substrates ADP and P(i), unbinding of product ATP, and ATP synthesis. This kinetic scheme has been analyzed mathematically, and a kinetic model has been obtained to explain the experimentally observed hyperbolic Michaellian dependence of the rate of ATP synthesis on the ADP concentration by ATP synthase under physiological steady-state operating conditions. The principal results of the kinetic model have been compared with the experimental data and an estimate of the enzymological kinetic parameters V(max), K(M), and K(I) has been determined. Mechanistic implications arising from further analysis of the kinetic model have been discussed. These biological implications provide deep insight into the sequence of events leading to ATP synthesis.  相似文献

Local protein synthesis in the synapse is required for synaptic plasticity and has been implicated in learning and memory. However, direct evidence that behavioral training induces local protein synthesis has been lacking. In this issue of Cell, Ashraf et al. (2006) observe persistent local protein synthesis in the antennal lobe synapses of the fruit fly following olfactory-avoidance learning. Excretory products of the tapeworm Hymenolepis diminuta, fed D-[13C6]glucose in vitro for 90 min, were studied using 1H and 13C nuclear magnetic resonance spectroscopy. Signals due to lactate, succinate, acetate, and alanine were identified in the spectra. Several differently labeled species were present for these metabolites; the variations of higher concentration were a consequence of metabolic factors while those of lower concentration could be accounted for by residual 12C in the glucose. The two major labeled lactates, U-13C and 2,3-13C2, were in the ratio 2:1, respectively, and the three major labeled succinates, 1,2,2'-13C3,2,2'-13C2, and U-13C, were present in the ratio 20:10:3, respectively. The different species of labeled end products are related to the overall glucose metabolism of H. diminuta.  相似文献
