堆肥对土壤重金属垂直分布的影响与污染评价   总被引:8,自引:1,他引:7  
对不同畜禽粪便堆肥与土壤重金属垂直分布的关系进行了研究.结果表明,在畜禽粪便堆肥过程中,粪堆下土壤pH值和有机质显著增加,其pH和有机质含量的垂直分布表现为从表层到底层逐渐降低.各种畜禽粪便粪堆下土壤Zn、Cd含量明显增高,且从表层到底层呈逐渐减小的趋势.鸡粪和猪粪堆下土壤Cu含量随土层深度增加而降低;牛粪堆下土壤Cu含量随土层深度增加没有明显的变化.自然条件下Cd和Zn在土壤系统中的迁移能力大于Cu.各粪堆下的部分土层Cu、Zn、Cd含量超过我国土壤环境质量一级标准.应用地质积累指数法对各土层污染评价的结果表明,只有肉鸡粪堆下0~10cm土壤和蛋鸡粪堆下0~40cm土壤受到轻度Zn污染,其它粪堆下各土层均未受到Cu、Zn、Pb和Cd污染.  相似文献

The number and proportion of CTX-M positive Escherichia coli organisms were determined in feces from cattle, chickens, and pigs in the United Kingdom to provide a better understanding of the risk of the dissemination of extended-spectrum β-lactamase (ESBL) bacteria to humans from food animal sources. Samples of bovine (n = 35) and swine (n = 20) feces were collected from farms, and chicken cecal contents (n = 32) were collected from abattoirs. There was wide variation in the number of CTX-M-positive E. coli organisms detected; the median (range) CFU/g were 100 (100 × 10(6) to 1 × 10(6)), 5,350 (100 × 10(6) to 3.1 × 10(6)), and 2,800 (100 × 10(5) to 4.7 × 10(5)) for cattle, chickens, and pigs, respectively. The percentages of E. coli isolates that were CTX-M positive also varied widely; median (range) values were 0.013% (0.001 to 1%) for cattle, 0.0197% (0.00001 to 28.18%) for chickens, and 0.121% (0.0002 to 5.88%) for pigs. The proportion of animals designated high-density shedders (≥1 × 10(4) CFU/g) of CTX-M E. coli was 3/35, 15/32, and 8/20 for cattle, chickens, and pigs, respectively. We postulate that high levels of CTX-M E. coli in feces facilitate the dissemination of bla(CTX-M) genes during the rearing of animals for food, and that the absolute numbers of CTX-M bacteria should be given greater consideration in epidemiological studies when assessing the risks of food-borne transmission.  相似文献

Dairy cattle have been identified as a principal reservoir of Escherichia coli O157:H7. The fate of this pathogen in bovine feces at 5, 22, and 37 degrees C was determined. Two levels of inocula (10(3) and 10(5) CFU/g) of a mixture of five nalidixic acid-resistant E. coli O157:H7 strains were used. E. coli O157:H7 survived at 37 degrees C for 42 and 49 days with low and high inocula, respectively, and at 22 degrees C for 49 and 56 days with low and high inocula, respectively. Fecal samples at both temperatures had low moisture contents (about 10%) and water activities ( < 0.5) near the end of the study. E. coli O157:H7 at 5 degrees C survived for 63 to 70 days, with the moisture content (74%) of feces remaining high through the study. Chromosomal DNA fingerprinting of E. coli O157:H7 isolates surviving near the completion of the study revealed that the human isolate strain 932 was the only surviving strain at 22 or 37 degrees C. All five strains were isolated near the end of incubation from feces held at 5 degrees C. Isolates at each temperature were still capable of producing both verotoxin 1 and verotoxin 2. Results indicate that E. coli O157:H7 can survive in feces for a long period of time and retain its ability to produce verotoxins. Hence, bovine feces are a potential vehicle for transmitting E. coli O157:H7 to cattle, food, and the environment. Appropriate handling of bovine feces is important to control the spread of this pathogen.  相似文献

A thermophilic bacterium, strain TAT105, was isolated from compost made of animal wastes. TAT105 had high tolerance to ammonium nitrogen up to 1200 mM, and highly assimilated nitrogen during the growth on swine feces. The strain was classified into Bacillus, close to Bacillus pallidus. To evaluate the effect of adding TAT105 to ammonia (NH3) emission during the composting process of animal wastes, laboratory scale composting was done. NH3 emission tended to be lower and nitrogen loss was smaller in the TAT105-added material than in the control material to which TAT105 was not added. Thermophilic ammonium-tolerant bacteria in the TAT105-added material increased to about 8x10(9) CFU/g of dry matter on the average during the tests, and most of them were judged to be TAT105 from morphological colony discrimination. These results suggested the possibility of reducing NH3 emission from composting of animal wastes by adding TAT105.  相似文献

AIM: To determine the persistence of Escherichia coli O157 in contrasting organic wastes spread to land and to assess the potential environmental risk associated with the disposal of these wastes to land. METHODS AND RESULTS: Twenty-seven organic wastes originating from slaughterhouses, wastewater treatment plants (raw and treated sewage), creameries and farms (bovine slurry), were inoculated with E. coli O157:H7 and incubated at 10 degrees C. Although pathogen numbers gradually declined in all the wastes, albeit at different rates even in the same waste type, E. coli O157:H7 was still viable in 77% of organic wastes tested after 2 months. CONCLUSIONS: Long-term storage of organic wastes led to a significant and gradual decline in E. coli O157:H7 numbers. Consequently, storage may be a useful means of reducing the pathogen load of wastes destined for land application. However, in most cases, long-term storage cannot be expected to completely eliminate E. coli O157:H7 from waste. SIGNIFICANCE AND IMPACT OF THE STUDY: Our results indicate that current legislation may be insufficient to protect the environment from E. coli O157:H7 contamination from untreated wastes spread to land.  相似文献

在实验室可控条件下,以碳氮比28.7∶1的农业有机废弃物(牛粪和稻秆)为赤子爱胜蚓(Eisenia foetida)的培养基质,研究蚯蚓的堆制作用对有机物料的化学及生物学特性的影响.结果表明: 蚯蚓堆制处理30 d后,基质pH值、碳氮比显著降低,全磷显著升高,而全氮、碱解氮、可溶性碳、速效磷、微生物生物量碳、呼吸速率和微生物熵分别提高8.5%、2.6%、18%、63%、212%、44%和300%,有机质、呼吸熵分别降低5.0%和21.9%.蚯蚓堆制处理后物料具有较高的转化酶、酸性和碱性磷酸酶活性,较低的过氧化氢酶和脲酶活性.多元数据分析结果显示,自然堆制和蚯蚓堆制处理物料的化学和生物学特性均呈现显著的差异性.蚯蚓堆制处理优于自然堆制处理,可以明显改善有机物料的化学、生物学性质,是一种高效率处理农业有机废弃物的技术.  相似文献

The aim of this study was to monitor some microbial indicators and pathogen contents (sulphite reducers clostridia, total enterobacteriaceae, total coliforms, faecal coliforms (Escherichia coli), enterococci, Staphylococcus aureus and Salmonella spp.) throughout the co-composting of wastes from the winery and distillery industry with other organic residues, as well as the effect of the composting system used. Seven different piles using mixtures of winery-distillery wastes with other organic materials were prepared. P1 and P2 were made using grape stalk (GS), grape marc (GM), exhausted grape marc (EGM) and sewage sludge (SS), whereas in P3 and P4 were also used exhausted grape marc with cow manure (CW) and poultry manure (PM), respectively, using the Rutgers system. Additionally, P2 was watered with vinasse (V). The rest of piles (P5, P6 and P7) were prepared with grape marc, exhausted grape marc, cow manure and poultry manure, using the turning system. The effectiveness of the composting process to reduce the pathogen content was higher in the static aerated piles than in those elaborated with the turning. The relatively high temperatures (50-60 degrees C) reached in some of the piles produced a notable decrease in some microbial groups, such as total and faecal coliforms (E. coli), but the characteristics of the raw materials used notably influenced the pathogen contents of the end-product.  相似文献

Twelve ruminally cannulated cattle, adapted to forage or grain diet with or without monensin, were used to investigate the effects of diet and monensin on concentration and duration of ruminal persistence and fecal shedding of E. coli O157:H7. Cattle were ruminally inoculated with a strain of E. coli O157:H7 (10(10) CFU/animal) made resistant to nalidixic acid (Nal(r)). Ruminal and fecal samples were collected for 11 weeks, and then cattle were euthanized and necropsied and digesta from different gut locations were collected. Samples were cultured for detection and enumeration of Nal(r) E. coli O157:H7. Cattle fed forage diets were culture positive for E. coli O157:H7 in the feces for longer duration (P < 0.05) than cattle fed a grain diet. In forage-fed cattle, the duration they remained culture positive for E. coli O157:H7 was shorter (P < 0.05) when the diet included monensin. Generally, ruminal persistence of Nal(r) E. coli O157:H7 was not affected by diet or monensin. At necropsy, E. coli O157:H7 was detected in cecal and colonic digesta but not from the rumen. Our study showed that cattle fed a forage diet were culture positive longer and with higher numbers than cattle on a grain diet. Monensin supplementation decreased the duration of shedding with forage diet, and the cecum and colon were culture positive for E. coli O157:H7 more often than the rumen of cattle.  相似文献

选择新鲜的牛粪、猪粪和鸡粪为堆肥原料,通过室内模拟直接堆肥试验,研究堆肥过程理化参数变化、种子发芽指数以及腐熟物红外光谱特征.结果表明: 堆肥过程温度变化明显,牛粪出现一次堆肥高温阶段,鸡粪和猪粪各两次,堆肥温度超过50 ℃以上时间均大于10 d,温度可以最直观地表征堆肥的腐熟程度.堆肥过程中pH值变化幅度较大,其中猪粪的pH值从6.63上升至7.74,鸡粪从7.73上升至8.66,牛粪则先从7.86上升至8.36再下降至7.52;有机碳含量逐渐降低,牛粪、猪粪和鸡粪分别下降23.3%、28.2%和31.7%;堆肥过程中,牛粪、猪粪和鸡粪的铵态氮含量分别下降87.8%、73.6%和79.7%,硝态氮含量分别增加至堆肥前的56.81、6.49和4.85倍,铵态氮/硝态氮均下降至2以下.温度、pH值、有机碳含量和铵态氮/硝态氮能较好地反映3种粪便高温堆肥的腐熟程度,且与种子发芽率相关性较高(P≤0.05).红外光谱分析表明,在堆肥过程中,牛粪和猪粪的脂肪族、多糖类物质减少,芳香结构化增强,腐殖化程度增加;鸡粪则相反.在建立理化指标体系的同时,应兼顾物质结构的稳定性.  相似文献

Genetic diversity, isolation frequency, and persistence were determined for Escherichia coli O157 strains from range cattle production environments. Over the 11-month study, analysis of 9,122 cattle fecal samples, 4,083 water source samples, and 521 wildlife fecal samples resulted in 263 isolates from 107 samples presumptively considered E. coli O157 as determined by culture and latex agglutination. Most isolates (90.1%) were confirmed to be E. coli O157 by PCR detection of intimin and Shiga toxin genes. Pulsed-field gel electrophoresis (PFGE) of XbaI-digested preparations revealed 79 unique patterns (XbaI-PFGE subtypes) from 235 typeable isolates confirmed to be E. coli O157. By analyzing up to three isolates per positive sample, we detected an average of 1.80 XbaI-PFGE subtypes per sample. Most XbaI-PFGE subtypes (54 subtypes) were identified only once, yet the seven most frequently isolated subtypes represented over one-half of the E. coli O157 isolates (124 of 235 isolates). Recurring XbaI-PFGE subtypes were recovered from samples on up to 10 sampling occasions and up to 10 months apart. Seven XbaI-PFGE subtypes were isolated from both cattle feces and water sources, and one of these also was isolated from the feces of a wild opossum (Didelphis sp.). The number of XbaI-PFGE subtypes, the variable frequency and persistence of subtypes, and the presence of identical subtypes in cattle feces, free-flowing water sources, and wildlife feces indicate that the complex molecular epidemiology of E. coli O157 previously described for confined cattle operations is also evident in extensively managed range cattle environments.  相似文献

Environmental survival of Escherichia coli O157 may play an important role in the persistence and dissemination of this organism on farms. The survival of culturable and infectious E. coli O157 was studied using microcosms simulating cattle water troughs. Culturable E. coli O157 survived for at least 245 days in the microcosm sediments. Furthermore, E. coli O157 strains surviving more than 6 months in contaminated microcosms were infectious to a group of 10-week-old calves. Fecal excretion of E. coli O157 by these calves persisted for 87 days after challenge. Water trough sediments contaminated with feces from cattle excreting E. coli O157 may serve as a long-term reservoir of this organism on farms and a source of infection for cattle.  相似文献

An assessment of the effect of the composting potential of Mater-Bi biodegradable plastic with green wastes, noted by GBIO, and degradable plastic (PDQ-H additive) with green wastes, noted by GDEG, was carried out in a lagged two-compartment compost reactor. The composting time was determined until constant mass of the composting substrates was reached. The green wastes composting process was used as control (G). After one week of composting, the biodegradable plastics disappeared completely, while 2% of the original degradable plastic still remained after about 8 weeks of composting. A net reduction in volatile solids contents of 61.8%, 56.5% and 53.2% were obtained for G, GBIO and GDEG, respectively. Compost quality was assessed in terms of nitrogen, potassium and phosphorus contents, which were found to be highest for GBIO compost. From the phytotoxicity test, it has been observed that a diluted extract of GBIO compost has produced the longest length of radicle. From the respiration test, no significant difference in the amount of carbon dioxide released by the composting of GDEG and G was observed. This study showed that the quality of the compost is not affected by the presence of the biodegradable and degradable plastics in the raw materials.  相似文献

The possible origin of beef contamination and genetic diversity of Escherichia coli populations in beef cattle, on carcasses and ground beef, was examined by using random amplification of polymorphic DNA (RAPD) and PCR-restriction fragment length polymorphism (PCR-RFLP) analysis of the fliC gene. E. coli was recovered from the feces of 10 beef cattle during pasture grazing and feedlot finishing and from hides, carcasses, and ground beef after slaughter. The 1,403 E. coli isolates (855 fecal, 320 hide, 153 carcass, and 75 ground beef) were grouped into 121 genetic subtypes by using the RAPD method. Some of the genetic subtypes in cattle feces were also recovered from hides, prechilled carcasses, chilled carcasses, and ground beef. E. coli genetic subtypes were shared among cattle at all sample times, but a number of transient types were unique to individual animals. The genetic diversity of the E. coli population changed over time within individual animals grazing on pasture and in the feedlot. Isolates from one animal (59 fecal, 30 hide, 19 carcass, and 12 ground beef) were characterized by the PCR-RFLP analysis of the fliC gene and were grouped into eight genotypes. There was good agreement between the results obtained with the RAPD and PCR-RFLP techniques. In conclusion, the E. coli contaminating meat can originate from cattle feces, and the E. coli population in beef cattle was highly diverse. Also, genetic subtypes can be shared among animals or can be unique to an animal, and they are constantly changing.  相似文献

The effect of raw materials and their proportions in initial mixtures on organic matter (OM) stabilization and nitrogen (N) availability during pit composting in Sub-Saharan Africa was assessed using biochemical fractionation and laboratory incubations to characterize composts sampled throughout the composting process. Stabilization of OM occurred more rapidly in mixtures with slaughter-house wastes, it was progressive in mixture with household refuses while tree leaves compost remained unstable. Carbon mineralization from compost samples was positively correlated to water soluble and hemicellulose-like organic fractions. Mixtures containing large proportions of household refuses reached the highest stability and total N but available N remained weak. Slaughter-house wastes in the initial mixtures made possible to reach good OM stabilization and the largest N availability. The nature of initial mixing influenced composting parameters, OM stabilization and N availability. It is suggested mixing household refuses and slaughter-house wastes with tree leaves to reach better amending and fertilizer qualities of composts.  相似文献

The distribution of Escherichia coli O157 in bovine feces was examined by testing multiple samples from fecal pats and determining the density of E. coli O157 in immunomagnetic separation (IMS)-positive fecal samples. The density of E. coli O157 in bovine feces was highly variable, differing by as much as 76,800 CFU g(-1) between samples from the same fecal pat. The density in most positive samples was <100 CFU g(-1), the limit of reliable detection by IMS. Testing only one 1-g sample of feces per pat with IMS may result in a sensitivity of detection as low as 20 to 50%. It is therefore probable that most surveys have greatly underestimated the prevalence of E. coli O157 shedding in cattle and the proportion of farms with shedding cattle. The sensitivity of the detection of E. coli O157 in bovine feces can be as much as doubled by testing two 1-g samples per pat rather than one 1-g sample.  相似文献

The study aimed to quantify and compare the incremental exposure of root crops, at point of harvest, to enteric pathogens from untreated vs. composted spent broiler litter (SBL)/bagasse mix in field-crop application. An exposure assessment based on the Source-Pathway-Receptor approach was developed for bacterial indicator species, total coliforms, faecal coliforms, Escherichia coli and faecal enterococci. Event trees were constructed to model the pathways leading to the partitioning of pathogens present in the SBL blend during composting and after land application. The main barriers are induction of composting, high-rate thermophilic phase, maturation phase, and, decay and dilution of the indicator pathogens in the soil. The computed exposures have been expressed in terms of the arithmetic mean. TC, FC, E. coli and FE levels on root crops were reduced to very remote fractions of 0.01826, 0.00046, 0.000132 and 0.000013 kg(-1), respectively. The degree of by-pass (pi) of the treatment at operational scale showed that less than 1-log reduction has been by-passed during each turning event, revealing the effectiveness of turning for process control. The predicted E. coli counts on root crops at point of harvest provided a basis for estimating the exposure potential by the beta-Poisson model. Probability of exposure was 0.782 for raw SBL mix compared to 1.40x10(-11) with composting. It can be concluded that there is a definite advantage in optimally composting SBL mix before land application. The exposure assessment may essentially require modification and fine tuning as and when further data become available.  相似文献

由废弃地整理复垦形成的耕地存在土壤有机质和有效养分低、土壤板结、微生物活性弱和土壤耕作性状不良等问题,快速、有效地提高土壤肥力质量是全面提升该类耕地质量和生产性能的重要组成部分.本文通过田间小区试验研究了城郊有机废弃物对新复垦耕地土壤培肥的综合效果,并比较了不同类型城郊有机废弃物在培育耕地质量方面的差异.试验设置了施用等量猪粪、鸡粪、水稻秸秆、蔬菜收获残留物、城市污泥、沼渣、猪粪/水稻秸秆堆肥、生活垃圾堆肥和对照(不施有机肥)9个处理(年用量30 t·hm-2),连续进行3年的定点试验.结果表明: 施用任何有机物对改善土壤肥力均有明显的作用.其中,提升土壤碳库管理指数以施用猪粪、鸡粪、猪粪/水稻秸秆堆肥、水稻秸秆和沼渣的效果最为显著;增加土壤水稳定性团聚体和降低土壤容重以施用猪粪/水稻秸秆堆肥和沼渣的效果最佳;施用污泥、猪粪/水稻秸秆堆肥和生活垃圾堆肥可增强土壤保蓄能力;施用猪粪、鸡粪和猪粪/水稻秸秆堆肥对增加土壤有效态养分的效果最为明显;各类有机物均显著提高了土壤微生物数量和酶活性.长期施用污泥、生活垃圾堆肥及畜禽粪存在着土壤重金属污染的风险,但短期施用对土壤环境质量影响不明显.总体上,对土壤肥力的改善效果由大至小依次为:猪粪/水稻秸秆堆肥>鸡粪>猪粪>沼渣>生活垃圾堆肥>水稻秸秆>城市污泥>蔬菜收获残留物;对土壤的相对污染程度由大至小为:城市污泥>生活垃圾堆肥>猪粪>鸡粪>沼渣>猪粪/水稻秸秆堆肥>蔬菜收获残留物>水稻秸秆.  相似文献

试验采用单因子随机分组试验设计,测定了施用复合微生物制菌剂Y30017后对蛋鸡舍内有害气体、鸡粪便排泄物成分的影响,分析该复合微生物菌剂对规模化养鸡场鸡舍环境净化的效果。结果表明:试验组鸡舍氨气浓度和硫化氢浓度分别下降了2.26 mg/kg和1.92 mg/kg(P<0.05);鸡粪排泄物pH值上升1.1%(P<0.05),粗蛋白含量提高了9.17%(P<0.05);每克盲肠内容物中大肠杆菌数量降低了37.17%(P<0.01),乳酸杆菌和双歧杆菌数量分别提高了11.49%和16.17%(P<0.05),未检出沙门氏菌。结果显示,鸡舍主要污染物得到有效控制,养殖环境得到较大改善,使用复合微生物菌剂Y30017控制规模化养鸡场污染物均具有良好的效果,同时可有效提高粪便废弃物的营养值,对养殖废弃物资源化利用具有良好的提质增效作用。  相似文献

Acid resistance (AR) is important to survival of Escherichia coli O157:H7 in acidic foods and may play a role during passage through the bovine host. In this study, we examined the role in AR of the rpoS-encoded global stress response regulator sigma(S) and its effect on shedding of E. coli O157:H7 in mice and calves. When assayed for each of the three AR systems identified in E. coli, an rpoS mutant (rpoS::pRR10) of E. coli O157:H7 lacked the glucose-repressed system and possessed reduced levels of both the arginine- and glutamate-dependent AR systems. After administration of the rpoS mutant and the wild-type strain (ATCC 43895) to ICR mice at doses ranging from 10(1) to 10(4) CFU, we found the wild-type strain in feces of mice given lower doses (10(2) versus 10(3) CFU) and at a greater frequency (80% versus 13%) than the mutant strain. The reduction in passage of the rpoS mutant was due to decreased AR, as administration of the mutant in 0.05 M phosphate buffer facilitated passage and increased the frequency of recovery in feces from 27 to 67% at a dose of 10(4) CFU. Enumeration of E. coli O157:H7 in feces from calves inoculated with an equal mixture of the wild-type strain and the rpoS mutant demonstrated shedding of the mutant to be 10- to 100-fold lower than wild-type numbers. The survival of Escherichia coli O157:H7 in feces from steers fed corn (CO) or barley (BA) was evaluated at -10, +4 and +22 degrees C. Fecal pats were inoculated with a four-strain mixture of nalidixic-acid resistant E. coli O157:H7 at two levels: 10(3) CFU g(-1) (low, L) and 105 CFU g(-1) (high, H). At -10 degrees C, duration of survival of E. coli O157:H7 was reduced (p < 0.05) in CO-L (35 days) compared to BA-L (49 days), likely due to the effects of fecal volatile fatty acids in combination with a fecal pH of <6.5. At 4 degrees C, E. coli O157:H7 was detected in BA-H, CO-H, CO-L and BA-L for 77, 77, 56 and 63 days, respectively, with no difference (p > 0.05) observed in the duration of survival or rate of decline of E. coli O157:H7 among treatments. Survival of E. coli O157:H7 was twice as likely (p < 0.05) at 22 degrees C than at 4 degrees C and -10 degrees C. While pH and dry matter content increased, and volatile fatty acid concentrations decreased over 84 days at all three temperatures, these changes were most pronounced at 22 degrees C. Survival of E. coli O157:H7 for extended periods of time in feces from both corn- and barley-fed animals was demonstrated, thus fecal material may serve as a vector for the transmission of the organism. The greater survival of E. coli O157:H7 at 22 degrees C suggests that temperature may play a role in the seasonality of transmission and prevalence of this bacterium in feedlot cattle. The reported greater prevalence of E. coli O157:H7 in cattle fed barley as compared to those fed corn does not appear to be related to elevated risk of transmission arising from differential survival of the bacterium in feces.  相似文献
