为了给鲤鱼遗传资源保护及利用提供较为准确的基础数据,利用10对微卫星标记分析了黄河鲤(Huanghe Cyprinus carpio,人工养殖群体)、长江野鲤(Changjiang wild Cyprinus carpio,安徽段)、安徽养殖鲤(Anhui cultured Cyprinus carpio,宿州市)群体遗传多样性。遗传多样性分析实验表明:3个群体均具有较高的遗传水平,且长江野鲤群体遗传多样性高于黄河鲤、安徽养殖鲤;卡方检验表明:3个群体在较大程度上受到人类活动及生态环境变化的影响,30个位点中76.67%的位点显著偏离Hardy-Weinberg平衡。遗传距离、遗传相似度、非加权组平均法(unweighted pair-group method with arithmetic means,UPGMA)聚类树及贝叶斯聚类分析均显示,长江野鲤与安徽养殖鲤亲缘关系较近,研究情况与实际生产中养殖户从长江里获得鲤鱼亲本用于繁殖苗种的现象相符。突变-漂移平衡分析显示,在TPM模式下,Wilcoxon符号秩次检验中黄河鲤群体显著偏离突变-漂移平衡,需进一步扩大选育群体。总体来说,黄河鲤、长江野鲤、安徽养殖鲤群体遗传多样性均较高,具有进一步的选育空间。 相似文献

该文从200个微卫星标记中筛选出149个多态性标记,并对镜鲤(Cyprinus carpio)子代个体数最多的家系进行了肌间刺数量的相关性分析。结果表明有8个微卫星标记与肌间刺数量显著相关(P<0.05)。其中,HLJ3086、HLJ2642及HLJ3515与肌间刺数量极显著相关(P<0.01)。多重比较同一标记不同基因型,得到与肌间刺数量相关的基因型。将得到的与肌间刺数量显著相关微卫星标记在NCBI上进行BLAST比对,结果显示HLJ2891与斑马鱼编码蜘蛛毒素亲和蛋白-2(latrophilin-2-like)基因相似,一致度达92%,HLJ3515与斑马鱼编码丝氨酸/苏氨酸激酶32B(serine/threonine-protein kinase 32B-like)基因相似,一致度达81%。该结果为镜鲤肌间刺数量的分子标记辅助育种(molecular marker assisted breeding)提供了有效依据。 相似文献

实验选取8 个多态性微卫星位点, 用于虾夷扇贝4 个不同壳色全同胞和半同胞家系160 个子代的亲权鉴定。在亲本未知和一亲本已知的情况下, 8 个微卫星位点累积排除概率分别为0.823 和0.961。鉴于亲权分析时子代的亲本在已知和未知情况下位点的累积排除概率不同, 实验采用了两种方法用于家系的亲权鉴定。方法1: 当子代的父母本情况未知时, 根据子代基因型数据, 通过CERVUS 2.0 软件计算子代所对应的候选亲本的LOD 值, 直接将候选亲本中具有最大的两个LOD 值的亲本确定为子代的父母本; 方法2: 在子代的亲本未知情况下, 视具有最大LOD 值的候选亲本为子代的第一候选亲本, 然后将该亲本视为已知, 通过CERVUS 2.0 软件重新计算每个侯选亲本的LOD 值, 再从中选择具有最大LOD 值的候选亲本作为该子代的第二候选亲本。结果表明, 采用方法2 得到的家系亲权鉴定成功率达到95%以上, 确定了微卫星标记在虾夷扇贝家系鉴定中的可行性。所检测的微卫星位点在子代中出现无效等位基因现象, 而无效等位基因存在会引起子代与亲本的错配。实验在家系鉴定时采用了无效等位基因存在(情况1)和缺失(情况2)两种子代基因型文件进行分析, 不同情况下同一方法家系鉴定成功率相差无几。这表明了基于多个微卫星位点计算候选亲本LOD 值大小寻找子代真实父母本可以降低由无效等位基因引起的错配的几率。研究表明了微卫星标记适合于不同壳色虾夷扇贝家系亲权鉴定工作。

经多重PCR扩增20个微卫星标记,对凉山半细毛羊进行亲权鉴定。荧光标记检测扩增产物,并用Cervus 2.0分析。结果表明:当双亲未知时,累积排除概率为0.998666 ,在父系半同胞基础上进行母子亲权鉴定,累积排除概率为0.999994,置信度为95%,最终从8个候选父亲中找出215个后裔的父亲,又从147个候选母亲中找出它们的母亲,构建了一个混合家系系谱,为进行连锁分析、QTL定位奠定了基础,并对研究绵羊群体结构、历史及多样性有重要意义。Abstract: The usefulness of multiplex polymerase chain reaction was evaluated and fluorescent detection with 20 microsatellites was used for paternity testing of Liangshan semi-fine wool sheep. The results in Cervus 2.0 indicated that the combined exclusion probability was 0.998666 when parents were both unknown. If father was known, combined exclusion probability was 0.999994, with confidence level of 95%. Finally, the biological parents of 215 individuals in offspring were found from 8 candidate fathers and 147 candidate mothers, and a mixed pedigree was constructed. It is useful for linkage analysis of sheep chromosomes and QTL location, also for the study of population structure, history, and diversity. 相似文献

微卫星DNA标记探讨镜鲤的种群结构与遗传变异 总被引:15,自引:2,他引:15
采用30个微卫星分子标记, 对5个镜鲤群体的观测杂合度(Ho)、期望杂合度(He)、多态信息含量(PIC)和有效等位基因数(Ae)等进行了遗传检测, 根据基因频率计算遗传相似系数和Nei氏标准遗传距离, 以c2检验估计Hardy-Weinberg平衡, 以近交系数(FST)和基因流(Nm)分析群体的遗传分化。同时, 使用PHYLIP3.63软件绘制基于Nei氏标准遗传距离的UPGMA聚类图, 并进行bootstrap自举检验验证进化树的可靠性。在德国镜鲤选育系(Scattered Cyprinus carpio L.)和来自4个不同养殖场(松浦、东岗、奉城和辽中)的德国镜鲤群体中共检测到7 083个扩增片段, 长度在102 ~ 446 bp之间, 在群体内扩增出等位基因1~16个不等, 共计356个等位基因。结果表明: (1)5个群体检测的有效等位基因数在1.07~12.30个不等, 平均多态信息含量为0.74、0.74、0.69、0.75和0.75, 无偏期望杂合度的平均值为0.74、0.78、0.70、0.76和0.78, 说明这几个群体属于高度多态, 遗传多样性水平较高。(2)群体间相似系数在0.52以上, 相似性较高。聚类分析显示, 东岗、奉城和辽中3个养殖场的德国镜鲤群体聚类成一个分支, 而德国镜鲤选育系与松浦群体聚类成另一分支。聚类的先后与它们在地理分布上距离远近有一定的相关性。(3)在与功能基因相关的多个微卫星基因座位上, 扩增产物呈现不同程度的缺失现象, 这些无效等位基因的产生可能与结构基因在育种中受到人工选择的影响较大有关。 相似文献

2017年7-8月和11-12月在贺兰山岩画风景区采集到369份岩羊新鲜粪便样本,用多态性较高的10个微卫星位点进行基因分型,共识别出282只岩羊个体。通过复合扩増和牙釉蛋白基因PCR法共鉴定出152只雄性,130只雌性。微卫星分析得到平均等位基因(Na)为10.1,平均观测杂合度(Ho)为0.7737,平均期望杂合度(He)为0.6512,平均多态信息(PIC)为0.6129。运用Cervus3.0软件进行亲权鉴定,并用Kingroup V2计算个体间的亲缘系数(r),作为亲权鉴定的补充,共鉴定出父-母-子7对,父-子21对,母-子19对。根据亲缘关系对岩羊的婚配制进行推测分析,表明岩羊婚配制应属一雄多雌。 相似文献

2014年4月至2015年1月,在古田山国家级自然保护区内共收集634份粪便样本,2份肌肉样本。通过严格筛选,最终获得390份可用于PCR扩增的样本。用多态性较高的8个微卫星位点进行基因型分型,共识别出177个小麂个体。SRY基因性别鉴定显示研究样本中雄性84只,雌性93只。所使用的8个微卫星位点在177个样本中,平均等位基因数(A)为11,平均观测杂合度(Ho)在0.960—1.000之间,平均值为0.9685,平均期望杂合度(He)在0.799—0.887之间,平均值为0.8429,多态信息含量(PIC)在0.766—0.872之间,平均多态信息含量为0.8214,基因杂合度水平较高,为遗传多样性丰富的种群。采用Cervus3.0进行亲权分析,当置信度为95%和80%时,8个微卫星位点的鉴定率均达到100%。共鉴定出父-母-子24对,母-子23对,父-子19对,涉及到104只个体。根据亲缘关系分析小麂的婚配制,结果发现小麂的婚配制属于1雄多雌,但并不是目前所知的亚型,而可能是一种被称作"检查策略"的一雄多雌制。 相似文献

关于鲤属鱼类的分类,过去已有两次比较系统的研究,但对亚属的划分、种下阶元的判定等仍有争议。原因是分类所循的外形性状变异较大,且多互相重叠。本文试从骨骼比较入手,寻找新的、较为稳定的分类依据。经比较分析,鲤属鱼类的第一脊椎横突、上颌骨、上筛骨、额骨、枕骨突、犁骨、下咽骨和下颌骨等存在明显差异,可作为划分亚属和种的鉴别特征。据此重新分类整理,认为Mesocyprinus亚属仍能成立,首次把产于洱海的厚唇鲤cyprinus crassilabris Chen et Hwang归入杞麓鲤Cyprinus carpio chilia Wu et al.的同物异名;Cyprinus pellegrini pellegrini Tchang,Cyprinus pellegrini barbatus Chen et Hwang,Cyprinns yunnanensis yunnanensis Tchang和Cyprinus yunnanensis daliensis Chen et Hwang在骨骼上有明显分化,由亚种提升为各自独立的种。整理后云南共有鲤属鱼类12个种和亚种,以骨骼特征做出检索表。 相似文献

Influences of traditional Chinese medicine on non-specific immunity of Jian Carp (Cyprinus carpio var. Jian) 总被引:2,自引:0,他引:2
The influence of traditional Chinese medicine (TCM) formulation from Astragalus Root (Radix astragalin seu Hedysari) and Chinese Angelica Root (R. Angelicae Sinensis) at a ratio of 5:1 (w/w) on non-specific immunity of Jian carp, Cyprinus carpio var. Jian was investigated. The number of NBT-positive cells in the blood and lysozyme and complement activities in the serum of Carp fed with commercial feed supplemented with 1.0% (diet 1) and 1.5% (diet 2) TCM at 10 day of post-feeding were not different from those of the control group fed with feed unsupplemented TCM 10 days post-feeding (P>0.05), but at 20 and 30 days they increased significantly (P<0.05). The values of diet 1 group and diet 2 group at 20 day and at 30 day were not significantly different (P>0.05) from each other. In addition, the TCM formula increased body weight of experimental fish by about 16.84% (diet 1) and 19% (diet 2) above that of the control group. Therefore, these data suggest that the TCM formula could elevate the function of non-specific immunity of Jian carp. The optimal dosage added to commercial carp feed was 1.0% (w/w) and the oral administration time as a course of treatment was 20 days. 相似文献

在基础饲料中添加0 g/kg、0.8 g/kg、1.2 g/kg、1.6 g/kg、2.0 g/kg、2.4 g/kg饲料的香菇多糖,连续饲喂幼建鲤Cyprinus carpio var.Jian 49 d后,检测香菇多糖对幼建鲤红细胞、白细胞数量及白细胞杀菌指数、血清溶菌酶活性、血清SOD活性及IgM含量的影响。结果表明,试验第49 d时,不同添加剂量的香菇多糖对各组实验鱼白细胞、红细胞数量影响不大,与对照组无显著差异(P>0.05)。在饲料中添加不同剂量香菇多糖后对幼建鲤白细胞杀菌指数、血清溶菌酶活性、血清SOD活性及IgM含量有不同程度影响。试验期间,0.8 g/kg、1.2 g/kg、1.6 g/kg、2.0 g/kg和2.4 g/kg组试验幼建鲤白细胞杀菌活性比对照组明显增强,差异极显著(P<0.01);血清溶菌酶活性逐渐升高,1.6 g/kg、2.0 g/kg和2.4 g/kg组极显著高于对照组和0.8 g/kg组(P<0.01),当香菇多糖添加量增加到1.6 g/kg时,对血清溶菌酶活力增加不显著(P>0.05)。在饲料中添加不同剂量香菇多糖后对幼建鲤血清SOD活性有不同程度影响。2.0 g/kg添加组对SOD增加明显,与对照组、0.8 g/kg、1.2 g/kg组差异极显著(P<0.01),与2.4 g/kg组差异显著(P<0.05)。试验期间,0.8 g/kg、1.2 g/kg、1.6 g/kg、2.0 g/kg和2.4 g/kg组试验幼建鲤血清IgM含量变化与对照组相比变化不明显,各组间差异不显著(P>0.05)。 相似文献

This study aimed to investigate the effects of threonine (Thr) on the digestive and absorptive ability, proliferation and differentiation of enterocytes, and gene expression of juvenile Jian carp (Cyprinus carpio var. Jian). First, seven isonitrogenous diets containing graded levels of Thr (7.4–25.2 g/kg diet) were fed to the fishes for 60 days. Second, enterocyte proliferation and differentiation were assayed by culturing enterocytes with graded levels of Thr (0–275 mg/l) in vitro. Finally, enterocytes were cultured with 0 and 205 mg/l Thr to determine protein synthesis. The percent weight gain (PWG), specific growth rate, feed intake, feed efficiency, protein retention value, activities of trypsin, lipase and amylase, weights and protein contents of hepatopancreas and intestine, folds heights, activities of alkaline phosphatase (AKP), γ- glutamyl transpeptidase and Na+/K+-ATPase in all intestinal segments, glutamate-oxaloacetate transaminase (GOT) and glutamate-pyruvate transaminase (GPT) activities in hepatopancreas, and 4E-BP2 gene expression in muscle, hepatopancreas and intestinal segments were significantly enhanced by Thr (p<0.05). However, the plasma ammonia concentration and TOR gene expression decreased (p<0.05). In vitro, Thr supplement significantly increased cell numbers, protein content, the activities of GOT, GPT, AKP and Na+/K+-ATPase, and protein synthesis rate of enterocytes, and decreased LDH activity and ammonia content in cell medium (p<0.05). In conclusion, Thr improved growth, digestive and absorptive capacity, enterocyte proliferation and differentiation, and protein synthesis and regulated TOR and 4E-BP2 gene expression in juvenile Jian carp. The dietary Thr requirement of juvenile Jian carp was 16.25 g/kg diet (51.3 g/kg protein) based on quadratic regression analysis of PWG. 相似文献

Microsatellite markers of the poly (CA) type in common carp ( Cyprinus carpio L.) are described. Clones containing a (CA) repeat were isolated from a common carp genomic library and sequenced. The number of repeats found was high compared to mammals but comparable with other teleost fishes. Classification of the repeats (perfect, imperfect and compound) are compared with the Atlantic cod ( Gadus morhua L.), rainbow trout ( Oncorhynchus mykiss ), and Atlantic salmon ( Salmo salar L.). A total of 41 primer sets were designed and tested for polymorphism on a test panel of eight animals (derived from outbred lines, inbred lines and gynogenetic clones). Thirty-two markers were found to be polymorphic. The heterozygosity in the outbred animals was 60·4%, 51·1% in the inbred animals and 0% in the gynogenetic clones. The average number of alleles among the eight animals was 4·7 per marker. Six markers (18·8%) gave an additional polymorphic amplification product besides the polymorphic amplification product in the expected size range. The possibility that these loci are tetraploid is discussed. The polymorphic loci described for common carp will be valuable as genetic markers for use in population, breeding, and evolutionary studies. 相似文献

ABSTRACT: BACKGROUND: The wild boar (Sus scrofa) is among the most widespread mammal species throughout the old world. Presently, studies concerning microsatellites in domestic pigs and wild boars have been carried out in order to investigate domestication, social behavior and general diversity patterns among either populations or breeds. The purpose of the current study is to develop a robust set of microsatellites markers for parentage analyses and individual identification. FINDINGS: A set of 14 previously reported microsatellites markers have been optimized and tested in three populations from Hungary, Portugal and Spain, in a total of 167 samples. The results indicate high probabilities of exclusion (0.99999), low probability of identity (2.0E-13 -- 2.5E-9) and a parentage assignment of 100%. CONCLUSIONS: Our results demonstrate that this set of markers is a useful and efficient tool for the individual identification and parentage assignment in wild boars. 相似文献

利用30个微卫星分子标记对海南鲤(HN)、长江鲤(CY)、月亮湖鲤(YL)、黑龙江鲤(FY)、呼伦湖鲤(ZL)、贝尔湖鲤(BR)6个野生鲤鱼群体的观测杂合度(Ho)、期望杂合度(He)、多态信息含量(PIC)和有效等位基因数(Ae)等进行了遗传检测,根据基因频率计算遗传相似系数和Nei氏标准遗传距离,χ2检验估计Hardy-Weinberg平衡,用近交系数(FST)和基因流(Nm)分析群体的遗传分化及其来源。同时,使用PHYLIP3.63软件绘制基于Nei氏标准遗传距离的UPGMA进化树。6个群体共检测到8,136个扩增片段,长度在125bp~414bp,30个基因座扩增出等位基因数从3~13个不等,共计210个等位基因,平均每个基因座扩增得到7个等位基因。结果显示:(1)6个野鲤群体的多态性指标均适中,多态信息含量依次0.44、0.52、0.53、0.57、0.63和0.64,有效等位基因数1.04~4.72个不等,平均有效等位基因数依次为2.19、2.60、2.42、2.43、2.45和2.33,无偏期望杂合度平均值为0.50、0.59、0.56、0.56、0.57和0.54;(2)遗传相似系数BR与ZL最高(0.8511),BR与HN最低(0.6688),聚类结果与地理分布呈一定相关性。 相似文献

1. The invasion of carp (Cyprinus carpio L.) in Australia illustrates how quickly an introduced fish species can spread and dominate fish communities. This species has become the most abundant large freshwater fish in south‐east Australia, now distributed over more than 1 million km2. 2. Carp exhibit most of the traits predicted for a successful invasive fish species. In addition, degradation of aquatic environments in south‐east Australia has given them a relative advantage over native species. 3. Derivation of relative measures of 13 species‐specific attributes allowed a quantitative comparison between carp and abundant native fish species across five major Australian drainage divisions. In four of six geographical regions analysed, carp differed clearly from native species in their behaviour, resource use and population dynamics. 4. Climate matching was used to predict future range expansion of carp in Australia. All Australian surface waters appear to be climatically suitable for carp. 5. This assessment strongly reinforces the need for immediate management of carp in Australia to include targeted control of human‐assisted dispersal, such as use of carp as bait by anglers, distribution to new locations by anglers and the use of the ‘Koi’ strain in the aquarium industry. 6. Given their historical spread, dispersal mechanisms and ecological requirements, the expansion of carp across most of the remainder of Australia is to be expected. 相似文献
