The interaction of main potato glycoalkaloids alpha-solanine and alpha-chaconine in inhibition of horse serum butyryl cholinesterases immobilized on the pH-sensitive field-effect transistors has been investigated. The method of isobol diagram of Loewe and Muishnek has been used for interpretation of results. It has been shown the alpha-chaconine inhibits the immobilized bytyryl cholinesterases more strongly than alpha-solanine, and their mixture has the addition effect.  相似文献

Butyryl cholinesterase from horse and human sera catalyzed the hydrolysis of monoacylglycerols containing fatty acids varying in chain length from 8 to 12 carbons; maximum activity was obtained with rac-glycerol 1-monodecanoate as substrate. Neither the triacylglycerols of these fatty acids nor the monoacylglycerols of longer chain length fatty acids were hydrolyzed at measurable rates in the system used. The enzyme was eserine sensitive and indistinguishable from butyryl cholinesterase as judged by purification, response to the several inhibitors tested, and heat inactivation. Data from mixed substrate experiments suggest a possible effector role for butyryl choline in accelerating the rate of rac-glycerol 1-monodecanoate hydrolysis. Fatty acid released during the course of rac-glycerol 1-monodecanoate hydrolysis may irreversibly inactivate the enzyme.  相似文献

Butyryl cholinesterase (BChE) has been seen as a key enzyme in the search for new strategies in the treatment of poisoning by organophosphates (OPs), since human BChE (HssBChE), complexed with the appropriate oxime, can be a suitable scavenger and deactivator for OPs in the blood stream. However, the efficacy of HssBChE is limited by its strict stoichiometric scavenging, slow reactivation, and propensity for aging. The improvement of the reactivation rate by new and more efficient oximes could contribute to mitigate this problem and increase the HssBChE efficiency as scavenger. Several oximes have been synthesized and tested with this goal, some with promising results, but the mechanistic aspects of the reactivation reaction are not fully understood yet. In order to better investigate this mechanism, docking and mixed quantum and molecular mechanics combined with principal components analysis were performed here to evaluate the capacity of reactivation and determine the preferred route for the reactivation reaction of two new oximes on HssBChE inhibited by the neurotoxic agents cyclosarin and sarin. Plots of potential energies were calculated and all the transition states of the reactional mechanism were determined. Our results showed a good correlation with experimental data and pointed to the most efficient oxime with both OPs. The protocol used could be a suitable tool for a preliminary evaluation of the HssBChE reactivation rates by new oximes.  相似文献

The cerebellum is subdivided into hundreds of discrete modules defined by their connectivity and molecular signatures. Cerebellar compartmentation arises very early in development through the formation of multiple populations of chemically distinct Purkinje cells that migrate in a coordinated fashion to form parasagittal bands of cells. Different Purkinje cell bands are then innervated by discrete subpopulations of cerebellar afferents. Because of its stereotyped and strikingly beautiful organization the cerebellum is an excellent model in which to explore genetic/epigenetic aspects of pattern formation in the central nervous system.  相似文献

Using 31P-NMR studies we have observed that 1. 2-Deoxyglucose leads into the brain in vivo and in superfused cortical slices in vitro to a maximum concentration at between 45 and 60 min, when 80% of the material is in the phosphorylated form. 2. The phosphorylated DOG6P disappears from the n.m.r. spectra with a half-life of ca 130 min. 3. Two resonances of DOG6P are observed in the actively metabolising tissue, whereas only one is visible in deproteinised tissue extracts. This suggests that the DOG6P is in two separate compartments which differ in pH. 4. Compartmentation between mitochondria, nerve endings and cytoplasm was concluded to be unlikely from subcellular fractionation studies, but the possibility of compartmentation between neurones and glia could not be so clearly assessed.  相似文献

Compartmentation is essential for the localization of biological processes within a eukaryotic cell. ATP synthase localizes to organelles such as mitochondria and chloroplasts. By contrast, little is known about the subcellular distribution of CTP synthase, the critical enzyme in the production of CTP, a high-energy molecule similar to ATP. Here I describe the identification of a novel intracellular structure con-taining CTP synthase, termed the cytoophidium, in Drosophila cells. I find that cytoophidia are present in all major cell types in the ovary and exist in a wide range of tissues such as brain, gut, trachea, testis, accessory gland, salivary gland and lymph gland. In addition, I find CTP synthase-containing cytoophidia in other fruit fly species. The observation of compartmentation of CTP synthase now permits a broad range of questions to be addressed concerning not only the structure and function of cytoophidia but also the organization and regulation of CTP synthesis.  相似文献

The life-history stages of Ganeo tigrinus Mehra & Negi, 1928 infecting the Indian bull frog Hoplobatrachus tigerinus (Daudin) are described, those from cercaria to egg-producing adult having been established in the laboratory. Non-virgulate xiphidiocercariae are released by the planorbid snail Indoplanorbis exustus (Deshayes). Metacercariae occur in the haemocoel of dragonfly nymphs and become infective to the frog H. tigerinus within 15 days. The pre-patent period is 45 days. Growth and development of both metacercariae and adults are described in detail. Comments on the systematic position of Ganeo Klein, 1905 are included.  相似文献

Individual amino acid levels and compartmentation in chick blood were measured on day 20 of incubation, at hatching (day 0), or after 1 or 5 days of free life, and compared with those of adult chickens. Blood cell amino acid concentrations were almost one order of magnitude higher than those of plasma, with higher values than those found in mammalian erythrocytes. This difference may be due to the capability for protein synthesis of the nucleated cells coupled with a postulated utilization of amino acids as fuel. The most common pattern of individual plasma amino acid levels was a slight rise at hatching followed by a large decrease, with minimal values for adults. The patterns in the cells did not always coincide with those for plasma. Total blood amino acid levels increased steadily during the period studied due to the increase in intracellular amino acids, giving rise to increasing blood-cell/plasma concentration ratios. These changes showed higher availability of plasma amino acids just after hatching, while the cell concentrations increased steadily to the maximum values in adults. The increase in alanine levels in cells with little changes in plasma can be correlated with the role of this amino acid as the main 2-amino nitrogen carrier in the avian bloodstream. The high amino acid levels in the cells suggest that these cells act as inter-organ transporters and reservoirs of amino acids, they have a different role in their handling and metabolism from those of mammals.  相似文献

Cannibalism is a recurrent behavior across the animal kingdom, with important ecological and evolutionary consequences due to its potential trade-offs on the fitness of involved individuals and demography dynamics. Cannibalism has been reported in eight species of several phylogenetic lineages within Liolaemus, a highly diverse genus of Neotropical lizards. Within the Liolaemus montanus series, two species are reported to eat juveniles in captivity. We observed a female Liolaemus orientalis, another L. montanus series member, which had ingested a juvenile of its own species in the wild. As it generally happens with other lizards, cannibalism in Liolaemus has been suggested to be performed by the larger sex, with L. chiliensis being the only other case observed until now in which the cannibalistic individual was a female. The fact that females are slightly smaller than males in L. orientalis means that our observation is an exception to previously observed trends. We suggest that there is a relationship between this behavior and the presence of a masculine trait in the cannibalistic female: developed precloacal pores. We also discuss how refuge availability may influence demography and the probability of dispersing juveniles encountering adults, increasing the chances for cannibalism to play some role in population density regulation.  相似文献
