Multi drug resistance (MDR) or cross resistance to drugs was initially explained on the basis that MDR cells express drug transporters that expel membrane-embedded drugs. However, it is now clear that transporters are a single piece from a complex puzzle. An issue that has been solved recently is, given that these transporters have to handle drugs, why should a membrane-embedded drug and a transporter meet? To solve this problem, a theory has been suggested considering the interaction between the cell membrane mechanical properties and the size of drugs. In simple terms, this theory proposes that an excess in the packing of lipid in the inner leaflet of the membrane of MDR cells is responsible for blocking drugs mechanically as a function of their sizes at the membrane level, thus impairing their flux into the cytosol. In turn it is expected that this would allow any membrane embedded drug to diffuse toward transporters. The study concluded that the size of drugs is necessarily important regarding the mechanical interaction between the drug and the membrane, and likely to be central to MDR. Remarkably, an experimental study based on MDR and published years ago concluded that the molecular weight (MW) of drugs was central to MDR (Biedler and Riehm in Cancer Res 30:1174–1184, 1970). Given that size and MW are linked together, I have compared the former theory to the latter experimental data and demonstrate that, indeed, basic membrane mechanics is involved in high levels of cross resistance to drugs in Pgp expressing cells.  相似文献

Corneal epithelial differentiation (primary stroma production) is dependent on the underlying extracellular matrix (ECM), for if the developing epithelium is enzymatically removed from the embryo, it fails to produce stroma in vitro unless it is cultured on collagenous ECM. We have previously shown that the stimulatory effect is mediated across Nucleopore filters in direct proportion to the surface area created by epithelial cell processes traversing the filter to contact ECM. Since collagenous ECM is insoluble under physiological conditions, transfilter stimulation of stroma production is probably due to an interaction of the epithelial cell surface with “inducer” ECM (killed lens capsule or purified collagen). We grew 5-day-old corneal epithelia on Nucleopore filters atop [3H]proline-labeled lens capsules and used both autoradiography and scintillation counting to show that radioactive collagen does not enter the epithelial cells in detectable amounts. We also show here that the stimulatory effect of collagen on collagen synthesis is not dependent on trapping of serum or binding of conditioned medium factors by ECM. Finally, we demonstrate that the stimulatory effect is reduced by removal of transfilter ECM after 6–12 hr in vitro. By 18–24 hr, however, cultured epithelium is less dependent on the substratum, probably because it has produced its own ECM. We conclude that: (1) the contact mediated collagen-cell surface interaction under study here requires the continuous presence of collagen in vivo and in vitro for maintenance of “stimulated” epithelial stroma synthesis; (2) the collagenous “inducer” interacts directly with epithelium rather than indirectly via trapped intermediates; (3) collagen acts at the epithelial cell surface without entering the cells.  相似文献

It is well established that the intraparietal sulcus (IPS) plays an important role in the processing and representation of numerical magnitude. Two recent studies by Piazza et al. and Cohen Kadosh et al. published in this issue of Neuron used fMRI adaptation to explore the extent to which parietal number processing is dependent upon or independent of a numbers' surface format. Forty-one accessions of the genusBeta representing wild and cultivated species of all sections were analyzed by DNA fingerprinting. Four sugar beet minisatellite DNA probes revealed characteristic banding patterns with Southern-hybridizedBeta DNA restricted withHindIII. A total of 111 polymorphic RFLP bands were scored across all accessions. Cluster analysis based on genetic similarity estimates for all 820 combinations of accessions revealed the following results. (1) All accessions could unambiguously be identified by a characteristic RFLP banding pattern. (2) The sugar beet cultivars examined displayed a low level of genetic diversity; they showed high similarity toB. Vulgaris ssp.maritima but low genetic similarity to the other wild species of section I. (3) In most cases, the present taxonomic classification of the genusBeta was confirmed. Species of sections II, III, and IV were clearly distinguishable from those of section I except forB. Macrocarpa, which showed high similarity to wild species of section II. In a second experiment, 108 single-copy RFLP probes from sugar beet were Southern hybridized withB. procumbens DNA. A surprisingly low degree of homology (34%) was found. The results are discussed with regard to the taxonomic classification of the genusBeta.  相似文献

“Bulk density” and “dry-matter content” are useful indices of dry-matter accumulation in plant organs. A theoretical equation describing the relationship between these two indices was put forward. To examine the reliability of this equation, the seasonal changes of these two indices were investigated in the leaves and stems of different ages ofAucuba japonica. In each organ both indices varied seasonally almost parallel to each other, but the seasonal changes of dry-matter content were less obvious than those of bulk density. The observed bulk density was always larger than that calculated from the observed dry-matter content by the theoretical equation. A drying experiment showed that this discrepancy was caused by the decrement of the volume of the plant material by water loss during the period from the weight measurement to the volume measurement. When the water loss was negligible, the equation described well the relationships between the two indices of the experimental materials. It was also shown that this equation was useful for the estimation of the amount of air space in plant materials.  相似文献

Impurities of free aromatic amino acids (Phe and Tyr) and the elastin protein were found in the heparin commercial drug (Hep) by spectral luminescent and spectrophotometric methods. The fluorescence quenching of the Trp, Tyr, and Phe amino acids by the Hep drug was studied, and the Stern-Folmer constants (K) that reflected stability of the Hep complexes with amino acids were determined. The stability of AA-Hep complexes increased in the following sequence: Trp < Tyr < Phe (K = 19 ± 2 < 39 ± 3 < 710 ± 70 M?1, respectively). These values probably determined the dominant contribution of the phenylalanine impurity in the heparin drug. The contamination of animal elastin whose structure differed from that of the human elastin is thought to be a reason for allergic reactions and even anaphylactic shock during medical treatment with this drug.  相似文献

The size, age,and sex composition of 2277 casual groups from eight shopping malls and two sidewalk shopping areas, and 1086 groups from other public environments, were recorded and analyzed to reveal age, sex, and environmental components of human grouping tendencies. Groups containing grade school or teenage children were significantly larger than groups of other ages. Adult females were found in larger groups than males; although females readily formed groups with either sex, adult males formed few groups containing one or two other males. Group size showed a significant, inverse correlation with social distances between first, second, and third nearest neighboring group members in a photo sample, but group size showed no correlation with social distances between strangers. These results are interpreted in the light of an ethological hypothesis that contemporary grouping patterns had their origins in adaptive, inborn aggregative tendencies that were later incorporated into the repertoire of nonverbal human behaviors.  相似文献

We investigated the influence of drone size and potential reproductive quality on caste interactions by adding large drones reared in drone cells (DC drones; considered to be of higher quality) and small drones reared in worker cells (WC drones; of lower quality) to two observation colonies and monitoring worker–drone interactions and acceptance by workers. When initially introduced into the colonies more DC drones received trophallaxis, whereas more WC drones received aggression and eviction attempts from workers. Nevertheless, WC and DC drones were equally likely to be accepted by workers. For both drone types accepted individuals had slightly, but significantly greater weights than rejected males. Thus, workers discriminated between drones of different sizes and potential quality upon initial encounter, although these discriminations were not strongly associated with acceptance decisions. After drones were accepted, workers either showed no preference for interacting with WC or DC drones, or if a preference was shown it tended to favor WC drones. Compared to accepted DC drones, significantly more WC drones received grooming for longer periods of time and also spent more time engaged in all interactions with workers combined. DC and WC drones did not differ in the likelihood of receiving trophallaxis or the vibration signal, although for both interactions slightly more WC drones were recipients. Thus, workers may bias some interactions with accepted drones to favor smaller individuals with potential developmental deficiencies, in a manner that could contribute to the production of a greater total number of competitive males and increased colony reproductive output.  相似文献

Summary Electron micrographs of Trichomonas vaginalis cells were examined morphometrically by means of an electronic image analyser. Cells from cultures in logarithmic growth contained smaller chromatic granules (hydrogenosomes ) and a higher proportion of sausage- or dumb-bell-shaped granules than did cells in stationary growth.These findings support the view that chromatic granules replicate by division of pre-existing granules and that this involves elongation and attenuation of their mid-region.The total area of cross sectioned granules occupied the same percentage of the cytoplasm of cells from cultures in logarithmic and stationary growth. The density of chromatic granules in the cytoplasm was identical for the two groups of cells.  相似文献

Multi-drug resistance (MDR) can be explained by a drug handling-type activity. In this context it is also necessary to consider the multi-specificity between drugs and drug transporters in order to explain MDR. Accordingly, the efficiency of drug efflux in MDR has always been a conceptual problem in biochemistry. Indeed, how one protein can expel, from cells, hundreds of compounds with high specificity is still controversial today. To safeguard the notion of biochemical specificity, many studies have suggested alternative mechanisms to Pgp-mediated drug resistance, which do not involve the handling of drugs. However, none of these studies have definitively ruled out the possibility concept of drug handling. Thus, until now it was not possible to imagine MDR without drug-transporter affinity or specificity. However, drug-transporter affinity is not always needed to generate what appears to be a very efficient chemical reaction. Indeed, based on the fact that bi-dimensional diffusion properties (i.e. diffusion in the membrane) are paramount to explain drug pumping-mediated MDR, it is possible to suggest how specific mathematical properties of random motions can be used to construct a model of Pgp-MDR, providing that Pgp oscillates between open/drug-accepting and closed/drug-expelling conformations. This different viewpoint highlights the fact that the multi-specificity of drug transporters and the “vacuum cleaner” hypothesis may actually be two sides of the same coin, both explained by the diffusion properties of drugs in the membrane. After retrieving basic results, predictions will be highlighted. Finally, the coherence of this model in the context of cancer biology will be discussed further.  相似文献

Summary Chloride cells in gill epithelium of Oligocottus maintained in sea water have a much branched system of agranular cytoplasmic tubules, numerous mitochondria, and a prominent apical crypt. The mitochondria are randomly dispersed and do not show preferential orientation with respect to the tubules.After brief exposure of fish to sea water diluted 1/100 with glass distilled water, the mitochondria and tubules become rearranged into parallel arrays and the apical crypts disappear. The appearance of these cells is similar to that of pseudobranch cells in Fundulus heteroclitus (Copeland and Dalton, 1959).These data suggest that chloride cells and pseudobranch cells represent different adaptive forms of a single cell type and that transformation from the chloride cell configuration to that of pseudobranch cells can be induced by osmotic stress.This work was performed in 1963 when the author was enrolled in the Fine Structure Training program of the Department of Biological Structure, University of Washington. Summary Segregation analyses were performed for plasma low density lipoprotein cholesterol (LDL-C), triglycerides (TG) and high density lipoprotein cholesterol (HDL-C) in five Christian Arab kindreds identified through probands with familial hypercholesterolemia. In this subset of the Christian Arab community, the results were consistent with major gene determination of LDL-C with allele frequency (q) of 0.042 (95% confidence interval 0.008–0.079) in addition to polygenic transmission (h 2= 0.34). The Lebanese allele was identified directly by polymerase chain reaction and HinfI restriction analysis. Analysis of this mutation permits direct diagnosis of familial hypercholesterolemia in most affected individuals although our results indicated the possible existence of an additional independent factor leading to elevated LDL-C levels. The segregation results for TG indicated the presence of a major effect, although the existence of a major gene could not be demonstrated. There was also no evidence of a major locus effect on HDL-C levels.  相似文献

The spindle assembly checkpoint (SAC) prevents anaphase onset until all chromosomes accomplish proper bipolar attachments to the mitotic spindle and come under tension, thereby ensuring the fidelity of chromosome segregation. Despite significant advances in our understanding of SAC signalling, a clear link between checkpoint signalling and the molecular mechanisms underlying chromosome attachment to microtubules has not been established so far. However, independent studies from many groups have interestingly found that the bone-a-fide Bub1, BubR1 and Bub3 SAC proteins are themselves required for proper kinetochore-microtubule (K-MT) interactions. Here, we review these findings and discuss the specific contribution of each of these proteins in the regulation of K-MT attachment, taking into consideration their interdependencies for kinetochore localization as well as their relationship with other proteins with a known role in chromosome attachment and congression.  相似文献

Summary In a pilot study, the possible influence of the inherited electroencephalogram (EEG) pattern on aspects of psychologic maturation was investigated. The twin sample consisted of 208 pairs, 110 monozygotic (MZ) and 98 dizygotic (DZ), mostly children and adolescents. The study showed a greater similarity between MZ compared with DZ twins in an immaturity score constructed from the Baum test (Koch 1976) and in a teacher's maturity scoring of essays. For both parameters, a correlation between occipital -frequency and the test criterium could be demonstrated. This correlation was in the direction expected, and it was significant statistically. This result suggests an influence of genetically determined variation of brain maturation on interindividual variability of psychologic variation. Due to shortcomings of the methods used in the study, more specific conclusions cannot be drawn; the problem needs further elucidation using modern methods.The study was supported by the Deutsche Forschungsgemeinschaft  相似文献
