During summer 2007, Arctic microphytobenthic potential primary production was measured at several stations around the coastline of Kongsfjorden (Svalbard, Norway) at ≤5 m water depth and at two stations at five different water depths (5, 10, 15, 20, 30 m). Oxygen planar optode sensor spots were used ex situ to determine oxygen exchange in the overlying water of intact sediment cores under controlled light (ca. 100 μmol photons m−2 s−1) and temperature (2–4°C) conditions. Patches of microalgae (mainly diatoms) covering sandy sediments at water depths down to 30 m showed high biomass of up to 317 mg chl a m−2. In spite of increasing water depth, no significant trend in “photoautotrophic active biomass” (chl a, ratio living/dead cells, cell sizes) and, thus, in primary production was measured at both stations. All sites from ≤5 to 30 m water depth exhibited variable rates of net production from −19 to +40 mg O2 m−2 h−1 (−168 to +360 mg C m−2 day−1) and gross production of about 2–62 mg O2 m−2 h−1 (17–554 mg C m−2 day−1), which is comparable to other polar as well as temperate regions. No relation between photoautotrophic biomass and gross/net production values was found. Microphytobenthos demonstrated significant rates of primary production that is comparable to pelagic production of Kongsfjorden and, hence, emphasised the importance as C source for the zoobenthos.  相似文献

Changes in oxygen consumption rate and Na+/K+-ATPase activity during early development were studied in the sea urchin Paracentrotus lividus Lam. The oxygen consumption rate increased from 0.12 μmol O2 mg protein−1 h−1 in unfertilized eggs to 0.38 μmol O2 mg protein−1 h−1 25 min after fertilization. Specific activity of the Na+/K+-ATPase was significantly stimulated after fertilization, ranging up to 1.07 μmol Pi h−1 mg protein−1 in the late blastula stage and slightly lower values in the early and late pluteus stages.  相似文献

Petroleum-related activities in Arctic waters are rapidly increasing parallel to the ongoing thinning of the Arctic sea ice. As part of a series of studies on petroleum-induced stress in polar cod Boreogadus saida, we tested the effects of acute (~60 min) and chronic (4 weeks) exposure to the water soluble fraction (WSF) of petroleum on whole body metabolism inferred from measurements of oxygen consumption rates. The exposure of polar cod to WSF leads to a statistically significant depression in routine metabolism in the order Control (0.260 mg O2 g fish−1 h−1; N = 6) > Chronic (0.191 mg O2 g fish−1 h−1; N = 6) > Acute (0.110 mg O2 g fish−1 h−1; N = 2), decoupling of routine metabolism and body mass but possibly also to a partial metabolic compensation after 4 weeks of exposure. The results are reviewed in context with similar studies on Antarctic and non-polar fishes.  相似文献

Toxic at low concentrations, phenol is one of the most common organic pollutants in air and water. In this work, phenol biodegradation was studied in extreme conditions (80°C, pH = 3.2) in a 2.7 l bioreactor with the thermoacidophilic archaeon Sulfolobus solfataricus 98/2. The strain was first acclimatized to phenol on a mixture of glucose (2000 mg l−1) and phenol (94 mg l−1) at a constant dissolved oxygen concentration of 1.5 mg l−1. After a short lag-phase, only glucose was consumed. Phenol degradation then began while glucose was still present in the reactor. When glucose was exhausted, phenol was used for respiration and then for biomass build-up. After several batch runs (phenol < 365 mg l−1), specific growth rate (μX) was 0.034 ± 0.001 h−1, specific phenol degradation rate (qP) was 57.5 ± 2 mg g−1 h−1, biomass yield (YX/P) was 52.2 ± 1.1 g mol−1, and oxygen yield factor ( \textY\textX/\textO 2 ) \left( {{\text{Y}}_{{{\text{X}}/{\text{O}}_{ 2} }} } \right) was 9.2 ± 0.2 g mol−1. A carbon recovery close to 100% suggested that phenol was exclusively transformed into biomass (35%) and CO2 (65%). Molar phenol oxidation constant ( \textY\textO 2 /\textP ) \left( {{\text{Y}}_{{{\text{O}}_{ 2} /{\text{P}}}} } \right) was calculated from stoichiometry of phenol oxidation and introducing experimental biomass and CO2 conversion yields on phenol, leading to values varying between 4.78 and 5.22 mol mol−1. Respiratory quotient was about 0.84 mol mol−1, very close to theoretical value (0.87 mol mol−1). Carbon dioxide production, oxygen demand and redox potential, monitored on-line, were good indicators of growth, substrate consumption and exhaustion, and can therefore be usefully employed for industrial phenol bioremediation in extreme environments.  相似文献

An isolated, perfused salmon tail preparation showed oxyconformance at low oxygen delivery rates. Addition of pig red blood cells to the perfusing solution at a haematocrit of 5 or 10% allowed the tail tissues to oxyregulate. Below ca. 60 ml O2 kg−1 h−1 of oxygen delivery (DO2), VO2 was delivery dependent. Above this value additional oxygen delivery did not increase VO2 of resting muscle above ca. 35 ml O2 kg−1 h−1. Following electrical stimulation, VO2 increased to ca. 65 ml O2 kg−1 h−1, with a critical DO2 of ca. 150 ml O2 kg−1 h−1. Dorsal aortic pressure fell to 69% of the pre-stimulation value after 5 min of stimulation and to 54% after 10 min. Microspheres were used to determine blood flow distribution (BFD) to red (RM) and white muscle (WM) within the perfused myotome. Mass specific BFD ratio at rest was found to be 4.03 ± 0.49 (RM:WM). After 5 min of electrical stimulation the ratio did not change. Perfusion with saline containing the tetrazolium salt 3-(4,5-Dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) revealed significantly more mitochondrial activity in RM. Formazan production from MTT was directly proportional to time of perfusion in both red and WM. The mitochondrial activity ratio (RM:WM) did not change over 90 min of perfusion.  相似文献

Chromosome set manipulation was used to produce rainbow trout, Oncorhynchus mykiss, with identical nuclear backgrounds, but different maternal backgrounds to determine mitochondrial effects on development rate and oxygen consumption. Significant differences in development rate and oxygen consumption were observed between groups from different females. Development rates ranged from a mean of 317.97 degree days (°d) to 335.25 °d in progeny from different females. Mean oxygen consumption rates ranged from 3.31 μmol O2 g− 1 wet mass h− 1 to 9.66 μmol O2 g− 1 wet mass h− 1. Oxygen consumption and development rate analysis revealed the two slowest developing groups had the highest oxygen consumption rates. Development rate differences between second generation clonal females indicate that mitochondrial genomes play a significant role on early development and are comparable to development rate differences between clonal lines of rainbow trout. These results indicate that selection for mitochondrial genomes could increase growth rates and possibly food conversion ratios in aquaculture species.  相似文献

A protocol to monitor respiration as O2 consumption in soil slurries using the Strathtox® respirometer was developed and tested on four soils from brownfield sites. Respiration rates (mg l−1 h−1) of soil slurries in the Strathtox® were compared with rates (μl min−1) of field moist soils analysed using the Columbus Oxymax® ER10 respirometer. One of the soils (99612B), historically contaminated with diesel, was further studied by monitoring the effect of inorganic NH4NO3 liquid nutrient on enhancing respiration rate. Soil microcosms were monitored continuously on the Oxymax® or sampled at 24, 48 and 72 h intervals, prepared as soil slurries, and analysed on the Strathtox®. On the full-scale remediation project (6000 m3) soil 99612B was treated as a biopile with spent mushroom compost (SMC) amendment and respiration rates monitored in samples over an 8-week period. In the laboratory microcosm experiment and full-scale bioremediation treatment described, correlation was found for respiration rates between the two respirometry systems.  相似文献

Nitrite accumulates during biological denitrification processes when carbon sources are insufficient. Acetate, methanol, and ethanol were investigated as supplementary carbon sources in the nitrite denitrification process using biogranules. Without supplementary external electron donors (control), the biogranules degraded 200 mg l−1 nitrite at a rate of 0.27 mg NO2–N g−1 VSS h−1. Notably, 1,500 mg l−1 acetate and 700 mg l−1 methanol or ethanol enhanced denitrification rates for 200 mg l−1 nitrite at 2.07, 1.20, and 1.60 mg NO2–N g−1 VSS h−1, respectively; these rates were significantly higher than that of the control. The sodium dodecyl sulfate polyacrylamide gel electrophoresis of the nitrite reductase (NiR) enzyme identified three prominent bands with molecular weights of 37–41 kDa. A linear correlation existed between incremental denitrification rates and incremental activity of the NiR enzyme. The NiR enzyme activity was enhanced by the supplementary carbon sources, thereby increasing the nitrite denitrification rate. The capacity of supplementary carbon source on enhancing NiR enzyme activity follows: methanol > acetate > ethanol on molar basis or acetate > ethanol > methanol on an added weight basis.  相似文献

Antibiotics regulate various physiological functions in cyanobacteria and may interfere with the control of cyanobacterial blooms during the application of algaecides. In this study, Microcystis aeruginosa was exposed to H2O2 and glyphosate for 7 d in the presence of coexisting mixed antibiotics (amoxicillin, spiramycin, tetracycline, ciprofloxacin, and sulfamethoxazole) at an environmentally relevant concentration of 100 ng · L?1. The mixed antibiotics significantly (P < 0.05) alleviated the growth inhibition effect of 15–45 μM H2O2 and 40–60 mg · L?1 glyphosate. According to the increased contents of chlorophyll a and protein, decreased content of malondialdehyde, and decreased activities of superoxide dismutase and glutathione S‐transferase, antibiotics may reduce the toxicity of the two algaecides through the stimulation of photosynthesis and the reduction in oxidative stress. The presence of coexisting antibiotics stimulated the production and release of microcystins in the M. aeruginosa exposed to low concentrations of algaecides and posed an increased threat to aquatic environments. To eliminate the secondary pollution caused by microcystins, high algaecide doses that are ≥45 μM for H2O2 and ≥60 mg · L?1 for glyphosate are recommended. This study provides insights into the ecological hazards of antibiotic contaminants and the best management practices for cyanobacterial removal under combined antibiotic pollution conditions.  相似文献

Schizothorax chongi (locally known as Xilian Yu), a fish species commonly found in Yalong River, has been declining quickly in recent years. One of the important factors, among many, is the interruption of the free flowing river by dams. To obtain data that can be applied to the design of a fishway for S. chongi and other species in the community, a laboratory study of juvenile S. chongi’s swimming energetics and kinematics was conducted in a flume-type respirometer equipped with a high speed video camera system to record swimming behavior. The aerobic metabolic rate, tail beat frequency (TBF) and tail beat amplitude (TBA) were measured during steady swimming at varying flow rates for fish of similar mass. A power function accurately describes the relationship between oxygen consumption rate (MO2) and swimming speed (U). The estimated standard metabolic rate (SMR) calculated from the power function was 445.34 mg O2 kg−1 h−1, similar to the experimental result of 431.5 mg O2 kg−1 h−1. The relationship between cost of transport (COT) and U was, characteristically, inverse bell-shaped, with COTmin = 44.6 J kg−1 m−1 at U opt = 5.5 body lengths per second (bl s−1). There was a significant positive linear correlation between TBF and U. The slope of the correlation (0.33) was lower than for many other species, implying that S. chongi swim efficiently. The TBA, ranging from 0.15 to 0.2 bl, was found to be independent of U. Kinematic analyses indicates that S. chongi primarily depends on the caudal fin to generate forward thrust and employs three velocity-dependent swimming gaits. This investigation provides data on the swimming ability of S. chongi that will add to the basic science required for fishway design.  相似文献

The production of compound K and aglycon protopanaxadiol (APPD) from ginsenoside Rd and ginseng root extract was performed using a recombinant β-glycosidase from Pyrococcus furiosus. The activity for Rd was optimal at pH 5.5 and 95°C with a half-life of 68 h at 95°C. β-Glycosidase converted Rb1, Rb2, Rc, and Rd to APPD via compound K. With increases in the enzyme activity, the productivities of compound K and APPD increased. The substrate concentration was optimal at 4.0 mM Rd or 10% (w/v) ginseng root extract; 4 mM of Rd was converted to 3.3 mM compound K with a yield of 82.5% (mol/mol) and a productivity of 2,010 mg l−1 h−1 at 1 h and was hydrolyzed completely to APPD with 364 mg l−1 h−1 after 5 h. Rb1, Rb2, Rc, and Rd at 3.9 mM in 10% ginseng root extract were converted to 3.1 mM compound K with 79.5% and 1,610 mg l−1 h−1 at 1.2 h and were hydrolyzed completely to APPD with 300 mg l−1 h−1 after 6 h. The concentrations and productivities of compound K and APPD in the present study are the highest ever reported.  相似文献

Benthic oxygen consumption and primary production were measured using the bell jar technique in deltaic and fringing mangrove forests of tropical northeastern Australia. In a deltaic forest, rates of sediment respiration ranged from 197 to 1645 mol O2 m–2 h–1 (mean=836), but did not vary significantly with season or intertidal zone. Gross primary production varied among intertidal zones and seasons, ranging from –281 to 1413 mol O2 m–2 h–1 (mean=258). Upon tidal exposure, rates of gross primary production increased, but respiration rates did not change significantly. In a fringing mangrove forest, benthic respiration and gross primary production exhibited strong seasonality. In both forests, rates of oxygen consumption and production were low compared to salt marshes, but equivalent to rates in other mangrove forests. The production:respiration (P/R) ratio varied greatly over space and time (range:–0.61 to 1.76), but most values were «1 with a mean of 0.15, indicating net heterotrophy. On a bare creek bank and a sandflat, rates of gross primary production and P/R ratios were generally higher than in the adjacent mangroves. Low microalgal standing stocks, low light intensity under the canopy, and differences in gross primary production between mangroves and tidal flats, and with tidal status, indicate that benthic microalgae are light-limited and a minor contributor to primary productivity in these tropical mangrove forests.  相似文献

Alginate production by Azotobacter vinelandii growing in chemostat cultures was evaluated under different O2 transfer rates (OTR). As a result of modifying the culture’s agitation rate from 300 to 500 rpm, the OTR increased from 9 to 15.1 mmol l−1 h−1 and a slight variation in the alginate production (1.7–2.2 g l−1) was observed. At a constant growth rate (0.1 h−1), the mean molecular mass of the alginate was strongly influenced by changes in the OTR, varying from 860 to 1,690 kDa. These results support a possible relationship between alginate polymerization-depolymerization process and the O2 uptake rate.  相似文献

In the root zone of rice plants aerobic methanotrophic bacteria catalyze the oxidation of CH4 to CO2, thereby reducing CH4 emissions from paddy soils to the atmosphere. However, methods for in situ quantification of microbial processes in paddy soils are scarce. Here we adapted the push–pull tracer-test (PPT) method to quantify CH4 oxidation in the root zone of potted rice plants. During a PPT, a test solution containing CH4 ± O2 as reactant(s), Cl? and Ar as nonreactive tracers, and BES as an inhibitor of CH4 production was injected into the root zone at different times throughout the circadian cycle (daytime, early nighttime, late nighttime). After a 2-h incubation phase, the test solution/pore-water mixture was extracted from the same location and rates of CH4 oxidation were calculated from the ratio of measured reactant and nonreactive tracer concentrations. In separate rice pots, O2 concentrations in the vicinity of rice roots were measured throughout the circadian cycle using a fiber-optic sensor. Results indicated highly variable CH4 oxidation rates following a circadian pattern. Mean rates at daytime and early nighttime varied from 62 up to 451 μmol l?1 h?1, whereas at late nighttime CH4 oxidation rates were low, ranging from 13 to 37 μmol l?1 h?1. Similarly, daytime O2 concentration in the vicinity of rice roots increased to up to 250% air saturation, while nighttime O2 concentration dropped to below detection (<0.15% air saturation). Our results suggest a functional link between root-zone CH4 oxidation and photosynthetic O2 supply.  相似文献

Bacterial density and productivity were investigated along four salinity gradients within the estuary Ria de Aveiro. Bacterial variables and environmental parameters were measured at three to four stations spanning the entire salinity gradient of the four channels. The rather high variation in bacterial productivity (0.16–7.6 μg C L−1 h−1) along the profiles of salinity indicates that bacterial activity shows a reactive behavior to environmental changing. Bacterial density (0.5–11.2 × 109 cells L−1) with a comparative smaller variation showed a more conservative behavior, mainly reflecting the phytoplankton distribution. Contrary to expectation, minimal values of bacterial productivity were not observed in November–December but in June. In fact, in November–December, the deep zone near the mouth showed the highest values of bacterial activity. At the upper stations, the highest values were observed in October. The relatively high values of bacterial production during the cold rainy season suggest that allochthonous substrates leached out from the surroundings by rain controlled the distribution of bacterial activity in the estuarine system. The substantial decrease in salinity during the rainy season negatively affected bacterial productivity, namely in the marine zone, where water column was highly stratified. Salinity seems to play an indirect role in the regulation of estuarine bacteria because there are different bacterial communities adapted to a wide salinity range.  相似文献

We have capitalised on the availability of eggs and adults of the naked dragonfish Gymnodraco acuticeps (Sub-order Notothenioidei, F. Bathydraconidae) near McMurdo Station, Antarctica to examine metabolic energy utilization at different stages of its life cycle. Average egg respiration rates were found to increase from 2.17±1.02 nmol O2 h−1 ind−1 at about 17 h post-fertilization (hpf) to 5.72±0.56 nmol h−1 ind−1 at about 24 hpf, during which time the eggs underwent first cleavage. The respiration rates of embryos from 2–20 days post-fertilization (dpf) averaged 4.11±1.47 nmol O2 h−1 ind−1. About 10 months post-fertilization, oxygen consumption rates of 27.14±3.92 nmol O2 h−1 ind−1 were recorded immediately prior to hatching, with a peak of 112.41±31.38 nmol O2 h−1 ind−1 at the time of hatch. Larvae aged 46–63 days post-hatch had an average respiration rate of 64.4±15.11 nmol O2 h−1 ind−1. Mass-specific respiration rates of hatched larvae (approximately 1–2 months old) were calculated using dry weights (DW) and averaged 16.1±3.4 nmol O2 h−1 mg−1 DW. Adult dragonfish respiration rates (corrected for a 100 g fish and using a 0.8 scaling exponent) averaged 0.91±0.36 mmol O2 kg−1 h−1 after a 48 h acclimatization period, which is not indicative of significant metabolic cold adaptation. The energy contents of dragonfish eggs and larvae were also measured by microbomb calorimetry and used, along with the respiration data, in an initial approach to estimate an energy budget. In order to balance the budget, the bulk of the available post-gastrulation respiratory energy (during 213 days of embryonic incubation) must be consumed at a relatively low average rate (7.1 nmol O2 h−1 ind−1), which supports the possibility that advanced dragonfish embryos overwinter in a relatively quiescent metabolic state while awaiting a suitable stimulus (such as the return of the sun) to initiate hatching.  相似文献

By applying planar optodes and imaging techniques to a benthic photosynthetic mat, we demonstrated an extensive vertical and horizontal variation in O2 concentrations, O2 consumption, and O2 production. In light, the oxic zone could be divided into three horizons: 1) an upper zone dominated by diatoms that had a moderate net O2 production, 2) another zone dominated by Microcoleus-like cyanobacteria with a high net O2 production, and 3) a lower zone with disintegrating microalgae and cyanobacteria with a high O2 consumption rate. From the O2 images, the net O2 production/consumption was calculated at a spatial resolution of 130 μM. This allowed us to identify microsites with high rates of O2 turnover within the photic zone. Sites with high net O2 consumption (>1.5 nmol·cm?3·s?1) were typically situated next to sites with a relatively high net production (>2 nmol·cm?3·s?1), revealing a mosaic in which the highest O2 consumption sites were surrounded by the highest O2 production sites. This suggested a tight spatial coupling between production and consumption of O2 within the photic zone. Light stimulated the O2 consumption within the photic zone. At irradiances above 400 μmol photons·m?2·s?1, the stimulated O2 production was almost completely balanced by enhanced O2 consumption at microsites exhibiting net consumption of O2 even at maximum irradiance (578 μmol photons·m?2·s?1). Our observations strongly supported the idea that light-stimulated respiration was caused by stimulated heterotrophic activity fueled by organic carbon leakage from the phototrophs. Despite microsites with high net O2 consumption, anoxic microniches were not encountered in the investigated mat. Images of gross photosynthetic rates also revealed an extensive horizontal variation in gross rates, with microsites of low or no photosynthesis within the otherwise photic zone. Calculations based on the obtained images revealed that at maximum light (578 μmol photons·m?2·s?1), 90% of the O2 produced was consumed within the photic zone. The presented data demonstrate the great potential offered by planar optode for studies of benthic photosynthetic communities.  相似文献

Little is known about the contributions of biomass feedstock storage to the net greenhouse gas emissions from cellulosic biofuels. Direct emissions of methane and nitrous oxide during decomposition in storage may contribute substantially to the global warming potential of biofuels. In this study, laboratory-scale bales of switchgrass and corn stover were stored under a range of moisture (13.0–32.9%) and temperature (5–35 °C) conditions and monitored for O2 consumption and CO2, CH4, and N2O production over 8 weeks. Gas concentrations and emissions rates were highly variable within and between experimental groups. Stover bales produced higher CO2 concentrations (P = 0.0002) and lower O2 (P < 0.0001) during storage than switchgrass bales. Methane concentrations (1.8–2100 ppm) were inversely correlated with bale moisture (P < 0.05), with emissions rates ranging from 4.4–914.9 μg kg−1 DM day−1. Nitrous oxide concentrations ranged from 0 to 31 ppm, and emissions from switchgrass bales inversely correlated with temperature and moisture (P < 0.0001). Net global warming potential from each treatment (0–2.4 gCO2e kg−1 DM) suggests that direct emission of methane and nitrous oxide from aerobically stored feedstocks have a small effect on net global warming potential of cellulosic biofuels.  相似文献
