The excretion of delta-aminolevulinic acid and porphobilinogen in the urine of 31 patients with multiple sclerosis did not differ significantly from that of 51 hospitalized control patients or eight patients with poliomyelitis. There was no relationship between exacerbations, remissions or duration of the illness, and levels of delta-aminolevulinic acid or of porphobilinogen. These assays therefore appear to be of no value in the differential diagnosis of multiple sclerosis or in following the severity or stage of this illness. Whereas demyelination does occur in acute porphyria where the levels of delta-aminolevulinic acid and porphobilinogen are elevated, the converse is not true; that is, demyelination is not always associated with an increase in the excretion of porphobilinogen or delta-aminolevulinic acid.  相似文献

Fatty acid (FA) composition of vacuolar membrane lipids from storage tissues of umbelliferous plants, viz., parsnip (Pastinaca sativa L.), parsley (Petroselinium crispum L.), and carrot (Daucus carota L.) is studied by gas-liquid chromatography and the FA biosynthetic pathways are considered. Vacuolar membrane lipids are characterized by high (78% of the total FA pool) content of unsaturated FA among which linoleic acid is predominant. Its content in vacuolar lipids of parsnip, parsley and carrot is 53.5, 55.1, and 54.9%, respectively. Parsnip and parsley vacuolar lipids contain large amounts of hexadecadienoic C16:2ω6 acid (8.0 and 4.6%, respectively). The content of α-linolenic acid in vacuolar lipids of tested plants varies from 4.8 to 7.3%. Palmitic acid (18.0–20.7%) predominates among saturated FA. High content of linoleic and hexadecadienoic acid in parsnip and parsley vacuolar lipids is suggestive of a crucial role of the microsomal ω6 fatty-acid desaturase fad2 gene in resistance and acclimation of plants to low temperatures.  相似文献

Measurements have been made of the fatty-acid composition of lipids in normal hamster sera compared with sera of hamsters infected with Schistosoma mansoni. The results show a consistent decrease of linoleic and eicosapentaenoic acids, and an increase in arachidonic acid in the phosphatidylcholine and lysophosphatidylcholine of the infected animals. Arachidonic acid was also increased in cholesterol ester, and docosahexaenoic acid was raised in free fatty acid and in phosphatidylcholine in sera from infected animals.  相似文献

A low protein diet affects amounts of linoleic and arachidonic acids in hepatic microsomal phospholipids of growing rats. Are the changes related to modifications in microsomal delta 6- and delta 5- linoleic acid desaturase activities? Two groups of Wistar rats weighing 80 +/- 5 g at the beginning of the experiment were used: Control group (T) was fed on a 16% gluten + 4% casein diet for 53 days; Experimental group (E) was fed on a 4% gluten + 1% casein diet for 26 days (MP) then Control diet for 27 days (RE). After 2, 14 and 26 days of MP and 2, 15 and 27 days of RE, rats of each group were sacrificed. Protein and water contents of liver, quantitative fatty acid, composition of total lipids in liver and hepatic microsomes were determined. delta 6- and delta 5- linoleic acid desaturase activities were estimated from incubation of liver microsomes with [1-14C] C 18: 2 n-6 or [2(14)C] C 20: 3 n-6 respectively. The low protein diet stops practically ponderal growth. The fatty-acid compositions of microsomal total lipids of E rats were affected in comparison with values of T rats. These modifications persist after 27 days of RE. The C 20: 4 n-6/C 18: 2 n-6 ratio in microsomal total lipids was slightly different between T and E rats but increased strongly during refeeding. Same modifications take place in the fatty-acid composition of hepatic total lipids. After two days of MP, delta 6- and delta 5- desaturase activities were depressed, phenomenon that not persist in the course of MP. These enzyme activities increase to higher values than those of the T after two days of RE.  相似文献

During a follow-up of five to seven years 33 out of 1222 middle-aged men initially free of coronary heart disease sustained fatal or non-fatal myocardial infarction or died suddenly. The fatty-acid composition of serum triglycerides, phospholipids, and cholesterol esters had been measured at the start of the surveillance in these men and in a control group of 64 men matched for age, serum cholesterol and triglyceride concentrations, blood pressure, obesity, smoking, and one-hour glucose tolerance. Palmitic and stearic acids of phospholipids were significantly higher and linoleic and most polyunsaturated fatty acids, including arachidonic acid and eicosapentaenoic acid, of phospholipids were lower in the subjects who sustained coronary events compared with the controls. Linoleic acid tended to correlate negatively with blood pressure while other polyunsaturated fatty acids, especially eicosapentaenoic acid, exhibited a negative correlation with blood pressure and relative body weight in the controls but not in the subjects who sustained coronary events. These findings suggest that the fatty-acid pattern of serum phospholipids is an independent risk factor for coronary heart disease.  相似文献

The yolk fatty-acid profiles of a range of species of insectivorous scincid lizards generally conform to a common pattern, typified by high proportions of linoleic acid (13.5–18.5% of total fatty acids), substantial proportions of -linolenic acid (2.4–8.2%), and significant amounts of the long-chain polyunsaturated fatty acids, arachidonic (1.6–3.3%), eicosapentaenoic (0.7–1.2%) and docosahexaenoic (0.7–1.6%) acids. We characterised the fatty-acid compositions of ten prey taxa that are eaten by female skinks during vitellogenesis. Linoleic acid is the major polyunsaturated fatty acid in all prey, excepting Orthoptera where -linolenic acid predominates. To varying extents, -linolenic acid is present in all the prey items. Arachidonic acid forms over 1% of total fatty acids for six of the prey items. Four of the prey items contain eicosapentaenoic acid at over 1%. Most notably, docosahexaenoic acid is essentially absent from all the prey items. There is a general similarity between the fatty-acid profiles of prey and yolk, suggesting that the linoleic, -linolenic, arachidonic and eicosapentaenoic acids required for egg formation can be supplied directly from the maternal diet. However, the docosahexaenoic acid of the egg lipids cannot derive from the diet and must, therefore, be formed by biosynthesis in the maternal liver, using dietary -linolenic and eicosapentaenoic acids as precursors.  相似文献

Erythrocyte membranes from multiple sclerosis (MS) patients and normal individuals were studied by electron spin resonance spectroscopy, osmotic fragility tests, scanning electron microscopy (SEM) and fatty acid analysis of membrane lipids. There was no significant difference in the membrane fluidity between MS and normal erythrocytes using fatty acid spin labels with the nitroxide moiety on carbons 5, 12, or 16 from the carboxyl group. Linoleic acid, which has been reported to decrease the absolute electrophoretic mobility of only MS erythrocytes, increased the fluidity of MS and normal erythrocyte membranes to a similar extent. The osmotic fragility of MS erythrocytes obtained from outpatients was similar to normal control cells but the osmotic fragility of erythrocytes obtained from hospitalized MS patients was greater than normal. Scanning electron microscopy of MS erythrocytes revealed no gross abnormalities. Cells incubated with linoleic acid had transformed from discocytes into sphero-echinocytes with prominent membrane surface indentations but MS and normal erythrocytes appeared identical. Of the fatty acid content of the total lipid extract, erythrocytes from most, but not all, MS hospitalized patients and some patients with other demyelinating diseases had relatively less (P<.001) 18:2 than the normal cells. These results indicate that at least some of the abnormalities reported in MS erythrocytes may only be found in hospitalized patients and may be due to other complications of the disease. They also indicate that the reported abnormal effects of linoleic acid on the electrophoretic mobility of MS erythrocytes may be caused by some other mechanism than an effect on the fluidity of the bilayer.  相似文献

The effects of hyperlipidic polyunsaturated fat diets (PF) and saturated fat diets (SF) versus control diet (CO) on total lipid, phospholipid (PL) and cholesteryl ester (CE) content and composition of pig serum lipoproteins were studied in Large White pigs who first underwent a PF period for 14 days followed by a CO period and third a SF feeding during a fortnight. PF and SF diets induced an increase of linoleic acid in serum total lipids, especially in HDL fraction; this increase in CE and PL involved a reverse change of oleic acid. The modifications induced by the amounts of dietary linoleic acid could be explained by an alteration of LCAT activity. The fatty acid pattern of PL which constitute the LCAT substrate lead to an enrichment of cholesterol linoleate.  相似文献

1. Three groups of female rats (8-12 weeks old) were maintained respectively on a linoleic acid-rich diet, a linoleic acid-poor predominantly saturated-fatty acid diet and a normal diet. Changes in the fatty acid compositions of serum, brain, brain mitochondria-rich fraction and myelin were observed. 2. Of the serum fatty acids, linoleic acid showed the greatest change in the percentage of the total acids in response to diet; the change in the proportion of oleic acid was considerable. The percentages of arachidonic acid in serum fatty acids in the groups on the linoleic acid-rich and linoleic acid-poor diets were similar, but higher than those in the normal group. 3. Changes in the proportions of linoleic acid, arachidonic acid and docosahexaenoic acid occurred in brain fatty acids that to some extent paralleled those occurring in the serum. Changes in the proportions of most other acids in the serum fatty acids were not accompanied by corresponding changes in the brain fatty acids. 4. The percentage fatty acid compositions of a mitochondria-rich fraction and myelin are given, and changes in the relative proportions of linoleic acid, arachidonic acid and possibly some docosapolyenoic acids were demonstrated to occur as a result of diet. 5. The results are discussed in relation to the possible aetiology of multiple sclerosis.  相似文献

The fatty acid pattern of blood serum lipids was examined by gas liquid-chromatography in 30 cases with bilharzial hepatic fibrosis, 11 cases with chronic active hepatitis accompanied by jaundice, and 28 healthy individuals as a comparison group of the same socioeconomic class of patients. In addition, the fatty acid patterns of the three major serum lipid classes, namely: cholesterol ester, phospholipids and triglycerides, were also investigated in seven cases of each group by gas liquid chromatography. The most remarkable differences were: a depression of the essential fatty acid level (linoleic and arachidonic) in both groups of patients together with a concomitant elevation of oleic acid in the bilharzial group and an elevation of oleic, palmitic, palmitoleic acids in the chronic active hepatitis group. The depression of linoleic and arachidonic acids was explained by the low fat diet intake, malnutrition, and the malabsorption factors which were frequent in all the patients studied. The elevation of monoethenoid acids was attributed to the decrease in the ability of the liver to desaturate the endogenous saturated and monounsaturated acids to polyunsaturated ones.  相似文献

Reduced reproductive performance of dairy cows during the summer is often associated with elevated temperature. Semen collected and cryopreserved during the summer may be of low quality and might contribute to the compromised fertility of dairy cows during this season. The present study examined the association between seasonality, semen quality and its potential to survive cryopreservation. A comparison between semen collected during the summer (July to August) and that collected during the winter (November to December) revealed the summer semen to be inferior, as reflected by low motility and high mortality of sperm. Furthermore, samples that were defined as good quality had changes in lipid concentration and fatty-acid composition in both the seminal plasma and cell compartment. In particular, semen collected during the summer had reduced levels of polyunsaturated arachidonic acid (20:4; P<0.05) and decreased levels of linoleic acid (18:2; P<0.05) in the cell compartment; corresponding reductions in cholesterol (P<0.06) and fatty-acid concentrations (P<0.001) were detected in seminal plasma of semen collected during the summer. In addition, we provided the first evidence for the existence of a very-low-density lipoprotein receptor (VLDLr) in bovine sperm, suggesting a mechanism for sperm utilization of extracellular lipids. Interestingly, the expression of VLDLr was three-fold greater in samples collected during the winter than in those collected in the summer (P<0.01) and was negatively associated with saturated fatty-acid concentration (P<0.018) but not with that of cholesterol. An opposite pattern was noted for samples obtained during the summer; mRNA expression of VLDLr was negatively associated with cholesterol concentration (P<0.01) but not with that of saturated fatty acids. Such modifications associated with extracellular lipid utilization and fatty-acid composition might explain, in part, the reduced quality of summer semen.  相似文献

The influence of linoleic acid upon the sedimentation of washed erythrocytes from multiple sclerosis patients and controls was investigated in the present study. Linoleic acid, especially in the concentration of 250 microM and 500 microM, caused slowing of erythrocyte sedimentation and this effect was significantly greater in multiple sclerosis than in controls. These findings with all probability reflect an unsaturated fatty acid-dependent membrane alteration in multiple sclerosis and are analogues to results obtained earlier by several authors by using cytopherometric measurements.  相似文献

One hundred and sixteen patients with acute remitting multiple sclerosis (MS) took part in a double-blind controlled trial of treatment with polyunsaturated fatty acids and were randomly allocated to one of four groups. Two groups received linoleic acid, one alone as a spread and one with gamma-linolenic acid in capsules (Naudicelle); and two control groups received oleic acid, one as a spread and one in capsules. Rates of clinical deterioration and frequencies of attacks were not significantly different between treated and control groups. Exacerbations were shorter and less severe in patients receiving a high dose of linoleic acid than in controls, but those receiving a lower dose--that is, Naudicelle--showed no such difference. Thus supplementing the diet with 20 g linoleic acid marginally affected the duration and severity of relapses of MS but had no effect on overall disability. The dose of Naudicelle used provided insufficient supplementation.  相似文献

Pregnant Wistar rats were fed a fatfree diet from day 16--22 of pregnancy. On day 22, the fatty acid components of cholesterol esters, triglycerides, free fatty acids and phospholipids of maternal (brain, muscle, serum, white adipose tissue, liver) and fetal (brain, carcass, serum, liver) tissues, including the placenta, were examined gaschromatographically for the participation of linoleic and arachidonic acid. In all fetal and maternal organs the linoleic acid levels in the fatty acid patterns were strongly reduced. The alterations nearly always involved all the lipid fractions of a tissue and were mostly equal within a tissue. The strongest decreases of linoleic acid occurred in the placenta, and the weakest, in the lipids of maternal muscle and maternal adipose tissue. The linoleic acid alterations were principally similar in fetal and the corresponding maternal tissues, while being less pronounced in case of maternal muscle. The participation of arachidonic acid in the fatty acid pattern is completely retained in the lipids of fetal organs, and is even enhanced in those of the placenta.  相似文献

To investigate mechanisms controlling the concentration of serum cholesterol we studied its relationship to the proportions of fatty acids in the lipids of adipose tissue of patients with multiple sclerosis. In 26 men the serum cholesterol concentration had a significant multiple linear regression on the proportion of palmitic, palmitoleic, and grouped longer-chain acids in adipose tissues. In 29 women the serum cholesterol had a significant regression on the proportion of palmitic acid and age. Whether these observations would hold for normal people must be determined in future studies.  相似文献

The gas chromatographic method was used to study the changes in the fatty acid composition of the sum of the blood serum and erythrocyte membrane lipids in healthy male volunteers under the conditions of simulation in the ground-based hyperbaric set of long dives at depths of up to 350 m in the oxygen-helium and oxygen-neon media and in the readaptation period. At a depth of 100 m, a decrease in the serum content of the sum of saturated acids (palmitic (16:0)n and stearic (18:0) acids) and a 1.5-fold increase in the linoleic acid content compared to the background was found. A comparison of the individual changes in the blood serum and erythrocyte membrane fatty acids, as well as the unsaturation index (UI, the ratio of the sum of the unsaturated fatty acid concentrations multiplied by the number of double bonds in the molecule to the sum of the saturated fatty acid concentrations) and the R index (the erythrocyte to the serum fatty acid content ratio), showed that, under the experimental conditions and in the period of readaptation, the main changes occurred in the erythrocyte membranes, in which the unsaturated fatty acid content increased, and the linoleic and, especially, the arachidonic acid contents changed in different directions, depending on the depth of the dive. In the readaptation period, the linoleic and arachidonic acid contents decreased twofold on the first day and exceeded even the background level on the fourth day. The data obtained give evidence for the adaptive role of the erythrocyte membrane lipids upon exposure of humans to a hyperbaric medium with a changed gas composition.  相似文献

Changes in the fatty-acid composition of human adipose tissue before birth and during infancy and childhood were studied in Tanzania and compared with data for British and Dutch infants in relation to their diet. From the 32nd to the 37th week of gestation in Tanzania the proportion in the body fat of the unsaturated fatty acid linoleic acid tended to rise, suggesting an adequate supply of this essential fatty acid from the mother to the fetus. At term 2.5% of the total fatty acids of the body fat was linoleic acid, which corresponded with values in Dutch newborn infants but was significantly higher than those in British infants. During infancy in Tanzania the composition of the fat showed a dramatic increase in the proportions of the saturated fatty acids lauric acid and myristic acid, which did not occur in Dutch and British infants. The proportion of linoleic acid increased to 8%. These changes were a reflection of the fatty-acid composition of the fat in the human milk that the infants received. During weaning (1-2 years of age) the fatty-acid composition changed only slightly. The specific fatty-acid composition of the fat in Tanzanian breast milk may have a beneficial influence on the extent of intestinal absorption in the newborn child.  相似文献

Rats were fed diets containing a high level of saturated fatty acids (hydrogenated beef tallow) versus a high level of linoleic acid (safflower oil) at both low and high levels of fish oil containing 7.5% (w/w) eicosapentaenoic and 2.5% (w/w) docosahexaenoic acids for a period of 28 days. The effect of feeding these diets on the cholesterol content and fatty acid composition of serum and liver lipids was examined. Feeding diets high in fish oil with safflower oil decreased the cholesterol content of rat serum, whereas feeding fish oil had no significant effect on the cholesterol content of serum when fed in combination with saturated fatty acids. The serum cholesterol level was higher in animals fed safflower oil compared to animals fed saturated fat without fish oil. Consumption of fish oil lowered the cholesterol content of liver tissue regardless of the dietary fat fed. Feeding diets containing fish oil reduced the arachidonic acid content of rat serum and liver lipid fractions, the decrease being more pronounced when fish oil was fed in combination with hydrogenated beef tallow than with safflower oil. These results suggest that dietary n-3 fatty acids of fish oil interact with dietary linoleic acid and saturated fatty acids differently to modulate enzymes of cholesterol and fatty acid metabolism.  相似文献

A semi-synthetic diet (Vivonex) was administered via nasogastric tube to three cystic fibrosis patients with pancreatic exocrine deficiency for 14 days to gain weight. Dietary essential fatty acids were provided as safflower oil, which constituted 1.3% of total calories. Plasma and red blood cells were analyzed for the content and composition of lipids at the start of the diet and at days 7 and 14 of the dietary period, and the results were correlated with the morphology of the cells. Feeding Vivonex to the patients led to an essential fatty acid deficiency, which was manifested in a 50% decrease in the linoleic acid content of the phosphatidylcholine of plasma and red blood cells at days 7 and 14 and in a 20% decrease in the linoleic acid content of red cell phosphatidylethanolamine at day 14. There was no significant alteration in the levels or composition of the other phospholipid classes and in the free cholesterol/phospholipid ratio. The decrease in the linoleic acid content of the erythrocytes was accompanied by a dramatic increase in the proportion of cells as echinocytes. We conclude that restricted linoleic acid availability in cystic fibrosis patients causes a change in red blood cell shape either directly by decreasing the linoleoylphosphatidylcholine content of the membrane or indirectly by affecting enzyme activity.  相似文献

Male SS/Jr rats were placed on a specially formulated, high-cholesterol, low-sodium diet at 3 weeks of age. Of the 50 animals on the diet, 40 developed skin lesions ranging from focal areas of alopecia to diffuse areas of moist dermatitis on the head, face, ear pinnae, and neck. Similar lesions were noted later in 17 of 36 SS/Jr rats in a second study group. Histopathologic findings from two affected animals revealed diffuse, hyperplastic, ulcerative dermatitis, with bacterial colonies of cocci in superficial crusts, as well as chronic hepatic inflammation with hepatocellular glycogen and sinusoidal macrophage aggregates suggestive of lipidosis. Results of a fatty-acid profile of the affected rats showed serum linoleic acid levels of 931 to 1566 micromol/liter, whereas those for control (SS/Jr) samples ranged from 2711 to 3145 micromol/liter. Dietary analysis of the specially formulated diet showed that it contained only 0.225% linoleic acid, which is below the recommended level of 0.3 to 0.6%. In light of the clinical and dietary findings, a diagnosis of linoleic acid deficiency was made. The food manufacturer revised its dietary formulation to increase the linoleic acid content to 1.05%, and no further cases of dermatitis developed in any subsequent groups of rats maintained under the same study protocol.  相似文献
