In this study, the morphology of sagittal otoliths of the plainfin midshipman fish Porichthys notatus was compared between populations, sexes and male alternative reproductive phenotypes (known as ‘type I males or guarders’ and ‘type II males or sneakers’). Sagitta size increased with P. notatus size and changes in shape were also detected with increasing body size. Porichthys notatus sagittae begin as simple rounded structures, but then elongate as they grow and take on a more triangular and complex shape with several prominent notches and indentations along the dorsal and caudal edges. Moreover, the sagittae of the two geographically and genetically distinct populations of P. notatus (northern and southern) differed in shape. Porichthys notatus from the north possessed taller sagittae with deeper caudal indentations compared to P. notatus from the south. Sagitta shape also differed between females and males of the conventional guarder tactic. Furthermore, guarder males had smaller sagittae for their body size than did sneaker males or females. These differences in sagittal otolith morphology are discussed in relation to ecological and life history differences between the sexes and male tactics of this species. This is the first study to investigate teleost otolith morphology from the perspective of alternative reproductive tactics.  相似文献

Understanding patterns of connectivity among marine fish populations with demersal adults and pelagic larvae is critical for effective conservation of west coast rockfishes. The brown rockfish (Sebastes auriculatus) occurs in nearshore habitat and is common from northern Baja California, Mexico to northern California, rare off the outer coast of Oregon and Washington and again common in the inland waters of Puget Sound, Washington. Here we examine patterns of microsatellite DNA diversity from throughout the species’ range as an indirect measure of long-term trends in larval dispersal. Genetic divergence was large and highly significant over all populations (F ST=0.056, P<0.0001), and was significantly correlated with geographic distance when considering coastal populations. The best estimates of mean coastal dispersal distance were on the order of 10 km or less per generation. Diversity was relatively low in the Puget Sound, suggesting that Puget Sound rockfish populations experienced a post-glacial founder effect followed by genetic isolation and low effective population size. Puget Sound individuals appeared to have recent mixed ancestry as a result of introgression with S. maliger and S. caurinus. Genetic isolation of Puget Sound fish provides a basis for consideration as a Distinct Population Segment (DPS) under the provisions of the Endangered Species Act. We recommend that coastal brown rockfish fisheries be managed at regional rather than coast-wide scales, and that design of marine reserve networks considers the surprisingly low realized dispersal distance of some species with high dispersal potential.  相似文献

Synopsis The midshipman fish, Porichthys notatus, is a visually active nocturnal predator. It must acquire exogenous sources of luciferin to remain luminescent and feeds on a variety of luminescent prey. Its ecomorphological adaptations for nocturnal predation were examined by observing predation on zooplankton illuminated by dinoflagellates. Its visual sensitivity is well adapted for detection of its own luminescence and its prey's luminescence. P. notatus can detect the luminescence of dinoflagellates and use this indirect light to increase predation on nonluminescent prey. Flash frequency and duration modulated predation rates. The interval between flash onset and strike initiation regulated strike success. High prey concentration decreased predation success due to increased optical and mechanosensory noise. Other ecomorphological adaptations of this predator to its special photic environment include a pigmented digestive tract, duplex retina with the capacity to discriminate emission spectra, contractible pupil, and the potential to counter illuminate. These morphological adaptations combined with an ambush predator style allow effective predation while minimizing exposure risk.  相似文献

Synopsis The painted greenling, Oxylebius pictus, a hexagrammid fish of shallow, northeastern Pacific rocky habitats was observed and collected near the center of its range at Monterey Bay, California, and near the northern limit of its distribution at Puget Sound. Life history characteristics were compared for breeding stocks from the two areas. Mean and maximum adult body sizes (lengths and wet weights) and longevities of both sexes are greater at Puget Sound. Also, both males and females reach sexual maturity at a smaller body length at Monterey. Age group 0 and I juveniles are twice as abundant as adults at Monterey but rare at Puget Sound. Monterey females have greater relative fecundities. Egg development rates are similar at equivalent temperatures; eggs are smaller, however, at Monterey, and thus embryos are smaller at hatching. Length increase is faster for Puget Sound juveniles; adult fish in the two areas grow in length at comparable rates, although weight per body length is consistently greater for Monterey fish. These findings reflect a poorer survivorship at Monterey, where natural predation on fertilized eggs and the mortality rates of adult males are greater.  相似文献

ABSTRACT

Nuttall's white-crowned sparrow Zonotrichia leucophrys nuttalli and the Puget Sound white-crowned sparrow Z. i. pugetensis intergrade through a zone of hybridization in northern California. The songs of the two differ in the sequence of syllable types and in the phonology of the complex syllables. We did a playback experiment to males of both subspecies in the field and to females of both subspecies in the laboratory to determine their response to the two subspecies songs. By making alterations of features of the songs, we also attempted to identify characteristics that account for subspecific recognition. Males of both subspecies were more responsive to their own subspecies song than to that of the other subspecies. Substitution of Puget Sound complex syllables into the Nuttall's song caused a reduction in response by male Nuttall's, but alteration of a Nuttall's song to a Puget Sound sequence of syllables had no effect. Females of both subspecies gave more sexual displays when played a song of their own subspecies than when played that of the other subspecies. Female Nuttall's gave an intermediate response when played Nuttall's songs with Puget Sound complex syllables or when played Nuttall's songs altered to a Puget Sound sequence. Puget Sound females did not discriminate between their own subspecies song and either of the altered stimulus songs separately. They did respond less to a song that combined both alterations than to their own subspecies song.  相似文献

The luciferin of the ostracod Vargula hilgendorfii (formerly Cypridina hilgendorfii) is often termed Cypridina luciferin in the scientific literature, but, to avoid ambiguity and based on a review of the literature, is best referred to by the more inclusive term cypridinid luciferin. This situation exemplifies how new knowledge in systematics and the resulting nomenclatural changes can result in evolutionary insights. Genetic substructure among groups of Pacific harbor seals, Phoca vitulina richardsi , along the western coast of the United States was investigated using mitochondrial DNA sequences. Blood and tissue samples were removed from 86 seals inhabiting Puget Sound and the Pacific coasts of Washington, Oregon, and California. A 320 base-pair segment of the control region was amplified using the polymerase chain reaction and directly sequenced. These data indicated a high level of diversity. Thirty variable sites were found that define 47 mitochondrial haplotypes. Among groups of P. v. richardsi sampled, 5 haplotypes were shared, but most (42) were unique to a locality. Haplotypic frequency and an Analysis of Molecular Variance (A mova ) revealed significant differences ( P = 0.001) among regions. Phylogenetic analysis indicated Puget Sound seals possess unique divergent lineages not found in seals from the coasts of Washington, Oregon, and California. These lineages may represent haplotypes from north of Washington, which is consistent with late reproductive timing of harbor seals from Puget Sound.  相似文献

Several rockfish species (genus Sebastes) along the northeastern Pacific Ocean have rapidly declined in abundance owing in part to overfishing. A striking exception is the dwarf-like Puget Sound rockfish Sebastes emphaeus, whose densities have increased by several orders of magnitude over the last several decades. To describe their genetic structure, we sequenced 395 bp from the mitochondrial control region of 128 S. emphaeus adults from 5 locations spanning approximately 120 km of the Northwest Straits of Washington state. We detected no significant genetic differentiation among these populations and substantial genetic variation within populations, a pattern that may indicate high levels of ongoing gene flow. Preliminary data from 2 microsatellite loci are also consistent with panmixia. The mtDNA sequences also suggest that Puget Sound rockfish populations have expanded substantially since the retreat of Pleistocene glaciers made habitat in Puget Sound region available approximately 12,000 years ago.  相似文献

A study of the oxygen consumed per lumen of luminescence during oxidation of Cypridina luciferin in presence of luciferase, gives 11.4 x 10–5 gm. oxygen per lumen or 88 molecules per quantum of λ = 0.48µ, the maximum in the Cypridina luminescence spectrum. For reasons given in the text, the actual value is probably somewhat less than this, perhaps of the order of 6.48 x 10–5 gm. per lumen or 50 molecules of oxygen and 100 molecules of luciferin per quantum. It is quite certain that more than 1 molecule per quantum must react. On the basis of a reaction of the type: luciferin + 1/2 O2 = oxyluciferin + H2O + 54 Cal., it is calculated that the total efficiency of the luminescent process, energy in luminescence/heat of reaction, is about 1 per cent; and that a luciferin solution containing 4 per cent of dried Cypridina material should rise in temperature about 0.001°C. during luminescence, and contain luciferin in approximately 0.00002 molecular concentration.  相似文献

The plainfin midshipman fish Porichthys notatus has both interand intra-sexual dimorphism in the sound-producing (vocal or sonic) muscles attached to the swimbladder wall. The “Type I” and “Type II” male morphs differ in that dramatic structural changes related to sexual maturity occur in the mass, the area of mitochondria-filled sarcoplasm, and the myofiber number of the sonic muscles of Type I males, but not in those of Type II males (nor of females). Androgen implantation for 9 weeks markedly increased the relative sonic muscle size in juvenile males, juvenile females, and Type II males, whereas estradiol or cholesterol treatment did not. The principal androgen effect on myofiber structure was an increase in the area of mitochondria-filled sarcoplasm. The ratio of sarcoplasm area to myofibril area (Sr/Mf) increased by 1.4- to 2-fold in myofibers of all androgen-treated groups, with the greatest structural change occurring in juvenile males. When androgen implants were removed from juvenile males, the muscle mass and Sr/Mf ratio reverted toward the unimplanted juvenile phenotype. Total fiber number in sonic muscle increased significantly in juvenile males following androgen implantation but did not detectably change in juvenile females or Type II males. These results suggest: (1) sonic muscle in Porichthys notatus is an androgen target tissue, (2) fiber structure and fiber number are androgen-sensitive features, and (3) there exist sex- and morph-specific patterns of sonic muscle responsiveness to androgen implants. © 1993 Wiley-Liss, Inc.  相似文献

Since the 1970s, Puget Sound, Washington State, USA, has experienced an increase in detections of paralytic shellfish toxins (PSTs) in shellfish due to blooms of the harmful dinoflagellate Alexandrium. Natural patterns of climate variability, such as the Pacific Decadal Oscillation (PDO), and changes in local environmental factors, such as sea surface temperature (SST) and air temperature, have been linked to the observed increase in PSTs. However, the lack of observations of PSTs in shellfish prior to the 1950s has inhibited statistical assessments of longer‐term trends in climate and environmental conditions on Alexandrium blooms. After a bloom, Alexandrium cells can enter a dormant cyst stage, which settles on the seafloor and then becomes entrained into the sedimentary record. In this study, we created a record of Alexandrium spp. cysts from a sediment core obtained from Sequim Bay, Puget Sound. Cyst abundances ranged from 0 to 400 cysts · cm?3 and were detected down‐core to a depth of 100 cm, indicating that Alexandrium has been present in Sequim Bay since at least the late 1800s. The cyst record allowed us to statistically examine relationships with available environmental parameters over the past century. Local air temperature and sea surface temperature were positively and significantly correlated with cyst abundances from the late 1800s to 2005; no significant relationship was found between PDO and cyst abundances. This finding suggests that local environmental variations more strongly influence Alexandrium population dynamics in Puget Sound when compared to large‐scale changes.  相似文献

Among sixteen groups of luminous forms investigated by the author, in only four (fireflies, Pholas, ostracods, and Odontosyllis) is it possible to demonstrate the luciferin-luciferase reaction. In many groups this is probably due to the small amount of these substances present in the luminescent organism or to their instability. In the medusæ and pennatulids, despite a large amount of luminescent material, luciferin and luciferase cannot be demonstrated. This does not appear to be due to the presence of luciferin and luciferase in equivalent proportion, or to their instability. In fact, one is led to the conclusion that luciferin and luciferase do not exist in these forms, but such a conclusion must be regarded as merely tentative, in view of the fundamental character of the luciferin-luciferase reaction. Luciferin of one form will not luminesce with the luciferase of another form or vice versa, unless very closely related (Cypridina and Pyrocypris). All experiments emphasize the specificity of the light producing substances of Cypridina.  相似文献

The macroscopic and microscopic diversity of potential food items available in the nests of plainfin midshipman Porichthys notatus were quantified and compared with items that were found in the stomach and intestine (digestive tract) of the guarding males. In this species, males occur as one of two possible reproductive morphs: guarder males that care for young and sneaker males that parasitize the courtship and care of guarder males. Although it was predicted that guarder males would have fewer feeding opportunities due to their confinement to the nest, they in fact had more food items in their digestive tracts than did sneaker males and females. Date in the breeding season (a proxy of care duration) and body condition were not correlated with the amount of food consumed by guarder males. The main type of food consumed was P. notatus embryos; 69% of all guarder males sampled had cannibalized offspring. By comparing the diet of both sexes and tactics, this study sheds light on some of the strategies designed to cope with the costs of providing parental care.  相似文献

There seems to be very little doubt but that luciferase is a protein or so closely associated with proteins that their removal destroys its characteristic properties. The particular group of proteins to which it belongs may be arrived at by a process of exclusion, and only the group of albumins has properties which agree completely with those of luciferase. Dubois believes Pholas luciferase to be an oxidizing enzyme similar to the oxydones of Battelli and Stern because it is readily destroyed by fat solvents such as chloroform, strong alcohol, etc. He has detected iron in a luciferase solution which has dialyzed against running water for a long time, and believes it to be made up of protein in combination with iron and to act as an "oxyzymase ferrique." Cypridina luciferase, on the other hand, is not readily destroyed by fat solvents. Toluene and chloroform are good preservatives, and I often make use of them for this purpose, keeping the luciferase solutions for many months. Professor A. H. Phillips of Princeton University has very kindly analyzed some whole dried Cypridinœ for me and finds iron, copper, and manganese but no zinc or vanadium to be present. Whether these metals are connected with the action of Cypridina luciferase is uncertain, but it is significant that all three of the metals thought to be concerned in organic oxidations are present. Although a large amount of luciferin mixed with a small amount of luciferase will use up all the latter, I agree with Dubois that luciferase has sufficient properties in common with the enzymes as a class to be considered an enzyme. The peroxidases are well known to be used up in the reactions they accelerate. All workers on enzymes agree that the more enzymes are purified the less active they become. The chemical procedures necessary to remove foreign material bring about irreversible changes in the enzyme itself, a characteristic also of many protein groups and of the colloidal state in general. This is true in the case of luciferase, for the crude luciferase solution is the most active preparation that can be obtained. I believe that Cypridina luciferase should be placed in a class of oxidizing enzymes by itself—a group having the chemical reactions of an albumin, possibly in combination with some heavy metal, and which as far as we know, acts specifically on only one substance, Cypridina luciferin. It resembles the plant peroxidases in resisting the action of chloroform, toluene, etc., but will not oxidize any of the hydroxyphenol or aminophenol compounds so readily oxidized by the peroxidases, nor will the peroxidases oxidize luciferin with light production. Dubois'' researches show that Pholas luciferase differs in some properties from Cypridina luciferase, and my own work indicates that firefly luciferase is more like that of Pholas. A comparative study of other species of luminous animals is needed in order to delimit more accurately the class of luciferases as a whole. Luciferin presents many characteristics in common with the proteins, but two, which, to say the least, throw doubt on its protein nature: (1) its peculiar solubility (in alcohols, esters, and glacial acetic acid), (2) and its resistance to digestion by proteases, even by trypsin which has almost universal digestive action. These two peculiarities have been discussed above. We can only say that if a protein, luciferin must belong to a new group differing from known natural proteins in these respects. In general characteristics this new group would fall somewhere on the border-line between the proteoses and peptones. It would not be surprising to find in nature proteoses or peptones soluble in absolute alcohol. We know also that some NH-CO linkages of proteins are broken down with great difficulty by trypsin as it is difficult to obtain a tryptic digest of protein which does not give the biuret reaction, and the work of Fischer and Abderhalden has shown that certain artificial polypeptides are not digested by pure activated pancreatic juice. We have, then, three possibilities. Luciferin is (1) either a natural proteose not attacked by trypsin, or (2) if attacked by trypsin, its decomposition products (presumably amino-acids) still contain the group oxidizable with light production, or (3) it is not a protein at all. I believe that luciferin has too many protein characteristics to conform to the last possibility. I have been unable to oxidize with light production various mixtures of amino-acids (from beef and casein) by means of luciferase and consequently am led to believe that luciferin is a new natural proteose, soluble in absolute alcohol and not digested by trypsin. Dubois believes Pholas luciferin to be a natural albumin with acid properties. Cypridina luciferin could not possibly be regarded as an albumin, but it is very likely that the luciferins of different species of luminous animals differ in certain characteristics. As in the case of the luciferases, we know that the luciferins are not identical substances, and only future work can determine in what particulars they differ. Some solubility, oxidation, reduction, and compound-forming characteristics of extracts of Cypridina luciferin have been presented. A method of purification has been described which increased the amount of luciferin per unit of dry weight, as measured by the total light emitted, to about two thousand times that in the dry starting material. Population genetic structure was determined for the planktonic diatom Ditylum brightwellii (West) Grunow in two connected estuaries—Puget Sound and the Strait of Juan de Fuca (WA, USA). Three genetically distinct populations were detected that were characterized by different microsatellite allele distributions and unique alleles. Isolates from the two most genetically diverged populations displayed identical full‐length 18S rDNA sequences suggesting that either a single or two recently diverged species were sampled. The extent of genetic differentiation between populations was not correlated with distance between water samples or time between sampling. Instead, distinct populations were associated with different estuaries. In Puget Sound waters, one population was detected three times over the course of 28 months. Cells from this population were likely maintained inside Puget Sound over long periods through water recirculation within the Sound. In Strait of Juan de Fuca waters, two additional populations were detected. Maximum growth rates of Puget Sound isolates were significantly different from Strait of Juan de Fuca isolates, indicating that populations were composed of cells with different physiological capabilities. The genetic and physiological differentiation observed between populations from intermixing estuaries suggested that genetic exchange between populations was restricted through differential selection. Despite the potential for widespread dispersal in planktonic organisms, it appears that populations with distinct genetic and physiological characteristics can be maintained over long time periods through a combination of hydrology and differential selection.  相似文献

Bioluminescent oxidation of Cypridina luciferin yields CO2 besides oxyluciferin and light. The exchange of oxygen between the CO2 and H2O of the solvent becomes significant when less than approximately 1 μmol of luciferin is reacted in 4 ml of buffer solution, and the exchanged oxygen in CO2 markedly increases by decreasing the amount of luciferin. Such an exchange is to be expected in any such system which produces CO2 in aqueous solution, and must be taken into account in interpreting the results of experiments.  相似文献

The chemiluminescence of the Cypridina luciferin analogue, 2-methyl-6-(p-methoxyphenyl)-3,7-dihydroimidazo[1,2-a]pyrazin-3-one (MCLA) was observed at 462nm in the presence of horseradish peroxidase (HRP) and the total spectrum of light emitted was found to depend linearly on HRP concentration. Methods for the determination of HRP concentration using the chemiluminescence was investigated. HRP could be detected in the range from 100 pmol/L to 100nmol/L under the optimum condition, H2O2 (10mmol/L) and MCLA (10μmol/L) at pH 5.8.  相似文献

The present study is a biochemical characterization of the photophore lenses of the midshipman fish, Porichthys notatus, a species that bears 800 photophores distributed over the body surface. The biochemical properties of the photophore lenses were compared with those of the eye lens with which they share a similar developmental origin and analogous function. To achieve a high refractive index, the vertebrate eye lens has a relatively high concentration of structural proteins (20–50%, depending on species) and a simple protein composition, that is, relatively few proteins are synthesized in comparison to other tissues. Similarly, the photophore lenses of P. notatus had a relatively high protein concentration (average = 29%, n = 5) and approximately 60% of the total soluble protein was represented by two subunit species of 33 kD and 35 kD on denaturing polyacrylamide gels. The structural proteins of the eye lens are of two principle types: 1) and polypeptides which belong to vertebrate lens-specific crystallin families, and, 2) enzymes recruited into the lens which take on the function of structural proteins. Here, we report that the two major photophore lens subunits of 33 kD and 35 kD are biochemically similar to each other, but are clearly distinct from any of the previously characterized crystallins. Therefore, we propose that photophore lenses appear to recruit a novel protein.  相似文献

Using the plainfin midshipman fish Porichthys notatus, a species with alternative reproductive tactics (ARTs), we investigated how sperm maturation shapes sperm competitive abilities. We compared sperm performance and morphology before and after final sperm maturation by sampling sperm from the testes and stripped ejaculates of guarders and sneakers. In accordance with sperm competition risk theory, ejaculates from sneaker males had three times as much sperm as ejaculates from guarder males and sneaker males produced faster swimming sperm than guarder males, but this was only the case after final sperm maturation had occurred. Additionally, fully mature sperm found in ejaculates had larger heads and midpieces than sperm found in the testes. These results emphasize the important role played by non-sperm components of an ejaculate in mediating sperm performance and potentially also morphology.  相似文献
