Antiradical properties of components of juice of the fresh ginger root and essence and oleoresin of ginger (Zingiber officinale R.) were studied and compared with the properties of the ionol synthetic antioxidant. A reaction with the stable free 2,2-diphenyl-1-picrylhydrazyl radical was used for the evaluation of the antiradical properties. Rates of the fast phase of this reaction and equivalent concentrations of the antiradical components in the ginger samples were determined from kinetic curves of this reaction phase. The values of EC50 and antiradical efficacy (AE) found for the ginger oleoresin were close to those of ionol and characteristic of highly active natural antioxidants, whereas the same values for the ginger essence and juice were two orders of magnitude lower. The ginger preparations belonged to the antiradical compounds of prolonged action according to their kinetic parameters.  相似文献

The bioactive compounds content and the antioxidant activity (AA) of twelve fruits native to the Cerrado were compared with the Red Delicious apple by means of the antiradical efficiency (using the 2,2-diphenyl-1-picrylhydrazil assay/DPPH), ferric reducing antioxidant power (FRAP) and the β-carotene/linoleic system. The antiradical efficiency (AE) and the kinetic parameters (Efficient concentration/EC50 and time needed to reach the steady state to EC50 concentration/TEC50) of the DPPH curve were also evaluated for comparison with the Trolox equivalent (TE) values. A strong, significant and positive correlation was observed between the TE and AE values, whereas a weak and negative correlation was observed between TE and EC50, suggesting that the values of AE and TE are more useful for the determination of antiradical activity in fruits than the widely used EC50. The total phenolic content found in the fruits corresponded positively to their antioxidant activity. The high content of bioactive compounds (flavanols, anthocyanins or vitamin C) relative to the apple values found in araticum, cagaita, cajuzinho, jurubeba, lobeira, magaba and tucum corresponded to the high antioxidant activity of these fruits. Flavanols and anthocyanins may be the main bioactive components in these Cerrado fruits. The daily consumption of at least seven of the twelve Cerrado fruits studied, particularly, araticum, cagaita, lobeira and tucum, may confer protection against oxidative stress, and thus, they may prevent chronic diseases and premature aging. The findings of this study should stimulate demand, consumption and cultivation of Cerrado fruits and result in sustainable development of the region where this biome dominates.  相似文献

We investigated the antioxidant and enzyme inhibitory activities and chemical composition of the hydro-distilled essential oil (0.35% yield) from aerial parts of Thymus spathulifolius. Antioxidant capacity of the oil was assessed by different methods including free radical scavenging (DPPH and ABTS), reducing power (FRAP and CUPRAC) and phosphomolybdenum assay. Inhibitory activities were analyzed against acetylcholinesterase (AChE), butyrylcholinesterase (BChE), α-amylase, α-glucosidase, and tyrosinase. Twenty-one constituents were identified representing 97.2% of the total oil with thymol (50.5%), borneol (16.7%) and carvacrol (7.7%) as the major components. The essential oil exhibited good antioxidant activity with IC50 values of 3.82 and 0.22?mg/mL determined by free radical scavenging DPPH and ABTS, respectively. EC50 values of FRAP and CUPRAC were found to be 0.12 and 0.34?mg/mL, respectively. The results of the present study support the uses of T. spathulifolius essential oil as a source of natural antioxidants and bioactivities for functional foods and phytomedicines.  相似文献

Thymus vulgaris L. (thyme), Origanum majorana L. (marjoram), and Origanum vulgare L. (oregano) were used to determine whether light modification (plants grown under nets with 40% shaded index or in un-shaded open field) could improve the quantity and quality of essential oils (EOs) and antioxidant activity. The yield of EOs of thyme, marjoram, and oregano obtained after 120 min of hydrodistillation was 2.32, 1.51, and 0.27 mL/100 g of plant material, respectively. At the same time under shading conditions plants synthetized more EOs (2.57, 1.68, and 0.32 mL/100 g of plant material). GC/MS and GC/FID analyses were applied for essential oils determinations. The main components of the thyme essential oil are thymol (8.05–9.35%); γ-terpinene (3.49–4.04%); p-cymene (2.80–3.60%) and caryophyllene oxide (1.54–2.15%). Marjoram main components were terpinene 4-ol (7.44–7.63%), γ-terpinene (2.82–2.86%) and linalool (2.04–2.65%) while oregano essential oil consisted of the following components: caryophyllene oxide (3.1–1.93%); germacrene D (1.17–2.0%) and (E)-caryophyllene (1.48–1.1%). The essential oil from thyme grown under shading (EC50 value after 20 min of incubation) have shown the highest antioxidant activity – 0.85 mg mL−1 in comparison to marjoram and oregano (shaded plants EC50 19.97 mg mL−1 and 7.02 mg mL−1 and unshaded, control plants EC50 54.01 mg mL−1 and 7.45 mg mL−1, respectively). The medicinal plants are a good source of natural antioxidants with potential application in the food and pharmaceutical industries. For production practice, it can be recommended to grow medicinal plants in shading conditions to achieve optimal quality parameters.  相似文献

The insecticidal activity of essential oils from 12 species of Eucalyptus (Myrtaceae) was evaluated on larvae of Aedes aegypti (L.) (Diptera: Culicidae), the most important vector of dengue and yellow fever in the Americas. Oils were obtained by hydrodistillation and their chemical composition was determined by gas chromatography coupled to mass spectrometry; yields ranged from 0.2 to 2.5%. Essential oils were mainly composed of 1,8‐cineole, α‐pinene, α‐phellandrene, β‐phellandrene, γ‐terpinene, 4‐terpineol, α‐terpineol, p‐cymene, and spathulenol. Larvicidal effects were tested on susceptible third or fourth stage Ae. aegypti larvae, determining median lethal concentration (LC50) and median effective concentration (EC50). Essential oils from Eucalyptus dunnii (Maiden), Eucalyptus gunnii (Hook), Eucalyptus tereticornis (Smith), Eucalyptus camaldulensis (Dehn), and Eucalyptus saligna (Smith) showed the best larvicidal activities with LC50 values of 25.2, 21.1, 22.1, 26.8, and 22.2, respectively. No significant differences were observed between LC50 and EC50 values of the same oil. Regression analysis revealed a significant relationship between total essential oil yields and 1,8‐cineole concentration. Significant relationships were also revealed between larval mortality and the concentration of 1,8‐cineole and p‐cymene. This indicated that Eucalyptus species with high oil yields have higher 1,8‐cineole concentrations and lower p‐cymene concentrations and have less effect on Ae. aegypti. Our results suggest the potential of controlled crossing methods to obtain Eucalyptus trees with chemical profiles having enhanced activity against this mosquito.  相似文献

Extracts from 48 marine macroalgae species (17 Chlorophyta, 8 Phaeophyta and 23 Rhodophyta) from the coasts of Yucatan and Quintana Roo (Mexico) were evaluated for antioxidant activity. The antioxidant activity was measured with the DPPH (2,2-diphenyl-1-picrylhydrasyl) method, and the phenolic content of each extract were also evaluated. All species exhibited a DPPH radical scavenging activity, and three species (Avrainvillea longicaulis, Chondria baileyana and Lobophora variegata) demonstrated great antioxidant potential with very low oxidation index EC50 (1.44 ± 0.01, 2.84 ± 0.07 and 0.32 ± 0.01 mg mL−1, respectively), significantly equivalent to EC50 of some commercial antioxidants such as α-tocopherol, ascorbic acid, BHA and BHT. Moreover, extracts of the most active species exhibited reducing activities, superoxide anion radical scavenging and inhibition of lipid peroxidation. These results suggest that some macroalgae from the Yucatan peninsula have a great antioxidant potential which could be considered for future applications in medicine, food production or cosmetic industry.  相似文献

In this study pro/anti-oxidant activities of Thymus revolutus Célak (T. revolutus C.) essential oil and its two main components on Hep G2 cells were determined. Twelve components were identified in T. revolutus C. essential oil by GC and GC-MS and the main components of the oil were cymene (32.57%) and γ-terpinene (17.18%). The essential oil showed more cytotoxicity than its two main components on Hep G2 after 24, 48 and 72 hours incubations. Membrane damage effects of the essential oil and its two main components on Hep G2 cells were increased by incubation with IC10, IC50 and IC70 concentrations. Lower essential oil (<IC50) concentrations had cytoprotective and membrane protective effects against H2O2. The free radical scavenging activity of the essential oil of T. revolutus C. (EC50 = 250±0.2 μg/mL) was superior to its two main components (neither not active). Oxidation of linoleic acid was effectively inhibited by T. revolutus C. (72.8%), while inhibition values of linoleic acid oxidation were calculated as 23.4.% and 26.7% for cymene and Γ-terpinene, respectively. These findings suggest that essential oils and its two main components had both pro-oxidant and protective (antioxidant) effects and they exhibited those effects depending on concentration.  相似文献

The present study aimed to evaluate the toxicity and biochemical changes of Tuta absoluta 3rd instar larvae affected by neemazal T/S, clove oil and ginger oil. These compounds were evaluated compared to the recommended pesticide, Coragen 20% SC. by means of sublethal concentrations, LC25 and LC50 under constant laboratory conditions. Results showed that neemazal T/S is more toxic than detected oils compared with higher toxicity of coragen with LC50 values of 57.52, 159.94, 633.38 and 930.71 μg mL?1 for coragen, neemazal, ginger oil and clove oil, respectively. There were highly significant differences between all treatments and untreated larvae. Neemazal possessed the greatest effect on activity level of most physiological parameters than selected oils. Larval content of digestive enzymes was decreased significantly 48 h after all treatments except for lipase, α-esterase and β-esterase (in case of coragen and clove oil). Also, total proteins, total carbohydrates, total lipids and total free amino acids take the same trend. Based on this study, these sublethal doses caused a significantly dose-dependent perturbation in determined components.  相似文献

The present study was designed to assess the influence of geographical factors on essential oil (EO) composition, along with antiradical potential and phytochemical contents of Ferulago angulata (Schltdl .) Boiss (Apiaceae) extracts for the first time. The aerial parts were hydrodistilled by Clevenger apparatus and subjected to gas chromatography coupled with flame ionization detector (GC/FID) and mass spectroscopy (GC/MS). The EO yields were significantly different from populations ‘Mongar’ (south‐slope, 3000 m) with 1.34±0.06 % and ‘Male‐Amiri’ (north slope, 2600 m) with 0.18±0.05 % of total oil. Thirty‐nine compounds were identified from the EOs of nine populations. α‐Pinene was the predominant component ranging from 20.84 to 49.06 % in ‘Gandomkar’ (north‐slope, 2500 m) and ‘Mongar’ (3000 m), respectively. The methanolic extract of ‘Mongar’ (north‐slope at 2500 m) possessed the highest total phenolic contents. Also, this population logically exhibited potent antiradical activity using both 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) and oxygen radical absorbance capacity (ORAC) assays with EC50 of 42.07±4.12 μg/mL and 8.34±0.21 mmol Trolox® equivalents/g, respectively. Due to its moderate free‐radical scavenging potential and high α‐pinene content, the population ‘Mongar’ might be considered as a perspective raw material in food and phytopharmaceutical industries.  相似文献

Sensitivity of 159 isolates of Zymoseptoria tritici collected from durum wheat fields in Tunisia in 2012 was analysed towards pyraclostrobin, fluxapyroxad, epoxiconazole, metconazole, prochloraz and tebuconazole using microtiter tests. All isolates were found to be highly sensitive to pyraclostrobin with EC50 <0.01 mg/l with the exception of three isolates from the same field with higher EC50 values (>0.5 mg/l). These three isolates carried a mutation in the cytochrome b gene encoding the G143A substitution. This is the first report of quinone outside inhibitors (QoI) resistance in Z. tritici in Tunisia. Sensitivity towards r fluxapyroxad was in a narrow range with EC50 values ranging between 0.013 and 0.125 mg/l, which can serve as baseline sensitivity data for the future. Demethylation inhibitors sensitivity varied across a broad range with the data indicating a slight shift in sensitivity when compared to a previous study on the 2010 population. No highly sensitive strains were isolated from samples from fields, which had received three or four DMI applications.  相似文献

The effect of two commonly used fertilizers, DAP (diammonium phosphate) and urea was studied on the freshwater flagellate Euglena gracilis using the automatic biotest ECOTOX. NOEC and EC50 values for various parameters like motility, velocity, cell shape and gravitaxis were calculated. The NOEC and EC50 values obtained for DAP were much lower than those for urea; i.e. DAP showed a stronger inhibitory effect as compared to urea. The inhibition caused by DAP increased with increasing exposure time over 24 h but urea showed no augmentation with increasing exposure time. Application of DAP resulted in an increased pH and high concentrations of ammonia but urea did neither affect the pH nor affect the ammonia concentration. Recovery experiments in deionized water after urea application showed a reconstitution of motility after 72 h. After an application of 1.35 g L−1 (24 h EC50 for motility) DAP motility recovered after 72 h but motility did not recover when the concentration was doubled (2.7 g L−1). The EC50 values obtained were compared with the EC50/LC50 values reported for other aquatic organisms and were found to be comparable with the reported values.  相似文献

EC50 and EC95 (the effective concentrations to cause inhibitions by 50 and 95%, respectively) are commonly used to express fungicide potency. Different methods are currently employed to calculate EC50 and EC95 values. In this study, EC50 and EC95 values for fungicide epoxiconazole against 34 isolates of Sclerotinia sclerotiorum were calculated with seven different methods. Results showed that for both EC50 and EC95 calculations, there was no significant difference among three statistical programs IBM spss ®, GraphPad Prism® and dps ® (P  0.066). Methods linear log (linear regression of mycelial growth inhibition vs. logarithmic concentration) and interpolation log (linear interpolation from inhibition and logarithmic concentration data) were not significantly different (P  0.058) from IBM spss in EC50 calculations. These results indicate that among the seven methods, the three statistical programs IBM spss , GraphPad Prism, dps and linear log method are appropriate for EC50 calculations. But for EC95 calculations, only the three statistical programs are recommended, and GraphPad Prism is likely to give a little higher values than spss and dps .  相似文献

Chitosans were prepared by H2O2 oxidative depolymerization from squid pens with low molecular weights (LMW) of 13,025, 7011, 4169, 2242 and 963 Da. The bile acid binding capacities and antioxidant properties of LMW chitosans were studied in vitro. LMW chitosans exhibited stronger bile acid binding capacities than that of chitosan. The scavenging ability of LMW chitosans against DPPH radicals improved with increasing concentration, and EC50 values were below 1.3 mg/mL. The EC50 values of LMW chitosans against hydroxyl radicals ranged from 0.93 to 3.66 mg/mL. All LMW chitosans exhibited a strong ferrous ion chelating effect and reducing power. At 1 mg/mL, the scavenging ability of chitosan-963 towards superoxide radicals was 67.76%. These results indicated that LMW chitosans which have stronger bile acid binding capacity and antioxidant activities may act as potential antioxidants in vitro.  相似文献

Many grass species exist in the oil exploration areas of North Dakota. The objective of this study was to evaluate seed germination of 65 grass species affected by crude oil. Germination of all species was reduced by crude oil, ranging from 4.3 to 100%. Twenty-eight species were tolerant, 29 moderately tolerant, 6 moderately sensitive, and 2 sensitive. Based on the tolerance levels, the following were used to further test the dose response to crude oil: strong creeping red fescue (Festuca rubra L. ssp. rubra), perennial ryegrass (Lolium perenne L.), orchardgrass (Dactylis glomerata L.), buffalograss [Buchloe dactyloides (Nutt.) Engelm.], little bluestem [Schizachyrium scoparium (Michx.) Nash], witchgrass (Panicum capillare L.), sand dropseed [Sporobolus cryptandrus (Torr.) Gray], Johnsongrass [Sorghum halepense (L.) Pers.], and smooth crabgrass [Digitaria ischaemum (Schreb.) Schreb. ex Muhl.]. The EC50 of germination and biomass was effective in ranking the 9 species. Buffalograss, sand dropseed, and orchardgrass were ranked as the most tolerant species with EC50 values of 0.1, > highest concentration tested, 0.05 m3 m?3 (P < 0.05), respectively. Smooth crabgrass and little bluestem were ranked as most sensitive with EC50 values of 0.03 and 0.04 m3 m?3 (P < 0.05), respectively. This study was undertaken to elucidate the effect of the essential oil from Alpinia speciosa (EOAs) on cardiac contractility and the underlying mechanisms. The essential oil was obtained from Alpinia speciosa leaves and flowers and the oil was analyzed by GC-MS method. Chemical analysis revealed the presence of at least 18 components. Terpinen-4-ol and 1,8-cineole corresponded to 38% and 18% of the crude oil, respectively. The experiments were conducted on spontaneously-beating right atria and on electrically stimulated left atria isolated from adult rats. The effect of EOAs on the isometric contractions and cardiac frequency in vitro was examined. EOAs decreased rat left atrial force of contraction with an EC50 of 292.2 ± 75.7 μg/ml. Nifedipine, a well known L-type Ca2+ blocker, inhibited in a concentration-dependent manner left atrial force of contraction with an EC50 of 12.1 ± 3.5 μg/ml. Sinus rhythm was diminished by EOAs with an EC50 of 595.4 ± 56.2 μg/ml. Whole-cell L-type Ca2+ currents were recorded by using the patch-clamp technique. EOAs at 25 μg/ml decreased ICa,L by 32.6 ± 9.2% and at 250 μg/ml it decreased by 89.3 ± 7.4%. Thus, inhibition of L-type Ca2+ channels is involved in the cardiodepressive effect elicited by the essential oil of Alpinia speciosa in rat heart.  相似文献

Propolis is known to exhibit various phytochemical compounds that aid in several biological activities. The current study investigates the phytochemical compounds of ethanolic extract of propolis of Tetrigona apicalis (EEP) using Q-TOF LC-MS, its antioxidant properties using DPPH and ABTS+ radical scavenging assays, total phenolic (TPC) and flavonoid content (TFC), using Folin-Ciocalteu and Aluminium Chloride method, respectively, as well as proapoptotic effects, based on the selected IC50 of the cytotoxic study conducted for EEP using annexin V-FITC assay. Terpene and polyphenol were among of 17 identified compounds. The EC50 of EEP for DPPH and ABTS+ was 1.78 mg/mL and 1.68 mg/mL, while the EEP exhibited TPC and TFC values of 31.99 mgGAE/g and 66.4 mgQCE/g, respectively in which the parameters were strongly correlated. The IC50 of EEP effectively induces apoptosis in MCF7 cells. In conclusion, EEP possessed important phytochemical compounds that work excellently as antioxidants and anticancer agents.  相似文献

A novel series of acetamide-substituted derivatives and two prodrugs of doravirine were designed and synthesized as potent HIV-1 NNRTIs by employing the structure-based drug design strategy. In MT-4 cell-based assays using the MTT method, it was found that most of the new compounds exhibited moderate to excellent inhibitory potency against the wild-type (WT) HIV-1 strain with a minimum EC50 value of 54.8?nM. Among them, the two most potent compounds 8i (EC50?=?59.5?nM) and 8k (EC50?=?54.8?nM) displayed robust activity against WT HIV-1 with double-digit nanomolar EC50 values, being superior to lamivudine (3TC, EC50?=?12.8?μM) and comparable to doravirine (EC50?=?13?nM). Besides, 8i and 8k shown moderate activity against the double RT mutant (K103N?+?Y181C) HIV-1 RES056 strain. The HIV-1 RT inhibition assay further validated the binding target. Molecular simulation of the representative compounds was employed to provide insight on their structure-activity relationships (SARs) and direct future design efforts. Finally, the aqueous solubility and chemical stability of the prodrugs 9 and 10 were investigated in detail.  相似文献

The hydrodistilled oil of Cryptocarya massoy bark was characterized by GC‐FID and GC/MS analyses, allowing the identification of unusual C10 massoia lactone ( 3 , 56.2%), C12 massoia lactone ( 4 , 16.5%), benzyl benzoate ( 1 , 12.7%), C8 massoia lactone (3.4%), δ‐decalactone ( 5 , 1.5%), and benzyl salicylate ( 2 , 1.8%) as main constituents. The phytotoxic activities of the oil, three enriched fractions (lactone‐rich, ester‐rich, and sesquiterpene‐rich), and four constituents (compounds 1, 2, 5 , and δ‐dodecalactone ( 6 )) against Lycopersicon esculentum and Cucumis sativus seeds and seedlings were screened. At a concentration of 1000 μl/l, the essential oil and the massoia lactone‐rich fraction caused a complete inhibition of the germination of both seeds, and, when applied on tomato plantlets, they induced an 85 and 100% dieback, respectively. These performances exceeded those of the well‐known phytotoxic essential oils of Syzygium aromaticum and Cymbopogon citratus, already used in commercial products for the weed and pest management. The same substances were also evaluated against four phytopathogenic bacteria and ten phytopathogenic fungi, providing EC50 values against the most susceptible strains in the 100–500 μl/l range for the essential oil and in the 10–50 μl/l range for compound 6 and the lactone‐rich fraction. The phytotoxic behavior was related mainly to massoia lactones and benzyl esters, while a greater amount of 6 may infer a good activity against some phytopathogenic fungi. Further investigations of these secondary metabolites are warranted, to evaluate their use as natural herbicides.  相似文献

Cereals are staple food for many countries and are grown on millions of hectares of land, but much of the harvest is wasted due to losses by pests. To minimize these losses, many pesticides are used which are damaging to the environment and human health. There are debates to get rid of these chemicals but they are still in use at large scale. An alternative control strategy for insect pests in storage houses is the use of botanicals. In this study, four plant essential oils, two plant extracts, two herbicides, and two insecticides were used against Tribolium confusum and the comparison of toxicity was made by calculating LC50 and LT50 values. LC50 values were higher for abamectin (2.09–10.23 mg/L) and cypermethrin (3.41–11.78 mg/L) insecticides followed by neem essential oil (7.39–19.24 mg/L) and citrus extract (10.14–24.50 mg/L). However, LC50 values were maximum in case of jaman plant extract (22.38–176.42 mg/L) followed by two herbicides, Logran (19.66–39.72 mg/L) and Topik (29.09–47.67 mg/L) However, LC50 values were higher for topic herbicide (24.098 ppm) and jaman essential oil (16.383 ppm) after four days of treatment. Abamectin and cypermethrin insecticides, neem essential oil and citrus plant extract also killed adults of T. confusum quicker as compared other essential oils, extracts and herbicides. Results revealed that botanical formulations being environmentally safe could be used instead of highly hazardous pesticides for stored products’ pests. This study also elaborates the non-host toxicity of herbicides commonly applied in our agroecosystem.  相似文献

To establish a baseline sensitivity of Botrytis cinerea to pyrrolnitrin, an antibiotic produced by several biological control agents, 204 isolates were tested for sensitivity to pyrrolnitrin using a spore germination assay. The results showed that the isolates exhibited a wide range of sensitivity to pyrrolnitrin, with an 8.4-fold difference in EC50 (effective concentration to reduce spore germination by 50% comparing to the control) values between the least and the most sensitive isolates. The model-based clustering analysis indicates that the distribution of the EC50 values best fit a normal mixture model with three components and unequal variance. The less sensitive isolates were also multidrug resistant isolates. The efficacy of the pyrrolnitrin-producing Pseudomonas chlororaphis ChPhzS24 strain was tested in vitro and on tomato plants with isolates of B. cinerea having different EC50 values. Whatever the EC50 value of the isolates tested, no significant differences in sensitivity were observed towards this bacterium indicating an absence of resistance to this biological control agent within B. cinerea isolates and suggesting also that additional mechanisms of action are probably operated by this biological control agent.  相似文献
