Isolation of microorganisms, screening for desirable characters and selection of efficient strains are important steps to optimize high crop yields and improve the sustainability of the ecosystem. The objective of this study was isolate and identify Azopirillum spp. with enhanced potential to promote plant growth among the natural bacterial population associated with rhizosphere soil, roots and stem of maize collected from five maize-growing regions within the southern state of Rio Grande do Sul in Brazil. Diazotrophic microorganisms were isolated using semi-solid N-free and solid selective media NFb. In order to select the most efficient isolates as candidates for plant growth promotion, the purified bacterial strains were studied for cell morphology, and Gram staining, streptomycin resistance, as well as screened for their potential for nitrogen fixation and auxin production under sterile conditions. Among 224 isolates obtained 121 were able to fix nitrogen and produce auxin. The 30 most promising isolates produced indole-3-acetic acid (IAA) ranging in concentration from 3.51 μg to 246.69 μg IAA mg−1. Nitrogen fixation ranged from 15.43 μg to 95.21 μg of N mg protein−1 day−1 From the 30 most productive isolates, chromosomal DNA was extracted and a portion of the nifH gene was amplified and sequenced. Twenty-nine isolates were found to be similar to the Azospirillum genus and one isolate was found to be similar to Herbaspirillum seropedicae. These bacterial isolates revealed potential to increase crop productivity, however field crop experiments in Rio Grande do Sul climatic conditions should be done in order to formulate recommendations for their use as inoculants.  相似文献

Prunings of Calliandra calothyrsus, Grevillea robusta, Leucaena diversifolia and farm yard manure were applied each cropping season at 3 and 6 t dry matter ha−1 to an Oxisol in Burundi. The field plots also received basal applications of nitrogen (N), phosphorus (P) and potassium (K). Application of the tree prunings or farm yard manure decreased the concentration of monomeric inorganic aluminium (Al) in soil solution from 2.92 mg Al dm−3 in the control plots to 0.75 mg Al dm−3 in the plots receiving 6 t ha−1 Calliandra prunings. The other organic materials also decreased the concentration of monomeric inorganic aluminium in the soil solution. The lowered Al concentration led to a corresponding decrease in the percentage Al saturation of the 0–10 cm soil layer from 80% to 68%. Grain yields of maize and beans were strongly inversely related to the percentage Al saturation of the soil. This confirms that soil acidity was the main constraint to maize and beans production. The yield improvement was mainly attributed to the ameliorating effects of the organic matter application on Al toxicity. The nutrient content had less effect presumably because of fertilizer use. In the best treatments, the yield of maize increased from 0.9 to 2.2 t ha−1 and the corresponding beans yield increased from 0.2 to 1.2 t ha−1. A C Borstlap Section editor  相似文献

Summary An all glass tight growth chamber, entirely sterilizable, has been constructed to carry out axenic and gnotobiotic cultures of rice plants (Oryza sativa L.). When grown in liquid medium and in the absence of combined nitrogen but in the presence of the diazotrophsAzotobacter vinelandii andRhodopseudomonas capsulata, rice plants exhibited a complete biological cycle from germination up to ear stage, during a period of time similar to the one encountered in french paddy soil of Camargue. In one experiment, mannitol was given to rice culture medium together withAzotobacter vinelandii andRhodopseudomonas capsulata. In another experiment, mannitol was not given together with Rhodopseudomonas, and still positive nitrogen gain was obtained, although it was less than culture with mannitol. When15N labeled cells of Rhodopseudomonas were added in rice culture medium,15N was partly transferred to rice plant. Among the nitrogen substances excreted from the bacteria in the rhizosphere medium, large organic molecules were shown to be the most abundant in our experimental conditions. Moreover, the concentration of free ammonia or aminoacids present in the rice rhizosphere were always compatible with a bacterial nitrogenase activity.  相似文献

The ability of the benthic cyanobacterium Lyngbya wollei to fix nitrogen was studied using field samples and axenic cultures. L. wollei was collected and isolated from Lake Okeechobee, Florida, where it forms extensive mats. Rates of acetylene reduction up to 39.1 nmol mg dry wt−1 h−1 were observed for field samples. The maximum observed rate of acetylene reduction in axenic laboratory cultures was 200 nmol mg dry wt−1 h−1. Aerobic conditions limited nitrogen fixation activity, but dark/light cycles promoted the development of activity. Reduced oxygen levels appeared to be required for the development of significant levels of nitrogenase activity. The level of irradiance also had a significant impact on the level of activity. The potential significance of nitrogen fixation to Lyngbya production is discussed.  相似文献

Summary The efficiency and balance of nitrogen from one year's application was studied in a long-term fertigation experiment. Enriched nitrogen fertilizer, K15NO3, was applied to a 22-year-old Shamouti orange tree with a history of high N applications (N3) and to an N-starved tree (N1). The distribution of N in the different parts of the trees and in the soil was determined after the experimental trees were excavated. Similar total recovery of the labeled fertilizer N was found in the trees and soil in both treatments (N1−61.7% N3−56%). However, the distribution between tree and soil was different. The amount of recovered residual fertilizer in the soil was much larger in the N3 treatment than in N1. The highest percentage of fertilizer N was found in the new organs,i.e. fruits, twigs and leaves. The roots and branches took up only 6–14% from the labeled fertilizer. Only 20.9% of the leaf N and 23.4% of the fruit N in the N3 tree originated in the labeled fertilizer, indicating translocation of N from older parts of the tree to new growth. Maize (Zea mays L.) and ricebean (Vigna umbellata [Thumb.] Ohwi and Ohashi) were grown in intercrop and monoculture on Tropaqualf soils under rainfed conditions in Northern Thailand yearly from 1983 to 1986. De Wit's replacement design was used to compare intercrops and monocultures with a constant plant density equivalent to 80 000 maize or 160 000 ricebean plants ha−1. Combined nitrogen was applied at varying levels to 200 kg N ha−1. In the final two seasons the intercrop ratio of maize: ricebean was also varied. At the time of maize maturity intercrops yielded upt 49 kg ha−1 more N in the above ground plant parts than the best monoculture. Dry matter, grain and nitrogen yield of maize and ricebean in intercrop relative to their monoculture yields (RY, relative yield) were significantly greater than their respective share of the plant population. Relative yield totals (RYT) for grain, dry matter and nitrogen were always greater than 1. Nitrogen uptake per maize plant increased with progressive replacement of maize by ricebean plants. This increase was similar to that obtained by applying combined N. Available soil nitrogen tended to decrease with increasing maize:ricebean ratio. Increasing the maize:ricebean ratio increased the % of nitrogen derived from fixation in ricebean, the increase being equivalent to that obtained by decreasing combined nitrogen application. Approximately the same amount of fertilizer and soil nitrogen was taken up by maize plus ricebean in intercrop as the maize monoculture. The results suggest that the improved nitrogen economy of the intercrop resulted from the strong competitiveness of maize in the use of mineral nitrogen and the enhancement of nitrogen fixation in intercropped ricebean which made it less dependent on the depleted pool of soil nitrogen.  相似文献

The role of salt marshes as nitrogen sink is examined taking into consideration the seasonal variation of above and belowground biomass of Spartina martima and Halimione portulacoides in two marshes from Tagus estuary, Pancas and Corroios, and the degradation rates of belowground litter. Total nitrogen was determined in plant components, decomposing litter and sediment. Biomass was higher in Corroios, the saltier marsh, with 7190 g m−2 y−1 dw of S. maritima and 6593 g m−2 y−1 dw of H. portulacoides and the belowground component contributed to 96% and 90% of total biomass, respectively. In the other marsh, Pancas, belowground biomass contributed to 56% and 76% of total biomass for S. maritima and H. portulacoides, respectively. Litterbag experiment showed that between 25% and 50% of nitrogen is lost within the first month and remained relatively constant in the next four months. Slower decomposition is observed in sediments with higher nitrogen concentration (max. 0.7% N in the saltier marsh). Higher concentrations of N were found in the sediment upper layers. Considering the sediment-root system, most of the nitrogen is stored in the sediment compartment and only about 1–4% of the total N was found in the roots. Considering these results, Tagus salt marshes act as a sink for nitrogen.  相似文献

Sesbania sesban was evaluated as green manure crop for lowland rice in the Dry Zone of Sri Lanka. The legume was grown during a fallow period before lowland rice (Oryza sativa) and ploughed under just before transplanting. Weight loss and nitrogen content in litterbags containing leaves, stems and roots of the legume were monitored. Comparisons were made between rice yields from 20 m2 plots after green manuring in combination with different nitrogen fertilizer levels (0, 2.4, 4.8 and 7.2 gm−2) and nitrogen fertilizer (9.6 gm−2) alone. Above-ground biomass ofS. sesban was 440 gm−2 (dry wt) when ploughed under after 84 days growth. N-content in leaves, stems and roots was 3.76%, 0.41% and 0.73%, respectively. This gave a N-input fromS. sesban of 9.2 gm−2 (8.3 g from above-ground parts and 0.9 g from roots). The corresponding K and P inputs were 7.3 and 0.6 gm−2 respectively. The nitrogen rich leaves, which contained 88% of the nitrogen in the above-ground parts, decomposed and released its nitrogen much more rapidly than the stems and roots. After only four days the leaves had released 5.3 g Nm−2 and after 14 days they had released 6.4 g Nm−2. The highest rice yield (505 gm−2) was obtained usingS. sesban and 4.8 gm−2 of N-fertilizer. The yields with only N-fertilizer or onlyS. sesban were 442 gm−2 and 396 gm−2, respectively. Due to the rapid decomposition of the nitrogen rich leaves,S. sesban did not behave as a slow release fertilizer. Thus, it is not necessary to apply nitrogen fertilizers as a basal dose.  相似文献

Fusarium verticillioides is the most important seed transmitted pathogen that infects maize. It produces fumonisins, toxins that have potential toxicity for humans and animals. Control of F. verticillioides colonisation and systemic contamination of maize has become a priority area in food safety research. The aims of this research were (1) to characterise the maize endorhizosphere and rhizoplane inhabitant bacteria and Fusarium spp., (2) to select bacterial strains with impact on F. verticillioides growth and fumonisin B1 production in vitro, (3) to examine the effects of bacterial inoculum levels on F. verticillioides root colonisation under greenhouse conditions. Arthrobacter spp. and Azotobacter spp. were the predominant genera isolated from maize endorhizosphere and rhizoplane at the first sampling period, whilst F. verticillioides strains showed the greatest counts at the same isolation period. All F. verticillioides strains were able to produce fumonisin B1 in maize cultures. Arthrobacter globiformis RC5 and Azotobacter armeniacus RC2, used alone or in a mix, demonstrated important effects on F. verticillioides growth and fumonisin B1 suppression in vitro. Only Azotobacter armeniacus RC2 significantly reduced the F. verticillioides root colonisation at 106 and 107 CFU g–1 levels under greenhouse conditions.  相似文献

Three separate experiments were conducted in the nursery using grassland soil as a growing medium. The first experiment was conducted to assess the nodulation of the two legume trees grown in unamended soil, the second was done to determine the effects of N-fertilizer application on the interaction of the five Rhizobium isolates withA. falcataria and the third experiment was conducted to determine the effects of liming on theRhizobium × A. mangium interaction. Three local Rhizobium isolates, A16, A18, and A14 were effective forA. falcataria with A16 as the most promising strain under the conditions described. In general, application of combined nitrogen suppressed nodulation ofA. falcataria. Nodulation in the absence of combined N exceeded those fertilized with 30 kg N ha−1 by 114.0%, 60 kg N ha−1 by 209.6% and 100 kg N ha−1 by 237.1%. Two local isolates, Am10 and Am2, and an introduced strain, NA 1533 from Australia were promising forA. mangium. Unlike inA. falcataria, the application of combined nitrogen at the rate of 100 kg N ha−1 did not suppress nodulation inA. mangium. Liming the soil to pH 6.5 regardless of nitrogen fertilizer application improved the performance of the Rhizobium—A. mangium symbiosis.  相似文献

The author studied the effect of different nickel concentrations (0, 0.4, 40 and 80 μM Ni) on the nitrate reductase (NR) activity of New Zealand spinach (Tetragonia expansa Murr.) and lettuce (Lactuca sativa L. cv. Justyna) plants supplied with different nitrogen forms (NO3 –N, NH4 +–N, NH4NO3). A low concentration of Ni (0.4 μM) did not cause statistically significant changes of the nitrate reductase activity in lettuce plants supplied with nitrate nitrogen (NO3 –N) or mixed (NH4NO3) nitrogen form, but in New Zealand spinach leaves the enzyme activity decreased and increased, respectively. The introduction of 0.4 μM Ni in the medium containing ammonium ions as a sole source of nitrogen resulted in significantly increased NR activity in lettuce roots, and did not cause statistically significant changes of the enzyme activity in New Zealand spinach plants. At a high nickel level (Ni 40 or 80 μM), a significant decrease in the NR activity was observed in New Zealand spinach plants treated with nitrate or mixed nitrogen form, but it was much more marked in leaves than in roots. An exception was lack of significant changes of the enzyme activity in spinach leaves when plants were treated with 40 μM Ni and supplied with mixed nitrogen form, which resulted in the stronger reduction of the enzyme activity in roots than in leaves. The statistically significant drop in the NR activity was recorded in the aboveground parts of nickel-stressed lettuce plants supplied with NO3 –N or NH4NO3. At the same time, there were no statistically significant changes recorded in lettuce roots, except for the drop of the enzyme activity in the roots of NO3 -fed plants grown in the nutrient solution containing 80 μM Ni. An addition of high nickel doses to the nutrient solution contained ammonium nitrogen (NH4 +–N) did not affect the NR activity in New Zealand spinach plants and caused a high increase of this enzyme in lettuce organs, especially in roots. It should be stressed that, independently of nickel dose in New Zealand spinach plants supplied with ammonium form, NR activity in roots was dramatically higher than that in leaves. Moreover, in New Zealand spinach plants treated with NH4 +–N the enzyme activity in roots was even higher than in those supplied with NO3 –N.  相似文献

A study was conducted to ascertain monthly changes in biomass of the plant and nutrient content in various organs of Nymphoides hydrophylla grown in a tropical pond during September 1999–August 2000 in relation to environmental factors. Biomass of N. hydrophylla ranged from 25 to 247 g dry weight m−2. Among the various organs, leaf blade showed highest nitrogen (3.0–4.6%) and phosphorus content (0.9–2.4%). Comparative data of three Nymphoides species showed that N. peltata, the temperate species, had maximum potential of biomass production while long flowering period, year around growth, higher nitrogen content in various organs and presence of other associated flora were unique features of tropical species (N. hydrophylla and N. indica). Both water temperature and water level together appeared to be the best environmental variables that significantly explained the variability in biomass of N. hydrophylla.  相似文献

A field study was carried out near Zürich (Switzerland) to determine the yield of symbiotically fixed nitrogen (15N dilution) from white clover (Trifolium repens L.) grown with perennial ryegrass (Lolium perenne L) and from red clover (Trifolium pratense L.) grown with Italian ryegrass (Lolium multiflorum Lam.). A zero N fertilizer treatment was compared to a 30 kg N/ha per cut regime (90 to 150 kg ha−1 annually). The annual yield of clover N derived from symbiosis averaged 131 kg ha−1 (49 to 227 kg) without N fertilization and 83 kg ha−1 (21 to 173 kg) with 30 kg of fertilizer N ha−1 per cut in the seeding year. Values for the first production year were 308 kg ha−1 (268 to 373 kg) without N fertilization and 232 kg ha−1 (165 to 305 kg) with 30 kg fertilizer N ha−1 per cut. The variation between years was associated mainly with the proportion of clover in the mixtures. Apparent clover-to-grass transfer of fixed N contributed up to 52 kg N ha−1 per year (17 kg N ha−1 on average) to the N yield of the mixtures. Percentage N derived from symbiosis averaged 75% for white and 86% for red clover. These percentages were affected only slightly by supplemental nitrogen, but declined markedly during late summer for white clover. It is concluded that the annual yield of symbiotically fixed N from clover/grass mixtures can be very high, provided that the proportion of clover in the mixtures exceeds 50% of total dry mass yield.  相似文献

A pot experiment was conducted to determine the effects of the application of 13C (1.256 atom%) and 15N (1.098 atom%) dual-labeled maize residue compost (MRC) on the nitrogen and carbon uptake by radish, komatsuna, and chingensai as compared with the effect of inorganic fertilizer (IF). The vegetables were grown over three consecutive growing seasons over 4 months; compost was applied at the rate of 24 g kg–1 soil. Nonlabeled nitrogen fertilizer was applied to the compost treatments in the second and third crops to compare the effects of blends of compost with N fertilizer to fertilizer alone. The N uptake and yield of vegetables were significantly higher with the recommended inorganic N treatment. The vegetables took up significantly (P < 0.05) lower amounts of N from MRC than from IFs during the three cultivations. The values of the N uptake derived by fertilizer application to the plant exhibited significant differences among different vegetables. Nitrogen recovered by komatsuna and chingensai from MRC was 7.3 (6.6%), 2.7 (1.8%), and 2.3, (1.7%) in the first, second, and third crops, respectively. Radish, komatsuna, and chingensai recovered significant amounts of C from MRC in the first and second crops, with negligible C recovery in the third crop. The initial loss of fertilizer C in soil at the first crop indicates that the microbial decomposition decoupled substantial amounts of 13C/15N-labeled compounds early in plant development, thus giving the microorganisms a preemptive competitive advantage in the acquisition of easily available 13C/15N-labeled substrates. It is concluded that a combination of compost and inorganic N did not supply sufficient plant-available N to increase vegetables yields or N uptake over those of fertilizer alone. The data suggested that higher productivity of vegetables might be achieved after the accumulation of a certain amount of residual compost N.  相似文献

Summary Differences in N2-fixation byPhaseolus vulgaris bean cultivars were successfully evaluated in the field using15N isotope dilution technique with a non-fixing test crop of a different species (wheat). The Phaseolus cultivars could have been similarly ranked for N2-fixation capacity from either seed yield or total nitrogen yield, but the isotope method provided a direct measure of N2-fixation and made it possible to estimate the proportion of fixed to total nitrogen in the crop and in plant parts. Amounts of nitrogen fixed varied between 24.59 kg N/ha for the 60-day cultivar Goiano precoce to 64.91 kg N/ha for the 90-day cultivar Carioca. The per cent of plant nitrogen due to fixation was 57–68% for the 90-day cultivars and 37% for Goiano precoce (60-day cultivar). Fertilizer utilization was 17–30% of a 20 kg N/ha fertilizer application. 100 kg N/ha fertilizer application decreased N2-fixation without suppressing it totally. Differences in yield between the highest yielding (Carioca) and the lowest (Moruna) 90-day cultivars were also due apparently to varietal differences in efficiency of conversion of nitrogen to economic matteri.e. seed, as well as to differences in capacity of genotypes for N2-fixation. Phellinus robustus produced both laccase (700–4,000 U l−1) and manganese peroxidase (MnP) (1,000–11,300 U l−1) in fermentation of nine food wastes, whereas Ganoderma adspersum produced only laccase (600–34,000 U l−1). Glucose provided high laccase and MnP activity of P. robustus but repressed enzyme production by G. adspersum. Ammonium sulphate and ammonium tartrate increased the P. robustus laccase yield (3-fold), whereas the accumulation of MnP was not enhanced by additional nitrogen.  相似文献

Summary The population size of diazotrophic bacteria naturally associated with the maize rhizosphere can be affected by biotic and environmental factors. In this work we have evaluated the effect of two genotypes of maize, with and without nitrogen fertilization, on the population dynamics and distribution of diazotrophic bacteria associated with maize plants over different plant ontogenic stages. The study was carried out in a field experiment with and without nitrogen fertilization, using two maize cultivars (Santa Helena 8447 and Santa Rosa 3063) previously selected from 32 maize cultivars for the lowest and highest response to nitrogen fertilization, respectively. Microbiological and molecular approaches were used to characterize the diazotrophic bacterial population structure. Bacterial population was assessed by the most probable number using semi-solid N-free media, and by DNA isolation from soil and plant tissue followed by amplification of nifH gene fragments using nested PCR, and by RFLP analysis using the restriction endonucleases TaqI and HaeIII. The dynamics of the diazotrophic bacterial population were affected by the ontogenic stage of the maize plants, but not by the cultivar type. Roots were the preferred site of colonization, independent of cultivar type and growth stage. During the first stage of maize growth, addition of nitrogen fertilizer negatively affected the diazotrophic bacterial population.  相似文献

The Salton Sea is one of the few saline, inland lakes in the world with a population of barnacles, Balanus amphitrite. It is also one of California’s most impaired water bodies due to excessive nutrient loading which leads to phytoplankton blooms and low dissolved oxygen. Currently, B. amphitrite growth is limited due to lack of hard substrate in and around the Sea. We have hypothesized that artificial substrate could support the growth of B. amphitrite and their filter-feeding would lead to improved water quality. Periodic harvesting of the barnacles would result in the permanent removal of nitrogen and phosphorus from the Sea. A 44-day in-situ experiment was carried out in the Salton Sea to assess the rate of barnacle growth and phosphorus and nitrogen sequestration on burlap sheets suspended vertically from a floating line. Burlap panels were collected weekly and the barnacles analyzed for Ca, total-P, inorganic-P, total-N, total-C, CaCO3, and organic matter content. After 44 days of growth, the barnacle mats weighed 7.4 kg m−2 on a dry weight basis, with 80% of the mass as shell material. The nutrient sequestration was 9.4 g P m−2 and 100 g N m−2. Approximately half of the P was inorganic and appears to be coprecipitated with the calcium carbonate shell material. Results indicate that harvesting barnacles grown on artificial substrate in the Salton Sea would not be an effective method for removing N or P from the lake because of the relative proportions of shell material and organic material. An integrated outdoor cultivation of two macroalgal species: Ulva rotundata (Chlorophyta) and Gracilariopsis longissima (Rhodophyta) was designed. The macroalgae were cultured in effluents from an intensive marine culture (growout phase) of gilthead seabream Sparus aurata. The biomass evolution of the algal tanks followed a logistic curve, where the approach to the maximum stocking density of seaweeds was governed by thalli self-shading, as nutrient limitation in the cultivation tank was unlikely. The maximum stocking density of the system was approximately 27.8 g U. rotundata L−1 (16.7 Kg m−2) and 11.9 g G. longissima L−1 (7.12 Kg m−2). Yield was more than 3 times higher in U. rotundata than in G. longissima. Overall, U. rotundata removed a greater percentage of phosphate (8.9%) and total dissolved inorganic nitrogen (54%) flowing into the algal tanks than G. longissima. The latter species biofiltered approximately 3.2% of phosphate and 17% of the total dissolved inorganic nitrogen input. However, mean nutrient uptake rates on wet weight basis were usually higher in G. longissima than in U. rotundata. The production of total oxidised nitrogen in the algal tanks, considered as being the nitrification rate occurring on the algal fronds by nitrifying bacteria, was less than half of the ammonium uptake by the macroalgae, suggesting that seaweeds competed efficiently for ammonia against the nitrifyers. The biofiltration during a diel cycle showed that mean phosphate biofiltration was lower than 4.5% in the two species whereas ammonium was biofiltered efficiently (up to 67%), especially in U. rotundata. The metal and heavy metal content in the algal tissue at the end of the monitoring period suggested no metal contamination of tissues so that both macroalgal species could be used in the food industry. The study reveals the value of ecological engineering techniques in reducing the dissolved nutrient content in effluents from the fish farm, with the prospect of a better management practises, based on integrated mariculture designs, being developed by the local farmers.  相似文献

Traditionally it has been thought that most boreal forest communities lack a significant input of biologically fixed nitrogen. Recent discoveries of nitrogen fixation by cyanobacteria associated with mosses have resulted in a re-evaluation of this view. While it is recognized that rates of nitrogen fixation in mosses can be highly variable, there is little understanding as to why this occurs. I monitored nitrogen fixation, using acetylene reduction, in wet lowland and dry upland boreal forest communities, in central Canada, over a growing season. At the peak of nitrogen fixation in mid summer, Sphagnum capillifolium had an 11 times higher rate of fixation than Pleurozium schreberi. Variation in canopy openness and precipitation had no effect on rates of fixation over the growing season. In P. schreberi fixation rates did not vary between sites. Temperature had a positive effect on fixation rates in both S. capillifolium and P. schreberi, but the effect was 4 times more pronounced in S. capillifolium. Seasonal rates of nitrogen fixation were estimated at 193 mg N m−2 for S. capillifolium and 23 mg N m−2 for P. schreberi. With moderate increases in climate warming, predicted increases in nitrogen fixation in S. capillifolium are sufficient to raise its decomposition rate. Increased temperatures may therefore act synergistically to change boreal systems from a sink to a source of carbon.  相似文献
