Six primary monosomes of the amphidiploid species Gossypium hirsutum have been identified as separate chromosomes of the complement. On the basis of cytogenetic tests involving translocations, a diploid species of the D genome group, and genetic markers, 4 of the 6 monosomes were designated chromosomes 1, 2, 4 and 6 of the A subgenome and 2 were designated chromosomes 17 and 18 of the D subgenome. Fourteen other monosomes were identified as duplicates of 3 of the above 6 primaries.  相似文献

Quiescent cottonseeds stored in a dry, anaerobic situation for over a year have been shown to contain cells whose contents are ultrastructurally similar to those of normal, fully hydrated plant cells. Plastids, mitochondria, and nuclei of the cells of cotyledon tissue in dry seeds possess normal-looking double membranes even under conditions of extreme desiccation. Previous reports have indicated on the basis of light microscopic work, that the cells of certain dry seeds do not possess nuclear membranes or mitochondria. The cells of the dry cottonseed do contain these structures, however. Dictyosomes have not been observed in the spongy parenchymal cells of the cotyledon tissue; it is suggested that they are concerned with translocation and/or utilization of material. The storage materials in the cells, protein and oil, are contained in vacuolar areas enclosed by a single membrane.  相似文献

Gossypium hirsutum has a large indigenous range encompassing most of Mesoamerica and the Caribbean, where it exhibits a diverse array of morphological forms spanning the wild-to-domesticated continuum. Modem, highly improved varieties (“Upland cotton”), which currently account for about 90% of world cotton commerce, are day-length neutral annuals derived from subtropical, perennial antecedents. To assess levels and patterns of genetic variation in the species and to elucidate the origin of Upland cotton, 538 accessions representing the full spectrum of morphological and geographical diversity were analyzed forallozyme variation at 50 loci. Levels of variation are modest overall but are low in Upland cotton. Relationships among accessions reflect pre-Columbian influences of aboriginal peoples and later European colonists superimposed on the preagricultural pattern. In contrast to expectations, two centers of diversity are evident, one in southern Mexico-Guatemala and the other in the Caribbean. Introgression of G. barbadense genes into G. hirsutum has been common in a broad area of sympatry in the Caribbean. The germplasm of present cultivars traces to Mexican highland stocks, which, in turn, were derived from material originally from southern Mexico and Guatemala. Despite the widespread belief that germplasm from several other species has been incorporated into modem Upland stocks through intentional breeding efforts, the 50 Upland cultivars examined contain no unique alleles, suggesting that retention of genes from transspecific sources has been minimal. The most recent infraspecific treatment, which recognizes seven races, does not adequately represent genetic relationships.  相似文献

在显微和超微水平不同层次上对陆地棉(Gossypium hisutum L.)再生植株中正常苗和玻璃苗叶及根端的形态结构进行了观察比较。结果表明:试管玻璃苗叶表面凹凸不平,气孔下陷,萎缩变形,表皮细胞形态不规则,排列不整齐,叶表面蜡质分布不均匀,其数量也较少;叶肉无明显的栅栏组织分化,细胞中叶绿体含量少、形态小,结构异常。超微结构显示,大部分叶肉细胞中叶绿体基粒片层分布少而含较多的基质片层,其它细胞器如线粒体、高尔基体、内质网等含量少或缺。叶脉维管组织发育不良,导管分子数目少,木质化程度低,根尖短而细,生长点仅由少数具分生能力的细胞组成,根冠细胞大,但数量少。正常苗形态结构基本类似于实生苗。  相似文献

Respiration and net photosynthetic O2 production by cotton cotyledons were determined from an early age through the senescent stage. Various treatments were applied to cotyledons to assess the importance of current photosynthesis as compared to translocation of reserves to seedling development. Rates of respiration and net photosynthesis per cm2 were high on 1-day-old cotyledons, but the rates decreased sharply with rapid expansion to reach a fairly stable rate. Respiration per cotyledon decreased linearly with age until the onset of senescence, then exhibited a distinct climacteric rise followed by a sharp decrease. Net photosynthesis per cotyledon increased until expansion was completed and then decreased linearly and steeply with age. Excision of cotyledons, inhibition of photosynthesis either chemically or by covering, and removal of the terminal bud indicated that current photosynthesis is a potent force behind early epicotyl growth.  相似文献

对原产于墨西哥的8个陆地棉半野生种系进行了核型分析。其根尖细胞的染色体数目均为52。“墨西哥棉”、“阔叶棉”、“帕默尔氏棉”和“尖斑棉”为1B 核型。其中以“墨西哥棉”最为原始,2n=4x=52=44m(6SAT) 8sm。“尤卡坦棉”、“玛利加郎特棉”、“莫利尔氏棉”和“雷奇蒙德氏棉”为2B 核型。其中以后两个种系最进化,2n=4x=52=32m(2或4SAT) 20sm。讨论了各种系之间的可能的演化关系。  相似文献

The arrangement of cellulosic fibrils in the cell walls of cotton fibers is very unusual; rather than exhibiting a continuous spiraling in one direction, they intermittantly reverse their gyre. Microtubules that line the periphery of the protoplasm, subjacent to the plasmalemma, tend to parallel the deployment of the cell wall microfibrils. It was not known whether this parallelism persisted through the reversal. By studying tangential sections of the cell wall/protoplasmic interfaces at the reversals, we show that congruity continues even through the reversals. Colchicine treatment did not appear to inhibit cellulose synthesis but it did abolish microtubules in the cotton fiber cells and deranged normal cell wall microfibrillar orientation. Previously, cotton fibers have been shown to possess most of the familiar organelles, but we found two new features not reported heretofore. They are microfilaments and peculiar “polygonal structures” that appear to be associated with the plasma membrane.  相似文献

Small (250 μm diam) palladium microprobes, either sharpened or blunt-tipped, were inserted into peduncles and petioles of field-grown cotton (Gossypium hirsutum L. cv. ‘Deltapine 61‘). Two weeks after microprobe insertion, tissue was harvested and processed for subsequent light-microscopic examination. Factors which influence structure and organization of wound response tissue, such as developmental stage of tissue at microprobe implantation site, are discussed. Patterns of differentiation in the wound response tissue which surrounds the microprobes are illustrated. There appeared to be direct rather than peripheral continuity between normal vascular tissue and vascular tissue formed within the wound callus surrounding microprobes implanted in petiole and peduncle. Apparently restoration of direct vascular connections between the original vascular tissue and the vascular tissue formed within the wound callus is a characteristic wound-healing response following microprobe implantation in cotton.  相似文献

The net rate of CO2 uptake for leaves of Gossypium hirsutum L. was reduced when the plants were grown at low concentrations of NO3-, PO42-, or K+. The water vapor conductance was relatively constant for all nutrient levels, indicating little effect on stomatal response. Although leaves under nutrient stress tended to be lower in chlorophyll and thinner, the ratio of mesophyll surface area to leaf area did not change appreciably. Thus, the reduction in CO2 uptake rate at low nutrient levels was due to a decrease in the CO2 conductance expressed per unit mesophyll cell wall area (gcellCO2). The use of gcellCO2 and nutrient levels expressed per unit of mesophyll cell wall provides a new means of assessing nutrient effects on CO2 uptake of leaves.  相似文献

采用MADS(MCM1-Agamous-Deficiens-SRF)盒基因家族功能区保守序列PCR引物,将水稻(Oryzasativa L.ssp indica)“珍汕97B”悬浮细胞、愈伤组织、分化愈伤组织和再生试管苗等不同形态发生的组织的mRNA反转录后选择性放大,经测序胶分离鉴定出一组差异表达的cDNA。对命名为RM1 cDNA的5'端序列测定表明,RM1与典型的MADS基因——拟南芥agamous蛋白保守区一级结构同源性达63%,模拟二级结构相似性显著,初步确认RM1属于MADS基因家族成员。分子杂交证实,RM1在悬浮培养细胞中不表达,而在愈伤组织、分化愈伤组织和再生试管苗中活跃表达。  相似文献

陆地棉(Gossypium hirsutum L.)胚性愈伤组织在两年多的继代培养过程中,由淡绿或灰色疏松状转变成灰黄色致密团块状,胚胎发生能力经历了一个由低到高再到低的过程。培养两年半后,从中分离出3种不同的愈伤组织系。第一种可产生大量的胚状体;第二种可产生少量胚状体;而第三种失去了胚胎发生能力。第一种团块颗粒小,后两种团块颗粒大。通过选择培养第一种愈伤组织系,建立了3 个陆地棉栽培品种的高频体细胞胚胎发生系。染色体观察显示:其变异频率随愈伤组织培养代数的增加而升高;3 种不同的愈伤组织系中,二倍体细胞的频率大小依次为:第一种> 第二种> 第三种。此外,第一种含有较多的超二倍体细胞,而后两种含较多亚二倍体细胞  相似文献

The hybrid of Gossypium hirsutum X G. gossypioides develops normally until 10–12 nodes have been produced at which time neoplasms form in the cortex, phloem, and pith of the lower portion of the shoot axis. Neoplasms are characterized by irregular whorls of unusually large cells the central portions of which contain apparently cytolysed cells and a reddish-brown substance that probably represents the products of cytolysis. Neoplastic invasion is followed by inactivation of the vascular cambium, disorganization of the phloem and cortex, and tylosing and plugging of many xylem vessels. Subsequent growth of the shoot, which continues at a very slow rate for up to several years, involves the proliferation of parenchymatous tissue which becomes secondarily vascularized.  相似文献

洋麻(Hibiscus cannabinus)一生中叶形态的变化有着一定的规律性(雅库希金1947)。茎上基部的叶子为锯齿卵形,中部为深裂叶,叶子的分裂程度随着叶子的层次逐渐加大,菜片由3裂、5裂、发展到7裂。以后分裂的程度开始下降由7裂降到5裂、3裂。顶端的叶子篇锯齿狭长叶。根据以上的现象,可以看出随着洋麻发育的进程叶片分裂的程度有着周期性的变化。洋麻可以用缩短光照的处理来加速它的开花结实。在短光照条件下的洋麻是否因为光照期的缩短加速了叶形态周期性的变化?当洋麻从短光照的条件下进入长光照之  相似文献

A partial complementary DNA (cDNA) (DSA8) for a P-type ATPase was obtained from the halotolerant alga Dunaliella salina (Dunal) Teod. (Chlorophyceae). The cDNA exhibited greater than 90% homology to the cDNA for a H+-ATPase in D. bioculata Butcher. The expression of the gene that corresponded to DSA8 was decreased strongly by increases in NaCl concentration. The expression of a gene that corresponded to another ATPase (DSA1; possibly for a Ca2+-ATPase) from D. salina did not show the same decrease as did the DSA8. However, increased osmotic pressure due to glycerol resulted in the same decrease in the DSA8 gene. Under salt or osmotic stress, the activity of a H+-ATPase from microsomes of this alga also decreased. We suggest that expression of the gene for the plasma membrane H+-ATPase of D. salina is regulated by osmotic pressure rather than by the concentration of NaCl.  相似文献
