目的观察中华眼镜蛇伤后不同时期应用抗蛇毒血清对机体保护作用的差异,为探索临床抗蛇毒血清使用的最佳有效时段提供依据。方法将SD大鼠随机分成6组(蛇毒组、40、60、80、100、120 min血清保护组),每组30只。动物经戊巴比妥腹麻,于单侧背部皮下及双侧小腿腓肠肌注射眼镜蛇毒以制备中华眼镜蛇毒挑战剂量(4×LD50)大鼠模型,分5个不同时段分别注射抗蛇毒血清,于注毒后连续观察3 h,统计各组平均存活时间、成活率及保护率。结果蛇毒组大鼠注入挑战剂量的眼镜蛇毒后,平均存活时间为(148.8±11.4)min,成活率仅为20%;其他血清保护组分别于注毒后40、60、80、100、120 min经腹腔注射精制抗眼镜蛇毒血清(125 u血清/mg蛇毒),40 min血清组保护率达80%;60 min血清组存活率及保护率分别达到70%、50%,平均存活时间为(172.8±7.2)min,与蛇毒组相比有明显差异(P<0.01);而801、00 min血清组保护率依次下降,分别为40%、30%,但仍较蛇毒组显著提高(P<0.01);120 min血清组存活时间及保护率与蛇毒组比较差异无显著性。结论利用中华眼镜蛇毒挑战剂量大鼠模型,通过不同时段施予同剂量抗血清,可显示出明显的机体保护时效性,该研究为探讨临床正确使用抗蛇毒血清提供了新的实验依据。 相似文献

目的建立中华眼镜蛇毒挑战剂量的大鼠模型,通过对多项电生理指标的动态观察,为蛇毒毒理及抗蛇毒制剂的研究提供实验数据。方法将SD大鼠分为蛇毒组、血清保护组和对照组,戊巴比妥腹麻,连续记录心率、血压、呼吸频率及心电图等电生理指标的动态变化,观察眼镜蛇毒的毒性反应及抗蛇毒血清对动物的保护作用。结果腹腔注入挑战剂量(4×LD50:4×0.636 mg/kg)的中华眼镜蛇毒后,大鼠平均生存时间为(3±0.33)h。蛇毒组给药30~60 min后,心率及呼吸频率均有轻度加快,Ⅱ导联ST段抬升幅度超过同期正常对照0.1 mV以上(P<0.05);130~150 min后,心率、呼吸频率及动脉血压均直线下降;达180 min时,心率、呼吸频率及动脉血压均已降到同期正常对照的30%以下,差异非常显著(P<0.01)。血清保护组给药后各指标相对平稳,与对照组比较无明显差异。结论以4倍LD50为挑战剂量造模的中华眼镜蛇伤大鼠模型,心率、血压、呼吸频率及心电图等电生理指标的动态变化与临床症状基本相符。眼镜蛇毒抗血清可有效地中和、保护眼镜蛇毒对机体的伤害。 相似文献

目的观察单价抗眼镜蛇毒血清溶液滴眼治疗中华眼镜蛇毒(Naja naja atra,Chinese cobra)不慎进人眼睛引起外伤中毒性急性角膜炎的临床效果。方法观察1992~2002年我院急诊救治眼镜蛇喷毒或加工蛇毒时不慎蛇毒进人眼睛引起外伤性急性角膜炎8例男性病人,从受伤到就诊时间最快15min,最慢50min。就诊后立即用生理盐水冲洗受伤眼睛,紧接着给予单价抗眼镜蛇毒血清溶液滴人患眼0.5h后,用氯霉素眼药水、可的松(或地塞米松)眼药水交替滴眼,直至痊愈。结果使用抗蛇毒血清滴眼后局部疼痛、异物感等症状在20min内得到缓解,3天内眼睑红肿、结膜充血等角膜炎症状消失。8例病人均治愈,未留有后遗症。结论:单价抗眼镜蛇毒血清滴眼治疗中华眼镜蛇毒致眼外伤是非常方便有效的方法,可能与其能迅速中和眼睛内残留的蛇毒有关。 相似文献

目的探讨我国华南亚种眼镜蛇(Naja naja atra)咬伤所致局部损伤的特点和治疗体会。方法回顾我院收治的102例眼镜蛇咬伤病人的相关资料,观察比较受伤局部损伤情况和临床表现以及治疗方法与疗效等情况。结果所有病例均表现为牙痕周围组织不同程度的坏死和周边较大范围的炎症反应和局域淋巴结肿大,部分病例伴有或发热、困倦以及血WBC增高等表现,少数病例出现心、肝、肾功能轻度损害和溶血表现,未见典型神经麻痹病例和凝血功能障碍病例;84例实施了清创术,28病例实施了植皮手术,9例实施了筋膜室切开减压,2例实施了截肢术,所有病例在(17±6.2)天内痊愈,致残5例,无死亡病例。结论华南亚种眼镜蛇咬伤的特点是局部组织变性坏死,神经毒和血液循环毒方面的表现轻而少见,早期使用抗眼镜蛇毒血清是预防和减少局部组织损伤的的关键措施,在此基础上,及时清除坏死组织、积极抗炎消肿减压、尽早植皮覆盖创面对抑制局部损伤发展和促进组织修复以及加快痊愈有重要意义。 相似文献

目的探索免疫鸡制备高效价抗眼镜蛇毒抗体的新方法。方法用中华眼镜蛇原毒作抗原免疫22周龄的莱航母鸡,水溶法粗提抗体,DEAE Sepharos FF柱纯化,切向流超滤膜脱盐及浓缩,免疫电泳及双向免疫扩散法进行鉴定及效价测定,采用BCATMProte in Assay K it测定蛋白含量。结果鸡卵黄经水溶法的粗提物与中华眼镜蛇毒即有较明显沉淀反应,其效价随着纯度的提高而增强。将马源性抗血清的蛋白质含量调至与浓缩的IgY相同(2mg/m l),经双向免疫扩散及免疫电泳鉴定,该抗体不但对中华眼镜蛇毒有特异性结合,与孟加拉眼镜蛇毒亦有较强的交叉免疫活性,其效价较马抗眼镜蛇毒血清高4倍以上。结论用中华眼镜蛇原毒制备的IgY抗体,其效价较马抗血清有显著提高,并与孟加拉眼镜蛇毒有高度交叉免疫。本实验为抗眼镜蛇IgY的应用及其它抗蛇毒IgY的制备奠定了基础。 相似文献

以章丘大葱为试验材料,从形态组织学及生理生化等方面探讨了大葱试管苗玻璃化发生的可能机理。结果表明,玻璃苗形态组织结构与正常苗有很大差异,玻璃苗组织含水量增加,过氧化物酶(POD)活性增强,叶绿素总量、叶绿素a、叶绿素b、过氧化氢酶(CAT)的活性、可溶性蛋白含量均降低。玻璃苗酯酶同工酶谱带与正常苗相比,增加了一条带,同时缺少了两条带;可溶性蛋白电泳分析表明,玻璃苗与正常苗间也存在明显差异。可见,当外界环境条件不适宜植物生长需要时,植物体在基因表达水平上将会受到影响,表达产物的差异可引起生理代谢功能上的紊乱,从而导致形态结构的畸型,进而产生玻璃化苗。 相似文献

Experiments are the foundation of empirical science, and experimental paradigms that are broadly applicable across settings and species are particularly useful for comparative research. Originally developed to address questions related to perception and cognition of pre‐verbal human infants, the looking time experimental paradigm has been increasingly used to study animal behavior and cognition, particularly in non‐human primates. Looking time experiments are based on the assumption that animals direct eye gaze toward objects or scenes based on their degree of interest, and use looking behavior to infer perceptual or cognitive characteristics of subjects. This paradigm can be used in a variety of contexts and is not based on species‐typical behaviors, allowing for intra‐ and interspecific comparisons. Here, we describe the history of use of looking time measures, provide an overview of the problems and controversies related to this method, and offer recommendations on how to implement looking time tasks, focusing on the preparation of stimuli, experimental procedures, and data analysis. Our overview focuses on non‐human primates, where most work has been carried out, but the issues discussed should be applicable to a wide range of species. We conclude that despite pertinent criticism, looking time tasks are practical when executed and interpreted properly. The further implementation of these methods in studies of animal behavior and cognition is likely to be fruitful. 相似文献

目的 利用鸭乙型肝炎病毒(DHBV)感染动物模型,评价亚甲蓝光化学病毒灭活方法对血液成分中DNA病毒的灭活效果。方法 将超离纯化的DHBV分别加入人血浆或人红细胞,经亚甲蓝光化学灭活病毒,将含不同基因组拷贝数DHBV的血浆成分经静脉感染1 d龄雏鸭。采用放射性核素核酸杂交法对血清中DHBV DNA进行检测,计算病毒灭活处理前、后人血浆及人红细胞中DHBV的半数感染计量(ID50)。结果 结果显示加入DHBV的血浆在未经灭活处理前对1 d龄雏鸭的ID50值为103.33,而经病毒灭活处理后ID50值为1010拷贝,灭活处理可使病毒感染性滴度下降达6个Log;加入DHBV的红细胞灭活前ID50值为103.35,经灭活处理后ID50值为108.35拷贝,灭活处理使病毒感染性滴度下降5个Log。结论 利用DHBV感染动物模型,可以检测到少量病毒在自然感染宿主体内的感染性,可用于评判血液成分中病毒灭活方法的效果,亚甲蓝光化学处理对血浆中DNA病毒的灭活效果较好于对红细胞中DNA病毒的灭活作用。 相似文献

Summary Reducing serum concentrations in media provides a potential cost advantage. To determine whether such media could be used
for applied mutagenesis assays, we measured cloning efficiency and growth parameters in suspension of Chinese hamster ovary
cells cultured in reduced serum with or without additives (1 μg/ml insulin, 3×10−7
M linoleic acid, 1×10−8
M H2SeO3) or bovine serum albumin (BSA, 1% wt/vol). With the additives and ≤0.5% fetal bovine serum (FBS), Ham’s F12 medium (without
hypoxanthine and thymidine) was more optimal than alpha Eagle’s minimum essential medium (MEM) (without ribosides and deoxyribosides)
for low density cloning and high density suspension growth. Acceptable cloning efficiencies were obtained with 2% FBS plus
BSA without additives in either medium; the addition of BSA resulted in improved colony size and more compact colony morphology.
In alpha MEM, satisfactory growth rates and maximum saturation densities in suspension culture were obtained only with 5%
FBS; in Ham’s F12, 1% FBS+deoxycytidine+BSA yielded satisfactory suspension growth. Spontaneous mutant frequencies were compared
for each medium containing 10% dialyzed FBS (DFBS), 1% FBS plus BSA, or 2% FBS plus BSA. The spontaneous frequency of azaadenine-resistant
phenotypes (mutant at theaprt locus) in 1% FBS plus BSA was significantly lower than the frequency observed in 2% FBS plus BSA or 10% DFBS. Frequencies
of spontaneous mutants resistant to thioguanine (hgprt locus) or fluorodeoxyuridine (tk locus) were similar with 10% DFBS, 1% FBS plus BSA, or 2% FBS plus BSA. Compared to alpha MEM with 10% DFBS, frequencies
of drug-resistant mutants induced by ethyl methanesulfonate or mitomycin C (MMC) were not significantly lower in alpha MEM
with 2% FBS plus BSA; observed mutant frequencies induced by dimethyl-nitrosamine or benzo(a)pyrene seemed to be decreased at lower survival levels.
