Four kinds of GA, GA1, GA2, GA3, and GA4, all inhibited sproutingin aerial tubers of Begonia evansiana. The sprout-inhibiting action of GA increased with incubationin a high O2 concentration, and decreased in a low O2 Concentration. Inhibition of sprouting by GA was reduced by incubation in thepresence of p-nitrophenol, resorcinol, salicylaldoxime, 2, 4-dinitrophenol,sodium azide and cycloheximide. The higher activity of polyphenol oxydase was observed in ahomogenate of GA treated tubers. Existence of counteracting two systems which were activatedby GA was considered. Measurements of the rate of photosynthesis of plants of Solanumtuberosum L. var. King Edward were made, using 14CO2, at weeklyintervals throughout their growth in a controlled environment.Leaf area and dry weight of sections of the plant were alsodetermined. The results are discussed in relation to existingtheories that photosynthesis can be limited by carbohydrateaccumulation in the leaves, and stimulated by the initiationof tubers.  相似文献

Changes in the amount of growth inhibitors by gibberellin (GA)treatment in tubers of Begonia evaniana was studied. Acidic,neutral and basic ethyl acetate-soluble and n-butanol-solublegrowth inhibitors increased with GA treatment and also withthe progress of natural dormancy. These inhibitors suppressedthe sprouting of nondormant aerial tubers. The increase of neutralethyl acetate-soluble inhibitors in the GA-treated tubers wassuppressed by incubation in low-O2 atmosphere. In early dormantstage tubers, the inhibitor increased remarkably during incubationat 28?C in light. The aerial tubers probably enter a dormantstate when sufficient amounts of inhibitors accumulate in them. (Received February 9, 1973; )  相似文献

Participation of phospholipase (PL) A2 in signal trans-ductionhas been reported to elicitate a resistance reaction in potatocells by inoculation of an incompatible race of Phytophthorainfestans, the late blight fungus, or by treatment with fungalelicitor hyphal wall components (HWC). Mastoparan, a genericG protein activator, has been shown to activate PLA in a G protein-dependentmanner in animal cells. We analyzed the effects of mastoparanand the inactive analog Mas-17 on PLA2 activity in potato tubers.In healthy potato tubers, the activation of PLA2 by mastoparanwas detected in the soluble fraction, but not micro-somal fraction.However, in potato tubers treated with HWC, PLA2 activity wasstimulated by mastoparan in both soluble and microsomal fractions.Pretreatment of the microsomal fraction with neomycin, a PLCinhibitor, and staurosporine, a protein kinase inhibitor, inhibitedthe mastoparan-induced activation of PLA2. This suggested thatthe PLA2 activation in potato tubers by mastoparan was mediatedby the PLC pathway and protein phospho-rylation. We also examinedthe accumulation of potato phytoalexin rishitin. Mastoparanstimulated rishitin accumulation induced by HWC, but did notinduce the accumulation. Potato plants (Solanum tuberosum L.) were grown in water culturein a controlled environment. Cooling (+8°C) of individualtubers decreased their growth rates and increased the growthrates of non-cooled tubers of the same plant. The carbohydrateconcentration in non-cooled and cooled tubers did not differsignificantly, but 14C-import from labelled photosynthate waslower in cooled than in non-cooled tubers. The markedly lowerconversion rate of ethanol-soluble 14C to starch in cooled,in comparison to non-cooled tubers, was not associated withsignificant differences in the in vitro activities of starchsynthase, ADPG-pyrophosphorylase and starch phosphorylase understandard assay conditions (+30°C). However, the Q10-valuesof the enzymes differed in vitro in the temperature range between30°C and 8°C, leading to a marked decrease in the activityratio of ADPG-pyrophosphorylase/starch phosphorylase in cooledtubers. In tubers differing in growth rates without manipulation, 14d after tuber initiation significant positive correlations werefound between 14C-concentration of tuber tissue and the in vitroactivities of starch synthase and ADPG-pyrophosphorylase anda significant negative correlation between 14C-concentrationand starch phosphorylase. In contrast, in tubers which wereanalysed 5 d after initiation, there were only small differencesbetween tubers in growth rate, 14C import and the activity ratioADPG-pyrophosphorylase/starch phosphorylase. From various directand indirect evidence it is concluded that the growth rate ofindividual tubers, and thus the sink strength, is at least inpart controlled by the activity of starch synthesizing enzymes. Intact tubers of potato (Solanum tuberosum L. cv. Irish Cobblerand an interspecific hybrid between S. tuberosum and S. demissumcv. Rishiri) contain a very low activity of 3-hydroxy-3-methylglutaryl(HMG)-CoA reductase. The activity increased first in responseto slicing, and again in response to additional treatments suchas inoculation with an incompatible race of Phytophthora infestans,application of a hyphal wall component of the fungus or HgCl2solution, and then decreased. Both the first and the secondincreases in activity in response to slicing and additionaltreatment with a hyphal wall component to elicit phytoalexinproduction were inhibited by blasticidin S. Compared with late cultivars, early potato cultivars allocatea larger part of the available assimilates to the tubers earlyin the growing season, leading to shorter growing periods andlower yields. A dynamic simulation model, integrating effectivetemperature and source –sink relationships of the crop,was used to analyse this relation, using data from experimentsin the Netherlands carried out over 5 years. Dry matter allocationto the tuber in these field experiments was simulated well whenthe tuber was considered as a dominant sink that affects earlinessof a potato crop in two ways: early allocation of assimilatesto the tubers stops foliage growth early in the season and reducesthe longevity of individual leaves. In a sensitivity analysisthe influence of tuber initiation, leaf longevity and the maximumrelative tuber growth rate (Rtb) on assimilate allocation andcrop earliness was evaluated. It was found that the maximumrelative tuber growth rate can influence crop earliness morethan the other two factors, but when conditions for tuber growthare optimal, the leaf longevity is most important. Warm temperatures (35°C day/30°C night) which inhibittuberization in potato (Solanum tuberosum L., cv. Sebago) increasedgibberellin activity in crude extracts from buds, but not frommature leaves, as determined by the lettuce hypocotyl bioassay.Changes in the growth of tubers and stolons indicate the occurrenceof basipetal movement of GA3 applied to the terminal bud ora mature leaf. 14C labelling from GA3 or mevalonic acid injectedjust below the terminal bud was recovered in the lower shoot,stolons and tubers, but the amount transported was greater atcool temperatures (20/15°C). The treatment of potato tubers with 150 µmol dm–3gibberellic acid (GA3) stimulated starch breakdown and hexoseaccumulation in tuber tissues and the transfer of dry matterto stems. These effects could not be accounted for by enhancedactivities of starch phosphorylase, amylase and acid invertase.Indeed enzyme activities either declined or remained relativelyconstant as starch degradation and hexose accumulation proceeded.Changes in the rate of starch depletion were related to changesin sink strength and sink type, the onset of tuber initiationin controls causing the rate of starch degradation to exceedthat in GA3-treated tissues, in which tuberization was inhibited. Potato plants (Solanum tuberosum L.) were grown in water culturein a controlled environment. The growth rates of individualtubers were closely reflected by their 14C-content 20 h after14CO2 had been applied to the aerial parts of the shoot for4 h. The 14C-content of the tuber (sink strength) was significantlycorrelated to the 14C-concentration of the tuber tissue (14Cg–1 fr. wt.=sink activity). The sink activity, which differedbetween individual tubers by up to a factor of 10, was alsoclosely related to the conversion rates of 14C into the starchand the remainder as well as to the 14C-content in the ethanolsoluble fraction. This indicates the simultaneous use of photosynthatefor growth and storage in the growing tubers. No preferenceof photosynthate utilization for either of these processes couldbe detected in relation to the sink activity of the tubers.Tubers with high sink activity imported 14C-labelled photosynthateat higher rates although their tissue contained higher concentrationsof reducing sugars and sucrose than the tissue of tubers withlow sink activity. Despite the close relationship between sinkactivity and the rate of starch synthesis (14C-conversion intostarch), no significant correlation was found between sink activityand the actual starch concentration of the tissue. The applicationof zeatin riboside directly onto individual tubers increasedtheir growth rates in comparison to non-treated tubers of thesame plant. The results indicate the importance of both growthand storage processes for the regulation of sink activity inyoung potato tubers. Potato plants (Solanum tuberosum L.) were grown in water culture.About 14 d after tuber initiation no significant differenceswere found between apical and basal tuber parts in 14C-uptakeand partitioning into various fractions from 14C-labelled photosynthate.Thus, the fresh weight of these tubers could be used as a parameterfor the sink size. The 14C-content per tuber (sink strength)20 h after 14CO2-supply to the foliage was significantly correlatedwith the tuber fresh weight. No correlation was found betweenthe 14C-concentration of the tuber (sink activity; ct. ming fr. wt.) and tuber fresh weight. Consequently, tuberfresh weight (sink size) per se must have been a factor whichinfluenced sink strength. Stolon parameters characterizing theirgrowth prior to tuber initiation (e.g. stolon volume) and theircapacity for photosynthate transport (diameter, length) weremeasured at the time of tuber initiation. Significant correlationswere found between these stolon parameters and subsequent growthof individual tubers. Anatomical studies on the proportion ofvarious tissues in the cross sectional area of stolons supportthe idea of a negative relation between growth of individualtubers and transport resistance in the phloem of the stolons.It is concluded that in the initial phase of tuber growth, mainlyfactors outside of the tuber determine its growth rate. In laterstages of tuber growth, when the sink strength increases, thecompeting strength of individual tubers for photosynthate isdominated mainly by factors within the tuber itself, such astheir sink size and sink activity. The Effect of Temperature on Leaf Appearance in Rice   总被引:10,自引:3,他引:10  
Temperature is the principal environmental determinant of cropleaf appearance. The objective of this study is to analyse whetherthere are different effects of day temperature (TD) and nighttemperature (TN) on main-stem leaf appearance in rice (OryzasativaL.). Plants of 12 rice cultivars were grown at five constant temperatures(22, 24, 26, 28 and 32 °C) and four diurnally fluctuatingtemperatures (TD/TN: 26 /22, 30 /22, 22 /26 and 22 /30 °C)with a constant photoperiod of 12hd-1. The leaf appearance onthe main stem was measured. A constant change in leaf appearance rate was observed duringontogeny. The relation between the number of emerged leavesand days from seedling emergence was described by a power-lawequation with only one cultivar-specific parameter. Values forthis parameter were estimated for the five constant temperaturetreatments, and the relation between this parameter and temperaturewas quantified by a nonlinear model. Leaf appearance for thefour fluctuating temperature treatments could be accuratelypredicted on the basis of these relations in each cultivar.This indicated that there were no specific effects ofTDandTNonleaf appearance in rice, in contrast with phenological developmentto flowering. The optimum temperature for leaf development wasfound to be substantially higher than for development to flowering. The final main-stem leaf number differed with diurnal temperatureconditions. When a diurnal temperature delayed flowering, itincreased the leaf number as well. This might explain whyTDandTNhada different effect on development to flowering but not on leafdevelopment. Photosynthetic and photorespiratoryenzymes in widely divergent plant species with special referenceto the moss Ceratodon purpureus: Properties of ribulose bisphosphatecarboxylase/oxygenase, phosphoenolpyruvate carboxylase and glycolateoxidase.—J. exp. Bot. 36: 1677–1684. Km(CO2) values and maximal velocities of ribulose bisphosphatecarboxylase/oxygenase (E.C. 4.1.1.39 [EC] ) were determined for sixplant species growing in the wild, consisting of a moss, a fernand four angiosperms. The maximum velocities of the RuBP carboxylasesvaried from 0.13 to 0.;62 µmol CO2 fixed min–1 mg–1soluble protein and the Km(CO2) values from 15 to 22 mmol m–3CO2. The highest Km(CO2) values found were for the moss, Ceratodonpurpureus, and the grass, Deschampsia flexuosa. These plantsalso had the highest ratios of the activities of RuBP carboxylaseto RuBP oxygenase. Glycolate oxidase (E.C. 1.1.3.1 [EC] ) activitieswere slightly lower in D.flexuosa, but not in C. purpureus,than for typical C3 species. Phosphoenolpyruvate carboxylase(E.C. 4.1.1.31 [EC] ) was not involved in the photosynthetic carboxylationby these two plants. However, another grass, Phragmites australis,was intermediate in PEP carboxylase activity between C3 andC4 plants The properties of RuBP carboxylase/oxygenase are discussedin relation to the activities of PEP carboxylase and glycolateoxidase and to the internal CO2 concentration. Elongation by dark-grown shoots of Potamogeton pectinatus tuberswas enhanced by the absence of oxygen. This promoting effectwas located in the stem and was stronger under water than ina gas phase and also stronger in unsparged water compared tosparged water. Anaerobic shoots elongated under water by almost13 cm in 5 d. This was the outcome of longer cells of the steminternodes and of some cell division coupled with leaf extensionwhich continued in the absence of oxygen, but at a slower rate.Continued attachment to a starch-filled tuber was required forsignificant anaerobic elongation, which could be sustained forat least 14 d. Switching intact, growing tubers from aerobicto anaerobic conditions stimulated stem extension within 24h. Conversely, stem elongation was slowed when tubers were transferredfrom an anaerobic to an aerobic environment. A marked gravitropicresponse occurred in anaerobic conditions, which involved bothstem and leaf tissue, and indicated that active internal growth-regulatingmechanisms continued to operate without oxygen. Shoot extension by tubers was also stimulated by hypo-aerobicconditions (5–8 kPa O2) compared with fully aerated solutions(20.8 kPa O2). This acceleration was smaller than that obtainedby the complete removal of oxygen, but still involved stem ratherthan leaf growth. Unlike elongation by apical shoots of tubers,that by shoot tips of rhizomes taken from mature light-grownplants was strongly inhibited by lack of oxygen, and in somecases shoot tips died within 5 d. All shoots and leaves werehighly aerenchymatous and the gas-filled lacunae were connectedby side-pores. The kinetic properties of ribulose 1,5-bisphosphate carboxylase(RuBPC) appear to have been modified during evolution of photosynthesisto adjust to changes in substrate availability. C4 plants areconsidered to have a higher concentration of CO2 available toRuBPC than C3plants. In this study, the Km(CO2 and catalyticcapacity (kcat) of RuBPC and the ratio of RuBPC protein to totalsoluble protein from several Flaveria species, including C3,C3-C4 intermediate, and C4 species, were determined. The C3and intermediate species had similar Km(CO2) values while theC4 species on average had higher Km(CO2) values. The mean ratioof Kcat/Km for species of each group was similar, supportingthe hypothesis that changes in Km and Kcat, are linked. Theallocation of total soluble protein to RuBPC was lowest in theC4 Flaveria species, intermediate in the C3-C4 species, andhighest in the C3 species. The results suggest that during evolutionof C4 photosynthesis adjustments may occur in the quantity ofRuBPC prior to changes in its kinetic properties. Plants of all eight isolines of three maturity genes (all combinationsof two alleles at the three lociE1/e1,E2/e2,E3/e3) of soyabean[Glycine max(L.) Merrill] were grown in four different photoperiods(12, 13, 14 or 15 h d-1) at 30/24 °C from first flower openingto harvest maturity. Photoperiod, isoline, and their interaction,affected significantly (P<0.01) the duration between firstand last flowering, and reproductive duration. The interactionsbetween genotype and photoperiod were sufficiently strong thatconsiderable differences in these durations were detected amongisolines in the least-inductive environment (15 h d-1) whereasdifferences were negligible in the most-inductive regime (12h d-1). There was a negative linear relation between photoperiodand both rate of progress from the appearance of the first tothe last flower, and rate of progress from first flowering toharvest maturity; sensitivity to photoperiod varied (P<0.05)six- and five-fold, respectively, among the extreme isolines(e1e2e3andE1E2E3). The three dominant allelesE1,E2andE3, singly,had comparatively little effect on post-flowering traits, butconsiderable epistasis (particularly betweenE1andE2) was detectedfor sensitivity to photoperiod in respect of rates of progressfrom the appearance of the first to the last flower, and fromfirst flower to harvest maturity. The possible roles of oxygen and carbon dioxide treatments inthe presence or absence of ethylene on tuber dormancy releasein potato (Solanum tuberosumL.) were examined. Using two gascompositions (I: 60% CO2–20% O2–20% N2and II: 20%CO2–40% O2–40% N2), the phase of tuber dormancyand previous storage temperature were demonstrated to be importantparameters for dormancy release by these gas mixtures. Gas Icaused decreased abscisic acid (ABA) levels within 24 h regardlessof previous storage temperature, although this effect was reversible.Exogenous C2H4, an effective dormancy release agent, also causeddecreased ABA levels within 24 h. It also enhanced dormancyrelease and further promoted ABA losses by gas I. Gas II treatmentled to slight reductions in ABA levels that were further decreasedby C2H4. Sprout length was modelled successfully by multipleregression analysis in terms of glucose and ABA levels withinthe apical eye tissues of Russet Burbank tubers immediatelyafter, and regardless of, previous gas treatments or storagetemperatures. The lower limit to the negative logarithmic relation betweenseed longevity and moisture content was determined in threesubspecies of rice (Oryza satwa L.) by storing seeds of fivecultivars at 65 °C with 11 different moisture contents (1.5–15.3%f. wt) for various periods up to 150 d and then testing forgermination. The estimates of the low-moisture-content limit(mc) were 4.3% for subsp. indica, 4.4% for subsp. japonica,and 4.5% for subsp. javanica. These moisture contents were inequilibrium with 10.5—12.0% r.h. No significant effectof moisture content between 1.5% and mc on longevity was detected(P > 0.05), while between mc and 15.3% there were negativelogarithmic relations between longevity and moisture content.There were no significant differences in the relations betweenlongevity and moisture either above or below mc between thetwo japonica cultivars or between the two javanica cultivars(P > 0.10). There was also no significant difference in theslope of the negative logarithmic relation between longevityand moisture above mc among the three subspecies (P > 0.25).However, there were significant differences in the standarddeviation of the frequency distribution of seed deaths in timeat any one moisture content, both above and below mc; this isa measure of seed longevity which is independent of pre-storageenvironment, and the differences observed show that there aregenetically determined differences in longevity among the threesubspecies (P < 0.005), indica being the longest and japonicathe shortest lived. The results provide no evidence for intra-specificvariation in mc and support the view that the maximum seed storagemoisture content which provides the maximum longevity is thatwhich is in equilibrium with about 10–11% r.h. It is concludedthat while the seed viability constant Cw of the seed viabilityequation is species specific and therefore applies to most,if not all, cultivars of rice, variation in the value of KEis the source of the differences in potential longevity of thethree subspecies. A comparison of the activity and properties of the enzyme phosphoenolpyruvatecarboxylase (PEPC) was made for plants of Sedum telephium L.grown under low (70 µmol m–2 s–1) or high(500µmol m–2 s–1) PPFD and subjected to varyingdegrees of water stress. Under well-watered conditions onlyplants grown under high PPFD accumulated titratable acidityovernight and the extractable activity of PEPC was almost 2-foldhigher in these plants than in plants grown under low PPFD.Increasing drought stress resulted in a substantial increasein the activity of PEPC extracted both during the light anddark periods and a decrease in the sensitivity to inhibitionby malic acid. The magnitude of these changes was determinedby the severity and duration of drought and by light intensity.A comparison of the kinetic properties of PEPC from severelydroughted plants revealed that plants droughted under high PPFDhad a lower Km for PEP than plants under low PPFD. Additionof 2·0 mol m–3 malate resulted in an increase inthe Km for PEP, with plants draughted under low PPFD havinga significantly higher Km in the presence of malic acid comparedto those under high PPFD. Response to the activator glc-6-P,which lowered the Km for PEP, also varied between plants grownunder the two light regimes. Under well-watered conditions PEPCextracted from plants under high PPFD was more sensitive toactivation by glc-6-P than those under low PPFD. After the severedrought treatment, however, the Km for PEP in the presence ofglc-6-P was similar for enzyme extracted from plants grown underboth light regimes. Soluble sugars and starch were depletedovernight and were both possible sources of substrate for PEPC.With increasing drought, however, the depletion of starch relativeto soluble sugars increased under both light regimes. The propertiesof PEPC and the characteristics of carbohydrate accumulation/depletionare discussed in relation to the regulation of CAM in S. telephiumgrown under different light and watering regimes. The influence of range of concentrations of potassium nitrate(Cm)in the medium surrounding the roots of decapitated Ricinuscommunis plants on: (a) the potassium concentration of the exudate(Cs), (b)the rate of exudation (fH2O), and (c) the flux of potassiuminto the exudate (fk) was investigated. A change in Cm couldinfluence exudation by altering the osmotic potential of themedium as will as by a change in potassium concentration, andthese two effect were separated in the following way. the effectof varying the potassium concentration was investigated usingmedia made isotonic with mannitol, whilst the osmotic effectwas investigated using mannitol solutions of various concentrationsbut of equal potassium concentration. An analysis of the resultsrevealed the existence of two salt-transfer processes, one independentof the water flux but dependent on the concentration of potassiumin the medium, the other linearly related to the flux of waterthrough the root system.
