Summary The effects of nodal explants collected at different plastochrones, use of various benzyladenine (BA) concentrations, sources of carbohydrates, and phases of the culture medium on shoot establishment and proliferation ofRosa hybrida L. andR. chinensis minima were evaluated. Higher numbers of shoots per explant were obtained fromR. hybrida cv. Carefree Beauty explants proximal to the apical meristem than those from distal nodes. However, proliferating shoots derived from plastochrones proximal to the apical meristem had a lower number of leaves/explant and were shorter than those derived from other distal plastochrones. Although shoot proliferation increased with higher BA concentration in the medium, a concentration of 4.4 μM BA was found optimum for axillary bud-break and shoot development forR. hybrida cvs. Adelaide Hoodless and Cuthert Grant. A higher shoot proliferation rate was observed forR. hybrida cv. Carefree Beauty explants grown on a medium containing 55.5 mM fructose than 58.4 mM sucrose. However, no differences were observed forR. hybrida cv. Cuthert Grant grown on a medium containing either fructose or sucrose. The mean number of shoots/explant was higher forR. chinensis minima cv. Red Sunblaze explants grown on a liquid (4.5) than on a solid medium (1.7) for the first reculture; while no significant differences between the two phases of the medium were observed for the second reculture. However, a higher mean number of shoots/explant was observed on solid-phase (4.0) than liquid-phase medium (3.4) for the third reculture. A higher mean number of leaves/shoot was obtained on a solidified medium rather than liquid medium in the first two consecutive recultures, while no differences were observed for the third reculture. Although a significant effect of BA concentration on mean number of shoots/explant was observed for Red Sunblaze nodal explants, the influence of BA concentration decreased in the two consecutive cultures for both phases of the medium. Hyperhydricity was observed on Red Sunblaze shoots grown on the liquid-phase medium.  相似文献

Summary An efficient protocol for in vitro propagation of the valuable medicinal plant, Wasabia japonica (Miq.) Matsumura is described through shoot tip proliferation and direct regeneration. Multiple shoots were induced from shoort tips cultured on Murashige and Skoog (MS) semi-solid medium containing various concentrations (0.5–50 μM) of N6-benzyladenine (BA), thidiazuron, kinetin, and zeatin. A comparison was made on shoot multiplication between semi-solid and liquid culture media. Well-developed shoots were obtained using full-strength MS semi-solid medium containing 5.0 μM BA. However, the greatest shoot proliferation was achieved on either full- or half-strength MS liquid media supplemented with 5.0 μM BA for 4 wk (15.3±0.9 and 15.0±0.7 shoots per explant, respectively), and on half-strength MS liquid medium for 6 wk (25.8±1.3 shoots per explant) in culture. In contrast, the maximum number of shoots per explant on full-strength MS semi-solid medium was achieved with either 5.0 μM BA (10.4±0.6 shoots per explant) or 10.0 μM kinetin (10.9±0.8 shoots per explant). Fresh weight of explants and length of shoots derived from full-strength MS liquid medium (1055±77 mg and 34.2±1.0 mm, respectively) were significantly higher than those derived from full-strength MS semisolid medium (437.6±17.3 mg and 15.4±0.7 mm, respectively). Quarter-strength MS liquid medium had no significant difference in shoot proliferation when compared to quarter-strength MS semi-solid medium. Elongated shoots were separated and rooted on half-strength MS semi-solid media fortified with 1-naphthaleneacetic acid (NAA), indole-3-butyric acid (IBA), or indole-3-acetic acid (IAA) ranging from 0.1 to 10.0 μM. Root formation was greatest with IBA when compared with IAA and NAA. One hundred percent of shoots were rooted on half-strength MS medium with 5.0 μM IBA, while vigorous roots were obtained with 10.0 μM IBA. Micropropagated plantlets were successfully established in soil with 95% survival rate after heardening.  相似文献

Micropropagation of tea (Camellia sinensis (L.) O. Kuntze) has been widely attempted but commercial exploitation of this method is limited by heavy losses during the hardening procedures. In the present study, optimization of time of harvesting (spring and early summer) of microshoots, shoot size, soil pH (4.0–6.4), plant growth regulator treatment (IBA; 500 mg l-1, 30 min) CO2 (9.09/10×10−5 mol l-1 to 10.22/10×10-5 mol l-1 and 20/11×10−5 mol l-1 to 80/13×10−7 mol l-1) enrichment and light (15 μ mol m-2 s-1) conditions in specially designed hardening chambers, made a significant impact on the percent of success for hardening. Following the standardized procedure, up to 71.6% root induction and 73% survival could be achieved. Successful field transfer was also accomplished. Tea (Camellia sinensis) phenylalanine ammonia-lyase (PAL) cDNA was cloned using labelled rice PAL cDNA as a probe. The PAL genes of the tea plant were investigated by restriction fragment length polymorphism (RFLP) analysis using tea PAL cDNA. PAL genetic variation in tea plants was much larger than predicted due to the presence of various hybridized fragments in the Assam hybrids, which are hybrids between C. sinensis var assamica and var sinensis. On the other hand, hybridized band patterns of Japanese green tea cultivars belonging to var sinensis could be divided into five groups. Furthermore, a short-length PAL probe, about 280 bp including the 3 untranslated sequence, detected 3 DNA fragments of different lengths, which were named A, B and D. An experiment tracing the PAL gene heredity showed that A, B and D fragments were inherited according to the Mendelian monogenic ratio. Therefore, PAL genes identifiable by A, B and D fragments are multiple alleles, and the PAL gene is present as a single gene in the tea haploid genome. It was also clear that five groups of Japanese green tea cultivars were characterized by the composition of these PAL fragments. From RFLP analysis using tea PAL cDNA, we succeeded in distinguishing Assam hybrids and Japanese green tea cultivars with high and low catechin content, respectively, and in grouping Japanese green tea at the cultivar level.  相似文献

Summary Nodal explants from hop were exposed to plant growth regulators to determine suitable media for initiation from axillary buds and subsequent micropropagation. Efficient culture establishment (96.6% of explants) was achieved on Murashige and Skoog (MS) medium (modified to contain 1 mg l−1 thiamine hydrochloride) supplemented with 0.57 μM indoleacetic acid (IAA) and 2.22 μM 6-benzylaminopurine (BA). Subsequent transfer of explants to treatments containing an auxin ([1-naphthaleneacetic acid], NAA or IAA) and BA, 6-[γ,γ-dimethylallylamino]purine (2iP), kinetin (KIN) or thidiazuron (N-phenyl-N′-1,2,3-thidiazol-5-ylurea [TDZ]) resulted in significantly different amounts of multiplication. Optimal TDZ-supplemented media elicited a greater than threefold increase in the number of shoots and nodes generated per explant compared to optimal media containing BA, 2iP and KIN. Shoots were successfully rooted using half-strength MS supplemented with 5.71 μM IAA and 4.9 μM indolebutyric acid (IBA), and regenerated plants were successfully transferred to soi. An overall micropropagation schedule, which can be implemented into hop commercialization programs, includes: (i) establishment in MS with 0.57 μM IAA and 2.22 μM BA; (ii) multiplication in MS with 0.57 μM IAA and 2.27 μM TDZ; (iii) elongation in MS; and (iv) rooting in half-strength MS with 5.71 μM IAA and 4.9 μM IBA.  相似文献

An efficient method was developed using floating membrane rafts (Liferaft) for the micropropagation of Aconitum napellus (Ranunculaceae), a cut flower crop with a low natural propagation rate. This was achieved by introducing shoot tips into culture on Murashige and Skoog's (1962) solid medium, or liquid medium-supported rafts, supplemented by different levels of benzyl adenine (BA). Optimum shoot proliferation on solid medium required 4mg/l BA, whereas for expiants supported on rafts optimal proliferation was achieved at 0.25mg/l BA. Maximum shoot proliferation was found using the floating rafts (propagation ratio of 4.2 per month), 45% higher than the maximum value on solid medium. A similar value could be obtained on solid medium after a period of 2 months. The optimal response to BA was similar for fresh weight gain and shoot length. Summary An in vitro propagation system was developed for Echinacea purpurea L. (purple coneflower), a medicinal plant commonly used in the treatment of colds, flu and related ailments. Echinacea seeds were found to be contaminated with systemic fungi and therefore an optimized minimal concentration of Plant Preservation Mixture (PPM) was incorporated in the seed germination medium to recover sterile seedlings. Regeneration was induced on petiole explants from 2-month-old sterile seedlings cultured on medium supplemented with benzylaminopurine (BAP) or thidiazuron (TDZ) in combination with indoleacetic acid (IAA). Two distinct forms of regeneration were identified in cultured petiole explants with histological and morphological observations, viz. the direct formation of somatic embryos on the epidermis and the de novo development of shoots from callus tissues formed in subepidermal cell layers. the results of this study have established a micropropagation system for E. purpurea that will provide sterile plant material for further investigations into medicinally active biochemicals and may facilitate mass production of high-quality E. purpurea plants for the commercial market.  相似文献

Summary Sugarcane (Saccharum spp. hybrid cv. CP 84-1198) embryogenic calluses were induced from young leaves cultured on modified Murashige and Skoog basal medium supplemented with 13.6 μM 2,4-dichlorophenoxyacetic acid. Five concentrations, 0.5, 1.0, 2.5, 5.0, and 10.0 μM, of five different growth regulators, 6-benzylaminopurine, kinetin, 6-γ,γ-(dimethylallylamino)purine, zeatin, and thidiazuron, were tested with or without 22.5 μM α-naphthaleneacetic acid to compare their ability to induce regeneration from embryogenic callus. After 4 wk on medium, the percentage of shoot meristem induction was evaluated, and after 10 wk the total number of shoots produced, as well as the percentage of shoots greater than 1 cm in length, was obtained. Although it had the lowest percentage of elongated shoots, medium containing thidiazuron alone performed better than all other growth regulators tested, with the highest percentage of shoot induction and the largest number of shoots, particularly at a concentration of 2.5 μM.  相似文献

An efficient adventitious shoot regeneration procedure was developed for Gypsophila paniculata L. Using cultivar Arbel, shoot regeneration from the three upper internodes of the stem was monitored on MS media supplemented with different cytokinins (thidiazuron, benzyladenine, kinetin or zeatin) and an auxin (naphthaleneacetic acid). Thidiazuron was found to be the most efficient cytokinin, with up to 100% of the explants forming shoots, at an average of up to 19 shoots per explant being regenerated. The highest percentage of shoot formation was observed in the stem explants originating from the first internode, with all cytokinins tested. The adventitious origin of shoots regenerated from stem explants was confirmed by scanning electron microscopy. The regeneration procedure was found to be applicable to five other gypsophila cultivars (Perfecta, Golan, Gilboa, Flamingo and Tavor). Regenerating plants were successfully transferred to soil, and did not differ in flower color, size or shape from standard vegetatively propagated plants derived from cuttings. This study reports on the biocontrol role birds play in caterpillar pest control of tea plantations of Northeast India. In this area large tracts of tea plantations have been extensively defoliated by the recent invasion of two forest-dwelling geometrid looper caterpillars, Hyposidra spp. and a lymantriid hairy caterpillar, Arctornis submarginata. This exacerbated tea herbivory by two resident pest caterpillars, Biston suppressaria and Eterusia magnifera. Currently there are no identified resident insect predators for any life stage of Hyposidra spp. and A. submarginata. Larvae of these pests drop from tea bushes using salivary thread, allowing caterpillars to escape from insect predators. The study identified 38 native insectivorous bird species in tea plantations, of which four species (Asian-pied starling, Chestnut-tailed starling, Jungle Myna, Red-vented Bulbul) could be potential control agents of looper and hairy caterpillar pests. These species had high population densities. Their cumulative abundances represented a major proportion of the total bird community during both the infested (86.44%) and non-infested phase (75.34%). They foraged in mixed-species flocks in both tea foliage and on the ground. This behavior is suited to capture foliage-living and dropped caterpillars that were flushed from tea bushes by foraging birds. Abundance and species richness of overall tea layer-foraging birds were higher in infested phase when compared to non-infested phase. The predation rate of four bird species of the foraging flock varied significantly. These results suggest that birds should be considered as important biological control agent of caterpillar pests of tea and considered in pest management plans.  相似文献

Water stress is a major limitation for plant survival and growth. Several physiological and antioxidative mechanisms are involved in the adaptation to water stress by plants. In this experiment, tea cultivars (TV-1, TV-20, TV-29 and TV-30) were subjected to drought stress by withholding water for 20 days followed by rehydration. An experiment was thus performed to test and compare the effect of dehydration and rehydration in growing seedlings of tea cultivars. The effect of drought stress and post stress rehydration was measured by studying the reactive oxygen species (ROS) metabolism in tea. Water stress decreased nonenzymic antioxidants like ascorbate and glutathione contents with differential responses of enzymic antioxidants in selected clones of Camellia sinensis indicating an oxidative stress situation. This was also apparent from increased lipid peroxidation, O2 and H2O2 content in water stress imposed plants. But the oxidative damage was not permanent as the plants recovered after rehydration. Comparatively less decrease in antioxidants, higher activities of POX, GR, CAT with higher phenolic contents suggested better drought tolerance of TV-1, which was also visible from the recovery study, where it showed lower ROS level and higher recovery of antioxidant property in response to rehydration, thus proving its better recovery potential. On the other hand, highest H2O2 and lipid peroxidation with decrease in phenolic content during stress in TV-29 suggested its sensitivity to drought. The antioxidant efficiency and biochemical tolerance in response to drought stress thus observed in the tested clones of Camellia sinensis can be arranged in the order as TV-30 > TV-1 > TV-29 > TV-20.  相似文献

从茶树内生真菌筛选产漆酶的菌株,分析不同营养因素和培养条件对菌株漆酶酶活力的影响。采用6种显色底物的平板初筛和酶活测定的复筛方法,从15株茶树内生真菌菌株中筛选获得1株产漆酶酶活较高的菌株CSN 4。单因素分析结果显示,液态发酵条件下菌株CSN-4适宜的主要培养基成分是麸皮和蛋白胨;菌株CSN-4分别在麸皮30 g/L、蛋白胨2.5 g/L、CuSO4·5H2O 0.015 g/L和茶水6 g/L时发酵产漆酶酶活最高。发酵条件试验结果表明,菌株CSN-4分别在接种量为6个菌饼(直径6 mm)、装液量60 mL/250 mL、pH 4.8、摇床转速120 r/min,培养温度为28 ℃时产漆酶酶活较高。在培养基中添加麸皮和茶水对菌株CSN-4产漆酶有明显的促进作用。经过培养基成分及培养条件优化后,菌株CSN 4产漆酶酶活显著升高,达到2 417 U/L。  相似文献

Summary The effect of ventilation during the multiplication stage on the development of propagules from different clones of jojoba [Simmondsia chinensis (Link) Schneider] was investigated. Variation in the response to ventilation was due to genotype, the extent of ventilation, and to the period of exposure (transfer number). With intermediate ventilation treatments, propagules elongated to a greater extent and produced more dry biomass than propagules grown without ventilation. In the highest ventilation treatment, however, growth parameters were negatively affected. More importantly, propagules grown with moderate ventilation produced more plant material suitable for further multiplication and for the elongation stage than those grown in sealed tubes—the vessels used in our original micropropagation system. In five of the seven clones studied, growth and multiplication rate were decreased by the highest ventilation treatment. Propagules from the second and third multiplication transfers into ventilated vessels became more sensitive to high ventilation. Ambient water loss was slower in propagules produced under ventilation, probably due to smaller stomatal apertures. As a result of improved growth and decreased hyperhydricity by ventilation, the micropropagation protocol should be modified to include Magenta boxes equipped with vented lids as the preferred growing vessels.  相似文献

Cytokinins, donor plants and their time in vitro as well as basal media were investigated for their influence on shoot regenerative capacity of American elm (Ulmus americana L.) leaves. Leaves excised from six 2-year-old seedlings formed adventitious shoots when placed on Driver and Kuniyuki Walnut (DKW) medium supplemented with 7.5, 15 or 22.5 M of benzyladenine (BA) or thidiazuron (TDZ). Thidiazuron induced significantly higher regeneration percentages on elm leaves than BA, regardless of concentration used. Donor plant also affected the efficiency of shoot regeneration, with certain seedlings having 1.5 to 7 times more explants forming shoots as compared to other seedlings tested. By subculture 15, the average number of shoots per regenerating explant increased at least 3-fold for leaves on media with BA or TDZ for the one donor plant that survived continued subculturing. Leaf explants from donor plants with the highest regenerative capacity had a higher percentage of shoot formation on DKW than MS medium. Explants from productive donor plants should be placed on DKW medium supplemented with TDZ to improve shoot regeneration efficiency from American elm leaves.  相似文献

Multiple shoots of Curcuma longa were induced by culture of bud explants for 1week in Murashige and Skoog (MS) liquid medium supplemented with 72.64M thidiazuron (TDZ) prior to culture on MS gelled medium without growth regulator for 8weeks. The regeneration rate was up to 11.4±1.7shoots/explant. Rooting was spontaneous and the regenerated plants were successfully transferred to soil. This protocol can be an alternative for rapid micropropagation of C. longa used for phytomedicine raw material production.  相似文献

The direct induction of adventitious shoots from leaf explants obtained from adult plants of Zhanhua winter jujube, an elite variety of Zizyphus jujuba Mill., is reported. The percentage of leaf explants producing shoots and the average number of shoots per explant were significantly improved when 10-day-old leaves were explanted onto Woody Plant Medium and maintained initially in the dark. The plant growth regulator thidiazuron (TDZ) was effective in stimulating shoot regeneration from leaf explants of Zhanhua winter jujube. The highest efficiency of shoot formation was observed with a 20-day culture in the dark on WPM containing 4.54 M TDZ and 2.85 M indoleacetic acid (IAA). The regenerated shoots were transferred to MS medium containing 0.89 M benzyladenine and 5.77 M gibberellic acid for growth. When the shoots were about 2 cm in height, they were transferred to Nitsch medium supplemented with 1.14 M IAA and 2.46 M indolebutyric acid to induce rooting. This system of adventitious shoot production from leaf explants of adult plants could be useful for the genetic engineering and polyploidization of winter jujube.  相似文献

Micropropagation of kiwifruit using non-axenic shoot tips   总被引:6,自引:0,他引:6  
Kiwifruit (Actinidia chinensis Planch.) shoot tips were subjected to a standard surface sterilization procedure and cultured on a Murashige and Skoog basal medium in the presence of two surviving bacterial contaminants. The fresh weight increase of the cultures and the number of shoots produced were greater in liquid medium than in medium solidified with 0.4 or 0.8% agar. A greater number of shoots was obtained with 125 ml than with 50, 250, or 500 ml Erlenmeyer flasks. A concentration of 2 mgl-1 N6-benzylaminopurine (BAP) gave a greater increase in fresh weight than either 0 or 4 mgl-1.Shoots cut from proliferating cultures were dipped in 0.05% indolebutyric acid (IBA) and rooted directly in a peat: vermiculite: perlite mix. Over 93 % of 907 plantlets produced were successfully acclimatized. The productivity of the method was comparable to that reported for the axenic culture of meristems. The contaminants which survived the initial surface sterilization procedure thus presented no major obstacle to the in vitro propagation of kiwifruit.  相似文献
