Two sulfated polysaccharides WF1 and WF3 were isolated from marine green algae Monostroma nitidum, and their structural characteristics were determined. Anticoagulant activities of WF1 and WF3 were evaluated by assays of the activated partial thromboplastin time (APTT), thrombin time (TT), prothrombin time (PT), antithrombin and anticoagulation factor Xa activities. The results showed that WF1 and WF3 had similar high contents of rhamnose, whereas their sulfate contents, sulfation positions, molecular sizes and linkage patterns of rhamnose residues were different. The bioassay results demonstrated that WF1 and WF3 had high anticoagulant activities, and were potent thrombin inhibitors mediated by heparin cofactor II, especially WF3. They also hastened thrombin and coagulation factor Xa inhibition by potentiating antithrombin III, but at a lower effectiveness. The differences of anticoagulant activities between WF1 and WF3 were directly due to their structural features discrepancy.  相似文献

The effect of structurally different sulfated polysaccharides from brown algae Fucus evanescens, Saccharina cichorioides, and Saccharina japonica on the morphofunctional changes of dendritic cells has been studied using flow cytometry and phase-contrast microscopy. It has been found that the dendritic cells are characterized by larger sizes, vacuolated cytoplasm, eccentrically located nucleus, the presence of numerous cytoplasmic pseudopodia of various forms; they do express surface markers, thus providing evidence for their maturation (CD83, CD11c, HLA-DR, CD86). Increased production of immunoregulatory (IL-12) and proinflammatory cytokines (TNF-α, IL-6) by dendritic cells polarizes the development of the Th-1 type immune response.  相似文献

A number of sulfated polysaccharides were tested for their ability to inhibit passively induced experimental allergic encephalomyelitis (EAE) in rats. Heparin and fucoidan both completely inhibited passive EAE even when treatment was begun 3 days after transfer of cells. Pentosan sulfate was partially inhibitory whereas chondroitin-4-sulfate had no effect. Inhibition was not merely due to killing of the cells since active sensitization 14 days after cell transfer resulted in an early onset of disease indicating the persistence of transferred cells as memory cells. Although all the inhibitory polysaccharides are anticoagulants, it would appear that this function alone is not the reason for inhibition since a heparin preparation devoid of anticoagulant activity also partially inhibited EAE. Actively induced EAE was also significantly delayed by treatment with heparin. The results are discussed in terms of the polysaccharides inhibiting the enzymatic dependent movement of lymphocytes across central nervous system vascular endothelium.  相似文献

Sulfated polysaccharides from the green algae Ulva conglobata were isolated and prepared by extraction in hot water, precipitation with ethanol and purification by ion-exchange and size-exclusion column chromatography. The characterizations of the sulfated polysaccharides were defined, and containing 23.04–35.20% sulfate ester groups, 10.82–14.91% uronic acid and 3.82–4.51% protein. Gas chromatography analysis shows that the sulfated polysaccharides from Ulva conglobata are mainly consisted of rhamnose with variable contents of glucose and fucose, trace amounts of xylose, glactose and mannose. The anticoagulant properties of the sulfated polysaccharides were compared with those of heparin by studying the activated partial thromboplastin time using normal human plasma. The sulfated polysaccharide from Ulva conglobata collected in Qingdao, China is the most potent among the sulfated polysaccharides tested. The mechanism of anticoagulant activity mediated by the sulfated polysaccharides is due to the direct inhibition of thrombin and the potentiation of heparin cofactor II.  相似文献

Summary We previously demonstrated that human keratinocyte cultures proliferate in the absence of polypeptide growth factors (autonomous growth) and that this autonomous growth is blocked by interaction of heparin with a human keratinocyte-derived autocrine factor (KAF) which we identified as amphiregulin (AR). In the present study, we demonstrate that sulfated polysaccharides other than heparin (low and high molecular weight dextran sulfates) also inhibit the AR-mediated autonomous proliferation of human keratinocytes. Furthermore, sulfated polysaccharides such as high and low molecular weight dextran sulfates, heparan sulfate and, to a lesser extent, chondroitin sulfates B and C were also shown to be inhibitors of human keratinocyte-derived AR (k-d AR)-stimulated DNA synthesis in quiescent murine AKR-2B cell cultures. Our results demonstrate that sulfation of polysaccharides is required for AR inhibitory activity, and that several sulfated polysaccharides (other than heparin) can act as inhibitors of AR-mediated autonomous proliferation in human epidermal keratinocytes and as inhibitors of k-d AR-mediated mitogenic activity in AKR-2B cells.  相似文献

Lichens are able to tolerate almost complete desiccation and can quickly resume metabolic activity after rehydration. In the desiccated state, photosynthesis is completely blocked and absorbed excitation energy cannot be used for electron transport, leading to a potential strong vulnerability for high light damage. Although desiccation and high insolation often occur simultaneously and many lichens colonize exposed habitats, these organisms show surprisingly little photodamage. In the desiccated state, variable chlorophyll fluorescence is lost, indicating a suspension of charge separation in photosystem II. At the same time, basal fluorescence (F (0)) is strongly quenched, which has been interpreted as an indication for high photoprotective non-radiative dissipation (NRD) of absorbed excitation energy. In an attempt to provide evidence for a photoprotective function of NRD in the desiccated state, isolated green lichen algae of the species Coccomyxa sp. and Trebouxia asymmetrica were used as experimental system. In contrast to experiments with intact lichens this system provided high reproducibility of the data without major optical artifacts on desiccation. The presence of 5?mM trehalose during desiccation had no effect but culture of the algae in seawater enhanced F (0) quenching in T. asymmetrica together with a reduced depression of F (V)/F (M) after high light treatment. While this effect could not be induced using artificial seawater medium lacking trace elements, the addition of ZnCl(2) and NaI in small amounts to the normal growth medium led to qualitatively and quantitatively identical results as with pure seawater. It is concluded that NRD indicated by F (0) quenching is photoprotective. The formation of NRD in lichen algae is apparently partially dependent on the presence of specific micronutrients.  相似文献

Summary The environmental conditions prevailing in Israel make marine algae an attractive crop for the production of valuable chemicals. A marine species of Porphyridium seems to fit this purpose.The unicellular red alga Porphyridium is encapsulated by a polysaccharide envelope that is present in the gel state. This polysaccharide is an acidic heteropolymer composed of sulfated sugars. It forms ionic bridges through divalent cations, thus reaching a very high molecular weight. The thickness of the polysaccharide capsule varies according to the phase of growth and the growth conditions. Its outer part dissolves in the growth medium, which becomes progressively more viscous. Sulfated polysaccharides form theramlly reversible gels similar to agar and carrageenan, which are usually extracted from marine macroalgae. These gels have been finding increasing use in commercial applications as gelling agents, thickeners, stabilizers, and emulsifiers.We have done experiments on the cultivation of a marine species of Porphyridium for the production of polysaccharides. This unicellular alga has an advantage over the macroalgae due to its relatively faster growth rate and the possibility to regulate its growth. The potential for production of the polysaccharide, both that dissolved in the external medium and that attached to the cell (including an intracellular fraction), and the effects of growth conditions on productivity were suudied in the laboratory. Porphyridium was also cultivated outdoors in seawater in 1-m2 ponds and its growth potential investigated.  相似文献

Microwave-assisted desulfation of sulfated polysaccharides   总被引:1,自引:0,他引:1  
Several sulfated polysaccharides were desulfated by heating their pyridinium salts dissolved in dimethyl sulfoxide in a microwave oven. The procedure was applied to different products like a λ-carrageenan, a partially cyclized μ/ν-carrageenan, an agar-like product like corallinan, a fucoidan and a chondroitin sulfate. When their pyridinium salts were generated in carefully chosen conditions, the desulfation proceeds smoothly after 1 min of heating, leading to a removal of 60–93% of the original sulfate present. Some depolymerization was found to occur, but its effect is moderate (yields of the recovered products are 64–89%), even for polysaccharides with labile bonds. An in situ methylation procedure was coupled with the microwave-aided desulfation method in order to facilitate the retrieval of structural data. Both spectroscopical and chemical studies showed that the integrity of the polysaccharides was not affected by this procedure (besides the loss of sulfate and the above-mentioned depolymerization).  相似文献

对超声辅助法提取的南瓜粗多糖,进行化学改性得到了硫酸化南瓜多糖和羧甲基南瓜多糖。用邻苯三酚自氧化法检测对超氧阴离子自由基的清除作用,用邻二氮菲-金属铁离子-H2O2体系检测对羟自由基的清除作用。结果表明,南瓜多糖对超氧阴离子自由基无抑制作用,经化学改性后的羧甲基南瓜多糖和硫酸化南瓜多糖对超氧阴离子自由基有明显抑制作用,最大清除率达44%和41.67%。以上3种多糖均能有效清除·OH,随着浓度的增加清除作用加强,最大清除率可达46.49%,22.61%和42.35%0。结果显示,经化学改性后的南瓜多糖有较强的抗氧化作用。  相似文献

An anticoagulant sulfated galactan isolated from the marine green alga, Codium cylindricum, was shown to have antiangiogeinic activity. This galactan suppressed microvessel formation in an ex vivo serum-free matrix culture model using rat aortic ring. It also inhibited human umbilical vein endothelial cells (HUVEC) tube formation on reconstituted basement membrane gel. These results show the value of algal sulfated galactans in the design of antiangiogenic agents.  相似文献

Simple separation of anticoagulant sulfated galactan from marine red algae   总被引:1,自引:0,他引:1  
In this study, hot water extracts of 22 red algal species were evaluated for their potential anticoagulant activities. The extracts from eight species (Grateloupia elliptica, Sinkoraena lancifolia, Halymenia dilatata, Grateloupia lanceolata, Lomentaria catenata, Martensia denticulata, Schizymenia dubyi, Chondrus crispus) showed potent activated partial thromboplastin time (APTT). Of these eight algae, the crude polysaccharide fraction (CpoF) from the hot water extracts of L. catenata and S. dubyi showed the highest APTT activity. Lomentaria catenata and S. dubyi were selected and an enzymatic digestion process which could effectively separate a crude polysaccharide fraction with higher yields from raw algae materials was applied. The 10 enzymes tested included five carbohydrases and five proteases. The Ultraflo and Celluclast digests of L. catenata and the AMG digest of S. dubyi exhibited the most potent anticoagulant activity. Furthermore, in both species, the active compounds were mainly concentrated in the >30 kDa fraction through ultrafiltration and showed strong APTT (>1000) and thrombin time (TT) >1000 activity. The active compounds were shown to be sulfated galactans with a greater than 80% galactose content and an 0.22 ∼ 0.31 sulfate to total sugar ratio.  相似文献

The composition, molecular weight and in vitro antioxidant activity of various sulfated polysaccharides obtained by anion exchange chromatography, acid hydrolysis and radical process degradation of the crude sulfated polysaccharide extracted from Laminaria japonica were compared. The low sulfated F-A2, with a peak-molecular weight (Mp) of 5–15 kDa, 14.5% sulfated ester and 21.8% glucuronic acid, exhibited a very strong antioxidant activity on superoxide and hydroxyl radicals, with activity even higher than that of large molecular weight fractions F-A and F-B. However, highly sulfated fractions with a peak-molecular weight below 15 kDa had much lower antioxidant activities than other fractions. These results indicated that the sulfate group of the low molecular weight fractions represents a physical block for the reaction with oxygen radicals. The chemical properties and antioxidant activities of sulfated polysaccharide fractions obtained by radical process degradation of crude sulfated polysaccharide were quite different from those obtained by acid hydrolysates. By radical process degradation, the high molecular weight was decreased to give LM2 (Mp 8 kDa) and LM1 (Mp 1.5 kDa), with a yield of 40% and 15%, respectively. LM2 was enriched with fucose and sulfated ester, while containing low amounts of glucuronic acid. The antioxidant activity showed that LM2 was unable to scavenge either superoxide or hydroxyl radical, which suggested that radical process degradation targeted mainly ascopyllan-like species rich in glucuronic acid, while the fraction rich in sulfated l-fucose remained unchanged. However, LM1 with Mp 1.5 kDa still retained apparent scavenging ability for superoxide radical, although it contained no glucuronic acid and certain amounts of galactose and mannose as main neutral sugars. These result suggest that the antioxidant activity of sulfated polysaccharides is apparently related not only to molecular weight and sulfated ester content, as previously determined, but also to glucuronic acid and fucose content.  相似文献

A homogeneous sulfated heterorhamnan was obtained by aqueous extraction, then by ultrafiltration from the green seaweed Gayralia oxysperma. Besides α-l-rhamnose it contains glucuronic and galacturonic acids, xylose and glucose. The structure was established by methylation analyses of the carboxyl-reduced, carboxyl-reduced/desulfated, carboxyl-reduced/Smith-degraded, and carboxyl-reduced/Smith-degraded/desulfated products and 1D, 2D NMR spectroscopy analyses. The heterorhamnan backbone is constituted by 3- and 2-linked rhamnosyl units (1.00:0.80), the latter being ∼50% substituted at C-3 by side chains containing 2-sulfated glucuronic and galacturonic acids and xylosyl units. The 3- and 2-linked rhamnosyl units are unsulfated (20%), disulfated (16%), and mostly monosulfated at C-2 (27%) and C-4 (37%). The branched and sulfated heterorhamnan had high and specific activity against herpes simplex virus.  相似文献

The in vitro antioxidant activities of the following six sulfated polysaccharides were investigated: iota, kappa and lambda carrageenans, which are widely used in the food industry, fucoidan (homofucan) from the edible seaweed Fucus vesiculosus and fucans (heterofucans) F0.5 and F1.1 from the seaweed Padina gymnospora. With respect to the inhibition of superoxide radical formation, fucoidan had an IC50 (the half maximal inhibitory concentration) of 0.058 mg·mL−1, while the IC50 for the kappa, iota and lambda carrageenans were 0.112, 0.332 and 0.046 mg·mL−1, respectively. All of the samples had an inhibitory effect on the formation of hydroxyl radicals. The results of peroxidation tests showed that fucoidan had an IC50 of 1.250 mg·mL−1 and that the kappa, iota and lambda carrageenans had an IC50 of 2.753 and 2.338 and 0.323 mg·mL−1, respectively. Fucan fractions showed low antioxidant activity relative to fucoidan. These results clearly indicate the beneficial effect of algal polysaccharides as antioxidants.  相似文献

Sulfated polysaccharides isolated from Capsosiphon fulvescens and fractionated using ion-exchange chromatography were investigated to determine their chemical and molecular characteristics and biological activities. The crude and fractionated polysaccharides (F(1), F(2), and F(3)) consisted mostly of carbohydrates (28.9-67.0%), uronic acids (1.6-9.2%) and sulfates (5.2-13.4%) with various amounts of proteins (2.1-53.7%). Their monosaccharide levels were significantly different including rhamnose (20.8-65.2%), xylose (13.0-37.1%) and mannose (11.6-65.1%). The polysaccharides contained one or two subfractions with molecular weights (M(w)) ranging from 401.7×10(3) to 6232×10(3)g/mol. These polysaccharides (the crude and fraction F(2)) strongly stimulated macrophage cells, RAW264.7 cell line, producing considerable amounts of NO, PGE(2) and cytokines which suggested that they could be strong immunostimulators.
