Awn absence was shown to be inherited as a dominant character in the tetraploid wheat species Triticum dicoccum (Schrank) Schuebl. and T. durum Desf. but as a recessive one in T. aethiopicum Jakubz. The monogenic control of the character was demonstrated for all studied species. In accessions of emmer and durum wheat, the character is controlled by the dominant gene B1, located on chromosome 5A, and in Ethiopian wheat, by a recessive gene, which we designated as awn. The recessive awn gene was localized on chromosome 3B of T. aethiopicum with the use of D-genome disomic substitution lines of cultivar Langdon.  相似文献

The inheritance of two taxonomically important characters was studied in hexaploid wheat species (2n = 6x = 42). The monogenic control of spherical grain was demonstrated for Triticum antiquorum Heer ex Udacz. The recessive gene controlling spherical grain in this species was assigned to chromosome 3D by monosomic genetic analysis and was shown to be allelic to the s gene determining the same character in the endemic Indian species T. sphaerococcum Perciv. The T. antiquorum and T. sphaerococcum dominant genes controlling compact ears proved to be nonallelic to the corresponding T. compactum Host. gene and were designated as C2. Cytogenetic analysis was conducted on a set of 67 Ethiopian wheat accessions collected by the expedition of N.I. Vavilov in 1927 and 85 years later by the Joint Ethiopian–Russian Biological Expedition (JERBE) in 2012 in the same sites of Ethiopia. The preservation of the polymorphism system of heterochromatic chromosome sites upon the change in the Ethiopian wheat population structure over the past period owing to a frequency shift of some specific chromosome variants and an increase in the proportion of genotypes carrying marker rearrangements was demonstrated. The unevenness of the geographical distribution of the 2A:4B translocation and of the 5A inversion was identified, and it was demonstrated that wheat accessions from Eritrea were cytogenetically the most isolated from the population from the central regions of Ethiopia. A low level of the Ethiopian wheat polymorphism was found along with the prevalence of the same chromosome rearrangement variants, which was indicative of monophyletic origin of the species. It was suggested that Triticum aethiopicum could have diverged from Ethiopian emmer as a result of hybridization with other wheat species, while subsequent evolution of these species occurred independently. Evidence for the participation of Ethiopian wheat in the formation of the gene pool of the unique Moroccan group of T. dicoccum was obtained.  相似文献

Background

Vernalization genes VRN1 play a major role in the transition from vegetative to reproductive growth in wheat. In di-, tetra- and hexaploid wheats the presence of a dominant allele of at least one VRN1 gene homologue (Vrn-A1, Vrn-B1, Vrn-G1 or Vrn-D1) determines the spring growth habit. Allelic variation between the Vrn-1 and vrn-1 alleles relies on mutations in the promoter region or the first intron. The origin and variability of the dominant VRN1 alleles, determining the spring growth habit in tetraploid wheat species have been poorly studied.

Results

Here we analyzed the growth habit of 228 tetraploid wheat species accessions and 25 % of them were spring type. We analyzed the promoter and first intron regions of VRN1 genes in 57 spring accessions of tetraploid wheats. The spring growth habit of most studied spring accessions was determined by previously identified dominant alleles of VRN1 genes. Genetic experiments proof the dominant inheritance of Vrn-A1d allele which was widely distributed across the accessions of Triticum dicoccoides. Two novel alleles were discovered and designated as Vrn-A1b.7 and Vrn-B1dic. Vrn-A1b.7 had deletions of 20 bp located 137 bp upstream of the start codon and mutations within the VRN-box when compared to the recessive allele of vrn-A1. So far the Vrn-A1d allele was identified only in spring accessions of the T. dicoccoides and T. turgidum species. Vrn-B1dic was identified in T. dicoccoides IG46225 and had 11 % sequence dissimilarity in comparison to the promoter of vrn-B1. The presence of Vrn-A1b.7 and Vrn-B1dic alleles is a predicted cause of the spring growth habit of studied accessions of tetraploid species. Three spring accessions T. aethiopicum K-19059, T. turanicum K-31693 and T. turgidum cv. Blancal possess recessive alleles of both VRN-A1 and VRN-B1 genes. Further investigations are required to determine the source of spring growth habit of these accessions.

Conclusions

New allelic variants of the VRN-A1 and VRN-B1 genes were identified in spring accessions of tetraploid wheats. The origin and evolution of VRN-A1 alleles in di- and tetraploid wheat species was discussed.

Segregation analysis of resistance to powdery mildew in a F2 progeny from the cross Chinese Spring (CS) × TA2682c revealed the inheritance of a dominant and a recessive powdery mildew resistance gene. Selfing of susceptible F2 individuals allowed the establishment of a mapping population segregating exclusively for the recessive resistance gene. The extracted resistant derivative showing full resistance to each of 11 wheat powdery mildew isolates was designated RD30. Amplified fragment length polymorphism (AFLP) analysis of bulked segregants from F3s showing the homozygous susceptible and resistant phenotypes revealed an AFLP marker that was associated with the recessive resistance gene in repulsion phase. Following the assignment of this AFLP marker to wheat chromosome 7A by means of CS nullitetrasomics, an inspection of simple sequence repeat (SSR) loci evenly spaced along chromosome 7A showed that the recessive resistance gene maps to the distal region of chromosome 7AL. On the basis of its close linkage to the Pm1 locus, as inferred from connecting partial genetic maps of 7AL of populations CS × TA2682c and CS × Virest (Pm1e), and its unique disease response pattern, the recessive resistance gene in RD30 was considered to be novel and tentatively designated mlRD30.Communicated by C. Genetic control of some morphological traits and the gliadin composition were examined in plants of two lines of common wheat carrying genes introgressed from the wild diploid cereal Aegilops speltoides. Leaf hairiness was shown to be controlled by a single introgressed dominant gene that was not allelic to the known common wheat gene Hl1. Waxlessness of the whole plant is controlled by the introgressed from Ae. speltoides inhibitor gene allelic to gene W1 I located on chromosome 2B. This gene was epistatic to the introgressed gene controlling spike waxlessness. The introgressed gene of spike color was shown to be allelic to Rg1 located on chromosome 1B of common wheat. Two Chinese wheat lines Jieyan 94-1-1 and Siyan 94-1-2 are resistant to all 120 isolates of Blumeria graminis f. sp. tritici maintained in Weihenstephan, Germany. Monosomic analyses employing the susceptible set of 21 Chinese Spring monosomic lines revealed that the line Jieyan 94-1-1 carries one dominant gene on translocated wheat/rye chromosome 1B/1R and one recessive gene on chromosome 7B, whereas line Siyan 94-1-2 possesses one recessive gene on chromosome 7B and one dominant gene on chromosome 5D. Allelism tests in combination with the use of specific isolates comfirmed that the dominant genes in Jieyan 94-1-1 and Siyan 94-1-2 are Pm8 and Pm2, respectively. The recessive genes present in each of the two lines are shown to be new alleles located on chromosome 7B at the pm 5 locus. The two genes are tentatively designated mljy in Jieyan 94-1-1 and mlsy in Siyan 94-1-2, respectively.  相似文献

Fluorescence in situ hybridization (FISH) was used to study the distribution of Spelt-1 repetitive DNA sequences on chromosomes of 37 accessions representing eight polyploidy wheat species of the Emmer evolutionary lineage: Triticum dicoccoides Körn, T. dicoccum (Schrank) Schuebel, T. durum Desf., T. polonicum L., T. carthlicum Nevski, T. aethiopicum Jakubz., T. aestivum L., and T. spelta L. Substantial polymorphism in the number, distribution, and the sizes of the Spelt-1 loci was revealed. On the chromosomes of the accessions examined, Spelt-1 tandem repeats were found in seven different positions (per haploid chromosome set). These were “potential hybridization sites”, including the subtelomeric regions of either short or long arms of chromosomes 2A and 6B, the short arm of chromosome 1B, and the long arms of chromosomes 2B and 3B. However, in individual genotypes, only from one to three Spelt-1 loci were revealed. Furthermore, no hybridization with Spelt-1 probe was detected on chromosomes from 12 accessions. Thus, the total number of Spelt-1 sites in karyotypes varied from zero to three, with the average number of 1.16. This was substantially lower than in the species of the Timopheevi section and diploid Aegilops speltoides Tausch, a putative donor of the B genome. The decrease of the content of Spelt-1 sequences in the genomes of the Emmer group wheats in comparison with the species of the Timopheevii group and diploid Ae. speltoides was assumed to result from the repetitive sequences reorganization during polyploidization and the repeat elimination during wheat evolution.  相似文献

A 4B/5R wheat-rye translocation line derived from the Danish wheat variety Viking was revealed to be highly copper efficient. The chromosomal exchange includes a very small terminal segment of chromosome arm 5RL of rye which was physically mapped by genomic DNA: DNA in situ hybridization and chromosome analysis. The gene for Cu efficiency (Ce) is linked to a dominant hairy neck character from rye (Ha1) and to two rye-specific leaf esterase loci (Est6, Est7), all of which are postulated to map to the distal part of 5RL. Genes coding for mugineic acid synthetase and 3-hydroxymugineic acid synthetase also on chromosome 5R are not included in the 4B/5R translocation and hence map outside the terminal 5R region. These genetic and molecular markers can be useful tools for large-scale screening in wheat breeding programmes.  相似文献

RFLP variation revealed by protein disulfide isomerase (PDI) coding gene sequences was assessed in 170 accessions belonging to 23 species of Triticum and Aegilops. PDI restriction fragments were highly conserved within each species and confirmed that plant PDI is encoded either by single-copy sequences or by small gene families. The wheat PDI probe hybridized to single EcoRI or HindIII fragments in different diploid species and to one or two fragments per genome in polyploids. Four Aegilops species in the Sitopsis section showed complex patterns and high levels of intraspecific variation, whereas Ae. searsii possessed single monomorphic fragments. T. urartu and Ae. squarrosa showed fragments with the same mobility as those in the A and D genomes of Triticum polyploid species, respectively, whereas differences were observed between the hybridization patterns of T. monococcum and T. boeoticum and that of the A genome. The single fragment detected in Ae. squarrosa was also conserved in most accessions of polyploid Aegilops species carrying the D genome. The five species of the Sitopsis section showed variation for the PDI hybridization fragments and differed from those of the B and G genomes of emmer and timopheevi groups of wheat, although one of the Ae. speltoides EcoRI fragments was similar to those located on the 4B and 4G chromosomes. The similarity between the EcoRI fragment located on the 1B chromosome of common and emmer wheats and one with a lower hybridization intensity in Ae. longissima, Ae. bicornis and Ae. sharonensis support the hypothesis of a polyphyletic origin of the B genome.  Stripe rust caused by Puccinia striifomis West. is one of the most devastating diseases relating to wheat production. Wild emmer wheat, Triticum dicoccoides, the tetraploid progenitor of cultivated wheat, has proven to be a valuable source of novel stripe-rust resistance genes for wheat breeding. For example, T. dicoccoides accessions from Mt. Hermon, Israel, are uniformly and highly resistant to stripe-rust. The main objective of the present study is to map a stripe-rust resistance gene, derived from the unique Mt. Hermon population of wild emmer, using microsatellite markers. An F2 mapping population was established by crossing stripe-rust resistant T. dicoccoides accession H52 from Mt. Hermon with the Triticum durum cultivar Langdon. The stripe-rust resistance derived from accession H52 was found to be controlled by a single dominant gene which was temporarily designated as YrH52. Out of 120 microsatellite markers tested, 109 (91%) showed polymorphism between the parental lines. Among 79 segregating microsatellite loci generated from 56 microsatellite primer pairs, nine were linked to YrH52 with recombination frequencies of 0.02–0.35, and LOD scores of 3.56–54.22. A genetic map of chromosome 1B, consisting of ten microsatellite loci and the stripe-rust resistance gene YrH52, was constructed with a total map length of 101.5 cM. YrH52 is also closely linked to RFLP marker Nor1 with a map distance of 1.4 cM and a LOD value of 29.62. Apparent negative crossover interference was observed in chromosome 1B, especially in the region spanning the centromere.  Low-temperature (LT) induced genes of the Wcs120 family in wheat (Triticum aestivum) were mapped to specific chromosome arms using Western and Southern blot analysis on the ditelocentric series in the cultivar Chinese Spring (CS). Identified genes were located on the long arms of the homoeologous group 6 chromosomes of all 3 genomes (A, B, and D) of hexaploid wheat. Related species carrying either the A, D, or AB genomes were also examined using Southern and Western analysis with the Wcs120 probe and the WCS120 antibody. All closely related species carrying one or more of the genomes of hexaploid wheat produced a 50 kDa protein that was identified by the antibody, and a Wcs120 homoeologue was detected by Southern analysis in all species. In the absence of chromosome arm 6DL in hexaploid CS wheat no 50 kDa protein was produced and the high-intensity Wcs120 band was missing, indicating 6DL as the location of Wcs120 but suggesting silencing of the Wcs120 homoeologue in the A genome. Levels of proteins that cross-reacted with the Wcs120 antibody and degrees of cold tolerance were also investigated in the Chinese Spring/Cheyenne (CS/CNN) chromosome substitution series. CNN chromosome 5A increased the cold tolerance of CS wheat. Densitometry scanning of Western blots to determine protein levels showed that the group 5 chromosome 5A had a regulatory effect on the expression of the Wcs120 gene family located on the group 6 chromosomes of all three hexaploid wheat genomes. The Russian wheat aphid is a significant pest problem in wheat and barley in North America. Genetic resistance in wheat is the most effective and economical means to control the damage caused by the aphid. Dn7 is a rye gene located on chromosome 1RS that confers resistance to the Russian wheat aphid. The gene was previously transferred from rye into a wheat background via a 1RS/1BL translocation. This study was conducted to genetically map Dn7 and to characterize the type of resistance the gene confers. The resistant line '94M370' was crossed with a susceptible wheat cultivar that also contains a pair of 1RS/1BL translocation chromosomes. The F2 progeny from this cross segregated for resistance in a ratio of 3 resistant: 1 susceptible, indicating a single dominant gene. One-hundred and eleven RFLP markers previously mapped on wheat chromosomes 1A, 1B and 1D, barley chromosome 1H and rye chromosome 1R, were used to screen the parents for polymorphism. A genetic map containing six markers linked to Dn7, encompassing 28.2 cM, was constructed. The markers flanking Dn7 were Xbcd1434 and XksuD14, which mapped 1.4 cM and 7.4 cM from Dn7, respectively. Dn7 confers antixenosis, and provides a higher level of resistance than that provided by Dn4. The applications of Dn7 and the linked markers in wheat breeding are discussed.Communicated by J. Using hybrid analysis and test-clone method, 102 accessions of Triticum monococcum L. from the collection of the Vavilov All-Russia Institute of Plant Industry have been studied. This species of wheat has been found to by considerably polymorphic with respect to the resistance to the fungus Erysiphe graminis DC. f. sp. tritici Marchal. causing powdery mildew. The resistance of most accessions to the fungus population and clones is determined by dominant genes. In rare cases, the resistance was determined by recessive genes or one, two, or three oligogenes. A group of einkorn wheat accessions has been found in which the resistance to powdery mildew was determined by the same dominant factor or different but closely linked ones. Recessive resistance genes of T. monococcum differ from the recessive gene pm5 determining the resistance of T. aestivum plants. The genome of T. monococcum contains various genes of resistance to powdery mildew and is a potential source of effective genes to be used when selecting cultivated species of wheat for immunity.  相似文献

Triticum monococcum accession TA2026 showed resistance to wheat powdery mildew. To identify the resistance gene and transfer it to common wheat, genetic analysis and molecular mapping were conducted using an F2 population and derived F3 families from the cross of TA2026 × M389. The results indicated that TA2026 possessed a recessive powdery mildew resistance gene. This gene was mapped to the terminal portion of chromosome 5AmL and flanked by SSR marker loci Xcfd39 and Xgwm126. Eight RFLP markers previously mapped to the terminal chromosome 5AmL were converted into STS markers. Three loci, detected by MAG1491, MAG1493 and MAG1494, the STS markers derived from RFLP probes CDO1312, PSR164 and PSR1201, respectively, were linked to this resistance gene with Xmag1493 only 0.9 cM apart from it. In addition, the STS marker MAG2170 developed from the tentative consensus wheat cDNA encoding the Mlo-like protein identified a locus co-segregating with Xmag1493. This is the first recessive powdery mildew resistance gene identified on chromosome 5Am, and is temporarily designated pm2026. We have successfully transferred it to a tetraploid background, and this resistance stock will now be used as the bridge parent for its transfer to common wheat.  相似文献

Microsatellite markers Xgwmand Xgdmwere used to map the S1, S2, and S3genes of the induced sphaerococcoid mutants of Triticum aestivumL. and to analyze the introgressive lines of common wheat, obtained by crossing several common wheat cultivars to T. timopheeviiZhuk.; these lines carry the Lrgenes of resistance to leaf rust. All sphaerococcoid genes were linked to centromeric markers of the short and long arms of chromosomes of homoeologous group 3 of T. aestivum: the S1locus was located between the markers Xgdm72and Xgwm456; the S2gene, betweenXgwm845and Xgwm566; and the S3was located between Xgwm2and Xgwm720. The introgressive lines of common wheat carry the following substitutions from T. timopheevii, most of 2A and 2B and telomeric region of the 5AL chromosome in the line 821, the same introgression and also the completely substituted chromosome 4B in line 837, and the partially substituted chromosomes 2A and 2B in line 842. The introgression of the genomic material fromT. timopheeviiinto the chromosomes of homoeologous group 2 was the common trait of all three lines resistant to leaf rust. The authors discuss the feasibility of using microsatellite-derived data for analyzing nonmapped wheat species, linking new genes to wheat molecular genetic maps, and analyzing wheat genomes of diverse hybrid origins.  相似文献

The leaf rust resistance gene Lr47 confers resistance to a wide spectrum of leaf rust strains. This gene was recently transferred from chromosome 7 S of Triticum speltoides to chromosome 7 A of hexaploid wheat Triticum aestivum. To facilitate the transfer of Lr47 to commercial varieties, the completely linked restriction fragment length polymorphism (RFLP) locus Xabc465 was converted into a PCR-based marker. Barley clone ABC465 is orthologous to the type-I wheat sucrose synthase gene and primers were designed for the conserved regions between the two sequences. These conserved primers were used to amplify, clone and sequence different alleles from T. speltoides and T. aestivum. This sequence information was then used to identify the T. speltoides sequence, detect allele-specific mutations, and design specific primers. Cosegregation of the PCR product of these primers and the T. speltoides chromosome segment was confirmed in four backcross-populations. To complement this dominant marker, a cleavage amplified polymorphic sequence (CAPS) was developed for the 7 A allele of Xabc465. This CAPS marker is useful to select homozygous Lr47 plants from F2or backcross-F2 segregating populations, and in combination with the T. speltoides-specific primers is expected to facilitate the deployment of Lr47 in new bread wheat varieties. A powdery mildew resistance gene from Triticum urartu Tum. accession UR206 was successfully transferred into hexaploid wheat (Triticum aestivum L.) through crossing and backcrossing. The F1 plants, which had 28 chromosomes and an average of 5.32 bivalents and 17.36 univalents in meiotic pollen mother cells (PMC), were obtained through embryos rescued owing to shriveling of endosperm in hybrid seed of cross Chinese Spring (CS) × UR206. Hybrid seeds were produced through backcrossing F1 with common wheat parents. The derivative lines had normal chromosome numbers and powdery mildew resistance similar to the donor UR206, indicating that the powdery mildew resistance gene originating from T. urartu accession UR206 was successfully transferred and expressed in a hexaploid wheat background. Genetic analysis indicated that a single dominant gene controlled the powdery mildew resistance at the seedling stage. To map and tag the powdery mildew resistance gene, 143 F2 individuals derived from a cross UR206 × UR203 were used to construct a linkage map. The resistant gene was mapped on the chromosome 7AL based on the mapped microsatellite makers. The map spanned 52.1 cM and the order of these microsatellite loci agreed well with the established microsatellite map of chromosome arm 7AL. The resistance gene was flanked by the microsatellite loci Xwmc273 and Xpsp3003, with the genetic distances of 2.2 cM and 3.8 cM, respectively. On the basis of the origin and chromosomal location of the gene, it was temporarily designated PmU.  相似文献
