The genome of a new SV40 strain (SV-IMB) isolated from a rhesus monkey was completely sequenced and compared with other isolates. The results showed that the whole genome contains 5246bp, and the average identity of SV-IMB was 98.1% as compared to other SV40 isolates. Its regulatory region is composed of a complete enhancer and a defective enhancer. Amino acid changes occurred to some extent in both the large T antigen (T-Ag) and VP1 region. The findings demonstrate that the SV-IMB is a new SV40 isolate.  相似文献

Parvoviridae is a family of the smallest viruses known with a wide variety of hosts. The capsid structure of the Aedes albopictus C6/36 cell densovirus (C6/36 DNV) at 1.2-nm resolution was obtained by elec-tron cryomicroscopy (cryoEM) and three-dimensional (3D) image reconstruction. Structure compari-sons between the C6/36 DNV and other parvoviruses reveal that the degree of structural similarity be-tween C6/36 DNV and the human parvovirus B19 is higher than that between C6/36 DNV and other in-sect parvoviruses. The amino acid sequence comparisons of structural and non-structural proteins also reveal higher levels of similarity between C6/36 DNV and parvovirus B19 than those between C6/36 DNV and other parvoviruses. These findings indicate that C6/36 DNV is closely related to the human virus B19, and the former might evolve from the human species other than from other insect viruses.  相似文献

Gene nanos is a maternal posterior group gene required for normal development of abdominal segments and the germ line in Drosophila. Expression of nanos-related genes is associated with the germ line in a broad variety of other taxa. In this study, the 5'-RACE method and the in silico cloning method are used to isolate the new nanos-like gene of Bombyx mor/and the gene obtained is analyzed with bioinformatics tools. The putative protein is expressed in Escherichia coli and the antiserum has been produced in New Zealand white rabbits. The result shows that the nanos cDNA is 1,913 bp in full length and contains a 954 bp open reading frame. The deduced protein has 317 amino acid residues, with a predicted molecular weight of 35 kDa, isoelectric point of 5.38, and contains a conserved nanos RNA binding domain. The conserved region of the deduced protein shares 73% homology with the nanos protein conserved region of Honeybee (Apis mellifera). This gene has been registered in the GenBank under the accession number EF647589. One encoding sequence of the nanos fragment has been successfully expressed in E. coli. Western blotting analysis indicates that homemade antiserum can specifically detect nanos protein expressed in prokaryotic cells.  相似文献

Classification of gene expression data is a pivotal research area that plays a substantial role in diagnosis and prediction of diseases. Generally, feature selection is one of the extensively used techniques in data mining approaches, especially in classification. Gene expression data are usually composed of dozens of samples characterized by thousands of genes. This increases the dimensionality coupled with the existence of irrelevant and redundant features. Accordingly, the selection of informative genes (features) becomes difficult, which badly affects the gene classification accuracy. In this paper, we consider the feature selection for classifying gene expression microarray datasets. The goal is to detect the most possibly cancer-related genes in a distributed manner, which helps in effectively classifying the samples. Initially, the available huge amount of considered features are subdivided and distributed among several processors. Then, a new filter selection method based on a fuzzy inference system is applied to each subset of the dataset. Finally, all the resulted features are ranked, then a wrapper-based selection method is applied. Experimental results showed that our proposed feature selection technique performs better than other techniques since it produces lower time latency and improves classification performance.  相似文献

The struggle for the existence of the biological species is a well-known Prey-Predator model study in the literature.In this study,we present an improved model of Jerri [J.Abdul,Introduction to Integral Equations with Applications,Vol.10 (Wiley,New York,1999)] by introducing the intra-species competition term between the same species in additiuu to the existing environmental changeb and lew other factors in the model.The derntmcl from the exiting (limited) retjources arid other requirements induces competition between the same species which may after the survival tactics among themselves.This intra species term provides strength to tho model ns it makes the model moro realistic.The governing equations are a system of two nonlinear delay integro differential pqimtions,which are solved using spectral collocation method.The role of intra-species coefficients denoting the logistic growth/decay of the two species and two other parameters affecting the population dynamics are analyzed with the three basis functions such as Chebyshev,Legendre and Jacobi polynomials.With the help of simple matrix analysis,the governing equations are converted into a system of nonlinear algebraic equations.Detailed error estiination is computed to compare our results witli the existing inethudb.It is shown with the help of tables and figures that the present method is very efficient,has better accuracy and has least computational cost.  相似文献

GPV is a Chinese serotype isolate of barley yellow dwarf virus (BYDV) that has no reactionwith antiserum of MAV, PAV, SGV, RPV and RMV. The sequence of the coat protein (CP) of GPV isolate of BYDV was identified and its amino acid sequence was deduced. The coding region for the putative GPV CP is 603 bases nucleotides and encodes a Mr 22218 (22 ku) protein. The same as MAV, PAV and RPV, GPV contained a second ORF within the coat protein coding region. This protein of 17024 Mr (17 ku) is thought to correspond to the Virion protein genome linked (Vpg). Sequence comparisons of the CP coding region between the GPV isolate of BYDV and other isolates of BYDV have been done. The nucleotide and ammo acid sequence homology of GPV has a greater identity to the sequence of RPV than those of PAV and MAV. The GPV CP sequence shared 83.7% of nucleotide similarity and 77.5% of deduced amino add similarity, whereas that of the PAV and MAV shared 56.9%. 53.2% and 44.1%. 43.8% respectively. BLCAP is a potential gene for suppression of cervical carcinoma, which was found by analysing the cervical carcinoma specimen with the oncogene and anti-oncogene cDNA microarray. Basing on the bioinformatical analyses, we try to predict the function of blcap gene. The results show that there are several genes that highly resemble with blcap. The comparability between the sequences of blcap and Homo sapiens mRNA (DKFZp564M053) or BC10 is 99% and 87%, respectively. The protein encoded by BLCAP is composed of Leu(19.5%), pro(9.19%), ser(8.04%)、 cys(8.04%) and other amino acids. The secondary structure of the N-terminal of BLCAP encoded protein is an alpha helix. In the C-terminal, it is beta sheet and in the middle, it is coil. The of the terminals is more hydrophobile than the middle region. Between 45-55aa, there is a transmembrane region. Therefore, we forecast the BLCAP is a member of transmembrane protein I. By analyzing the signal peptide and the procedure of blcap gene with the program of SignalP (V1.1), we found a cleavage site in 59-66aa. By using the program of Netpho, we predicted there might be three phospholate sites at 68aa, 73aa and 78aa. At 78-81aa, we found a typical [ST]-X [2] -[DE] structure—the phospholate site of tyrosine protein kinase, which might be related to its function. Bioinformatic studies of blcap provided the foundation for the function researches of BLCAP in laboratory.  相似文献

Although mitochondria provide eukaryotic cells with certain metabolic advantages, in other ways they may be disadvantageous. For example, mitochondria produce reactive oxygen species that damage both nucleocytoplasm and mitochondria, resulting in mutations, diseases, and aging. The relationship of mito-chondria to the cytoplasm is best understood in the context of evolutionary history. Although it is clearthat mitochondria evolved from symbiotic bacteria, the exact nature of the initial symbiosis is a matter of continuing debate. The exchange of nutrients between host and symbiont may have differed from that be-tween the cytoplasm and mitochondria in modern cells. Speculations about the initial relationships includethe following. (1) The pre-mitochondrion may have been an invasive, parasitic bacterium. The host did notbenefit. (2) The relationship was a nutritional syntrophy based upon transfer of organic acids from host tosymbiont. (3) The relationship was a syntrophy based upon H2 transfer from symbiont to host, where thehost was a methanogen. (4) There was a syntrophy based upon reciprocal exchange of sulfur compounds.The last conjecture receives support from our detection in eukaryotic cells of substantial H2S-oxidizing activity in mitochondria, and sulfur-reducing activity in the cytoplasm.  相似文献

The cloning of several plant genes directly involved in triggering a disease resistance response has shown that numerous resistance genes in the nucleotide binding site (NBS)/leucine-rich repeat (LRR) class have similar conserved amino acid sequences. In this study, we used a short soybean DNA sequence, previously cloned based on its conserved NBS, as a probe to identify full-length resistance gene candidates. Two homologous, but genetically independent genes were identified. One gene maps to the soybean molecular linkage group (MLG) F and a second is coded on MLG E. The first gene contains a 3,279 nucleotide open reading frame (ORF) sequence and possesses all the functional motifs characteristic of previously cloned NBS/LRR resistance genes. The N-terminal sequence of the deduced gene product is highly characteristic of other resistance genes in the subgroup of NBS/LRR genes which show homology to the Toll/Interleukin-1 receptor genes. The C-terminal region is somewhat more divergent as seen in other cloned disease resistance genes. This region of the F-linked gene contains an LRR region that is characterized by two alternatively spliced products which produce gene products with either a four-repeat or a ten-repeat LRR. The second cloned gene that maps to soybean MLG E contains 1,565 nucleotides of ORF in the N-terminal domain. Despite strong homology, however, the 3′ region of this gene contains several in-frame stop codons and apparent frame shifts compared to the F-linked gene, suggesting that its functionality as a disease resistance gene is questionable. These two disease resistance gene candidates are shown to be closely related to one another and to the members of the NBS/LRR class of disease resistance genes.
