Citizen science has emerged as an important tool in biology, ecology, and conservation studies. In this paper, we examined YouTube (YT) videos featuring the behaviour of red (Sciurus vulgaris) and grey (Sciurus carolinensis) squirrels in Europe, in both urban and forest habitats. Our study shows that specific behaviours of these squirrels, such as aggression, grooming and calling, have frequently been recorded on YT videos. The present paper uses this open data for the first time to compare differences between the frequency of some types of behaviour in two habitats. Based on detrended correspondence analysis, we show a significant difference in sets of behaviours between two squirrel species. Our investigation show that YT can be a source for monitoring the behaviour of wild species, especially in urban habitats, thus affording insights into the species plasticity of urban individuals. YT, as part of citizen science, is a potential source of information in behavioural ecology. 相似文献

Infection is responsible for approximately 25% of all deaths in patients with systemic lupus erythematosus (SLE), making it a leading cause of mortality among patients. Ruiz-Irastorza and colleagues, in a recent issue of Arthritis Research & Therapy, report the clinical predictors of major infections found in a prospective study of patients with SLE. Similar patterns of infection and pathogens as reported in previous studies were seen; what is striking, however, was the protective effect seen with anti-malarial use. Many infections in patients with SLE could be prevented with timely vaccinations, reducing exposure to contagious contacts, screening for latent infections, minimizing exposure to corticosteroids, targeted prophylaxis for high risk patients, and, unless contraindicated, anti-malarial therapy as standard of care. 相似文献

The European ground squirrel (EGS) (Spermophilus citellus) populations of Vojvodina (Serbia) represent the southernmost part of its distribution in the Pannonian lowland. For species with low dispersal abilities a presence of even weak barriers can have significant influence on genetic structure among adjacent populations. We examined here the effects of habitat fragmentation and river barriers on the genetic structure of the EGS based on 12 microsatellite loci. Bayesian clustering methods were used as additions to classical population genetic approaches. We found that EGS populations in Vojvodina are highly fragmented, but their genetic variation is still higher than in peripheral populations in Central Europe. Populations in Vojvodina consistently grouped into three genetic clusters. The Danube, but not the Tisza River, represents an important barrier to gene flow. EGS populations in the studied area did not show the signs of recent genetic bottlenecks, as would be expected from observations of recent population declines. Conservation strategy should be focused on maintenance of remained suitable habitats and optimal population sizes. 相似文献

Species distribution models (SDMs) are popular in conservation and management of a wide array of taxa. Often parameterized
with coarse GIS-based environmental maps, they perform well in macro-ecological settings but it is debated if the models can
predict distribution within broadly suitable “known” habitats of interest to local managers. We parameterized SDMs with GIS-derived
environmental variables and location data from 82 GPS-collared female red deer (Cervus elaphus) from two study areas in Norway. Candidate GLM models were fitted to address the effect of spatial scale (landscape vs. home
range), sample size, and transferability between study areas, with respect to predictability (AUC) and explained variance
(Generalized R
2 and deviance). The landscape level SDM captured variation in deer distribution well and performed best on all diagnostic
measures of model quality, caused mainly by a trivial effect of avoidance of non-habitat (barren mountains). The home range
level SDMs were far less predictable and explained comparatively little variation in space use. Landscape scale models stabilized
at the low sample size of 5–10 individuals and were highly transferrable between study areas implying a low degree of individual
variation in habitat selection at this scale. It is important to have realistic expectations of SDMs derived from digital
elevation models and coarse habitat maps. They do perform well in highlighting potential habitat on a landscape scale, but
often miss nuances necessary to predict more fine-scaled distribution of wildlife populations. Currently, there seems to be

The human exploitation of land resources (land use) has been considered the major factor responsible for changes in biodiversity within terrestrial ecosystems given that it affects directly the distribution of the fauna. Reptiles are known to be particularly sensitive to habitat change due to their ecological constraints. Here, the impact of land use on reptile diversity was analysed, choosing Catalonia (NE Iberia) as a case study. This region provides a suitable scenario for such a biogeographical study since it harbours: 1) a rich reptile fauna; 2) a highly diverse environment showing strong variation in those variables usually shaping reptile distributions; and 3) good species distribution data. Potential species richness was calculated, using ecological modelling techniques (Ecological Niche Factor Analysis – ENFA). The subtraction of the observed from the potential species richness was the dependent variable in a backwards multiple linear regression, using land use variables. Agriculture was the land use with the strongest relation with the non-fulfilment of the potential species richness, indicating a trend towards a deficit of biodiversity. Deciduous forest was the only land use negatively related with the subtracted species richness. Results indicate a clear relationship between land use and biodiversity at a mesoscale. This finding represents an important baseline for conservation guidelines within the habitat change framework because it has been achieved at the same spatial scale of chorological studies and management policies. 相似文献

Genetic stochasticity due to small population size contributes to population extinction, especially when population fragmentation disrupts gene flow. Estimates of effective population size ( N e ) can therefore be informative about population persistence, but there is a need for an assessment of their consistency and informative relevance. Here we review the body of empirical estimates of N e for wild populations obtained with the temporal genetic method and published since Frankham's (1995 ) review. Theoretical considerations have identified important sources of bias for this analytical approach, and we use empirical data to investigate the extent of these biases. We find that particularly model selection and sampling require more attention in future studies.
We report a median unbiased Ne estimate of 260 (among 83 studies) and find that this median estimate tends to be smaller for populations of conservation concern, which may therefore be more sensitive to genetic stochasticity. Furthermore, we report a median N e / N ratio of 0.14, and find that this ratio may actually be higher for small populations, suggesting changes in biological interactions at low population abundances. We confirm the role of gene flow in countering genetic stochasticity by finding that N e correlates strongest with neutral genetic metrics when populations can be considered isolated. This underlines the importance of gene flow for the estimation of N e , and of population connectivity for conservation in general. Reductions in contemporary gene flow due to ongoing habitat fragmentation will likely increase the prevalence of genetic stochasticity, which should therefore remain a focal point in the conservation of biodiversity. 相似文献

Most species are structured and influenced by processes that either increased or reduced gene flow between populations. However, most population genetic inference methods assume panmixia and reconstruct a history characterized by population size changes. This is potentially problematic as population structure can generate spurious signals of population size change through time. Moreover, when the model assumed for demographic inference is misspecified, genomic data will likely increase the precision of misleading if not meaningless parameters. For instance, if data were generated under an n-island model (characterized by the number of islands and migrants exchanged) inference based on a model of population size change would produce precise estimates of a bottleneck that would be meaningless. In addition, archaeological or climatic events around the bottleneck''s timing might provide a reasonable but potentially misleading scenario. In a context of model uncertainty (panmixia versus structure) genomic data may thus not necessarily lead to improved statistical inference. We consider two haploid genomes and develop a theory that explains why any demographic model with structure will necessarily be interpreted as a series of changes in population size by inference methods ignoring structure. We formalize a parameter, the inverse instantaneous coalescence rate, and show that it is equivalent to a population size only in panmictic models, and is mostly misleading for structured models. We argue that this issue affects all population genetics methods ignoring population structure which may thus infer population size changes that never took place. The aim of this study is to assess anthropometric changes and menstrual cycle characteristics during the maturation process of an adolescent female sample in Madrid, Spain. The new demographic context of Madrid, with a 33.9% of girls coming from Central and South America, makes this study relevant in terms of new epidemiological situations that could possible develop. The sample consists of 284 girls, ages 9 to 16 years, measured and interviewed in four school centres of Madrid. Results show that menarche is slightly earlier in the Spanish girls, but there are no other important differences regarding the characteristics of their menses. However, the Spanish girls have a significantly higher intake of menarcheal pain related drugs. The anthropometric changes accompanying menarche are greater in the immigrants, especially in terms of trunk fatness, leading to an "overweight" characterisation of this sub-sample. These maturational profiles show the need for educational programs, especially focussed on the foreign adolescent population, to cope with health risks related to overweight. 相似文献

Grey partridge populations showed drastic decreasing numbers throughout Europe. Existing knowledge of the causes of decline and the effectiveness of conservation measures was reviewed. Population studies from the UK indicated three periods: a stable population before 1950, sharply decreasing numbers in 1950–1970 and a continued decline after 1970. Other European studies fitted into this picture, with a 10-year lag in each period. The onset of population decrease corresponded with a sharp decrease in chick survival mainly caused by reduced insect availability due to pesticide use. Several factors caused the continued decreasing numbers after 1970, such as decreased hatching success and an increased role of predation. Measures to restore partridge numbers should firstly focus on the main cause of population decline, that is, improve foraging conditions to increase chick survival rate. Next to creation of special partridge habitat, conventional agriculture offers good opportunities to improve foraging conditions. Only when an integrative approach is adopted may large-scale habitat improvements be realised to restore population level to the level before 1950. 相似文献

Variation in DNA repair is a factor in cancer susceptibility: a paradigm for the promises and perils of individual and population risk estimation? 总被引:26,自引:0,他引:26
The repair of DNA damage protects the genome of the cell from the insults of cancer causing agents. This was originally demonstrated in individuals with the rare genetic disease, xeroderma pigmentosum, the prototype of cancer genes, and subsequently in the relationship of mismatch repair to colon cancer. Recent studies suggests that individuals with less dramatic reductions in the capacity to repair DNA damage are observed at polymorphic frequency and these individuals have an increased susceptibility to several types of cancer. Screening of individuals for DNA sequence variation in the exons of 9 DNA repair genes has resulted in identification of 15 different polymorphic amino acid substitution variants. Although the studies to relate these variants to reduced DNA repair capacity and cancer status have not been completed, the available information is sufficient to suggest that DNA repair genes should be incorporated into molecular epidemiology and cancer susceptibility studies. The availability of molecular epidemiology data presents exciting opportunities for refinement of risk estimation models and identification of individuals at increased risk of disease, with resultant opportunities for effective surveillance and early intervention and treatment. The opportunities to acquire susceptibility data are associated with possible perils for establishment of regulations for permissible exposures to carcinogenic agents and also stigmatization of ‘at risk’ individuals that may result in decreased access to employment opportunities and health care. 相似文献

The detailed characterization of the overall free energy landscape associated with the folding process of a protein is the ultimate goal in protein folding studies. Modern experimental techniques provide accurate thermodynamic and kinetic measurements on restricted regions of a protein landscape. Although simplified protein models can access larger regions of the landscape, they are oftentimes built on assumptions and approximations that affect the accuracy of the results. We present a new methodology that allows to combine the complementary strengths of theory and experiment for a more complete characterization of a protein folding landscape. We prove that this new procedure allows a simplified protein model to reproduce remarkably well (correlation coefficient > 0.9) all experimental data available on free energies differences upon single mutations for S6 ribosomal protein and two circular permutants. Our results confirm and quantify the hypothesis, recently formulated on the basis of experimental data, that the folding landscape of protein S6 is strongly affected by an atypical distribution of contact energies. 相似文献

The life history of Daphnia exposed to fish kairomone at different developmental stages was examined in a laboratory experiment. The strongest life history
response to the applied predation threat was observed in females exposed during the 4th instar. Compared to Daphnia experiencing the presence of fish at earlier or later instars, these individuals reached maturity at a smaller size and released
fewer neonates. Moreover, their offspring also demonstrated the strongest reaction to predation threat, exhibiting the broadest
phenotypic plasticity in the life history response to predation. The breadth of their reaction norm was, on average, two times
larger comparing with individuals from other treatments. Broader phenotypic plasticity may offer clear selective advantages
under the unpredictable predation regime. This finding highlights the adaptive role of maternal effect in shaping life history

Mammalian hepatic cytochromes P450 (P450s) are endoplasmic reticulum (ER)-anchored hemoproteins engaged in the metabolism of numerous xeno- and endobiotics. P450s exhibit widely ranging half-lives, utilizing both autophagic-lysosomal (ALD) and ubiquitin-dependent 26S proteasomal (UPD) degradation pathways. Although suicidally inactivated hepatic CYPs 3A and "native" CYP3A4 in Saccharomyces cerevisiae are degraded via UPD, the turnover of native hepatic CYPs 3A in their physiological milieu has not been elucidated. Herein, we characterize the degradation of native, dexamethasone-inducible CYPs 3A in cultured primary rat hepatocytes, using proteasomal (MG-132 and MG-262) and ALD [NH4Cl and 3-methyladenine (3-MA)] inhibitors to examine their specific degradation route. Pulse-chase with immunoprecipitation analyses revealed a basal 52% 35S-CYP3A loss over 6 h, which was stabilized by both proteasomal inhibitors. By contrast, no corresponding CYP3A stabilization was detected with either ALD inhibitor NH4Cl or 3-MA. Furthermore, MG-262-induced CYP3A stabilization was associated with its polyubiquitylation, thereby verifying that native CYPs 3A were also degraded via UPD. To identify the specific participants in this process, cellular proteins were cross-linked in situ with paraformaldehyde (PFA) in cultured hepatocytes. Immunoblotting analyses of CYP3A immunoprecipitates after PFA-cross-linking revealed the presence of p97, a cytosolic AAA ATPase instrumental in the extraction and delivery of ubiquitylated ER proteins for proteasomal degradation. Such native CYP3A-p97 interactions were greatly magnified after CYP3A suicidal inactivation (which accelerates UPD), and/or proteasomal inhibition, and were confirmed by proteomic and confocal immunofluorescence microscopic analyses. We review commonly used population definitions under both the ecological paradigm (which emphasizes demographic cohesion) and the evolutionary paradigm (which emphasizes reproductive cohesion) and find that none are truly operational. We suggest several quantitative criteria that might be used to determine when groups of individuals are different enough to be considered 'populations'. Units for these criteria are migration rate (m) for the ecological paradigm and migrants per generation (Nm) for the evolutionary paradigm. These criteria are then evaluated by applying analytical methods to simulated genetic data for a finite island model. Under the standard parameter set that includes L = 20 High mutation (microsatellite-like) loci and samples of S = 50 individuals from each of n = 4 subpopulations, power to detect departures from panmixia was very high ( approximately 100%; P < 0.001) even with high gene flow (Nm = 25). A new method, comparing the number of correct population assignments with the random expectation, performed as well as a multilocus contingency test and warrants further consideration. Use of Low mutation (allozyme-like) markers reduced power more than did halving S or L. Under the standard parameter set, power to detect restricted gene flow below a certain level X (H(0): Nm < X) can also be high, provided that true Nm < or = 0.5X. Developing the appropriate test criterion, however, requires assumptions about several key parameters that are difficult to estimate in most natural populations. Methods that cluster individuals without using a priori sampling information detected the true number of populations only under conditions of moderate or low gene flow (Nm < or = 5), and power dropped sharply with smaller samples of loci and individuals. A simple algorithm based on a multilocus contingency test of allele frequencies in pairs of samples has high power to detect the true number of populations even with Nm = 25 but requires more rigorous statistical evaluation. The ecological paradigm remains challenging for evaluations using genetic markers, because the transition from demographic dependence to independence occurs in a region of high migration where genetic methods have relatively little power. Some recent theoretical developments and continued advances in computational power provide hope that this situation may change in the future. 相似文献

In mainland Europe, the mosquito species Culex modestus Ficalbi (1890) is a bridge vector for West Nile virus (WNV) from its natural bird-mosquito cycle to mammals. The present study assessed the genetic diversity of Cx. modestus, as well as related Culex species, using the mitochondrial COI DNA barcoding region and compared this with the population structure across Europe. A haplotype network was mapped to determine genealogical relationships among specimens. The intraspecific genetic diversity within individual Culex species was below 2%, whereas the interspecific genetic divergence varied from 2.99% to 13.74%. In total, 76 haplotypes were identified among 198 sequences. A median-joining network determined from 198 COI sequences identified two major lineages that were separated by at least four mutation steps. A high level of intraspecific genetic diversity was not detected in Cx. modestus in samples submitted from different European populations, which indicates that morphologically identified specimens represent a single species and not a species complex. Therefore, it is deduced that different populations of Cx. modestus will show a similar potential to transmit WNV, lending support to concerns that the population present in southeast England represents a risk of transmission to humans. 相似文献

A few studies indirectly support the existence of an intermediate in the transition of Ca(2+)-saturated bovine alpha-lactalbumin (alpha-LA) from the native (N) to the acidic (A) state, known as the molten globule state. However, direct experimental evidence for the appearance of this intermediate has not been obtained. The signal of circular polarization of luminescence (CPL) is sensitive to fine conformational transitions because of its susceptibility to changes in the environmental asymmetry of fluorescent chromophores in their excited electronic states. In the present study, CPL measurements were applied using the intrinsic tryptophan fluorescence of alpha-LA as well as the fluorescence of 8-anilino-1-naphthalenesulfonic acid (ANS) bound to alpha-LA. CPL of tryptophan and ANS was measured in the pH range of 2.5-6 in order to find direct experimental evidence for the proposed intermediate. CPL (characterized by the emission anisotropy factor, g(em)) depends on the asymmetry of the protein molecular structure in the environment of the tryptophan and the ANS chromophores in the excited electronic state. The pH dependence of both the gab, absorption anisotropy factor determined by CD, and the ANS steady state fluorescence, showed a single transition at pH 3-3.7 as already reported elsewhere. This transition was interpreted as being a result of a change of the alpha-LA tertiary structure, which resulted in a loss of asymmetry of the environment of both the tryptophan residues and the ANS hydrophobic binding sites. The pH dependence of the tryptophan and ANS g(em) showed an additional conformational transition at pH 4-5, which coincided with the pKa of Ca2+ dissociation (pKa 5), as predicted by Permyakov et al. (1981, Biochem Biophys Res Commun 100:191-197). The titration curve showed that there is a pH range between 3.7 and 4.1 in which alpha-LA exists in an intermediate state between the N- and A-state. We suggest that the intermediate is the premolten globule state characterized by a reduced Ca2+ binding to the alpha-LA, native-like tertiary structure, and reduced asymmetric fluctuation of the tertiary structure on the nanosecond time scale. This intermediate resembles the "critical activated state" theoretically deduced by Kuwajima et al. (1989, J Mol Biol 206:547-561). The present study demonstrates the power of CPL measurements for the investigation of folding/unfolding transitions in proteins. 相似文献

Age of fish at maturation depends on the species and environmental factors but, in general, investment in growth is prioritized until the first sexual maturity, after which a considerable and increasing proportion of resources are used for reproduction. The present study summarizes for the first the key elements of the maturation of European smelt (Osmerus eperlanus) young of the year (YoY) in the North-eastern Gulf of Riga (the Baltic Sea). Prior to the changes in climatic conditions and collapse of smelt fishery in the 1990s in the Gulf of Riga, smelt attained sexual maturity at the age of 3–4 years. We found a substantial share (22%) of YoY smelt with maturing gonads after the collapse of the smelt fisheries. Maturing individuals had a significantly higher weight, length and condition factor than immature YOY, indicating the importance of individual growth rates in the maturation process. The proportion of maturing YoY individuals increased with fish size. We discuss the factors behind prioritizing reproduction overgrowth in early life and its implications for the smelt population dynamics. 相似文献

Since homology among the fusogenic sequences of the F glycoprotein of paramyxoviruses is high compared with other hydrophobic sequences such as transmembrane sequences and signal peptides, specific binding of this segment to some membrane component was suspected. Thus, temperature-dependent binding of cholesterol to F protein was found. A virus inhibitory peptide (Z-D-Phe-L-Phe-Gly) was found to bind to virus particles also temperature dependently. These dependency are very similar to that of the fusion reaction. Binding of cholesterol and the peptide to the fusogenic sequence of F protein was suggested based on construction of molecular models. Importance of these bindings for understanding the fusion mechanism is discussed. 相似文献
