The functioning of the photosynthetic apparatus during leaf senescence was investigated in alstroemeria cut flowers by a combination of gas-exchange measurements and analysis of in vivo chlorophyll fluorescence. Chlorophyll loss in leaves of alstroemeria cut flowers is delayed by light and by a treatment of the cut flowers with gibberellic acid (GA3). The maximal photosynthesis of the leaves was approximately 6 μmol CO2 m−2 s−1 at I 350 μmol m−2 s−1 (PAR) which is relatively low for intact C3 leaves. Qualitatively the gas-exchange rates followed the decline in chlorophyll content for the various treatments, i.e. light and GA3-treatment delayed the decline in photosynthetic rates. However, when chlorophyll loss could not yet be observed in the leaves, photosynthetic rates were already strongly decreased. In vivo fluorescence measurements revealed that the decrease in CO2 uptake is (partly) due to a decreased electron flow through photosystem II. Furthermore, analysis of the fluorescence data showed a high nonphotochemical quenching under all experimental conditions, indicating that the consumption of reducing power in the Calvin cycle is very low. The chlorophyll, remaining after 9 days incubation of leaves with GA3 in the dark should be considered as a 'cosmetic' pigment without any function in the supply of assimilates to the flowers.  相似文献

The oxygen requirement for stomatal opening in maize plants ( Zea mays L. hybrid INRA 508) was studied at different CO2 concentrations and light intensities. In the absence of CO2, stomatal opening always required O2, but this requirement decreased with increasing light intensity. In darkness, the lowest O2 partial pressure needed to obtain a weak stomatal movement was about 50 Pa. This value was lowered to ca 10 Pa in light (320 μmol m−2 s−1).
On the other hand. in the absence of O2, CO2enabled stomatal opening to occur in the light, presumably due to the evolved photosynthetic O2. Thus, CO2, which generally reduced stomatal aperture, could induce stomatal movement in anoxia and light. The effect of CO2 on stomatal opening was closely dependent on O2 concentration and light intensity. Stomatal aperture appeared CO2-independent at an O2 partial pressure which was dependent on light intensity and was about 25 Pa at 320 umol m−2 s−1.
The presence of a plasmalemma oxidase, in addition to mitochondrial oxidase, might explain the differences in the O2 requirement at various light intensities. The possible involvement of such a system in relation to the effect of CO2 is discussed.  相似文献

Abstract. The uptake and accumulation of inorganic carbon has been investigated in Chlorella ellipsoidea cells grown at acid or alkaline pH. Carbonic anhydrase (CA) was detected in ceil extracts but not in intact cells and CA activity in acid-grown cells was considerably less than that in alkali-grown cells. Both cell types demonstrates low K1/2 (CO2) values in the range pH 7.0–8.0 and these were unaffected by O2 concentration. The CO2 compensation concentrations of acid- and alkali-grown cells suspended in aqueous media were not significantly different in the range of pH 6.0–8.0, but at pH 5.0, the CO2 compensation concentrations of acid-grown cells (57.4cm3 m−3) were lower than those of alkali-grown cells (79.2cm3 m−3). The rate of photo-synthetic O2 evolution in the range pH 7.5–8.0 exceeded the calculated rate of CO2 supply two- to three-fold, in both acid- and alkali-grown cells, indicating that HCO3 was taken up by the cells. Accumulation of inorganic carbon was measured at pH 7.5 by silicone-oil centri-fugation, and the concentration of unfixed inorganic carbon was found to be 5.1 mol m−3 in acid-grown and 6.4mol m−3 in alkali-grown cells. These concentrations were 4.6- and 5.9-fold greater than in the external medium. These results indicate that photorespiration is suppressed in both acid- and alkali-grown cells by an intracellular accumulation of inorganic carbon due, in part, to an active uptake of bicarbonate.  相似文献

Pinus banksiana seedlings were grown for 9 months in enclosures in greenhouses at CO2 concentrations of 350 or 750 μmol mol−1 with either low (0.005 to 0. 3 W m−2) or high (0.25 to 0. 90 W m−2) ultraviolet-B (UV-B) irradiances. Total seedling dry weight decreased with high UV treatment but was unaffected by CO2 enrichment. High UV treatment also shifted biomass partitioning in favor of leaf production. Both CO2 and UV treatments decreased the dark respiration rate and light compensation point. High UV light inhibited photosynthesis at 350 but not at 750 μmol mol−1 CO2 due to a UV induced increase in ribulose-1, 5-bisphosphate carboxylase/oxygenase efficiency and ribulose-1, 5-bisphosphate regeneration. Stomatal density was increased by high UV irradiance but was unchanged by CO2 enrichment.  相似文献

Abstract. Poplar shoots ( Populus euramericana L.) obtained from cuttings were exposed for 6 or 8 weeks to NH3 concentrations of 50 and 100 μgm−3 or filtered air in fumigation chambers. After this exposure the rates of NH3 uptake, transpiration, CO2 assimilation and respiration of leaves were measured using a leaf chamber. During the long-term exposure also modulated chlorophyll fluorescence measurements were carried out to obtain information about the photosynthetic performance of individual leaves. Both fluorescence and leaf chamber measurements showed a higher photosynthetic activity of leaves exposed to 100 μg NH3 m−3. These leaves showed also a larger leaf conductance and a larger uptake rate of NH3 than leaves exposed to 50 μg m−3 NH3 or filtered air. The long-term NH3 exposure did not induce an internal resistance against NH3 transport in the leaf, nor did it affect the leaf cuticle. So, not only at a short time exposure, but also at a long-term exposure NH3 uptake into leaves can be calculated from data on the boundary layer and stomatal resistance for H2O and ambient NH3-concentration. Furthermore, the NH3 exposure had no effect on the relation between CO2-assimilation and stomatal conductance, indicating that NH3 in concentrations up to 100 μg m−3 has no direct effect on stomatal behaviour; for example, by affecting the guard or contiguous cells of the stomata.  相似文献

The effect of long-term exposure to different inorganic carbon, nutrient and light regimes on CAM activity and photosynthetic performance in the submerged aquatic plant, Littorella uniflora (L.) Aschers was investigated. The potential CAM activity of Littorella was highly plastic and was reduced upon exposure to low light intensities (43 μmol m−2 s−1), high CO2 concentrations (5.5 mM, pH 6.0) or low levels of inorganic nutrients, which caused a 25–80% decline in the potential maximum CAM activity relative to the activity in the control experiments (light: 450 μmol m−2 s−1; free CO2: 1.5 mM). The CAM activity was regulated more by light than by CO2, while nutrient levels only affected the activity to a minor extent. The minor effect of low nutrient regimes may be due to a general adaptation of isoetid species to low nutrient levels.
The photosynthetic capacity and CO2 affinity was unaffected or increased by exposure to low CO2, irrespective of nutrient levels. High CO2, low nutrient and low light, however, reduced the capacity by 22–40% and the CO2 affinity by 35-45%, relative to control.
The parallel effect of growth conditions on CAM activity and photosynthetic performance of Littorella suggest that light and dark carbon assimilation are interrelated and constitute an integrated part of the carbon assimilation physiology of the plant. The results are consistent with the hypothesis that CAM is a carbon-conserving mechanism in certain aquatic plants. The investment in the CAM enzyme system is beneficial to the plants during growth at high light and low CO2 conditions.  相似文献

The effects of K+ concentration, light intensity and CO2 levels on the volume of Commelina communis L. guard cell protoplasts were studied. Two degrees of swelling response were observed, both dependent on an external supply of K+, but not necessarily on the supply of a permeant anion. The presence of K+ itself, independent of light or CO2 level, stimulated swelling at a relatively slow rate. When K+, light and low CO2 conditions were supplied together, the swelling was relatively rapid and of high magnitude. The rapid swelling was specific for K+ and Rb+ giving a half maximal effect after 2 h at a KCl concentration of about 18 mmol m−3. The addition of CaCl2 at 1 mol m−3 inhibited K+-dependent swelling under all conditions tested. The response to light and low CO2 levels by Commelina guard cell protoplasts is thought to reflect a high degree of physiological integrity.  相似文献

Seedlings of two tree species from the Atlantic lowlands of Costa Rica, Ochroma la-gopus Swartz, a fast-growing pioneer species, and Pentaclethra macroloba (Willd.) Kuntze, a slower-growing climax species, were grown under enriched atmospheric CO2 in controlled environment chambers. Carbon dioxide concentrations were maintained at 350 and 675 μl 1−1 under photosynthetic photon flux densities of 500 μol m−2 s−1 and temperatures of 26°C day and 20°C night. Total biomass of both species increased significantly in the elevated CO2 treatment; the increase in biomass was greatest for the pioneer species, O. lagopus . Both species had greater leaf areas and specific leaf weights with increased atmospheric CO2. However, the ratio of non-pho-tosynthetic tissue to leaf area also increased in both species leading to decreased leaf area ratios. Plants of both species grown at 675 μl 1−1 CO2 had lower chlorophyll contents and photosynthesis on a leaf area basis than those grown at 350 μl 1−1. Reductions in net photosynthesis occurred despite increased internal CO2 concentrations in the CO2-enriched treatment. Stomatal conductances of both species decreased with CO2-enrichment resulting in significant increases in water use efficiency.  相似文献

Abstract. A field study was conducted to determine the relationship of solar-excited chlorophyll a fluorescence to net CO2 assimilation rate in attached leaves. The Fraunhofer line-depth principle was used to measure fluorescence at 656.3 nm wavelength while leaves remained exposed to full sunlight and normal atmospheric pressures of CO2 and O2. Fluorescence induction kinetics were observed when leaves were exposed to sunlight after 10 min in darkness. Subsequently, fluorescence varied inversely with assimilation rate. In the C4 Zea mays , fluorescence decreased from 2.5 to 0.8 mW m-2 nm-1 as CO2 assimilation rate increased from 1 to 8 μmol m-2 s-1 (r2= 0.520). In the C3 Liquidambar styraciflua and Pinus taeda , fluorescence decreased from 6 to 2 mW m-2 nm-1 as assimilation rate increased from 2 to 5 or 0 to 2 μmol m-2 s-1 (r2= 0.44 and 0.45. respectively). The Fraunhofer line-depth principle enables the simultaneous measurement of solar-excited fluorescence and CO2 assimilation rate in individual leaves, but also at larger scales. Thus, it may contribute significantly to field studies of the relationship of fluorescence to photosynthesis.  相似文献

Abstract. Mass spectrometry has been used to measure the rates of CO2 uptake of acid- and alkali-grown cells of the green algae Chlorella ellipsoidea (UTEX 20) and C. saccharophila (UTEX 27). The time course of CO2 formation on addition of 100mmol m−3 K2CO3 to cells in the dark was used as an assay for external carbonic anhydrase (CA). No external CA was detected in acid-grown cells of either species or in alkali-grown cells of C. ellipsoidea but was present in alkali-grown C. saccharophila . In the absence of external CA, or when it was inhibited by 5mmol m−3 acetazolamide, cells of both species, on illumination, rapidly depleted the free CO2 in the medium at pH 7.5 to near zero concentrations before maximum photosynthetic O2 evolution rates were established. Addition of bovine CA rapidly restored the equilibrium CO2 concentration in the medium, indicating that the cells were selectively taking up CO2. Transfer of cells to the dark caused a rapid increase in the CO2 concentration in the medium largely due to the efflux of inorganic carbon from the cells as CO2. This rapid light-dependent CO2 uptake takes place against pH and concentration gradients and, thus, has the characteristics of active transport.  相似文献

The floating angiosperm Lemna gibba L. was exposed for 2 h to various combinations of photosynthetic photon flux densities and temperature. The extent of photoinhibition of photosynthesis was assayed by measuring the net CO2 uptake before and after a photoinhibitory treatment, and the time course for photoinhibition was studied. It was found that the maximum quantum yield and the light-saturated rate of CO2 uptake were affected by the interaction between light and temperature during the photoinhibitory treatment. At a constant photon flux density of 650 μmol m−2 s−1 the extent of photoinhibition increased with decreasing temperature showing that even a chilling-resistant plant like L. gibba is much more susceptible to photoinhibition at chilling temperatures. About 60% photoinhibition of the quantum yield for CO2 uptake could be obtained either by a high photon flux density of 1 750 μmol m−2 s−1 and 25°C or by a moderate photon flux density of 650 μmol m−2 s−1 and 3°C. The time courses of recovery from 60% photoinhibition produced by either of these two treatments were similar, indicating that the nature of the photoinhibition was intrinsically similar. The extent of photoinhibition was related to the amount of light absorbed in excess to what could be handled by photosynthesis at that temperature. The vital importance of photosynthesis in alleviating photoinhibition is discussed.  相似文献

Elevated CO2 appears to be a significant factor in global warming, which will likely lead to drought conditions in many areas. Few studies have considered the interactive effects of higher CO2, temperature and drought on plant growth and physiology. We grew canola ( Brassica napus cv. 45H72) plants under lower (22/18°C) and higher (28/24°C) temperature regimes in controlled-environment chambers at ambient (370 μmol mol−1) and elevated (740 μmol mol−1) CO2 levels. One half of the plants were watered to field capacity and the other half at wilting point. In three separate experiments, we determined growth, various physiological parameters and content of abscisic acid (ABA), indole-3-acetic acid and ethylene. Drought-stressed plants grown under higher temperature at ambient CO2 had decreased stem height and diameter, leaf number and area, dry matter, leaf area ratio, shoot/root weight ratio, net CO2 assimilation and chlorophyll fluorescence. However, these plants had increased specific leaf weight, leaf weight ratio and chlorophyll concentration. Elevated CO2 generally had the opposite effect, and partially reversed the inhibitory effects of higher temperature and drought on leaf dry weight accumulation. This study showed that higher temperature and drought inhibit many processes but elevated CO2 partially mitigate some adverse effects. As expected, drought stress increased ABA but higher temperature inhibited the ability of plants to produce ABA in response to drought.  相似文献

In vitro shoots of cv. Doyenne ďHiver pear ( Pyrus communis L.) were irradiated under controlled environments for 6 h per day at 5 different levels of biologically effective UV-B radiation (UV-BBE). UV-B exposure caused a progressive increase in apical necrosis above background levels and stimulated leaf abscission. Shoots grown for 2 weeks at 7. 8 mol m−2 day −1 of photosynthetic photon flux (PPF) and treated with 8. 4 or 12. 0 kJ m−2 day −1 UV-BBE produced up to 4 times more ethylene than those given 2. 2 or 5. 1 kJ m−2 day−1 UV-BBE or untreated controls. Exposure of shoots to 12 kJ m−2 day −1 of UV-BBE caused an increase in free putreseine content after 4 to 14 days of irradiation. Shoots showed a decrease in CO2 uptake after 3 days of UV-B: thereafter, they appeared to recover their photosynthetic capacity. Under typical PPF conditions used in micropropagation (90 μmol m−2 S−1). 8. 4 kJ m−2 day −1 of UV-B radiation was injurious to realatively tender tissues of in vitro pear shoots: increasing the level of UV-BBE to 12 kJ m−2 day−1 produced even more adverse effects.  相似文献

Proliferating cultures of Actinidia deliciosa A. Chev., C. F. Liang and A. R. Ferguson cv. Tomuri (♂) were grown under photosynthetic photon flux density (PPFD) rates ranging from 30 to 250 μmol m−2 s−1 in order to determine certain physiological parameters in vitro: CO2 evolution, photosynthesis at three CO2 atmospheric concentrations (330, 1450 and 4500 μl l−1), fresh and dry matter accumulation and proliferation rate.
A proportional response in dry weight, dry/fresh weight ratios and PPFD was found. The proliferation rate increased up to 120 μmol m−2 s−1 but decreased at higher rates. At the highest PPFD, the CO2 released from cultures and accumulated in the vessels reached 200 μl l−1 of; at the lowest rate the CO2 concentration reached 10500 μl l−1 after 28 days of culture. The photosynthetic rate at 1450 and 4500 μl l−1 of CO2 was nearly 4 times higher than at the lowest concentration tested.  相似文献

I considered the possibility that changes in fruit photosynthesis obscure the occurrence of the climacteric rise in respiration in tomato fruits attached to the plant. Internal CO2 and ethylene concentrations in tomatoes ( Lycopersicon esculentum Mill. cv. OH 7814) were analyzed after direct sampling through polyethylene tubes implanted in the external pericarp. Fruits which were shaded with aluminium foil contained up to 60 ml 1−1 CO2, until the internal ethylene concentration exceeded 1 μl l−1, when CO2 concentration declined to below 40 ml l−1; the CO2 concentration in fruits exposed to light only occasionally exceeded 40 ml 1−1. The internal CO2 concentration of detached fruits first declined and then increased along with ethylene concentration, as expected for the climacteric. Detached green fruits under continuous low photosynthetic photon flux density (100 μmol m−2 s−1) contained almost no internal CO2 and produced no CO2. Changes in photosynthesis and an associated CO2-generating system in green fruits are thought to obscure the climacteric rise in tomato fruits developing on the plant.  相似文献

The effects of the ratio of Rubisco activase to Rubisco (activase/Rubisco ratio) on light dependent activation of CO2 assimilation were investigated during leaf aging of rice. Changes of photosynthetic CO2 gas exchange rates in relation to step increases of light intensity from two photon flux densities of 60 µmol m−2 s−1 (low initial PFD) and 500 µmol m−2 s−1 (high initial PFD) to saturated PFD of 1 800 µmol m−2 s−1 were measured. These photosynthetic activation processes were considered to be limited by the Rubisco activation rate when analyzed by the relaxation method. The relaxation time of low initial PFD gradually declined from 3 to 33 days after leaf emergence and showed high and negative correlation to the activase/Rubisco ratio. The initial rate of Rubisco activation under low initial PFD linearly correlated to the amounts of Rubisco activase, whereas these were almost constant from 3 to 23 days after leaf emergence. But these correlations could not be recognized in the case of high initial PFD. Moreover, the relaxation times were more sensitive to intercellular CO2 concentration (Ci) under high initial PFD than under low initial PFD, especially, at Ci below 300 µl l−1. These results suggest the involvement of the activase/Rubisco ratio in the photosynthetic activation under relatively low initial PFD, and the limitation of photosynthetic activation under relatively high initial PFD by Rubisco carbamylation during leaf aging of rice.  相似文献

Abstract: The concentration dependency of the impact of elevated atmospheric CO2 concentrations on Arabidopsis thaliana L. was studied. Plants were exposed to nearly ambient (390), 560, 810, 1240 and 1680 μl I-1 CO2 during the vegetative growth phase for 8 days. Shoot biomass production and dry matter content were increased upon exposure to elevated CO2. Maximal increase in shoot fresh and dry weight was obtained at 560 μl I-1 CU2, which was due to a transient stimulation of the relative growth rate for up to 3 days. The shoot starch content increased with increasing CO2 concentrations up to two-fold at 1680 μl I-1 CO2, whereas the contents of soluble sugars and phenolic compounds were hardly affected by elevated CO2. The chlorophyll and carotenoid contents were not substantially affected at elevated CO2 and the chlorophyll a/b ratio remained unaltered. There was no acclimation of photosynthesis at elevated CO2; the photosynthetic capacity of leaves, which had completely developed at elevated CO2 was similar to that of leaves developed in ambient air. The possible consequences of an elevated atmospheric CO2 concentration to Arabidopsis thaliana in its natural habitat is discussed.  相似文献

Carbon dioxide induces increases in guard cell cytosolic free calcium   总被引:10,自引:0,他引:10
The hypothesis that increases in cytosolic free calcium ([Ca2+]i) are a component of the CO2 signal transduction pathway in stomatal guard cells of Commelina communis has been investigated. This hypothesis was tested using fura-2 fluorescence ratio photometry to measure changes in guard cell [Ca2+]i in response to challenge with 700 µl l−1 CO2. Elevated CO2 induced increases in guard cell [Ca2+]i which were similar to those previously reported in response to abscisic acid. [Ca2+]i returned to resting values following removal of the CO2 and further application of CO2 resulted in a second increase in [Ca2+]i. This demonstrated that the CO2-induced increases in [Ca2+]i were stimulus dependent. Removal of extracellular calcium both prevented the CO2-induced increase in [Ca2+]i and inhibited the associated reduction in stomatal aperture. These data suggest that Ca2+ acts as a second messenger in the CO2 signal transduction pathway and that an increase in [Ca2+]i may be a requirement for the stomatal response to CO2.  相似文献

Calcification in aquatic plants   总被引:1,自引:0,他引:1
Abstract. The CaCO3 deposits of aquatic plants may be intra-, inter- and extracellular. Calcification is mainly the result of photosynthetic CO2 or HCO3 assimilation. This raises the local pH and CO2−3 concentration resulting from shifts in the dissolved inorganic carbon equilibrium, due to either net CO2 depletion as in Halimeda or localized OH efflux (or H+ influx) as in Chara. The plant cell wall may be important in CaCO3 nucleation by acting as an epitaxial substratum or template, or by creating a microenvironment enriched in Ca2+ compared to Mg2+. Hypotheses on the reason for the lack of calcification in many aquatic plants are presented.  相似文献

Responses of apple leaf stomata to environmental factors   总被引:5,自引:4,他引:1
Abstract. Stomatal conductances ( g s) were measured on the leaves of 3–4 year old Golden Delicious trees and of seedlings of two other cultivars. Measurements were made on container grown trees in the field with a diffusion porometer in 1975 and 1976, and in controlled conditions in a leaf chamber in the laboratory in 1976. Stomatal densities in the Golden Delicious leaves were assessed from scanning electron micrographs. Stomatal density on extension shoot leaves was higher than on other leaf types after June.
The response to irradiance shown by both the porometer and the leaf chamber results could be described by a rectangular hyperbola: where g max is maximum conductance and β indicates the sensitivity of gs to photon influx density ( Q p). The values of β were in the range 60–90 μmol m−2 s−1.
There was no evidence that apple stomata are sensitive to temperature per se, but g s was reduced by increasing leaf to air vapour pressure deficits ( D ). There was a linear relationship between g s and D which was not attributable to feed-back to leaf water potential (ψL) as the latter did not affect g s until a threshold of about −2.0 to −2.5 MPa was reached. Conductance generally declined with increasing ambient CO2 concentration.  相似文献
