The influence of resistant and susceptible potato cultivars on Globodera rostochiensis population density changes was studied at different nematode inoculum levels (Pi) in the greenhouse and field. Soil in which one susceptible and two resistant cultivars were grown and fallow soil in pots was infested with cysts to result in densities of 0.04-75 eggs/cm³ soil. A resistant cultivar was grown in an infested field with Pi of 0.7-16.7 eggs/cm³ soil. Pi was positively correlated with decline of soil population densities due to hatch where resistant potatoes were grown in the greenhouse and in the field but not in fallow soil. However, Pi was not correlated with in vitro hatch of G. rostochiensis cysts in water or potato root diffusate. Under continuous culture o f a resistant cultivar, viable eggs per cyst declined 60-90% per plant growth cycle (4 weeks) and the number of cysts containing viable eggs had decreased by 77% after five cycles. The rate of G. rostochiensis reproduction on both resistant and susceptible cultivars was negatively correlated with Pi. These data were used to predict the effect of resistant and susceptible potato cultivars on G. rostochiensis soil population dynamics.  相似文献

A new technique is described for establishing different numbers of the potato cyst-nematode Globodera rostochiensis in field soil, which leaves the soil homogeneous in nutrient status. Field plots established in this way were used to compare yield losses in four potato cultivars (Maris Piper, Pentland Crown, Pentland Dell and Désirée) associated with different numbers of G. rostochiensis. Over the range of 7.4 to 148.4 eggs g-1 soil at planting, yield losses were 18.7% (Maris Piper), 53.2% (Désirée), 55.7% (Pentland Crown) and 63.5% (Pentland Dell). Similar results were obtained in another experiment on the same field in a different year using only lightly and heavily infested plots. Treating the seedbed soil with oxamyl before planting prevented significant injury to potatoes by G. rostochiensis but increased the yield of Pentland Dell and perhaps Désirée (but not Maris Piper or Pentland Crown) more than expected from nematode control alone. Treating heavily infested soil with such a nematicide cannot therefore be recommended as part of a valid procedure for establishing lightly and heavily infested plots for comparing tolerances of attack by potato cyst-nematodes in a range of potato genotypes. In peaty loam soils moderately or heavily infested with G. pallida, oxamyl at 5.6 kg a.i. ha-1 incorporated into the seedbed before potatoes were planted generally increased tuber yields, though the effects varied considerably with the cultivar grown. Increase of G. pallida in these soils was controlled better by growing potatoes bred for resistance to it (ZB 35 – 29, Caxton, Santé, Morag, 11233 ab 22, Fingal, A27/23, Cromwell). Increase of G. pallida on susceptible cultivars varied greatly and Romano increased G. pallida no more than the resistant Morag. G. pallida is probably controlled best in peaty loam by growing a resistant cultivar in soil treated with a granular (non-fumigant) nematicide.  相似文献

Eight trials were conducted in commercial potato fields infested with the white potato cyst nematode (wPCN, Globodera pallida) and one in a field infested with the yellow PCN (yPCN, Globodera rostochiensis). Our aims were to produce data to validate and refine a computer‐based program (The Model) for the long‐term management of PCN, to determine nematicide effectiveness and to assess rates of PCN population decline between potato crops. Prior to planting, each farmer applied an overall nematicide treatment to his field, except for ten untreated plots that were widely spaced to encompass a range of PCN population densities. Each untreated plot was paired with a similar plot in the adjacent treated area and all plots were intensively sampled for PCN population densities at planting (Pi) and again at harvest (Pf) when tuber yields were determined. Four trials were re‐sampled 2–4 years later to determine PCN population decline rates. Regressions that form the basis of ‘The Model’ and described the relationship between Pi and tuber yield and PCN population density at harvest were fitted to the results from both the untreated and nematicide treated plots. These regressions also enabled us to estimate the yield potential at each site in the absence of PCN and showed that nematicide treatment generally did not increase yield potential and that both tuber yield and PCN multiplication decreased with increasing Pi. However, there were major differences between sites and cultivars. When untreated, the yield of cv. Maris Piper was hardly affected in a highly organic soil with Pi > 200 eggs g?1 whereas the yield of partially resistant cv. Santé was decreased from a potential of c. 60 t ha?1 to c. 20 t ha?1 in a light silt with Pi = 20 egg g?1 soil. Similarly, untreated wPCN multiplication rates at a low Pi ranged from 46‐fold to >100‐fold. Nematicide effectiveness was estimated from the regressions and, at several sites, yield was decreased despite nematicide treatment. Control of wPCN multiplication was even poorer. In only two of seven trials planted with susceptible cultivars was more than 50% control achieved – maximum populations in treated plots usually exceeded 250 eggs g?1. Partially resistant Santé decreased the multiplication rate of wPCN in the two trials where it was planted. An alternative analysis using Genstat indicated that The Model tended to underestimate the maximum multiplication rate and overestimate the maximum population density. When four sites were re‐sampled 2–4 years after harvest the populations of wPCN had declined by between 15% and 33.5% per annum with a mean of 26% per annum. Modelling indicated that rotations longer than 8 years were required to control wPCN unless other effective control measures, such as growing a partially resistant cultivar, were used.  相似文献

Cropping systems in which resistant potato cultivars were grown at different frequencies in rotation with susceptible cultivars and a nonhost (oats) were evaluated at four initial nematode population densities (Pi) for their ability to maintain Globodera rostochiensis at a target level of <0.2 egg/cm³ of soil. At a Pi of 0.1 to 1 egg/cm³ of soil, cropping systems with 2 successive years of a resistant cultivar every 3 years of potato production reduced and maintained G. rostochiensis at <0.2 egg/cm³ of soil. At a Pi of 1 to 4 eggs/cm³ of soil, 2 successive years of a resistant cultivar followed by 1 year of oats for every 4 years of production were necessary to reduce and maintain G. rostochiensis populations at <0.2 egg/cm³ of soil. At a Pi greater than 4 eggs/cm³ of soil, 2 successive years of a resistant cultivar plus 1 year of oats reduced G. rostochiensis densities to <0.2 egg/cm³ of soil, but the population increased above that density after cropping 1 year to a susceptible cultivar. The numbers of cysts and eggs per cyst in the final population (Pf) of G. rostochiensis were influenced by initial density and the frequency of growing a susceptible cultivar in a cropping system. The lowest number of cysts and eggs per cyst in the final G. rostochiensis population occurred with a cropping system consisting of 2 successive years of a resistant cultivar followed by oats with a susceptible cultivar grown the fourth year of production.  相似文献

The effects of a root-knot nematode-resistant tomato cultivar and application of the nematicide ethoprop on root-knot nematode injury to cucumber were compared in a tomato-cucumber double-cropping system. A root-knot nematode-resistant tomato cultivar, Celebrity, and a susceptible cultivar, Heatwave, were grown in rotation with cucumber in 1995 and 1996. Celebrity suppressed populations of Meloidogyne incognita in the soil and resulted in a low root-gall rating on the subsequent cucumber crop. Nematode population densities were significantly lower at the termination of the cucumber crop in plots following Celebrity than in plots following Heatwave. Premium and marketable yields of cucumbers were higher in plots following Celebrity than in plots following Heatwave. Application of ethoprop through drip irrigation at 4.6 kg a.i./ha reduced root galling on the cucumber crop but had no effect on the nematode population density in the soil at crop termination. Ethoprop did not affect cucumber yield. These results indicate that planting a resistant tomato cultivar in a tomato-cucumber double-cropping system is more effective than applying ethoprop for managing M. incognita.  相似文献

The population dynamics of Globodera pallida were studied on susceptible potato plants. Oxamyl treated and untreated plots were planted with a G. pallida susceptible crop, from the first early maturity class, under polythene at the end of February and immediately followed, after harvest in June, by a susceptible second crop. Other nematicide treated and untreated plots were sown in April with a G. pallida susceptible cultivar. The results were variable but showed that reproduction of G. pallida under double cropping conditions was greater than on a single main crop.  相似文献

The fecundity of Globodera rostochiensis (R₁A) females that developed on resistant Rosa and susceptible Katahdin potato cultivars were compared. Cysts collected from each cultivar were bulked, separated into four sizes (> 500 μm, 355-500 μm, 250-355 μm, and < 250 μm), and crushed to determine fecundity as measured by viable egg content (VEC). Fewer and generally smaller cysts developed on Rosa than on Katahdin. Although cyst size significantly (P = 0.01) influenced VEC, cyst age (8 or 13 weeks) had no effect. Regardless of size, cysts produced on Rosa contained significantly fewer viable eggs than did cysts produced on Katahdin. The fecundity of progeny from cysts produced on Rosa was significantly reduced compared with that of progeny from cysts produced on Katahdin. After two generations on Katahdin, the VEC of cysts from a population originating from Rosa was significantly less than that of cysts from a population originating from Katahdin, indicating that in the presence of a pure population of G. rostochiensis R₁A, the females that develop on the resistant cultivar Rosa represent a diminished rather than a superior selected population.  相似文献

In field trials Cara, Brio, Maris Piper and Pentland Javelin were consistently more tolerant of damage by Globodera rostochiensis yielding more than Corsair, Pentland Dell, Maris Anchor and Maris Peer, in untreated, heavily infested soil and giving the smallest increases to nematicide treatment. No yield or growth differences were found between plants in untreated and nematicide treated plots at a nematode-free site. The most tolerant cultivars all had a gene (H1) for resistance to G. rostochiensis derived from Solanum tuberosum ssp. andigena and in soil infested with G. pallida the tolerance of at least one resistant cultivar (Maris Piper) appeared to be lessened. However, some resistant cultivars were comparatively intolerant, even to G. rostochiensis. Early cultivars were generally less tolerant than late maturing cultivars but there were exceptions. Amongst cultivars with resistance derived from Solanum vernei the early cultivar Guardian was more tolerant than the main crop cultivar Corsair. The effect on the yield of several cultivars of a range of densities of G. rostochiensis, produced either by applying different rates of a nematicide or by cropping in the previous year, was examined at two sites. The results indicated that the slope of the regression for yield in relation to nematode density was less for tolerant than intolerant cultivars. At sites infested with G. rostochiensis Maris Piper was found to be consistently more tolerant than Pentland Crown.  相似文献

Heterodera glycines is a cyst nematode that causes significant lost soybean yield in the U.S. Recent studies observed the aphid Aphis glycines and H. glycines interacting via their shared host, soybean, Glycine max. A greenhouse experiment was conducted to discern the effect of A. glycines feeding on H. glycines reproduction. An H. glycines-susceptible cultivar, Kenwood 94, and a resistant cultivar, Dekalb 27–52, were grown in H. glycines-infested soil for 30 and 60 d. Ten days after planting, plants were infested with either zero, five, or ten aphids. At 30 and 60 d, the number of H. glycines females and cysts (dead females) and the number of eggs within were counted. In general, H. glycines were less abundant on the resistant than the susceptible cultivar, and H. glycines abundance increased from 30 to 60 d. At 30 d, 33% more H. glycines females and eggs were produced on the resistant cultivar in the ten-aphid treatment compared to the zero-aphid treatment. However, at 30 d the susceptible cultivar had 50% fewer H. glycines females and eggs when infested with ten aphids. At 60 d, numbers of H. glycines females and cysts and numbers of eggs on the resistant cultivar were unaffected by A. glycines feeding, while numbers of both were decreased by A. glycines on the susceptible cultivar. These results indicate that A. glycines feeding improves the quality of soybean as a host for H. glycines, but at higher herbivore population densities, this effect is offset by a decrease in resource quantity.  相似文献

The potato cyst nematodes (PCNs) Globodera pallida and Globodera rostochiensis are important parasites of potato. PCNs undergo complex biotrophic interactions with their hosts that involve gene expression changes in both the nematode and the host plant. The aim of this study was to determine key genes that are differentially expressed in Globodera pallida life cycle stages and during the initiation of the feeding site in susceptible and partially resistant potato genotypes. For this purpose, two microarray experiments were designed: (i) a comparison of eggs, infective second‐stage juveniles (J2s) and sedentary parasitic‐stage J2s (SJ2); (ii) a comparison of SJ2s at 8 days after inoculation (DAI) in the susceptible cultivar (Desirée) and two partially resistant lines. The results showed differential expression of G. pallida genes during the stages studied, including previously characterized effectors. In addition, a large number of genes changed their expression between SJ2s in the susceptible cultivar and those infecting partially resistant lines; the number of genes with modified expression was lower when the two partially resistant lines were compared. Moreover, a histopathological study was performed at several time points (7, 14 and 30 DAI) and showed the similarities between both partially resistant lines with a delay and degeneration in the formation of the syncytia in comparison with the susceptible cultivar. Females at 30 DAI in partially resistant lines showed a delay in their development in comparison with those in the susceptible cultivar.  相似文献

Three clones of potatoes bred from Solanum vernei were tested for their resistance to the white potato cyst-nematode, Heterodera pallida, both by root-ball counts and by estimates of total cysts produced. Two susceptible clones were used as controls. Root-ball counts suggested a higher degree of resistance than that based on total cysts produced. There were differences in pathogenicity between the two populations. The degree of resistance found was such that in practice the resistant clones would be best used in conjunction with nematicide treatment in a system of integrated control.  相似文献

Six potato cultivars with different levels of resistance to the white potato cyst nematode (PCN) Globodera pallida Pa2 were grown for three seasons in field plots to which G. pallida Pa2 cysts had been introduced earlier. There were two planting times, corresponding to early and maincrop commercial planting times, and two initial PCN population densities, high and low. The effect of cultivar on PCN population density was far greater than the effect of planting time or initial nematode population. The final PCN populations for the cultivars Ilam Hardy, Wha, 4696A(2), Sovereign, D40/6 and V390 were 151, 74, 27, 1.4, 0.2 and 0.06 eggs per g of soil respectively. It is concluded that resistant potato cultivars can be very effective in controlling G. pallida Pa2 in the field.  相似文献

Globodera rostochiensis population densities and potato root growth were measured in field plots of one susceptible and two resistant potato cultivars. Root growth and nematode densities were estimated from soil samples taken at three depths between plants within the rows, three depths 22.5 cm from the rows, and at two depths midway between rows (furrows). Four weeks after plant emergence (AE), nematode densities in the rows had declined 68% in plots of the susceptible cultivar and up to 75% in plots of both resistant cultivars. Significant decline in nematode densities in the furrows 4 weeks AE occurred only in plots of the susceptible cultivar. Total decline in nematode density in fallow soil was 50%, whereas in plots of the resistant cultivars, decline was more than 70% in the rows and more than 50% in the furrows. Nematode densities increased in the rows of the susceptible cultivar but declined in the furrows. We conclude that G. rostochiensis decline or increase is correlated with host resistance and the amount of roots present at any particular site.  相似文献

Clones of two partially resistant and two susceptible white clover, Trifolium repens, genotypes were exposed to eggs of Heterodera trifolii and nematode development in stained roots measured at 2, 4, 7, 11, 18, 23, and 37 days after inoculation. The differences in development between nematode populations in resistant and susceptible genotypes showed that resistance operated after infection during feeding and development. At 7 days after inoculation, counts of second-stage juveniles did not differ between genotypes, whereas at 37 days more adults had developed in the susceptible than in the resistant genotypes. In a separate experiment, cysts hosted by susceptible genotypes were larger and contained more eggs than those on resistant genotypes so that the product of the values for cysts per plant and for eggs per cyst resulted in a more sensitive measure of resistance than from using cysts per plant alone.  相似文献

Populations ofHelicoverpa (=Heliothis) zea (Boddie),Heliothis virescens (F.),Manduca sexta (L.) andM. quinquemaculata (Haw.) and their egg and larval parasitoids were sampled in field plots of the: insect-resistant wild tomato,Lycopersicon hirsutum f. glabratum C. H. Mull, accession PI 134417; susceptible commercial tomato cultivar ‘Better Boy’; F1 hybrid; and selected, moderately resistant backcross genotype. Densities ofH. zea andH. virescens eggs and small larvae were higher on resistant genotypes than on susceptible genotypes, but densities of large larvae were similar on all genotypes. Densities ofManduca spp. larvae were too low to permit similar analyses of the effects of plant genotype. Rates of egg parasitism byTrichogramma spp. andTelenomus sphingis (Ashmead) were reduced on insect-resistant genotypes. Rates of parasitism by the larval parasitoidsCampoletis sonorensis (Cameron) andCotesia congregata (Say) were reduced on resistant genotypes. No consistent effects on parasitism rates byCotesia marginiventris (Cresson) were observed and parasitism rates byCardiochiles nigriceps Viereck were unaffected.  相似文献

The increase of 35 English field populations of potato cyst-nematodes (Globodera rostochiensis and/or G. pallida) was measured on Désirée, Maris Piper, Caxton (A25/11), Cromwell (A27/20) and clone 11233 ab 22 in pots of sandy, silty or peaty loam soil. Désirée was susceptible to all populations tested and, as in field soils, the final population (Pf) was inversely related to the initial population of potato cyst-nematode eggs (Pi) in the soil. Maris Piper and Cromwell were resistant to all G. rostochiensis populations, with one possible exception. Maris Piper was susceptible to all G. pallida populations. Caxton was susceptible to some and fairly resistant to other populations of G. rostochiensis, indicating the existence either of two biotypes within the one pathotype (Rol) as yet encountered in Britain, or the existence of an additional pathotype. Caxton and Cromwell were fairly resistant to G. pallida. Clone 11233 ab 22 was only moderately resistant to both species. Resistance to potato cyst-nematode increase varied considerably, especially in Caxton (to G. rostochiensis) and in 11233 ab 22 (to both species). Oxamyl greatly reduced the increase of G. rostochiensis populations on Désirée potatoes, with the notable exception of one population but it generally had much less effect on G. pallida populations, regardless of soil type. The difference in effect on the two species may be due to a longer period of hatching in G. pallida than in G. rostochiensis and also perhaps to a second generation in G. pallida.  相似文献

Six cultivars of potato (Santé, Morag, Paladin, Glenna and Fingal bred for resistance to both potato cyst-nematodes (Globodera rostochiensis and G. pallida) and Valiant bred for resistance to G. pallida alone) were exposed to 28 English populations of G. pallida and eight English populations of G. rostochiensis in pots. Susceptible cv. Désirée potatoes served as controls for all 36 populations. Inoculum (Pi) was 12000 eggs in cysts per 400ml pot of soil. Average increase of G. rostochiensis (Pf/Pi) on cv. Désirée was 23.5 but on cvs Sante, Glenna and Fingal it was < 1.0 and on cv. Morag it was 2.2. In contrast, cvs Paladin and Valiant were susceptible (average Pf/Pi = 17.4 and 26.5, respectively). Against G. pallida populations, average Pf/Pi for cv. Désirée was 21.7; on cvs Paladin, Santé and Glenna it was 2.9, 2.6 and 2.4, respectively; cvs Morag and Fingal were less resistant (7.4 and 5.6, respectively) and cv. Valiant was quite susceptible (11.0). Resistance to the different populations of G. pallida and G. rostochiensis varied but for the most resistant cultivars (Santé, Glenna and Paladin) the variation was usually small. The value of the six resistant cultivars studied to the integrated control of potato cyst-nematodes in England and the genetic diversity of the nematode populations to which they were exposed are discussed.  相似文献

Cotton plants resistant to Meloidogyne incognita had roots characterized by fewer and smaller galls, and females that produced fewer egg masses containing fewer eggs than did susceptible plants. Many galls on resistant roots contained no nematodes at the time of examination. Penetration of the resistant cultivar was equal to that of the susceptible cultivar and independent of the number of nematodes in the inoculum. Fewer nematodes penetrated resistant or susceptible plants with eight leaves than those with fewer leaves. Reciprocal grafts of resistant and susceptible plants failed to confer resistance or susceptibility to the rootstock.  相似文献

The two species of the potato cyst nematodes (PCN) Globodera pallida and G rostochiensis are the most problematic pests of the potato crop in the UK. There are no commercially available cultivars with full resistance to G. pallida and both crop rotation and granular nematicides are less effective at controlling this species than G. rostochiensis. In situations of very high PCN levels it may be possible to reduce populations and yield losses by using an autumn application of the soil fumigant 1,3-dichloropropene (1,3-D) followed by a spring application of a granular nematicide. Two field experiments were done to look at the integration of methods for the control of PCN. The Common Field experiment (G. rostochiensis infested) compared the use of 1,3-D with the granular nematicides aldicarb, oxamyl and fosthiazate when growing the susceptible cv. Estima. The Four Gates experiment (infested with both PCN species but mainly G rostochiensis) compared the performance of cv. Santé (partially resistant to G. pallida, fully resistant to G. rostochiensis) with that of the susceptible cv. Estima when treated with 1,3-D and oxamyl at full and half-rates. The results of the experiments show that an integrated approach to nematode control on heavily infested sites, including granular and fumigant nematicides and cultivar resistance, can lead to significant decreases in nematode population densities and reduce yield losses. An economic evaluation of the experiments modelled the gross margins from the different nematicide treatments. In Common Field, the highest gross margins were achieved with the combined use of fumigant and granular nematicides. In Four Gates, there was a clear economic benefit for both cultivars from the use of 1,3-D. In this experiment, oxamyl was of economic value to Estima but not to Sante and full-rate oxamyl was of more benefit than half-rate to Estima.  相似文献

The assessment of larval emergence from cysts, second stage larval behaviour in soil, invasion of roots, development within roots and the final population provides a technique which can be used for evaluating the efficacy of any non-volatile organophosphate or carbamate nematicide.
Oxamyl delayed emergence from cysts, disorientated second stage larvae in soil, delayed and reduced invasion of roots, reduced the number of eggs in the final population and markedly reduced the final population of cysts.  相似文献
