脑中风是严重危害我国中老年人身体健康的主要疾病之一,对中风相关基因单核苷酸多态性位点在人群中分布情况的研究将有助于深入认识中风发病机制及提高防治能力。该研究用寡核苷酸芯片方法检测了8个多态性位点与中风易感性之间的关联。结果发现:寡核苷酸芯片技术作为一种很好的基因分型方法,具有高通量、平行性、微量化、自动化和快速灵敏等的特点,适用于大规模基因分型平台的建立;实验初步发现与中风易感性相关的有血管紧张素原(AGT)基因M235T(OR=3.437,CI=1.211,9.709),载脂蛋白E(APOE)基因cys158arg (OR=9.434,1.686-83.3),亚甲基四氢叶酸还原酶(MTHFR)基因C677T(OR=2.591,1.002-6.711),血管紧张素ⅡⅠ型受体(AT1R)基因A1166C(OR=0.213,0.057-0.799)。  相似文献

以欧洲黑杨的雄花枝为材料,采用温室切枝水培和醋酸洋红染色制片法,观察花粉母细胞减数分裂进程及其与花芽发育外部形态的关系。结果表明:(1)欧洲黑杨花粉母细胞的减数分裂进程快慢与温度高低关系密切;(2)减数分裂发生时其花芽的外部形态发育没有明显变化;(3)花粉母细胞减数分裂不同步性突出,同一花药内的花粉母细胞处于不同的减数分裂时期,且同一花芽尖端发育明显滞后于中部和基部;(4)减数分裂过程中有少量的核仁异常现象,虽观察到较多平行纺锤体现象,但未发现天然2n花粉。该研究结果揭示了欧洲黑杨减数分裂的遗传变异规律,为诱变育种、倍性育种以及分子水平的进一步研究奠定了细胞学基础。  相似文献

利用PCR检测了转基因欧洲黑杨当代和杂交后代群体 ,外源Bt基因在 7年生的转基因欧洲黑杨 1 4个无性系中稳定存在 ,Bt基因在杂交后代中分离比例为 1∶1。转基因欧洲黑杨作母本进行杂交时有落花、落果现象 ,可能是基因改造影响了它们的某些生理功能。  相似文献

为获得林木植物核基质附着区(MARs)序列,诱导欧洲黑杨(Populus nigra)愈伤组织并制备悬浮细胞,裂解释放细胞核,除去非MARs的游离DNA片段,得到核基质。碱性缓冲液分离残留DNA与结合蛋白,回收并克隆残留DNA,测序后得到两个具有MARs序列特征的DNA片段A7和A23。序列特征分析结果表明它们都含有MARs的特征基元,通过与其他已发表的MARs序列比较,认为是从杨树中分离到的两个MARs片段,并预测了其功能。该研究是首次从木本植物获得MARs序列。  相似文献

利用RT-PCR技术研究了美洲黑杨(Populus deltoides)材性相关候选基因细胞分裂素结合蛋白(cytokininbin ding protein)基因PdCYTOB的表达谱,结果显示,在未成熟木质部、未成熟韧皮部和韧皮部中PdCYTOB基因具有较高水平的表达量。对导入反义PdCYTOB的山新杨(Populus davidiana×P.bolleana)植株进行Southern杂交和RT-PCR检测,证实反义PdCYTOB基因已整合到杨树基因组中并表达。对大田转基因株系及对照植株进行表型观察、组织切片和微纤丝角的测定,结果表明,转反义PdCYTOB基因植株的高度明显增加,木质部、韧皮部变宽,微纤丝角明显变小,初步表明转基因杨树在造纸性能上有所改良。这些研究结果对于阐明PdCYTOB在美洲黑杨木材形成中的分子作用机制具有重要的理论意义。  相似文献

目的:探索老年冠心病多基因遗传易感性基础及相关的危险因素。方法:采用病例-对照研究方法,共入选老年汉族冠心病患者246例,非冠心病患者185例,纳入性别、年龄、吸烟,饮酒,高血压史、糖尿病史、高脂血症史、同型半胱氨酸、氨基末端脑钠肽前体、超敏C反应蛋白、抗凝血酶III、胆固醇、甘油三酯、高密度脂蛋白、低密度脂蛋白共15种危险因素与冠心病的关联性进行logistic回归分析。同时使用美国Sequenom高通量基因多态性分型技术研究了10种基因11个单核苷酸基因多态性(SNP)位点与冠心病的关联性。结果:15种危险因素中,发现增龄、高血压、抗凝血酶III(ATIII)下降是冠心病主要的危险因素,P<0.05。11个SNPs中3个SNP,血小板糖蛋白GP1BA rs2243093(-5T/C),血管紧张素转化酶ACE rs4332(547C/T)与ATIII rs2227589(893C/T)与老年汉族患者冠心病相关联。rs2243093(-5T/C)突变基因型CC与TT+AT比较,P=0.029(OR=3.41,CI:1.19-9.75);rs4332(547C/T)杂合型TC与CC+TT相比,P=0.003(OR=0.56,CI:0.38-0.82);rs2227589(893C/T),CT+CT与野生基因型CC相比较,P=0.003(OR=1.79,CI:1.22-2.63)。结论:增龄、抗凝血酶III下降、高血压是影响老年冠心病的主要危险因素,血小板、抗凝血系统、肾素-血管紧张素系统三种机制参与了老年冠心病的发生与发展。  相似文献

目的从单核苷酸多态性(single nucleotide polymorphism,SNP)水平探索小鼠生化标记基因Hbb多态性的形成机理。方法从DNA、RNA和蛋白多肽3个方面分析研究Hbb基因的多态性。结果基因组DNA中有4个SNP位点与Hbbd/s多态性相关:分别为外显子1中的2个T(G),外显子2中的G(A)和外显子3中的A(G);RNA水平有4个SNP位点与Hbbd/s多态性相关:分别对应为外显子1中的2个T(G),外显子2中的G(A)和外显子3中的A(G);蛋白多肽水平第13、20和139位氨基酸残基,即Cys/Gly、Ser/Ala和Thr/Ala间的转换与Hbb/s多态性相关,分别对应于外显子1中的2个T(G)和外显子3中的A(G)。结论第13、20和139位氨基酸残基,即Cys/Gly、Ser/Ala和Thr/Ala间的转换可能是Hbbd/s多态性的形成原因。  相似文献

目的:探讨单羧酸转运蛋白基因(monocarboxylate transporter,MCT)单核苷酸多态性(single nucleotide polymorphism,SNPs)与肝细胞肝癌(hepatocellular carcinoma,HCC)根治术患者预后的关系。方法:运用Sequenom i PLEX分型技术对830例原发性HCC患者MCT家族(MCT1、MCT2和MCT4)基因上的8个功能性SNP位点进行基因分型,并分析这些SNP与HCC患者预后的相关性。结果:MCT1基因rs1049434位点和MCT2基因rs995343位点基因型与HCC患者总体生存期及无复发生存期均显著相关(P0.05)。携带MCT1 AT基因型或TT基因型的患者死亡及复发风险均显著低于携带AA基因型的患者(HR=0.72;P=0.042或HR=0.64;P=0.002);携带MCT2 CT基因型或TT基因型的患者死亡及复发风险均显著高于携带CC基因型的患者(HR=1.64;P=0.018或HR=1.52;P=0.026)。而且,MCT1基因rs1049434位点和MCT2基因rs995343位点对HCC预后存在显著的累积效应,携带2个危险基因型的患者死亡及复发风险分别是没有危险基因型患者的2.16倍和2.54倍。此外,携带2个危险基因型的HCC患者在术后行TACE辅助治疗后死亡及复发风险均显著降低(P0.05)。结论:MCT1和MCT2基因上的功能性SNP位点有可能作为HCC根治术后预后评估和TACE辅助治疗反应预测的独立标志物。  相似文献

LALBA基因SNP与内蒙古白绒山羊经济性状的关联   总被引:1,自引:0,他引:1  
利用PCR-SSCP和DNA测序技术检测452份内蒙古白绒山羊α-乳白蛋白(LALBA)基因单核苷酸多态性(SNP), 并分析SNP与产绒量、绒厚、绒长和体重性状的关联。结果表明, 仅P2引物位点存在SSCP多态, 其外显子3区域存在1个突变位点: M63868:g.1897T>C。内蒙古白绒山羊群体LALBA基因M63868:g.1897位点以TT型为主, T等位基因频率为0.983, 且处于Hardy-Weinberg平衡状态(P>0.05)。方差分析表明, LALBA基因M63868:g.1897位点多态仅与产绒量存在显著相关(P=0.017); 1897位点TC基因型个体产绒量比TT基因型个体多产绒142.68 g, 高26.21%, 且差异显著(P<0.05)。因此, TC基因型可作为山羊产绒性状标记辅助选择的有效DNA标记。  相似文献

目前有很多证据证明RELN基因在世界人群中是一个精神分裂症的致病基因.基于之前报道过的RELN基因在精神分裂症患者中表达下降的事实,可以推测在RELN基因启动子区可能包含影响精神分裂症发生的多态位点.该研究分析了中国西南地区病例——对照人群中(940位患者和1369位正常人)RELN基因启动子区的3个单核苷酸多态性位点与精神分裂症的相关性.研究结果显示,这些多态位点都不与精神分裂症相关,表明RELN基因的致病位点并不在其启动子区.将前人研究结果与该研究结果进行综合分析(共2843个样本),结果仍不显著.因此,该研究表明,RELN基因启动子区的单核苷酸多态性位点在中国人群中并不与精神分裂症相关.  相似文献

Black poplar (Populus nigra L.) is a tree of ecological and economic interest. A better knowledge of P. nigra genome is needed for an effective protection and use of its genetic resources. The main objective of this study is the construction of a highly informative genetic map of P. nigra species including genes of adaptive and economic interest. Two genotypes originated from contrasted natural Italian populations were crossed to generate a F1 mapping pedigree of 165 individuals. Amplification fragment length polymorphism (AFLP), simple sequence repeat (SSR), and single nucleotide polymorphism (SNP) markers were used to genotype 92 F1 individuals, and the pseudo-test-cross strategy was applied for linkage analysis. The female parent map included 368 markers (274 AFLPs, 91 SSRs, and 3 SNPs) and spanned 2,104 cM with 20 linkage groups, and the male parent map, including 317 markers (205 AFLPs, 106 SSRs, 5 SNPs, and sex trait), spanned 2,453 cM with 23 main linkage groups. The sex, as morphological trait, was mapped on the linkage group XIX of the male parent map. The generated maps are among the most informative in SSRs when compared to the Populus maps published so far and allow a complete alignment with the 19 haploid chromosomes of Populus sequence genome. These genetic maps provide informative tools for a better understanding of P. nigra genome structure and genetic improvement of this ecologically and economically important European tree species. This study describes growth performance and productivity of poplars (Populus nigra L.) on a 5-year rotation in two soil types. Two adjacent and even-aged stands of poplar (PS1 and PS2) with different productivity were selected in poplar plantations of Tehran, Iran. On a 5-year rotation, growth parameters of poplar trees were measured and productivity was computed. A transect sampling method with 10% intensity was used for measurement of the growth parameters. A composite soil sample was taken for all transects, and soil physico-chemical properties were determined. Texture, EC, organic matter percent, the contents of some macro and micro nutrients and some heavy metal concentrations of soil had significant differences between two stands; as a consequence growth performance and productivity of poplars were significantly difference between PS1 and PS2. In PS1, productivity of 5-year trees was 2.5 times higher than in PS2. Soil with Clay Loam texture, more contents of organic matter and nutrients, lower EC and lower contents of Ca, Mg, S and heavy metals provided more suitable conditions for growth of poplars. The growth parameters were positively correlated with the contents of nutrients, clay and organic matter in the soil, but they were negatively related to EC and the contents of silt, Ca, Mg, S and heavy metals of soil. Careful selecting sites based on soil properties before planting can be useful for increasing productivity of poplars.  相似文献

In this study six simple sequence repeats (SSR or microsatellites) were selected for their ability to fingerprint a total of 60 commercial clones of Populus deltoides Marsh. and Populus x canadensis Moench (typically derived from crosses between Populus nigra L and P. deltoides) and to characterize a natural population of P. nigra growing along the Ticino river in the North of Italy. Out of six SSRs used, four microsatellite loci were found to have alleles which were species-specific to P. deltoides and could therefore be used as markers for introgression of P. deltoides into P. nigra. In the studied region hybrid poplars and P. deltoides commercial clones are cultivated as monoclonal stands close to the area where black poplar has its natural habitat. SSR analysis was performed to investigate whether there was evidence of introgression between the natural population and the monoclonal plantations of hybrids and P. deltoides clones cultivated in the surrounding area. Three stages of the natural population were analysed: a group of old trees about a hundred years old, a younger population (aged 2-30 years) and the seedlings of three females of this population. Alleles specific to P. deltoides were detected only in the old cohort of the natural population, while no introgression was observed in the younger individuals and their progenies. These results were also confirmed by isozyme analysis of loci PGI-B, PGM and LAP-A, which were previously identified as diagnostic for P. nigra, P. deltoides and P.xcanadensis.  相似文献

The evolution of genotypic diversity with population age remains poorly explored in clonal plant populations despite the potential for important shifts to occur through the course of time. Woody sprouting species are particularly under-represented in studies investigating intra-specific variations in levels of clonality from one locality to the next and through time. In this study we sought to determine the incidence and frequency of replicate genotypes in natural Populus nigra L. (Salicaceae) stands of different ages. Ten stands of this woody riparian sprouting species were selected in each of three distinct age groups (young, middle-aged and old) along a 30 km stretch of the River Garonne (south-west France). Leaf samples were collected from 15 neighbouring trees in each stand (450 samples in total) and replicate genotypes were identified using five SSR markers. Replicate genotypes were identified in two-thirds of all stands sampled (i.e. 50 of young stands, 100 of middle-aged stands and 50% of old stands). Young stands had significantly fewer replicated genotypes than middle-aged or old stands, while middle-aged stands had the greatest number of replicated genotypes. Replicate genotypes were most often found to occur as nearest neighbours and formed relatively small, discrete units (i.e. 2–4 trees growing in close proximity to one another). This suggests that asexual regeneration frequently occurs through flood-training in this species, although asexual regeneration from translocated fragments also evidently occurs as evidenced by 11 cases of replicate genotypes occurring in widely separated stands (up to 19 km apart). The results of this study highlight the need for a hierarchical sampling strategy in space and across age groups for an accureate understanding of the genotypic structure of woody sprouting species populations. Conservation and management of effective population sizes will benefit from better insight into not only spatial, but also temporal variations in levels of genotypic diversity.Co-ordinating editor: J. Insect-resistant poplar (Populus nigra L.) plants have been produced by infecting leaves withAgrobacterium tumefaciens strains carrying a binary vector containing different truncated forms of aBacillus thuringiensis (B.t.) toxin gene under a duplicated CaMV 35S promoter. Putative transgenic plants were propagated by cuttings at two experimental farms (in Beijing and Xinjiang, China). At 2–3 years after transformation, 17 of them were selected on the bases of insect-tolerance and good silvicultural traits, and evaluated for insect resistance, for the presence of theB.t. toxin DNA fragment (Southern blots and PCR) and for the expression of the transgene (western and northern blots). Somaclonal variation, as suggested by the appearance of permanent changes in the shape of the leaves, was also investigated with molecular tools (RFLP (restriction fragment length polymorphism), RAPD (random amplified polymorphic DNA) and microsatellite DNA).Bioassays withApochemia cineraius andLymantria dispar on the leaves of the selected clones showed different and, in some cases, high levels of insecticidal activity. The molecular analysis demonstrated integration and expression of the foreign gene. Somatic changes were correlated to extensive genomic changes and were quantified in dendrograms, in terms of genomic similarity. The analysis of control plants suggested that genomic changes were correlated to thein vitro culture step necessary forA. tumefaciens-mediated gene transfer, rather than to the integration of the foreign genes.Three transgenic clones (12, 153 and 192), selected for insect resistance, reduced morphological changes and promising silvicultural traits, are now under large-scale field evaluation in six different provinces in China.  相似文献

Summary Lipophilic material produced by buds and young leaves of Populus nigra L. contains flavonoid aglycones. Under precise experimental conditions a green-yellow fluorescent light can be detected from these phenolic compounds. A cytophotometric technique using this property has been established: localisation and quantitative estimation of intracellular flavonoid material are realized by fluorometry. The accumulation and localisation of flavonoids in the different cellular compartments (plasts, vacuoles, cytoplasm, cell walls) have been determined for each period of the secretory cycle. These results were obtained from, in situ, investigations or from experiments with ethanolic extracts of cellular compartments.  相似文献

近年来,危害小黑杨Populus simonii×P.nigra的害虫发生严重,给林业生产造成很大损失。为了提高小黑杨的抗虫能力,避免使用杀虫剂带来的污染,用农杆菌介导法将澳大利亚漏斗蛛Atrax robustus的毒蛋白基因和苏云金芽孢杆菌CryⅠA(b)基因C末端的融合基因BGT转化入小黑杨。PCR 和Southern印记分析转基因植株,结果表明,BGT杀虫基因已经整合在小黑杨基因组上。活性实验表明,取食转基因杨树6天和9天后,舞毒蛾Lymantria dispar 2龄幼虫的校正死亡率分别是37.0%和92.6%。方差分析表明取食转基因和对照杨树的舞毒蛾幼虫体重差异显著。这些结果显示转基因杨树上的舞毒蛾的发育速率受到影响。  相似文献
