Oscillatory shear rheometry has been used to study the gelation of beta-lactoglobulin at ambient in 50% v/v trifluoroethanol (TFE)/pH 7 aqueous buffer and in 50% v/v ethanol (EtOH)/water at pH 2. In contrast to what was found on heating aqueous solutions at pH 2 (Part 2 of this series), a more expected "chemical gelation"-like profile was found with modulus components G' and G' ' crossing over as the gels formed and then with G' ' passing through a maximum. In addition, for the EtOH system, there was a significant modulus increase at long time, suggestive of a more complex two-step aggregation scheme. Modulus-concentration relationships were obtained for both systems by extrapolating cure data to infinite time. For the TFE gels, this data was accurately described by classical branching theory, although it could also be approximated by a constant power--law relationship. Only the latter described the modulus--concentration data for the gels in ethanol, but there were problems here of greater frequency dependence of the modulus values and much less certain extrapolation. Gel times for the TFE systems showed higher power laws in the concentration than could be explained by the branching theory in its simplest form being similar, in this respect, to the heat-set systems at pH 2. Such power laws were harder to establish for the EtOH gels as for these there was evidence of gel time divergence close to a critical concentration. Reduced G'/G'inf versus t/tgel data were difficult to interpret for the gels in ethanol, but for the TFE system they were consistent with previous results for the heat-set gels and approximated master curve superposition. The frequency and temperature dependences of the final gel moduli were also studied. In general, the networks induced by alcohols appeared more flexible than those obtained by heating.  相似文献

A micro analysis system for the electrochemical determination of the activity of protease along with pH sensing was fabricated aiming for its use in telemetric micro analysis systems targeting the testing of the stomach and intestines. The system consisted of a pH-sensing site and two protease assay sites formed in polydimethylsiloxane (PDMS) micro flow channels. To introduce sample solutions, valves were formed with gold electrodes in the inlets, which functioned on the basis of electrowetting. An external sample solution could be introduced into the sensing sites by switching on the valves at appropriate times. In the pH-sensing site, a pH-indicator electrode changed its electrode potential immediately after a sample solution reached an internal liquid-junction reference electrode. The slope of the calibration plot was -74.5 mVpH(-1). Bovine serum albumin (BSA) was used as the substrate for the enzyme and was spotted on the wall of the flow channel that faced the pH-indicator electrode of the protease assay sites. The release of protons accompanying the hydrolysis of BSA by the enzyme was detected using the pH-indicator electrode. When trypsin was contained in the sample solution as a test enzyme, a distinct decrease in pH, which was dependent on the trypsin activity, was observed, indicating that enzymatic hydrolysis was proceeding. The initial rate of potential change varied in proportion to the activity in a range between 1.0 and 51.7 Uml(-1). The integration of the microfluidic and sensing functions provides significant advantages for the use of this system as an isolated telemetric micro system that might operate with small batteries.  相似文献

The killing of Listeria monocytogenes following exposure to low pH, organic acids, and osmotic stress was enhanced by the addition of 5% (vol/vol) ethanol. At pH 3, for example, the presence of this agent stimulated killing by more than 3 log units in 40 min of exposure. The rate of cell death at pH 3.0 was dependent on the concentration of ethanol. Thus, while the presence 10% (vol/vol) ethanol at pH 3.0 stimulated killing by more than 3 log units in just 5 min, addition of 1.25% (vol/vol) ethanol resulted in less than 1 log unit of killing in 10 min. The ability of 5% (vol/vol) ethanol to stimulate killing at low pH and at elevated osmolarity was also dependent on the amplitude of the imposed stress, and an increase in the pH from 3.0 to 4.0 or a decrease in the sodium chloride concentration from 25 to 2.5% led to a marked reduction in the effectiveness of 5% (vol/vol) ethanol as an augmentative agent. Combinations of organic acids, low pH, and ethanol proved to be particularly effective bactericidal treatments; the most potent combination was pH 3.0, 50 mM formate, and 5 % (vol/vol) ethanol, which resulted in 5 log units of killing in just 4 min. Ethanol-enhanced killing correlated with damage to the bacterial cytoplasmic membrane.  相似文献

For the first time, Pulsed Field Gradient-Nuclear Magnetic Resonance, a powerful noninvasive tool for studying the dynamics and structure of complex gels, has been used to measure diffusion of probe molecules in aqueous solutions/gels of noncommercial purified pig gastric mucin (PGM), in a concentration range up to 5 wt %. Complementary data were obtained from rheology measurements. The combination of techniques revealed a strong pH dependency of the structure of the PGM samples while changes in concentration, ionic strength, and temperature appeared to induce less pronounced alterations. Viscosity was found to vary in a nonmonotonous way with pH, with the more viscous solutions found at intermediate pH. We propose that this finding is due to a reduced charge density at lower pH, which is expected to continuously increase the relative importance of hydrophobic associations. The results suggest a loose network of expanded fully charged PGM molecules with considerable mobility at neutral pH (pH 7.4). At intermediate pH (pH 4), a three-dimensional expanded network is favored. At pH 1, the charge density is low and microphase separation occurs since hydrophobic associations prevail. This leads to the formation of clusters concentrated in PGM molecules separated by regions depleted in PGM. The results obtained increase our knowledge about the gastric mucosal layer, which in vivo contains mucin in the same concentration range as that of the samples investigated here.  相似文献

The aggregation and gelation properties of beta-lactoglobulin (BLG), a globular protein from milk, was studied in aqueous ethanol solutions at room temperature. The phase state diagrams as a function of pH and ethanol concentration showed that a gel structure appeared after a period ranging from 1 min to 1 week, depending on the physico-chemical conditions. The in-situ kinetics of aggregation were followed by several methods in order to obtain a better understanding of the building of aggregates by the addition of ethanol. It was shown that the aggregation kinetics highly depended upon the pH, the process being fastest at pH 7. Viscoelasticity and infrared measurements indicated that alcohol-induced gelation would proceed via a two-step mechanism: small aggregates loosely connected between them were first built up; a real network took place in a second step. The coarse and irregular structures formed in aqueous ethanol gels revealed by confocal laser scanning microscopy could be analysed in terms of a phase separation. This observation was supported by a syneresis phenomenon visible in the final gel state. BLG in water-ethanol solution would undergo either an inhibition of the demixing by gelation or a binary phase separation accompanied by an irreversible gelation transition.  相似文献

On-line fluorescence-monitoring of the methanogenic fermentation   总被引:2,自引:0,他引:2  
On-line in situ fluorescence measurements of the methanogenic fermentation were conducted with reactors receiving either glucose or a mixture of volatile fatty acids as the substrate. The reactors were perturbed from steady-state conditions in order to assess the response of fluorescencemonitoring probes. Two fluorescence-monitoring probes were evaluated over a period of 8 months; they performed in a consistent manner, and their response was not significantly affected by the changes in pH and redox potential encountered during routine reactor operation. A commercially available probe, designed to measure NAD(P)H, demonstrated particular promise for detecting imbalance caused by the entry of air, inhibitor addition and was capable of distinguishing between different substrates. This fluorescence-monitoring probe detected imbalance more rapidly than other on-line measurements such as pH, Eh, or gas production, or off-line measurements such as volatile fatty acid concentration or gas composition. An experimental fluorescence-monitoring probe, designed to measure coenzyme F(420), also showed some promise in this regard. The response of the fluorescence-monitoring probes also revealed details of the metabolic routes in the reactors and the probes represent a useful research tool. For example, a failure to observe the characteristic response of the NAD(P)H-monitoring probe to formate addition during the metabolism of acetate, propionate, or glucose strongly suggests that any formate liberated during their catabolism is degraded via a different route to exogenously added formate.  相似文献

The aim of this work was to determine the potential application of dried sewage sludge as a biosorbent for removing phenol from aqueous solution. Results showed that biosorption capacity was strongly influenced by the pH of the aqueous solution with an observed maximum phenol removal at pH around 6-8. Biosorption capacity increased when initial phenol concentration was increased to 110 mg/L but beyond this concentration, biosorption capacity decreased suggesting an inhibitory effect of phenol on biomass activity. Biosorption capacity decreased from 94 to 5 mg/g when biosorbent concentration was increased from 0.5 to 10 g/L suggesting a possible competitive effect of leachable heavy metals from the sludge. The effect of Cu2+ on biosorption capacity was also observed and the results confirmed that the phenol biosorption capacity decreased when concentration of Cu2+ in the sorption medium was increased up to 15 mg/L. Desorption of phenol using distilled deionized water was less than 2% suggesting a strong biosorption by the biomass.  相似文献

A lightweight telemetric mobility sensing system was used to study the relationship between high levels of motor activity during free-play and school performance. Among the 21 normal children, there was a significant correlation between high ankle motor activity during free-play, poor school achievement, the presence of neurological soft signs, and a poor self-image. Those normals whose free-play ankle activity was above the mean, also had significantly more errors and performed at a significantly lower level on the Bender Visual Motor Gestalt Test than children whose activity was below the mean. This preliminary study suggests that the telemetric mobility sensing system can be easily applied to children to assess clinically relevant components of psychomotor activity.  相似文献

The effects of aqueous solutions of ethanol, acetonitrile and 1,4-dioxane in the concentration range 10–90% (v/v) on the activity of porcine pepsin were studied. The enzyme retained its activity in aqueous ethanol and aqueous acetonitrile with increasing organic solvent concentration up to 60%, and in aqueous 1,4-dioxane up to 30%, but thereafter a considerable decrease in activity was observed. The changes caused in the catalytic activity by the water-miscible organic solvents may be related to structural changes, which were followed by means of intrinsic fluorescence and circular dichroism spectroscopy measurements.  相似文献

Summary D-Xylose was fermented to ethanol by a strain ofPachysolen tannophilus in yields greater than 0.3g ethanol per g xylose consumed. Ethanol production was influenced by xylose concentration and was at a maximum at 10%, w/v. Ethanol formation occurred at pH 2.75-2.50 but the yeast would not grow at this pH when the initial pH of the medium was less than 3.0. Ethanol was consumed by the yeast when the xylose concentration became limiting. L-Arabinose, D-glucose, D-fructose, cellobiose, D-glucuronic acid, but not sucrose,were also fermented to ethanol byPachysolen tannophilus. Kinetic studies on xylose fermentation established various parameters involved in growth, substrate utilization and ethanol formation when the yeast was fermenter grown.  相似文献

The novelty of this approach was hydrolysis of the raw starch in ground corn to fermentable sugars that are simultaneously fermented to ethanol by yeast in a non-sterile environment. Thus, the conventional cooking step can be eliminated for energy conservation. A koji of Aspergillus niger grown on whole corn for 3 days was the crude enzyme source. A ratio of 0.2 g dry koji/g total solids was found sufficient. Optimum pH was 4.2. Ethanol concentration was 7.7% (w/w) in the aqueous phase with 92% raw starch conversion. Agitation increased rate. Sacharification was the rate-limiting step. The initial ethanol concentration preventing fermentation was estimated to be 8.3% by weight.  相似文献

Extractive fermentation is a technique that can be used to reduce the effect of end product inhibition through the use of a water-immiscible phase that removes fermentation products in situ. This has the beneficial effect of not only removing inhibitory products as they are formed (thus keeping reaction rates high) but also has the potential for reducing product recovery costs. We have chosen to examine the ethanol fermentation as a model system for end product inhibition and extractive fermentation and have developed a computer model predicting the productivity enhancement possible with this technique together with other key parameters such as extraction efficiency and residual glucose concentration. The model accommodates variable liquid flowrates entering and leaving the system, since it was found that the aqueous outlet flowrate could be up to 35% lower than the inlet flowrate during extractive fermentation of concentrated glucose feeds due to the continuous removal of ethanol from the fermentation broth by solvent extraction. The model predicts a total ethanol productivity of 82.6 g/L h if a glucose feed of 750 g/L is fermented with a solvent having a distribution coefficient of 0.5 at a solvent dilution rate of 5.0 h(-1). This is more than 10 times higher than for a conventional chemostat fermentation of a 250 g/L glucose feed. The model has furthermore illustrated the possible trade-offs that exist between obtaining a high extraction efficiency and a low residual glucose concentration.  相似文献

1,4-Diacetoxy-2,3-dicyanobenzene (ADB) has been increasingly used for measurement of intracellular pH by flow cytometry. ADB rapidly enters cells and is cleaved to the fluorescent pH indicator 2,3-dicyano-hydroquinone (DCH). We have analyzed several potential problems that can affect its usefulness as a pH indicator. Hydrolysis of ADB in aqueous solutions reveals the temporary presence of a fluorescent species blue-shifted from DCH at the same pH. The presence of this species with DCH can lead to erroneous pH measurements. Stable pH measurements with ADB depend on the incubation conditions and esterase activity. Heated cells required 20 min for stable measurements, whereas control cells required 5 to 10 min. The reproducibility of pH measurements was excellent, with a resolution of less than or equal to 0.05 pH units in the range of 6.4 to 8.0. Absolute calibration curves of intracellular pH using the ionophore nigericin depended on matching the intracellular K+ concentration with the buffer, but relative measurements of intracellular pH were insensitive to K+. ADB was nontoxic to Chinese hamster ovary cells at up to 20 micrograms/ml. However, when cells loaded with dye were passed through a UV laser beam, concentrations of dye greater than 5 micrograms/ml were highly toxic. Viable cells could be sorted on the basis of intracellular pH if ADB were used at low concentrations.  相似文献

A composite phyco-biomass including four different marine macroalgae species (Chaetomorpha sp., Polysiphonia sp., Ulva sp., and Cystoseira sp.) was evaluated as a novel biosorbent for the biosorption of manganese ions from aqueous solution. The experimental studies were performed to optimize the operational factors including solution pH, biosorbent amount, initial manganese concentration, and reaction time in a batch-mode biosorption system. The removal yield of the biosorbent for manganese ions increased with increasing pH, manganese ion concentration, and reaction time, while it decreased as the biosorbent dose increased. The obtained kinetic data indicated that the removal of manganese ions by the biosorbent was best described by the pseudo-second-order model and the pore diffusion also contributed to the biosorption process. The results of isotherm and thermodynamic studies showed that the Freundlich model represented the biosorption equilibrium data well and this biotreatment system was feasible, spontaneous, and physical. The maximum manganese uptake capacity of used biosorbent was found to be 55.874 mg g?1. Finally, a single-stage batch manganese biosorption system was designed and its kinetic performance was evaluated. All these findings revealed that the prepared composite macroalgae biosorbent has a fairly good potential for the removal of manganese ions from the aqueous medium.  相似文献

研究了用十六烷基三甲基溴化铵(CTAB)/正己醇/正辛烷反胶束溶液萃取和反萃取商业用胰激肽原酶时,水相pH值、离子强度和种类、CTAB浓度和助表面活性剂浓度等因素对分离效率的影响,并从反胶束微观结构给予解释。结果表明:[CTAB]=0.02 mol•L-1,正己醇/正辛烷(V/V)=1:5,萃取pH=9.0,反萃pH=7.0,萃取[KBr]=0.1 mol•L-1,反萃[KBr]=1.5 mol•L-1,反萃取加15%乙醇(V/V)时,萃取率接近100%,反萃取活性回收得率在80%以上。商业用酶的纯化倍数最高为1.97倍,粗酶为7.15倍,且粗酶纯化后比活在200U/mg以上,电泳分析证实了纯化效果,显示了很好的工业前景。  相似文献

The present study investigates the effect of pH and intermediate products formation on biological hydrogen production using Enterobacter cloacae IIT-BT 08. Initial pH was found to have a profound effect on hydrogen production potential, while regulating the pH 6.5 throughout the fermentation was found to increase the cumulative hydrogen production rate and yield significantly. Modified Gompertz equation was used to fit the cumulative hydrogen production curves to obtain the hydrogen production potential P, the hydrogen production rate R and lag phase λ. At regulated pH 6.5, higher H(2) yield (3.1molH(2)mol(-1) glucose), specific hydrogen production potential (798.1mL/g) and specific rate of H(2) production (72.1mLL(-1)h(-1)g(-1)) were obtained. The volatile fatty acid profile showed butyrate, ethanol and acetate as the major end metabolites of fermentation under the operating pH conditions tested; however, their pattern of distribution was pH dependent. At the optimum pH of 6.5, the acetate to butyrate ratio (A/B ratio) was found to be higher than that at any other pH. The study also investigates the effect of sodium ions on biohydrogen production potential. It was also found that sodium ion concentration up to 250mM enhanced the hydrogen production potential; however, any further increase in the metal ion concentration had an inhibitory effect.  相似文献

An amperometric method was using a nickel catalytic electrode in aqueous base solution for detecting acetylcholine (ACh). A sensing mechanism was developed in which ACh was hydrolyzed in base aqueous solution to produce the acetic anion and choline. The alcohol group of choline was oxidized to the corresponding carboxylic acid by Ni(OH)2/NiOOH catalytic system. The amperometric response resulted from the current generated by ACh oxidation in response to step changes in ACh concentration. The potential window of limiting current of ACh anodic oxidation at the Ni interface was determined in NaOH electrolyte. The effect of NaOH electrolyte concentration on sensitivity was also discussed. At the optimum operating condition, the method exhibits a good linear relationship between the response current and the ACh concentration. The response time of the ACh sensing system was 10 s. Scanning electrochemical microscopy (SECM) with platinum micro-tips was used to investigate the diffusion layer thickness of Ni electrode.  相似文献

A central composition design was developed to study the influence of process variables (temperature, pulping time and ethanol concentration) on the properties of the pulp produced (yield and holocellulose, alpha-cellulose and lignin contents) and the pH of the resulting wastewater, in the ethanol pulping of olive tree trimmings. The proposed equations reproduce the experimental results for the dependent variables with errors less than 5% for the holocellulose and alpha-cellulose contents, yield and wastewater pH, and less than 15% for the lignin content. Obtaining pulp with acceptably high yield (37.6%), high holocellulose and alpha-cellulose contents (above 88.8% and 46.9%, respectively), and low lignin contents (below 7.2%), entails operating at a pulping temperature of 200 degrees C, using an ethanol concentration of 75% and a pulping time of 60 min.  相似文献

The influence of increasing ethanol concentration in the feed on growth and physiological activity of the yeast Candida utlis was studied. The measurements were made at steady states of continuous culture under constant values of dilution rate, temperature, and pH in all stages of the fermentor; Synthetic ethanol was used as the sole source of carbon and energy in the concentration range 10-100 g/liter. The maximum biomass concentration in the effluent and maximum productivity was achieved at 75 g ethanol/liter in the feed. In respect to ethanol losses in the outlet and biomass yield, the optimum ethanol concentration in the input of the growth medium was found to be about 50 g/liter using a four-stage system.  相似文献
