Aim Mirabilis himalaica (Nyctaginaceae) is an endangered medicinal plant mainly distributed in the plateau region of northern Tibet, China. The outer surface of M. himalaica achenes is covered by a pectinaceous mucilaginous layer upon hydration. However, the role of the achene mucilage is poorly understood. In this study, we investigated the effects of mucilage on achene germination and sprout growth under abiotic stress to explain how M. himalaica survive the alpine environment.Methods We investigated the effect of mucilage on achenes germination by contrast the capacity of water absorption, dehydration and respiration of intact achene and the achene with mucilage removal. We performed abiotic stresses experiments including drought stress, salt stress, cold stress and high temperature stress, and quantified the effects of mucilage removal on achene germination rate, root and shoot lengths of seedlings.Important findings Mucilage is extremely hydrophilic, and the mass of intact achenes can be 9-fold greater than that of demucilaged achenes. The removal of the mucilaginous layer did not significantly change final germination percentages under ideal conditions, but intact achenes (i.e. with mucilage) took longer to germinate. The mucilage significantly decreased seed respiration rates by acting as a physical barrier that prevented oxygen diffusion. Germination rates, shoot and root growth of intact achenes were higher than those of demucilaged ones during exposures to cold, heat, osmotic and salt stresses. Achene mucilage presumably plays an ecologically important role in the life cycle of M. himalaica by aiding the critical achene germination and early seedling growth in the stressful habitats of the plateau region of northern Tibet.  相似文献

Despite proposed ecological importance of mucilage in seed dispersal, germination and seedling establishment, little is known about the role of mucilage in seed pre-germination processes. Here we investigated the role of mucilage in assisting achene cells to repair DNA damage during dew deposition in the desert. Artemisia sphaerocephala achenes were first treated γ-irradiation to induce DNA damage, and then they were repaired in situ in the desert dew. Dew deposition duration can be as long as 421 min in early mornings. Intact achenes absorbed more water than demucilaged achenes during dew deposition and also carried water for longer time following sunrise. After 4-d dew treatment, DNA damage of irradiated intact and demucilaged achenes was reduced to 24.38% and 46.84%, respectively. The irradiated intact achenes exhibited much higher DNA repair ratio than irradiated demucilaged achenes. Irradiated intact achenes showed an improved germination and decreased nonviable achenes after dew treatment, and significant differences in viability between the two types of achenes were detected after 1020 min of dew treatment. Achene mucilage presumably plays an ecologically important role in the life cycle of A. sphaerocephala by aiding DNA repair of achene cells in genomic-stressful habitats.  相似文献

Post-dispersal seed removal by animals can lead to extensive seed loss and thus is an important factor in structuring plant communities. However, we know much less about post-dispersal seed predation than about other forms of herbivory. Mucilage plays many ecological roles in adaptation of plants to diverse environments; nevertheless, until now the role of mucilage in ant-mediated seed movement remains largely hypothetical. We studied the role of mucilage in seed removal of Artemisia sphaerocephala by ants in Mu Us Sandland in Inner Mongolia, China. Messor aciculatus was the most active seed predator of Artemisia sphaerocephala. Time to first ant collecting (T 1st) of wet intact seeds was longest and significantly different from that for dry intact seeds, wet demucilaged seeds, and dry demucilaged seeds; number of seeds removed to ant nests was lowest for wet intact seeds. After they were collected by ants, 5 % of wet intact seeds were dropped during transport. Our results indicate that seed mucilage of Artemisia sphaerocephala may play a significant role in post-dispersal seed removal by (1) making seeds less attractive to ants, thus resulting in a delay of collection time; (2) forming a strong bond to soil particles, making it difficult for ants to remove seeds; and (3) making seeds more likely to be dropped during transport, thereby allowing them to escape from predation even after collection by ants. This study demonstrates the importance of mucilage in reducing seed removal by ants and thus in anchoring seeds of desert plants in the vicinity of mother plants.  相似文献

Artemisia sphaerocephala and A. ordosica are two dominant shrub species in Mu Us sandy land (Inner Mongolia, China) and are widely used for vegetation restoration. However, there are two different conclusions about the effect of light on their germination: light promotes germination versus light inhibits germination. The aim of this study was to evaluate these two conclusions and relate the results to instances of failure of these two species to germinate well when air-dispersed in revegetation projects. The effects of fluctuating temperature, light/dark, source (population), position on mother plant, storage condition, and storage time were tested on germination of achenes of these two species. At low temperature, final percent germination (FPG) of achenes in dark and nearly dark conditions was significantly higher than those in light. At 10:20°C, achenes of both A. sphaerocephala and A. ordosica had higher FPG in dark than in light regardless of source, position on mother plants or storage condition. At suboptimum (5:15°C) and supraoptimal (25:35°C) temperatures, germination of A. sphaerocephala and A. ordosica achenes was inhibited in both light and darkness. It was concluded that light inhibits germination of A. sphaerocephala and A. ordosica achenes at low (10:20°C) temperature but not at high (15:25°C) temperature. Since the temperature in Mu Us sandy land is around 10:20°C in early June, when air sowing is done, achenes should germinate best when they are covered by a thin layer of sand.  相似文献

Aims

Seeds of Henophyton deserti (Brassicaceae), an endemic saharan shrub in south Tunisia, produce a pectinaceous mucilage layer that can imbibe a large amount of water when wetted. The aim of this study was to explore the role of mucilage in seed germination of this shrub under heterogeneous stressful environments.

Methods

Germination of both intact and demucilaged seeds was tested over wide ranges of temperature, and in iso-osmotic solutions of NaCl and PEG. Recovery of germination after NaCl and Polyethylene Glycol (PEG)-6000 treatment was also tested. The effect of mucilage on water uptake was measured and the structure of the seed investigated.

Results

A considerable proportion of seed mass (30 %) is made up of mucilage, which is extremely hydrophilic and able to increase seed mass by 550 % over dry seeds. Mucilage water uptake appears to be unaffected by salt concentration, while higher concentrations of PEG inhibit mucilage hydration. Mucilage decreases germination specifically at 10 °C and this effect can be interpreted in relation to oxygen uptake. High concentrations of NaCl and PEG decrease both germination percentage and rate, with some greater tolerance at 15 °C and 20 °C versus 25 °C. Recovery was higher from higher concentrations of NaCl and PEG and lower temperatures, with a clear inhibitory effect of mucilage.

Conclusions

The study has shown that the mucilage of H. deserti may act as a physical barrier for regulating diffusion of water and oxygen to the inner tissue of the seed and thereby prevent germination under unsuitable conditions.  相似文献

To test the role of the seed mucilage of Plantago minuta Pall. in regulating germination under osmotic stress and cycles of hydration and dehydration, two experiments were carried out using seeds with intact mucilage and mucilage‐free seeds. In Experiment 1 seeds were immersed in a range of iso‐osmotic polyethylene glycol solutions (?1.15 to 0 MPa) for 14 days; any ungerminated seeds were transferred to deionized water to investigate the recovery germination. In Experiment 2 seeds were immersed in deionized water for 24 h, and were then incubated on filter paper for an additional 13 days to ensure complete desiccation before reimbibition to test the germination recovery percentage. Under mild osmotic stress (?0.73 to 0 MPa), the intact seeds with mucilage were shown to have higher germination rates than the mucilage‐free seeds, indicating that the mucilage led to a “fast sprouting” germination strategy under mild osmotic stress. However, when seeds were exposed to high osmotic stress (?1.15 MPa), the mucilage apparently slowed the germination rate, resulting in a “risk‐balancing” germination strategy. Extreme drought induced by polyethylene glycol solution and the desiccation pretreatment accelerated germination rates compared to non‐pretreated seeds; both germination potential and recovery percentage of the mucilage seeds were significantly higher than that of the mucilage‐free seeds. Our results revealed that the seed mucilage of P. minuta plays a crucial role in regulating seed germination rates and the germination strategies adopted by controlling seed water absorption when the seeds experience different osmotic stresses or alternating wet and dry conditions.  相似文献

通过等渗的NaCl和PEG溶液模拟盐分和水分胁迫,设置渗透梯度,在控制条件下对杠柳种子的萌发过程中总萌发率、幼苗鲜重、胚根生长、种子活力、发芽值等指标系统研究,对种子萌发率与渗透势之间关系进行回归分析,主要结果包括:(1)杠柳种子萌发过程中总萌发率、幼苗鲜重、活力指数和发芽值四项指标均随NaCl和PEG溶液的渗透势降低逐渐下降,综合研究活力指数和发芽值表明渗透势≥-0.5 MPa和≤-1.4 MPa时,PEG对种子萌发抑制作用大于NaCl,其他情况相反;(2)杠柳种子逐日萌发率和胚根日变化研究表明,与NaCl相比,PEG推迟杠柳种子萌发,并且对胚根增长抑制作用较大;(3)建立盐分和水分胁迫条件下种子萌发率与渗透势回归方程,发现杠柳种子在PEG胁迫下的萌发临界值和极限值为-1.0和-1.4 MPa,在NaCl胁迫下是-0.9和-1.3 MPa,解除胁迫条件,不同处理的杠柳种子复水萌发率均达到100%。说明杠柳具有良好的耐盐抗旱的特性。  相似文献

The success of seedling establishment of desert plants is determined by seedling emergence response to an unpredictable precipitation regime. Sand burial is a crucial and frequent environmental stress that impacts seedling establishment on sand dunes. However, little is known about the ecological role of seed mucilage in seedling emergence in arid sandy environments. We hypothesized that seed mucilage enhances seedling emergence in a low precipitation regime and under conditions of sand burial. In a greenhouse experiment, two types of Artemisia sphaerocephala achenes (intact and demucilaged) were exposed to different combinations of burial depth (0, 5, 10, 20, 40 and 60 mm) and irrigation regimes (low, medium and high, which simulated the precipitation amount and frequency in May, June and July in the natural habitat, respectively). Seedling emergence increased with increasing irrigation. It was highest at 5 mm sand burial depth and ceased at burial depths greater than 20 mm in all irrigation regimes. Mucilage significantly enhanced seedling emergence at 0, 5 and 10 mm burial depths in low irrigation, at 0 and 5 mm burial depths in medium irrigation and at 0 and 10 mm burial depths in high irrigation. Seed mucilage also reduced seedling mortality at the shallow sand burial depths. Moreover, mucilage significantly affected seedling emergence time and quiescence and dormancy percentages. Our findings suggest that seed mucilage plays an ecologically important role in successful seedling establishment of A. sphaerocephala by improving seedling emergence and reducing seedling mortality in stressful habitats of the sandy desert environment.  相似文献

Artemisia sphaerocephala is widely used for vegetation rehabilitation, but its germination is very low after air seeding of achenes. We explored effects of light, temperature and water stress on germination. Results show that both final percent germination and germination rate were increased by temperature increment, with the highest values occurring at 15: 25°C (night: day) in dark and 20: 30°C under light. Light inhibited germination, especially at lower alternating temperatures (5: 15°C and 10: 20°C). The alternating temperature window for germination was slightly narrower under light than in dark, and germination was slower under light than in dark across the temperature range. Achenes incubated in the dark and at constant temperatures had over 80% germination at 10 to 25°C, with an optimum at 20°C. Under dark and 25μmol m‐2 s‐1 light flux density at 10: 20°C, final percent germination was over 94%, but if the light flux density was increased to 100 and 400 μmol m‐2 s‐1, final percent germination was significantly lower (64% and 38% respectively). However, achenes could keep their germination capacity for a long time (over 50 days) and germinate well after going back to the dark. Germination was also lower under water stress and few achenes germinated at ‐1.4 MPa. This was more pronounced at high and low temperatures. Given these findings and the prevailing climatic characteristics, the most suitable time for air seeding of achenes may be mid‐May.  相似文献

分布于科尔沁沙地西部的固沙先锋植物乌丹蒿近些年呈现出衰退趋势。本研究选取了该地区3种主要飞播植物白沙蒿、柠条、羊柴以及乌丹蒿自身进行了化感作用的研究,结果表明:3种飞播植物及乌丹蒿自身的水浸提液对乌丹蒿的生长均有不同程度的化感作用,并且不同部位(根、叶、果皮或总苞片)中存在的化感物质的作用不同。主要表现为:乌丹蒿种子的萌发率在各供体的根浸提液中均下降,除羊柴外均达到显著,白沙蒿总苞片和羊柴果皮浸提液也有着同样的结果,而乌丹蒿叶浸提液则有一定的促进作用;4种植物不同部位的水浸提液对乌丹蒿种子发芽速度及幼根的生长均有显著的抑制作用。从综合效应来看,4种植物对乌丹蒿均有抑制作用,抑制作用由强到弱的顺序为:白沙蒿>柠条>乌丹蒿>羊柴。  相似文献

In contrast to the extensive understanding of seed mucilage biosynthesis, much less is known about how mucilage is biodegraded and what role it plays in the soil where seeds germinate. We studied seed mucilage biodegradation by a natural microbial community. High‐performance anion‐exchange chromatography (HPAEC) was used to determine monosaccharide composition in achene mucilage of Artemisia sphaerocephala. Mucilage degradation by the soil microbial community from natural habitats was examined by monosaccharide utilization tests using Biolog plates, chemical assays and phospholipid fatty acid (PLFA) analysis. Glucose (29.4%), mannose (20.3%) and arabinose (19.5%) were found to be the main components of achene mucilage. The mucilage was biodegraded to CO2 and soluble sugars, and an increase in soil microbial biomass was observed during biodegradation. Fluorescence microscopy showed the presence of mucilage (or its derivatives) in seedling tissues after growth with fluorescein isothiocyanate (FITC)‐labelled mucilage. The biodegradation also promoted early seedling growth in barren sand dunes, which was associated with a large soil microbial community that supplies substances promoting seedling establishment. We conclude that biodegradation of seed mucilage can play an ecologically important role in the life cycles of plants especially in harsh desert environments to which A. sphaerocephala is well‐adapted.  相似文献

In order to examine the response characteristics and possible reasons of Atriplex lentiformis and Atriplex undulata under salt stress at stage of seed germination, the seeds were treated with different concentrations of NaCl (0, 50, 100, 200 and 300 mmol⋅L−1), 20 mmol⋅L−1 LiCl or mannitol whose iso-osmotic concentrations corresponding to 200 mmol⋅L−1 NaCl. The results showed that the germination rate of two species of saltbush was depressed with the increase of NaCl concentration, and A. lentiformis showed greater salt tolerance compared with A. undulata. After removal of salt stress, the final germination ratio of A. lentiformis was over 93%, while that of A. undulata was only 56%. Evans blue staining revealed that 200 mmol⋅L−1 NaCl did not damage membrane permeability of A. lentiformis seed embryos, but significantly increased the membrane permeability of A. undulata seed embryos and caused irreversible damage to them, especially radicles. The results on water uptake indicated that the inhibition of NaCl on seed germination was mainly due to osmotic stress instead of ionic toxicity, and A. lentiformis exhibited higher salt tolerance due to its greater resistance to osmotic stress.  相似文献

Background and Aims

Seed germination is negatively affected by salinity, which is thought to be due to both osmotic and ion-toxicity effects. We hypothesize that salt is absorbed by seeds, allowing them to generate additional osmotic potential, and to germinate in conditions under which they would otherwise not be able to germinate.

Methods

Seeds of barley, Hordeum vulgare, were germinated in the presence of either pure water or one of five iso-osmotic solutions of polyethylene-glycol (PEG) or NaCl at 5, 12, 20 or 27 °C. Germination time courses were recorded and germination indices were calculated. Dry mass, water content and sodium concentration of germinating and non-germinating seeds in the NaCl treatments at 12 °C were measured. Fifty supplemental seeds were used to evaluate the changes in seed properties with time.

Key Results

Seeds incubated in saline conditions were able to germinate at lower osmotic potentials than those incubated in iso-osmotic PEG solutions and generally germinated faster. A positive correlation existed between external salinity and seed salt content in the saline-incubated seeds. Water content and sodium concentration increased with time for seeds incubated in NaCl. At higher temperatures, germination percentage and dry mass decreased whereas germination index and sodium concentration increased.

Conclusions

The results suggest that barley seeds can take up sodium, allowing them to generate additional osmotic potential, absorb more water and germinate more rapidly in environments of lower water potential. This may have ecological implications, allowing halophytic species and varieties to out-compete glycophytes in saline soils.  相似文献

Marathrum schiedeanum and Marathrum rubrum are annual Podostemaceae, thus their seeds are important to their dispersal and persistence in their habitat. We assessed the effect on germination of (1) light (white, red and far red) and darkness, (2) temperature (15, 20, 25, 30 °C and alternating 20/30 °C), (3) osmotic potential (0 to −0.8 MPa), (4) proximity to moisture sources and (5) seed storage. Seeds of M. schiedeanum and M. rubrum were non-dormant and had a high germination capacity (96%). Seeds were positive photoblastic; at 15 °C germination drop to zero, and germination rate was slower at 20 °C and at 20/30 °C than at 25 °C. A small proportion of seeds of both species germinated even at osmotic potentials as low as −0.6 MPa (11%) for M. rubrum and −0.8 MPa (70%) for M. schiedeanum. Seeds germinated only when near to the source of moisture (91.3–87.1% and 53.3–35.6% for M. schiedeanum and M. rubrum, respectively) and 2 years in dry storage did not modify their capacity to germinate. At the beginning of the rainy season, light and temperature in the rivers may be high enough for germination. The ability to germinate at low osmotic potential may be related to early germination during the rainy season. This may be because the seed mucilage assists in diffusion of water from the substrate to the seed. Both species germinated faster at −0.06 MPa, than in distilled water, which may indicate appropriate conditions for germination of these short-lived species.  相似文献

白沙蒿(Artemisia sphaerocephala Krasch.)是中国西北部沙漠的流动及固定沙丘上广泛分布的优势种灌木。瘦果小而轻,借助显微镜和扫描电镜对瘦果的形态结构进行了观察,其种子的种皮与果皮愈合,果皮外层为很厚的粘液层,其粘液层在遇湿后迅速吸水膨胀,其重量增至原来的589倍。在自然生境中,粘液物质将沙粒粘附于瘦果的周围,使瘦果的重量发生变化。瘦果能长时间在水上漂浮,粘液物质有助于种子的萌发和苗的发育。白沙蒿产生三种不同颜色的瘦果,其种子具有不同的萌发速率但最终达到相同的萌发率。研究表明瘦果的粘液物质对白沙蒿种子的传播与萌发具有重要的生态意义。  相似文献

The differential germination responses of ray and disc achenes of Hemizonia increscens (Asteraceae) were compared in field and laboratory investigations in order to gain insight into the ecological and evolutionary significance of heterocarpy. In the field, 200 ray and 200 disc achenes were placed in native, sterilized soil in a series of cleared, randomized, replicated plots. In a nearby plot a similar number of achenes were placed in plastic petri dishes in which high moisture conditions were maintained. Disc achene germination occurred under relatively minimal moisture conditions (<1 cm rainfall for 19 days) and relatively mild temperature regimes (21–7 C). Disc achene germination began three days after planting in the field plot and four days after they were put into the field petri dishes. In contrast, the onset of ray achene germination occurred 21 days after planting in the field plot and 19 days after planting in the field petri dishes. Averages of 2.05 and 2.71 disc achenes/day germinated in the field plot and field petri dishes, respectively. These contrasted with averages of 0.57 and 0.50 ray achenes/day germinated in the field plot and in the field petri dishes, respectively. A total of 67.5% and 69% disc achenes germinated in the field plot and the field petri dishes, but only 18% and 16.5% ray achenes germinated in the field plot and field petri dishes, respectively. Three separate treatments, using 100 ray and 100 disc achenes in each, were performed in laboratory growth chambers: 1) nicking the fruit coat, 2) excising the embryo, and 3) leaving the fruit coat intact. Onset of germination for all disc achene treatments occurred after three days. No significant differences were found among the three disc achene treatments in timing, rate, or germination percentage. All three disc treatments in the laboratory closely paralleled those for disc achenes in the field plots in time and germination percentage, but rates of germination were not as high. Germination of the nicked and excised ray achenes treatments began after four days, while germination of the untreated ray achenes began after 27 days. Untreated ray achenes in the laboratory paralleled the ray achenes in both field experiments in rate and germination percentage, but were delayed in time of germination. The nicked and excised ray achene treatments, however, were similar to the disc achene treatments in time of germination, and were not significantly different from disc achenes in rate of germination. These data suggest 1) that ray and disc achenes are markedly different in germination under identical conditions in field and laboratory experiments, and 2) this difference in germination response may be due to the thicker pericarp of the ray achenes.  相似文献

疏齿千里光(Senecio subdentatus)是分布在新疆北部古尔班通古特沙漠中的一种具异形瘦果的菊科一年生短命植物。将野外观测与室内实验相结合, 对该物种异形瘦果的形态、扩散和萌发特性, 以及异形瘦果产量与植株大小的关系进行了研究, 并对其生态学意义进行了探讨。结果表明: 疏齿千里光果序中的外围果和中央果均为柱形, 但前者为淡黄色, 后者为褐色, 且二者在大小、冠毛长度及果皮微形态等方面均存在明显差异。两种瘦果均以单个果实为扩散单元, 且在静止空气中的降落速度和在1 m·s-1与2 m·s-1风速下的扩散距离无显著差异, 说明虽然果实大小和冠毛长度对瘦果扩散具有不同的影响, 但对其整体扩散能力无明显影响。在各温变周期(5/2、15/2、20/10、25/15和30/15 ℃)处理中, 淡黄色外围果的萌发率均高于褐色中央果的, 且不同温度间两种瘦果的萌发率均存在显著差异, 但光照条件对其无显著影响。果序中的中央瘦果数明显多于外围果的, 且植株中外围果所占比例与植株大小间呈显著负相关关系, 而中央果所占比例与植株大小间呈显著正相关关系。这些特点说明, 该物种的小植株倾向于产生较多较易萌发的外围果, 大植株倾向于产生较多不易萌发的中央果。在古尔班通古特沙漠不可预测的极端环境中, 疏齿千里光可通过异形瘦果间的萌发差异及调节其不同大小植株中异形瘦果的比例, 来减少同胞后代之间的竞争, 增加其对不同微环境条件的生态适应性。  相似文献

Kosteletzkya virginica (L.) Presl. (Malvaceae) is a perennial that grows in saline or brackish water, and is salt-tolerant in its mature state, but less tolerant during germination. The seeds show a very low permeability to water that increases during storage. The permeability to water differs in seeds harvested in different years. Optimal temperature for germination is 28–30 C. The effect of salinity on imbibition is largely osmotic, but germination is inhibited, apparently, by the combined osmotic and “ionic” effects, especially at high NaCl concentrations. Inhibition of germination by high NaCl concentrations is relatively more severe in scarified than in intact seeds, indicating that the seed coat acts as a partial barrier to Na+ influx. External application of proline or betaine did not improve germination under saline conditions. Dry seeds contain a significant amount of betaine and low levels of proline, but during germination and in the presence of NaCl the betaine content decreased while the proline content increased. Thus, the likely compatible solute in the germinating seed seems to be proline.  相似文献

A total of ten rare indigenous rice landraces of West Bengal were screened for germination potential and seedling growth under varying concentrations of sodium chloride (NaCl) and polyethylene glycol (PEG) solutions as osmotic stress inducing agents. Among the studied rice landraces Kelas and Bhut Moori showed highest degree of tolerance to induced osmotic stresses. Proline content of the studied lines was also determined. Genetic relationship among the studied rice landraces was assessed with 22 previously reported osmotic stress tolerance linked Simple Sequence Repeat (SSR) markers. The identified allelic variants in form of amplified products size (molecular weight) for each SSR marker were documented to find out allele mining set for the linked markers of the studied genotypes in relation to osmotic stress tolerance. A Microsatellite Panel was constructed for the different allelic forms (size of amplified products) of each used marker. Among 22 SSR markers, ten showed unique alleles in form of single specific amplified product for the studied four genotypes which can be used for varietal identification. Genetic relationship among the studied rice lines was determined and a dendrogram was constructed to reveal their genetic inter-relationship. Polymorphism Information Content (PIC) for each used marker was also calculated for the studied rice lines.  相似文献
