The aim of the current investigation was to study the effect of lithium on circadian rhythms of pineal - testicular hormones by quantitations of pineal and serum serotonin, N-acetylserotonin and melatonin, and serum testosterone at four time points (06.00, 12.00, 18.00 and 24.00) of a 24-hr period under normal photoperiod (L:D), reversed photoperiod (D:L), constant light (L:L) and constant dark phase (D:D) in rats. Circadian rhythms were observed in pineal hormones in all the combinations of photoperiodic regimens, except in constant light, and in testosterone levels in all the photoperiodic combinations. Pineal and serum N-acetylserotonin and melatonin levels were higher than serotonin at night (24.00 hr), in natural L:D cycle, in reversed L:D cycle or similar to normal L:D cycle in constant dark phase, without any change in constant light. In contrast, testosterone level was higher in light phase (12.00 hr through 18.00 hr) than in the dark phase (24.00 hr through 06.00 hr) in normal L:D cycle, in reversed L:D cycle, similar to normal L:D cycle in constant dark (D:D), and reversed to that of the normal L:D cycle in constant light (L:L). Lithium treatment (2 mEq/kg body weight daily for 15 days) suppressed the magnitude of circadian rhythms of pineal and serum serotonin, N-acetylserotonin and melatonin, and testosterone levels by decreasing their levels at four time points of a 24-hr period in natural L:D or reversed D:L cycle and in constant dark (D:D). Pineal indoleamine levels were reduced after lithium treatment even in constant light (L:L). Moreover, lithium abolished the melatonin rhythms in rats exposed to normal (L:D) and reversed L:D (D:L) cycles, and sustained the rhythms in constant dark. But testosterone rhythm was abolished after lithium treatment in normal (L:D)/reversed L:D (D:L) cycle or even in constant light/dark. The findings indicate that the circadian rhythm exists in pineal hormones in alternate light - dark cycle (L:D/D:L) and in constant dark (D:D), but was absent in constant light phase (L:L) in rats. Lithium not only suppresses the circadian rhythms of pineal hormones, but abolishes the pineal melatonin rhythm only in alternate light - dark cycles, but sustains it in constant dark. The testosterone rhythm is abolished after lithium treatment in alternate light - dark cycle and constant light/dark. It is suggested that (a) normal circadian rhythms of pineal hormones are regulated by pulse dark phase in normal rats, (b) lithium abolishes pineal hormonal rhythm only in pulse light but sustains it in constant dark phase, and (c) circadian testosterone rhythm occurs in both pulse light or pulse dark phase in normal rats, and lithium abolishes the rhythm in all the combinations of the photoperiod. The differential responses of circadian rhythms of pineal and testicular hormones to pulse light or pulse dark in normal and lithium recipients are discussed.  相似文献

Plasma melatonin concentrations were measured in marmots (Marmota flaviventris) maintained under three short-day (4L:20D; 8L:10D; 10L:14D) and one long-day (14L:10D) photoperiod(s). Each animal had a daily rhythm of plasma melatonin with elevated plasma melatonin levels occurring during the dark period of the lighting cycle. There were no significant differences between any peak values during the night. The mean duration of elevated night melatonin concentrations was significantly different between long-day (16L:8D) and 8L:16D or 4L:20D animals (P less than 0.01). Daytime plasma melatonin levels were not significantly different among the photoperiods. These results characterize plasma melatonin rhythms in a sciurid rodent and demonstrate that this rhythm is modified by photoperiod. Therefore, the plasma melatonin profile could convey information about day length to the animal or, alternatively, the rhythm may be acting as a time-keeping mechanism for other physiological functions.  相似文献

The adult emergence rhythm of Telenomus busseolae, an egg parasitoid of Sesamia nonagrioides, was examined when parasitoids were exposed to different light-dark regimes. Most of the adult parasitoids emerged throughout the whole period of the photoperiodic cycle. Peak male emergence occurred 2–5 hours earlier than that of females. Adult emergence was asynchronous in continuous darkness or light. However, regimes of alternative light and dark phases such as L4:D20, L8:D16, L12:D12, L16:D8 and L20:D4 h generated a population rhythm with a period length of 24 hours. The peak of the emergence activity moves from the scotophase to the middle of the photophase with an increase of the photophase from 4 to 20 h. Rhythmical activity of adults was synchronised within 2 cycles when immature stages of parasitoid grow under continuous light conditions (LL) and then transferred to L12:D12. Moreover, emergence rhythm persisted and continued in a free-run with a period length of less than 24 hours by transferring a rhythmic culture from L12:D12 h to LL or RR (continuous red light) conditions, indicating the existence of a circadian rhythm. The ecological implications of the expression rhythm relate to better survival of the parasitoids.  相似文献

To demonstrate the involvement of circadian rhythm in photoperiodic time measurement, photosensitive female blackheaded buntings were kept under different photoperiodic schedules consisting of 6 h of main photophase coupled with scotophases of various durations. Ovarian mass and circulating plasma estradiol concentration were found to be elevated in cycles of 6L:6D, 6L:36D, 6L:54D and in control 15L:9D groups. But cycles of 6L:18D, 6L:42D and 6L:66D did not stimulate ovarian growth or elevate circulating plasma estradiol concentration. These results are consistent with the Bünning hypothesis according to which a photoperiodic response is elicited as a result of the coincidence of light with the photoinducible phase of an endogenous circadian rhythm. The results thus indicate the involvement of a circadian rhythm of photoinducibility in ovarian growth and estradiol secretion.  相似文献

In Neurospora crassa, a circadian rhythm of conidiation (asexual spore formation) can be seen on the surface of agar media. This rhythm has a period of 22 hr in constant darkness (D/D). Under constant illumination (L/L), no rhythm is visible and cultures show constant conidiation. However, here we report that strains with a mutation in the vivid (vvd) gene, previously shown to code for the photoreceptor involved in photo-adaptation, exhibit conidiation rhythms in L/L as well as in D/D. The period of the rhythm of vvd strains ranges between 6 and 21 hr in L/L, depending upon the intensity of the light, the carbon source, and the presence of other mutations. Temperature compensation of the period also depends on light intensity. Dark pulses given in L/L shift the phase of the rhythm. Shifts from L/L to D/D show unexpected after effects; i.e., the short period of a vvd strain in L/L gradually lengthens over 2–3 days in D/D. The rhythm in L/L requires the white collar (wc-1) gene, but not the frequency (frq) gene. FRQ protein shows no rhythm in L/L in a vvd strain. The conidiation rhythm in L/L in vvd is therefore driven by a FRQ-less oscillator (FLO).  相似文献

Roots of sunflower plants (Helianthus annuus L. var. Mammoth Russian) subjected to L12:D12, L18:D6, and L12:D12 followed by continuous light all display rhythms of about 12 hours for glutamine synthetase (GS) activity (transferase reaction) with one peak in the `light phase' and one in the `dark phase.' Root energy charge (EC = ATP+½ADP/ATP+ADP+AMP) is directly correlated with GS, but the GS rhythm is better explained as the result of a rhythmic adenine nucleotide ratio (ATP/ADP+AMP) that regulates enzyme activity through allosteric modification. When L12:D12 plants are subjected to free-running conditions in continuous darkness, only diurnal rhythms for GS and EC, with peaks in the dark phase, remain. The 12-hour root rhythms for GS and EC appear to be composed of two alternating rhythms, one a diurnal, light-dependent, incompletely circadian light phase rhythm and the other a light-independent, circadian dark phase rhythm.

Only glutamine, of the root amino acids, displays cyclical changes in concentration, maintaining under all conditions a 12-hour rhythm that is consistently synchronized with, but nearly always inversely correlated with, GS and EC rhythms.

To investigate the daily rhythm of rectal and vaginal temperature, we used six mongrel dogs with a mean body weight of 15±3 kg, aged between 2 and 3 years old. Rectal and vaginal temperatures were recorded every 3 h over 48-h period during three different lighting regimes: natural light/dark (L/D) cycle (sunrise 06:25, sunset 17:05), constant light (L/L) and constant darkness (D/D). A daily rhythm of rectal temperature was observed in both days of monitoring in all experimental conditions. Vaginal temperature showed a daily rhythmicity in L/D and D/D cycle. During the L/L cycle, daily rhythm was disrupted. Rhythmic parameters (MESOR, amplitude, acrophase and robustness) did not change between the different photoperiod and the site of temperature collection. In conclusion, the monitoring of vaginal temperature can be considered a valid alternative to the monitoring of body temperature as well as rectal temperature under natural lighting conditions in canine medicine.  相似文献

Although a significant body of literature has been devoted to the chronobiology of aquatic animals, how biological rhythms function in molluscan bivalves has been poorly studied. The first objective of this study was to determine whether an endogenous circadian rhythm does exist in the oyster, Crassostrea gigas. The second objective was to characterize it in terms of robustness. To answer these questions, the valve activity of 15 oysters was continuously recorded for 2 mo in the laboratory under different entrainment and free-running regimes using a high-frequency noninvasive valvometer. The present work demonstrates the presence of a circadian rhythm in the oyster Crassostrea gigas. First, oysters were entrained by 12?L:12 D conditions. Then, free-running conditions (D:D and L:L) indicated that the most frequently observed period ranged from 20 to 28?h, the circadian range. That endogenous circadian rhythm was characterized as weak. Indeed, the period (τ) of the individual animals exhibited high plasticity in D:D and L:L, and the animals immediately followed a 4-h phase advance or delay. Additionally, C. gigas appeared as a dual organism: all oysters were nocturnal at the beginning of the laboratory experiment (January), whereas they were diurnal at the end (March). That shift was progressive. Comparison with a full-year in situ record showed the same behavioral duality as observed in the laboratory: the animals were nocturnal in autumn–winter and diurnal in spring–summer. Abstract

A study was made on the developmental rhythms of Ephestia kuehniella during different photoperiods: constant lighting (L/L), under conditions of 12 hours light per day (L/D) and in constant darkness (D/D). Observations were made every 2 hrs, for 3 successive 24‐hr periods, of the number of wandering larvae emerging from food, the number of pupations and of imaginai moults. Emergence of wandering larvae is rhythmic only under L/D conditions, and in addition this is also affected by the density of the larvae. Rhythm is least apparent with greatest density of the larvae. Pupation in E. kuehniella is a non‐rhythmic process with each of the photoperiods examined, whereas the imaginai moult is characterized by an endogenous (existing under D/D conditions) rhythm. Emergence of imagines is non‐rhythmic under L/L, and also when pupae under L/D fail to receive the final period of darkness. The rhythm of the imaginai moult is possibly controlled by the biological clock, which stops under conditions of constant light.  相似文献

A biphasic circadian rhythm in the content of liver lipid peroxidation products has been demonstrated in male Wistar rats housed under the conditions of 12L: 12D, with 3 hours of morning and evening twilight. Maxima of the concentration of the products were observed in the morning and early at night. The rhythm of lipid anti-oxidative activity was found in an anti-phase. Inversion of both the L: D cycle and glucocorticoid circadian rhythms (cortisol injections) led after 14-16 days to the same shifts in the rhythm of anti-oxidative activity. The data indicate that glucocorticoids modulate the diurnal rhythms of lipid anti-oxidative activity and may be responsible for the shifts in the rhythms of free radical oxidation, induced by inversion of the L: D cycle.  相似文献

Reproduction in most fish is typically a seasonal process, as spawning takes place usually at a given time of the year, depending on the reproduction strategy of the species, to ensure maximal survival of offspring. Nevertheless, fish reproduction cannot be considered an exclusively annual phenomenon, because spawning may also show daily rhythmicity. In this study, we investigated the existence of a daily spawning rhythm in gilthead seabream (Sparus aurata L) exposed to different light‐dark (LD) cycles and at different times of the year using an automatic and programmable egg collector. Floatability and fertilization rates were analyzed at different times throughout the 24 h. The results showed a daily spawning rhythm with spanning occurring from 14:30 to 21:30 h, with the acrophase (peak time) being 18:29 and 18:08 h in fish exposed to an artificial photoperiod of 9L∶15D in winter and in spring, respectively. Nevertheless, in fish exposed to a natural photoperiod of 12L∶12D in spring, the acrophase of the rhythm was recorded later, at 21:28 h. The average fertilization rate was 87%, and no significant differences were found between the different hours of spawning. Moreover, when the LD cycle (9L∶15D) was shifted by 12 h, the daily spawning rhythm gradually re‐synchronized, resuming a stable phase‐relationship after 4–5 transient days, which is characteristic of a endogenous circadian rhythm. Our results clearly demonstrated the existence of a 24 h period of spawning in gilthead sea bream, with a peak anticipating the forthcoming night, and its capacity to gradually re‐synchronize after a 12 h shift in the LD cycle, pointing to the endogenous nature of this rhythm. These findings will be valuable for better understanding the reproductive physiology of this species and for optimizing the protocols of egg collection and larvae production in aquaculture. It is known that contemporary space station revolves at the altitude of 400-500 km in the outer space. The present study was undertaken to investigate the effects of simulated space rhythm (L:D = 0.75 h:0.75 h) at this altitude on circadian rhythm in tree shrews (Tupaia belangeri chinensis) and the effects of endogenous sleep inducing neuropeptide Asp5-alpha-DSIP on the space-rhythm-entrained circadian rhythm. This primitive stock serves as one species of Tupaiidae and is a unique native of South China. Our previous studies have shown that this species showed striking differences in natural circadian rhythm between day and night (e.g. 3.03 degrees C of rectal temperature). Results showed that the above mentioned space rhythm (L:D = 0.75h:0.75h) could drastically disturb the inherent circadian rhythm of Tupaia belangeri chinensis. The maximal peak of motor activity dropped significantly in the morning (0600-1200) and a new enhanced peak (more than 20 times greater than that of the control) was found between 1800-2400, whereas the maximal trough of motor activity (2400-0600) remained basically unchanged. Concurrently, the total amount of 24-h motor activity was significantly decreased and the recovery after the cessation of space rhythm was slow. Experimental results also demonstrated that consecutive administration of Asp5-alpha-DSIP (30 micrograms/kg, i.p.) for 5 days (2 days before and 3 days during space rhythm) did not prevent the basic disturbance of circadian rhythm of Tupaia belangeri chinensis caused by the space rhythm (L:D = 0.75h:0.75h). The objective of this study was to investigate the entrainment of melatonin rhythms in rams using symmetrical light-dark cycles of different period length. Five groups of six He de France rams were kept in 12L: 12D for 7 weeks and then (i) 12L: 12D, (ii) 11L: 11D, (iii) 10L: 10D, (iv) 13L: 13D and (v) 14L: 14D for a further 3 weeks. Environmental factors other than the light dark cycle were not controlled. The onset and offset of the plasma melatonin rhythm in DD after 3 weeks of the respective light treatments was assessed for 48 hr, immediately after transferring to DD. The duration of secretion in DD was positively related to the length of the previous dark phase. The phase of the melatonin rhythm with respect to the anticipated dark phase suggested entrainment with no change in phase-relationship to the zeitgeber by 12L: 12D and 13L : 13D. Entrainment with a phase-delay or a phase-advance was apparent after 11L: 11D and 14L: 14D, but the individual rhythms were not all synchronized with respect to each other after 10L: 10D. Activity recordings for 2-3-week periods during 12L: 12D, 10L: 10D and 14L: 14D all showed a major 24-hr component at all times, with activity during the light phase in 12L: 12D. It appears that melatonin may be readily desynchronized from overt activity-rest cycles in sheep. The upper and lower entrainment limits are probably greater than 28 hr and close to 20 hr cycles, respectively.  相似文献

The influence of the daily distribution of light on the duration of the breeding season of the barbel was investigated. Interrupted night photoperiods (6L:2D:2L:14D or 6L:8.5D:2L:7.5D) allowed spawnings of male and female barbel (previously maintained under 10L:14D or 16.5L:7.5D) to continue, whereas a 2 or 8.5 h shorter photoperiod (8L: 16D) inhibited both sexes. The number of spawnings for each female was significantly higher under 6L:2D:2L:14D or 6L:8.5D:2L:7.5D (respectively 2.9 and 3.6 spawnings on average) than in control groups maintained under 8L:16D (0.6 and 2.1 spawnings on average). These results suggest that there is a daily rhythm of photosensitivity in the barbel.  相似文献

The seasonal maturation of four species of helminths, Acanthocephalus clavula, A. lucii, Camallanus lacustris and Bunodera luciopercae, from the perch Perca fluviatilis is briefly described. It is noted that A. clavula, A. lucii and C. lacustris are found as mature worms in the intestine of the fish host throughout the year, whereas in Bunodera luciopercae there is a limited season of maturation. As a result of experimental studies it is suggested that B. luciopercae uses host gonadotrophin to initiate and stimulate the early phase of gametogenesis. The gonadotrophin may also activate the parasites' own endocrine system, which then takes over control of subsequent spermatogenesis and oogenesis. This formation is used for a hypothesis for seasonal maturation of helminths in the fish definitive host. It also provides a potential explanation for seasonal maturation of helminths of fish in polar and tropical zones of the world.  相似文献

The present study was conducted to describe the impact of circadian rhythm on melatonin levels and redox statusunder three photoperiods (12L:12D, 0L:24D, and 24L:0D) in head and hemolymph of Spodoptera litura. Melatonin is an powerful antioxidant and controls the reproduction of organisms. In this study, melatonin levels, Arylalkylamine N-acetyltransferase(AA-NAT), and antioxidant enzyme activities were analyzed. Results showed melatonin, AA-NAT levels in hemolymph were significantly (p < 0.05) higher during the dark period than during LL regime. HPLC chromatogram of the insect head and hemolymph showed 5 peaks while hemolymph showed 6 peaks in LD, and LLregimes. The day–night changes of melatonin increased the antioxidant enzymes (GST, CAT, POX) persisted in the insect hemolymph, but were suppressed by constant light. The present study leads us to speculate that synthesis and release of melatonin in the S.litura head occur as circadian rhythm and light has an inhibitory effect on melatonin synthesis.  相似文献

Abstract

The Locomotory activity of the Plains Garter snake was determined under L/D: 12/12 conditions at five constant temperatures and three light intensities during the light period. The snakes were diurnal at low temperatures with nocturnal activity increasing in various amounts at higher temperatures. The different light intensities had relatively small effects on the activity rhythm.

Activity was recorded under four constant light conditions at five constant temperatures and the characteristics of the free‐running rhythm measured. A comparison of the characteristics of the free‐running rhythm to Aschoff's circadian rule indicates that this snake is an exception to this rule.

Increase light intensity decreased total activity under all conditions. Under a L/D: 12/12 cycle the decrease in activity was more pronounced during the dark period than the light period.

It is suggested that crepuscular or nocturnal activity shown by snakes at high temperatures may be an effect the temperature level has on the biological clock and activity controlling mechanisms rather than temperature selection by the snake.  相似文献

We studied the influence of light-dark (L:D) cycle reversal on daily variations in the brown adipose tissue (BAT) capacity for nonshivering thermogenesis (NST) in Siberian hamsters (Phodopus sungorus). Continuous and simultaneous measurements of BAT temperature (T(BAT)) and preferred ambient temperature (PT(a)) were made after noradrenaline (NA) injections administered every 4 hr. First, hamsters were acclimated for 4 weeks to an ambient temperature (T(a)) of 23 degrees C and 12L:12D, and then to a reversed photoschedule 12D:12L for 8 weeks. The same was done after a 4- and 8-week acclimation period at the same T(a). We found that after photoschedule reversal, the re-entrainment of T(BAT) and PT(a) rhythms preceded re-entrainment of the NST rhythm. The daily rhythms of T(BAT) and PT(a) were fully re-entrained after 4 weeks of acclimation to the reversed photoschedule, but rhythmicity of the response to NA disappeared. This rhythm was restored in hamsters acclimated to a reversed photoschedule for 8 weeks. We suggest that the daily rhythm of NST capacity is not responsible for generating the rhythm of body temperature (T(b)). Rather, it is a result of the daily rhythm of T(b), but adjusts to the new environment more slowly than the T(b) rhythm. When a daily rhythm of NST was present, the increase in T(BAT) after NA injection was inversely correlated with the pre-injection T(BAT). In addition, NA-induced changes in PT(a) reflected the intensity of NST in BAT; namely, increased T(BAT) was correlated with the post-injection decrease in PT(a). When the increase in T(BAT) was large, animals chose a lower T(a) to dissipate excessive heat and prevent overheating. In the course of the experiments, we recorded a decreased mean NST capacity and increased body mass of hamsters. These changes are representative of the time of photorefractoriness and a transition to a summer status. Despite prolonged exposure to an intermediate day length (12 hr of light) and photoschedule reversal, hamsters continued to change towards their summer condition and were able to acclimate to the new D:L cycle.  相似文献

To examine the importance of the inductive light period of a skeleton photoperiod in relation to the endogenous circadian rhythm of photoinducibility mediating photoperiodic induction, P. domesticus were exposed for 28 weeks to a series of skeleton photoperiods, viz. 6L:4D:1L:13D, 6L:6D:1L:11D. 6L:8D:1L:9D and 6L:14D:1L:3D. The inductive effects of 1 hr light pulse at night varied depending on the time of its placement. To compare the inductive effects of complete and its corresponding skeleton photoperiods, birds in the second experiment were subjected for 20 weeks to 12L:12D and 6L:5D:1L:12D given daily or interposed on alternate days with constant darkness (12L:12D/DD and 6L:5D:1L:12D/DD). There was a difference in the rate and magnitude of response between the complete and skeleton photoperiods. It appears that the subtropical house sparrow uses photoperiodic strategy in regulation of its seasonal testicular responses similar to that is reported for its temperate population.  相似文献
