In unrestrained cats clozapine in increasing doses (1--5 mg/kg) caused a behavioural depression and suppression of amphetamine-induced stereotypy of behaviour against the background of marked vegetative shifts. Similarly to chlorpromazine clozapine intensified the behavioural arrest reaction) and electrographic (neocortical caudate spindle) indices of the arrest function of the caudate nucleus. Arrest reaction changed more distinctly in stimulation of the ventral parts of the head of the nucleus. Clozapine also eliminated the weakening of the caudate responses caused by the stereotypical doses of amphetamine.  相似文献

Large doses of d,1-amphetamine produce in cats a stereotype behaviour: its chronic administration results in low variability of the behaviour of one and the same animal and a stable set of motor automatisms. This makes it possible to use the cyclography for an objective estimation of the d,1-amphetamine-induced stereotypy. Low-frequency stimulation of the caudate nucleus head weakens or completely blocks the sterotype movements when current intensity is subthreshold for behavioral arrest reaction. The pecularities of the caudate control its similarity to the action of haloperidol and the absence of influence of the stimulation of the capsula interna and some thalamic nuclei on the stereotypy lead to the assumption that it is due to the depression of the inhibitory function of the caudate nucleus brought about by the intensification of the nigro-striatal dopaminergic transmission.  相似文献

In experiments performed on 9 dogs with alimentary method, extinctive inhibition was deepened and its elaboration was aceelerated by electrical stimulation of somatosensory and motor cortical areas and ventral segment of the caudate nucleus head. The extinction of the conditioned reflex was slowed down by stimulation of the anterolateral gyrus and the central segment of the caudate nucleus head. General motor excitation of animals during stimulation of the dorsal zone of caudate nucleus head impaired the elaboration of extinctive inhibition.  相似文献

The influence of preliminary subthreshold activation of thalamic intralaminary nuclei on achievement of instrumental reflex and on inhibitory effects, caused by stimulation of the caudate nucleus head, were studied in chronic experiments on 5 dogs with a model of instrumental defensive conditioned reflexes, providing for maintainance of a given posture. It was shown that the preceding high-frequency electrostimulation of intralaminary nuclei activates motor components of the instrumental response (shortened latency, EMG and mechanogram of the response, increased amplitude of instrumental response) and significantly lowers threshold strength of the current, necessary for obtaining "caudate pause". The greatest increase in inhibitory influences, caused by stimulation of the caudate nucleus head, was observed when thalamic stimulation preceded stimulations, localized in the dorsolateral segment of the caudate nucleus head. The obtained data are discussed in aspect of Buchwald et al. hypothesis on the existence of "caudate loop".  相似文献

In unrestrained cats parachlorophenylalanine, an inhibitor of serotonin synthesis, increased the thresholds of the contraversive reaction and arrest reaction accompanied by spindle-waves in the sensory-motor cortex in the stimulation of the rostro-ventral areas of the caudate nucleus. The reactions evoked from the dorso-medial areas of the head and the body of the nucleus changed differently. On the contrary, 5-hydroxytryptophane, serotonin precursor, increased most of the indices of the caudate activity independently of the site of stimulation. It is suggested that the serotoninergic mechanisms (antagonistic to the dophaminergic system) existing in the ventral part of the caudate nucleus may trigger some caudate functions.  相似文献

Using models of electrical self-stimulation of the positive emotiogenic zones and stimulation of the negative emotiogenic zones of the hypothalamus in rats, we demonstrated that both these stimulations increase the noradrenaline level in the frontal cortex. This shows a nonspecific nature of activation of the dorsal noradrenergic bundle, resulting from motivational excitation. When the frequency of self-stimulation reaction remained stable, activation of the dorsal noradrenergic bundle was moderate, and at decay of the above reaction it returned to the control level. Behavior connected with activation of the motor functions was characterized by an increase in the dopamine and noradrenaline levels in the caudate nucleus. In theglobus pallidum, the dopamine content changed only under conditions of stimulation of the negative emotiogenic zones: these were an increase in the reaction of active avoidance and a decrease in passive avoidance.  相似文献

A conditioned food-procuring reaction previously elaborated to an acoustic stimulus was reproduced in chronic experiments on six cats by means of direct electrical stimulation of the posterior parts of the lateral hypothalamus. Folloiwng extensive bilateral electrolytic ablation of the caudate nucleus, conditioned food-procuring reaction to the stimulation of the hypothalamus could not be reproduced for 40 to 70 days. The conditioned foor-procuring reflex to the acoustic stimulus disappeared for 14 to 30 days to be subsequently spontaneously restored. After caudatotomy, a diminution of the average amplitude of background oscillations and of evoked potentials to acoustic stimuli was recorced in the examined zones of the lateral hypothalamus. The part played by the caudate nucleus in the processes of alimentary behaviour activation is discussed.  相似文献

Properties were studied of elaboration of conditioned alimentary secretory reflexes to threshold electric stimulation of the caudate nucleus in dogs. The rate of formation of such a reflex and its magnitude were considerably lower than in reflexes elaborated to distant stimuli. Latencies were longer; dynamics of conditioned and unconditioned secretory reactions was similar to that of reflexes to a weak stimulus--light. The motor component of the conditioned alimentary reaction, as a rule, was absent. Characteristics were revealed of formation of alimentary reflexes to stimulation of the dorsal and ventral parts of the head and body of the caudate nucleus.  相似文献

Adenosine plays an important role in neuromodulation and neuroprotection. Recent identification of transient changes in adenosine concentration suggests adenosine may have a rapid modulatory role; however, the extent of these changes throughout the brain is not well understood. In this report, transient changes in adenosine evoked by one second, 60 Hz electrical stimulation trains were compared in the caudate–putamen, nucleus accumbens, hippocampus, and cortex. The concentration of evoked adenosine varies between brain regions, but there is less variation in the duration of signaling. The highest concentration of adenosine was evoked in the dorsal caudate–putamen (0.34?±?0.08 μM), while the lowest concentration was in the secondary motor cortex (0.06?±?0.02 μM). In all brain regions, adenosine release was activity-dependent. In the nucleus accumbens, hippocampus, and prefrontal cortex, this release was partly due to extracellular ATP breakdown. However, in the caudate–putamen, release was not due to ATP metabolism but was ionotropic glutamate receptor-dependent. The results demonstrate that transient, activity-dependent adenosine can be evoked in many brain regions but that the mechanism of formation and release varies by region.  相似文献

During chronic experiments on unanesthetized cats neuronal response in the caudate nucleus to the presentation of local photic stimuli and electrical stimulation of the specific (field 17) and the association (Clare-Bishop) areas were compared. Stimulation of the Clare-Bishop area proved more effective than stimulating field 17 for neurons of the caudate nucleus; a response was produced in 47% of test neurons in comparison with 8% of units only in the specific area. Lower average values were observed for latency of neuronal response to stimulation of the Clare-Bishop area. An insignificant number of caudate nucleus neurons were activated as a result of stimulation of both cortical areas. A comparison between the response of one set of neurons to electrical cortical and visual stimulation showed that cells responding to visual stimulation were more highly activated by stimulating the Clare-Bishop area than by stimulation of field 17. This type of neuron predominated in the caudate nucleus. A discussion follows of the possible involvement of the Clare-Bishop area in shaping neuronal response to visual stimulation.A. A. Evoked potentials were recorded in the caudate nucleus of adult rabbits and young rabbits aged 2–30 days in response to stimulation of the ipsilateral motor cortex. The response of the caudate nucleus in the adult rabbit consisted of a positive-negative complex with latent period of 3–5 msec. Maximal amplitude of the response was observed in the dorsorostral region of the nucleus. As the recording electrode was inserted deeper, the amplitude of the response gradually decreased but without reversal of its polarity. Responses of the caudate nucleus to stimulation of the motor cortex were recorded as early as on the 3rd day after birth. These responses were indistinguishable in configuration from responses of the nucleus of adult rabbits. Their latent period was about 10 msec. Between the 16th and 20th day after birth the latent period of the response decreased considerably — from 9 to 5 msec, and by the 30th day of life it had reached its definitive value. With age the amplitude of the response increased but the threshold of stimulation decreased, The results indicate early functional maturation of connections of the motor cortex with the caudate nucleus and they agree with the results of morphological investigations of the structural development of the afferent systems of this nucleus.Brain Institute, Academy of Medical Sciences of the USSR, Moscow. Unitary responses of the caudate nucleus to stimulation of various parts of it were investigated by extracellular recording. Latent periods of response discharges varied from 3.5 to 40 msec. Most neurons were excited by stimulation of the most rostral part of the head of the caudate nucleus. Irrespective of the site of stimulation, in most cases responses consisted of initial excitation in the form of one or, less frequently, two discharges followed by a period of depression of spontaneous activity. Recovery of activity took place gradually, without postinhibitory facilitation. No afterdischarges or periodic repetitions of spikes were observed after the initial response. Repetitive stimulation of the caudate nucleus showed that the neurons of this nucleus reproduce frequencies of stimulation badly above 30/sec, and under these circumstances in many cases they continued to discharge on average at a frequency of 5–15/sec. The results are examined from the standpoint of participation of the caudate nucleus in the formation of spindle activity.A. A. Two models of postural tremor — induced by cold (shivering) and by injection of oxotremorine into the caudate nucleus (caudate tremor) — were compared in experiments on cats. Both models shared the same electromyographic picture of tremor; the frequency of activity of motor units working independently was 4–12 spikes/sec. Both types of tremor were potentiated by cold stimulation of temperature receptors and were inhibited by electrical stimulation of the medial preoptic region of the hypothalamus and by intravenous injection of cholinomimetics. Caudate tremor, unlike shivering, ceased after intravenous injection of scopolamine (an antiparkinsonism drug). On the basis of data on motoneuron function it is concluded that both types of tremor are analogs of physiological postural tremor. The selective inhibitory action of scopolamine on caudate tremor reflects only the mediator organization of structures in the caudate nucleus and cannot therefore serve as the basis for the conclusion that this disturbance of motor activity is a model of pathological postural tremor.O. Mechanisms of the amygdala central nucleus (CNA) influence on gastric motor reflex activity were studied in electrophysiological and neuroanatomical experiments in Wistar rats. In the anaesthetized animals, electrical stimulation of the CNA affected spontaneous gastric motility and caused inhibitory as well as excitatory changes of vagus-induced gastric relaxation. The most significant and mainly inhibitory effects were observed under the stimulation of the medial CNA. Microinjection of the anterograde tracer Phaseolus vulgaris-leucoagglutimn (PHA-L) into the different divisions of the CNA revealed direct projections from its dorso-medial portion to the gastric related area of the dorsal vagal complex. Electrical stimulation of this amygdaloid area was found to change activity of the bulbar gastric related neurons. Inhibitory and excitatory changes of their vagus-induced responses under the amigdala stimulation were manifested as a general modulation of all phases of the reaction or a selective modulation of some of them. These mechanisms may underlie the amygdalo-fugal modulation of gastric motor reflex activity.  相似文献

Weak electrical stimulation of emotiogenic brain structures was found to lead to the development of different general non-specific adaptation reactions. Stimulation of nucleus lateralis septi was found to evoke mainly the development of activation reaction, while stimulation of globus pallidum caused primarily the development of training reactions. Stress reactions were considerably less frequent in electrical stimulation of both kinds of structures than in the control. This can be explained by small values of the electrical current applied. Morphofunctional activity of thymus lymphatic system depended not only on the type of non-specific reaction but also on the character of the stimulated emotiogenic structure--the functional activity was higher with the stimulation of nucleus lateralis-septi. However, the functional activity of thymus lymphatic system in rats with globus pallidum stimulation was higher than in control rats that were not subject to any stimulation.  相似文献

Responses of 137 neurons of the rostral pole of the reticular and anterior ventral thalamic nuclei to electrical stimulation of the ventrolateral nucleus and motor cortex were studied in 17 cats immobilized with D-tubocurarine. The number of neurons responding antidromically to stimulation of the ventrolateral nucleus was 10.5% of all cells tested (latent period of response 0.7–3.0 msec), whereas to stimulation of the motor cortex it was 11.0% (latent period of response 0.4–4.0 msec). Neurons with a dividing axon, one branch of which terminated in the thalamic ventrolateral nuclei, the other in the motor cortex, were found. Orthodromic excitation was observed in 78.9% of neurons tested during stimulation of the ventrolateral nucleus and in 52.5% of neurons during stimulation of the motor cortex. Altogether 55.6% of cells responded to stimulation of the ventrolateral nucleus with a discharge of 3 to 20 action potentials with a frequency of 130–350 Hz. Similar discharges in response to stimulation of the motor cortex were observed in 30.5% of neurons tested. An inhibitory response was recorded in only 6.8% of cells. Convergence of influences from the thalamic ventrolateral nucleus and motor cortex was observed in 55.7% of neurons. The corticofugal influence of the motor cortex on responses arising in these cells to testing stimulation of the ventrolateral nucleus could be either inhibitory or facilitatory.A. A. 为了理解啮齿类动物的脑功能连接,本文利用9.4T fMRI获得轻度麻醉状态下大鼠静息状态及刺激激活的数据,通过互相关分析构建节点之间的相关系数矩阵并计算相应的网络参数.结果发现:给予前爪电刺激时,刺激对侧初级感觉皮层(S1)、丘脑(Tha)有较强的正激活,双侧尾状壳核(CPu)有较强的负激活.静息状态时大鼠感觉/运动皮层内部、丘脑内部的连接性较强,而感觉/运动皮层与丘脑之间的连接较弱,双侧感觉运动系统之间存在较强的同步低频振荡,感觉运动系统在静息态时的脑网络具有小世界属性.结果提示,啮齿类动物在大脑信息处理中的功能分离和整合可能与人类存在某些相似性,支持哺乳动物中枢神经系统的基本功能存在遗传保守性的观点.  相似文献

Abstract: The effects of (+)-amphetamine on carrier-mediated and electrically stimulated dopamine release were investigated using fast cyclic voltammetry in rat brain slices incorporating the nucleus accumbens, and in the caudate putamen. In the caudate putamen, dopamine release either increased with increasing frequency of local electrical stimulation (hot spots) or did not increase significantly (cold spots); dopamine release increased with increasing frequency of electrical stimulation in the nucleus accumbens. Local pressure application of (+)-amphetamine from a micropipette caused dopamine efflux at all sites examined, and this was not affected by sulpiride, indicating that efflux of dopamine caused by (+)-amphetamine is not regulated by dopamine D2 autoreceptors. (+)-Amphetamine reduced single-pulse electrically stimulated dopamine release at all sites; sulpiride reversed this decrease, indicating that endogenous dopamine released by (+)-amphetamine activates dopamine D2 autoreceptors. In nucleus accumbens and hot spots, (+)-amphetamine did not affect 20-pulse 50-Hz-stimulated dopamine release, whereas in cold spots it potentiated 20-pulse 50-Hz-stimulated dopamine release. We conclude that (+)-amphetamine modifies electrically stimulated dopamine release by uptake inhibition or by indirect activation of D2 autoreceptors; the precise mechanism is determined by site and duration of electrical stimulation.  相似文献

In view of evidence indicating that endogenous opiate-like peptides have epileptiform effects, we examined the effect of the opiate antagonist naloxone on the kindling of seizures produced by repeated electrical stimulation of the amygdala or the caudate nucleus in rats. Naloxone had no effect on the threshold for local after-discharge in the two areas and failed to retard the rate of kindling of clinical seizures. These results suggest that an interaction of opiate-like peptides with central opiate receptors does not play any critical role in the kindling of seizures.  相似文献

In cats with a preliminarily learned conditioned avoidance reaction, stimulation of the caudate nucleus inhibited behavioral response by lengthening its latency and reducing the number of conditioned reactions. Intensity of the inhibitory effect did not substantially depend on the localization of electrodes in the head or body of the nucleus and weakened within a few experimental days. D,1-amphetamine (0.5 to 4 mg/kg) and caffeine (10 to 10 mg/kg) suppressed the caudate inhibition, but the action of the drugs was manifested in different ways. The influence of large doses of d,1-amphetamine was characterized by a serious disturbance of behaviour and caudate inhibition of conditioned responses.  相似文献
