AIMS: To determine the influence of alternansucrase-derived oligosaccharides (AOS) and other carbohydrates on alpha-galactosidase and alpha-glucosidase activity in Bifidobacterium adolescentis. METHODS AND RESULTS: Activities for alpha-galactosidase and alpha-glucosidase were determined from cell extracts of B. adolescentis grown on 18 test carbohydrates including AOS. alpha-galactosidase activity was enhanced on a variety of alpha-linked or beta-linked carbohydrates regardless of a galactoside or glucoside. alpha-glucosidase, however, was enhanced only on alpha-linked carbohydrates. AOS significantly enhanced enzyme activity compared with most of the carbohydrates tested. Most of the AOS showed significant increases in activity for both enzymes over that displayed by their corresponding acceptor carbohydrates. CONCLUSIONS: alpha-galactosidase may serve as a biomarker for microbial metabolic activity within the large intestine for potential prebiotics composed of alpha-linked or beta-linked oligosaccharides whereas alpha-glucosidase activity may be restricted to assessing the influence of only alpha-linked carbohydrates. The AOS synthesis process provided a value-added component to carbohydrates by increasing metabolic activity (via alpha-galactosidase and alpha-glucosidase) over certain acceptor carbohydrates. SIGNIFICANCE AND IMPACT OF THE STUDY: Fundamental knowledge of enzyme activity in Bifidobacterium may aid in the design of more effective prebiotics and may also help identify enzyme indicators of metabolic activity when assessing influence within the intestine.  相似文献

There is little doubt that carbohydrates are crucial to the functions of NK cells. Exogenous carbohydrates alter the cytolytic capabilities of these cells, treatment of NK cells with glycosidases or inhibitors of glycosylation affect function, NK cells can bind selective sugars, and NK cells can be identified and subdivided into functionally distinct subsets on the basis of cell surface carbohydrates. Yet, despite the large number of observations which have been made concerning carbohydrates and NK cells, there is little consensus regarding these studies and few investigations which satisfactorily demonstrate specific mechanisms for the observed effect of the carbohydrates. Almost certainly, the number of diverse observations implies roles for carbohydrates in multiple areas of NK function, probably including target recognition, tissue distribution and post-binding events in the lytic cascade. Hopefully, these observations made to date will be viewed as exciting preliminary studies which will entice more sophisticated investigations designed to elucidate the precise roles of carbohydrates in the functions of NK cells.  相似文献

The number of roots formed in cuttings of pea ( Pisum sativum L. cv. Alaska) was regulated both by the temperature and by IAA, whereas the time to the appearance of the first roots was regulated only by the temperature. Cuttings treated with 10-3 M IAA had a smaller content of extractable carbohydrates than the control ones irrespective of the temperature. In the bases of cuttings rooted at 25°C the content of extractable carbohydrates was lower than in those rooted at 15°C. Cuttings treated with IAA showed up to elevenfold increase of extractable carbohydrates in the bases at day 3. This increase of soluble sugars was not correlated with the number of roots formed or the speed of rooting. It is concluded that IAA affects the accumulation of carbohydrates, and this is not connected with the rooting ability of the cuttings.  相似文献

The effect of the bacterium Azospirillum brasilense jointly immobilized with Chlorella vulgaris or C. sorokiniana in alginate beads on total carbohydrates and starch was studied under dark and heterotrophic conditions for 144h in synthetic growth medium supplemented with either d-glucose or Na-acetate as carbon sources. In all treatments, enhanced total carbohydrates and starch content per culture and per cell was obtained after 24h; only jointly immobilized C. vulgaris growing on d-glucose significantly increased total carbohydrates and starch content after 96h. Enhanced accumulation of carbohydrate and starch under jointly immobilized conditions was variable with time of sampling and substrate used. Similar results occurred when the microalgae was immobilized alone. In both microalgae growing on either carbon sources, the bacterium promoted accumulation of carbohydrates and starch; when the microalgae were immobilized alone, they used the carbon sources for cell multiplication. In jointly immobilized conditions with Chlorella spp., affinity to carbon source and volumetric productivity and yield were higher than when Chlorella spp. were immobilized alone; however, the growth rate was higher in microalgae immobilized alone. This study demonstrates that under heterotrophic conditions, A. brasilense promotes the accumulation of carbohydrates in two strains Chlorella spp. under certain time-substrate combinations, producing mainly starch. As such, this bacterium is a biological factor that can change the composition of compounds in microalgae in dark, heterotrophic conditions.  相似文献

Myelin-associated carbohydrates were studied by means of histochemical techniques in the central nervous system of birds and mammals. Polianions in the surface of myelin and in interfascicular oligodendroglia were detected using histochemical techniques. Glycoproteins were studied by means of concanavalin A. The Con-A-PO-DAB sequence was used. Concanavalin-A-binding sites were detected in oligodendroglia and on the myelin surface. Similar results were observed in both birds and mammals. The processes of the interfascicular oligodendroglia also contain carbohydrates. A close association between the carbohydrates of these glial processes and the myelin surface carbohydrates was demonstrated, and their probable identity is assumed.  相似文献

Rabbit follicular oocytes were cultured at 37 °C for 18–24 h in a basic salt medium containing 0.4% bovine serum albumin (BSA), carbohydrates and amino acids in various combinations. Osmolarity of the medium was maintained at 308 mOsm. The carbohydrates, pyruvate, lactate and glucose were all about equally beneficial, but not essential for rabbit oocyte maturation. Glutamine and proline, but not methionine or phenylalanine stimulated oocyte development. Glutamine stimulated more follicular oocytes to develop to prophase and metaphase II than did any of the three carbohydrates tested alone or in combination. Ammonia production after 24 h of culture was highest in media containing glutamine (15.2 μg/ml) but this was not inhibitory to maturation. Negligible amounts of ammonia were found with the other amino acids added. With 0, 0.08, 0.4, 2, 10 and 50 mM of glutamine in the basic salt medium, plus 0.4 % BSA, but without carbohydrates, 30, 73, 70, 71, 59 and 45 % of the follicular oocytes developed to the prophase or metaphase II stage. It is concluded that the optimum level of glutamine ranged from 0.08 to 2 mM and that no carbohydrate need be added to the medium for culturing oocytes when glutamine is included.  相似文献

In testis tissues from patients with idiopathic male infertility, complex carbohydrates have been studied by means of light microscopic histochemical methods, comparisons being made with the normal testes. In the infertile testes, histochemical reactions for acidic and neutral complex carbohydrates were in general weaker in intensity than in the normal testes. In the seminiferous tubules and interstitium of the infertile testes, a histochemical finding of primary importance was obtained with the demonstration of galactose deficiency in the complex carbohydrates involved. These glycoproteins contained only small numbers of sulfhydryl and disulfide groups. The possible histophysiological significance of these complex carbohydrates is discussed, with special reference to their histochemical properties determined in the present study.  相似文献

Diatoms are abundant in biofilms developed on surfaces immersed in sunlit waters. In both the planktonic and the biofilm mode of growth, diatoms produce carbohydrate polymers which perform several functions including motility, protection, production of macro-aggregates and detoxification. However, little is known about the differences, if any, in the production and characterization at the molecular level of carbohydrates in planktonic and biofilm cells. In order to identify the differences in these two modes of growth, the concentration of total carbohydrates, carbohydrate fractions, neutral carbohydrates, uronic acids and amino sugars in planktonic and biofilm cells of Amphora rostrata were measured. The results showed that the distribution of carbohydrate fractions, uronic acids and amino sugars was different in biofilm and planktonic cells. Cell normalized concentrations of these components were two to five times greater in planktonic cells compared with biofilm cells. The concentrations of glucose and glucosamine decreased, whereas fucose increased in planktonic cells over the period of cultivation. Conversely, the concentrations of glucose and glucosamine increased while that of fucose decreased in attached cells. The study suggests that marked differences exist between the carbohydrates of the planktonic and the biofilm cells of A. rostrata.  相似文献

The rate of turnover of membrane proteins and membrane-bound carbohydrates in exponentially growing and in confluent contact-inhibited cultures of strain MK-2 cells was investigated. Cells were labelled with [14-C]leucine and [3-H]glucosamine, incubated in isotope-free medium and, at various times thereafter, the cells were harvested and membranes isolated from them. The rate of decay of the protein and carbohydrate components was determined from specific activity dilution of the labeled components in the isolated membranes. Although the rate of membrane synthesis is different in exponential and contact-inhibited cells, the rate of degradation (turnover) of membrane proteins and carbohydrates was found to be the same (25% per generation (42 h) or 0.6%/h).  相似文献

Ligands for the leukocyte adhesion molecule L-selectin are expressed not only in lymph node high endothelial venules (HEV) but also in the renal distal tubuli. Here we report that L-selectin-reactive molecules in the kidney are chondroitin sulfate and heparan sulfate proteoglycans of 500-1000 kDa, unlike those in HEV bearing sialyl Lewis X-like carbohydrates. Binding of L-selectin to these molecules was mediated by the lectin domain of L-selectin and required divalent cations. Binding was inhibited by chondroitinase and/or heparitinase but not sialidase. Thus, L-selectin can recognize chondroitin sulfate and heparan sulfate glycosaminoglycans structurally distinct from sialyl Lewis X-like carbohydrates.  相似文献

In 2 two‐factorial experiments, each conducted on 80 growing male rats, the effects of substituting 10% raw potato starch (PS), pectins (PEC), or cellulose (CEL) for wheat starch (WS) and the addition of tannic acid to WS (WSTA) were studied using diets differing in protein quality. Casein unsupplemented or supplemented with DL‐methionine and gluten unsupplemented or supplemented with lysine, methionine and tryptophan were used as protein sources in Experiment 1 and 2, respectively. Parameters indicative of caecal fermentation intensity (pH, acetic, propionic and butyric acid contents, digesta and tissue weight) and of protein metabolism (urea blood concentration, faecal and urinary nitrogen excretion) were determined. Ten‐day balance experiments were preceded by a 10‐day adaption period to respective carbohydrates given in a diet containing balanced protein.

In both experiments the type of carbohydrates affected the caecal concentration of individual and total SCFA and other parameters of fermentation intensity. Pectins and potato starch were fermented more intensively than cellulose. Faecal N excretion was increased by all carbohydrates substituted for cereal starch, and by tannic acid. Urinary excretion was greater on CEL than on PEC and WSTA containing casein and on other diets containing gluten. In both experiments urinary N excretion was the lowest on PEC diets. Protein quality had the greatest effect on apparent biological value and net protein utilization but all indices of protein utilization were also affected by carbohydrates. It is concluded that not only the amount of N excreted in faeces but also in urine is affected by the type and fermentability of carbohydrates.  相似文献

Most strains of Escherichia coli K-12 lacking the enzyme adenylate cyclase showed normal chemotaxis toward carbohydrates taken up and phosphorylated by the phosphoenolpyruvate-dependent carbohydrate: phosphotransferase system. The normal reaction was observed even in the absence of externally added cyclic adenosine 3',5'-phosphate, provided that the enzyme II chemoreceptors and the flagella were synthesized. In the CA8306 series of strains, however, the cya-854 deletion abolished chemotaxis toward phosphotransferase system carbohydrates even though growth on and transport of these carbohydrates were not affected. This abnormal phenotype was due to the presence of a specific mutation in strain CA8306 which mapped in or close to the crp locus and apparently prevented expression of a hitherto unidentified molecule involved in enzyme II-mediated signal transduction. This molecule is neither a pts protein nor a cyclic adenosine 3',5'-phosphate-binding protein.  相似文献

Recent ethnobotanical exploration of henequen (Agave fourcroydes) in the Peninsula of Yucatan, Mexico, finds that inflorescence peduncles are used as emergency food and in the preparation of a fermented drink. Bromatological analysis and determination of total carbohydrates were made for the two length classes (ca. 3.30 m and ca. 0.60 m) which are consumed. The analysis of both the cultivated plant and its putative wild ancestor (Agave angustifolia) suggests that utilization of the inflorescence peduncles as food may have been involved in the initial stages of the history of its evolution under artificial selection, because the wild and the cultivated plants have similar palatability. The subsequent agricultural prevalence of annual crop species in the region was possibly responsible for the abandonment of henequen in the local diet. No significant differences are observed between the bromatological and total carbohydrate values of domesticated and wild plants. The preference for small inflorescence peduncles as a vegetable is a consequence of its significantly minor content of raw fiber and its larger content of total carbohydrates. As a fermented drink, longer peduncles are preferred because they provide more substrate material and because fiber can be eliminated by filtering. This agricultural byproduct, almost totally wasted, has potential value as a source of carbohydrates and raw fiber.  相似文献

In the intercellular matrix of the granulosa layer of the mouse ovarian follicles, ultrastructural features of acidic complex carbohydrates have been studied by means of dialyzed iron (DI) staining in combination with procedures of digestion with Streptomyces and testicular hyaluronidases. In the intercellular matrix, DI reactive structures containing acidic complex carbohydrates consist of layers of a variable thickness coating the plasma membrane of the granulosa cells and reticular elements distributed in the spaces between the cells. The latter exists in two appearances; one is clumped masses of irregular shapes and different sizes, whereas the other being filamentous figures radiating from the masses. The effects of digestion with Streptomyces and testicular hyaluronidases upon the DI staining of the tissues indicate that the DI reactive structures in the intercellular matrix contain at least three types of acidic complex carbohydrates; hyaluronic acid, isomeric chondroitin sulfates and other acidic glycosaminoglycans. The histophysiological activities played by these particular complex carbohydrates have been briefly discussed.  相似文献

The rumen holotrich protozoa Isotricha intestinalis and I. prostoma showed chemotaxis to sucrose, glucose and fructose. They attached themselves, by means of an organelle on the anterior cell surface, to particulate sources of these carbohydrates provided soluble protein was present in the medium. The concentration of protein eliciting attachment varied with the species and the state of nutrition of the cell, but was between 20 and 150 microgram ml-1. Attachment occurred only if the concentration of carbohydrate, at its source, exceeded the chemotaxis threshold concentration (50 micrometer for sucrose) and if it was less than 1 mM. At concentrations exceeding 1 mM, indiscriminate attachment to gas-liquid and solid-liquid interfaces occurred, provided the protein concentration was high enough to elicit attachment. In the rumen, soluble carbohydrates diffusing from food particles may attract the protozoa which attach themselves to the particles in the presence of soluble plant protein at less than 20 microgram ml-1; these conditions exist in the host animal soon after feeding when fed infrequently. The attachment mechanism may confer an ecological advantage on the Isotricha spp. over other rumen organisms dependent on soluble carbohydrates as energy and carbon sources.  相似文献

Determinations were made of carbohydrates in hemolymph collected from adult female mosquitoes (Anopheles stephensi). First the hemolymph was fractionated by extraction and precipitation procedures, after which qualitative and quantitative determinations of carbohydrates were made by thin layer chromatography. The most abundant sugars found in the hemolymph were glucose and trehalose, though maltose, glucuronic acid, and inositol could be found after the mosquitoes took blood meals. After the mosquitoes ingested a noninfected blood meal, their hemolymph sugar levels rose almost 4-fold. There was less of an increase following a blood meal infected with the rodent malaria parasite, Plasmodium berghei. Depletion of sugars in the hemolymph of infected mosquitoes may result from direct utilization of sugar by the malaria parasite developing within the mosquito.  相似文献

The production of glycoproteins with carbohydrates of defined structure and minimal heterogeneity is important for functional studies of mammalian carbohydrates. To facilitate such studies, several Chinese hamster ovary mutants that carry between two and four glycosylation mutations were developed. All of the lines grew readily in culture despite the drastic simplification of their surface carbohydrates. Therefore, both endogenous glycoproteins and those introduced by transfection can be obtained with specifically tailored carbohydrates. The lectin resistance properties of the mutants showed that each line expresses a novel array of cell surface carbohydrates useful for identifying specific roles for carbohydrates in cellular interactions. In addition, they showed that the epistatic relationships among different glycosylation mutations are not entirely predictable, providing insight into the complexity of the carbohydrate structures at the Chinese hamster ovary cell surface.  相似文献

Each perennial woody plant is a highly integrated system of competing carbohydrate sinks (utilization sites). Internal competition for carbohydrates is shown by changes in rates of carbohydrate movement from sources to sinks and reversals in direction of carbohydrate transport as the relative sink strengths of various organs change. Most carbohydrates are produced in foliage leaves but some are synthesized in cotyledons, hypocotyls, buds, twigs, stems, flowers, fruits, and strobili. Although the bulk of the carbohydrate pool moves to sinks through the phloem, some carbohydrates are obtained by sinks from the xylem sap. Sugars are actively accumulated in the phloem and move passively to sinks along a concentration gradient. The dry weight of a mature woody plant represents only a small proportion of the photosynthate it produced. This discrepancy results not only from consumption of plant tissues by herbivores and shedding of plant parts, but also from depletion of carbohydrates by respiration, leaching, exudation, secretion, translocation to other plants through root grafts and mycorrhizae and losses to parasites. Large spatial and temporal variations occur in the use of reserve- and currently produced carbohydrates in metabolism and growth of shoots, stems, roots, and reproductive structures. A portion of the carbohydrate pool is diverted for production of chemicals involved in defense against fungi, herbivores, and competing plants. Woody plants accumulate carbohydrates during periods of excess production and deplete carbohydrates when the rate of utilization exceeds the rate of production. Stored carbohydrates play an important role in metabolism, growth, defense, cold hardiness, and postponement or prevention of plant mortality.  相似文献
