Inhibition of photosynthetic electron transport in isolated chloroplasts by lead salts has been demonstrated. Photosystem I activity, as measured by electron transfer from dichlorophenol indophenol to methylviologen, was not reduced by such treatment. However, photosystem II was inhibited by lead salts when electron flow was measured from water to methylviologen and Hill reaction or by chlorophyll fluorescence. Fluorescence induction curves indicated the primary site of inhibition was on the oxidizing side of photosystem II. That this site was between the primary electron donor of photosystem II and the site of water oxidation could be demonstrated by hydroxylamine restoration of normal fluorescence following lead inhibition.  相似文献

Irradiation of tris-washed chloroplasts with moderate intensities of red light caused a partial bleaching of chloroplast pigments and an inhibition of the hydroquinone-supported photoreduction of NADP. The presence of an electron donor for photosystem 2 (PS2) during the irradiation prevented the bleaching and inhibition. It is concluded that the strong oxidant produced by PS2 accumulates in tris-washed chloroplasts during irradiation and an electron donor for PS2 protects against the photooxidation reactions.  相似文献

During protein import into chloroplasts, one of the Hsp70 proteins in pea (Hsp70-IAP), previously reported to localize in the intermembrane space of chloroplasts, was found to interact with the translocating precursor protein but the gene for Hsp70-IAP has not been identified yet. In an attempt to identify the Arabidopsis homolog of Hsp70-IAP, we employed an in vitro protein import assay to determine the localization of three Arabidopsis Hsp70 homologs (AtHsp70-6 through 8), predicted for chloroplast targeting. AtHsp70-6 and AtHsp70-7 were imported into chloroplasts and processed into similar-sized mature forms. In addition, a smaller-sized processed form of AtHsp70-6 was observed. All the processed forms of both AtHsp70 proteins were localized in the stroma. Organelle-free processing assays revealed that the larger processed forms of both AtHsp70-6 and AtHsp70-7 were cleaved by stromal processing peptidase, whereas the smaller processed form of AtHsp70-6 was produced by an unspecified peptidase.  相似文献

The translocons at the outer envelope membrane of chloroplasts (TOCs) initiate the import of thousands of nucleus-encoded proteins into the organelle. The identification of structurally and functionally distinct TOC complexes has led to the hypothesis that the translocons constitute different import pathways that are required to coordinate the import of sets of proteins whose expression varies in response to organelle biogenesis and physiological adaptation. To test this hypothesis, we examined the molecular basis for distinct TOC pathways by analyzing the functional diversification among the Toc159 family of TOC receptors. We demonstrate that the N-terminal A-domains of the Toc159 receptors regulate their selectivity for preprotein binding. Furthermore, the in vivo function of the two major Toc159 family members (atToc159 and atToc132) can be largely switched by swapping their A-domains in transgenic Arabidopsis thaliana. On the basis of these results, we propose that the A-domains of the Toc159 receptors are major determinants of distinct pathways for protein import into chloroplasts.  相似文献

Ferricyanide can be reduced in Photosystem II of spinach chloroplasts at 2 separate sites, both of which are sensitive to 3-(3,4-dichlorophenyl)-1,1-dimethylurea, but only one of which is sensitive to dibromothymoquinone. Data presented in this paper emphasize ferricyanide site II of Photosystem II, which is sensitive to thiol inhibition and may reflect a cyclic pathway around Photosystem II. Ferricyanide reduction sites 1 and 2 also differ from each other in fractions isolated from discontinuous sucrose gradients, from fragmented chloroplasts, and upon trypsin treatment. Sucrose density gradient centrifugation shows that ferricyanide reduction site 1 activity at pH 6 decreases from 30 to 50% in various isolated fractions, while the dibromothymoquinone-insensitive activity at pH 8 (site 2) is stimulated from 15 to 35%.  相似文献

We have investigated the interactions of two nuclear-encoded preproteins with the chloroplast protein import machinery at three stages in import using a label-transfer crosslinking approach. During energy-independent binding at the outer envelope membrane, preproteins interact with three known components of the outer membrane translocon complex, Toc34, Toc75, and Toc86. Although Toc75 and Toc86 are known to associate with preproteins during import, a role for Toc34 in preprotein binding previously had not been observed. The interaction of Toc34 with preproteins is regulated by the binding, but not hydrolysis of GTP. These data provide the first evidence for a direct role for Toc34 in import, and provide insights into the function of GTP as a regulator of preprotein recognition. Toc75 and Toc86 are the major targets of cross-linking upon insertion of preproteins across the outer envelope membrane, supporting the proposal that both proteins function in translocation at the outer membrane as well as preprotein recognition. The inner membrane proteins, Tic(21) and Tic22, and a previously unidentified protein of 14 kD are the major targets of crosslinking during the late stages in import. These data provide additional support for the roles of these components during protein translocation across the inner membrane. Our results suggest a defined sequence of molecular interactions that result in the transport of nuclear-encoded preproteins from the cytoplasm into the stroma of chloroplasts.  相似文献

After acid-treatment of spinach (Spinacia oleracea) chloroplasts, various partial electron transport reactions are inactivated from 25 to 75%. Divalent cations in concentrations from 10 to 50 millimolar can partially restore electron transport rates. Two cation-specific sites have been found in photosystem II: one on the 3-(3,4-dichlorophenyl)-1, 1-dimethylurea-insensitive silicomolybdate pathway, which responds better to restoration by Mg2+ than by Ca2+ ions, the other on the forward pathway to photosystem I, located on the 2,5-dimethylbenzoquinone pathway. This site is selectively restored by Ca2+ ions. When protonated chloroplasts are treated with N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)aziridine, a carboxyl group modifying reagent, presumed to react with glutamic and aspartic acid residues of proteins, restoration of electron transport at the Ca2+-selective site on the 2,5-dimethylbenzoquinone pathway is impaired, while no difference in restoration is seen at the Mg2+ site on the 3-(3,4-dichlorophenyl)-1,1-dimethylurea-insensitive silicomolybdate pathway.

Trypsin treatment of chloroplasts modifies the light-harvesting pigment-protein complex, destroys the dibromothymoquinone-insensitive 2,5-dimethyl-benzoquinone reduction, but does not interfere with the partial restoration of activity of this pathway by Ca2+ ions, implying that the selective Ca2+ effect on photosystem II (selective Ca2+ site) is different from its effects as a divalent cation on the light-harvesting pigment-protein complex involved in the excitation energy distribution between the two photosystems.

Synthesis and assembly of photosystems (PS) I and II polypeptides in etiochloroplasts isolated from greening wheat (Triticum aestivum L. cv Norin 61) seedlings were studied. The isolated etiochloroplasts synthesized PSI polypeptides of 66 and 15 kilodaltons, PSII polypeptides of 46 and 42 kilodaltons, and atrazine-binding 34 to 32 kilodalton polypeptide. Their assembly processes in the thylakoid membrane were studied by pulse-chase labeling with [35S]methionine, mild solubilization of the thylakoid membrane with Triton X-100, sucrose density gradient centrifugation, and polyacrylamide gel electrophoresis. The newly synthesized polypeptides of 66, 46, 42, 34, and 32 kilodaltons were first integrated into the complexes of 7.5, 5.9, 7.5, 6.3, and 7.5 Svedberg units, respectively, in 20 minutes. After the chase with excess amount of methionine for 100 min, they were found in complexes of 9.5, 9.1, 9.1, 9.1, and 9.1 Svedberg units, respectively. In this condition, stained polypeptides of PSI and PSII were found in the complexes of 11.1 and 10.3 Svedberg units, respectively. These results indicated that newly synthesized PSI or PSII polypeptides are integrated into intermediate complexes, but not complete complexes in the isolated etiochloroplasts. The relationship between the processing of the atrazine-binding 32 kilodalton polypeptide and its assembly into the PSII complex is also discussed.  相似文献

The post-translational import of nucleus-encoded preproteins into chloroplasts occurs through multimeric translocons in the outer (Toc) and inner (Tic) membranes. The high fidelity of the protein import process is maintained by specific recognition of the transit peptide of preproteins by the coordinate activities of two homologous GTPase Toc receptors, Toc34 and Toc159. Structural and biochemical studies suggest that dimerization of the Toc receptors functions as a component of the mechanism to control access of preproteins to the membrane translocation channel of the translocon. We show that specific mutations that disrupted receptor dimerization in vitro reduced the rate of protein import in transgenic Arabidopsis compared with the wild type receptor. The mutations did not affect the GTPase activities of the receptors. Interestingly, these mutations did not decrease the initial preprotein binding at the receptors, but they reduced the efficiency of the transition from preprotein binding to membrane translocation. These data indicate that dimerization of receptors has a direct role in protein import and support a hypothesis in which receptor-receptor interactions participate in the initiation of membrane translocation of chloroplast preproteins as part of the molecular mechanism of GTP-regulated protein import.  相似文献

Cadmium ions, as an environmental pollution factor, significantly inhibited the photosynthesis especially, photosystem Ⅱ activity in isolated spinach chloroplasts. The presence of 5 mmol/l Cd2+ inhibited the O2-evolution to 53%. Cd2+ reduced the activity of photoreduction of DCIP and the variable fluorescence of chloroplasts and PSⅡ preparation. The inhibited DCIP photoreduction activity could only be restored slightly by the addition of an artificial electron donor of PSII, DPC, and the inhibited variable fluorescence could not be obviously recovered by the addition of NH2OH, another artificial electron donor of PSⅡ. It is considered that, besides the oxidizing side of PSI1, Cd2+ could also inhibit directly the PSⅡ reaction center. The inhibitory effect of Cd2+ on the whole chain electron transport (H2O→MV) was more serious than on O2-evolution (H2O→DCMU). It is suggested that the oxidizing side of PSⅡ is not the only site for Cd2+ action. There may be another site inhibited by Cd2+ in the electron transport chain between PSⅠ and PSⅡ.  相似文献

The room temperature fluorescence induction of chloroplasts was utilized as a probe to locate the site of inhibition on PSII by copper. It was found that, while the initial fluorescence yield was hardly affected, the variable fluorescence yield was lowered without significant change in its kinetics. Addition of DCMU, or abolishing oxygen evolution capability by Tris treatment, did not alter this basic inhibition pattern. Copper was also found to lower the fluorescence yield of chloroplasts treated with linolenic acid which inhibited the secondary electron transport on both oxidizing and reducing sides of PSII. The data indicate that copper adversely affects the primary charge separation at the PSII reaction center. We suggest that the inhibition is due to creation of a lesion close to the reaction center, leading to increased dissipation of incoming excitation energy to heat.  相似文献

Heat shock protein 70s (Hsp70s) are encoded by a multigene family and are located in different cellular compartments. They have broad-ranging functions, including involvement in protein trafficking, prevention of protein aggregation, and assistance in protein folding. Hsp70s work together with their cochaperones, J domain proteins and nucleotide exchange factors (e.g., GrpEs), in a functional cycle of substrate binding and release accompanied by ATP hydrolysis. We have taken advantage of the gene targeting capability of the moss Physcomitrella patens to investigate the functions of chloroplast Hsp70s. We identified four Hsp70 genes and two GrpE cochaperone homolog genes (CGE) in moss that encode chloroplast proteins. Disruption of one of the Hsp70 genes, that for Hsp70-2, caused lethality, and protein import into heat-shocked chloroplasts isolated from temperature-sensitive hsp70-2 mutants was appreciably impaired. Whereas the double cge null mutant was not viable, we recovered a cge1 null/cge2 knock down mutant in which Hsp70-2 was upregulated. Chloroplasts isolated from this mutant demonstrated a defect in protein import. In addition, two different precursors staged as early import intermediates could be immunoprecipitated with an Hsp70-2–specific antibody. This immunoprecipitate also contained Hsp93 and Tic40, indicating that it represents a precursor still in the Toc/Tic translocon. Together, these data indicate that a stromal Hsp70 system plays a crucial role in protein import into chloroplasts.  相似文献

Under anaerobic atmosphere where the gas phase was simply replacedby N2, photo-inhibition of PS I of isolated spinach chloroplastswas insignificant. However, when dithionite was included inthe irradiation mixture, severe photoinhibition of the NADP+and the MV photo-reduction occurred. Neither P700 determinedby continuous illumination-induced difference spectroscopy,Fe-S centers determined by EPR under cryogenic temperatures,nor vitamin K-l determined by HPLC analysis were significantlydecreased under these photoinhibition conditions. Although photobleachingof antenna chlorophylls occurred to more or less extent, NADP+photoreduction activities were markedly depressed even undersaturating actinic light. The maximal amplitude of the flashinduced absorbance changes of P700 in ms range decreased almostin parallel with the loss of NADP+ photoreduction activity.These results indicate that although the Fe-S centers of thephotoinhibited chloroplasts were reducible by continuous illumination,to almost the same extents as that of the control chloroplasts,the efficiency of reduction by each flash was much lower thanthat of the control chloroplasts. The photosystem II core complex (TSF-IIa) is composed of polypeptides of molecular weight 54-, 47-, 42-, and 30 kilodaltons (kD) and cytochrome b-559. After treatment with trypsin or α-chymotrypsin for 20 hours, the TSF-IIa particles still retained their photochemical activity and the light-induced cytochrome b-559 signal, although all of the polypeptides of the complexes, except the 30 kD unit were extensively degraded. Proteolytic treatment decreased the apparent molecular weight of the complex from 250,000 to 100,000 daltons as determined by gel filtration, and also decreased the protein to chlorophyll ratio by 40%. Chlorophyll a appeared to be associated with the 47- and 42 kD polypeptides. Proteolysis of the complex produced a single chlorophyll a band with a slightly higher electrophoretic mobility. This band was not equivalent to the 30 kD polypeptide. Proteolysis also reduced the sensitivity of the TSF-IIa particles to 3-(3′,4′-dichlorophenyl)-1,1-dimethylurea (DCMU), but did not completely abolish it.  相似文献

The mitochondrion is arguably the most complex organelle in the budding yeast cell cytoplasm. It is essential for viability as well as respiratory growth. Its innermost aqueous compartment, the matrix, is bounded by the highly structured inner membrane, which in turn is bounded by the intermembrane space and the outer membrane. Approximately 1000 proteins are present in these organelles, of which eight major constituents are coded and synthesized in the matrix. The import of mitochondrial proteins synthesized in the cytoplasm, and their direction to the correct soluble compartments, correct membranes, and correct membrane surfaces/topologies, involves multiple pathways and macromolecular machines. The targeting of some, but not all, cytoplasmically synthesized mitochondrial proteins begins with translation of messenger RNAs localized to the organelle. Most proteins then pass through the translocase of the outer membrane to the intermembrane space, where divergent pathways sort them to the outer membrane, inner membrane, and matrix or trap them in the intermembrane space. Roughly 25% of mitochondrial proteins participate in maintenance or expression of the organellar genome at the inner surface of the inner membrane, providing 7 membrane proteins whose synthesis nucleates the assembly of three respiratory complexes.TO think about how mitochondrial proteins are synthesized, imported, and assembled, it is useful to have a clear picture of the organellar structures that they, along with membrane lipids, compose and the functions that they carry out. As almost every schoolchild learns, mitochondria carry out oxidative phosphorylation, the controlled burning of nutrients coupled to ATP synthesis. Since Saccharomyces cerevisiae prefers to ferment sugars, respiration is a dispensable function and nonrespiring mutants are viable [although they cannot undergo meiosis (Jambhekar and Amon 2008)]. However, mitochondria themselves are not dispensable. A substantial fraction of intermediary metabolism occurs in mitochondria (Strathern et al. 1982), and at least one of these pathways, iron–sulfur cluster assembly, is essential for growth (Kispal et al. 2005). Thus, any mutation that prevents the biogenesis of mitochondria by, for example, preventing the import of protein constituents from the cytoplasm, is lethal (Baker and Schatz 1991).The mitochondria of S. cerevisiae are tubular structures at the cell cortex. While the number of distinct compartments can range from 1 to ∼50 depending upon conditions (Stevens 1981; Pon and Schatz 1991), continual fusion and fission events among them effectively form a single dynamic network (Nunnari et al. 1997). The outer membrane surrounds the tubules. The inner membrane has a boundary domain closely juxtaposed beneath the outer membrane and cristae domains that project internally from the boundary into the matrix (Figure 1A). The matrix is the aqueous compartment surrounded by the inner membrane. However, electron tomography of mitochondria from several species, including yeast, shows that cristae actually emanate from the boundary membrane as narrow tubular structures at sites termed “crista junctions” and expand as they project into the matrix (Frey and Mannella 2000; Mannella et al. 2001) (Figure 1B). It seems clear that the boundary and cristae domains of the inner membrane have distinct compositions with respect to the respiratory complexes that are embedded preferentially in the cristae membrane domains, as well as other components (Vogel et al. 2006; Wurm and Jakobs 2006; Rabl et al. 2009; Suppanz et al. 2009; Zick et al. 2009; Davies et al. 2011).The outer and inner boundary membranes are connected at multiple contact sites, at least some of which are involved in protein translocation and may be transient (Pon and Schatz 1991). In addition, there appear to be firm contact sites, not directly involved with protein translocation, preferentially colocalized with crista junctions (Harner et al. 2011a).Overall, there appear to be ∼1000 distinct proteins in yeast mitochondria (Premsler et al. 2009). One series of proteomic studies on highly purified organelles identified 851 proteins thought to represent 85% of the total number of species (Sickmann et al. 2003; Reinders et al. 2006; Zahedi et al. 2006). Another study identified an additional 209 candidates (Prokisch et al. 2004). A computationally driven search for candidates involved in yeast mitochondrial function, coupled with experiments to assay respiratory function and maintenance of mitochondrial DNA (mtDNA), identified 109 novel candidates, although many of these may not be mitochondrial per se (Hess et al. 2009). Taking the boundary and cristae domains together, the inner membrane is the most protein-rich mitochondrial compartment, followed by the matrix (Daum et al. 1982).Only eight of the yeast mitochondrial proteins detected in proteomic studies are encoded by mtDNA and synthesized within the organelle. They are hydrophobic subunits of respiratory complexes III (bc1 complex or ubiquinol-cytochrome c reductase), IV (cytochrome c oxidase), and V (ATP synthase), as well as a hydrophilic mitochondrial small subunit ribosomal protein. The remaining ∼99% of yeast mitochondrial proteins are encoded by nuclear genes, synthesized in cytoplasmic ribosomes, and imported into the organelle.An overview of known nuclearly encoded mitochondrial protein functions (Figure 2) reveals that ∼25% of them are involved directly in genome maintenance and expression of the eight major mitochondrial genes (Schmidt et al. 2010). The functions of ∼20% of the proteins are not known. Fifteen percent are involved in the well-known processes of energy metabolism. High rates of light-dependent fatty acid synthesis from acetate were measured in isolated chloroplasts that were permeabilized to varying extents by resuspension in hypotonic reaction medium. The reactions in hypotonic medium unsupplemented with cofactors were linear with time and were directly proportional to chlorophyll concentration, suggesting that the enzymes and cofactors of fatty acid synthesis remained tightly integrated and thylakoid associated within disrupted chloroplasts. Permeabilized chloroplasts expanded to at least twice the volume of intact chloroplasts, lost about 50% of their stromal proteins in the medium, and metabolized exogenous nucleotides. However, neither acetyl-coenzyme A (CoA) nor malonyl-CoA inhibited fatty acid synthesis from acetate; nor were [1-14C]acetyl-CoA and [14C]malonyl-CoA significantly incorporated into fatty acids. Fatty acid synthesis from acetate was independent of added cofactors but was totally light dependent. Changes in the products of fatty acid synthesis were consistent with the loss of endogenous glycerol-3-phosphate from permeabilized chloroplasts. However, in appropriately supplemented medium, the products of acetate incorporation by spinach (Spinacia oleracea) chloroplasts were similar when reactions were carried out in either isotonic or hypotonic medium. Taken together, the results of this study suggest that the enzymes of fatty acid synthesis with chloroplasts are organized into a multienzyme assembly that channels acetate into long-chain fatty acids, glycerides, and CoA esters.  相似文献

We previously reported (A Reinero, RN Beachy 1986 Plant Mol Biol 6:291-301) that coat protein (CP) of tobacco mosaic virus (TMV) accumulates in chloroplasts of systemically infected leaves. To determine the significance of such interaction we examined electron transport rates in chloroplasts containing different levels of TMV-CP. Tobacco (Nicotiana tabacum L.) plants were infected with either a TMV strain inducing chlorosis or with a strain inducing mild symptoms, and both the accumulation pattern of TMV-CP inside chloroplasts as well as the rates of photosynthetic electron transport were followed. The CP of the TMV strain inducing chlorosis was detected inside chloroplasts 3 days after infection, and thereafter accumulated at a rapid rate, first in the stroma and then in the thylakoid membranes. On the other hand, the CP of the TMV strain that caused only mild symptoms accumulated in chloroplasts to lower levels and little CP was associated with the thylakoids. In vivo and in vitro measurements of electron transport revealed that photosystem II activity was inhibited in plants infected with the aggressive TMV strain while no reduction was observed in plants infected with the mild strain. The capacity of chloroplasts to synthesize proteins was equivalent in organelles isolated from healthy and virus-infected leaves. The possibility that a large accumulation of TMV-CP inside chloroplasts may affect photosynthesis in virus-infected plants by inhibiting photosystem II activity is discussed.  相似文献

In eukaryotic cells, both lysosomal and nonlysosomal pathways are involved in degradation of cytosolic proteins. The physiological condition of the cell often determines the degradation pathway of a specific protein. In this article, we show that cytosolic proteins can be taken up and degraded by isolated Saccharomyces cerevisiae vacuoles. After starvation of the cells, protein uptake increases. Uptake and degradation are temperature dependent and show biphasic kinetics. Vacuolar protein import is dependent on cytosolic heat shock proteins of the hsp70 family and on protease-sensitive component(s) on the outer surface of vacuoles. Degradation of the imported cytosolic proteins depends on a functional vacuolar ATPase. We show that the cytosolic isoform of yeast glyceraldehyde-3-phosphate dehydrogenase is degraded via this pathway. This import and degradation pathway is reminiscent of the protein transport pathway from the cytosol to lysosomes of mammalian cells.  相似文献

利用荧光光谱学等方法结合高压力技术研究了光合作用系统II中的一个外周蛋白——— 2 3kD(以P2 3k表示 )蛋白的去折叠。热力学研究表明 ,在 2 0℃、180MPa(1MPa =10 .0大气压 )可使该蛋白质完全去折叠 ,而在3℃ ,16 0MPa即可使该蛋白质完全去折叠 ,这是迄今为止有关研究中最易被高压力去折叠的一个蛋白质。在2 0℃ ,该蛋白质在常压下去折叠反应的标准自由能与标准体积变化分别为 2 3.4 5kJ mol和 - 15 0 .3ml mol;动力学研究揭示该蛋白质的折叠反应的活化体积ΔV f 为正值 (84 .1ml mol) ,而去折叠反应的活化体积ΔV u 为负值(- 6 6 .2ml mol)。在常压下 ,折叠和去折叠反应的速度常数 (K0f,K0u)分别为 1.87s- 1 和 1.3× 10 - 4s- 1 ,这些结果为解释该蛋白质易被压力去折叠提供了线索  相似文献
