The germination of intact, dehusked, and peeled seeds (caryopses) of the japonica rice cultivar Sasanishiki, harvested 30, 40, 47 and 60 days after anthesis, and of the indica rice cultivar Assam IV, harvested 14 and 28 days after anthesis, was examined. Dehusking strongly inhibited germination of Sasanishiki seeds, with the exception that seeds harvested 30 days after anthesis gave minimal germination percentages even when left intact. Peeling (removal of the pericarp and testa) restored or enhanced germination, and 60–100% of seeds germinated after 10 days. By contrast, the rank order of germination of Assam IV seeds was intact, dehusked, and peeled seeds, with peeled seeds yielding germination percentages of 100%. In Sasanishiki, inhibition of germination of peeled seeds was observed at reduced oxygen concentrations (1–4% oxygen). This inhibition might explain the inhibitory effects of dehusking on germination of seeds from the japonica cultivar. It is possible that the testa and pericarp, which cover the embryos of dehusked seeds, acted as a barrier to the diffusion of oxygen to the embryo.  相似文献

An examination was made of the effects of ethanol at 0.2–6.0%(v/v) on the germination, under aerobic conditions, of intactand dehusked seeds of indica rice (cv. Assam IV), which hadbeen harvested 14, 21 and 28 d after anthesis, and of the japonicarice (cv. Sasanishiki), which had been harvested 30 and 60 dafter anthesis. The inhibition of germination caused by dehuskingjaponica rice was overcome by 0.5–5% ethanol, with maximumgermination (frequently 100%) achieved at 3–5% (30 d afteranthesis) or 1–4.5% (60 d after anthesis) ethanol. Furtherincreases in the ethanol concentration reduced germination.The germination of dehusked indica rice was slightly inhibitedat 0.5 and 1% ethanol, whilst the promotion of germination by2% ethanol increased as the seeds matured. At all harvests germinationwas greatest at 3% ethanol, and at 5–6% ethanol germinationfell to 0%. Inhibition, no effect, or minimal stimulation ofthe germination of intact seeds of both japonica and indicarice by ethanol was observed at the concentrations examined.The absence of oxygen stimulated germination of dehusked japonicarice, but this germination was inhibited by ethanol. In contrastethanol had little or no effect on the failure of dehusked indicaseeds to germinate in anaerobic conditions. Thus ethanol treatmentmay help break the strong dormancy of dehusked seeds of indicaand japonica rice. The possible role of ethanol in stimulatinggermination in rice is discussed. Differences in germination of intact and dehusked seeds of two japonica and two indica cultivars of rice (Oryza sativaL.) were examined during the development and maturation of seeds both under high-temperature (30/23°C) and low-temperature (20/13°C) regimes and a 14-h photoperiod in growth cabinets. As described previously for seeds grown in the paddy fields, germination of freshly harvested japonica rice seeds that developed and matured in growth cabinets was inhibited by dehusking. We observed a roughly consistent triphasic pattern with respect to the germination of intact and dehusked seeds during the development and maturation of seeds of indica and japonica rice. The triphasic pattern consisted of: (a) an initial phase, during which germination was stimulated by dehusking both in indica and japonica rice; (b) a second phase during which almost no dehusked seeds of indica or japonica rice germinated; and (c) a third phase during which intrinsic differences between indica and japonica rice were observed, with dehusking stimulating germination of indica rice but inhibiting that of japonica. Temperature regimes did not affect this triphasic pattern, but the time from the day of anthesis to mass maturity was affected by temperature regimes, and the effect was more pronounced in indica than in japonica rice.  相似文献

NaCl salinity stress consistantly decreased the rate of germinationof wheat. GA alone or in combination with kinetin alleviatedthe inhibitory effect of salinity on germination. However, kinetinfurther decreased the rate of germination under NaCl salinitystress. NaCl salinity increased accumulation of Na+ and Clwhile it decreased K+ accumulation in germinating seeds. GAcaused an increase in K+ accumulation and a decrease in Claccumulation in the germinating seeds while kinetin increasedCl accumulation in salinity stressed plants. 1. The effects of gibberellin, kinetin, potassium nitrate, andtheir interactions in the germination of light-sensitive tobaccoseeds were studied. 2. Gibberellin was very effective in inducing the dark germinationof tobacco seeds, and a linear relation was found to exist betweenthe germination rate and the concentration of gibberellin ifa suitable temperature was chosen for germination. This linearrelation was, however, changed by illumination and by a raisedtemperature in the manner that the germination on lower concentrationsof gibberellin was increased by the former and decreased bythe latter. 3. Kinetin was effective in promoting the tobacco seed germinationonly when the seeds were irradiated. This light effect was,however, limited only to the longer wavelengths in the visibleregion. The red effect was found to be reversed by infra-redirradiation under the influence of kinetin. 4. The combination of gibberellin and kinetin was synergisticin promoting the tobacco seed germination both in the lightand in the dark, whereas the combination of potassium nitrateand gibberellin or kinetin was synergistic only in the light. Sensitivity to ultraviolet-B (UVB) radiation (280-320 nm) varies widely among rice cultivars. We previously indicated that UV-resistant rice cultivars are better able to repair cyclobutane pyrimidine dimers (CPDs) through photorepair than are UV-sensitive cultivars. In this paper, we report that UVB sensitivity in rice, in part, is the result of defective CPD photolyase alleles. Surjamkhi (indica) exhibited greater sensitivity to UVB radiation and was more deficient in CPD photorepair ability compared with UV-resistant Sasanishiki (japonica). The deficiency in CPD photorepair in Surjamkhi resulted from changes in two nucleotides at positions 377 and 888 in the photolyase gene, causing alterations of two deduced amino acids at positions 126 and 296 in the photolyase enzyme. A linkage analysis in populations derived from Surjamkhi and Sasanishiki showed that UVB sensitivity is a quantitative inherited trait and that the CPD photolyase locus is tightly linked with a quantitative trait locus that explains a major portion of the genetic variation for this trait. These results suggest that spontaneously occurring mutations in the CPD photolyase gene cause different degrees of sensitivity to UVB in rice, and that the resistance of rice to UVB radiation could be increased by increasing the photolyase function through conventional breeding or bioengineering.  相似文献

UV-B responses of three rice (Oryza sativa L.) cultivars (Sasanishiki, Norin 1 and Surjamkhi) with different photolyase activity were investigated. Carbon dioxide assimilation data support that Sasanishiki was less sensitive to UV-B than Norin 1 and Surjamkhi. UV-B radiation sharply decreased the content of Rubisco protein in Surjamkhi and has no effect in Sasanishiki. The photochemical activities of photosystem (PS) 1 and PS 2 was slightly affected by UV-B treatment. The content of H2O2 and the activities of antioxidant enzymes, catalase (CAT), peroxides (POX) and superoxide dismutase (SOD) were enhanced after UV-B treatment. The activities of CAT and POX isoenzymes in Sasanishiki were more enhanced by UV-B radiation than those in Norin 1 and Surjamkhi.  相似文献

Theseeds couldgerminate under otherwise non-inductive photoperiods,when thedark period was interrupted with a short period ofillumination.Thus the photoperiodic behaviour of Begonia seedsin germinationis similar to that of typical long-day plantsin flowering.

The application of gibberellin brought aboutno germinationin complete darkness, but markedly reduced thecritical daylengthfor germination, even 1-minute photoperiodsbeing inductive.The germination under continuous light wasalso favoured bygibberellin application. Immunocytological studies in this laboratory have suggested that NADH-dependent glutamate synthase (NADH-GOGAT; EC 1.4.1.14) in developing organs of rice (Oryza sativa L. cv. Sasanishiki) is involved in the utilization of glutamine remobilized from senescing organs through the phloem. Because most of the indica cultivars contained less NADH-GOGAT in their sink organs than japonica cultivars, over-expression of NADH-GOGAT gene from japonica rice was investigated using Kasalath, an indica cultivar. Several T0 transgenic Kasalath lines over-producing NADH-GOGAT under the control of a NADH-GOGAT promoter of Sasanishiki, a japonica rice, showed an increase in grain weight (80% as a maximum), indicating that NADH-GOGAT is indeed a key step for nitrogen utilization and grain filling in rice. A genetic approach using 98 backcross-inbred lines (BC(1)F(6)) developed between Nipponbare (a japonica rice) and Kasalath were employed to detect putative quantitative trait loci (QTLs) associated with the contents of cytosolic glutamine synthetase (GS1; EC 6.3.1.2), which is probably involved in the export of nitrogen from senescing organs and those of NADH-GOGAT. Immunoblotting analyses showed transgressive segregations toward lower or greater contents of these enzyme proteins in these BC(1)F(6). Seven chromosomal QTL regions were detected for GS1 protein content and six for NADH-GOGAT. Some of these QTLs were located in QTL regions for various biochemical and agronomic traits affected by nitrogen recycling. The relationships between the genetic variability of complex agronomic traits and traits for these two enzymes are discussed.  相似文献

Summary Dormancy of intact sycamore (Acer pseudoplatanus) seeds was broken by chilling (5°C) for several weeks in moist conditions. Treatment of unchilled seeds with kinetin induced some germination, but gibberellin was ineffective. This stimulation by kinetin was not suppressed by the added presence of abscisic acid during incubation.The chilling requirement of intact seeds was eliminated by removal of the testa, and the naked embryos developed with no morphological abnormalities. During early growth of isolated embryos in the light, two distinct developmental processes were recognised. One involved initial elongation of the radicle accompanied by geotropic curvature and was stimulated by kinetin but not by gibberellin, while the other involved unrolling of the cotyledons, which was accelerated by gibberellin but much less by kinetin. Abscisic acid strongly suppressed both developmental processes when applied alone, inhibited cotyledon expansion in the presence of gibberellin, but failed to overcome the promotory effects of kinetin on radicle growth. Experiments with CCC indicated that under natural conditions the unrolling of the cotyledons is dependent upon endogenous gibberellin. Radicle growth of isolated embryos was unimpaired by incubation in the dark, but cotyledon expansion of water incubated embryos was poor, and although it was accelerated by gibberellin, the responses in all treatments were slower than in the corresponding light grown samples.It is suggested that endogenous cytokinins are primary factors in the initiation of radicle growth, while gibberellins are important in cotyledon expansion. Abscisic acid appears to have an inhibitory role in both processes, and the interactions of these regulators in the control of germination and development are discussed.  相似文献

The effects of CO2 on dormancy and germination were examinedusing seeds of cocklebur (Xanthium pennsylvanicum Wallr.) andgiant foxtail (Setaria faberi Herrm.). The rate of germinationof the giant foxtail seeds as well as cocklebur was promotedby exogenously applied CO2 at a concentration of 30 mmol mol-1regardless of the sowing conditions. However, seeds which failedto germinate in the presence of CO2, entered a secondary phaseof dormancy under unfavourable germination conditions. If CO2was applied to seeds under conditions such as water stress imposedwith a 200 mol m-3 mannitol solution, a hypoxic atmosphere of100 mmol mol-1 O2 or a treatment of 0·1 mol m-3 ABA,development of secondary dormancy was accelerated. These contrastedeffects of CO2 were observed in ecological studies. Under naturalfield conditions germination of buried giant foxtail seeds respondedpositively to CO2 during a period of release from primary dormancyfrom Feb. to May, but CO2 accelerated secondary dormancy commencingin early Jun. In other words, in the presence of CO2, both theenvironmental conditions and the germination states of the seedsclearly showed secondary dormancy-inducing effects. Instances of flowering of bamboo species Dendrocalamus strictus are few and far between which are taken as opportunity by nurserymen to collect seeds for propagation. Germination of seeds is reported to be poor. Therefore, different seed priming treatments were applied to D. strictus seeds collected from Ranchi in order to obtain uniform and high germination. Under laboratory conditions, dehusking of seeds before sowing ensured cent percent germination. Seed priming with KNO3 1% solution resulted in 80.4% increase in germination followed by hydropriming by 16 h (73.1% increase). In field conditions, dehusked seeds gave 23.0% germination without any priming treatment. Priming treatment with KNO3 1% gave the highest rise in germination (39.1%) followed by hydropriming for 16 h (26.1%). Seeds with their seed coats intact could give germination of 9.5% when germinated without any treatment. A rise of 115.8% in germination was obtained by priming with KNO3 1% (final germination count 20.5%). The next best treatment was hydropriming for 16 h (final germination 18.5%, a rise of 94.7%). KNO3 1% also induced the earliest and the most rapid germination. When seedlings germinated in laboratory were transferred to soil, all seedlings from all treatments established successfully without any mortality whatsoever. Therefore, it is recommended that seeds should be primed for 8 h with 1% KNO3 and germinated in laboratory or in farm house under normal atmospheric condition before transplanting the seedlings to soil.  相似文献

Gibberellin A3 (GA3), kinetin and zeatin promoted germinationof conditioned witchweed (Striga asiatica [L.] Kuntze) seedssimilar to dl-strigol and a natural stimulant from corn rootexudates. GA3 promoted germination only with scarified seeds.However, these regulators, as well as dl-strigol and naturalstimulants, inhibited the conditioning of seeds and their subsequentgermination. In indica rice, heating followed by chilling followed by heating resulted in decreased mean germination time, time to start germination, electrical conductivity of seed leachates, and time to 50% germination, as well as increased germination index, energy of germination, radicle and plumule length, root length, root/shoot ratio, root fresh and dry weight, radicle and plumule growth rate, and shoot fresh weight. The effects of 4 chemicals on the germination promotion of stratified and unstratified seeds of loblolly pine (Pinus taeda) and baldcypress (Taxodium distichum) were studied. The chemicals used were gibberellic acid, kinetin, potassium nitrate and thiourea, each at 3 different concentrations. Stratification promoted the germination of both seed species. Certain concentrations of gibberellic acid, potassium nitrate and thiourea improved the germination of unstratified loblolly pine and baldcypress seeds while kinetin had no stimulatory effect. All 4 chemicals at specific concentrations promoted the germination of loblolly pine seeds stratified for a short period of time. Considering both speed and completeness of germination, best results were obtained when 21-day stratified seeds were treated with either gibberellic acid (100 mg/1) or kinetin (10 mg/1). In baldcypress, on the other hand, none of these chemicals had any stimulatory effect on the germination of stratified seeds. Germination of both species of seeds was either partially or completely inhibited by the highest concentration of thiourea (30,000 mg/1) used.  相似文献

To understand the genetic basis of yield-related traits of rice, we developed 39 chromosome segment substitution lines (CSSLs) from a cross between an average-yielding japonica cultivar, Sasanishiki, as the recurrent parent and a high-yielding indica cultivar, Habataki, as the donor. Five morphological components of panicle architecture in the CSSLs were evaluated in 2 years, and 38 quantitative trait loci (QTLs) distributed on 11 chromosomes were detected. The additive effect of each QTL was relatively small, suggesting that none of the QTLs could explain much of the phenotypic difference in sink size between Sasanishiki and Habataki. We developed nearly isogenic lines for two major QTLs, qSBN1 (for secondary branch number on chromosome 1) and qPBN6 (for primary branch number on chromosome 6), and a line containing both. Phenotypic analysis of these lines revealed that qSBN1 and qPBN6 contributed independently to sink size and that the combined line produced more spikelets. This suggests that the cumulative effects of QTLs distributed throughout the genome form the major genetic basis of panicle architecture in rice. All seeds of Aeluropus lagopoides and Urochondra setulosa germinated under non-saline conditions except for Sporobolus ioclados which showed only 40 % germination. Increase in salinity substantially inhibited germination and few seeds germinated at 400 mM NaCl. Germination at 200 mM NaCl was alleviated in U. setulosa by the application of gibberellic acid and fusicoccin, in A. lagopoides by thiourea, betaine, kinetin, fusicoccin and ethephon, and in S. ioclados by gibberellin and ethephon. High salinity (400 mM NaCl) induced germination inhibition was alleviated by proline, kinetin, fusicoccin and ethephon only in A. lagopoides.  相似文献

The donor-recipient protoplast fusion method was used to produce cybrid plants and to transfer cytoplasmic male sterility (CMS) from two cytoplasmic male-sterile lines MTC-5A and MTC-9A into a fertile japonica cultivar, Sasanishiki. The CMS was expressed in the cybrid plants and was stably transmitted to their progenies. Only cytoplasmic traits of the male-sterile lines, especially the mitochondrial DNAs, were introduced into the cells of the fertile rice cultivar. More than 80% of the cybrid plants did not set any seeds upon selfing. Sterile cybrid plants set seeds only when they were fertilized with normal pollen by hand and yielded only sterile progenies. This maternally inherited sterility of the cybrid plants showed that they were characterized by CMS. The CMS of cybrid plants could be restored completely by crossing with MTC-10R which had the single dominant gene Rf-1 for restoring fertility. These results indicated that CMS was caused by the mitochondrial genome introduced through protoplast fusion. The introduced CMS was stably transmitted to their progenies during at least eight backcross generations. These results demonstrate that cybrids generated by the donor-recipient protoplast fusion technique can be used in hybrid rice breeding for the creation of new cytoplasmic male-sterile rice lines.  相似文献

Effects of three phytohormones (IAA1, GA3 and kinetin) on grain-fillingand the pattern of 32P translocation from individual leavesto grains were studied at intervals of 7 days during the progressof reproductive development of rice (Oryza saliva L. cv. Jaya).The plants were sprayed with 100 µg ml–1 aqueoussolutions of the hormones at 100 days, when the plants wereentering the reproductive stage. Kinetin produced a pronouncedeffect on grain-filling as well as on 32P mobilization fromindividual leaf to grains and increased yield, possibly by increasingleaf longevity. GA3 and IAA also increased the grain-fillingand 32P mobilization significantly over control but the effectswere less marked than those of kinetin.
