This study analysed the microhabitat use of three endangered fish species, Andalusian toothcarp Aphanius baeticus , Iberian loach Cobitis paludica and sand smelt Atherina boyeri , in a coastal stream stretch. Plastic minnow traps were set both during daytime and at night on the bottom and at the surface. Depth and presence of effective refuge were recorded for each trap. To assess size‐related changes in microhabitat use individuals of each species were classified in three size classes. The three species preferentially used bottom positions in the water column, though this behaviour was more evident in the case of Iberian loach. While large Iberian loach remained active at night Andalusian toothcarp and sand smelt were strongly diurnal, especially larger individuals. The three species showed a clear ontogenetic change in microhabitat preferences towards deeper waters. Small Andalusian toothcarp and medium‐sized Iberian loach used deeper microhabitat in the presence of refuge. Large Andalusian toothcarp consistently preferred exposed microhabitat at any time. Andalusian toothcarp using refuge were smaller at any time, while Iberian loach followed this pattern only at night. The differential vulnerability of these species to different predators (aerial and aquatic; diurnal and nocturnal) could explain the observed patterns in microhabitat use. Fish tended to co‐occur in microhabitats either due to habitat characteristics independently of species or due to species independently of habitat. Andalusian toothcarp segregated spatio‐temporally from sand smelt and Iberian loach, but these species occurred independently of each other. According to these co‐occurrence patterns, Andalusian toothcarp would be more sensitive than Iberian loach or sand smelt to interspecific interactions.  相似文献

Young‐of‐the‐year smelt Osmerus eperlanus in Lake Tuusulanjärvi were examined for Glugea hertwigi cysts. Cysts were visible on smelt in the beginning of August and showed a peak at the end of August. Glugea hertwigi may cause mortality among the most heavily infected young‐of‐the‐year smelts.  相似文献

A protein kinase activity associated with maize root plasma membranes was partially purified and characterized. Biochemical properties, such as calcium dependence, inhibition by calmodulin antagonists, and absence of calmodulin stimulation, indicated that the enzyme belongs to the calcium‐dependent protein kinase (CDPK) family. By means of an in‐gel phosphorylation assay the molecular mass of active polypeptides was determined: two bands of 55 and 51 kDa became labelled. The same proteins were also immunodecorated by monoclonal antibodies against soybean CDPK. Results from in vitro assays demonstrated that maize H+‐ATPase was a suitable substrate for this protein kinase and that the phosphorylation site was located at the C‐terminal domain of the enzyme. This result was confirmed by using as substrate in phosphorylation assays the isolated C‐terminal domain of the H+‐ATPase expressed in Escherichia coli as a glutathione‐transferase fusion protein.  相似文献

Leucocyte lysates from rainbow trout Oncorhynchus mykiss showed an iron‐binding activity that was retained even if the samples were exposed to an acid pH (4·5). Iron‐binding activity of leucocyte supernatants was enhanced by the presence of 1 μg ml−1 phorbol‐12‐myristate‐13‐acetate in the cell medium.  相似文献

Inter‐individual differences in rates of routine (non‐feeding) metabolism and growth were evaluated in young‐of‐the‐year (YOY) juvenile Atlantic cod Gadus morhua . Rates of O2 consumption, CO2 production and ammonia (TAN) excretion were measured in 64, 25–43 mm standard length ( L S) YOY growing at different rates (0·27–0·47 mm day−1) in a common rearing tank. Parameter rates ( y ) increased allometrically ( y = a·Mb ) with increasing body mass ( M ) with b ‐values for O2 production, CO2 consumption and TAN excretion equal to 0·81, 0·89 and 0·56, respectively. In some cases, residuals from these regressions were significantly negatively correlated to fish growth rate. In no cases did residuals of parameter rates increase with increasing growth rate. These data suggest that, during unfed periods, relatively fast‐growing fish were more metabolically efficient than slower‐growing fish from the same cohort. The fish condition factor, derived from     , also significantly decreased with increasing growth rate. Results indicated differences in both the rates of routine energy loss and the patterns of growth allocation among YOY Atlantic cod. Since these physiological attributes were positively correlated with growth rate, they may be indicative of 'survivors' in field populations.  相似文献

The physiological and molecular events of ethylene‐induced abscission in mature fruit calyx, laminar and floral abscission zones of cv. Valencia orange were examined. Continuous exposure of fruit explants to 5 µl 1−1 ethylene for 2 to 40 h resulted in marked increases in endo‐1,4‐β‐glucanase (cellulase) and polygalacturonase (PG) activities in calyx abscission zones. Two abscission‐related cellulases and one PG were found. The major peak of cellulase activity corresponded to a pI of 8.0 and molecular weight of 51 kDa, whereas the minor cellulase peak had a pI of 5.5. The abscission polygalacturonase had a pI of 5.5. Calyx abscission zone RNA was amplified with degenerate primers based on sequence of the purified Valencia orange calyx abscission cellulase, and cloned. The two partial cellulase cDNA clones were 59% identical at the nucleotide level. Genomic Southern analysis suggested that Valencia orange contained two groups of cellulase genes. A full‐length cDNA clone from each group was isolated from a cDNA library prepared from ethylene‐induced calyx abscission zone mRNA. Both genes were expressed in ethylene‐induced calyx, laminar and floral abscission zones, but were not expressed in non‐induced abscission zones or mature leaves treated with or without ethylene, young bark or young fruit of Valencia.  相似文献

2‐Cysteine‐peroxiredoxins (2‐CP) constitute a ubiquitous group of enzymes which reduce toxic alkyl hydroperoxides. In higher plants it was shown that the nuclear encoded 2‐CPs are posttranslationally imported into the chloroplasts, the site of most active oxidative metabolism in plants (Baier and Dietz 1997, Plant J. 12; 179‐190). The genome of the bluegreen alga Synechocystis (EMBL acc. # D64000) encodes a 2‐CP which shares 60% homology to higher plant 2‐CPs on the gene level and about 70% on the level of the mature protein. In order to elucidate the physiological significance of 2‐CPs for photosynthetic organisms, the 2‐CP gene was mutated in Synechocystis sp. PCC 6803 by insertion of a kanamycin gene cartridge. Following complete segregation mutant lines were analyzed for growth and photosynthetic parameters. The mutants revealed decreased growth rates as compared to the wild type. Growth inhibition was relieved after lowering the concentration of Fe or trace elements in the growth medium. Chlorophyll a fluorescence transients as induced by saturating light pulses were used as indicator for the state of photosynthesis. The effective quantum yield decreased at lower light intensities in the mutants as compared to the wild type Synechocystis . Simultaneously, electron transport rates saturated at lower light intensities in the mutants. These data provide the first evidence that 2‐CPs play a pivotal protective role in photosynthesis.  相似文献

Fifty‐three one‐sea‐winter Atlantic salmon Salmo salar (45–63 cm L T) were radio‐tagged in the Tana fjord, Barents Sea, in 1995. Thirty‐seven fish (70%) entered the freshwater zone of the River Tana in an average of 3 days after release in the fjord. The migration speeds in the lowest river section below the first riffle area were significantly higher than in the subsequent river section below the second riffle area. Similarly, the observed time spent in the first riffle area was significantly lower than in the next riffle area. The majority of Atlantic salmon entered the river during the hours of high tide and the subsequent ebb tide. In addition, most river entries were recorded around midnight. No effects of river flow on the river entry or migration speed were detected, but the migration speed of Atlantic salmon in both river sections examined was greater at lower temperatures. Twenty‐eight fish (72%) were recaptured in the river, 71% of them with weirs and gillnets, and 29% by rod and line. Over half of the Atlantic salmon (54%) were recaptured within 3 weeks following river entry, and within the first 100 km of the river (56%). The results are discussed in relation to earlier studies on multi‐sea‐winter Atlantic salmon in the River Tana.  相似文献

Literature data for 19 populations from 16 different studies of six species of stream‐dwelling salmonids were analysed to test the hypothesis that density‐dependent growth is stronger at low rather than at high population density. Fifteen of 19 populations showed evidence of a significant decrease in growth rate with increasing density. In 11 of these 15 populations, the pattern of density‐dependent growth was better described by a negative power curve than by a linear regression ( i.e . Akaike Information Criterion, AIC linear −  AIC power > 2), whereas only one population was better described by a linear regression than by a negative power curve; three populations were adequately described by both models ( AIC  < 2). In 10 of the 11 populations that were best described by a negative power curve, most of the decrease in growth rate occurred at population densities <1 fish m−2, when space limitation is unlikely. This analysis provides broad support for the hypothesis that density‐dependent growth in stream salmonids occurs primarily at low population densities, probably due to exploitative competition.  相似文献

Direct day and night underwater observations of juvenile Atlantic salmon Salmo salar during summer and autumn showed a duality in response to temperature between 7 and 11° C for young‐of‐the‐year (YOY) Atlantic salmon. They were predominantly diurnal in early summer and nocturnal in late summer although water temperatures were similar. Post‐YOY Atlantic salmon did not show a strong response to temperature as they were mostly nocturnal during the study period. It is suggested that the difference in activity patterns between YOY and post‐YOY Atlantic salmon can be explained by size‐dependent trade‐off between growth and predation risk.  相似文献

Stem elongation can be suppressed by a temperature drop at the onset of the photoperiod (DROP) or with a cooler day than night temperature (DT and NT, respectively), commonly described as DIF (DT ‐ NT). To test our hypothesis that phytochrome A (phyA) mediated the reduction of stem elongation caused by −DIF and DROP, we conducted experiments with photomorphogenic mutants of tomato ( Solarium lycopersicon L.) and transgenic potato ( Solarium tuberosum L.). The plants studied were tomato mutants fri 1 (deficient in phyA) and tri 3 (deficient in phytochrome B1 [phyBl]) and their isogenic wild‐type (WT) cv. Moneymaker, nontransformed potato, and two lines each of antisense phyA (15‐9 and 15‐11) and overexpressed phyA (PS‐2 and PS‐4). Plants were placed in three temperature regimens with a daily mean of 20°C: a constant 20°C (0 DIF), an 8°C DROP for 3 h, and a ‐ 8°C DIF. For all tomato genotypes, −DIF and DROP reduced intemode length by ≥ 21% and stem elongation by 30% compared to that of plants at 0 DIF. Interactions between temperature treatment and genotype were nonsignificant. For potato, −DIF, but not DROP, significantly reduced intemode length of WT (by 39%) and both antisense lines (by 36 or 48%) but only one of the two lines of overexpressed phyA plants (by 18%). The −DIF significantly reduced stem length for only antisense phyA (by 36 or 48%) and WT (by 35%) plants. Thus, at least for tomato and potato, it appears that phyA does not control stem extension in relation to cool‐temperature treatments.  相似文献

Seasonal and diurnal patterns of larval and juvenile fish drift were investigated in the Marchfeldkanal, a man‐made side branch of the Danube River near Vienna, Austria. A clear seasonal pattern with peak densities in mid‐June was found. Species composition varied over time, showed a site specific pattern and was dominated by tubenose goby Proterorhinus marmoratus . Water temperature was the main factor responsible for the increase of drift densities until the median drift date and repeated occurrence of early larval stages in drift indicated repeated spawning for many species. Significant differences in drift densities between different time periods of the day (day, dusk, night and dawn) were found for common bream Abramis brama , barbel Barbus barbus , chub Leuciscus cephalus , tubenose goby and roach Rutilus rutilus . The highest drift rates occurred at night (2200–0400 hours), with 86% of all larvae drifting during the hours of darkness. Fish larvae of different lengths drifting at different phases of the day were found for common bream, bleak Alburnus alburnus and chub, with largest larvae drifting during dusk (chub) and day (bleak and common bream). For bleak, all gudgeon species Gobio spp., tubenose goby, roach and for all cyprinid species combined, one 2 h night sample was found to be sufficient to predict the total 24 h drift.  相似文献

A number of recent papers have investigated the potential of familiarity to organize the distributions of free‐ranging animals. It is not clear, however, to which extent individual recognition or a more general recognition of a group odour is responsible for familiarity preferences. First, we tested the sensory basis of the recognition of familiars in three‐spined stickleback ( Gasterosteus aculeatus ). When allowed to choose between a familiar and an unfamiliar stimulus shoal on the basis of both visual and chemical communication, visual communication only and chemical communication only, the preference of focal fish for familiars was shown to be dependent on the presence of chemical cues. We subsequently investigated the mechanisms underlying such association preferences, specifically the effects of recent habitat and diet on preferences. Experimental fish were divided into four treatment groups consisting of two environment treatments (saline and freshwater) and two diet treatments ( Daphnia spp. and chironimid larvae). Focal fish subsequently showed significant association preferences for groups of unfamiliar fish that had undergone the same environment or diet treatment as themselves, suggesting self‐referent matching. These data indicate that individual recognition is not a pre‐requisite for the expression of familiarity preferences.  相似文献

Atlantic salmon Salmo salar were followed from egg to smoltification using genetic analysis to identify individuals and to link observations from pre‐ and post first feeding. Egg size and hatch timing significantly influenced alevin size at first feed but neither egg size, hatch timing or alevin sizes were correlated to size, condition factor or smolt status post first feed. In a hatchery environment the potential advantage gained by early hatching, larger alevin does not persist after first feeding. Different physiological and genetic complexes appear to influence growth in these two distinct phases of the Atlantic salmon's life‐cycle.  相似文献

The aim of this study was to co‐evaluate c‐ erbB ‐2 and p53 protein expression in breast cancer fine needle aspirates (FNA) and to compare this with histological variables and the immunohistochemical phenotype of the tumours. Furthermore, we assessed the relationship of c‐ erbB ‐2 and p53 immunocytochemical expression to tumour prognostic factors. We examined 124 breast cancer FNAs and 79 matched surgical specimens using the avidin–biotin complex (ABC) and the alkaline phosphatase immunocytochemical techniques. C‐ erbB ‐2 immunopositivity was detected in 37.9% of the FNAs, while 31.7% were positive for p53. A statistically significant correlation was observed between p53 negativity and absence of c‐ erbB ‐2 immunostaining in the FNAs ( P =0.0007). Smears from infiltrating ductal carcinomas tended to be more frequently positive for p53 (36.7%) than those from lobular carcinomas (11.7%) ( P =0.054). In matched tumour tissues, c‐ erbB ‐2 was positive in 16.7% and p53 in 19% of cases. The immunocytochemical results for both c‐ erbB ‐2 and p53 were significantly correlated with the immunohistochemical results. There was no correlation between c‐ erbB ‐2 and p53 immunostaining, in both FNAs and tissues, and patients' menopausal status, tumour size, grade and lymph node status.  相似文献

Specific receptor and fibre types of rainbow trout Oncorhynchus mykiss involved in the detection and discrimination of amino acids and a heterogeneous collection of compounds were investigated by recording the electrical activity of the maxillary branch of the facial nerve innervating taste buds inside the upper jaw. Proline (Pro), alanine (Ala), leucine (Leu), betaine (Bet) and 2‐amino‐3‐guanidinopropionic acid (Agp) were the major amino acids detected by the gustatory system. The two experimental approaches, concentration‐response curves and cross‐adaptations, showed that all amino acids were detected by three independent receptor types: Pro ‐, Agp/Bet ‐ and Leu ‐receptors. Bile acids, the most potent stimulants recorded, were detected by a single receptor type independent of those for amino acids, with threshold concentrations of 10−12 M. Strychnine, quinine and tetrodotoxin may have partially shared a single receptor mechanism. The gustatory sensibility narrowly tuned towards the amino acid spectrum compared to those for a diverse array of non‐feeding stimulant chemicals, combined with feeding behaviour triggered primarily by vision and olfaction, suggest that in rainbow trout, and possibly other salmonid species, gustatory chemical cues, in addition to food finding and intake, play an important role in detecting poisonous prey and substances.  相似文献

The population structure of 'lake‐type' and 'river‐type' sockeye salmon Oncorhynchus nerka , primarily in transboundary rivers in northern British Columbia, was examined with a survey of microsatellite variation. Variation at 14 microsatellite loci was surveyed from c . 3000 lake‐type and 3200 river‐type sockeye salmon from 47 populations in six river drainages in British Columbia. The mean F ST for the 14 microsatellite loci and 47 populations was 0·068, and 0·034 over all river‐type populations. River‐type sockeye salmon were more genetically diverse than lake‐type sockeye salmon, with expected heterozygosity of river‐type sockeye salmon 0·72 and with an average 12·7 alleles observed per locus, whereas expected heterozygosity of lake‐type sockeye salmon was 0·65 with and average 10·5 alleles observed per locus. River drainage of origin was a significant unit of population structure. There was clear evidence of genetic differentiation among river‐type populations of sockeye salmon from different drainages over a broad geographic range in British Columbia.  相似文献

The aim of this study was to test the hypothesis that newly‐acquired boron (B) undergoes rapid xylem‐to‐phloem transfer in plants with restricted mobility. Analysis of the element accumulation and water usage by shoots of intact broccoli ( Brassica oleracea var. italica Plenck cv. Commander) and lupin ( Lupinus albus L. cv. Ultra) plants provided with a non‐deficient supply of B, revealed that the concentration of various mineral elements (K, P, Mg, Ca, B, Fe, Zn, Mo, Cu, Mn) in xylem sap of intact plants ranged from 0.3 µ M to 3.5 m M , with B being present at 2.9‐3.5 µ M . For each element assayed, the concentration was higher in phloem exudate (1.6 µ M to 91 m M ) than in xylem sap; B was present at about 0.4 m M . Intact broccoli and lupin plants or detached transpiring broccoli shoots were supplied simultaneously with enriched 10B, strontium (a xylem marker) and rubidium (a xylem/phloem marker) during early reproductive growth. The contents of these three compounds were determined in foliage and florets or fruits as a function of time (i.e. up to 12 h and 4 days for broccoli and lupin plants, respectively), and the content in florets or fruits was expressed as a percent of the total recovered. In general, the percent recovery of both 10B and rubidium in florets or fruits was similar and markedly greater than that for strontium, even at the earliest harvest times (within 2 h for broccoli and 1 day for lupin). The data indicate that in plants with restricted B mobility, B is supplied to sink tissues in the phloem, and the extent of B xylem‐to‐phloem transfer is closely determined by current uptake.  相似文献

The influence of solar UV‐A and UV‐B radiation at Beltsville, MD, USA, on growth of Lactuca sativa L. (cv. New Red Fire lettuce) was examined during early summer of 1996 and 1997. Plants were grown from seed in plastic window boxes covered with Llumar to exclude UV‐A and UV‐B, polyester to exclude UV‐B, or tefzel (1996) or teflon (1997) to transmit UV‐A and UV‐B radiation. After 31–34 days, plants grown in the absence of solar UV‐B radiation (polyester) had 63 and 57% greater fresh weight and dry weight of tops, respectively, and 57, 72 and 47% greater dry weight of leaves, stems and roots, respectively, as compared to those grown under ambient UV‐B (tefzel or teflon). Plants protected from UV‐A radiation as well (Llumar) showed an additional 43 and 35% increase, respectively, in fresh and dry weight of tops and a 33 and 33% increase, respectively, in dry weight of leaves and stems, but no difference in root biomass over those grown under polyester. Excluding ambient UV‐B (polyester) significantly reduced the UV absorbance of leaf extracts at 270, 300 and 330 nm (presumptive flavonoids) and the concentration of anthocyanins at 550 nm as compared to those of leaf extracts from plants grown under ambient UV‐A and UV‐B. Additional removal of ambient UV‐A (Llumar) reduced the concentration of anthocyanins, but had no further effect on UV absorbance at 270, 300 or 330 nm. These findings provide evidence that UV‐B radiation is more important than UV‐A radiation for flavonoid induction in this red‐pigmented lettuce cultivar. Although previous workers have obtained decreases in lettuce yield under enhanced UV‐B, this is the first evidence for inhibitory effects of solar UV‐A and UV‐B radiation on lettuce growth.  相似文献
