Of the several species of Biomphalaria snails worldwide that serve as the intermediate host for Schistosoma mansoni, Biomphalaria alexandrina is a species that is indigenous to Egypt. Recently, there has been much debate concerning the presence of Biomphalaria glabrata and the hybrid of the species with Biomphalaria alexandrina. Due to this debate, the absence of a clear explanation for the presence of B. glabrata in Egyptian water channels and the probability that they may be reintroduced, we conducted this field study to identify Biomphalaria species present in Alexandria water channels. Laboratory-adapted susceptible snails to Schistosoma mansoni of the following species were used as a reference; Biomphalaria alexandrina, Biomphalaria glabrata and their hybrid. These snails were used to perpetuate the Schistosoma life cycle at the Theodor Bilharz Research Institute (TBRI), Cairo, Egypt. Morphological and molecular studies were conducted on these reference snails as well as on the first generation of Biomphalaria snails from two areas in the Alexandria governorate. The morphological study included both external shell morphology and internal anatomy of the renal ridge. The molecular study used a species-specific PCR technique.The results demonstrated that there was an absence of Biomphalaria glabrata and the hybrid from Alexandria water channels. Moreover, the susceptibility patterns of these reference snails were studied by measuring the different parasitological parameters. It was found that Biomphalaria glabrata and the hybrid were significantly more susceptible than Biomphalaria alexandrina to the Egyptian strain of Schistosoma mansoni. The results demonstrated that if Biomphalaria glabrata was reintroduced and adapted to the local environment in Egypt, it would have important epidemiologic impacts that would have a serious effect on the health of Egyptian people.  相似文献

Summary Grand Etang lake in Guadeloupe is a focus of intestinal schistosomiasis characterized by the virtual absence of human contact and the presence of a heavily infested rat population. The lake, with a surface area of 0.03 km2, is situated in rain forest at an altitude of 450 m. Originally it contained an extensive belt of the floating plant Pistia stratiotes which provided a favourable habitat for Biomphalaria glabrata, the intermediate host snail of Schistosoma mansoni. In 1974 and 1975, a low prevalence (about 3%) of S. mansoni was observed in B. glabrata while a high prevalence (about 60%) was recorded in rats. The rat population was infected with another trematode, Ribeiroia guadeloupensis, which requires two intermediate hosts, B. glabrata and the fish Tilapia mossambica. In 1976, the prosobranch snail, Ampullaria glauca, was accidentally introduced into the lake. By 1980, this snail had spread over half of the lake and had caused marked regression of the P. stratiotes layer. Concomitantly, the population of B. glabrata declined to such a point that in 1985 only a small colony remained in the northern section of the lake. The consequences of these changes are reflected in a rapid decline of S. mansoni in the rat population from 61.3% in 1980 to 34.8% in 1981, 6.5% in 1982, 2.8% in 1983, 11.6% in 1984, and 0% in 1985 and 1986.  相似文献

The percentage of water in the total living body weight is restricted within a narrow range for each species, even in snails cultured under differing controlled ionic concentrations in the medium. The water level in Helisoma trivolvis is consistently higher than in Biomphalaria pfeifferi and B. glabrata under the same culture conditions.The whole cationic fraction of the total osmoconcentration of the haemolymph is maintained at a constant hypertonic level, even though individual cation levels vary with the cationic composition of the surrounding medium.  相似文献

The single cell gel eletrophoresis or the comet assay was established in the freshwater snail Biomphalaria glabrata. For detecting DNA damage in circulating hemocytes, adult snails were irradiated with single doses of 2.5, 5, 10 and 20 Gy of 60Co gamma radiation. Genotoxic effect of ionizing radiation was detected at all doses as a dose-related increase in DNA migration. Comet assay in B. glabrata demonstrated to be a simple, fast and reliable tool in the evaluation of genotoxic effects of environmental mutagens.  相似文献

This study showed for the first time changes in the reproductive biology of Biomphalaria glabrata experimentally infected with Angiostrongylus cantonensis. The values of all the parameters analyzed (total number of eggs, number of egg masses, number of eggs/mass, number of eggs/snail, percentage of viable eggs and galactogen content in albumen gland) changed with progressive infection. The results indicate the occurrence of partial parasitic castration of B. glabrata by A. cantonensis larvae, probably in response to the depletion of energy reserves, with no injuries to the gonadal tissues.  相似文献

Eggs of the basommatophoran snails Physa marmorata and Biomphalaria glabrata were cultured in low concentrations of calcium to determine effects on growth and development. In both species there was some development in media with 0.12 mg/l Ca2+ but embryos were unable to hatch. 61.04% of embryos of P. marmorata could develop to hatching in 0.22 mg/l Ca2+ but those of B. glabrata required a level of 0.42 mg/l Ca2+, to attain even a 31.07% hatch. Marked effects on growth rate, embryo size and on time taken to achieve hatching were noted in both species at very low calcium levels. The possibility of cation-controlling mechanisms in the egg membrane is discussed.  相似文献

The efficiency of S. mansoni miracidia in locating and infecting Biomphalaria pfeifferi in Gezira canals has been studied under field conditions. When S. mansoni eggs were introduced into clean stagnant water in small field channels, the miracidia hatched to infect 100% of 30 snails in cages at the release point. Fifteen metres upstream and downstream 13% of caged snails were infected but no infections were found in snails 20 m away.When eggs were released into the same canal in flowing water (8.3 cm · s–1), no infections were detected in any of the caged snails placed 0–100 m downstream. Releasing hatched miracidia instead of eggs resulted in infections in all cages at 5 m intervals from 0-100 m. The release of eggs into flowing water was likened to the method by which S. haematobium eggs are deposited during urination. The 0% infection suggests that eggs will be swept away from the point of contamination by the flow. Thus only urination into stagnant water will lead to heavy snail infection rates.When eggs were released into a small pond-like minor canal tail end snail infection rates were only 3%. This was probably due to the larger water volume, smaller number of caged snails, and the presence of vegetation and other fauna which may be decoys or predators.The results highlight how very high snail infection rates can be produced under ideal conditions but also show how large snail and miracidia numbers are required in natural situations.  相似文献

Thin layer chromatography was used to analyze the glucose and maltose concentrations of the digestive gland–gonad complex (DGG) of uninfected-estivated Biomphalaria glabrata snails and estivated B. glabrata patently infected with Schistosoma mansoni. All snails were estivated in a most chamber at a relative humidity of 98 ± 1% and a temperature of 23 ± 1 °C for 14 days. Carbohydrates were extracted from the DGG with 70% aqueous ethanol, and extracts were analyzed on silica gel preadsorbent plates using ethyl acetate–glacial acetic acid–methanol–water (60:15:15:10) mobile phase, α-naphthol–sulfuric acid detection reagent, and quantification by densitometry. The concentrations of glucose and maltose were significantly reduced in both uninfected-estivated snails and infected-estivated snails.  相似文献

Summary The tissues of Biomphalaria glabrata contain three types of calcium cells which can be differentiated by their location and the size and number of their contained corpuscles. X-ray analysis has confirmed the presence of calcium and magnesium in the corpuscles. A model developed in a previous paper (Coutinho and Coutinho,Bull. Math. Biophysics,30, 553–563, 1968) is used to study populations ofBiomphalaria glabrata in fast-flowing rivers and channels. The hemoglobin of Biomphalaria glabrata was purified to homogeneity by gel filtration column followed by anion exchange chromatography. The dissociation products were analyzed by a 5–15% gradient polyacrylamide gel electrophoresis containing sodium dodecyl sulfate (SDS-PAGE) giving a band of 270 kDa and a band of 180 kDa after reduction with β-mercaptoethanol. The same profile was obtained in a 3.5% agarose gel electrophoresis containing SDS (SDS-AGE) but showed additional bands of higher molecular weight. These bands were proposed to be monomers, dimers and trimers, since they showed a good correlation in a plot of Rf versus log Mr. After partial reduction in a two-dimensional SDS-AGE, the proposed monomers and dimers produced two and four bands, respectively, likely indicating one to four chains crosslinked by disulfide bridges. Digestion with four different proteases yielded several equivalent fragments with molecular weights multiples of its minimum molecular weight (17.7 kDa). The circular dichroism spectrum of the protein showed a characteristic high α-helix content (70%). It was proposed that this hemoglobin is a pentamer with a molecular weight of aproximately 1.8×103 kDa, assembled by five 360-kDa subunits, each formed by two 180-kDa chains linked in pairs by disulfide bridges and each of these chains, in turn, comprised by ten heme binding domains linked in tandem. These data are compared to the published information for other planorbid extracellular hemoglobins.  相似文献

Mature Biomphalaria glabrata, submitted to four weeks of varied starvation times (0, 1, 3, 5, 6 & 7 d.week–1, were thereafter refed during four weeks. The different intermittent starvation times had no significant effect on snails survival. As weekly starvation increased, the rate of change in body weight and fecundity decreased. In snails fed one or two d.week–1, the rate of change in body weight was negative, while fecundity remained at a low level. Continuous hunger stopped oviposition.Starvation had no further effect on body growth after the first week of refeeding; however, its effect on fecundity remained significant over the two first weeks.  相似文献

Although gastropod myoglobin has been extensively studied, there are knowledge gaps in its biological characteristics. We describe for the first time the presence of a myoglobin in the triturative stomach of Biomphalaria gastropods. We compared biochemical parameters of myoglobins of stomach and radular muscles of Biomphalaria glabrata and Biomphalaria tenagophila. Apomyoglobin and holomyoglobin were obtained. Myoglobin was the most abundant protein in the stomach (85.0%) and radular muscles (80.0%) of the two Biomphalaria species evaluated. The Molecular mass and isoeletric point of stomach myoglobins were 16,124.93 Da and 7.98 and 16.095.28 Da and 7.77 for B. glabrata and B. tenagophila, respectively. Stomach myoglobins of B. glabrata and B. tenagophila rate autoxidation were equal to 8.0 × 10−4 h−1 ± 0.0002 and 1.0 × 10−4 h−1 ± 0.00142, respectively. Analysis of N-terminal amino acid sequencing revealed that stomach myoglobins are blocked in this region for a chemical group. Concluding, the differences we observed in the biochemical properties of stomach and radular myoglobins of B. glabrata and B. tenagophila suggest they may be isoform representing an evolutionary event related to the adaptation of these proteins.  相似文献

A sample of mud snails Hydrobia ulvae (Prosobranchia) from an intertidal population revealed that the shells of trematode-infected specimens were especially likely to be fouled with epibionts. Experimentally trematode-infected Biomphalaria glabrata (Pulmonata) appeared to be especially prone to develop epigrowth in comparison with uninfected conspecifics as well. These findings suggest an interaction between trematode infections and epibiosis in aquatic gastropods. The two most likely explanations for this are (1) that trematode infections weakens the snails' natural defences against epibionts, or (2) that the defences against epibionts also are effective against invading trematodes, causing snail specimens with a particularly good fouling defence to be less likely to become infected.  相似文献

Summary Strain SS86-4 was one of 40Bacillus brevis strains shown to be molluscicidal to the schistosomiasis snail vectorBiomphalaria glabrata. When grown in mB4 medium in 2-L fermentors, SS86-4 was molluscicidal only if fructose or phenylalanine was present in the medium. This is reminiscent of secondary fermentation factor effects, in this case an antioxidant effect. In vivo proteases also were capable of reducing molluscicidal activity. The molluscicidal toxin has an LC50 of 1 g toxin protein ml–1 (approx. 1 p.p.m.) and may be described as a small proteinaceous, heat-stable, oxygen-sensitive entity associated with the particulate portion of the cell wall fraction ofB. brevis that is formed prior to sporulation. Initial information indicates that its HPLC signature shows major peaks at 148.37 and 163.96 s and consists of two bands of approximately 5.3 kDa and 8.7 kDa on PAGE gel.  相似文献

Starvation of immature snails of Biomphalaria pfeifferi and B. glabrata results in an arrest of growth and animals remain immature. Spermatogenesis is limited to spermatogonia (B.g.) or spermatocytes 1 (B.p.). The number and the size of oocytes remain inferior to that of controls. Animals show reduced genital tracts.Once feeding is restored, growth is resumed but wet weight and shell diameter do not reach the same level as in controls. Fecundity and gametogenesis do not differ from that in controls. Genital tracts weight is proportional to body weight.  相似文献
