Abstract

In this study, four Co(III)-, Cu(II)-, Zn(II)- and Pd(II)-based potent antibacterial complexes of formula K3[Co(ox)3]·3H2O (I), [Cu(phen)2Cl]Cl·6.5H2O (II), [Zn(phen)3]Cl2 (III) and [Pd(phen)2](NO3)2 (IV) (where ox is oxalato and phen is 1,10-phenanthroline) were synthesized. They were characterized by elemental analysis, molar conductivity measurements, UV–vis, Fourier transform infrared (FT-IR) and proton nuclear magnetic resonance (1H-NMR) techniques. These metal complexes were ordered in three combination series of I+II, I+II+III and I+II+III+IV. Antibacterial screening for each metal complex and their combinations against Gram-positive and Gram-negative bacteria revealed that all compounds were more potent antibacterial agents against the Gram-negative than those of the Gram-positive bacteria. The four metal complexes showed antibacterial activity in the order I > II > III > IV, and the activity of their combinations followed the order of I+II+III+IV > I+II+III > I+II. The DNA-binding properties of complex (I) and its three combinations were studied using electronic absorption and fluorescence (ethidium bromide displacement assay) spectroscopy. The results obtained indicated that all series interact effectively with calf thymus DNA (CT-DNA). The binding constant (Kb), the number of binding sites (n) and the Stern–Volmer constant (Ksv) were obtained based on the results of fluorescence measurements. The calculated thermodynamic parameters supported that hydrogen bonding and van der Waals forces play a major role in the association of each series of metal complexes with CT-DNA and follow the above-binding affinity order for the series.

Interrelationships between H. vulgare (2x=14) and H. bulbosum (2x=14; 4x=28) were estimated on the basis of the karyotypes and the pairing behaviour of the chromosomes in diploid, triploid and tetraploid hybrids obtained with the aid of embryo culture. — A comparison of the karyotypes of the two species revealed similarities as well as differences. It was concluded that at least 4 or more of the chromosomes were similar in morphology and probably closely related. — Diploid and tetraploid hybrids are rarely obtained and their chromosome numbers tend to be unstable whereas triploid hybrids (1 vulgare + 2 bulbosum genomes) were stable and relatively easy to produce. In the diploid hybrid only 40% of the meiotic cells contained 14 chromosomes while the numbers ranged from 7 to 16 in other cells. All hybrids exhibited pairing between the chromosomes of the two species. Diploid hybrids had a mean of 5.0 and a maximum of 7 bivalents per cell in those cells having 14 chromosomes. Triploid hybrids from crosses between 2x H. vulgare and 4x H. bulbosum exhibited a mean of 1.5 and a maximum of 5 trivalents per cell. In a hexaploid sector found following colchicine treatment of a triploid the mean frequencies of chromosome associations per cell were: 5.5I+8.0II+0.7III+3.7IV+0.3V+0.4VI. One unstable 27 chromosome hybrid obtained from crosses between the autotetraploid forms had a mean of 1.1 and a maximum of 4 quadrivalents per cell. The chromosome associations observed in these hybrids are consistent and are taken as evidence of homoeologous pairing between the chromosomes of the two species. Interspecific hybridization between these two species also reveals that chromosome stable hybrids are only obtained when the genomes are present in a ratio of 1 vulgare2 bulbosum. Based upon the results obtained, the possibility of transferring genetic characters from H. bulbosum into cultivated barley is discussed.  相似文献

An unidentified species of the genus Austroagalloides is shown to have 7II+1IV instead of the normal 11II+XO configuration at metaphase I of meiosis in males. The quadrivalent manifests two types of pairing which ensures regular disjunction; normal chiasma-type pairing and distance pairing. It is suggested that the 7II+1IV form is derived from the 11II+XO form by a series of fusions.  相似文献

Crosses between the diploids G. oxylepis var. eligulata Steyermark (Mexico) and G. havardii Steyermark (New Mexico) and the tetraploid G. aphanactis Rydb. (New Mexico) were made. With G. aphanactis as the pistillate parent and G. havardii as the pollen parent a triploid hybrid was obtained in which the maximum meiotic configuration observed was 6m. The plant was 10 % fertile. Two triploid hybrid plants were also obtained when G. aphanactis was used as the pistillate parent and G. oxylepis var. eligulata was the pollen parent. One plant was about 20 % fertile and the other had a maximum configuration of 3II + 4III. The reciprocal cross produced a tetraploid which had a maximum configuration of 6II + 3IV and was 8 % fertile. The tetraploid plant undoubtedly resulted from the union of an unreduced gamete from the 2n parent and a normally reduced gamete from the tetraploid. Morphology of the hybrids was usually intermediate when compared with the parental species, although some characters in the triploids were those of the diploid parent. Chromosome end arrangements of the respective genomes and putative pairing relationships are presented and phylogenetic implications are discussed. It is concluded that G. aphanactis is more closely related to G. havardii than to G. oxylepis var. eligulata.  相似文献

Summary Plants with a pair of extra homologous telocentric chromosomes in addition to the normal chromosome complement are called ditelotetrasomics. Six types of ditelotetrasomics of barley have been obtained. Four types obtained in the selfed progenies of telotrisomics (Triplo 2S, Triplo 5S, Triplo 6S, and Triplo 7S) are reported in this paper. The ditelotetra 2S showed a stronger expression of the diagnostic characteristics of Triplo 2S. It was weak and small, with narrow, short, dark-green leaves, and was almost completely pollen and seed sterile. However, three other ditelotetrasomics (ditelotetra 5S, 6S, and 7S) did not show specific diagnostic characteristics and were similar to normal diploid plants, with the exception of ditelotetrasomic 5S, which showed some effects. At meiotic diakinesis and metaphase I, these ditelotetrasomic plants showed chromosome configurations of 6II+1IV, 7II+1 teloII, 6II+1III+1teloI, or 7II+2 teloI. Most of the sporocytes at anaphase and telophase in the first and second meiotic divisions showed almost normal chromosome behavior. Quartets were mostly normal with no micronuclei. Approximately 30% of the selfed progenies of these three ditelotetrasomics were ditelotetrasomics and almost 50% were telotrisomics, indicating a high percentage of male and female transmission of the extra telocentric chromosomes.  相似文献

The karyotype and meiosis of the 12-ploid plants—one of the offspring of the natural F1 hybrid (Aster ageratoides subsp.ovatus (2n=36) ×Kalimeris incisa (2n=72), 2n=72)—were examined. The 2n=108 chromosomes of the 12-ploids were found to consist of 18 large chromosomes and 90 small chromosomes. In meiosis of the PMCs of the 12-ploid, chromosome configurations of 3III+46II+7I, 2III+48II+6I and 3III+47II+5I were observed. All the univalents and trivalents were small, and among the 46–48 bivalents nine were large and the remaining 37–39 were comparatively small. The large bivalents were found to represent autosyndetic pairings, and the small bivalents and trivalents were probably formed by autosyndetic pairings. The large chromosomes of the 12-ploids were found to coincide with the large chromosomes ofovatus, and the 90 small chromosomes to correspond to small chromosomes ofovatus andK. incisa. The 12-ploids were concluded to have been produced by a fusion of an unreduced gamete of the F1 plant and a reduced gamete ofK. incisa which was growing in proximity to the F1s. Thus the 12-ploids were regarded to be an amphidiploid having 36 chromosomes ofovatus and 72 chromosomes ofK. incisa.  相似文献

Bioconversion of quercetin glucosides using four generally recognized as safe (GRAS) organisms (Aspergillus oryzae, Bacillus subtilis, Lactobacillus plantarum, and Saccharomyces cerevisiae) was evaluated by measuring changes in the levels of quercetin compounds of onion. Of the four organisms, S. cerevisiae increased the content of quercetin-3-O-β-d-glucoside (III; isoquercitrin) and quercetin (IV), whereas decreasing quercetin-3,4′-O-β-d-glucoside (I) and quercetin-4′-O-β-d-glucoside (II). Also, S. cerevisiae converted authentic compound I to III, and II to IV, respectively. These results suggest that S. cerevisiae can be used to increase the levels of isoquercitrin (III), the most bioavailable quercetin compound in onion.  相似文献

 Reactions (25  °C) of galactose oxidase, GOaseox from Fusarium NRRL 2903 with five different primary-alcohol-containing substrates RCH2OH:- D-galactose (I) and 2-deoxy-d-galactose (II) (monosaccharides); methyl-β-d-galactopyranoside (III) (glycoside);d-raffinose (IV) (trisaccharide); and dihydroxyacetone (V) have been studied in the presence of O2. The GOaseox state has a tyrosyl radical coordinated at a square-pyramidal CuII active site, and is a two-equivalent oxidant. Reactant concentrations were [GOaseox] (0.8–10 μM), RCH2OH (1.0–6.0 mM), and O2 (0.14–0.29 mM), with I=0.100 M (NaCl). The reactions, monitored at 450 nm by stopped-flow spectrophotometry, terminated with depletion of the O2. Each trace was fitted to the competing reactions GOaseox+RCH2 OH → GOaseredH2+RCHO (k 1), and GOaseredH2+O2→ GOaseox+H2O2 (k 2), with GOaseredH2 written as the doubly protonated two-electron-reduced CuI product. It was necessary to avoid auto-redox interconversion of GOaseox and GOasesemi . Information obtained at pH 7.5 indicates a 5 : 95 (ox : semi) "native" mix equilibration complete in ∼3 h. At pH >7.5, rate constants 10–4k 1 / M–1 s–1 for the reactions of GOaseox with (I) (1.19), (II) (1.07), (III) (1.29), (IV) (1.81), (V) (2.94) were determined. On decreasing the pH to 5.5, k 1 values decreased by factors of up to a half, and acid dissociation pK as in the range 6.6–6.9 were obtained. UV-Vis spectrophotometric studies on GOaseox gave an independently determined pK a of 6.7. No corresponding reactions of the Tyr495Phe variant were observed, and there are no similar UV-Vis absorbance changes for this variant. The pK a is therefore assigned to protonation of Tyr-495 which is a ligand to the Cu. The rate constant k 2 (1.01×107 M–1 s–1) is independent of pH in the range 5.5–9.0 investigated, suggesting that H+ (or H-atoms) for the O2 → H2O2 change are provided by the active site of GOasered . Natural intergeneric hybrids betweenAster ageratoides subsp.ovatus (2n=36) andKalimeris incisa (2n=72) were found. All of the hybrids studied were found to have 2n=72, 18 more chromosomes than a regular F1 hybrid. The hybrids were found to be of two types: one having 18 large chromosomes ofovatus, and the other having 9 large chromosomes of the same subspecies. In meiosis of the PMCs of the hybrid with 18 large chromosomes, a regular chromosome configuration, 36II, was observed. In PMCs of the hybrid with 9 large chromosomes an irregularity of chromosome pairings was observed, showing varied chromosome configurations: 35II+2I, 34II+4I, 33II+6I, IIII+33II+3I, 1IV+32II+4I, 32II+8I, 31II+10I, 29II+14I, 3III+29II+5I. Most univalents were large, but a few were small. The hybrids with 18 large chromosomes were found to be partial amphidiploid and possessing double chromosome complements ofovatus. The hybrids with 9 large chromosomes were found to be the first backcrossed generation between the hybrid with 18 large chromosomes andK. incisa.  相似文献

Interspecific hybridization was achieved between cowpea (Vigna unquiculata [L.] Walp) and a hairy wild relative (V. pubescens). Hybrid embryos which otherwise would have shrivelled and failed to germinate were excised prematurely and cultured on a solidified MS medium. The F1 plants were vigorous in growth, partially sterile, slightly hairy and showed some intermediate features between the two parental types. Cytological investigations on F1 pollen mother cells showed average chromosome associations per cell of 0.66I+10.00II+0.34IV.  相似文献

Summary A total of 1475 individuals belonging to 43 natural populations of seven diploid (2x) blueberry species (Vaccinium section Cyanococcus) and two natural interspecific 2x hybrid populations were evaluated for unreduced pollen production. Significant differences were found in the frequency of unreduced pollen producers between species and within and between populations of the same species. Individuals with 1% or more unreduced pollen were considered unreduced pollen producers. The average frequency of unreduced pollen producers in these diploid species was 13.5%, ranging from 7.4% (V. corymbosum) to 18.4% (V. darrowii). The frequency of unreduced pollen grains in individual clones varied from <-1% to 28.6%. The production of unreduced pollen was not associated with male fertility. Antibacterial activity was tested for each of these four metal complexes and their combinations against Gram-positive and Gram-negative bacteria. All compounds were more potent antibacterial agents against the Gram-negative than those of the Gram-positive bacteria. The four metal complexes showed antibacterial activity in the order I > II > III > IV and the activity of their combinations followed the order of I + II + III + IV > I + II + III > I + II. CT-DNA binding studies of complex I and its three combinations were carried out using UV–vis spectral titration, displacement of ethidium bromide (EB), and electrophoretic mobility assay. The results obtained from UV–vis studies indicated that all series interact effectively with CT-DNA. Fluorescence titration revealed that the complexes quench DNA-EB strongly through the static quenching procedures. The binding constant (Kb), the Stern–Volmer constant (Ksv), and the number of binding sites (n) were determined at different temperatures of 293, 300, and 310 K, respectively. The calculated thermodynamic parameters supported that hydrogen binding and Van der Waals forces play a major role in association of each series of metal complexes with CT-DNA and follow the above-binding affinity order for the series.  相似文献

Summary Intergeneric hybrids recovered through plant regeneration from embryo callus culture and their colchicine-induced amphiploids were obtained from a cross of Elymus canadensis with Secale cereale (cv Gazeller). The embryo-callus-regenerated F1 plants grew vigorously to maturity and regrew well after clipping, while the embryo-rescued F1 died of hybrid necrosis before maturity. Most of the morphological characters of the sterile F1 hybrids were intermediate between the parents, but tiller number and dry matter yield were higher than the parents. Amphiploids from these F1 plants had improved fertility but were less vigorous than the F1 plants. The predominance of univalents in the F1 and bivalents in the amphiploids indicated that the genomes S, H, and R were distinct. However, the occasional occurrence of multivalents reflected a random, intergenomic or intragenomic pairing. The mean chromosome associations of the F1 (2n=21, SHR), the C0 amphiploids (2n=42, SSHHRR), and the C1 amphiploid (2n=40) at metaphase I (MI) were 16.51I+2.05II+0.06III+0.02IV+0.02V, 2.20I+19.87II +0.02IV, and 7.10I+16.37II+0.04III+0.02IV, respectively. These amphiploids could be exploited as new germplasm for forage crop improvement by controlled introgression and backcrosses to the parents.  相似文献

To study K+ channels in the basolateral membrane of chloride-secreting epithelia, rat tracheal epithelial monolayers were cultured on permeable filters and mounted into an Ussing chamber system. The mucosal membrane was permeabilized with nystatin (180 μg/ml) in the symmetrical high K+ (145 mm) Ringer solution. During measurement of the macroscopic K+ conductance properties of the basolateral membrane under a transepithelial voltage clamp, we detected at least two types of K+ currents: one is an inwardly rectifying K+ current and the other is a slowly activating outwardly rectifying K+ current. The inwardly rectifying K+ current is inhibited by Ba2+. The slowly activating K+ current was potentiated by cAMP and inhibited by clofilium, phorbol 12-myristae 13-acetate (PMA) and lowering temperature. This is consistent with the biophysical characteristics of I SK channel. RT-PCR analysis revealed the presence of I SK cDNA in the rat trachea epithelia. Although 0.1 mm Ba2+ only had minimal affect on short-circuit current (I sc) induced by cAMP in intact epithelia, 0.1 mm clofilium strongly inhibited it. These results indicate that I SK might be important for maintaining cAMP-induced chloride secretion in the rat trachea epithelia. Received: 1 March 1996/Revised: 5 August 1996  相似文献

Most individuals of the loach Misgurnus anguillicaudatus reproduce bisexually, but cryptic clonal lineages reproduce by natural gynogenesis of unreduced diploid eggs that are genetically identical to maternal somatic cells. Triploid progeny often occur by the accidental incorporation of a sperm nucleus into diploid eggs. Sex reversal from a genetic female to a physiological male is easily induced in this species by androgen treatment and through environmental influences. Here, we produced clonal tetraploid individuals by two methods: 1) fertilization of diploid eggs from a clonal diploid female with diploid sperm of a hormonally sex-reversed clonal diploid male and 2) artificial inhibition of the release of the second polar body in eggs of clonal diploid females just after initiation of gynogenetic development. There is no genetic difference between the clonal diploid and tetraploid individuals except for the number of chromosome sets or genomes. Clonal tetraploid males never produced unreduced tetraploid sperm, only diploid sperm that were genetically identical to those of a clonal diploid. Likewise, clonal tetraploid females did not form unreduced tetraploid eggs, just diploid eggs. However, the eggs' genotypes were identical to those of the original clone, and almost all the eggs initiated natural gynogenesis. Thus, gametogenesis of the clonal tetraploid loach is controlled by the presence of two chromosome sets to pair, thereby preserving the normal meiotic process, i.e., the formation of bivalents and subsequently two successive divisions.  相似文献

The majority of tetraploid peonies are allopolyploids derived from crosses between phylogenetically distinct diploid lineages. Tetraploid Paeonia obovata was previously considered to be an autopolyploid because it is morphologically indistinguishable from the diploid of the same species. The presence of the Adh2 gene in tetraploid P. obovata but the inability to amplify the Adh2 gene from Chinese diploids of P. obovata, however, suggests that the tetraploid was not an autotetraploid derivative of the geographically adjacent diploid populations in China. The Adh gene phylogenies rather suggest that the tetraploid originated from crosses between two geographical races of diploid P. obovata distributed in China and Japan. The intermediate status of tetraploid P. obovata between auto‐ and allopolyploidy highlights the need for population genetic analyses of polyploid origins along the continuous range of genomic divergence. Here we present a model that describes the probabilities of polyploid formation and establishment as a function of genomic divergence between diploid progenitors. The probability of polyploid formation (Pf) is obtained from the multiplication of the probability of production of unreduced gametes (Pg) and the probability of ‘hybridization’ (Ph). Pf stays relatively stable when the genomic divergence is low, and then decreases progressively rapidly with the increase of genomic divergence between diploid progenitors. The probability of polyploid establishment (Pe), which depends on the rate of appearance of stable beneficial gene combinations and the rate of fertility restoration, is positively correlated with the genomic divergence of diploid parents. Muscarinic receptor-linked G protein, G i , can directely activate the specific K+ channel (I K(ACh)) in the atrium and in pacemaker tissues in the heart. Coupling of G i to the K+ channel in the ventricle has not been well defined. G protein regulation of K+ channels in isolated human ventricular myocytes was examined using the patch-clamp technique. Bath application of 1 μm acetylcholine (ACh) reversibly shortened the action potential duration to 74.4 ± 12.1% of control (at 90% repolarization, mean ±sd, n= 8) and increased the whole-cell membrane current conductance without prior β-adrenergic stimulation in human ventricular myocytes. The ACh effect was reversed by atropine (1 μm). In excised inside-out patch configurations, application of GTPγS (100 μm) to the bath solution (internal surface) caused activation of I K(ACh) and/or the background inwardly-rectifying K+ channel (I K1) in ventricular cell membranes. I K(ACh) exhibited rapid gating behavior with a slope conductance of 44 ± 2 pS (n= 25) and a mean open lifetime of 1.8 ± 0.3 msec (n= 21). Single channel activity of GTPγS-activated I K1 demonstrated long-lasting bursts with a slope conductance of 30 ± 2 pS (n= 16) and a mean open lifetime of 36.4 ± 4.1 msec (n= 12). Unlike I K(ACh), G protein-activated I K1 did not require GTP to maintain channel activity, suggesting that these two channels may be controlled by G proteins with different underlying mechanisms. The concentration of GTP at half-maximal channel activation was 0.22 μm in I K(ACh) and 1.2 μm in I K1. Myocytes pretreated with pertussis toxin (PTX) prevented GTP from activating these channels, indicating that muscarinic receptor-linked PTX-sensitive G protein, G i , is essential for activation of both channels. G protein-activated channel characteristics from patients with terminal heart failure did not differ from those without heart failure or guinea pig. These results suggest that ACh can shorten the action potential by activating I K(ACh) and I K1 via muscarinic receptor-linked G i proteins in human ventricular myocytes. Cytology of a pentaploid hybrid (2n=60) produced by crossing an autotetraploid (2n=48) of a diploid race of Solanum nigrum L. as seed parent with the naturally occurring hexaploid S. nigrum (2n=72) was studied which showed approximately 24II+12I. Chromosome pairing in the gametic complement of the hexaploid was deduced to be 12II+12I and on this basis it is concluded that the natural hexaploid is an autoallohexaploid, which has three sets (genomes) of 12 chromosomes each, two of which are to a large extent homologous to each other. Theoretically the genomic formula can be tentatively written as AAAABB. Since the natural hexaploid does not show any quadrivalents between the four homologous A genomes, chromosome pairing appears to be restricted to bivalent formation by a special genetic mechanism. The probability that S. nigrum could be an autohexaploid, parallelling Pheleum pratense, has been discussed.  相似文献

The present study investigated both HCO 3 and Cl secretions in a human pancreatic duct cell line, CAPAN-1, using the short-circuit current (I sc ) technique. In Cl/HCO 3-containing solution, secretin (1 μm) or forskolin (10 μm) stimulated a biphasic rise in the I sc which initially reached a peak level at about 3 min and then decayed to a plateau level after 7 min. Removal of external Cl abolished the initial transient phase in the forskolin-induced I sc while the plateau remained. In HCO 3/CO2-free solution, on the contrary, only the initial transient increase in I sc was prominent. Summation of the current magnitudes observed in Cl-free and HCO 3-free solutions over a time course of 10 min gave rise to a curve which was similar, both in magnitude and kinetics, to the current observed in Cl/HCO 3-containing solution. Removal of external Na+ greatly reduced the initial transient rise in the forskolin-induced I sc response, and the plateau level observed under this condition was similar to that obtained in Cl-free solution, suggesting that Cl-dependent I sc was also Na+-dependent. Bumetanide (50 μm), an inhibitor of the Na+-K+-2Cl cotransporter, and Ba2+ (1 mm), a K+ channel blocker, could reduce the forskolin-induced I sc obtained in Cl/HCO 3-containing or HCO 3-free solution. However, they were found to be ineffective when external Cl was removed, indicating the involvement of these mechanisms in Cl secretion. On the contrary, the HCO 3-dependent (in the absence of external Cl) forskolin-induced I sc could be significantly reduced by carbonic anhydrase inhibitor, acetazolamide (45 μm). Basolateral application of amiloride (100 μm) inhibited the I sc ; however, a specific Na+-H+ exchanger blocker, 5-N-methyl-N-isobutylamiloride (MIA, 5–10 μm) was found to be ineffective, excluding the involvement of the Na+-H+ exchanger. However, an inhibitor of H+-ATPase, N-ethylmaleimide did suppress the I sc (IC50= 22 μm). Immunohistochemical studies also confirmed the presence of a vacuolar type of H+-ATPase in these cells. H2DIDS (100 μm), an inhibitor of Na+-HCO 3 cotransporter, was without effect. Apical addition of Cl channel blocker, diphenylamine-2,2′-dicarboxylic acid (DPC, 1 mm), but not disulfonic acids, DIDS (100 μm) or SITS (100 μm), exerted an inhibitory effect on both Cl and HCO 3-dependent forskolin-induced I sc responses. Histochemical studies showed discrete stainings of carbonic anhydrase in the monolayer of CAPAN-1 cells, suggesting that HCO 3 secretion may be specialized to a certain population of cells.
