Eleven pairs of Undaria pinnatifida (Harv.) Suringar gametophytes were identified with random amplified polymorphic DNA (RAPD) technique. After screening 100 primers, 20 ten-base primers were determined for the RAPD analysis. A total of 312 polymorphic loci were obtained, of which 97.7% were polymorphic. The primer S198 was found to distinguish all the selected Undaria
pinnatifida gametophytes. The genetic distances between each two of the twenty-two U.
pinnatifida gametophytes ranged from 0.080 to 0.428, while the distances to the Laminaria was 0.497 on average. After reexamination, two sequences characterized amplification region (SCAR) markers were successfully converted, which could be applied to U.
pinnatifida germplasm identification. All these results demonstrated the feasibility of applying RAPD markers to germplasm characterization and identification of U. pinnatifida gametophytes, and to provide a molecular basis for Undaria breeding. 相似文献

The physiological performances of tetrasporophytes and gametophytes of Gelidium canariensis (Grunow) Seoane-Camba were compared to estimate whether the field predominance of tetrasporophytes is due to lower fitness of gametophytes. No significant differences between tetrasporophytes and gametophytes were detected for calorific content, protein and pigment concentrations, NADH-Diaphorase, alkaline phosphatase and glucose-6-phosphate dehydrogenase activities and photosynthesis and respiration at 15, 20 and 25 °C, and pH 6.5, 8.2 and 9.2. Our results indicate that these physiological characteristics are not responsible for the scarcity of gametophytes in the field populations of G. canariensis.Abbreviations ALP =

The cell wall polysaccharides of two species of red algae, which are adapted to both freshwater and marine environments, were analysed to determine the effect of these widely different environments on their commercially important agarocolloids and to investigate the possible role of the cell wall in environmental adaptation. Cell wall polymers of freshwater isolates of Bangia atropurpurea (Roth) C. Agardh and cultured freshwater and marine Bostrychia moritziana (Sonder ex Kützing) J. Agardh were isolated and the polysaccharides chemically fractionated and characterized. Wall polysaccharides of freshwater B. atropurpurea were similar to those previously reported for marine isolates with repeating disac-charide units of agarose and porphyran predominant in the hot water extracts. In the insoluble residues, 3-iinked galactosyl and 4-linked mannosyl residues were predominant. Bostrychia moritziana wall polysaccharides included agarocolloids with various patterns of methyl ether substitution similar to those previously described for other Ceramiales. Differences in the position of methyl ether substituents were detected in the hot water extracts of the freshwater and marine specimens. Polymers of freshwater ß. moritziana cultures were composed of a complex mixture of repeating disaccharide units including 2′-O-methyl agarose, 6-O-methyI agarose and 2′-O-methyl porphyran. Polymers of marine isolates of ß. moritziana differ in that they contain only trace amounts of 2-O-methyl saccharides and increased amounts of 6-O-amethyl saccharides. The hot water insoluble residues of both freshwater and marine isolates of ß. moritziana contain a mixture of 3-linked galactosyl and 4-linked glucosyl residues. These results indicate that the adaptive response of B. moritziana to changing osmotic and ionic conditions may include changes in cell wall chemistry: notably, the pattern of methyl ether substitution. 相似文献

Summary The red alga,Antithamnion kylinii Gardner, was found to have needle-shaped inclusions about 10 m long and less than 0.4 m thick. They ranged in abundance from one or a few in young cells to hundreds in fully enlarged cells. Under polarized light, the inclusions were birefringent, indicating crystallinity. Solubility tests suggested that the inclusions were composed of calcium oxalate: they dissolved in 1 N hydrochloric acid and in a saturated solution of aqueous cupric acetate, but they were not soluble in 10 N acetic acid or 5.25% sodium hypochlorite. X-ray microanalysis confirmed the presence of calcium. Calcium oxalate crystals were present in cells of indeterminate axes, but cells of determinate lateral filaments lacked them. Light and electron microscopic study demonstrated that the crystals were associated with the parietal cytoplasm. Calcium oxalate crystals were also present inA. defectum Kylin, but they were not found in ten more distantly related taxa. 相似文献

The paleozoic marine algae of Central Asia were recently investigated and attributed to warm-water and Tethyan microflora. If the majority of the taxa (114 out of 117) were already described from other basins, a few elements merit attention. Two chlorophyte species are new (Atractyliopsis nuratauensis nov. sp., Amarellina moscoviensis nov. sp.) and a new Hoeegonites sp. is left in open nomenclature. The flora also contains representatives of the puzzling Moniliporelleae, a rhodophyte family only reported from Kazakhstan with the genus Furcatoporella. An unusual in situ cluster of calcispheres is also illustrated. 相似文献

In this first sequence analysis of ‘zooxanthellae’ (symbiotic algae of the genus Symbiodinium) in scleractinian corals in Africa, seven Kenyan species sampled in 2001–2002 were analysed by RFLP and sequencing of a PCR-amplified fragment of the LSU rRNA gene. Zooxanthellae of phylotypes A, C and D, all described previously in corals from other regions of the world, were detected. All sequences of phylotype D were identical, while phylotype C was variable, with 14 distinct sequences, seven of which clustered in a previously unreported subgroup of phylotype C, among the 22 samples. These data on the diversity of zooxanthellae in Kenyan corals 3–4 years after the 1998 bleaching event are of potential value for longitudinal studies of temporal changes in zooxanthella diversity in Kenyan corals, especially in relation to future large-scale bleaching episodes. 相似文献

Marine, estuarine and freshwater isolates of Caloglossa leprieurii (Montagne) J. Agardh exhibit a high salinity tolerance, reflected by broad cell viability and growth rate. Osmotic adjustment is shown to rely to a large extent on ion-transport systems, with K+ and Cl- accumulated in osmotically- significant quantities and active Na+ extrusion. The ion concentrations contribute a large proportion (67–94%) to internal osmotic pressure. The concentration of the organic osmolyte mannitol in all populations was strongly salinity dependent. Mannitol made a lower contribution to the internal osmotic pressure, when compared to ion concentrations, but nonetheless represented an important proportion of the internal osmolality. The response of the three isolates is discussed in relation to the salinity of their respective environments. 相似文献

Red algae genus Laurencia (Rhodomelaceae, Ceramiales) are known to produce a wide range of chemically interesting secondary halogenated metabolites. This investigation delves upon extraction, isolation, structural elucidation and antibacterial activity of inherently available secondary metabolites of Laurencia majuscula Harvey collected from two locations in waters of Sabah, Malaysia. Two major halogenated compounds, identified as elatol (1) and iso-obtusol (2) were isolated. Structures of these compounds were determined from their spectroscopic data such as IR, 1H-NMR, 13C-NMR and optical rotation. Antibacterial bioassay against human pathogenic bacteria was conducted using disc diffusion (Kirby-Bauer) method. Elatol (1) inhibited six species of bacteria, with significant antibacterial activities against Staphylococcus epidermis, Klebsiella pneumonia and Salmonella sp. while iso-obtusol (2) exhibited antibacterial activity against four bacterial species with significant activity against K. pneumonia and Salmonella sp. Elatol (1) showed equal and better antibacterial activity compared with tested commercial antibiotics while iso-obtusol (2) only equaled the potency of commercial antibiotics against K. pneumonia and Salmonella sp. Further tests conducted using dilution method showed both compounds as having bacteriostatic mode of action against the tested bacteria. 相似文献

Material of the red alga Odonthalia floccosa (Esper) Fal-kenberg (Rhodomelaceae, Ceramiales), collected from California, was cultured in the laboratory and its life-history was completed. Tetraspores grew into bipolar sporelings that differentiated into a colorless rhizoidal portion and a pigmented upright shoot. The sporelings became compressed apically and formed lateral branches in a regularly distichous manner that were congenitally fused with the main axis. These tetraspore germlings grew into diecious gametophytes. Male ga-metophytes produced numerous spermatangia on modified fertile branchlets (male trichoblasts) that possessed three to four monosiphonous, proximal segments. Female gametophytes formed a single pro-carp on the suprabasal segment of unbranched female trichoblasts. Cystocarps developed on the female gametophytes cocultured with male gametophytes and released viable carpospores that developed into fertile te-trasporophytes. Tetrasporangia were produced from the third and fourth periaxial cells in each of 12–45 successive fertile segments and provided three (two lateral and one basal) cover cells. The occurrence of both spermatangia and procarps on fertile trichoblasts in O. floccosa suggests that the alga is the most derived in these two characters among the species of the genus Odonthalia. This species is distributed in cold temperate regions in the North Pacific, and it should be excluded from the North Atlantic marine algal flora. 相似文献

The red alga Laurencia nipponica Yamada (Rhodomelaceae, Ceramiales) is known to contain several chemical races, each of which is characterized by a particular, major halogenated secondary metabolite. Both field-collected and cultured plants of a population of this species found recently at Shikanoshima Island, Fukuoka Prefecture, southern Japan, produced C15 bromoethers, (3E)-laureatin and (3E)-isolaureatin, and sesquiterpenoids, 2,10-dibromo-3-chloro-9-hydroxy-α-chamigrene and 2,10-dibromo-3-chloro-α-chamigrene. Laurencia nipponica can be referred to as a further chemical race that is characterized by the production of two C15 bromoethers, (3E)-laureatin and (3E)-isolaureatin, and a sesquiterpenoid, 2,10-dibromo-3-chloro-9-hydroxy-α-chamigrene as major compounds. 相似文献

On Normandy coasts, the red alga Delesseria sanguinea perennates by its stipe; fronds grow in January and disappear in June. Seasonal variations in sterol composition in relation to the biology of D. sanguinea are reported. Sterols in cellular membranes are free or conjugated by esterification with fatty acids, heterosides or lipid complexes like phospholipids. Both kinds of sterols were analyzed by GC-MS. The major sterol (80%) found in fronds was cholesterol whereas in stipes, cholesterol was also the major sterol in spring, but in September, an important reduction in cholesterol yield was noted with proportional increase in sitosterol content. It appears that cholesterol is synthesized in fronds in spring, then transferred to the stipe, which loses an important amount of cholesterol with loss of the blades. 相似文献

Photosynthesis and growth rates of Laurencia brongniartii J. Agardh (Rhodophyta, Ceramiales) in preparation for cultivation 总被引:1,自引:1,他引:1
Laurencia brongniartii is usually found at depths below 4 m, but can be found in shallow subtidal areas in crevices and on the walls of a coral reef in Amami Oshima Island, Kagoshima Prefecture, Japan, where irradiances were significantly lower than those at similar depths in open water. In preparation for the possible cultivation of this species for its antibiotic compounds, the effects of temperature and irradiance on photosynthesis and growth were measured. Photosynthesis and growth rates of L. brongniartii explants were highest at 26 and 28 °C, which closely corresponded to temperatures found during August to late December when it was most abundant. The estimated maximum photosynthesis rate (P
max) was 4.41 mol photon m–2 s–1 at 26 °C and 4.07 mol photon m–2 s–1 at 28 °C. Saturating irradiance occurred at 95 mol photon m–2 s–1 at 26 °C and 65 mol photon m–2 s–1 at 28 °C. In contrast, growth experiments at 41.7 mol photon m–2 s–1 caused bleaching of explants and the maximum growth rate observed during the study was 3.02 ± 0.75% day–1 at 28 °C and 25 mol photon m–2 s–1. The difference in the saturating irradiance for photosynthesis and the irradiance that caused bleaching in growth experiments suggests that long-term exposure to high irradiance was detrimental and should be addressed before the initiation of large scale cultivation. 相似文献

The growth of Laurencia okamurae and the content of laurinterol and debromolaurinterol were influenced by various factors.
Temperature influenced growth raere te with a maximum at 25°C, regardless of daylength. Maturity depended on temperature rather
than daylength; the plants grew without maturing at 15°C, while the plants matured within two weeks at 25°C. The plants were
able to grow at salinities of 14–50‰ with maximum growth at 26‰. The salinities growth rash;50‰te reduced with decreasing
concentration of nitrate below 1.2 × 10-3 M, and of total phosphate below 7.5× 10-8 M. Bromide concentration had no effect on growth, and the plants grew without bromide. Variation in temperature between 15–25°C
and daylength produced no obvious change in laurinterol and debromolaurinterol contents. On the other hand, an increase in
salinity led to an increase for both. The plants cultured in a completely artificial medium, modified ASP12NTA, showed a marked drop in their content of these metabolites. An increase in concentrations of nitrate, total phosphate
or bromide did not restore the content.

Macroscopic red algae were sampled simultaneously with diatom sampling from 60 locations in central Finnish rivers and at some rivers in south coast rivers in the summers 1986, 1993 and 1994. In those samples 17 red algal taxa from 5 genera ( Audouinella (3). Batrachospermum (7). Lemanea (4), Sirodotia (2) and Tuomeya (1)) were identified. Reproductive structures of Laurencia pelagosae are described for the first time. Tetrasporangia, in parallel arrangement, are cut off from the mother cells adaxially; antheridial branches, which are unramified, are inserted in depressions lacking a row of axial cells on their bases; cystocarps are sessile and subspherical. The occurrence of secondary pit-connections between epidermal cells and of lenticular thickenings in the walls of medullary cells is documented. Gametophytes of Pellaea viridis that appeared spontaneously on the surface of substratum originating from an ultramafic area were found to form mycothallic symbiosis with arbuscular mycorrhizal fungi (AMF) under laboratory conditions. In gametophytes and sporophytes grown with Glomus tenue, abundant arbuscule formation was observed at both stages. In gametophytes, the fungus was found in the region where the rhizoids are initiated. If G. intraradices was added to the soil, the gametophytes were colonised mostly by G. tenue, and roots of sporophytes were colonised by G. intraradices. The presence of AM fungi in both gametophytes and sporophytes of P. viridis
resulted in the development of larger leaf area and root length of the sporophyte. The analysis of gametophytes from the Botanical Garden in Krakow (Poland) showed that cordate gametophytes of Pteridales, namely Pellaea viridis (Pellaeaceae), Adiantum raddianum and A. formosum (Adiantaceae), were also mycothallic. 相似文献

Purification of mannitol-l-phosphatase, an enzyme catalyzing the final step of mannitol biosynthesis, was first achieved
in the mannitol-accumulating red alga Caloglossa continua (Okamura) King et Puttock. The enzyme was shown to be a monomer, since gel filtration and sodium dodecyl sulfate–polyacrylamide
gel electrophoresis gave close values of apparent molecular weights of 28,500 and 30,200, respectively. The protein exhibited
an isoelectric point of 4.8. The substrate specificity for mannitol-l-phosphate (MIP) was very high, and that for K
m(MIP) was 0.41 mM. The catalytic activity was optimal at pH 7.4. The enzyme was activated by Mg2+, but was strongly inhibited by Ca2+, NaF, N-ethylmaleimide, and p-hydroxymercuribenzoic acid. Seawater levels of NaCl and physiological levels of mannitol also inhibited the activity by 50%
or more. Changes in the concentrations of those ions and metabolites may regulate the biosynthesis of mannitol as an osmoregulant
in vivo.

Habitat forming algae play an important role in the ecology of temperate reefs worldwide. Despite this, our understanding of levels of gene flow within and among populations of algae is largely limited to studies on intertidal species; we know comparatively little about important habitat-forming subtidal algae. Here, we develop eight polymorphic microsatellite markers for the characterisation of population genetic diversity and structure in the subtidal kelp, Ecklonia radiata. This large macroalga is the most abundant habitat-forming kelp on the subtidal rocky reefs of temperate Australia and New Zealand where it forms extensive forests that support an astounding diversity of associated taxa. 相似文献

Early detection is the most effective way to mitigate the effects of harmful algal blooms (HAB). Cell counts based on examination of microplankton samples using settling chambers and visual inspection with an inverted microscope are tedious and time consuming, and counting precision is generally poor at low cell densities. The FlowCAM is a continuous imaging flow cytometer designed to characterize particles in the microplankton size range (20–200 μm diameter). In this study we examined the ability of the FlowCAM to improve routine monitoring protocols for HAB species by automatically recording information on size and fluorescence per cell. This will eliminate the need to examine cells outside the ranges of these measurements for our target species, Karenia brevis. We also tested the ability of image comparison software to match images of cells in mixed assemblages to images of the target species. For simple mixtures of cultured dinoflagellates, the ability of the image matching software to discriminate target cells varied greatly depending on how similar the two species were in size and shape. When target cells were added to natural plankton samples, the image recognition software correctly identified 80–90% of the target cells, but misidentified 20–50% of non-target cells in the size range of the target species. We conclude that the FlowCAM is less tedious and time-consuming than microscopy, allowing for examination of more cells for greater counting precision. The cell recognition software helps reduce the numbers of cells that must be screened, but images must still be examined by a trained operator to identify the HAB species of interest. 相似文献
