高山生态系统的主要气候特征是温度变化幅度较大,目前研究主要集中于高山植物的抗冻机制,而很少关注其对极端高温(高于45℃)的适应性。本研究发现高山物种圆锥南芥跟拟南芥比,具有更强的基础耐热性和获得性耐热性。通过叶绿素荧光检测发现在极端高温下,圆锥南芥具有更稳定的光系统II和更有效的能量耗散机制来维持更高水平的光合效率。通过电导率和丙二醛含量的检测发现圆锥南芥的膜伤害更小,膜流动性与脂肪酸链的长度和不饱和度紧密相关,圆锥南芥脂肪酸具有更低的16∶3含量,更长的碳链,不饱和度没有明显的变化,这些可能有助于维持膜的稳定。另外,更高表达量的HSP101和HSP70可能为圆锥南芥提供了更好的保护作用。以上结果表明,圆锥南芥能利用生理生化活动的调整来适应高山环境中的极端高温,这种耐热策略与低地耐热植物相似,因此圆锥南芥具有系统性耐热能力,可以作为研究植物耐热分子机制的模式物种。  相似文献

Low non-freezing temperature is one of the major environmental factors that affect metabolism, growth, development and geographical distribution of chilling-sensitive plants, Jatropha curcas, a chilling-sensitive plant, which is considered as a sustainable energy plant with great potential for biodiesel production. Our previous studies showed that short-term chilling shock at 5 °C for 4 h and long-term chill hardening at 12 °C 1 or 2 days could improve chilling tolerance of J. curcas seedlings, but lipidomic response to chilling shock and chill hardening has not been elucidated. In this study, membrane lipid composition change in J. curcas seedlings during chilling shock and chill hardening was investigated by liquid chromatography-electrospray ionization-mass spectrometry (LC–ESI–MS) approach. The results indicated that the relative abundances of nine classes and 72 species of membrane lipids, such as phosphatidylethanolamine (PE), phosphatidylcholine (PC), phosphatidylglycerol (PG), phosphatidylinositol (PI), lysophosphatidylcholine (lysoPC) and lysophosphatidylglycerol (lysoPG), two glycolipids digalactosyldiacylglycerol (DGDG) and monogalactosyldiacylglycerol (MGDG) and a sulfoquinovosyldiacylglycerol (SQDG), were significantly changed, and the degree of unsaturation of above-mentioned cellular membrane lipids with fatty acid differing in chain lengths and the number of double bonds also increased in varying degrees during chilling shock and chill hardening. These results suggested that remodeling and increase in the degree of unsaturation of membranes lipids may be a common physiological basis for short-term chilling shock- and long-term chill hardening-induced chilling tolerance of J. curcas seedlings.  相似文献

Polar lipids were extracted from the leaves of Passiflora species which varied in their resistance to chilling injury. The fatty acid compositions of the 8 major polar lipid classes from P. caerulea (chill-resistant) were generally similar to those of the corresponding lipids from P. flavicarpa (chill-sensitive). Using ESR spectroscopy, the motion of spin-labelled molecules was measured in phospholipids isolated from a range of Passiflora species. The temperature dependence of the motion of the spin labels showed a change at 1° for lipids of the most chill-resistant species and at 9° for the lipids of the most chill-sensitive species. Lipids from other species showed changes at intermediate temperatures, and the greater the chilling sensitivity of the species, the higher was the temperature of the change. It is concluded that pronounced differences in chilling sensitivity of the Passiflora species are correlated with physical differences in their membrane lipids; however, the degree of unsaturation of the lipids is not a reliable guide to chilling sensitivity.  相似文献

In plants, two lipid desaturation pathways exist. A so-called prokaryotic pathway is active in plastids and responsible for unsaturation of 16 carbon fatty acids. An eukaryotic one, in the endoplasmic reticulum, acts on 18 carbon fatty acids. Desaturase activities are affected in stressed plants, and conversely, they have an impact on the capability of plants to adapt to stress. So knowing lipid unsaturation is important for physiological studies. Analysis of lipids by mass spectrometry, in the multiple reaction mode, gives access to the molecular species present in each membrane lipid class. We illustrate the powerfulness of this technique by applying it to phospholipids and galactolipids extracted from plants where the desaturation pathways are present at variable level.  相似文献

Cell suspension cultures of Catharanthus roseus were used to study the effect of temperature on plant cell lipids and indole alkaloid accumulation. Lowering the cultivation temperature increased the total fatty acid content per cell dry weight relative to that at higher temperatures, mainly because of increased accumulation of unsaturated C18 acids. In addition, an increase in the relative proportion of phosphatidylcholine and phosphatidylethanolamine was observed. Within individual lipids, the degree of unsaturation was increased and the mean fatty acid chain length decreased with reducing temperature. These changes may be interpreted as modifying the cell membrane fluidity to keep it optimal for growth and metabolism at each temperature. Selected lipid properties were compared between populations of Carex aquatilis occupying adjacent hot spring and permafrost-dominated sites in interior Alaska. Analyses were carried out on a variety of plant parts and results correlated with prevailing ambient temperature. Phospholipid concentrations were higher in plants from permafrost than from hot spring areas, even though there was no consistent interpopulation difference in total lipid content. The population differences in phospholipid content were greatest in those plant parts that experienced the greatest interpopulation temperature difference. Despite lower soil phosphorus availability, permafrost plants had higher total phosphorus concentrations, and a larger proportion of their phosphorus was contained in phospholipid than in hot spring plants. There was no consistent relationship between the proportion of various classes of phospholipids present and prevailing ambient temperature. Fatty acid chain length and degree of unsaturation of the phospholipid fraction were inversely correlated with temperature both between populations and among plant parts. No such relationship was found in the fraction containing neutral lipids. We suggest that the high tissue phosphorus content that characterizes northern plants may be necessary in part to maintain large quantities of phospholipid-containing membrane. Increased amounts of membrane in turn may be a prerequisite for the high rates of membrane-associated processes characteristic of plants in the cold subarctic environment.  相似文献

Vegetable oils promoted mycelial growth ofVolvariella volvacea. Ethyl esters of major components of saponified fatty acids (palmitic, stearic, oleic and linoleic acid) from vegetable oils were stimulatory. The stimulatory effect of these fatty acids varied with concentration and degree of unsaturation; relatively high concentrations being inhibitory. Mycelial growth appears to be promoted by low concentrations of fatty acids. Supplementation of growth medium with sunflower oil altered membrane permeability and this resulted in an increased uptake of glucose. The total mycelial lipids accounted for only 30% of consumed lipids, the remainder being metabolized. The failure of the fungus to adjust the degree of unsaturation in membrane lipids when it was transferred to 0°C may partially explain its susceptibility to chilling injury.  相似文献

Direct analysis of membrane lipids by liquid chromatography-electrospray mass spectrometry was used to demonstrate the role of unsaturation in ether lipids in the adaptation of Methanococcoides burtonii to low temperature. A proteomics approach using two-dimensional liquid chromatography-mass spectrometry was used to identify enzymes involved in lipid biosynthesis, and a pathway for lipid biosynthesis was reconstructed from the M. burtonii draft genome sequence. The major phospholipids were archaeol phosphatidylglycerol, archaeol phosphatidylinositol, hydroxyarchaeol phosphatidylglycerol, and hydroxyarchaeol phosphatidylinositol. All phospholipid classes contained a series of unsaturated analogues, with the degree of unsaturation dependent on phospholipid class. The proportion of unsaturated lipids from cells grown at 4 degrees C was significantly higher than for cells grown at 23 degrees C. 3-Hydroxy-3-methylglutaryl coenzyme A synthase, farnesyl diphosphate synthase, and geranylgeranyl diphosphate synthase were identified in the expressed proteome, and most genes involved in the mevalonate pathway and processes leading to the formation of phosphatidylinositol and phosphatidylglycerol were identified in the genome sequence. In addition, M. burtonii encodes CDP-inositol and CDP-glycerol transferases and a number of homologs of the plant geranylgeranyl reductase. It therefore appears that the unsaturation of lipids may be due to incomplete reduction of an archaeol precursor rather than to a desaturase mechanism. This study shows that cold adaptation in M. burtonii involves specific changes in membrane lipid unsaturation. It also demonstrates that global methods of analysis for lipids and proteomics linked to a draft genome sequence can be effectively combined to infer specific mechanisms of key biological processes.  相似文献

The effect of change in ambient temperature on fatty acid unsaturation has been studied in the cyanobacteriumAnabaena variabilis. When cells isothermally grown at 22°C are compared with those grown at 38°C, the relative content of oleic acid decreases and that of linolenic acid increases in all of the lipid classes. After a temperature shift from 38 to 22°C, palmitic acid is rapidly desaturated in monogalactocyldiacylglycerol, but in no other lipids, and oleic acid is slowly desaturated in most lipid classes. When cells ofAnacystis nidulans are exposed to low temperature such as 0°C, they lose physiological activities and finally die. This low-temperature damage is initiated by the phase transition of lipids in the plasma membrane. The phase transition of thylakoid membrane that occurs at intermediate temperature produces loss of activity related to photosynthesis. This is, however, recovered when the cells are rewarmed to growth temperature. A model for the mechanism of the low-temperature damage in the cyanobacterial cells is proposed.  相似文献

Environmental temperature change may induce modifications in membrane lipid properties and composition, which account for different physiological responses among plant species. Coffee plants, as many tropical species, are particularly sensitive to cold, but genotypes can present differences that can be exploited to improve crop management and breeding. This work intended to highlight the changes promoted by low non-freezing temperatures (chilling) in phospholipid (PL) composition of chloroplast membranes of genotypes from two Coffea species, Coffea arabica cv. Catuaí (moderately tolerant) and Coffea canephora cv. Conilon (Clone 153, more susceptible), and relate them with cold sensitivity differences. Such evaluation was performed considering a gradual temperature decrease, chilling (4 °C) exposure and a recovery period under rewarming conditions. Catuaí presented an earlier acclimation response than Clone 153 (CL 153). It displayed a higher metabolic activity during acclimation (total fatty acids and total PL increases) and chilling (phosphatidylglycerol increases), and an overall better recovery. Catuaí also showed the highest phosphatidylglycerol unsaturation (higher double bond index) after chilling, in contrast with CL 153 (gradual unsaturation decrease). Higher unsaturation degree in Catuaí than in CL 153 was also observed for phosphatidylcholine and phosphatidylinositol, resulting, mainly, from raises in unsaturated C18:2 and C18:3. It is suggested that an enhanced PL synthesis and turnover induced by a gradual cold exposure, as well as unsaturation increases in major PL classes, is related to decreased Catuaí susceptibility to low temperatures and strongly contributes to sustain photosynthetic activity in this genotype under chilling conditions, as reported in previous work by this team.  相似文献

The effects of cadmium (Cd) uptake on ultrastructure and lipid composition of chloroplasts were investigated in 28-day-old tomato plants (Lycopersicon esculentum var. Ibiza F1) grown for 10 days in the presence of various concentrations of CdCl2. Different growth parameters, lipid and fatty acid composition, lipid peroxidation, and lipoxygenase activity were measured in the leaves in order to assess the involvement of this metal in the generation of oxidative stress. We first observed that the accumulation of Cd increased with external metal concentration, and was considerably higher in roots than in leaves. Cadmium induced a significant inhibition of growth in both plant organs, as well as a reduction in the chlorophyll and carotenoid contents in the leaves. Ultrastructural investigations revealed that cadmium induced disorganization in leaf structure, essentially marked by a lowered mesophyll cell size, reduced intercellular spaces, as well as severe alterations in chloroplast fine structure, which exhibits disturbed shape and dilation of thylakoid membranes. High cadmium concentrations also affect the main lipid classes, leading to strong changes in their composition and fatty acid content. Thus, the exposure of tomato plants to cadmium caused a concentration-related decrease in the fatty acid content and a shift in the composition of fatty acids, resulting in a lower degree of fatty acid unsaturation in chloroplast membranes. The level of lipid peroxides and the activity of lipoxygenase were also significantly enhanced at high Cd concentrations. These biochemical and ultrastructural changes suggest that cadmium, through its effects on membrane structure and composition, induces premature senescence of leaves.  相似文献

This study aimed to evaluate the effect of Cd exposure (100 μmol/L) on polar lipid composition, and to examine the level of fatty acid unsaturation in maize (Zea mays L.). In roots, the level of 16:0 and monounsaturated fatty acids (16:1 + 18:1) decreased in phosphatidylcholine (PC) and phosphatidylethanolamine (PE). In contrast, the proportion of unsaturated 18-C fatty acid species showed an opposite response to Cd. The content, on the other hand, of PC, PE, digalactosyldiacylglycerol (DGDG), and steryl lipids increased in roots (2.9-, 1.6-, 5.3-, and 1.7-fold increase, respectively). These results suggest that a more unsaturated fatty acid composition than found in control plants with a concomitant increase in polar lipids may favor seedling growth during Cd exposure. However, the observed increase in the steryl lipid (SL) : phospholipid (PL) ratio (twofold), the decrease in monogalactosyldiacylglycerol (MGDG) : DGDG ratio, as well as the induction of lipid peroxidation in roots may represent symptoms of membrane injury. In shoots, the unsaturation level was markedly decreased in PC and phosphatidylglycerol (PG) after Cd exposure, but showed a significant increase in sulfoquinovosyldiacylglycerol (SQDG), MGDG and DGDG. The content of PG and MGDG was decreased by about 65%, while PC accumulated to higher levels (4.4-fold increase). Taken together, these changes in the polar lipid unsaturation and composition are likely to be due to alterations in the glycerolipid pathway. These results also support the idea that the increase in overall unsaturation plays some role in enabling the plant to withstand the metal exposure.  相似文献

A detailed analysis of the low temperature-induced alterations of Dunaliella salina (UTEX 1644) microsomal membrane lipids was carried out. Microsomal membranes were isolated from cells grown at 30 degrees C, from cells shifted to 12 degrees C for 12 hours, and from cells acclimated to 12 degrees C. Fatty acid analyses of the major lipid classes demonstrated significant changes in the fatty acid composition of phosphatidylcholinemine (PE) and phosphatidylglycerol (PG) but not phosphatidylcholine (PC) during the initial 12 hours at low temperature. These changes did not entail enhanced desaturation of linoleic acid. Subsequent to 12 hours, the proportions of linolenic acid increased in all phospholipids.Molecular species analyses of the phospholipids demonstrated that the most immediate changes following a shift to low temperature were limited to several molecular species of PE and PG. The changes observed in PE included a decrease in C(30) species and concomitant increases in C(34) and C(36) species. Compositional changes associated with PG entailed the emergence of a new molecular species (18:1/18:1) not found at 30 degrees C. The retailoring of molecular species resulted in an increase in the number of species having two unsaturated acyl chains and did not reflect a simple enhancement of desaturase activity as suggested by the fatty acid analysis. We conclude that the initial alterations in response to low temperature stress involve discrete changes in certain molecular species. These and further alterations of molecular species following acclimation to low temperature would appear to augment increases in acyl chain desaturation as a means of modifying membrane properties in response to low temperature stress.  相似文献

Understanding how wheat (Triticum aestivum L.) plants under high temperature (HT) regulate lipid composition is critical to developing climate‐resilient varieties. We measured 165 glycerolipids and sterol derivatives under optimum and high day and night temperatures in wheat leaves using electrospray ionization‐tandem mass spectrometry. Levels of polar lipid fatty acyl chain unsaturation were lower in both heat‐tolerant genotype Ventnor and susceptible genotype Karl 92 under HT, compared with optimum temperature. The lower unsaturation was predominantly because of lower levels of 18:3 acyl chains and higher levels of 18:1 and 16:0 acyl chains. Levels of 18:3‐containing triacylglycerols increased threefold/more under HT, consistent with their possible role in sequestering fatty acids during membrane lipid remodelling. Phospholipids containing odd‐numbered or oxidized acyl chains accumulated in leaves under HT. Sterol glycosides (SG) and 16:0‐acylated sterol glycosides (ASG) were higher under HT than optimum temperatures. Ventnor had lower amounts of phospholipids with oxidized acyl chains under HT and higher amounts of SG and 16:0‐ASG than Karl 92. Taken together, the data demonstrate that wheat leaf lipid composition is altered by HT, in which some lipids are particularly responsive to HT, and that two wheat genotypes, chosen for their differing physiological responses to HT, differ in lipid profile under HT.  相似文献

Cultivation of Catharanthus roseus hairy root cultures at different temperatures was found to have an effect on growth rate and indole alkaloid content as well as lipid composition. When lowering the temperature, the roots responded by increasing the degree of unsaturation of cellular lipids, which was mainly due to an increased proportion of linolenic acid in the main lipid classes. The modifications in lipid composition were obviously necessary for the roots to retain the proper cell membrane fluidity at each temperature. Despite of changes in membrane lipids, no effect on the distribution of indole alkaloids between the roots and the medium could be detected. The fatty acid compositions of plastidic and extra-plastidic membrane lipids of two metribuzin-resistant cell lines L4 and L7 of Chenopodium rubrum were determined after growth in the absence and in the presence of the herbicide and compared with those of wild type cells. Fatty acid biosynthesis was markedly affected in all cell lines by metribuzin treatment. In the absence and in the presence of metribuzin alterations of the fatty acid composition of the various lipid classes were, as compared to wild type cells, generally lower in the highly resistant L4 cells than in the less resistant L7 cells. The two resistant cell lines demonstrated a higher degree of unsaturation within the plastidic monogalactosyldiacylglycerols (L4 cells also within plastidic digalactosyldiacylglycerols) and, particularly, within the predominantly extra-plastidic phosphatidylcholines (L7 cells also within the predominantly extra-plastidic phosphatidylethanolamines), whereas the degree of unsaturation was slightly altered in the plastidic phosphatidylglycerols. Within the two metribuzin-resistant cell lines, the highly resistant L4 cells differed from the less resistant L7 cells by increased alpha-linolenic acid/palmitic acid ratios in both the plastidic and extra-plastidic membrane lipids suggesting that particularly in L4 cells higher proportions of linolenate are formed as a result of selection pressure. On the other hand, the proportion of linoleate was increased predominantly in extra-plastidic membrane lipids of both L4 and L7 cells which explains a raise in linoleic acid/palmitic acid ratios in both cell lines as compared to wild-type cells. Moreover, in the absence of metribuzin decreased proportions of trans-3-hexadecenoic acid were found in phosphatidylglycerols of L4 and, particularly, of L7 cells as compared to the wild type cells. It is suggested that L4 and L7 cells--having multiple mutations in the psbA gene as observed earlier--are additionally characterized by increased degree of unsaturation of acyl moieties in various polar lipids, e.g. linoleoyl moieties in L4 and L7 cells as well as linolenoyl moieties particularly in highly resistant L4 cells. This increase gives rise to a change in membrane fluidity and may finally lead to increased metribuzin resistance.  相似文献

The influence of the acclimation temperature on the thermotropic behaviour of mitochondrial respiration and on the degree of unsaturation of mitochondrial membrane lipids has been studied. The mitochondria were isolated from red muscle, white muscle and liver of goldfish acclimated to 5, 20 and 30 degrees C. ADP-activated succinate oxidation was measured at different temperatures and resulted in non-linear Arrhenius-plots with breaks between 10 and 23 degrees C. As for the break-temperatures, there was found a shift downwards in preparations of decreased acclimation temperatures. This could be caused by a changed composition of membrane lipids and a simultaneous shift of the membrane phase transition temperature. Therefore, the fatty acid composition of all membrane preparations was analyzed. However, no consistent change of the degree of unsaturation due to a changed acclimation temperature could be found.  相似文献
